金藻3011和8701与球等鞭金藻的18S rRNA基因研究

摘    要:对金藻3011和8701的18S rRNA基因序列进行测定,分别获得1695 bp和1684 bp的DNA序列。应用DNAMAN,DNAstar和RnaViz 2.0生物软件,将获得的DNA序列与球等鞭金藻的18S rRNA基因序列进行DNA序列和RNA二级结构对比分析。DNA序列分析显示,三者的18S rRNA基因序列非常保守,相似性在99.5%以上,三者应同为球等鞭金藻;RNA二级结构分析显示,三者的RNA二级结构既存在球等鞭金藻种的特异性茎环结构,又存在明显的差异,并且从相近水域(山东)分离出来的3011和8701相对于采自挪威水域的球等鞭金藻具有更大的结构相似性;相对于3011,8701可能与球等鞭金藻具有更高的同源性。由此可见,单纯的18S rRNA基因序列分析可能不适用于球等鞭金藻种下水平的研究,但是其RNA二级结构分析对球等鞭金藻的物种鉴定,甚至是种下地理株的研究都具有非常重要的意义。

Abstract:The 18S rRNA gene sequences of Isochrysis 3011 and Isochrysis 8701 were determined,and 1695 bp and 1684 bp DNA sequences were found,respectively.The DNA sequence and RNA secondary structure in 18S rRNA genes were comparatively analyzed in Isochrysis 3011,Isochrysis 8701 and I.galbana by bio-soft DNAMAN,DNAstar and RnaViz 2.0.The results showed that the 18S rRNA gene sequences of Isochrysis 3011,Isochrysis 8701 and I.galbana are quite conservative,with the similarity above 99.5%,indicating that the Isochrysis 3011,Isochrysis 8701 and I.galbana are all the same species.The results of RNA secondary structure analyses indicated that their RNA secondary structures have specific stem and hairpin loop which only I.galbana exists,while the different stem hairpin loop exist distinctly among them.Besides,the 3011 and 8701 collected from nearby waters in Shandong province,China,are more similar than I.galbana sampled from Norway in the RNA secondary,and for Isochrysis 3011,Isochrysis 8701 may be more homologous to I.galbana.In overall,only 18S rRNA gene sequence analyses can be unadvisable to use for down-species of I.galbana study.But their RNA second structure analyses are useful for the study on species of I.galbana identification,even the analyses among different geographical strains of I.galbana.
