贵州地方黄牛不同组织中PPARγ基因表达水平研究

摘    要:试验旨在研究威宁牛、思南牛、关岭牛和黎平牛不同组织中过氧化物酶体增殖物激活受体γ(peroxisome proliferators-activated receptors γ,PPARγ)基因mRNA的表达差异。以这4个贵州地方品种黄牛为试验动物,提取心脏、肝脏、脾脏、肺脏、肾脏、脂肪和背最长肌组织的总RNA,设计PPARγ基因的实时荧光定量引物,以牛GAPDH基因为内参,应用实时荧光定量PCR技术检测PPARγ基因在4个品种不同组织中mRNA的相对表达量。结果显示,PPARγ基因在4个品种黄牛的心脏、肝脏、脾脏、肺脏、肾脏、脂肪和背最长肌组织中均有表达,但在脂肪组织中的表达量高于其他组织,且差异极显著(P<0.01),在脾脏和肺脏中也有较高表达,而在肾脏、肝脏、心脏和背最长肌中表达量较低;不同品种PPARγ基因的表达在部分组织中存在差异。研究表明,PPARγ基因在不同组织中的表达量存在差异,可能与其在不同组织中的功能有关,而品种对于PPARγ基因的表达影响不大。

Abstract:The objective of this study was to investigate the expression level of PPARγ gene mRNA in different tissues from Weining cattle, Sinan cattle, Guanling cattle and Liping cattle in Guizhou province. Total RNA was extracted from the heart, liver, spleen, lung, kidney, adipose and longissimus dorsi of the cattle resources. Primers of PPARγ gene were designed. The developmental changes of PPARγ gene mRNA relative expression in different tissues from 4 kinds of cattle breeds were detected by Real-time PCR, taking GAPDH gene as the internal reference. The results showed that PPARγ gene was expressed in all tissues. The mRNA expression in adipose tissue was extremely significantly higher than that in other tissues (P<0.01), with a higher expression in the spleen and lung as well, while a lower expression in the kidney, liver, heart and longissimus dorsi.The expression of PPARγ gene in the different cattle resouces was different in partial tissues. The results suggested that PPARγ gene mRNA expression existed differences in the tissues,which might be due to its function in different tissues, but different cattle resouces did not basically influence the PPARγ gene mRNA expression.
