凝结芽孢杆菌芽孢高产固体发酵的研究

摘    要:为研究凝结芽孢杆菌(Bacillus coagulans)固体发酵大量生成芽孢的方法,就固体发酵的不同底物(麦麸、糠粉、玉米粉和谷草粉)、湿度、初始pH值和添加物质(碳源、氮源和盐类)等因子进行单因素试验,探索芽孢形成的最佳的培养条件;其次采用正交设计法对固体发酵培养基优化.结果表明底物的理化性质对固体发酵生产芽孢影响明显,其中以麦麸为底物进行固体发酵时,促进芽孢形成的能力最强.在以麦麸为底物,温度37℃,湿度为60%,初始pH7.5,添加外源物质为葡萄糖1.00%、蛋白胨2.00%,KH2PO40.50%,MnSO40.05%所形成的固体培养条件下进行48 h的发酵,芽孢生成进一步提高,优化验证试验结果表明凝结芽孢杆菌的芽孢产量可以达3.80×1010个/克以上,明显高于对照组2.98×109个/克.

Abstract:In order to study the method for spore production of Bacillus coagulans using solid state fermentation (SSF),the SSF conditions involving the substrate(wheat bran,rice bran,corn flour and straw powder),moisture content,initial pH and additive (carbon sources,nitrogens sources and salts),was investigated through single-factor test,and orthogonal design was used to optimize the SSF medium of B.coagulans.The results showed that spore production of B.coagulans using SSF was influenced by physiological and chemical nature of the agriculture by-product.The optimized SSF conditions such as substrate wheat bran,moisture content 60%,temperature 37℃,initial pH 7.5.1.0% glucose,2.0% peptone,0.50% KH2PO4,0.05% MnSO4 were determined.When SSF was adopted the optimized conditions,the spore biomass concentration of B.coagulans increased from 3.80 × 1010 cfu/g to 2.98× 109 cfu/g under the condition of temperature 37 ℃ in 48 h.
