羊瘤胃乳酸菌的分离鉴定及抗逆性筛选

摘    要:为开发羊源乳酸菌菌种资源,从健康成年散养山羊瘤胃内容物中分离乳酸菌,经生化试验、基因测序以及同源性分析、构建遗传进化树等方法对分离出的乳酸菌进行鉴定,并通过耐酸和耐胆盐试验筛选抗逆性良好的乳酸菌。结果表明:从散养山羊瘤胃内容物中分离出11株乳酸菌,将其编号为R1~R9、R11和R12;其中R1、R2为杜氏肠球菌,R3、R7为乳酸肠球菌,R4、R5、R6、R8、R9为屎肠球菌,R11为鼠李糖乳杆菌,R12为莱菲氏肠球菌;R4(屎肠球菌JQ067694.1)在pH值为2时存活率超过80%,在0.9%浓度胆盐下,存活率超过30%,抗逆性优于其他菌株,生产应用潜力较好。

Abstract:The purpose of this paper is to develop the resources of lactic acid bacteria from sheep.Lactic acid bacteria were isolated from the rumen contents of healthy adult free-range goats,and the isolated lactic acid bacteria were identified by biochemical test,gene sequencing,homology analysis,construction of genetic evolution tree,and so on.The lactic acid bacteria with good stress resistance were screened by acid tolerance and bile salt tolerance test.The results showed that 11 strains of lactic acid bacteria were isolated from the rumen contents of free-range goats and numbered as R1-R9,R11 and R12.Among them,R1 and R2 were Enterococci,R3 and R7 were Enterococci,R4,R5,R6,R8,R9 were Enterococcus faecium,R11 was Lactobacillus rhamnoides,and R12 was Enterococcus Leifei.The survival rate of R4(Enterococcus faecium JQ067694.1)in pH=2 was more than 80%,and in 0.9%concentration of bile salt,the survival rate was more than 30%,and the stress resistance was better than other strains,indicating that strain R4 had good production and application potential.
