降解黄曲霉毒素枯草芽孢杆菌的解毒性、抗菌性及抗逆性研究

摘    要:试验对筛选得到的高效降解黄曲霉毒素B1、G1和M1的枯草芽孢杆菌(霉立解060菌)进行解毒活性、抗菌活性及抗逆性研究。将枯草芽孢杆菌发酵液分成上清液、菌悬液和胞内液组分,验证各组分对黄曲霉毒素的解毒效率。试验结果表明,枯草芽孢杆菌发酵后的上清液对AFB1、AFG1和AFM1的降解活性最高,确定发酵后的上清液中存在解毒活性组分;对解毒活性物质进行加热和蛋白酶K变性处理后,解毒活性显著降低,表明起解毒作用的活性物质是其分泌的一种胞外酶;试验进一步证明枯草芽孢杆菌具有显著抑制大肠杆菌、沙门氏菌以及金黄色葡萄球菌,抵抗胃酸、胆盐、高温等不利环境的特性,具有很强的抗菌性和抗逆性。

Abstract:Detoxification,antimicrobial activity and resistance to the simulated gut environments of Bacillus subtilis(strain MLJ060) possesing aflatoxin B1,G1 and M1 degradation activity were preliminarily studied in this research.Broth culture of MLJ060 were divided into different components including culture supernatant,bacteria cell and cell extracts.Results implied that the activity of aflatoxins degradation mainly happened in the culture supernatant of strain MLJ060 rather than in its cells or cell extracts.Characteristic research of active component suggested that the effect of supernatant from strain MLJ060 on aflatoxin might be due to the enzymatic reactions.In the present research,strain MLJ060 showed the antimicrobial activities against escherichia coli,salmonella typhimurium,staphylococcus aureus and provided resistance to the simulated gut environments.Hence strain MLJ060 can not only be used as probiotics but also as antidote for aflatoxins.
