胚胎期大鼠性腺生长与分化的研究

摘    要:本试验旨在研究胚胎期大鼠性腺生长与分化的情况。选取12.5~15.5 dpc SD大鼠胚胎为研究对象,运用PCR技术进行大鼠胚胎性别鉴定,采用H-E技术对大鼠性腺分化形态进行观察。结果表明:12.5 d的鼠胚肾管已经开始形成,生殖嵴已经建立,此时仍无明显的性别分化形态;13.5 d的鼠胚开始出现性别分化的迹象,雄性的原始性索开始形成,雌性性腺分化比雄性稍晚,此期仍不易辨别出典型的卵巢特征结构;14.5 d的鼠胚性腺形态初步成型,此期性别明显分化,雄性的原始性索开始分化为实心原始生精小管,雌性胚胎中性腺分为两层,初步形成卵巢特征;15.5 d的鼠胚,雄性胚胎性腺中已经具有明显的曲精小管的雏形,雌性胚胎卵巢特征也开始明显。大鼠胚胎性腺从13.5 d胚龄时开始分化,15.5 d胚龄性腺特征明显。

The study of the growth and differentiation of embryonic rat gonads
Abstract:The experiment was to study the growth and differentiation of rat embryonic gonads. In this experiment, six nest of 12.5~15.5 dpc embryos of SD rats, were selected and the sex was identified by PCR technology. Observation of gonadal differentiation char-acteristics was conducted on three male rats and three female rats of each embryo ages by using H-E techniques. On the 12.5 days' embryo, the renal tubular and genital ridge began to form, but still no clear sex differentiation morphology was observed in the rat em-bryos. On the 13.5 days' embryo, sex differentiation of rat embryos began to appear, the original male sex cord began to form, but it was still not easy to identify the typical ovarian characterized structure in female gonadal. But the initial shape of mouse embryo go-nadal morphology was obviously found on the 14.5 days' embryo. The original male sex cord began to differentiate to the solid original spermatogenic tubules. But the female embryonic gonads had two layers, and showed the initial formation of ovarian characteristics. On the 15.5 days' embryo, the prototype of the seminiferous tubules had been obviously observed in the gonads of male embryonic rats, and the female embryos ovarian characteristics were also clearly found. Therefore, the time of the rat gonads differentiation was on the 13.5th day and the gonad characteristics significantly was on the 15.5th day.
