猪伪狂犬病病毒基因缺失灭活疫苗(LA-A株)的最小免疫剂量和制品保存期的研究

摘    要:本研究将伪狂犬病病毒(Pseudorabies virus,PRV)变异株(PRV AH02LA株)的gE基因缺失株(LA-A株)接种BHK-21细胞,经纯悬浮培养制备抗原,甲醛灭活后制备油乳剂灭活疫苗,并确定该灭活疫苗的最小免疫剂量和效力检验方法,以及在2~8℃保存期。结果显示:猪伪狂犬病病毒基因缺失灭活疫苗(LA-A株)的效力检验方法为以2.0 mL(抗原含量108.20TCID50)接种4~5周龄PRV阴性健康仔猪,颈部肌肉注射,间隔28 d以相同剂量和方法加强免疫,加强免疫后第21 d,免疫猪血清PRV抗体中和指数应不低于10000,攻毒保护率应不低于80%;最小免疫剂量为1.0 mL(抗原含量107.90 TCID50);制品保存期:在2~8℃保存期为18个月。该研究结果为新型疫苗的研制提供了重要的试验依据。

Abstract:In this study,suspension cultures of BHK-21 cells were inoculated with a gE gene-deleted strain LA-A of Pseudorabies virus(PRV)that derived from an emerging PRV variant strain AH02 LA.The harvested viral fluid was inactivated by formaldehyde and formulated into water-in-oil emulsion.This inactivated oil adjuvanted LA-A vaccine was tested for its minimum immunization dosage in pigs and storage stability at 2-8℃.Healthy PRV-antibody-negative piglets at age of 4-5 weeks old were intramuscularly injected twice with 2.0 mL of the LA-A vaccine produced from viral fluid(108.20 TCID50)at an interval of 28 days.The vaccinated pigs were challenged with virulent AH02 LA strain 21 days post the booster vaccination.The efficacy of the LA-A vaccine met the satisfactory requirements including serum PRV antibody neutralization index at no less than 10000 and the protection rate at no lower than 4/5 pigs vaccinated/challenged.Accordingly,the minimal immunization dosage of the LA-A vaccine was determined as 1.0 mL prepared from viral fluid containing 107.90 TCID50.The efficacy of this LA-A vaccine was tested satisfactory for 21 months stored at 2-8℃.Therefore,the preservation period of this LA-A vaccine was determined as 18 months.
