Protection provided by live and inactivated virus vaccination against challenge with the virulent nephropathogenic infectious bronchitis virus (NIBV) strain PA/Wolgemuth/98 was assessed. Vaccinations with combinations of live attenuated strains Massachusetts (Mass) + Connecticut (Conn) or Mass + Arkansas (Ark) were given by eyedrop to 2-wk-old specific-pathogen-free leghorn chickens. After live infectious bronchitis virus (IBV) vaccination, some chickens at 6 wk of age received an injection of either an oil emulsion vaccine containing inactivated IBV strains Mass + Ark or an autogenous vaccine prepared from NIBV PA/Wolgemuth/98. Challenge with PA/Wolgemuth/98 was given via eyedrop at 10 wk of age. Serum IBV enzyme-linked immunosorbent assay antibody geometric mean titers (GMTs) after vaccination with the combinations of live attenuated strains were low, ranging from 184 to 1,354, prior to NIBV challenge at 10 wk of age. Both inactivated vaccines induced an anamnestic response of similar magnitudes with serum GMTs of 6,232-12,241. Assessment of protection following NIBV challenge was based on several criteria virus reisolation from trachea and kidney and renal microscopic pathology and IBV-specific antigen immunohistochemistry (IHC). Live attenuated virus vaccination alone with combinations of strains Mass + Conn or Mass + Ark did not protect the respiratory tract and kidney of chickens after PA/Wolgemuth/98 challenge. Chickens given a live combination vaccination of Mass + Conn and boosted with an inactivated Mass + Ark vaccine were also susceptible to NIBV challenge on the basis of virus isolation from trachea and kidney butshowed protection on the basis of renal microscopic pathology and IHC. Live IBV-primed chickens vaccinated with an autogenous inactivated PA/Wolgemuth/98 vaccine had the highest protection against homologous virulent NIBV challenge on the basis of virus isolation.  相似文献

Despite the existence of an active vaccination program, recently emerged strains of nephropathogenic infectious bronchitis virus (IBV) in Korea have caused significant economic losses in the poultry industry. In this study, we assessed the pathogenic and antigenic characteristics of a K-IIb type field strain of IBV that emerged in Korea since 2003, such as Kr/Q43/06. Specific pathogen free 1-week-old chickens exhibited severe respiratory symptoms (dyspnea) and nephropathogenic lesions (swollen kidneys with nephritis and urate deposits) following challenge with the recent IBV field strain. The antigenic relatedness (R value), based on a calculated virus neutralization index, of the K-IIb type field strain and K-IIa type strain KM91 (isolated in 1991) was 30%, which indicated that the recent strain, Kr/Q43/06, is a new variant that is antigenically distinct from strain KM91. This report is the first to document the emergence of a new antigenic variant of nephropathogenic IBV in chicken from Korea.  相似文献

Two serological tests, the virus-neutralization (VN) test in tissue culture using a tissue-cell-adapted virus and the enzyme-linked immunosorbent assay (ELISA), were compared to detect antibodies against Massachusetts 41 and Connecticut 46 strains of Infectious Bronchitis Virus (IBV). The VN test was conducted in wells of microplates by the usual procedure. The two strains of IBV were adapted after 20 serial passages to induce CPE in 24 hours in chickens embryos kidney cells (CEKC). The ELISA test was carried out using partially virus following ultracentrifugation of each stain of IBV as antigen. The ELISA test detected higher geometric mean antibody titers (GMT) against both strains of IBV than did the VN test. One hundred four serum samples taken at 1, 3, 5, 9, 22, 24, and 26 weeks of age from a flock of chickens vaccinated with the Mass strain three times and the Conn strain of IBV two times during the growing period showed higher antibody titer responses to the Conn 46 than to the Mass 41 strain. Maternal antibodies in chicks one week of age were readily detected by the ELISA test, whereas low or insignificant titers were found by the VN test. Sera of vaccinated chickens collected following challenge with Mass 41 or Conn 46 strain of IBV showed that the ELISA was more sensitive and showed higher titers than did the VN test. Although the VN test showed no rise in GMT in the same sera tested with the heterologous virus, the ELISA showed a slight increase or cross-reaction. The serum samples from the unchallenged control group showed no change in GMT with either test or IBV strain.  相似文献

从山东省发病鸡群中分离鉴定了一株鸡传染性支气管炎病毒(Infectious bronchitis virus,IBV)强毒株SDIB821/2012,对其进行S1基因序列测定分析和免疫保护试验。S1基因遗传进化分析结果显示,SDIB821/2012属于以QXIBV为代表的基因型,与同属一个基因型的IBV参考株氨基酸同源性为91.6%~98.5%,与疫苗株491同源性为77.6%,与H120和MA5同源性均为74.8%。免疫保护试验结果显示,根据试验鸡临床症状和发病死亡情况,弱毒活疫苗491对SDIB821/2012的保护率为90%,而H120和MA5对SDIB821/2012的保护率分别仅为40%和33%。攻毒后各免疫组喉头、泄殖腔棉拭样品以及气管、肺脏和肾脏组织均可检测到病毒,表明3种IB疫苗均不能对SDIB821/2012提供完全的免疫保护。  相似文献

Six isolates originating from acute outbreaks of infectious bursal disease recently reported in broiler and pullet flocks in France were studied with respect to their pathogenicity and their antigenic relatedness to the Faragher 52/70 reference strain. Although the mortality experimentally induced in susceptible chickens by the field strains was sometimes four times higher than that which followed the inoculation of the reference strain (16 to 48% versus 12%), neither mortality nor morbidity were observed in chickens previously vaccinated with a commercial live vaccine and then challenged under the same conditions. Agar gel precipitation tests demonstrated the existence of common antigens in the different strains, and high cross-neutralization indices measured in embryonated specific pathogen free eggs showed them all to belong to serotype I. These data are discussed with reference to previous European and North-American studies on the antigenic status of infectious bursal disease virus.  相似文献

The protective efficacy of three infectious bronchitis virus (IBV) vaccines for sentinel chickens raised with commercial Delmarva broiler chickens was evaluated during winter 1987. Specific-pathogen-free leghorn sentinel chickens were vaccinated with Massachusetts (Mass) alone, Mass and JMK, or Mass and Arkansas (Ark) combination live vaccines, or they remained unvaccinated. Four weeks post-vaccination, sentinels were placed on broiler farms at weekly intervals for 3 weeks corresponding to weeks 4, 5, and 6 of the broiler growing cycle. Vaccine efficacy was evaluated based on IBV reisolation attempts from tracheal swabbings following a 1-week field exposure period. Sentinel chickens vaccinated with Mass and Ark combination vaccine were best protected against IBV field challenge. Only four IBV isolations were made out of a 3-week total of 36 attempts, for an 11% isolation rate. IBV vaccines containing either Mass alone or Mass and JMK offered much lower levels of protection.  相似文献

Infectious bronchitis virus (IBV) causes avian infectious bronchitis, an important disease that produces severe economic losses in the poultry industry worldwide. Recent IBV infections in Sweden have been associated with poor growth in broilers, drop in egg production and thin egg shells in layers. The complete spike gene of selected isolates from IBV cases was amplified and sequenced using conventional RT-PCR. Nucleotide and amino acid sequence comparisons have shown that the recent isolates bear 98.97% genetic similarity with strains of the QX-like genotype. The phylogenetic analysis revealed that strains predominant in the nineties, which were of the Massachusetts type, have been replaced by D388/QX-like strains, however the evolutionary link could not be established. The homology between the two genotypes was 79 and 81%. Remarkably, a strong positive selection pressure was determined, mostly involving the S1 subunit of the S gene. This strong selective pressure resulted in recombination events, insertions and deletions in the S gene. Two new isolates generated from recombination were found with nucleotide sequence diverging 1.7-2.4% from the D388/QX-like branch, indicating the emergence of a new lineage. The study demonstrates a constant evolution of IBV that might be in relation to increased poultry farming, trade and vaccine pressure. The findings underscore the importance of continuous monitoring to control spread of infections, as well as to timely adjust diagnostic methods, molecular epidemiological studies, development and use of vaccines that are adapted to the changing disease scenario.  相似文献

In this study, we characterized three variant infectious bronchitis virus (IBV) strains isolated in 2003 and 2004 from broiler chickens in California and compared them to previously isolated California variant viruses and to common vaccine serotypes used in the United States. We conducted genetic, serologic, and pathogenicity studies on all three isolates, then tested different vaccines against one of the viruses. Genetically the three variant IBV strains, designated CA557/03, CA706/03, and CA1737/04, were not related to each other. GenBank BLAST database search and phylogenetic analysis of the hypervariable region of the S1 subunit of the spike gene to determine the most closely related viruses to the three variants showed the CA557/03 variant to be 81.8% similar to the CAV/CA56b/91 whereas the CA706/03 and CA1737/04 variant viruses were only distantly related to Dutch/D1466/81 (72.2%), a vaccine strain used in Europe, and Korea/K142/02 (72.7%), a Korean field isolate, respectively. Cross virus-neutralization testing showed that none of the 2003-04 California IBV variant viruses were serologically related to each other or to Ark, Conn, or Mass vaccine strains. In addition the CA1737/04 isolate was also tested against DE072 and found not to be serologically related. All three variant viruses were pathogenic in 1-wk-old broilers and vaccination with Mass/Conn followed by Holland/Conn provided 80% protection against the CA1737/04 virus. The 2003-04 California variant viruses were not compared with variants isolated in California during 1970s and 1980s because, to our knowledge, no genetic information is available and those viruses are no longer obtainable. This study shows that the CA557/03 virus was distantly related to the CAV-type viruses isolated in California in the early 1990s, but that none of the 2003-04 viruses were similar genetically or serologically to the CAL99-type viruses, indicating that new IBV variants continue to emerge and cause disease in commercial chickens in California.  相似文献

将鸡传染性支气管炎病毒肾型 T 株 S1 基因c D N A 连接于pc D N A3 的 Hind I I I与 Ba m H I位点之间构建含有 C M V 启动子及 B G Hpoly A 信号序列的鸡传染性支气管炎病毒 S1 蛋白真核表达质粒。实验证明, S P F 鸡肌注免疫后血清 Ig G 抗体逐渐升高,至第35 日龄左右达到高峰,攻毒后血清 Ig G 抗体先下降而后升高,血清 Ig G 抗体升高幅度不及 I B 油苗免疫组。质粒 D N A 免疫鸡攻毒后有40 % 的鸡可耐过强毒的攻击,说明 S1 基因在体内获得了表达并使鸡产生了一定的免疫力。  相似文献

A stable recombinant fowl poxvirus (rFPV) expressing the C-terminal region (119 amino acids) of the nucleocapsid (N) protein of an infectious bronchitis virus (IBV) strain Ch3 was constructed by inserting the coding sequence within the thymidine kinase gene of fowl poxvirus (FPV) by homologous recombination. The N protein was expressed under control of the vaccinia virus promoter P7.5 in chicken embryo fibroblast cell cultures as seen in immunofluorescence assay and in rFPV-inoculated specific-pathogen-free (SPF) chickens by detecting antibodies with enzyme-linked immunosorbent assay (ELISA). A homologous IBV strain (Ch3) and two heterologous IBV strains (Ch5 and H4) were used to inoculate SPF chickens in a challenge to examine the protective efficacy of the rFPV. When the chickens were challenged with IBV Ch3 or Ch5, the control birds had respiratory signs of infections bronchitis, whereas all the vaccinated birds were clinically normal although low levels of the IBV infection were detected by a differential ELISA. In contrast, in the chickens challenged with IBV H4, all control birds and vaccinated birds suffered from the highly lethal IBV H4 infection. Our results suggest that the C-terminal 119 amino acid of the nucleocapsid expressed by FPV is a host-protective antigen and may induce cross-protective immunity against illness among some IBV strains.  相似文献

Serial passage of two infectious bronchitis virus (IBV) vaccine strains in chickens enhanced their capacity to increase the incidence and severity of Mycoplasma synoviae (MS) airsacculitis. Included in this report were the mild Massachusetts-type Connaught strain and the Arkansas 99 vaccine strain of IBV. The Connaught strain and one of two Ark 99 vaccine strains passaged in chickens increased the incidence of airsacculitis markedly compared with nonpassaged virus. The other Ark 99 vaccine virus already exacerbated MS airsacculitis, before passage in chickens, and its influence did not increase on passage. All IBV strains studied to date have either possessed this trait or reacquired it on passage in the natural host.  相似文献

Despite the long-term vaccination programs implemented in China, H9N2 avian influenza viruses (AIVs) continue to persist in chicken populations, even in vaccinated flocks. We previously demonstrated that H9N2 AIV isolated from chickens in China also underwent antigenic drift and evolved into distinct antigenic groups (C, D and E). To understand whether antigenic drift of viruses away from the vaccine strain partially contributed to the circulation of H9N2 AIV in China, we evaluated the protective efficacy of a commercial vaccine against different antigenic groups of H9N2 AIV. Challenge experiments using vaccinated chickens indicated that the vaccine prevented shedding of antigenic group C viruses, but not those of the more recent groups D and E. Vaccinated chickens, even those with vaccine-induced HI titers of 1:1024, shed virus after being infected with A/chicken/Shandong/ZB/2007, a representative virus of antigenic group D. Genetic analysis showed that the representative viruses of antigenic groups D and E possessed greater numbers of amino acid substitutions in the hemagglutinin protein compared to the vaccine strain and the antigenic group C virus, and many of which were located in antigenic sites. Our results indicated that the persistence of H9N2 AIV in China might be due to incomplete vaccine protection, and that the avian influenza vaccine should be regularly evaluated and updated to maintain optimal protection. Furthermore, the avian influenza vaccination policy also needs to be re-assessed, and increased veterinary biosecurity on farms, rather than vaccine application alone, should be implemented to prevent and control avian influenza.  相似文献

In order to verify a commonly held assumption that only Massachusetts (Mass) serotype of infectious bronchitis virus (IBV) was prevalent in the United States between the 1930s (when IBV was first isolated) and the 1950s (when the use of commercial IBV vaccines began), we examined 40 IBV field isolates from the 1940s. Thirty-eight of those isolates were recognized as Mass serotype viruses based on their reactivity to Mass-specific monoclonal antibody (Mab) and neutralization by Mass-specific chicken serum. The remaining two isolates, N-M24 and N-M39, that did not react with Mass-specific Mab, resisted neutralization by Mass-specific chicken serum, and were neutralized only by homologous chicken antibody were identified as non-Mass IBV. When the first 900 nucleotides (nt) from the 5'-end of the spike (S1) glycoprotein gene and their deduced amino acid (aa) sequences were compared, the two non-Mass isolates differed from each other by 24% and 28%, respectively. In a similar comparison, the non-Mass viruses N-M24 and N-M39 differed from M28, a Mass-type isolate from the 1940s, by 21% and 22% (nt) and 28% and 27% (aa), respectively. These data indicate that antigenic and genetic diversity among IBV isolates existed even in the 1940s. Interestingly, when the N-terminal region of the S1 of M28 was compared to that of M41, a prototype Mass virus that has undergone countless number of in vivo and in vitro host passages, the two viruses differed by only 2% (nt) and 4% (aa). This finding suggests that frequent genetic changes are not inherent in all IBV genomes.  相似文献

2015年,从安徽合肥某养鸡场分离出一株H9N2亚型禽流感病毒(AIV),命名为HF株。该毒株鸡胚半数感染量(EID50)为109.17/0.1 mL,最小致死量的平均死亡时间(MDT)为87 h。对其HA基因分析发现,其氨基酸裂解位点为RSSR↓GLF,符合低致病性AIV特征;HA基因的遗传进化分析结果表明,该分离株属于h9.4.2.5谱系,符合当前毒株流行趋势。将HF株与2006-2018年分离自全国各地的10株H9N2亚型AIV分离株同时制备灭活疫苗,免疫SPF鸡,制备阳性血清,通过交叉血凝抑制试验分析病毒抗原性,结果显示HF株与2014年之前毒株抗原相关性介于0.50~0.56之间,与2014年及之后毒株抗原相关性介于0.89~1.00之间,表明该分离株与2014年之后的流行毒株具有良好的抗原相关性。用0.2%甲醛灭活HF株病毒液,其HA效价在灭活前后未发生变化;用灭活抗原制备油乳剂灭活疫苗免疫SPF鸡,免疫后21 d HI抗体效价几何平均值达到9.0log2以上,可使免疫鸡完全抵抗H9亚型AIV的感染,提供100%的攻毒保护。研究结果表明,HF株具有良好的免疫原性,可作为疫苗候选株用于H9N2亚型禽流感疫苗的研制。  相似文献

A model system was used to study infectious bronchitis virus (IBV) and Mycoplasma synoviae (MS) interaction. The system involved exposure of chickens to IBV, followed by exposure to MS 2-5 days later. The chickens were subjected to a cold environment (10 +/- 2 C) for 3 weeks starting one day before MS exposure. Under these conditions, differences in the capacity of various strains of IBV to exacerbate MS airsacculitis was demonstrated. Exposure to IBV field isolates generally resulted in more air-sac lesions than did higher-egg-passaged laboratory strains and vaccine strains. Use of lower-egg-passaged vaccines resulted in a higher incidence of airsacculitis than did higher-egg-passaged vaccines. When chickens were IBV-vaccinated before being used in the model system, the incidence of airsacculitis was lowered, even though the chickens became infected by the challenge virus. Vaccination of MS-free chickens with IBV had no effect on airsacculitis incidence when MS exposure occurred after the vaccine reaction was past.  相似文献

Between 2003 and 2005, four strains of infectious bronchitis virus (IBV) were isolated from the vaccinated chicken flocks in China. The results from chicken embryo cross-neutralization assays showed that all the four isolates were relative to strain A2 of IBV, which was isolated in 1996 in Beijing and related to strain 4/91. The S1 gene of the spike protein was amplified and sequenced. The nucleotide and amino acid sequence of the S1 gene had a similar degree of identity (88.98-99.28%) among the four Chinese IBV isolates. The identity of the S1 protein gene between the four Chinese IBV isolates and 14 strains of other IBVs varied from 70.06 to 81.59%. Phylogenetic analysis suggested that there are at least four groups of IBVs circulating in China and the disease outbreaks might have been caused by infection of multiple strains of IBV.  相似文献

In 2015,an H9N2 subtype avian influenza virus (AIV) strain was isolated from a chicken farm in Hefei,Anhui,and named HF strain.The results of the chicken embryo proliferation characteristics study showed that the half infection rate of chicken embryo (EID50) was 109.17/0.1 mL,and the mean time to death for minimum lethal dose(MDT) was 87 h.The analysis result of HA gene showed that its amino acid cleavage site was located in RSSR↓GLF,which accorded with the characteristics of low pathogenic avian influenza.The genetic evolution analysis of HA gene revealed that the isolate belonged to the h9.4.2.5 lineage,which accorded with the current virus strain epidemic characteristics.The HF strain was prepared with 10 H9N2 subtype AIV isolates which isolated from all over the country from 2006 to 2018 to prepare inactivated vaccines,immunize SPF chickens,prepare positive sera,and analyze the virus antigenicity by cross hemagglutination inhibition test.The results showed that the correlation between the HF strain and the virus antigens before 2014 and was between 0.50-0.56,and the virus antigen correlation after 2014 was 0.89-1.00.This showed that the isolate had good antigenic correlation with epidemic strains in recent years.Inactivate HF strain virus solution with 0.2% formaldehydel,and its HA titer did not change before and after inactivation.After the inactivated virus solution was prepared into an oil emulsion inactivated vaccine to immunize SPF chickens,21 days after immunization,the average value of the HI antibody titer reached 9.0log2.It could make immune chicken completely resistant to H9 subtype AIV infection and provide 100% protection from challenge.The above research results showed that the HF strain had good immunogenicity and could be used as a vaccine candidate strain for the prevention of H9N2 subtype AIV.  相似文献
