将禽巴氏杆菌荚膜粗提物经SephadexG—200分子筛凝胶过滤层析,获得纯化荚膜抗原;然后用反相蒸发法制备脂质体-荚膜抗原(A组),并以单纯荚膜抗原(B组)、禽霍乱“731”菌苗(C组)作对照,进行免疫试验,以酶标单克隆抗体间接ELISA,分别检测IgM和IgG的效价.结果,免疫后3个组的IgM效价相差都不显著(P>0.05),A组的IgG效价明显增高,与其他两组差异非常显著(P<0.01).3个组的IgM、IgG消长规律一致,IgM于免疫后7d开始上升,峰值在14d,其后下降迅速,第8周后降至免疫前水平,IgG于免疫后14d开始明显上升,峰值在28d,其后缓慢下降.保护率,A、B、C组分别为67%、33%、33%.结果表明,脂质体对禽巴氏杆菌荚膜抗原具有明显的佐剂作用.  相似文献

为了研究副鸡禽杆菌荚膜多糖(CPS)-血凝素(HA)蛋白的免疫原性,试验采用B型副鸡禽杆菌CPS与HA蛋白分别对SPF鸡进行单独免疫和联合免疫的方法,比较攻毒后的保护效果。结果表明:CPS单独肌肉注射或眶下窦注射免疫对鸡的保护率不超过30%;HA单独肌肉注射或眶下窦注射免疫对鸡的保护率不超过40%;CPS+HA联合肌肉注射免疫的保护率为60%,眶下窦免疫的保护率达80%。CPS+HA联合免疫组血清抗体水平高于相应免疫方式HA、CPS单独免疫组;与肌肉注射相比,攻毒试验结果显示眶下窦免疫的保护效果好。说明副鸡禽杆菌CPS-HA蛋白具有较好的免疫原性。  相似文献

用酶联免疫吸附试验(ELISA)对抗绿脓杆菌血清学的一种共同抗原(原始内毒素蛋白质)。蛋白酶和弹性蛋白酶的免疫球蛋IgM和IgG抗体的检测。对马血清中绿脓杆菌抗体滴度的测定。利用辣根过氧化物酶标记兔抗马IgM和IgG抗体作为酶标记抗体结合物。利用5氨基水杨酸(5—Aminosalicylic(?)acid)和H_2O_2作为底物。以免疫马、新生驹和72匹健康赛马的血清作为ELISA和被动血球凝集研究的抗绿脓杆菌抗体。通过ELISA,免疫马IgG抗体滴度的变化是清楚的。在新生驹中,出生后第一天,IgM和IgG抗体的数量主要是来自初乳,同时指出,新生驹的抗体水平开始低然后增加。抗绿脓杆菌原始内毒素蛋白质,蛋白酶和弹性蛋白酶的抗体,几乎存在于所有被研究的健康赛马中。  相似文献

为探讨鸡新城疫、禽流感(H9N2 HP株)二联灭活疫苗在商品蛋鸡和商品肉鸡上的免疫效果,以鸡新城疫、禽流感二联灭活疫苗免疫7日龄商品蛋鸡与商品肉鸡,在免疫后第7,14,21,28,35天分别采血检测新城疫与禽流感抗体水平;并于免疫后第14天和第21天分别进行禽流感病毒(H9N2 HP株)攻毒,观察其攻毒保护效果。结果表明:商品蛋鸡免疫组在免疫后第7天新城疫抗体与禽流感抗体开始快速上升,第14天时抗体水平高于4 lb,且抗体峰值高;商品肉鸡免疫组免疫后第14天新城疫抗体与禽流感抗体才开始快速上升,第21天时抗体高于4 lb,抗体峰值低于商品蛋鸡组的峰值。商品蛋鸡免疫组第14天与第21天的攻毒保护率均达到100%,而商品肉鸡组免疫组第14天的攻毒保护率达到了90%,第21天的攻毒保护率也达到了100%,两者均有较好的免疫保护效果。  相似文献

以家畜伊氏锥虫弱毒株免疫家兔12只,然后进行攻虫试验。其中11只被保护,1只发病死亡。以BA-ELISA检测家兔免疫后及攻虫后特异性IgG、IgM的变化,发现免疫后IgG、IgM均于第21d达到峰值;攻虫后IgM于第6d达到峰值,IgG于第9d达到峰值。攻虫后,随着免疫组家兔体内特异性IgG、IgM的不断上升,其血液内的锥虫也同时被清除。体外中和试验也表明,特异性抗体对锥虫的感染力具有明显的抑制作用。  相似文献

鸡传染性支气管炎ELISA抗体检测试剂盒的研制   总被引:1,自引:0,他引:1  
采用加蔗糖垫离心纯化鸡传染性支气管炎病毒(IBV)制备抗原,用提纯的IBV抗原包被微量板,建立了检测鸡传染性支气管炎(IB)抗体的ELISA试剂盒。抗原、被检血清和酶标结合物的最佳工作浓度分别为10ug/ml、1:200和1:3200。与IDEXX试剂盒相比,其敏感性、重复性、特异性均接近国外同类产品水平。对SPF鸡血清、实验免疫与攻毒的SPF鸡血清进行检测,结果表明所建立的ELISA特异性为95.6%,与IDEXX试剂盒符合率为95.6%。用于检测抗IBV特异性IgG抗体发现在免疫接种IB弱毒苗后,第4天即可检测到IgG抗体,峰值在第3周。试验鸡在通过滴鼻、点眼途径人工感染IBV强毒后,第5天抗体滴度明显上升。我们认为,该法是目前我国SPF鸡质量监测、养鸡生产中进行IB监测较好的方法。  相似文献

对8批禽霍乱、新城疫二联灭活疫苗进行新城疫组分的效力检验,在检测新城疫血凝抑制效价(HI)的同时,进行新城疫病毒强毒攻击试验,旨在寻找抗攻毒保护与HI抗体效价之间的平行关系。结果表明:免疫剂量与HI抗体效价和攻毒保护率呈正相关,免疫40、20、10、5μL剂量组攻毒保护率和HI抗体效价分别为94.9%(75/79)、5.2 log_2,87.5%(70/80)、4.4 log_2,29.1%(23/79)、2.7 log_2,11.4%(9/79)、2.1 log_2;对照组攻毒保护率为0(0/10),HI抗体效价≤2.0 log_2;新城疫HI抗体效价与攻毒保护率也呈正相关,当HI3.0 log_2,攻毒保护率为4.1%;HI=3.0 log_2,攻毒保护率为22.9%;HI=4.0 log_2,攻毒保护率为87.7%;HI=5.0 log_2,攻毒保护率为94.4%;HI=6.0 log_2,攻毒保护率为100%。本研究为制定疫苗质量标准中新城疫病毒株的效力检验方法和标准提供实验室理论依据。  相似文献

为了研究成都天邦生物制品有限公司生产的鸡新城疫(基因Ⅶ型)、禽流感(H9亚型)二联灭活疫苗(aSG10株+G株)对蛋鸡的安全性和免疫效果,在北京市华都峪口禽业有限责任公司京红1号蛋鸡养殖场进行相关试验。结果显示:疫苗经不同免疫途径(皮下注射和肌肉注射)、不同免疫剂量(单剂量和超剂量)接种115日龄蛋鸡后,未见任何不良反应,注射部位疫苗吸收良好,产蛋率与空白对照组无明显差异。试验鸡免疫后14d,新城疫HI抗体平均滴度为10.9log2,H9亚型禽流感HI抗体平均滴度为9.1log2,免疫后6个月新城疫HI抗体水平不低于6.9log2,H9亚型禽流感HI抗体水平不低于7.4log2。分别于免疫后21d和6个月对免疫鸡进行攻毒,新城疫攻毒保护率为100%,禽流感攻毒保护率为95%。表明该二联苗对蛋鸡安全、有效,且免疫持续期长,可用于蛋鸡的免疫。  相似文献

三批禽脑脊髓炎(AE)油佐剂灭活疫苗免疫开产前种鸡,种鸡免疫2~3个月后所产种蛋对禽脑脊髓炎病毒(AEV)鸡胚易感性试验的受保护率达100%,而同期所产种蛋孵出的仔代雏鸡,在出雏后3周内对AEVVR株攻毒的受保护率亦达100%;种鸡免疫6~7个月后所产种蛋对AEVVR鸡胚易感性试验的受保护率达75%~80%,而同期所产种蛋孵出的仔代雏鸡,在出雏后3周内对AEVVR株攻毒的受保护率达70%~90%。试验结果表明,AE灭活疫苗对AEV鸡胚易感性试验的鸡胚保护率与同期雏鸡对AEVVR肌肉途径攻毒的保护率基本相同。  相似文献

本研究应用间接ELISA方法对新城疫LaSota和V4疫苗免疫SPF鸡及免疫后攻毒SPF鸡的血清中新城疫病毒(NDV)特异性HI抗体、IgM和IgG抗体水平的动态变化进行了检测。结果表明,V4较LaSota疫苗免疫鸡HI抗体提前3天左右出现,但除高峰期(1周左右)外,其HI抗体水平均低于LaSota免疫鸡。LaSota免疫鸡。LaSota疫羁免疫鸡血清中NDV特异性IgM和IgGr抗体高峰较V4免疫鸡提前约2周出现,攻毒后,LaSota免疫鸡血清中特异性IgG和IgG回忆应答显著,而V4免疫鸡IgM和IgG回忆应答不明显。  相似文献

雏鸡1日龄感染鸡贫血病毒,8日龄接种Lasota疫苗,以未感染免疫雏鸡为对照,于免疫后7、14、28d检测血清免疫球蛋白IgG、IgM、IgA,在凝抑制抗体(HI)滴度;胸腺、法氏囊、脾脏T细胞、IgG^ 、IgM^ 、IgA^ ,抗体生成细胞数量及T、B细胞增殖反应;胸腺、脾脏细胞因子IL-2、IFN活性的变化。结果发现,感染CAV雏鸡Lasota疫苗免疫后,其血清IgG、IgM、IgA免疫球蛋白含量明显减少,HI抗体滴度降低;胸腺、法氏囊、脾脏T细胞、抗体生成细胞数量降低及T、B细胞增殖反应减弱,胸腺、脾脏IL-2及TNF诱生活性降低,表明其细胞免疫和体流免疫功能以及细胞因子免疫调节作用均未感染免疫雏鸡明显减弱。  相似文献

Four different protocols were tested for the induction of IgA deficiency in chickens: (I) inoculation of anti-alpha intra-peritoneally (i.p.) on alternate days after hatching up to a period of three weeks; (II) bursectomy within 6 h and at 24 h after hatching; (III) in-ovo injection of anti-alpha on the 17th day of embryonation followed by bursectomy at 24 h of hatch and a single injection of anti-alpha i.p. on the day of hatching; (IV) as in III above but bursectomy within 6 h of hatch, followed by three further injections of anti-alpha on days 3, 10 and 34 after hatching. Treatment (I) produced temporary dysgammaglobulinemia during the period of treatment. Bursectomy at 24 h of hatch rendered 75% of the chicks IgA deficient up to four weeks of age. Early bursectomy within 6 h of hatch resulted in substantial improvement of IgA suppression. Such chicks, when tested at 4, 6 and 10 weeks of age, were found to be 81, 72 and 58.3% IgA deficient, respectively. With treatment (III) all the treated birds were IgA deficient at four weeks of age. However, as the birds grew older, IgA appeared in the serum so that at the age of 12 weeks only 27.3% were deficient. Treatment (IV) completely suppressed the IgA system of 13 out of 14 chickens. These chickens lacked both serum and secretory IgA as well as IgA-containing cells in their intestinal mucosa. Both IgG and IgM continued to be produced.  相似文献

Total serum protein levels, serum protein fraction levels, and specific serum immunoglobulin class or subclass levels were measured in colostrum-fed (CF) and colostrum-deprived (CD) calves during the first 144 hours after birth. Total serum protein values increased at 24 hours in the CF group and then decreased slightly at 144 hours. The increase in total serum protei5) in beta1-, beta2-, and gamma-globulins. The beta2- and gamma-globulin levels decreased by 144 hours, while the serum level of beta1-globulin continued to increase. The CD calves exhibited a significant decrease (P less than 0.05) in total serum protein at 24 hours, folhours, the level of beta1-globulin inlowed by a significant increase (P less than 0.05) at 144 hours. At 24 hours, the level of beta1-globulin decreased slightly, and the level of beta2- and gamma-globulins increased slightly. At 144 creased, and the level of gamma-globulin decreased. The beta2-globulin level did not change. At birth, immunoglobulin (Ig) M was detected in 5 of the 10 calves, IgG1 in 6 of the 10 calves, and IgG2 in 3 of the 10 calves. By 24 hours after birth, all CF calves had detectable levels of IgM, IgG1, and IgG2, and there were significant increases (P less than 0.01) in the mean serum levels of all 3 immunoglobulins. By 144 hours after birth, the serum levels of IgM, IgG1, and IgG2 decreased to various degrees. At 24 hours, the IgM level had not increased in CD calves; however, the level of IgG2 appeared to increase slightly, and the mean IgG1 level increased by approximately 50%. By 144 hours after birth, there was a significant increase (P less than 0.01) in the mean level of serum IgM. The level of IgG, also appeared to increase substantially, while the level of IgG2 appeared to increase slightly.  相似文献

The concentrations of IgG, IgM and IgA in sera collected from 3855 sows (3208 pregnant and 647 lactating) at a single time point were determined. This experimental design allowed changes in serum immunoglobulin over the reproductive cycle to be studied without bias from seasonal influence. The concentrations of the three immunoglobulins changed independently during the reproductive cycle. Serum levels of IgM and IgG began a progressive postpartal decline during the 14th–17th week of gestation. At the onset of lactation serum IgG levels progressively increased while IgM levels continued to decline, the latter reaching their lowest level during the third week of lactation. In contrast to IgM and IgG, serum IgA levels increased 35% during weeks 14–17 of gestation and continued to increase throughout lactation, reaching their highest serum levels in the third week of lactation; the serum IgA concentration at this time was twice that observed during the first 13 weeks of gestation. Results of these studies allowed the reproductive cycle to be classified into four phases on the basis of serum immunoglobulin concentrations: (1) weeks 1–4 of gestation; (2) weeks 5–13 of gestation; (3) weeks 14–17 of gestation and (4) lactation.  相似文献

The transfer of immunoglobulin G (IgG) and immunoglobulin M (IgM) from ewe to lamb was quantitated to determine the occurrence of failure in passive transfer. Concentrations of IgG and IgM in ewe serum did not correlate with those in the colostrum. Colostrum from all ewes contained abundant amounts of immunoglobulins when compared with serum values, with IgG being selectively concentrated over IgM. Absorption through the intestinal tract of the lamb appeared to be a nonselective process, lacking predilection for IgG and IgM. All lambs tested 24 hours after birth absorbed colostral immunoglobulins to some extent; however, 13 (14%) of 91 clinically normal lambs demonstrated some failure of passive transfer. In contrast, failure of passive transfer was found in 27 (46%) of 59 lambs dying of natural causes between 24 hours and 5 weeks of age. Evidence presented emphasizes the importance of absorption of adequate amounts of immunoglobulins to enable the newborn lamb to survive the first few weeks of life.  相似文献

将16只SPF鸡分成4组,每组4只,分别接种新城疫油乳剂苗,新城疫、传染性支气管炎、传染性法氏囊病三联油乳剂苗,LaSota弱毒苗和Mukteswer苗。接种后每隔2d采血1次,用ELISA测定新城疫特异性IgM和IgG抗体。结果:油乳剂灭活苗接种组均无特异性IgM抗体出现,特异性IgG抗体高峰期出现在接种后22d,LaSota弱毒苗和Mukteswer苗接种组均有特异性IgM抗体出现,高峰期为接种后第9d。各接种组经强毒攻击后,均出现IgM反应,IbG呈典型的回忆反应。进一步试验用LaSota灭活苗经肌肉、静脉接种和加氢氧化铝胶佐剂后肌肉接种,经测定,接种鸡均无或仅有很低水平的特异性IgM抗体产生。  相似文献

本研究采用阻断ELISA(B-ELISA)和血凝抑制试验(HI)方法,检测了鸡体分别人工感染副鸡嗜血杆菌Hp8株(A型)和H668株(C型)后0-9周的血清抗体消长情况,并绘制了示意抗体曲线。结果表明,Hp8人工感染后,从第2周到第9周,B-ELISA阳性检出率一直保持100%,其中抗体效价在感染后第4周达到效价高峰,平均约为15.2。而H688人工感染鸡则在感染后第7周检出率最高,此后急剧下降。AI方法对A型抗体的阳性检出率亦在在第4-5周达到高峰,而对C型抗体的检出率一直较低。结果进一步显示了B-ELISA良好的特异性和敏感性,为本方法及其试剂盒产品的进一步应用提供了参考。  相似文献
