The formalin-ethyl acetate sedimentation procedure was used to detect ova of the giant liver fluke, Fascioloides magna, in feces of farmed wapiti in Alberta. Twenty (3.2%) of the 629 fecal samples examined contained ova of F. magna. Thirteen (33.3%) of the 39 farms surveyed had wapiti positive for F. magna. The presence of F. magna in farmed wapiti north of the North Saskatchewan River is confirmed, and 3 areas where the infection has become endemic are identified.  相似文献

Four monoclonal antibodies (mAbs) (9.49, 24.27, 46.71 and 179.57) were produced against Fasciola hepatica excretory-secretory products. Isotype analysis revelead the antibodies to be IgM, IgG3, IgG1, and IgM. In immunoblot assays, the mAbs recognized different antigenic polypeptides migrating between 29 and 180 kDa. Specificity of the mAbs was evaluated by ELISA against antigens of Fascioloides magna, Anoplocephala magna, Stichorchis subtriquetrus, Haemonchus contortus, sheep liver extract (SLE), bovine liver extract (BLE), bovine serum albumin (BSA), bovine viral diarrhea virus (BVDV), and Madin-Darby bovine kidney (MDBK) cells. Monoclonals 9.49 and 24.27 were specific, and reacted only with Fasciola hepatica antigens. However, mAb 46.71 cross-reacted with antigens of Fascioloides magna, A. magna, Stichorchis subtriquetrus, and H. contortus but not with SLE, BLE, BSA, BVDV or MDBK cells. Isoelectric focusing was performed on the soluble proteins of whole-body and excretory-secretory products (ESP) of Fasciola hepatica and Fascioloides magna. Adult F hepatica flukes were recovered from experimentally infected sheep and ESP obtained from the flukes; portions of liver were cut and frozen at -70 C. Fascioloides magna adults were collected from naturally infected white-tailed deer and ESP obtained; portions of liver were collected from noninfected white-tailed deer. Adult flukes and their host tissues were homogenized and centrifuged; protein concentrations with their ESP were determined and adjusted to less than 2.50 mg/ml. Seven ESP samples from F hepatica and 1 from Fascioloides magna were subjected to isoelectric focusing with the 2 species of fluke and their respective host liver homogenates. After separation, gels were stained with silver and scanned on a laser densitometer. Protein banding patterns of the 2 species of flukes were dissimilar. In the pH range of 3.5 to 9.6, the body protein had approximately 30 peaks and ESP about 23 peaks in both species. Overall banding patterns of the body protein and ESP of both species were distinct from those of respective host tissues. Of the peaks reported as dominant, 3 of the body protein and 2 of ESP were shared between the 2 species. Fascioloides magna had more dominant peaks than F hepatica. This technique of soluble protein isoelectric focusing is simple and reproducible, and the 2 fluke species can easily be differentiated by this technique, as well as by morphologic characteristics.  相似文献

During 1989-1990, the United States Department of Agriculture (USDA) meat inspection records were used to determine the distribution and incidence of liver flukes (Fasciola hepatica and Fascioloides magna) in Montana cattle. Of the cows and bulls slaughtered in USDA-inspected packing plants during a 12 month time period, 17.24% had livers that were condemned because of liver flukes. This was a 12% increase over USDA liver condemnations reported for 1973. Infected animals have been reported from 26 counties in Montana, mostly located in the south-central and western half of the state. Forty-nine percent of the 2.4 million cattle in Montana are raised in these counties. Lymnaeid snail species that may serve as intermediate hosts for Fasciola hepatica were found in most of the counties where liver flukes were reported. The principal vectors believed to be responsible for the transmission of Fasciola hepatica in Montana are species of the genus Fossaria. Stagnicola montanensis and Lymnaea stagnalis, which may serve as intermediate hosts for this parasite have also been collected. A known intermediate host for Fascioloides magna, Stagnicola caperata, was also found in several locations.  相似文献

During the past decade, Fascioloides magna, the large American liver fluke, has spread within free-living deer in wetlands of the Danube in Lower Austria. The aim of this study was to determine the current infection rates with F. magna and other digenean parasites in the intermediate host snail Galba truncatula from risk areas in Lower Austria. A total of 3444 G. truncatula were collected and examined microscopically for the presence of digenean trematodes. A set of randomly selected snails and isolated trematode stages were also investigated molecular biologically by PCR and sequencing. Digenean parasites were detected with a prevalence of 2.41% (1.83% Paramphistomoidea; 0.46% Echinostomatoidea; 0.09% Strigeida; 0.06% Plagiorchiida). F. magna was found with an overall prevalence of 0.23%, which may indicate a recovery of the parasite population in spite of an ongoing triclabendazole treatment programme. Moreover, high risk areas and a seasonality of infections were observed.  相似文献

In a study to evaluate the efficacy of clorsulon against Fascioloides magna infection in sheep, 12 ewes were inoculated orally with 100 metacercariae of F magna, and 6 were treated with clorsulon (15 mg/kg of body weight) 8 weeks after inoculation. The sheep were euthanatized 16 weeks after inoculation, flukes were recovered, and the liver and other tissues were subjectively scored for the severity of lesions (0 to 4+). The number of flukes recovered from the clorsulon-treated group (3.8 +/- 1.2 flukes) was significantly (P = 0.025) lower than the number of flukes recovered from the group of untreated controls (10.0 +/- 6.6 flukes). The severity of lesions was significantly (P = 0.004) reduced (45.9%) in the treated group (2.0 +/- 1.1), compared with that in the untreated controls (3.7 +/- 0.5). In the untreated group, 3 sheep died and 1 became moribund 14 to 16 weeks after inoculation. The data suggested that a single treatment with clorsulon at a dosage of 15 mg/kg 8 weeks after inoculation was not effective in preventing F magna infection in sheep, because the survival of only a few F magna is potentially fatal in sheep within 6 months after infection.  相似文献

The presence of American liver fluke (Fascioloides magna) in Croatian wild ruminant species was detected for the first time in January 2000. At the same time, the problem of adequate parasitological monitoring and treatment appeared in the captive deer population. Quarantine and health screening protocols, as well as migration and transportation influence had to be evaluated in red deer husbandry. Non-invasive methods were introduced to estimate the prevalence of F. magna in the semi-farm rearing system. Coprological analysis has been performed on 264 faecal samples. The most effective antiparasitic treatment was implemented on herd and individual treatment. Treatment was extended to the free-ranging population of deer in the same region.  相似文献

A feedlot steer died suddenly without exhibiting signs of clinical illness. Cysts containing Fascioloides magna were found in the liver and lung. Fatal pulmonary hemorrhage was associated with these flukes. Neither death nor clinical pulmonary disease has been previously attributed to infections of adult cattle by F. magna.  相似文献

Infection with Fascioloides magna, the large American liver fluke, was diagnosed in two moose (Alces alces) and six wapiti (Cervus elaphus) from central Saskatchewan. This is believed to be the first record of the parasite in the province. Fecal samples collected from wild wapiti at five sites in the commercial forest zone in Saskatchewan contained eggs believed to be those of F. magna. Trematode eggs were not found in feces from five captive herds of wapiti in the province, nor in samples from wild wapiti in Moose Mountain Provincial Park. Operculate eggs were found in feces of wild wapiti from Cypress Hills, but these were believed to be from trematodes other than F. magna. The distribution and significance of F. magna in Canada, based partially on responses to a mail questionnaire, are reviewed.  相似文献

A survey among 1.12 million cattle slaughtered in 357 packing plants in Michigan during 1977 to 1981 was conducted to determine the prevalence of liver fluke infection. The condemnation rate of liver fluke-infected livers was 0.41, 3.7, and 13.9% in the southern, northern-lower, and upper regions of Michigan, respectively. The same trend in infection rates was observed in white-tailed deer that had been examined in diagnostic laboratories in the state. A serologic survey among 50 randomly selected dairy farms detected Fascioloides antibody-carrying cattle in 3 farms. The data on prevalence in slaughter cattle were slightly biased, because many slaughter cattle originated from other states in which F magna may be less common.  相似文献

The efficacy of diclazuril against intestinal and hepatic coccidiosis was studied in artificially infected rabbits. Prophylaxis against intestinal coccidiosis was evaluated using a mixed infection of Eimeria intestinalis, Eimeria magna and Eimeria perforans. Continuous medication in the feed at 1 p.p.m. was 100% effective in reducing oocyst output and faecal scores, and weight gain and feed efficiency were normal. Hepatic coccidiosis induced by Eimeria stiedai was prevented at 0.5 and 1 p.p.m. as shown by negative oocyst counts, normal liver weight, absence of liver lesions, and normal body-weight gain and feed efficiency. Medication at 1 p.p.m. for 7 consecutive days during the prepatent phase of hepatic coccidiosis resulted in large reductions in oocyst counts and lesion scores with a normal liver weight and growth performance. Diclazuril at 1 p.p.m. in the feed prevented both intestinal and hepatic coccidiosis in rabbits and can be advocated for safe mass medication.  相似文献

Fascioloides magna,the giant liver fluke, is an introduced parasite imported to Europe with infected game which is endemic in the Austrian wetlands of the Danube from Vienna to the Slovakian border. Due to its pathogenicity (especially in roe deer) and its potential of transmission to domestic ruminants a project on the epidemiology and control of this parasite was carried out between 2000 and 2005. To assess distribution faecal droppings from red deer were collected in an area along the Danube and livers of red and roe deer shot or found dead were examined. Simultaneously, triclabendazole-medicated feed was offered repeatedly in several areas of known fluke presence from 2001. The average prevalence of infection as demonstrated by Fascioloides eggs in droppings (n = 145) was 6.3%, with > 50% in some areas. Infection rates decreased from 11.3% (01/2001) to 1.5% (04/2002) in the whole area. In Fischamend, a highly endemic area, prevalences in monthly samples (n = 25) decreased from 70% (01/2001) to less than 1% (01/2005). Similarly, the number of eggs per gram of faeces was significantly reduced. Of 457 red deer livers 15.8% were positive. In the Fischamend area, a reduction of positive livers was also found (2000:100%; 2005:13%), as well as a reduction of the numbers of flukes/liver. Control of F. magna in the Danubian wetlands by anthelmintic treatment proved to be successful although eradication was not achieved.  相似文献

Efficacy of oral administration of 20 mg of triclabendazole/kg of body weight was evaluated against 12-week Fascioloides magna infections in 12 sheep, each inoculated orally with 250 viable metacercariae. From 6 sheep treated with triclabendazole, 1 immature F magna was recovered, whereas 116 F magna with a mean length of 19 +/- 6.5 mm were recovered from 6 untreated control sheep. Efficacy of triclabendazole was 99.14%. Signs of toxicosis or illness were not observed in the sheep.  相似文献

分别用4种兔艾美耳球虫经口接种45日龄无球虫感染兔,接种剂量为1×104个卵囊/兔。感染后4d~20d,以麦克马斯特氏法计数每天排出的卵囊。结果表明,肠艾美耳球虫排卵量最多,为365.76×106个,中型艾美耳球虫为317.26×106个、黄艾美耳球虫为304.36×106个,大型艾美耳球虫排卵量最少,为200.12×106个。大型艾美耳球虫在感染后第6.5天有卵囊排出,第10.5天达到高峰(66.50×106个/只),占总量的33.23%;肠艾美耳球虫在感染后第9天有卵囊排出,第13天达到高峰(138.40×106个/只),占总量的37.83%;黄艾美耳球虫在感染后第9天有卵囊排出,第11天达到高峰(102.80×106个/只),占总量的33.78%;中型艾美耳球虫在感染后第4.5天有卵囊排出,第6.5天达到高峰(167.8×106个/只),占总量的52.89%。  相似文献

The pathogenesis of early prepatent Fascioloides magna infection was investigated in seven fawns (Odocoileus virginianus) given 500 metacercariae and examined at one, two, three, five, eight, 12 and 13 weeks postinoculation. Blood samples were taken from eight inoculated deer every two weeks up to 16 weeks postinoculation. Eosinophilia with a mild transitory anemia were the main clincopathological features. Postmortem examination at two weeks postinoculation revealed extensive migration of immature flukes. Subcapsular tracks in the liver, nodules on the blind sacs of the rumen, as well as retroperitoneal granulomas on flanks and necrotic tracks on the diaphragm were found. Evidence of penetration of flukes into the lung was found at two weeks postinoculation and there was early granuloma formation at three weeks postinoculation. Flukes migrating into tissues other than the liver were destroyed in large granulomas, although remnants of degenerating parasites were not found. At eight weeks postinoculation, widespread granuloma formation characterized the infection with this lesion present in nodes along the gastrointestinal tract, in the mesentery, flanks, psoas muscles, diaphragm, between the ribs and in the lungs. By 12 weeks postinoculation subcapsular tracks were observed in the liver.  相似文献

从河北省兔场分别单卵囊分离孢子化大型艾美耳球虫卵囊、黄艾美耳球虫卵囊及肠艾美耳球虫卵囊,接种无球虫兔后获得纯种卵囊,CTAB法提取孢子化卵囊基因组DNA。利用艾美耳属球虫18SrDNA和5.8SrDNA保守引物,PCR扩增3种兔球虫ITS-1片段,产物纯化后测序。将3种球虫ITS-1测序结果与GenBank发布的兔球虫ITS-1序列进行比对和遗传距离比较,绘制系统发育树。结果表明,大型艾美耳球虫、黄艾美耳球虫及肠艾美耳球虫河北株分别扩增出424、455、434bp的ITS-1片段。大型艾美耳球虫、黄艾美耳球虫及肠艾关耳球虫河北株与GenBank中发布的同种兔球虫ITS-1序列相似性分别为97.4%、97.9%和96.9%。系统发育树显示兔球虫ITS-1序列形成1个单系群,该单系群根据寄生部位分为2个姊妹群。  相似文献

The purpose of this study was to examine the prevalence and distribution of grossly visible lesions of tuberculosis in a herd of 344 North American elk (Cervus elaphus) depopulated during a three-month period in 1991. Abattoir inspection detected mycobacterial lesions in 134 (39.8%) of the 337 animals received for slaughter. The prevalence of lesions increased with the age of the animals. Lesions were predominantly suppurative rather than caseous, and mineralization was less evident than in tuberculous lesions in cattle. Lesions occurred predominantly in lymph nodes, and lungs were the only organs in which mycobacterial lesions were found. The distribution of lesions suggested that aerosol transmission was the most significant means of spread of the disease within the herd. Giant liver flukes (Fascioloides magna) were observed in approximately 80% of the adult elk.  相似文献

Cerebrospinal fluid analyses are important for diagnosis of neurologic problems in rabbits and for translational research projects using rabbits as models. Blind puncture of the cisterna magna is the current standard technique for sampling cerebrospinal fluid in this species. However, the complexity and small size of the cisterna magna and surrounding structures are limitations of this technique. Aims of this prospective, anatomic, pilot study were to (1) describe the normal anatomy of the atlanto‐occipital region, (2) describe ultrasonographic anatomic landmarks, and (3) develop and evaluate a technique for ultrasound‐guided puncture of the cisterna magna for cerebrospinal fluid sampling in rabbits. Thirty healthy rabbits were included and the study was conducted in three stages. Three rabbit cadavers were used for the first stage of the study. Then, the second stage was completed using 13 rabbit cadavers. Finally, the third stage was completed in 14 live rabbits. The ultrasound‐guided puncture performed in 13 cadavers was successful at the first attempt in 10 cases, and at the second attempt in the remaining three cases. In the in vivo study, the ultrasound‐guided puncture was successful in all 14 cases, without signs of complications. Findings supported the use of ultrasound‐guided puncture of the cisterna magna as a safe technique that may be used routinely or when the sample of cerebrospinal fluid cannot be obtained with the blind technique in rabbits.  相似文献
