将两株鸡传染性法氏囊病病毒(IBDV)弱毒株在SPF鸡体内连续传代至第5、第6代时,出现明显的法氏囊萎缩和B:B指数下降,表明IBDV弱毒株在鸡体内连续传代后毒力增强。为进一步阐释哪些基因位点导致了上述毒力的变化,本试验测定了基础弱毒株及其在鸡体内传代后各个代次毒的基因组序列,比对分析后发现VP2蛋白253位氨基酸发生了由H到Q或N的变异,表明VP2蛋白253位氨基酸的替换可能会增强传染性法氏囊病病毒在鸡体内的致病性。  相似文献

本研究旨在探讨二甲双胍(metformin)处理对鸡巨噬细胞HD11天然免疫相关基因表达及禽白血病病毒(Avian leukosisvirus,ALV)复制水平的影响。用2mmol/L二甲双胍处理鸡巨噬细胞HD11,处理48h后收集细胞,荧光定量PCR(RT-qPCR)检测天然免疫基因和内源性反转录病毒的表达。另外,将J亚群禽白血病病毒(J subgroup Avian leukosis virus,ALV-J)(MOI=5)接种于细胞板中,病毒感染细胞24 h后进行二甲双胍处理, 48 h后收集HD11细胞和上清分别进行RT-qPCR检测和ELISA(enzyme linked immunosorbent assay)检测。结果表明,二甲双胍显著上调HD11细胞天然免疫相关基因TLR3(P<0.01)、IFIH1(P<0.01)、IRF7(P<0.01)、STAT1(P<0.05)、IFN-α(P<0.01)、IFN-β(P<0.01)表达水平,并激活了鸡内源性反转录病毒的表达,抑制ALV-J复制水平。综上,二甲双胍可激活鸡巨噬细胞的天然免疫反应,从而提高鸡抗禽白血病病毒的能力。本研究在体外实验中验证二甲双胍具有抑制ALV增殖的作用,对家禽业的疾病防控有一定的应用价值。  相似文献

从我国新疆维吾尔自治区某鸡场发病症状及病理变化疑似鸡传染性支气管炎的病鸡有出血点病变的腺胃组织中分离病毒,在9-11日龄SPF鸡胚上连续传代9次,通过病毒对鸡胚的致病作用、病毒在电镜下的形态观察、病毒在鸡胚中的增殖动态变化、病毒在CEF中的增殖特性、凝集鸡红细胞的特性以及动物回归试验等来研究我国鸡传染性支气管炎病毒地方流行株的生物学特性。结果表明,该毒株的第一代尿囊液对鸡胚无肉眼可见的致病作用,当继代到第5代后,胚体严重病变;电镜观察该病毒为典型的冠状病毒;病毒在鸡胚中随着接种病毒时间的延长,其效价增高,96小时可达到48小时的1倍,该毒株可在CEF上生长,但不能形成明显的蚀斑;并且经1%胰酶处理后可疑集鸡红细胞;鸡胚的第4代尿囊液病毒回归动物体,可致鸡病变病死鸡肾脏病变尤为明显,呈典型的花斑肾,腺胃则未见肉眼可见的病变,接种鸡、同居鸡和对照鸡之间NDV疫苗免疫后其HI抗体水平无明显差异,但从发病症状来看,IBV对ND疫苗具有干扰作用。  相似文献

Antibody titers for infectious bursal disease virus (IBDV), infectious bronchitis virus, Newcastle disease virus, and reovirus from chicks with chicken anemia agent (CAA) antibodies were compared with antibody titers from their CAA-antibody-negative counterparts. These comparisons were made in 396 chickens that were 1 day, 2 weeks, 8-9 weeks, 10 weeks, 17 weeks, or 29-32 weeks old. Only one serum sample was collected from any given chick or chicken. There were no significant differences between the antibody titers at any age for any antigen, with one exception: at 29-32 weeks, the IBDV titers were higher (t = 2.62, df = 142, P less than 0.01) in chickens with CAA antibody. Although not at all likely, we believe that the observation of high IBDV antibody titers in CAA-antibody-positive chicks could have been a spurious one.  相似文献

Attempts to isolate and identify budgerigar papovavirus (BPV) were made during three separate outbreaks of disease diagnosed on pathological grounds. Direct electron microscopy was successful only when large areas of skin were extensively disrupted to release virus and then extracted with fluorocarbon to remove lipids. Direct inoculation of budgerigar tissue suspensions into chicken embryos or chicken cell cultures failed to produce detectable virus. However, when primary cultures of liver and kidney were prepared from affected budgerigars, BPV could be detected by electron microscopy and by the production of a cytopathic effect at the third or fourth passage in cell cultures. The isolated virus was pathogenic for 10-day-old but not 11- or 12-day-old chicken embryos. Inoculated 11- and 12-day-old embryos produced antibodies to BPV that were detectable 2 weeks after hatching by agar-gel-immunodiffusion tests.  相似文献

2001年在我国黑龙江省某鸡场疑似鸡传染性支气管炎的发病鸡群中,分离到一株病毒,将该病毒接种10日龄SPF鸡胚,取72h的尿囊液进行电镜观察并用1%胰酶处理,结果发现尿囊液中存在典型的冠状病毒,用胰酶处理过的尿囊液能凝集SPF鸡的红细胞,初步鉴定为鸡的传染性支气管炎病毒。将该病毒尿囊液再次接种10日龄SPF’鸡胚,通过病毒对鸡胚的致病作用、病毒超微形态特征以及病毒凝集鸡红细胞的特性等对该毒株进行研究,结果表明:经胰酶处理后的病毒尿囊液可凝集鸡红细胞。鸡胚的第二代病毒尿囊液(命名为LH2,/01/10)分别接种1日龄和15日龄的SPF鸡,发现对不同日龄的鸡表现不同的致病性,对1日龄接种鸡和同笼饲养的同居对照鸡致病力高,发病率为11/11,致死率分别为4/6和2/5;15日龄接种和同居感染鸡发病率为9/9,致死率分别为1/5和1/4。实验应用反转录一聚合酶链式反应技术对LH2/01/10的膜蛋白基因进行扩增、克隆和序列测定,结果表明该基因具有IBVM基因的共有分子特征.与IBV标准株M41的M基因核苷酸的同源性为90%,氨基酸的同源性为91%。这从分子水平进一步证实引起鸡群死亡的病毒为鸡传染性支气管炎病毒。  相似文献

从市场上随机购得不同厂家各两批次的商品化禽流感(H9)油乳剂灭活疫苗,按瓶签说明免疫禽流感母源抗体为阴性的蛋鸡。当疫苗抗体下降至5log2左右时部分鸡再二免,并做田间试验。结果表明,免疫1周出现血清抗体,并逐日上升,3周达高峰。首次免疫8周后抗体下降至5log2左右,二次免疫15周后抗体下降至5log2左右。这四批疫苗的免疫原性较高,安全可靠,两次免疫保护期可保护至开产后。  相似文献

Chickens vaccinated at one and 14 days of age with a live vaccine containing the Massachusetts and Connecticut strains of infectious bronchitis virus were protected against challenge with a Massachusetts strain administered at 5 weeks of age. The virus recovery method in which tracheal swabs taken 4 days post-challenge were used to inoculate chicken embryos was compared with the method based on ciliary activity in tracheal explants. The results showed that evaluation of ciliary activity effectively measured immunity to infectious bronchitis virus. Serological results obtained in a microneutralization test using chicken embryo kidney cells correlated with the immune status of the birds as well as with the alpha neutralization results obtained using chicken embryos.  相似文献

A total of 291 eight-week-old chickens were exposed to chickens infected with either of two Australian lentogenic strains (V4 and AVL NDV-1) of Newcastle disease virus (NDV). At 3 weeks after exposure, all chickens exposed to V4 infected chickens had developed haemagglutination-inhibition (HI) antibody. All chickens exposed to AVL NDV-1 virus infected chickens had developed HI antibody 5 weeks later. This sudden late appearance of HI antibody, to titres higher than those observed with V4 chickens, was explained by V4 virus being introduced to the AVL NDV-1 group of chickens. When groups of these chickens were challenged with Roakin virus (mesogenic NDV) at 3 weeks and Fontana 1083 virus (viscerotropic velogenic NDV) and Texas GB virus (neutrotropic NDV) at 3, 5, 10 and 21 weeks only three chickens developed clinical illness one of which died. These chickens were one AVL NDV-1 chicken contact challenged with Fontana 1083 virus at 3 weeks, one V4 chicken oronasally challenged with Texas GB virus at 5 weeks and one V4 chicken challenged oronasally with Fontana 1083 virus at 10 weeks. Susceptible non-vaccinated chickens died soon after challenge. Challenge by oronasal infection with 10(7.0) ELD50 of virus or contact with susceptible infected chickens enabled virulent virus to be isolated from most chickens and was accompanied by a large anamnestic increase in serum HI antibody.  相似文献

鸡传染性支气管炎病毒(IBV)具有不同致病特性,将IBV XDC-2株接种9日龄SPF鸡胚培养,可引起鸡胚死亡和出现侏儒胚,病毒EID50达5×10-5.33/mL。将IBV XDC-2株接种18日龄SPF鸡,饲养观察14 d,病鸡临床症状表现为:精神沉郁,羽毛凌乱,双翅下垂,轻微腹泻,多数拉白色水样稀粪。病死鸡出现肾肿大、呈花斑状、大量尿酸盐沉积。鸡发病率为100%,死亡率为25%。死亡鸡肺脏、肾组织制作组织切片,发现病理变化明显,主要为:肾小管扩张,上皮细胞呈玻璃样变性,部分管腔内可见坏死脱落之上皮细胞,于肾间质可见大量单核细胞浸润,肾间质有充血、出血现象;肺内动脉、毛细血管充血,淋巴细胞浸润。死亡鸡肺脏、肾组织接种鸡胚分离病毒,RT-PCR检测结果为阳性,表明该分离株为鸡传染性支气管炎病毒,具有很强的嗜肾性。  相似文献
