猪瘟病毒囊膜糖蛋白DNA疫苗的研制   总被引:6,自引:0,他引:6  
构建了含有猪瘟病毒石门株囊膜糖蛋白(E2)基因的重组真核表达质粒pRCSE2。将pRCSE2和空载体pRC分别免疫小鼠,ELISA检测结果表明:仅pRCSE2免疫鼠可以产生猪瘟病毒的特异抗体。DNA疫苗在许多传染病研究中已取得可喜成果,因此猪瘟病毒DNA疫苗可能会成为防制猪瘟的一条新的途径。  相似文献

The time periods and the behavioural pattern of mink euthanized with carbon dioxide (CO2), 100% and 70%, carbon monoxide (CO), 4%, and nitrogen (N2), 100%, are described. The time between the placement of the animal in a glass box and the first symptoms of incoordination (phase I), the period of loss of consciousness (phase II), and, finally, the coma phase until cessation of respiration (phase III) using three groups each of 10 pastel male mink each were recorded. Phase I times ranged from 14 s for CO2, 31 s for N2, and 49 s for CO. In phase II, the difference was even more pronounced, being 5 s for CO2, 15 s for CO, and 45 s for N2. In phase III, the time was 58 s for N2, 134 s and 151 s for CO2 and CO respectively. Hence, the critical period from the time when the animals were placed in the glass box till unconsciousness occurred (phase I and II) was 19 s for CO2, 64 s for CO, and 76 s for N2. The total course of euthanasia was 153 s for CO2, 215 s for CO, and 134 s for N2. The following minimum times will be required before it can be judged to be safe to remove the animals: CO2 and N2: 5 min, and for CO: 6.5 min. In a supplementary experiment, involving a mixture of 70% CO2 and 30% atmospheric air, it was not possible within the allotted time to kill adult male mink.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

ObjectiveTo compare the accuracy of transcutaneous (tc) to arterial partial pressure of carbon dioxide (PaCO2) and partial pressure of oxygen (PaO2) in anesthetized rabbits.Study designProspective, randomized, experimental study.AnimalsEight healthy adult female New Zealand white rabbits weighing 4.05 ± 0.30 kg.MethodsIsoflurane anesthetized rabbits received six treatments in random order; PaCO2 < 35, 35-45, and >45 mmHg and PaO2 < 80, 100-200, >200 mmHg. Arterial and transcutaneous measurements were taken after 15 minutes of stabilization at each condition. Linear regression, correlation and Bland-Altman analysis were performed to compare PtcCO2 to PaCO2 and PtcO2 to PaO2.ResultsOver a range of measured PaCO2 values from 21 to 67 mmHg (n = 24) mean bias for PtcCO2 was -1 mmHg and the 95% limits of agreement were -7 to 5 mmHg. The correlation between PtcCO2 and PaCO2 was strong with R2 value of 0.9454. Over the entire range of measured PaO2 values (46-508 mmHg) mean bias for PtcO2 was -61 mmHg and the 95% limits of agreement were -226 to 104 mmHg. Correlation was poor with R2 = 0.5969. Comparing PtcO2 to PaO2 over a narrower range [PaO2 < 150 mmHg (n = 13)] improved the correlation, with an R2 value of 0.8518, mean bias of -7 mmHg and 95% limits of agreement from -33 to 19 mmHg.Conclusions and clinical relevanceIn healthy anesthetized rabbits, PtcCO2 closely approximated PaCO2. In contrast PtcO2 underestimated PaO2, particularly at high values. The PtcCO2 sensor may be a useful noninvasive way to assess adequacy of ventilation in anesthetized rabbits.  相似文献

Endogenous prostaglandin (PG) E(2) plays important roles in renal homeostasis. Immunoexpressions of PGE(2) biosynthesis-related enzymes, cyclooxygenase (COX)-2 and microsomal PGE(2) synthetase (mPGES)-1 and EP4 (a PGE(2) receptor), were investigated in renal development. Kidney tissues were obtained from fetuses on gestation days 18 and 21 and neonates on days 1 to 18. In fetuses and early neonates, the expressions of COX-2, mPGES-1 and EP4 were observed in developing renal tubules, indicating that COX-2 and its product, PGE(2), play important roles in blastemal cell-derived renal tubular development via EP4. Cyclin D1 expression was seen in both the nucleus and cytoplasm of the developing tubules. These findings differed from the decreased COX-2 expression and exclusive nuclear expression of cyclin D1 seen in abnormal epithelial regeneration of injured renal tubules in cisplatin-treated rats in our previous articles. Collectively, PGE(2), induced by COX-2, regulates renal tubular epithelial formation via EP4.  相似文献

Mycoplasma mycoides strains were screened for the ability to produce H(2)O(2) from glucose and glycerol metabolism using rapid and simple colorimetric assays. In quantitative assays, H(2)O(2) production by washed cell suspensions was detected by the oxidation of o-dianisidine in the presence of peroxidase. In qualitative assays, a 3,3'-diaminobenzidine-peroxidase reagent was applied to colonies on agar plates. Both methods enabled differentiation of European subsp. mycoides SC (small colony) isolates from other M. mycoides strains by their inability to produce H(2)O(2) from glycerol metabolism. In addition, two strains of subsp. capri were identified which produced large amounts of H(2)O(2) from glucose oxidation. In lysed cells of these strains, NADH oxidation gave approximately 1 mol H(2)O(2) per mol NADH oxidised whereas in 36 subsp. mycoides and 10 other subsp. capri strains, the quantity produced was 0.01-0.20mol H(2)O(2) per mol NADH oxidised.  相似文献

铜是动物必需的一种微量元素,参与血红素的合成和红细胞的成熟及动物成骨过程、毛发和皮毛的色素沉着和角质化过程,在动物体内作为几种重要酶的成分(或作为辅因子)而发挥作用。饲料中添加高铜(250mg/kg)能提高断奶仔猪的日增重、采食量和饲料效率(Edmonds等,1985;Cromwell等,1991、1997),因此目前国内在猪饲料中大多添加高剂量的硫酸铜。但在生产应用中,由于硫酸铜易溶于水、易潮解、氧化还原能力强,在饲料贮存过程中极易导致饲料结块、饲料营养成分损失。实践证明,饲料中维生素对铜的催化十分敏感,饲料中的脂肪也因高铜发生酸败而导致饲料…  相似文献

为探讨猪圆环病毒2型(PCV2)ORF2/猪白介素-2(PoIL-2)嵌合重组表达质粒(rpcDNA3.1/PCV2-linker-PoIL-2)在猪体内的免疫效果和免疫保护效果进而研制高效PCV2核酸疫苗,将35只10日龄健康仔猪平均分成7组分别以rpcDNA3.1/PCV2-linker-PoIL-2重组表达质粒或PCV2ORF2基因原核表达的rCap蛋白进行免疫及免疫保护试验。共进行4次肌肉注射免疫,每次间隔2周;于第4次免疫后3周通过口腔和鼻腔途径感染PCV2细胞强毒。分别于第4次免疫后和攻毒后不同时间通过检测免疫猪血清抗体水平和外周血T淋巴细胞增殖活性、辅助性T细胞(Th)和细胞毒性T细胞(Tc)亚群的百分含量、排毒率和病毒血症阳性维持时间等指标以评价其免疫和免疫保护效果。结果表明,各免疫组猪均产生了抗PCV2特异性ELISA免疫抗体,但rCap蛋白免疫组猪抗体水平较低;rpcDNA3.1/PCV2-linker-PoIL-2质粒对猪体的免疫和免疫保护效果显著优于rpcDNA3.1/ORF2质粒;在rpcDNA3.1/PCV2-linker-PoIL-2质粒或rpcDNA3.1/ORF2质粒中添加rCap蛋白对重组质粒免疫及免疫保护效果无明显影响。因此,选取rpcDNA3.1/PCV2-linker-PoIL-2质粒为下一步研制PCV2核酸疫苗的主要成分。  相似文献

Anelloviruses are small, non-enveloped viruses with circular single stranded DNA, which infect a number of animal species as well as humans. In pigs, two distinct Torque teno sus virus (TTSuV) species have been described so far, being one of them linked to disease occurrence. Specifically, TTSuV2 loads in serum have been found increased in pigs suffering from postweaning multisystemic wasting syndrome (PMWS). Since this pathological condition is able to be controlled by means of porcine circovirus type 2 (PCV2) vaccination, it was hypothesized the possibility that such vaccination would have an impact on TTSuV2 prevalence and loads. A total of 150 pigs were divided in two study groups. Half of them received a PCV2 commercial vaccine, while the other half remained as non-vaccinated controls. PCV2 infection was monitored at 3-4, 8, 12, 16 and 21 weeks of age by means of an standard PCR, while TTSuV2 loads were determined at 8, 16 and 21 weeks of age by a quantitative PCR. No obvious PMWS clinical signs were observed among studied animals, although PCV2 infection was confirmed in both groups of pigs. Almost all pigs got TTSuV2 infection throughout the study period, independently of the PCV2 vaccination status of animals. Moreover, TTSuV2 load did not show significant differences between different pig groups at each sampling time, but mean viral load increased with age. Taking into account that previous results suggest that TTSuV2 load in serum is increased in the background of PMWS, the present study suggests that this is not the case in a PCV2 subclinical infection scenario. Therefore, vaccination of PCV2 subclinically infected pigs did not modify the outcome of TTSuV2 infection.  相似文献
