为确定益生菌FGM发酵黄芪多糖的最佳提取工艺,本试验以发酵黄芪多糖含量为考察指标,采用响应面法对工艺的提取参数进行优化,并建立回归模型。结果显示,提取时间、提取温度和料液比对发酵黄芪多糖的含量影响显著,并且两两因素间存在一定的交互作用;影响发酵黄芪多糖提取含量的工艺因素按主次顺序排列为:料液比 > 提取温度 > 提取时间;乳酸菌发酵黄芪液多糖提取最佳工艺为:提取时间65 min、提取温度80℃、料液比为1:9,在此条件下,发酵黄芪多糖的含量为6.72 mg/mL。试验证明该工艺可行,可为新兽药开发提供参考。 相似文献

[目的]优化饲料中粗脂肪超声提取工艺参数。[方法]以草鱼膨化配合饲料为试验材料,设计单因素试验,考查分析4个因素(液料比、浸泡时间、超声时间和超声次数)不同水平对饲料粗脂肪含量测定的影响。在单因素试验基础上,设计4因素3水平L9(34)正交试验,优化饲料粗脂肪超声提取工艺参数。采用优化后的超声提取工艺测定草鱼膨化配合饲料的粗脂肪含量,并与索氏提取法测定结果进行比较,重复6次,以此验证最佳超声提取工艺参数。超声提取工艺参数确定后,分别采用超声提取法和索氏提取法测定13种不同饲料样品的粗脂肪含量,重复6次,分析比较两种方法在测定饲料原料和常见动物性饲料产品粗脂肪含量的差异。[结果]影响饲料粗脂肪含量测定各因素的排序为超声次数>液料比>超声时间>浸泡时间;最佳超声提取工艺为液料比37.5 mL/g,浸泡0 min,超声时间30 min,超声提取2次;最佳超声提取工艺下,草鱼膨化配合饲料的粗脂肪含量为4.76%(n=6),而索氏提取法粗脂肪含量为4.77%(n=6),试验证明超声提取工艺稳定且可行;分别采用超声提取法和索氏提取法测定13种不同饲料样品的粗脂肪含量,两种方法测定... 相似文献

为了确定红芪中红芪多糖提取工艺的最佳条件,采用单因素法考察了浸提次数、料水比、浸提时间、浸提温度对红芪多糖提取的影响。试验采用常规法提取红芪多糖,苯酚—硫酸比色法测定多糖含量。结果表明,浸提温度80℃,浸提次数3次,浸提时间1.0 h,料液比1∶12时,红芪多糖的提取率较高。 相似文献

选用甘肃省道地药材红芪作为试验材料,通过水提醇沉法提取红芪多糖,并以单因素试验与响应面法相结合的方法优化红芪多糖的提取工艺。单因素试验设置提取温度、提取时间、提取次数、液料比和醇沉比5个因素,每个因素设置5个水平。在此基础上以多糖提取率为衡量指标,进行Box-Behnken试验设计,通过回归模型的建立及显著性检验,得到红芪多糖提取工艺的最佳参数,再利用该工艺参数条件进行验证。结果显示,红芪多糖的最佳提取条件为提取温度为80℃,提取时间为3 h,提取次数为4次,液料比为17 mL/g,醇沉比75%。最佳条件下红芪多糖的平均提取率为7.70%,与理论提取率8.01%无显著性差异(P0.05)。证明所建立的数学模型是合理的,认为采用单因素试验结合响应面法优化红芪多糖的工艺稳定可行,为红芪多糖的有效提取提供理论依据,为进一步分离纯化红芪多糖奠定基础。 相似文献

采用正交试验法优选女贞子中特女贞苷的最佳提取工艺条件,以加水量、提取时间与提取次数为考察因素,特女贞苷含量为评价指标对提取工艺进行考察。结果显示,特女贞苷的最佳提取工艺为加10倍药材质量的水,提取3次,每次2 h。该工艺为提高特女贞苷的提取率提供了实验基础,并为进一步的工业化生产提供科学的理论依据。 相似文献

The experiment was conducted to study the best extract process of mulberry leaf polysaccharide,three factors and three levels orthogonal test was designed to optimize the extraction process.Three factors were considered,including ratio of solid to liquid,decoction time,the time of extracting and the yield of mulberry leaf polysaccharide was determined with phenol-sulfurie acid colorimetry as the evaluate index.Meanwhile,the influence of ethanol concentrations on mulberry leaf polysaccharide extract was studied.The results showed that decoction times could greatest affect the extraction percent of the polysaccharides.The extraction percent of polysaccharide was the highest (4.94%) when the rate of solid to liquid was 25,the time of extracting was 2 h and decoction times was 2.While the most appropriate process was 25 times for the rate of sdid to liquid,2 h for extracting and decoction times was 3,considering with practical situations.The concentration of ethanol would significantly affect polysaccharides yield and purity.When the concentration of ethanol was 70%,the yield of mulberry leaf polysaccharid was highest.When the concentration of ethanol was 40%,the content of mulberry leaf polysaccharide was highest (41.5%),significantly higher than other ethanol concentration (P < 0.05).It suggested that the higher the ethanol concentration,the lower the polysaccharide content,but the higher the yield.The optimum extraction of mulberry leaf polysaccharides were that the rate of solid to liquid was 25,extractign for 2 h,decoction 3 times and 70% ethanol precipitation. 相似文献

为探究桑叶粗多糖最佳提取工艺,利用正交试验设计对桑叶粗多糖的提取工艺进行优化。以桑叶粗多糖得率为评价指标,以加水倍数、煎煮时间、煎煮次数为考察因素,设计3因素3水平正交试验,采用苯酚一硫酸比色法进行桑叶粗多糖含量测定,比较得出桑叶水提物的最佳制备工艺,并在此基础上考察不同浓度乙醇对桑叶粗多糖提取的影响。结果显示,煎煮次数对桑叶粗多糖提取率影响最大,当加水倍数为25倍,煎煮时间为2 h,煎煮2次时桑叶水提物中桑叶粗多糖提取率最高,为4.94%。但结合影响因素分析,最终确定桑叶水提物的最佳提取工艺为加25倍水,煎煮2 h,煎煮3次。乙醇浓度对桑叶粗多糖的含量及得率影响很大,70%乙醇醇沉所得水提物中桑叶粗多糖质量最大,40%乙醇醇沉所得桑叶粗多糖含量最高,平均含量达41.5%,显著高于其他乙醇浓度(P < 0.05),提示乙醇浓度越高,桑叶粗多糖含量越低,但得率越高。桑叶粗多糖最佳提取工艺为:加水倍数为25倍,煎煮时间2 h,煎煮3次,采用70%乙醇醇沉。 相似文献

To optimize the extraction technology of Rehmannia glutinosa polysaccharide and catalpol from Radix Rehmanniae simultaneously,a single factor test and orthogonal test were used in this experiment.Content of Rehmannia glutinosa polysaccharides and catalpol were determined by phenol-sulfuric acid method and HPLC,respectively.The yields of Rehmannia glutinosa polysaccharide and catalpol were used as the evaluating parameters,and the effects of the solid to liquid ratio,extraction temperature and extraction time on the yield were examined.The optimum extraction technological parameters were as follows:solid to liquid ratio was 1:8,the extraction time was 1.5 h,and the extraction temperature was 90 ℃.This technique was stable and feasible,and was appropriate for industrial production,and would provide references for the development and utilization of new veterinary drugs. 相似文献

Response surface method was used to optimize main process parameters for the polysaccharides extractions from Astragalus membranaceus fermented by probiotic FGM strain.A regression model equation was fitted.The results showed that extraction time, extraction temperature and solid-liquid ratio were significant factors on polysaccharide content fermented from Astragalus membranaceus and there was interactive effects between every two factors.Polysaccharide content was significantly affected by solid-liquid ratio,followed by extraction temperature and extraction time.The optimum conditions were found to be that the extraction time was 65 min, extraction temperature was 80℃,solid-liquid ratio was 1:9,and the polysaccharide content was 6.72 mg/mL at this condition.The results showed that the model was practical that could provide a base for exploitation new medicine. 相似文献

为充分利用紫花苜蓿(Medicago sativa L.)中的黄酮类物质,应用微波辅助提取技术,分别对影响其黄酮提取量的乙醇浓度、微波功率、固液比、提取时间和提取次数进行单因素试验,并在此基础上对各因素的互作效率进行正交试验。结果表明:固液比和乙醇浓度是影响提取量的主要因素,微波功率和其它因素影响较小;微波提取苜蓿黄酮的最佳工艺为:40%乙醇浓度,1:30固液比,微波500 W功率下作用60s;在此工艺条件下,黄酮提取量的理论值为4.92mg/g,接近于实际测定值4.88mg/g。 相似文献
