The effect of colostral antibodies on the immune response in lambs following adenovirus vaccination was studied. Young lambs of different ages, born from vaccinated ewes, were vaccinated with an inactivated and aluminium hydroxide-adsorbed sheep adenovirus vaccine. In a group of lambs vaccinated at 5–7 days of age, the titre of humoral antibodies declined in parallel to unvaccinated controls. In lambs vaccinated at 24–35 days of age, antibody titres to adenovirus stabilised and persisted after an initial fall. Cell mediated immunity, as measured by blastogenic responses of circulating lymphocytes, was stimulated in both groups, but higher numbers of IgG-producing cells became evident in the group of lambs vaccinated at 24–35 days of age.  相似文献

The level of antigen-specific interferon-gamma (IFN-gamma) production can be used as an indicator of cellular immunity. In this study, we investigated the role of cellular immune response in protection against classical swine fever virus (CSFV). Pigs were vaccinated once with CSFV vaccine and challenged 6 days post-vaccination (dpv). Vaccinated animals had significantly higher CSFV-specific IFN-gamma secreting cells than the unvaccinated pigs (p<0.05) at the time of challenge and were protected against CSFV infection, whereas the control pigs died within 14 days post-infection (dpi). In the second experiment, pigs were vaccinated once with either CSFV vaccine or CSFV vaccine combined with Aujeszky's disease (AD) vaccine and challenged at 140 dpv. All vaccinated pigs developed both CSFV-specific, cellular and antibody responses and were protected against CSFV infection. However, differences in cellular, but not antibody, responses were observed in the two vaccinated groups. The group vaccinated with CSFV vaccine developed a significantly higher number of CSFV-specific, IFN-gamma secreting cells (p<0.05), exhibited a shorter fever period and less pathological changes, when compared with the group vaccinated with the combined vaccine. The kinetics of IFN-gamma production, following challenge in the two vaccinated groups, were also different. Taken together, our results indicated that CSFV-specific, IFN-gamma production could be detected early after antigen exposure and correlated with protection against CSFV challenge. Our findings highlight the role of cellular immune responses in porcine anti-viral immunity.  相似文献

The serological response of young and adult sheep vaccinated conjunctivally with Rev-1 vaccine was assessed by fluorescence polarization assay (FPA), Rose Bengal test (RBT), complement fixation test (CFT), modified Rose Bengal test (m-RBT), indirect ELISA (i-ELISA) and competitive ELISA (c-ELISA), at different post vaccination intervals. One hundred and thirty six adult sheep and 64 lambs were used in the study. The vaccinated animals were bled prior to vaccination (0 day) and thereafter at 21st, 42nd, 35th, 63rd, 91st, 125th, 159th, and 223rd and 330th day post vaccination. The majority of animals (young and adult) showed positive reaction by FPA, RBT, CFT, m-RBT and c-ELISA 21 days post vaccination, whereas by i-ELISA at 42 days. All tests perform equal when animals vaccinated as young are tested 125 days (4 months) post vaccination. In case of animals vaccinated at adulthood, FPA, RBT, CFT and c-ELISA perform equal if the animals are tested 223 days (approximately 8 months) post vaccination. I-ELISA and m-RBT show low specificity if ewes vaccinated at adulthood are tested 330 days (11 months) post vaccination. If control of brucellosis in sheep is based on conjunctivally vaccination of lambs with Rev-1, the vaccinated animals can be tested by any test used for diagnosis of B.melitensis infection accurately at least 4 months post vaccination. If brucellosis control is based on mass vaccination the use of m-RBT and i-ELISA is not recommended for testing adult animals at least for 330 days (11 months) post vaccination due to tests low specificity. Further research is needed so the appropriate cut-offs to be established for FPA, c-ELISA or i-ELISA to become valuable tools for the eradication of Brucella spp. infection in small ruminants in areas where vaccination is practiced.  相似文献

Lambs which received colostrum from ewes vaccinated with contagious ecthyma (CE) virus and other lambs vaccinated with CE virus were compared for serum anti-CE immunoglobulin (Ig)G levels, delayed-type hypersensitivity (DTH) responses to CE viral antigen, and protective immunity to challenge with CE virus. Ewes vaccinated 3-4 weeks prior to parturition transferred CE antibody to lambs via colostrum. Although these lambs had higher levels of antibody at challenge than lambs vaccinated when 1-4 days old, only the vaccinated lambs were protected against challenge with CE virus at 1 month of age. Furthermore, the presence of colostrum-derived maternal antibody prevented an active antibody response in lambs to vaccination and/or challenge with CE virus, except where pre-inoculation titres were low. In contrast, the DTH response to CE viral antigen and induction of protective immunity by CE vaccination were not impaired by passively-acquired antibody. Actively immunised lambs could be distinguished from those only receiving passively-acquired antibody by the DTH response to heat-killed CE viral antigen.  相似文献

To evaluate the immune efficacy of porcine reproductive and respiratory syndrome (PRRS) attenuated live vaccine,this study selected 4 litters from two small pig farms (two litters each farm) and vaccinated with porcine reproductive and respiratory syndrome virus (PRRSV) attenuated live vaccine.Sera were collected on different days post vaccination (dpv) to detect PRRSV nucleic acid by RT-PCR,and PRRSV antibody using two different ELISA Kits,i.e.N-ELISA and G-ELISA.The results showed that PRRSV nucleic acid were positive on 0 dpv until 30 dpv in piglets and also positive on 0 dpv in the corresponding sows.All piglets were negative for PRRSV antibody on 0 dpv,but were positive on 30 dpv until 150 dpv.The positive rates detected by N-ELISA Kit were higher than those of G-ELISA Kit on 30 and 60 dpv,but lower than those of G-ELISA Kit on 120 and 150 dpv.A total of 216 sera were detected respectively by two ELISA Kits and the coincidence rate of the results was 95.83%.The P value of χ2 test was more than 0.05,showing there was no significant difference between the results of two Kits.The Kappa value was 0.87,showing there was strong consistence between them.The correlation coefficient was 0.605,showing there was significantly linear correlation between them.The results indicated that the wild-type PRRSV in the previously infected pig herds could be eliminated by vaccination with attenuated live vaccine and both N-ELISA and G-ELISA Kits could be used to estimate the immune efficacy of the attenuated live vaccine effectively.  相似文献

为评估猪繁殖与呼吸综合征(porcine reproductive and respiratory syndrome,PRRS)弱毒活疫苗的免疫效果,本研究在两个小型猪场各选取两窝仔猪,在免疫接种猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)弱毒活疫苗后不同时间采集血清,应用RT-PCR检测PRRSV核酸,应用N-ELISA和G-ELISA两种试剂盒检测PRRSV抗体。结果发现,免疫前0 d可以从仔猪血清中检测到PRRSV野毒株核酸,可持续到免疫后30 d;相对应的母猪在免疫前0 d可从血清中检测到PRRSV野毒株核酸。试验仔猪在免疫前0 d未能检测到PRRSV抗体,但在免疫后30~150 d均可检测到PRRSV抗体,其中N-ELISA试剂盒的阳性检出率在免疫后30、60 d高于G-ELISA试剂盒的阳性检出率,而在免疫后120、150 d则低于G-ELISA试剂盒的阳性检出率。使用两种ELISA试剂盒共同检测216份血清样品,检测结果的总体符合率为95.83%;配对χ2检验,P>0.05,两者差异不显著;Kappa值为0.87,属于极强一致性;两者相关系数r为0.605,具有显著线性相关。表明免疫接种弱毒活疫苗可以有效清除野毒感染猪群中的野毒株,N-ELISA和G-ELISA两种ELISA试剂盒均可用于评估弱毒活疫苗的免疫效果。  相似文献

Selection of resistant animals and host immunization have been proposed as alternative methods for the control of gastrointestinal nematode parasites. However, a better knowledge of the mechanisms involved in protective immunity against these parasites is required for the development of optimal strategies. In this study, 3 month old INRA 401 lambs (n = 81) were allocated into three groups: uninfected control, challenged either once (primary-infected animals) or twice (previously-infected animals) with 10,000 Haemonchus contortus L3. Uninfected control and challenged animals were sequentially sacrificed at 3, 7, 15 and 28 days post challenge. In both challenged groups, a clear Th2-oriented immune response was observed in the abomasal lymph node and mucosa. IL-4 and IL-13 mRNA over-expression, recruitment of eosinophils, mast cells and globule leukocytes and production of specific systemic IgG and mucosal IgA were observed earlier in previously-infected animals than in primary-infected ones. At 28 days post infection, no differences between intensities of these responses were observed between the challenged groups. Worm establishment rates were similar in previously-infected and primary-infected lambs. However, reductions of worm development, female fecundity and fecal egg output were observed in previously-infected sheep. We conclude that H. contortus infection in young INRA 401 lambs induced an unequivocal Th2 immune response resulting in the regulation of worm egg production without affecting their establishment.  相似文献

The objective of this study was to compare the induction of humoral and cell-mediated immune responses by four commercially available single-dose porcine circovirus type 2 (PCV-2) vaccines. A total of 50 3-week-old piglets were assigned to five groups (10 pigs per group). Four commercial PCV-2 vaccines were administered according to the manufacturer's instructions and the piglets were observed for 154 days post vaccination (dpv). Inactivated chimeric PCV-1-2 vaccines induced higher levels of PCV-2-specific neutralizing antibodies (NA) and interferon-γ-secreting cells (IFN-γ-SC) in pigs than did the other three commercial PCV-2 vaccines. The proportions of CD4+ cells were significantly higher in animals vaccinated with inactivated chimeric PCV-1-2 and PCV-2 vaccines than in animals vaccinated with the two subunit vaccines. To our knowledge, this is the first comparison of humoral and cell-mediated immunity induced by four commercial single-dose PCV-2 vaccines under the same conditions. The results of this study demonstrated quantitative differences in the induction of humoral and cell-mediated immunity following vaccination.  相似文献

Eight capuchin monkeys (Cebus apella) were vaccinated against rabies with an inactivated suckling mouse brain vaccine (SMBV). Three 1-ml doses of 2% brain tissue suspension were given by i.m. injection at 0, 30, and 60 days. Blood samples were collected at 0, 30, 60, 90, 150, 210, 240, 300, and 365 days and were tested by simplified fluorescence inhibition to titer-neutralizing antibodies. All of the animals developed neutralizing antibodies with titers >0.5 IU/ml after vaccination, but the immune response persisted for only 122.3 +/- 32.6 days. The SMBV was able to induce immune response in the capuchin monkeys, but protection was short-lived.  相似文献

A field trial was undertaken from 1999 until 2004 to determine the efficacy of a killed M. a. paratuberculosis vaccine, Gudair, for the control of ovine Johne's disease (OJD) in merino sheep run under Australian pastoral conditions. On each of three farms experiencing significant OJD losses (5-15% per annum), 200 merino lambs (age 1-4 months) were vaccinated, and 200 lambs were left unvaccinated. Animal assessments and sample collections were conducted twice yearly until 4 or 5 years of age. The impact of vaccination on mortality rate, faecal shedding of M. a. paratuberculosis (by pooled and individual faecal culture), liveweight, wool productivity, vaccine injection site lesions and cellular (BOVIGAM) and humoral (PARACHEK) immunity was examined. Vaccination stimulated cell-mediated and humoral immune responses, reduced mortalities due to OJD by 90% and delayed faecal shedding for the first year post-vaccination. Thereafter, the prevalence of shedders among vaccinates was reduced by 90%. The numbers of M. a. paratuberculosis excreted by the vaccinated groups were also reduced by at least 90% at most sampling times. However, high levels of excretion by vaccinates occurred on some occasions, and although only 7 of 600 vaccinates died from OJD, all 7 had multibacillary lesions. Thus there remains a risk that some vaccinated sheep will transfer the disease. Small reductions in liveweight were found in vaccinated lambs in the first year, but there was little effect on wool production. Vaccine injection site lesions were detected in almost 50% of sheep after 2 months, and these persisted for at least 4 years in 20-25% of vaccinates. Data from this trial enabled the registration of Gudair in Australia in 2002 and underpins the pivotal role of vaccination in the current management of OJD.  相似文献

Expression of CD25 (interleukin-2 receptor alpha chain) was used to monitor antigen-specific activation of T lymphocyte subsets (CD4+, CD8+, and gamma delta T cells) from cattle immunized with modified-live virus (MLV) bovine viral diarrhea virus (BVDV) vaccines. Two groups of 15 animals each were vaccinated with one dose of either BVDV genotype 1 (BVDV-1) or BVDV-1 and BVDV genotype 2 (BVDV-1/2). Six animals negative for both BVDV antibody and BVDV virus were used as negative controls. Three animals vaccinated 7 and 5 weeks before the start of the experiment with MLV BVDV-1 vaccine served as positive controls. Blood samples were taken from the negative control group, the positive control group, and the BVDV-1/2 group 0, 21, 35, 60, and 90 days after vaccination. Blood samples were taken from the BVDV-1 group 0, 21, and 90 days after vaccination. Isolated peripheral blood lymphocytes from immunized and control animals were incubated for 5 days with and without BVDV-1 or BVDV-2. Compared with nonvaccinated animals, a significant (P <.05) increase in expression of CD25 by CD4+ (60 days), CD8+, and gammadelta T (35 to 90 days) lymphocytes from the group given BVDV-1/2 was detected following in vitro exposure to BVDV-1 or BVDV-2 after vaccination. The CD8+ and gammadelta T cells from the group vaccinated with BVDV-1 had significantly (P <.05) increased expression of CD25 compared with nonvaccinates following postvaccination exposure to in vitro BVDV-1 but not to BVDV-2. There was no significant difference between the two vaccinated groups in CD25 expression on any of the T cell subsets in response to BVDV-1 or BVDV-2 exposure. A single administration of MLV BVDV vaccine may be more effective at stimulating CD8+ and gammadelta T cell-specific immune responses to the homologous genotype than to the heterologous genotype.  相似文献

Mycobacterium avium subspecies paratuberculosis (MAP) is the causative agent of paratuberculosis (ParaTB) also known as Johne’s disease (JD) in ruminants, which is characterized by chronic intestinal inflammation. A similar counterpart has been observed in the form of Crohn's disease in humans. The present study is the first trail in goats to understand the peripheral cellular immune responses following experimental MAP infection and vaccination. Fifteen apparently healthy male kids (3–6 months old) of Barbari breed were included in this study. In the experimental study, 5 kids were infected with ‘S 5’ strain of MAP (“Indian Bison Type”), 5 were vaccinated (Indigenous Vaccine) against MAP infection (Singh et al., 2007) and the remaining 5 kids were uninfected and non-vaccinated controls. Kids were observed for a period of 180 days post exposure (infection and vaccination) and were tested for development of infection. Cellular immune responses (in blood) were recorded post-exposure by three assays. We measured the frequencies of CD4 and CD8T cells, estimated plasma IFNγ and TNα and in the third assay, in vitro cytokine production by peripheral blood mononuclear cells (PBMCs) from vaccinated, infected and controls were examined in response to polyclonal stimulation. The frequencies of peripheral CD4 and CD8T cells were comparable in control, infected and vaccinated animals except around day 49 post-infection where MAP infected animals showed a trend towards significantly reduced frequencies of CD4 T cells compared to apparently healthy controls. Significantly reduced plasma TNFα levels were also observed in infected animals compared to vaccinated animals,during the course of infection. Diminished levels (although non significant) of TNFα were observed in the supernatants from polyclonally stimulated PBMCs at around day 49 post infection. It is conceivable that the diminished cellular immune responses may coincide with an impairment (immune exhaustion) of perhaps antigen-specific CD4T cells that might, in the course of infection, contribute to the progressive nature of caprine paratuberculosis.  相似文献

At present, two types of vaccines against classical swine fever (CSF) virus are commercially available: E2 sub-unit marker vaccines and the conventional attenuated live C-strain vaccines. To evaluate the reduction of the horizontal virus transmission, three comparable experiments were carried out in which groups of weaner pigs (vaccinated with a marker vaccine or a C-strain vaccine) were challenged with CSF virus at 0, 7, and 14 days post-vaccination (dpv). Virus transmission was prevented totally when the challenge occurred at 14 dpv with an E2-marker vaccine (0/12 contact pigs positive in virus isolation (VI); R = 0 (0; 1.5)). At 7 dpv, transmission was reduced slightly (5/12 contact pigs positive in VI; R = 1.0 (0.3; 3.0)), whereas at 0 dpv, vaccination had no effect on transmission (10/12 contact pigs positive in VI; R = 2.9 (1.5; 10.8)). In the C-strain-vaccinated pigs, no virus transmission was detected even when the challenge was performed at the same day as the vaccination (0/12 contact pigs positive in VI; R = 0 (0; 1.5)).  相似文献

An experiment was conducted on 30 male Nellore lambs with average body weight (BW) of 15.45 +/- 0.06 kg to determine the level of the copper (Cu) supplementation in diet from inorganic and organic sources required for optimum immunity and its effect on copper dependent enzymes by allotting them randomly to five groups in completely randomized design. The dietary treatments were viz., basal diet (no Cu supplementation, BD), other four groups were offered BD supplemented with 7 or 14 ppm Cu from copper sulphate (CuSO(4)) and Cu-proteinate, respectively. The lambs were fed the respective diets at 3.5 per cent BW to meet the requirements except Cu for 180 days. The humoral immune response against Brucella abortus and chicken RBC was assessed after 90 days of feeding. The in vivo delayed type hyper sensitivity reaction against PHA-P and in vitro lymphocyte proliferation against Con A indicative of cell mediated immune response (CMI) was assayed at 180 days of feeding. At the end of experiment four lambs from each group were slaughtered for estimation of liver superoxide dismutase activity (SOD). The ceruloplasmin and RBC-SOD activities were higher (P < 0.05) in 14 ppm Cu supplemented lambs from Cu-proteinate at 90 and 180 days, while the liver SOD activity was higher (P < 0.05) in lambs fed 14 ppm Cu from CuSO(4). The STAT titres against B. abortus were higher in Cu supplemented lambs, with no effect of dose of supplementation. Lambs supplemented with Cu-proteinate had higher titers than CuSO(4) on 7 and 14 days of post sensitization. The total immunoglobulin concentration and the CMI response against PHA-P and Con-A was higher (P < 0.05) in lambs fed 14 ppm Cu-proteinae diet. The IgM level was though high in Cu supplemented lambs, no dose or source effect were observed. The study indicated that Cu dependent enzymes activity and immune response were highest and respond better against stress in lambs on 14 ppm supplemented Cu from Cu-proteinate.  相似文献

Antibody responses in indigenous village and commercial chickens vaccinated with 12 thermostable Newcastle disease (ND) vaccine and protection levels against challenge with a virulent field isolate were determined. The antibody response of village chickens vaccinated by eye drop revealed that 30, 60 and 90 days after primary vaccination, the mean log2 HI titres were 6.1, 5.4 and 3.6, respectively, whereas for commercial chickens, the antibody response after 14, 30 and 90 days were 8.2, 5.1 and 4.2, respectively. Village chickens vaccinated orally via drinking water had mean log2 HI titres of 3.4 after 30 days. After booster vaccination, the mean HI titre was 5.4 and 3.3 after 30 and 60 days post-secondary vaccination (i.e. 60 and 90 days after primary vaccination). Antibody response of mean log2 HI titres of 2.6 was recorded 30 days after primary vaccination orally through food; 30 and 60 days after secondary vaccination (i.e. 60 and 90 days after primary vaccination), mean log2 HI titres were 5.3 and 3.2, respectively. All commercial and village chickens vaccinated by eye drop survived the challenge trial whereas village chickens vaccinated through drinking water and food had protection levels of 80% and 60% 30 days after primary vaccination, respectively. However, 30 days after booster vaccination, the protection level was 100%. At 60 days after secondary vaccination, the protection level dropped again to 80% for chickens vaccinated orally. All control chickens used in the challenge trials developed clinical ND and died 3-5 days after inoculation with the virulent virus. Supported by laboratory findings, I2 strain of NDV seemed to be avirulent, immunogenic and highly protective against virulent isolates of NDV. It may be a suitable vaccine to use in village chickens to vaccinate them against ND in rural areas.  相似文献

Twenty helminth-free lambs were fed diets containing either 169 g crude protein (CP) kg-1 dry matter (DM) or 88 g CP kg-1 DM from the age of seven months. One month later five lambs from each dietary group were vaccinated against Haemonchus contortus by the oral administration of 10,000 irradiated larvae on two occasions, four weeks apart. Four weeks following the administration of the second dose of irradiated larvae both the vaccinated and unvaccinated lambs were exposed to an experimental infection of 10,000 non-irradiated H contortus larvae Faecal egg output and haematological changes were monitored throughout the study. The lambs were slaughtered 28 days after challenge when worm burdens were assessed. Vaccination was equally successful in inducing a strong resistance to the challenge infection regardless of dietary status. It was concluded that dietary protein does not influence the response to vaccination with irradiated H contortus larvae of lambs more than seven months old.  相似文献

The aim of this study was to determine the benefits of growth hormone-releasing factor (GRF) on growth and feed conversion efficiency (FCE) in buffaloes. Twelve Murrah buffalo heifers (Bubalus bubalis) of mean age 24.8 months and mean body weight 302.4kg were divided into two groups (treatment and control) with six animals in each group. The buffaloes were given intravenous injections of bovine GRF (bGRF) at a dose rate of 10microg/100kg body weight or an equal volume of saline at 15-day intervals for a period of 9 months. Plasma growth hormone (GH) responses to bGRF challenge were measured in blood samples collected at 90-day intervals on days 1, 90, 180 and 270 and samples were taken at -60, -30, 0, +10, +20, +30, +60, +120 and +180min relative to bGRF injection. Blood samples were also collected weekly by jugular venepuncture for the quantification of plasma GH. The average growth rate (AGR) and FCE of all animals were recorded at 15-day intervals. Plasma GH concentrations increased (P=0.001) steadily following bGRF challenge, peaking 10-20min after challenge and declining to baseline by 180min. In the treatment group, there were no significant differences (P>0.05) in either the peak heights of the GH response or the area under the curve (AUC) of the GH response after bGRF challenge on any of the four occasions of intensive bleeding. There were overall increases in plasma GH concentrations (P<0.01), AGR (P<0.01) and FCE (P=0.05) in the treatment group compared with the control animals. The study showed that GH responsiveness to administration of bGRF at 15-day intervals over 9 months of treatment remained unchanged in buffalo heifers. Exogenous bGRF treatment for a long period can therefore enhance GH release leading to higher growth rates and better feed conversion efficiency in buffalo heifers.  相似文献

Humoral immune responses to vaccination, mean daily body-weight gains, morbidity, and mortality were compared in groups of beef replacement heifers from weaning to 4 months after weaning. The only difference in management among groups of heifers was the number and type of vaccines they received. Heifers were vaccinated at weaning (mean age, 205 days) and again 28 days later against 0, 1, 9, 10, 17, or 18 antigens, using commercially available monovalent and multivalent vaccines. The common vaccine component in all treatment groups was a modified-live bovine respiratory syncytial virus. Mean daily gain, morbidity, mortality, and serum neutralization antibody titers to bovine respiratory syncytial virus did not differ among treatment groups. Although the study revealed the safety of vaccinating beef heifers against 18 antigens at weaning, our data emphasized the need for serial vaccination to induce a measurable serum antibody response.  相似文献

Real-time ultrasound technology offers the possibility of estimating carcass characteristics in live animals and represents a potential method for selection of breeding stocks. A total of 745 live lambs born during 2001–2003 into two flocks was used to estimate rib muscle and fat depth by ultrasound. Lambs came from 559 ewes and 97 rams of a fat-tailed breed, known as the ‘Barbarine’ in North Africa. Ultrasound measurements of external fat thickness (UFD) and muscle (UMD), taken at the 12–13th rib and palpation of body conditions (loin and tail scores) were made for 150 days until lambs 520 days old of lamb ages. Main results showed that UMD and UFD had the same trend as live weights from 150 to 240 days old. They decreased from 150 to 200 days old, and then increased. The same trend was seen for loin and tail scores. Average differences in live weights between male and female lambs were 1, 3 and 6 kg at 30, 90 and 120 days old, respectively. The highest muscle depth for males was reached at day 180 and the lowest fat depth was recorded between 180 and 200 days. Average loin scores became greater for males than females from 240 days of age. Male lambs had greater tail scores at all ages, from 150 to 520 days old. Differences became greater for ages more than 180 days, showing that males have a tendency to store more fat in their tails than females. Phenotypic correlation was 0.70 between muscle depth and loin scores, indicating that, at 180 days old, the Barbarine breed has more muscle and less fat. The optimum slaughter age was defined between 180 and 200 days old, producing carcasses with more muscle and less fat. In order to investigate Rev-1 vaccination immunogenicity, 60 indigenous (Makuii) non-infected female lambs 3–4 months of age in northwestern Iran were systematically divided into two groups: 40 into a treatment (T) group and 20 into a control (C) group. Lambs in Group T received 1 ml of Rev-1 vaccine by s.c. injection and blood samples were collected from both groups at 15 days and 2, 4, 6, 8, 10, 12 and 14 months after vaccination. The seroagglutination test (SAT) and the 2-mercaptoethanol test (2-MET) were carried out on the sera. Control lambs were negative in all the examinations. The results of the SAT showed that the mean( ± SD) antibody titre of lambs in Group T was maximum (725 ± 112 IUml−1) at Day 15, then declined gradually until the fourth month and was in the normal range (31 IU ml−1) thereafter. The results of the 2-MET were negative in Group C but in Group T the antibody titre was at a maximum level (161.7 ± 46.6 IUml−1) at Day 15, decreased to 1.2 ± 2.5IUml−1 at 4 months post-vaccination and was negativ; thereafter. In the second part of the trial, 20 vaccinated and ten control lambs received 0.25 ml melitin by i.d. injection 10.5 months after vaccination. The control group showed no reaction but 13 (65%) of the vaccinated lambs showed an oedematous swelling at the site of injection within 48 h, which indicated that the delayed-type hypersensitivity test (DTHT) could be useful in the diagnosis of Brucella infection in lambs. In the third stage of the experiment, four vaccinated and three control lambs were injected with Brucella melitensis strain 16M at a rate of more than 123 × 106 bacteria per lamb 14 months after vaccination. Rectal temperature (Tem), heart-rate (HR) and respiration rate (RR) were measured three times a day (at 08:00, 14:00 and 21:00 h) 3 days before until 9 days after injection. SAT was carried out on the day before injection and at the time of slaughter. The mean Tem, HR and RR did not significantly change but the mean( ± SD) SAT titre in the T lambs 9 days after injection was 47.5 ± 9.9 IU ml−1 (vs. 8.66 ±6.1 IU ml−1 in the control lambs) (P < 0.01). All lambs were slaughtered on the ninth day. Fewer bacterial cultures of samples taken from the uterus, spleen, liver and lymph nodes were positive in T lambs compared with the controls. The lower proportion of Brucella-positive lambs in the vaccinated group compared with the control group indicates that vaccination with Rev-1 improved the resistance of lambs to the experimental infection.  相似文献
