Use of live and dead probiotic cells in tilapia Oreochromis niloticus   总被引:3,自引:0,他引:3  
ABSTRACT:   To investigate the effect of live and dead probiotic cells on the non-specific immune system of tilapia Oreochromis niloticus , probiotics were introduced by feeding either in the form of live or dead cells, or supplying live cells to the rearing water in a closed recirculating system. The probiotics treatment enhanced non-specific immune parameters such as lysozyme activity, migration of neutrophils and plasma bactericidal activity, resulting in improvement of resistance to Edwardsiella tarda infection. Especially, oral administration of live cells seemed to be more effective compared with other probiotic treatments such as oral administration of dead probiotic cells and supply of live probiotic cells to the rearing water. These results indicate that probiotics treatment is promising as an alternative method to antibiotics for disease prevention in aquaculture, and the viability of probiotic bacteria is a key factor to induce more potential effect of probiotics used for fish production.  相似文献

The objective of this experiment was to test the efficacy of a probiotic (Efinol®L) during transportation of marbled hatchetfish, Carnegiella strigata . Wild specimens were captured from a small stream and transported for 24 h in plastic fish boxes with a probiotic (10 mg L−1) and probiotic-free water. The boxes were sampled at 3, 12 and 24 h of transport. At the end of the experiment, the survival rate was close to 100% in both treatments. Dissolved oxygen diminished with time in both treatments, but the probiotic group had significantly higher levels. Conductivity, pH and ammonia increased significantly during the transport, demonstrating higher levels in the probiotic-free group. Fish from both treatments presented very high net Na+ and K+ effluxes after 3 h of transport. At 24 h, net K+ effluxes in fish of the probiotic treatment reached values close to zero and a significantly lower Na+ efflux was observed. Cortisol levels in both treatments at 3 and 12 h were significantly higher than that in control samples. Higher body cortisol levels were observed in the probiotic-free group than that in the probiotic group at 3 and 12 h. The results demonstrate that addition of a probiotic during fish transport improves water quality and leads to fish presenting a lower stress response intensity.  相似文献

ABSTRACT:   Experiments were conducted to determine the acute tolerance of juvenile (mean weight ± standard error, 9.9 ± 0.9 g) black sea bass Centropristis striata to environmental un-ionized ammonia-nitrogen (NH3-N) and nitrite-nitrogen (NO2-N) exposure at various salinities. Specifically, median lethal concentrations (LC50 values) of NH3-N and NO2-N at 24, 48 and 96 h of exposure were determined at salinities of 10, 20 and 30 g/L at 22°C. With the exception of LC50 values determined at 48 h, median lethal concentrations of NH3-N to black sea bass were not influenced by environmental salinity; 24, 48, and 96 h LC50 values ranged from 0.81 to 0.85, 0.65–0.77, and 0.46–0.54 mg-NH3-N/L, respectively. In contrast, tolerance of black sea bass to environmental NO2-N was compromised at reduced salinities. Median lethal concentrations of NO2-N to fish at 24, 48 and 96 h of exposure ranged from 288.3 to 429.0, 258.4 to 358.8 and 190.0 to 241.9 mg-NO2-N/L, respectively. Results indicate that while juvenile black sea bass are relatively sensitive to acute NH3-N exposure, they are highly resistant to NO2-N exposure.  相似文献

The effects of two selected probiotics ( Pseudomonas synxantha and Pseudomonas aeruginosa ) on the specific growth rate (SGR), survival and immune parameters of juvenile western king prawns ( Penaeus latisulcatus ) were examined for 84 days. Pseudomonas synxantha and P. aeruginosa were either applied into the rearing medium or supplemented in the formulated feed at a concentration of 105 colony forming unit (CFU) mL−1 for each probiotic. The results showed that applications of the probiotics resulted in no significant difference ( P >0.05) in the SGR and survival of the prawns, but significantly decreased the food conversion ratios ( P <0.05) compared with the control where no probiotics were applied. The supplement of the probiotics with the formulated feed enhanced the probiotic numbers in the intestines of the prawns and low bacteria numbers in the haemolymph. The prawns were significantly ( P <0.05) healthier, as indicated by the immune response obtained when both probiotics were given in combination compared with when individual probiotics were used. When applied singly, P. aeruginosa resulted in a higher health status of the prawns compared with P. synxantha . Overall, equal combinations (105 CFU mL−1) and volumes of P. synxantha and P. aeruginosa used as a supplement in the formulated feed resulted in an improvement in the health of the juvenile western king prawns.  相似文献

This experiment was carried out to evaluate the effects of the probiotic, Lactobacillus acidophilus , on the growth performance, haematology parameters and immunoglobulin concentration in African catfish Clarias gariepinus fingerling. Two experimental diets were formulated to contain 35 g kg−1 crude protein and 10 g kg−1 lipids accordingly and fed three times daily for 12 weeks to 25 C. gariepinus fingerlings per fibreglass tank in 12 replicates each. The control diet was prepared with no probiotic supplementation whereas the second diet was prepared supplemented with a probiotic, L. acidophilus , containing about 3.01 × 107 colonies/g of diet. The results show that growth performance [specific growth rate (SGR) and relative growth rate (RGR)], nutrient utilization [protein efficiency ratio (PER) and feed conversion ratio (FCR)] and survival were significantly ( P <0.05) higher in fish maintained on the probiotic-supplemented diet compared with those on the control diet. Haematology parameters (packed cell volume, haemoglobin, erythrocyte sedimentation rate, red blood cell and white blood cell, total serum protein, Ca2+, Mg2+, Cl, glucose and cholesterol) and total immunoglobulin concentrations were also significantly better in fish fed the probiotic-supplemented diet than in the control. Although the water quality parameters monitored were better in the fish fed the probiotic-supplemented diet than in the control, the parameters were not significantly different ( P >0.05). From the results of this experiment, we conclude that L. acidophilus can be used as a probiotic agent in African catfish culture, to enhance fish health, survival and better feed efficiency and growth performance.  相似文献

The objective of this experiment was to test the probiotic Efinol®L during transportation of cardinal tetra, Paracheirodon axelrodi (Schultz). For the transportation, fish were distributed in 18 plastic tanks, of which nine contained the Efinol®L (10 mg L−1; probiotic treatment) and the remaining had no probiotic (control treatment). Transport lasted 24 h and three different boxes of each treatment were sampled at 3, 12 and 24 h. Up to the 12-h sampling period, no significant difference in the survival was observed; on the other hand, survival was higher at the end of the transport (24 h) in the probiotic treatment. No significant difference was found in dissolved oxygen and temperature between treatments. Conductivity, pH and alkalinity increased along the transport, but without a difference between treatments. Ammonia increased in all treatments, although it was significantly lower in the probiotic group at 24 h. Cortisol levels were significantly higher in all transported fish when compared with the basal values. At 24 h, cortisol levels in control fish were significantly higher than those in the probiotic treatment. With the observed results, we are able to conclude that the probiotic Efinol®L is efficient during cardinal transport, lowering the mortality and helping maintain water quality possibly by lowering metabolic wastes.  相似文献

ABSTRACT:   Fast skeletal muscles of Japanese flounder Paralichthys olivaceus and red sea bream Pagrus major were examined for quantitative and qualitative changes of mitochondrial ATP synthase (FoF1-ATPase) in association with rearing temperatures. The specific activities of FoF1-ATPase from Japanese flounder reared at 10°C, 15°C and 25°C for 4 weeks were determined to be 81 ± 11, 74 ± 13 and 83 ± 11 nmol/min·mg mitochondrial protein, respectively. The corresponding activity from red sea bream reared at 8°C for 5 weeks was determined to be 65 ± 9 nmol/min·mg mitochondrial protein, which was higher than 33 ± 9 nmol/min·mg mitochondrial protein in fish reared at 23°C. The contents of α- and β-F1-ATPase in total mitochondrial proteins were not significantly different between fish reared at different temperatures for the two fish species. However, the contents of β-F1-ATPase in the total fast skeletal muscle extracts, prepared from Japanese flounder reared at 10°C, were 2.1- and 2.9-fold higher than those for fish reared at 15°C and 25°C, respectively. The corresponding content from red seabream reared at 8°C was 2.2-fold higher than that for fish reared at 23°C. Therefore, the changes in FoF1-ATPase depending on rearing temperatures were species-specific.  相似文献

Growth and amino acid oxidation studies were conducted to estimate methionine requirement of juvenile Japanese flounder, Paralichthys olivaceus , by using the purified diets containing 500 g kg–1 crude protein from casein, gelatine and crystalline amino acids (CAA). Diets with six graded levels of methionine (5.3, 8.3, 11.3, 14.3, 17.3 and 20.3 g kg–1 diet) were fed to triplicate groups of the juvenile (initial weight 2.8 ± 0.05 g) twice a day for 40 days. To prevent leaching losses, CAA were precoated using carboxymethylcellulose (CMC), and further diets were bound by CMC and κ-carrageenan. Based on broken-line analysis of percentage weight gain and feed conversion efficiency, the methionine requirements of Japanese flounder in the presence of 0.6 g kg–1 of cystine were 14.9 and 14.4 g kg–1 dry diet, respectively. After the growth study was finished, a direct estimate of methionine requirement was made by examining the influence of dietary methionine level on 14C-methionine oxidation by determining radioactive carbon dioxide, protein and nonprotein fractions of the whole body. The dose–response curve between expired radioactive CO2 and dietary methionine levels showed that the optimum methionine level for the flounder was estimated to be within the range of 14.3–17.3 g kg–1 of diet in high agreement with values obtained from the growth study.  相似文献

ABSTRACT:     To improve stock enhancement strategies, the influences of repeated cross-breeding on several traits were studied in hatchery-reared juveniles of the Japanese flounder. Ten families (seven caught in the wild and three from cross-bred brood fish) were produced by artificial fertilization. Ninety-four days after hatching, we investigated settlement, burying in the sand and escape behavioral traits, and then measured tolerance of high salinity and six morphological traits. Juveniles from the repeatedly cross-bred brood fish showed significantly higher growth and tolerance than those from the wild-caught brood fish. Among the behavioral traits, a significantly lower settling speed and incidence of burying in the sand were observed in the juveniles from the repeatedly cross-bred families. Heritability ( h 2) of growth traits was more than 0.2 in both origins, while that of other traits was close to zero. These results suggest that repeated cross-breeding of Japanese flounder under hatchery conditions improves growth performance and leads to the loss of certain behavioral traits that are considered essential to survival in wild environments.  相似文献

ABSTRACT: The present study was conducted to investigate the effects of dietary vitamin A on reproduction and egg quality in Japanese flounder Paralichthys olivaceus . Broodstock were fed experimental pellets containing two levels of vitamin A [11 × 103 IU/100 g (control diet; CD), 337 × 103 IU/100 g (experimental diet; ED)] for approximately 2 months before spawning and during the spawning period. Two groups of five females (average weight 1.4 kg) and 10 males (average weight 0.7 kg) were randomly allocated to two 30 m3 indoor tanks. Total egg production of the CD group was slightly higher than the ED group. Percentage of buoyant eggs and hatching rate of the ED group were significantly higher than the CD. In other egg quality parameters, such as percentage abnormal larvae and starvation tolerance of larvae, no notable difference was found between these two groups. At the end of the experiment, the skin color of broodstock in the ED group was darker than that of the CD group. Vitamin A content in eggs of the ED group was significantly higher than that of the CD group. However, the difference in vitamin A content in eggs between the ED and CD groups was much smaller than that in the liver of the females between the two groups. These results indicate that feeding broodstock a higher level of vitamin A increases the vitamin A content in eggs but does not affect egg quality in Japanese flounder because excess dietary vitamin A was stored mainly in the broodstocks' liver.  相似文献

Five iso-nitrogenous (300 g crude protein kg−1 diet) semi-purified diets with graded levels of carbohydrate at 220 (D-1), 260 (D-2), 300 (D-3), 340 (D-4) and 380 (D-5) g kg−1 diet were fed ad libitum to Puntius gonionotus fingerlings (average weight 0.59±0.01 g) in triplicate groups (20 fish replicate−1) for a period of 90 days to determine the effect of the dietary carbohydrate level on the growth, nutrient utilization, digestibility, gut enzyme activity and whole-body composition of fish. Fifteen flow-through cement tanks of 100 L capacity with a flow rate of 0.5 L min−1 were used for rearing the fish. The maximum weight gain, specific growth rate, protein efficiency ratio, RNA:DNA ratio, whole-body protein content, protease activity, protein and energy digestibility and minimum feed conversion ratio (FCR) were found in the D-2 group fed with 260 g carbohydrate kg−1 diet. The highest protein and energy retention was also recorded in the same group. However, from the second-order polynomial regression analysis, the maximum growth and nutrient utilization of P. gonionotus fingerlings was 291.3–298.3 g carbohydrate kg−1 diet at a dietary protein level of 300 g kg−1 with a protein/energy (P/E) ratio of 20.58 −20.75 g protein MJ−1.  相似文献

ABSTRACT:   An 8-week feeding trial was conducted to determine the optimum dietary phosphorus level of juvenile Japanese flounder Paralichthys olivaceus reared in the recirculating system. The basal diet containing 0.33% total phosphorus (TP) was supplemented with NaH2PO4·H2O to provide 0.33, 0.51, 0.71, 0.94, 1.10 and 2.12% TP for the experimental diets. Fish averaging 2.0 ± 0.04 g (mean ± SD) were fed in triplicate groups with the experimental diets for 8 weeks. After 8 weeks, fish fed the 0.51% TP diet showed significantly higher weight gain (WG), feed efficiency (FE), specific growth rate (SGR), and protein efficiency ratio (PER) than those fed the 0.33% and 2.12% TP diets ( P  < 0.05). Meanwhile, no difference was observed in WG, FE, SGR and PER of fish fed the 0.51, 0.71, 0.94 and 1.10% TP diets. Fish fed the 0.33% TP diet showed phosphorus deficiency symptoms such as poor growth and a deformed head by the end of 8 weeks. Ash and phosphorus contents of bone increased with the increase of dietary phosphorus concentrations. anova test, the quadratic regression analysis, and the broken line analysis suggested that the optimum dietary phosphorus level could be between 0.45 and 0.51% TP for maximum WG in juvenile Japanese flounder.  相似文献

The use efficiency and feed conversion of extruded and pelletized diets were compared. Eight isoproteic diets (220 g kg−1 digestible protein) were assayed for 90 days in a 2 × 2 × 2 multifactorial design with two carbohydrate levels (400 and 500 g kg−1), two lipids levels (40 and 80 g kg−1) and two diet processing (pelletization and extrusion) with three repetitions. The growth of Piaractus mesopotamicus fed with these diets and the quality control indices of diets were gauged. The density of extruded diets was lower as carbohydrate level was 400 g kg−1 and lipid 40 g kg−1. The interaction carbohydrate and diet processing presented higher leaching value for low carbohydrate level in extruded diet. Fish fed with extruded diets presented the best feed conversion and protein efficiency ratio. When high levels of carbohydrate and lipid are combined, the weight gain is impaired. The interaction between diet processing diet and lipid levels resulted in the best fish performance when pelletized diets with 40 g kg−1 lipid or extruded diets with 80 g kg−1 lipid were considered. The protein efficiency ratio increased with the increment of carbohydrates in the pelletized diets. The fish show low tolerance to lipids and a preference for carbohydrate when the lipid productive values are taken into account.  相似文献

Five iso-nitrogenous (300 g kg−1 diet) purified diets with graded level of lipid at 40 (D-1), 60 (D-2), 80 (D-3), 100 (D-4) and 120 (D-5) g kg−1 diet were fed to Puntius gonionotus fingerlings for 90 days to determine their dietary lipid requirement. Two hundred and twenty-five fingerlings (average weight 2.34 ± 0.03 g) were equally distributed in five treatments in triplicate groups with 15 fish per replicate. Fifteen flow-through cement tanks of 100 L capacity with a flow rate of 0.5 L min−1 were used for rearing the fish. Specific growth rate (SGR), feed conversion ratio (FCR), nutrient digestibility, retention, digestive enzyme activity, RNA : DNA ratio and whole-body composition were considered as the response parameters with respect to dietary lipid levels. Maximum SGR and minimum FCR with highest RNA : DNA ratio, whole-body protein content and digestive enzyme activity was found in D-3 group fed with 80 g kg−1 diet lipid. Nutrient digestibility was similar in all the groups irrespective of the dietary lipid level. Maximum protein and energy retention was recorded at 80 g kg−1 dietary lipid fed group. However, from the second-order polynomial regression analysis, the maximum growth of P. gonionotus fingerlings was found at 96.9 g lipid kg−1 diet.  相似文献

Five iso-nitrogenous (300 g protein kg−1 diet) and iso-lipidic (80 g kg−1 diet) semi-purified experimental diets with variable energy levels of 10.5 (D-1), 12.5 (D-2), 14.6 (D-3), 16.7 (D-4) and 18.8 (D-5) MJ kg−1 diets were fed to Puntius gonionotus fingerlings (average weight 1.79 ± 0.02 g) in triplicate groups (15 healthy fishes per replicate) for a period of 90 days to assess the optimum dietary energy level and protein-to-energy ratio (P/E). Fifteen flow-through cement tanks of 100 L capacity with a flow rate of 0.5 L min−1 were used for rearing the fish. Maximum specific growth rate, protein efficiency ratio, protein productive value, RNA : DNA ratio, whole body protein content, digestive enzyme activity and minimum feed conversion ratio was found in fish-fed diet D-3 with 14.6 MJ kg−1 energy level. There were no improvements in all these parameters with the further rise in dietary energy level. Hence, it may be concluded that the optimum dietary gross energy level for maximum growth and nutrient utilization of silver barb is 14.6 MJ kg−1 diet with a resultant P/E ratio of 20.2 g protein MJ−1 diet, when the dietary protein and lipid are maintained at optimum requirement levels of 300 and 80 g kg−1 diet, respectively, for this species.  相似文献

This experiment, designed for the first time in the Middle East region to sustain Coregonus lavaretus production, examined the efficacy of a live organism ( Brachionus plicatilis ), an artificial diet (48% crude protein) and a mixed feed for the first 3 month of rearing fry. Nine rearing cages were floated on the surface of a natural pond supplied with hyporheic-zone river water, each containing 150 numbers of white fish fry, where they were treated with each of the three diets in triplicates. The fry were fed eight times daily starting at the onset of exogenous feeding. Results showed that fry utilized live organisms more efficiently than the artificial and mixed diets. Rotifer-fed group consistently showed higher growth rate during the entire experiment. Length increment (29.6 mm), weight gain (572.3 mg) after 12 weeks of feeding were higher ( P  < 0.05) in this group. Also, their mean weekly specific growth rate (4.97% day−1) was the highest and feed conversion rate (2.97) was the lowest among all treatments. Survival rates of the fry declined from 75% to 61.4%, from 74.3 to 45.8% and from 72.7 to 54.5% for live feed, mixed diet and artificial diet, respectively.  相似文献

ABSTRACT:   The effectiveness on water quality, population density of bacteria, and shrimp production in ponds treated with commercial probiotics was tested in Penaeus vannamei ponds in Hai-yan, China. Six ponds with replicates for treatment and control were used. Results showed that the probiotics could improve the population density of beneficial bacterial flora, reduce concentrations of nitrogen and phosphorus, and increase yields of shrimp. The average counts of Bacillus sp., ammonifying bacteria, and protein mineralizing bacteria were found to be significantly higher in treated ponds compared to control ponds ( P  < 0.05). In control ponds, an increase in presumptive vibrios was observed and the average density was up to 2.09 × 103 cfu/mL, whereas that was only 4.37 × 102 cfu/mL in treated ponds ( P  < 0.05). The use of probiotics also significantly increased dissolved oxygen ( P  < 0.05) and reduced dissolved reactive-phosphorus, total inorganic nitrogen and chemical oxygen demand ( P  < 0.05). An average of 8215 ± 265 kg shrimp/ha was obtained in treated ponds with a feed conversion ratio (FCR) of 1.13 ± 0.05 and survival rate of 81.00 ± 6.25% compared with 4985 ± 503 kg shrimp/ha, 1.35 ± 0.12 and 48.67 ± 3.51%, respectively, in control ponds. This indicates that the addition of the commercial probiotics had a noticeable influence on water quality of shrimp ponds and shrimp production.  相似文献

ABSTRACT:     The acute toxicities of boron were examined for Japanese flounder Paralichthys olivaceus and red sea bream Pagrus major in terms of fish size and water temperature. Japanese flounder of 0.1–70.0 g and red sea bream of 0.6–20.3 g were exposed to different concentrations of boron for 96 h at 20°C under semi-static conditions. In both fish species, the median lethal concentration (LC50) for 96 h of boron increased linearly with increasing fish size, ranging from 108 to 252 mg B/L for the flounder, and from 97 to 172 mg B/L for the sea bream. The effect of water temperature on acute toxicity was examined for Japanese flounder of 0.6 and 1.5 g at 10, 15, 20 and 25°C, and for red sea bream of 0.6 and 2.4 g at 12, 15, 20 and 25°C. The toxicity of boron for the flounder increased linearly with increasing water temperature. The 96 h LC50 values ranged from 299 to 108 mg B/L for the 0.6 g flounder and from 350 to 113 mg B/L for the 1.5 g flounder. A similar trend was shown for the 2.4 g red sea bream; however, the relationship for the 0.6 g red sea bream was not significant.  相似文献

Olive flounder artificially infected with lymphocystis disease virus (LCDV) were reared at 10, 20 and 30 °C for 60 days, to compare LCD-incidence. In the fish reared at 20 °C, lymphocystis cells appeared on the skin and fins at 35 days post-challenge, and the cumulative LCD-incidence was 80% at 60 days. High levels of LCDV, with a mean polymerase chain reaction (PCR) titre of 106 PCR-U mg−1 tissue, were detected in the fins and skin of LCD-affected fish at 20 °C, but were not detected in the spleen, kidney, brain and intestinal tissues of these fish. No LCD clinical signs were observed in the fish reared at 10 °C and 30 °C; however, a low level of LCDV (103 PCR-U mg−1 tissue) was detected in the fins and skin of these fish. By increasing the rearing temperature from 10 to 20 °C, lymphocystis clusters appeared on the skin and fins of the fish with no previous LCD clinical signs within 33 days after the temperature change. It was shown that permissive cells for LCDV infection exist in the epidermis of olive flounder. At low temperatures, small amounts of LCDV were able to persist over a period extended for a further 45 days in the fish epidermis, even though the fish showed no LCD clinical signs. The optimum growth temperature of LCDV is near 20 °C.  相似文献

Two trials were conducted to determine the effects of dietary enrichments with the microalga Parietochloris incisa , rich in arachidonic acid (ARA), on stress resistance in guppies Poecilia reticulata . The microalga was added to commercial diets as a neutral lipid (NL) extract and its fractions or as broken cells. Experimental diets were applied for a period of 14 days. In trial 1, commercial diets were supplemented with NL (containing 25 mg ARA and 0.11 mg β-carotene g−1 feed), its triacylglycerol (TAG) fraction (containing 25 mg ARA g−1 feed and no β-carotene) and the β-carotene fraction (containing 0.11 mg carotenoid g−1 feed and minute amounts of ARA). Neutral lipid-fed fish demonstrated the highest resistance ( P <0.05) to osmotic stress (32-ppt NaCl), followed by fish fed with diets supplemented with TAG and β-carotene alone, which were more resistant than control ( P <0.05). In trial 2, fish fed diets supplemented with higher levels of broken alga (26.1 mg ARA g−1 feed) were more resistant ( P <0.05) to stress as compared with fish fed lower ARA (16.3 mg g g−1) or an unsupplemented control diet. We suggest a dietary supplementation with broken P. incisa cells to enhance stress resistance in guppies before a stressful event.  相似文献
