The genetic diversity among 126 exotic (108) andreference array (RA) melon (Cucumismelo L.) accessions (18) was assessed byvariation at 49 random amplified polymorphic DNA marker bands(putative loci) using 29 10-mer primers. Africanaccessions of unknown melon market classes were compared to the RAaccessions from a broad range of C.melo subsp. melo groups(Cantalupensis, Conomon, Inodorus and Flexuosus). Althoughdifferences in groupings occurred after multidimensional scaling andcluster analysis, both analyses placed African accessions into twogroups, which were separate from RA groupings. One African group of33 accessions containing accessions from Zimbabwe (5),Zambia (24), Mali (1), one of two Senegalaccessions and two of three South African accessions examined. Thesecond group, which consisted of 67 accessions containing collectionsfrom Egypt (40), Tunisia (6), Libya(13), Morocco (1), Algeria (2),Ethiopia (1), Niger (1), Sierra Leone(1), S. Africa (1), Zambia (1) andZimbabwe (1). Depending on the multivariate analysistechnique employed, accessions from Kenya, Senegal and Ghana formedeither unique groupings or were grouped with accessions(Cantalupensis) from the RA. Both analyses indicate thatthe genetic differences inherent between the African gene pools isassociated with the geographic proximity of African countries(northern vs. central-southern Africa) in thegermplasm array examined. Moreover, these data indicate that thegenetic diversity of U.S. and European commercial RA germplasm(Cantalupensis and Inodorus) could be enhanced by theintroduction of genetic variation from African accessions, and thatit would be advantageous to acquire more accessions from thisgeographically and ecologically varied region to ensure the retentionof existing genetic diversity.  相似文献

Genetic variation in cucumber (Cucumis sativus L. var. sativus) accessions from China was assessed by examining variation at 21 polymorphic isozyme loci. One hundred and forty-six Chinese accessions acquired by the U.S. National Plant Germplasm System (NPGS) before 1994 were compared to 67 recently collected Chinese accessions. Of the 67, 39 were collected during a 1994 U.S.–China expedition and 28 were donated in 1996. Isozymic profiles of all of these Chinese accessions were also compared with 853 previously examined NPGS cucumber accessions. Principal component analysis of allele frequencies identified two distinct groups of Chinese accessions: one containing accessions collected in 1994 (Group 1) and the other containing accessions received by NPGS in 1996 (Group 2) (P<0.01). Variation at Gpi, Gr, Mdh-2, Mpi-2, Pep-gl and Pep-la was important in the detection of this difference. Isozymic variation in U.S. NPGS accessions of Chinese origin acquired before 1994 differed significantly (P<0.01) from those collected during 1994 and those received in 1996. Eleven loci (Fdp-1, Gpi, Gr, Idh, Mpi-2, Pep-gl, Pep-la, Pep-pap, Per, Pgm and Pgd-1), were important in the detection of this difference. When Chinese accessions taken collectively (i.e., those acquired before 1994, and those during 1994 and 1996) were compared with an array of 853 C. s. var. sativus accessions examined previously, relationships between accessions grouped by country or subcontinent differed from those found in the previous work. Accessions from China and India were distinct from each other and from all other groupings and the C. s. var. hardwickii (feral form of C. s. var. sativus) accessions examined. Thus, Chinese and Indian accessions represent the most diverse genetic variation present in the NPGS cucumber collection. Further collection in India and China would be strategically important for increasing genetic diversity of the U.S. NPGS cucumber collection.  相似文献

The genetic diversity among 67 melon (C. melo L.) cultivars from five Japanese seed companies was assessed using 25 10-mer RAPD primers (56 bands) and nine SSR (36 alleles) markers. These cultivars belong to three horticultural varieties (synom. Groups) spanning eight melon market classes: Group Cantalupensis (market classes Earl’s, House, Galia, Charentais, and Ogen), Group Inodorus [Honeydew and Casaba melons (market classes Amarillo, Piel de Sapo, Rochet, Negro, Crenshaw, and Tendral)], and Group Conomon (market class Oriental). Genetic variation among these cultivars was compared to variation in a reference array (RA) consisting of 34 selected melon accessions from previous studies. Cluster analysis resulted in 11 of 15 Japanese Oriental accessions forming a group with South African RA accessions. The remaining Group Conomon Japanese accessions grouped either with Casaba or with Honeydew cultivars. Japanese Group Conomon accessions and South African RA accessions formed a genetic group that was distinct from all other accessions studied, and suggests either an Asiatic origin for the South African melon germplasm examined or an independent domestication involving similar ancestors. The majority of Japanese House and Earl market class accessions shared genetic affinities, and were genetically different from the Japanese Group Inodorus accessions examined. These Japanese accessions were most similar to Casaba RA accessions. Japanese Galia accessions were similar to either House and Earl’s market classes or to Galia, Ogen, Casaba, and Honeydew RA accessions. Genetic differences exist between melon types that were domesticated from wild, ‘free-living’ subspecies agrestis and from melo. Genetic variation within the U.S. cucumber collection (Cucumis sativus var. sativus L. and var. hardwickii (Royle) Alef.) was assessed by examing the variation at 21 polymorphic isozyme loci and comparing the results of this investigation with a similar previous analysis of 14 loci. About 29% (15 of 51) of the enzyme systems examined in an initial survey were polymorphic. Seven loci (Ak-2, Ak-3, Fdp-1, Fdp-2, Mpi-1, Pep-gl and Skdh) which were not previously used to estimate genetic diversity, were assessed. On average, 1.4 loci were polymorphic per enzyme system and 2.2 alleles were present per polymorphic locus. The frequency of polymorphisms was relatively low for Fdp-1(2) (0.01), Mpi-1(1) (0.03), and Skdh(1) (0.02). Principal component and cluster analyses of allelic variation at polymorphic loci separated a diverse array of 757 cucumber accessions from the U.S. National Plant Germplasm Systesm's (NPGS) collection into distinct groups by country (45 nations examined). All accessions of C. s. var. sativus were isozymically distinct from C. s. var. hardwickii, which were themselves dissimilar from each other. Data suggest that C. s. var. hardwickii is not a feral derivative of extant C. s. var. sativus populations. The allelic profile of C. s. var. sativus accessions originating from Burma, Thailand, Indonesia, Hong Kong, Zimbabwe and Ethiopia were distinct from the other accessions examined. Allelic fixation has occurred at Pgd-2 in accessions from Burma, and at Ak-2 in accessions from Zimbabwe and Ethiopia. Some of the countries examined that were in close geographic proximity (e.g., Thailand, Indonesia and Hong Kong) contained accessions with similar isozyme profiles. Accessions are fixed for certain alleles [e.g., Gr(1) (100%), Fdp-1(1) (100%) and Mpi-2(2) (50%) for accessions from Thailand, Indonesia and Hong Kong]. Grouping countries by continent or sub-continent (i.e., North and South American, China, Eastern Europe, Western Europe) and by numbers of accessions examined (i.e., India/Burma, Iran, Japan, Turkey, and remaining accessions) was used to identify accessions with unique allozymic profiles [PIs 209064 (USA), 257486 (China), 188749 (Egypt), 285607 (Poland), 369717 (Yugoslavia), 357844 (Poland), 255936 (Netherlands), 183127 (India), 200818 (Burma), 200815 (Burma), 137836 (Iran), 227013 (Iran), 227235 (Iran), 451976 (Japan), 181752 (Syria), 181874 (Syria), 169383 (Turkey), 171613 (Turkey)].  相似文献

Genetic variation in cucumber (Cucumis sativus L. var. sativus) accessions from India was assessed by examining variation at 21 polymorphic isozyme loci. Forty-six accessions acquired by the U.S. National Plant Germplasm System (NPGS) before 1972 were compared with 146 accessions collected during a 1992 U.S.–India expedition to the states of Rajasthan, Madhya Pradesh and Uttar Pradesh, India. Isozymic profiles of these Indian accessions were also compared with 707 previously examined U.S. NPGS cucumber accessions. Two distinct groups (Group 1 and Group 2) were identified within accessions collected in 1992 (0.025 > P > 0.01). Variation at Ak-2, Fdp-2, Gr, Mdh-2, Mpi-1, Per, Pgm and Skdh was important in the detection of this difference. A previously unreported Pgm allele [Pgm (3) – 105] was detected in accessions collected in a market in Pali, Rajasthan. Group 1 contained 37 (27 Madhya Pradesh + 10 Uttar Pradesh) accessions and Group 2 contains 102 (84 Rajasthan + 18 Madhya Pradesh) accessions. Seven accessions (5 Madhya Pradesh + 2 Rajasthan) were not associated with either group. The accessions 20664 (Tonk, Rahjasthan), 20666 (Jaipur, Rahjasthan), 20872 (Sehore, Madhya Pradesh), 20881 (Ashtok, Sehore, Madhya Pradesh) and 21026 (Bhatta, Dehra Dun, Uttar Pradesh) were heterozygous for at least nine loci and represent the genetic diversity within this collection. Isozymic variation in U.S. NPGS accessions acquired before 1972 differed significantly (P > 0.005) from those collected during 1992. All loci were important in the detection of this difference, except Ak-2, Pep-pap, and Pgd-2. When Indian accessions taken collectively (i.e., those acquired before 1972 and during 1992) were compared with an array of 707 C. sativus accessions examined previously, relationships between accessions grouped by country or subcontinent differed from those found in previous work.  相似文献

The molecular diversity and population structure present in the Ethiopian sorghum collection maintained at the USDA–ARS National Plant Germplasm System (NPGS) has not been studied. In addition, 83 % of the accessions in the Ethiopian collection lack passport information which has constrained their evaluation and utility. Therefore, 137 Ethiopian accessions from NPGS were randomly selected and characterized with 20 strategically selected simple sequence repeat markers. These markers indentified 289 alleles with average polymorphic information content of 0.78. The allele frequency distribution reflects that 62 % of the alleles were rare (<0.05), 17 % range from 0.05 to 0.10, and 22 % were higher than 0.10. Expected and observed heterozygosity were estimated at 0.78 and 0.23, respectively, demonstrating Ethiopia has high sorghum genetic diversity germplasm. Population structure analysis indentified two subpopulations of 77 and 41 accessions, respectively, while a third group was constituted by 19 accessions whose classifications were not defined (i.e. hybrids). Analysis of molecular variances determined variation within subpopulations as the major source of variation. Likewise, genetic differentiation between subpopulations was moderate (Fst = 0.10). These results indicated that a continuous exchange of genes between subpopulations of sorghum exists in Ethiopia. The absence of a well defined population structure positioned this germplasm as an important resource for the study and dissection of agricultural traits by association mapping. However, genetic redundancy analysis indicated the presence of highly related accessions, therefore, strategic selected accessions must be consider prior to phenotype evaluation of larger number of accessions. The Ethiopian collection is composed of highly genetically diverse germplasm, and the genetic information presented herein is valuable to ex situ and in situ conservation programs to promote the use of this germplasm for breeding programs.  相似文献

Over the past three centuries, maize has become adapted to complex environmental conditions in the highlands of Ethiopia. We analyzed 62 traditional Ethiopian highland maize accessions, using 20 simple sequence repeat (SSR) markers and 15 morphological traits, to assess genetic diversity and relationships among these accessions and to assess the level of correlation between phenotypic and genetic distances. The accessions varied significantly for all of the measured morphological traits. The average number of alleles per locus was 4.9. Pair-wise genetic dissimilarity coefficients ranged from 0.27 to 0.63 with a mean of 0.49. Ward minimum variance cluster analysis showed that accessions collected from the Northern agroecology were distinct from the Western and Southern agroecologies. However, there was no differentiation between the Western and Southern accessions. This suggested gene flow between these regions. The relationship between morphological and SSR-based distances was significant and positive (r = 0.43, p = 0.001). The high genetic diversity observed among these set of accessions, suggests ample opportunity for the development of improved varieties for different agroecologies of Ethiopia. From conservation perspective, sampling many accessions from all agroecologies would be an effective way of capturing genetic variation for future collections and conservation.  相似文献

Colonial bentgrass (Agrostis capillaris L.) is a cool-season grass, native to temperate Asia and Europe. It has good tolerance to low temperatures and partial shade and is well suited to golf course fairways and tees. Little information is available regarding levels and patterns of genetic variation among populations of colonial bentgrass, which would be useful for breeding programs. To study the genetic relationships among 27 colonial bentgrass accessions obtained from the US National Plant Germplasm System (NPGS), randomly amplified polymorphic DNA (RAPD) markers were scored and analyzed. Out of 80 primers screened, 16 were selected for further analysis, which yielded a total of 120 polymorphic bands used to differentiate the accessions. Dice's similarity coefficients for pair-wise comparisons ranged from 0.23 to 0.84 based on the RAPD data. Since there was no similarity coefficient value close to 1 between any two accessions, there was no apparent duplication among the sampled accessions. A dendrogram constructed on the basis of the Unweighted Pair Group Method with Arithmetic average (UPGMA) clustering algorithm clearly separated 26 of the accessions into three clusters with one accession distinct from the rest. The least similar pair of accessions was PI 204397 from Turkey and PI 628720 from Bulgaria, and the most similar pair was PI 509437 from Romania and PI 491264 from Finland. Clustering patterns based on principal components analysis (PCA) corresponded well with the dendrogram. A high cophenetic correlation (r = 0.82) was found between the RAPD data matrix and cophenetic matrix. The accession PI 628720, from Bulgaria, did not cluster with any other accessions.  相似文献

Tef is the most important and well adapted cereal crop in Ethiopia and grows better than other cereals both in biotic and abiotic stress conditions. In this study 93 tef accessions representative of various agro-ecologies of Ethiopia along with four improved varieties, one local check and six wild relatives were studied. The materials were genotyped with 12 simple sequence repeat markers (SSRs) to investigate the extent and structure of the genetic diversity of tef. A total of 152 alleles were detected indicating high polymorphism in the studied accessions. The number of alleles per polymorphic loci ranged from seven for CNLT 538 to 29 for CNLT150 with an average of 12.67. High average polymorphic information content (0.82) and average expected heterozygosity (0.86) also signifies the suitability and efficiency of SSR markers to discriminate among tef genotypes. Cluster, principal coordinate and structure analyses grouped the 98 tef genotypes and six wild relatives into four distinct classes. The groupings showed no special pattern despite the different regions of origins for the accessions; however, the five wild relatives were grouped together. The analysis of molecular variance and fixation index showed low genetic differentiation and high gene flow between regions revealing that much of the variability was attributed to within populations variation. Generally, the present study showed that there is high genetic variability with unstructured genetic pattern among the studied tef genotypes which implies the vitality of conserved tef germplasms at Ethiopian Biodiversity Institute and the impact of the seed exchange system on tef breeding activities in Ethiopia.

As an oilseed crop, the cultivation of Ethiopian mustard (Brassica carinata) is restricted only to Ethiopia. Even though geographic diversity is a potent source of allelic diversity, the extent of genetic diversity among germplasm material of Ethiopian mustard from different countries has not been assessed. Forty-three accessions, comprising 29 accessions from eight different geographic regions of Ethiopia and 14 exotic accessions from Australia, Pakistan, Spain, and Zambia were analysed for their genetic diversity using random amplified polymorphic DNA (RAPD) technique. A set of 50 primers yielded a total of 275 polymorphic bands allowing an unequivocal separation of every Ethiopian mustard accession. The usefulness of the 50 RAPD primers in measuring heterozygousity and distinguishing accessions was variable such that polymorphic information content (PIC) varied from 0.05 to 0.40, band informativeness (BI) from 0.05 to 0.65 and primer resolving power (RP) from 0.15 to 6.83. Jaccard's similarity coefficients ranged from 0.44 to 0.87 indicating the presence of a high level of genetic diversity. On the average, Australian and Ethiopian accessions were the most similar while, Spanish and Zambian accessions were the most distant ones. Cluster analysis grouped the 43 accessions into four groups, which has quite a high fit (r = 0.80) to the original similarity matrix. With no prior molecular information, the RAPD technique detected large genetic diversity among the 43 accessions from five different countries and their grouping by dendrogram and principal coordinate analysis (PCoA) was inclined towards geographic differentiation of RAPD markers. Conversely, RAPD differentiation along geographic origin was not apparent within the Ethiopian accessions.  相似文献

Melon (Cucumis melo L.) landraces of the Madrid provenance, Spain, have received national distinction for their high fruit quality and sensorial attributes. More specifically, a unique array of Group Inodorus landraces have been continuously cultivated and conserved by farmers in the municipality of Villaconejos since the 19th century. Their genetic relationships to other Group Inodorus and Flexuous melon market classes is not known, and, thus, a study was designed to determine their genetic relationships using 52 simple sequence repeat (SSR) markers, and then make genetic comparisons between these accessions and a previously published “Standard Reference Germplasm Array” (RA) containing Group Inodorus (14 Spanish and one USA), Flexuosus (1 Spanish), and Cantalupensis ( 2 USA) melon accessions. This subset consisted of 15 Spanish Group Inodorus landraces that circumscribed the genetic variation of major Spanish melon market classes (Groups Inodorus and Flexuosus), and USA commercial varieties (Groups Cantalupensis and Inodorus). Based on genetic distances, Villaconejos (Madrid) genotypes differed substantially from RA subset accessions, thus defining their genetic uniqueness. Principal component analysis (PCA) partitioned the accessions examined into four distinct groups revealing that Villaconejos black epidermis melons (landraces ‘Largo’, ‘Largo Negro Escrito’ and ‘Puchero’) were distinctly different from all other accessions examined, as cluster analysis separated Rochet market type Villaconejos’ accessions (landraces ‘Mochuelo’, ‘Mochuelo Tradicional’ and ‘Melón de Villaconejos’) from RA of the same market type. Genetic assessment of principal Spanish market classes revealed comparatively low intra-market heterogeneity in Piel de Sapo type accessions and high heterogeneity in Black and Yellow market type accessions. While a relatively high level of genetic introgression was detected between Yellow and Green market types, black epidermis market types were genetically unique. Given the uniqueness and high genetic diversity resident in Villaconejos landraces, this germplasm pool should be considered as a genetic source for broadening the comparatively narrow genetic base of Group Cantalupensis and Inodorus melon market types, especially standard commercial Spanish Group Inodorus market types (e.g., Piel de Sapo, Rochet, and Canari).  相似文献

The USDA Agricultural Research Service (ARS) National Small Grains Collection (NSGC) has 207 landrace barleys obtained from a nursery grown in the Ukraine in 1930 by N.I. Vavilov, many of which have multiple resistance (MR) to disease similar to accessions from Ethiopia. The original collection locations of the accessions from the Vavilov nursery are unknown. The purpose of this study was to determine, using molecular markers, if the MR accessions of unknown origin from the Vavilov nursery are genetically related to MR accessions from Ethiopia. The genetic relatedness among susceptible accessions of unknown origin and among a selection of landrace accessions from the NSGC barley core subset was also assessed. Simple sequence repeat (SSR) and Diversity Array Technology PL (DArT) marker data were used to generate similarity matrices with Dice’s similarity coefficients. Cluster analysis from these results showed that the unknown-origin MR accessions grouped with accessions from Ethiopia. Susceptible accessions of unknown origin were genetically similar to accessions from western Asia, especially near the Caucasus, and from both southern and northern Europe. Based on similarity of marker profiles, some accessions from the core collection are likely duplicates. Future work will seek to identify the probable origin of the remainder of the unknown origin accessions and to more thoroughly characterize the genetic diversity within NSGC barley core subset.  相似文献

In this study, we investigated the genetic diversity and relationships in C. olitorius by analysing populations representing different distribution areas, and developed a hypothesis on the origin and spread of the species in the pantropics. We employed amplified fragment lengths polymorphism (AFLP) and morphometric analyses in a total of 101 C. olitorius accessions. Results of both data sets are mostly congruent. The molecular analysis indicated generally low genetic diversity within populations and the Nei’s gene diversity (He) ranged from 0.0457 to 0.0955 with a mean of 0.0763. Qualitative traits, especially related to leaf morphology, branching habit and stipule color were the taxonomically most informative characters. The highest morphological variability occurred within African accessions, indicating that this species originally evolved in Africa. In both analyses, the Asian materials were nested within African populations, especially with those from North and East Africa. This indicates an African origin of the species and we hypothesise that dispersal occurred via the Mediterranean–Indian trade routes instead of natural migration along the coasts from western Africa to the Indian subcontinent. Both analyses revealed materials from Ethiopia to be quite distinct. The highest number of private bands (N?=?35), percent polymorphism (29.29%), and gene diversity (0.0955) were also detected in wild accessions collected from Ethiopia, suggesting a long-term spatial isolation of C. olitorius populations in the country. Germplasm samples from this region could therefore be a useful source of genetic variation in jute breeding programs.  相似文献

Genetic diversity was investigated in 348 accessions and subaccessions of grasspea (Lathyrus sativus L.) from 10 geographical regions. Polymorphism for 20 isozymes of 13 enzyme systems was studied to estimate the genetic diversity. The Near East and North Africa regions included the most variability for these isozyme systems, suggesting that the center of diversity (center of origin) for grasspea is in this general area. The lowest variability was found in accessions and subaccessions from South America, followed by those from Sudan–Ethiopia. Diversity was measured for individual loci over regions and EST-1 and SKDH had the highest genetic diversity. The closest genetic diversity was observed for LAP-2, followed by AAT-1 and PGM. The closest genetic distance existed between populations from the Near East and North Africa. Populations from South Asia and Sudan–Ethiopia, though geographically widely separated, exhibited a closer genetic distance from each other than from other regions.  相似文献

Summary Isozyme variation in 25 accessions of wild and cultivated Vigna unguiculata, 49 accessions of seven wild species belonging to section Vigna, and 11 accessions of V. vexillata (subgenus Plectrotropis) was scored at 17 putative loci to assess genetic relationships within and among species. The wild species selected for this study are among those which carry important agronomical traits useful in cowpea (V. unguiculata) breeding programs. Allelic frequencies were calculated and Nei's genetic distances were obtained. Low levels of intraspecific variation were observed for V. heterophylla, V. luteola and V. racemosa, whereas the other species showed a higher polymorphism. Vigna unguiculata possessed intraspecific genetic distances comparable to those previously found by other authors. Most of the isozyme variation was apportioned among species. Although V. luteola and V. marina had an interspecific genetic distance resembling the range observed at intraspecific level, all the other species showed very high interspecific distances. The present work was carried out to study genetic diversity among 17 populations of wild emmer wheat sampled from South-eastern Turkey, considered to be an important region for domestication of wheat. Eleven RFLP clones and 4 restriction enzymes combinations were used to probe the genomic DNA. A total of 151 polymorphic loci were obtained from the enzyme-probe combinations. The Genetic Distance (GD) values were from 0.019 (Gaziantep-3 and Sanliurfa-4) to 0.200 (Gaziantep-1 and K. Maras). Cluster analysis results showed that populations formed 2 clades within the dendrogram. Population Gaziantep-1 was unique and genetically most diverse from the remaining 16 populations. The results of average genetic distance (GD) among populations suggested that narrow genetic variability exist among 17 populations in the present study.  相似文献

The genetic relationships among 33 coconut germplasm accessions were analyzed using RAPD markers. The germplasm accessions were collected from various coconut growing regions viz. South Asia (SA), South East Asia (SEA), South Pacific (SP), Atantic and America, and Africa. Forty-five random primers produced a total of 399 polymorphic markers. The Polymorphism Information Content (PIC) ranged from 0.031 to 0.392 and the Marker Index (MI) ranged from 6.28 to 0.031 among the primers. Based on the MI a set of 5, 10 and 15 informative and reproducible primers were identified. The mantel matrix correlation was calculated to compare the similarity matrices of a set of reproducible informative primers and global primers. There was significant correlation among the similarity matrices (r ≥ 0.50). The similarity matrix based on 399 polymorphic markers was used to construct the dendrogram to show the genetic relationship among the accessions. Similarity values ranged between 0.573 and 0.846. There was less genetic similarity (based on Jaccard's coefficient) among South Pacific and South East Asian accessions. The clustering pattern obtained in the present study was in agreement with the earlier reports based on RFLP, SSRs and AFLPs.  相似文献

Asian cotton (Gossypium arboreum L.) was once widely cultivated in China. It has also been a valuable source of genetic variation in modern cotton improvement. In this study, the genetic diversity of selected G. arboreum accessions collected from different regions of China was evaluated by microsatellite (simple sequence repeats, SSRs) analysis. Of the 358 microsatellite markers analyzed, 74 primer pairs detected 165 polymorphic DNA fragments among 39 G. arboreum accessions examined. Twelve accessions could be fingerprinted with one or more SSR markers. With the exception of two accessions, DaZiJie and DaZiMian, genetic similarity coefficients among all accessions ranged from 0.58 to 0.87 suggesting high level of genetic variation in the G. arboreum collections. The UPGMA dendrogram constructed from genetic similarity coefficients revealed positive correlation between cluster groupings and geographic distances. In addition, comparison of the microsatellite amplification profiles of the diploid G. arboreum and tetraploid Gossypium hirsutum L. found that size distribution of amplified products in G. arboreum was dispersive and that of G. hirsutum was relatively concentrated. A study was conducted to determine the extent of genetic diversity among African cassava (Manihot esculenta Crantz) accessions resistant to the cassava mosaic virus disease (CMD), using simple sequence repeat (SSR) markers. The accessions included a breeding stock (clone 58308), five improved lines, 62 CMD resistant and 10 CMD susceptible landraces. Genetic diversity was assessed among accessions in five cluster groups derived from UPGMA analysis on data from 18 SSR primer pairs. Average gene diversity, He, was high in all cluster groups, with an average heterozygosity of 0.591 ± 0.061. The estimator of inbreeding Fis revealed a low level of inbreeding within groups and averaged −0.262 ± 0.142. Gene diversity among all accessions was 51.4% and gene diversity within cluster groups was 46.6%, while 4.8% was due to diversity between the different cluster groups. The amount of genetic differentiation measured by Gst and Fst were 9.6% and 12.1% respectively, indicating a weak genetic structure.  相似文献

One hundred and forty-four heterogeneous tef germplasm accessions collected from 10 major tef growing areas in Ethiopia were evaluated for 18 quantitative traits in a simple lattice design, at Holetta and Ginchi, in 2001 main growing season to study the genetic diversity in tef. The combined analysis of variance revealed the presence of significant variation among germplasm accessions for all traits studied. Cluster analysis revealed the overlapping of the germplasm accessions from different origin and the accessions were grouped into eight distinct clusters of 1–78. Generalized distance square confirmed the presence of significant genetic distance between clusters. In principal component (Prin) analysis, the first four principal components with latent root values above one accounted for about 80.6% of the variability existing among the germplasm accessions. Prin1 constituted about 55% of the variability mainly from almost equal contribution of 10 quantitative traits, indicating that most of the traits are equally important in tef diversity. In this study, the regions and altitudes of origin were not found to have a substantial effect on the genetic diversity in tef germplasm. Diversity within the regions was found to be significant and, hence, an opportunity for exploitation in tef improvement.  相似文献
