Cytochrome P450 is involved in drug metabolism. Subfamily CYP3A shows a degree of similarity across different animal species. However, little information is available about its expression and activity in broiler chickens. A RT‐PCR method was developed for the quantification of CYP3A37 expression in the liver and small intestine of broilers. A higher expression in the jejunum was observed compared with that in the ileum. In the liver, a significantly lower expression compared with that in the jejunum was noticed. Thus, the role of the small bowel in drug metabolism cannot be neglected in broilers. CYP3A activity was studied in vitro using midazolam as a substrate. Two protocols for the preparation of intestinal microsomes were compared. Mincing of the tissues before ultracentrifugation seemed to be more appropriate than a protocol based on ethylenediaminetetra‐acetic acid separation. CYP3A activity revealed to be the highest in the duodenum with a decreasing trend towards the ileum. Activity in liver was comparable to duodenal activity.  相似文献

为研究温和气单胞菌(A.sobria)溶血素基因的特性,本研究根据GenBank登录的气单胞菌属溶血素基因序列设计一对引物,经PCR扩增得到大小约1.5kb的A.sobria RC-07-KA株溶血基因片段,将该片段克隆到pGEM-T载体,测序结果显示:溶血素基因片段大小为1 467 bp,编码487个氨基酸残基,遗传进化分析结果表明该基因与气单胞菌属中的A.hydrophila菌株Sb (AY611033)、NLEPA-1607 (AF410466)、AEF (HM853019),A.sobria菌株357 (AY157998)、人源分离株(EF620533)和A.salmonicida菌株17-2 (X65048)的溶血素基因亲缘关系较近,同源性大于95%,而与其他菌株的同源性较低.通过构建溶血素重组表达质粒pET-HIy,诱导表达并通过western blot鉴定表达蛋白,结果显示重组菌能高效表达重组溶血素,而且纯化的重组溶血素具有溶解鲤鱼红细胞的活性.为该菌进一步深入研究奠定了基础.  相似文献

In this study the tonB2 gene was cloned from Actinobacillus pleuropneumoniae JL01 (serovar 1) and expressed as a glutathione-S-transferase (GST) fusion protein in Escherichia coli BL21(DE3). The GST fusion protein was recognized by antibodies in serum positive for A. pleuropneumoniae by Western blot analysis. Purified soluble GST-TonB2 was assessed for its ability to protect BALB/c mice against A. pleuropneumoniae infection. Mice were vaccinated with GST-TonB2 subcutaneously and challenged intraperitoneally with either ~4.0 × 10(5) colony-forming units (CFU) or ~1.0 × 10(6) CFU of A. pleuropneumoniae 4074. They were examined daily for 7 d after challenge. The survival rate of the TonB2-vaccinated mice was significant higher than that of the mice given recombinant GST or adjuvant alone. These results demonstrate that A. pleuropneumoniae TonB2 is immunogenic in mice and should be further assessed as a potential candidate for a vaccine against A. pleuropneumoniae infection. In addition, an indirect enzyme-linked immunosorbent assay (ELISA) based on the GST-TonB2 recombinant protein was developed. Compared with the ApxIVA ELISA, the TonB2 ELISA provided earlier detection of antibodies in pigs at various times after vaccination with A. pleuropneumoniae live attenuated vaccine. When compared with an indirect hemagglutination test, the sensitivity and specificity of the TonB2 ELISA were 95% and 88%, respectively. The TonB2 ELISA provides an alternative method for rapid serologic diagnosis of A. pleuropneumoniae infection through antibody screening, which would be especially useful when the infection status or serovar is unknown.  相似文献

Blood samples and ticks were collected from 48 cattle and 74 horses from seven sites in the Peten region of Guatemala. Data on body condition, mucous membrane capillary refill time and tick infestation levels were recorded for each animal in the study. Horses had significantly higher levels of tick infestation than cattle, as well as poorer body condition scores. Seroprevalence of Babesia spp. was 95.8% for B. bovis in cattle, 89.6% for B. bigemina in cattle, and 92.7% for B. equi in horses. Seroprevalence of Anaplasma marginale in cattle was 87.5%, similar to reports in animals from other regions of Central America. This is the first time that A. phagocytophilum has been reported in animals from this region, with overall PCR-prevalence of 27.6% in cattle and horses, and seroprevalence of 28.4% (52% in cattle and 13% in horses). An agent was identified with serological cross-reactivity and close genetic relatedness to Ehrlichia ruminantium, but further testing confirmed that the agent in Guatemalan cows was not the agent of heartwater. Ticks were identified to species with the predominant species identified on cattle as Boophilus microplus and Amblyomma cajennense, while Anocentor nitens and A. cajennense were most commonly found on horses. Prevalence of infection, tick infestation levels, host factors and environmental data were analyzed for association; A. nitens was significantly associated with A. phagocytophilum prevalence by village.  相似文献

An eight-month-old domestic shorthair cat was presented with chronic vomiting for three months, with an acute increase in frequency during the past two days. A diagnosis of megaoesophagus was made by chest radiography. Diagnostic work-up for megaoesophagus was performed. A gastro-oesophageal intussusception was identified during endoscopy. The importance of Anoplocephala perfoliata in horses with colic was studied in 139 horses referred for colic and 139 control horses with no signs of colic for at least three years. The serodiagnostic method of Proudman and Trees, which measures the level of A. perfoliata antibody, was used to detect A. perfoliata infection. Thirty-two horses were examined at necropsy, to determine whether the presence of A. perfoliata in the ileocaecal region was associated with the A. perfoliata antibody level. The mean A. perfoliata antibody level was significantly higher in horses with colic than in horses without colic (P < 0.001), indicating a relationship between A. perfoliata infection and colic in general. There was no relation between age and A. perfoliata antibody level. The mean A. perfoliata antibody level in 12 horses with ileocaecal disorders was significantly higher than that in control horses (P < 0.001). Of the 32 horses examined at necropsy, 7 horses with tapeworms in the ileocaecal region had a significantly higher mean A. perfoliata antibody level than the 25 horses without the parasite (P = 0.030). Lastly, examination of faeces to detect the presence of A. perfoliata infection was not useful in the present study.  相似文献

OBJECTIVE: To compare a recently developed recombinant MSP-5 competitive inhibition ELISA with a card agglutination test for detection of antibodies to Anaplasma marginale and Anaplasma centrale in Australian cattle. MATERIALS AND METHODS: The ELISA was compared with the card agglutination test using 208 sera from cattle in Anaplasma-free herds, 86 sera from cattle experimentally infected with A marginale or A centrale and 757 sera from cattle in areas endemic for A marginale. RESULTS: The specificity of the ELISA, based on testing 208 sera from cattle in Anaplasma-free areas, was 99.5%, and the sensitivities for detection of antibodies to A marginale and A centrale in sera from the experimentally infected cattle were 98.0% and 100%, respectively. For the same sets of sera, the specificity of the card agglutination test was 98.6% and the sensitivities for detection of antibodies to A marginale and A centrale were 98.0% and 100%, respectively. For the 757 sera collected from cattle in areas endemic for A marginale, the agreement between the ELISA and the card agglutination test depended on the positive threshold selected for the ELISA. The maximum achievable agreement was 91.5% (kappa = 0.73; 95% confidence interval 0.66, 0.79). CONCLUSION: We conclude that the competitive inhibition ELISA is a useful alternative to the card agglutination test for detection of A marginale or A centrale infection in cattle. The assay should be particularly useful for epidemiological applications such as prevalence studies and control programs.  相似文献

本试验采用PCR-RFLP方法检测脂滴包被蛋白基因(perilipin,PLIN)在济宁百日鸡、汶上芦花鸡等5个地方鸡种和1个培育品系中的遗传多态性,并分析了多态位点不同基因型与鸡胴体及脂肪性状的相关性。结果发现,在6个供试群体中检测到1个Pst Ⅰ酶切突变位点,测序证实为新发现的鸡PLIN基因2224 bp处G→T突变,该突变位点在供试群体中共检测到两种基因型(A1A1和A1A2),等位基因A1在所有供试群体中均表现为优势等位基因。关联分析结果表明,PLIN基因2224多态位点对鸡胴体性状和脂肪性状影响均不显著(P>0.05)。多重比较结果表明,A1A1基因型个体在胴体和脂肪性状上总体优于A1A2基因型个体,但各性状均值在两种基因型个体间差异均不显著(P>0.05)。本研究结果表明,该位点不能作为鸡胴体和脂肪性状的有效分子标记。  相似文献

A 1-year-old, intact female, domestic shorthaired cat was presented for dysuria resulting from perineal scarring subsequent to injuries incurred during a cat fight. A prepubic urethrostomy was performed to manage the dysuria. Eleven months later, the cat was re-presented with a 3-day history of pyrexia and inappetence. A pinpoint opening extending 0.5 cm ventral to the anus on midline, exuding a clear discharge, was noted in the perineal region. A contrast fistulogram was performed, and a vaginoperineal fistula was diagnosed. The fistulous tract was a result of vulvar stricture from the trauma of the cat fight. A vaginoplasty was performed to create an opening for vaginal secretions. This is the first published report of a prepubic urethrostomy performed in a female cat.  相似文献

Effect of different genomic relationship matrices on accuracy and scale   总被引:1,自引:0,他引:1  
Phenotypic data on BW and breast meat area were available on up to 287,614 broilers. A total of 4,113 birds were genotyped for 57,636 SNP. Data were analyzed by a single-step genomic BLUP (ssGBLUP), which accounts for all phenotypic, pedigree, and genomic information. The genomic relationship matrix (G) in ssGBLUP was constructed using either equal (0.5; GEq) or current (GC) allele frequencies, and with all SNP or with SNP with minor allele frequencies (MAF) below multiple thresholds (0.1, 0.2, 0.3, and 0.4) ignored. Additionally, a pedigree-based relationship matrix for genotyped birds (A(22)) was available. The matrices and their inverses were compared with regard to average diagonal (AvgD) and off-diagonal (AvgOff) elements. In A(22), AvgD was 1.004 and AvgOff was 0.014. In GEq, both averages decreased with the increasing thresholds for MAF, with AvgD decreasing from 1.373 to 1.020 and AvgOff decreasing from 0.722 to 0.025. In GC, AvgD was approximately 1.01 and AvgOff was 0 for all MAF. For inverses of the relationship matrices, all AvgOff were close to 0; AvgD was 2.375 in A(22), varied from 11.563 to 12.943 for GEq, and increased from 8.675 to 12.859 for GC as the threshold for MAF increased. Predictive ability with all GEq and GC was similar except that at MAF = 0.4, they declined by 0.01 for BW and improved by 0.01 for breast meat area. Compared with BLUP, EBV in the ssGBLUP were, on average, increased by up to 1 additive SD greater with GEq and decreased by 2 additive SD less with GC. Genotyped animals were biased upward with GEq and downward with GC. The biases and differences in EBV could be controlled by adding a constant to GC; they were eliminated with a constant of 0.014, which corresponds to AvgOff in A(22). Unbiased evaluation in the ssGBLUP may be obtained with GC scaled to be compatible with A(22). The reduction of SNP with small MAF has a small effect on the real accuracy, but it may falsely increase the estimated accuracies by inversion.  相似文献

本研究旨在阐明Toll样受体6(TLR6)基因的多态性与荷斯坦母牛乳房炎抗性之间的关系.采用PCR-SSCP和PCR-RFLP技术检测208头荷斯坦母牛TLR6基因多态性,用最小二乘法分析该基因多态性与荷斯坦母牛钵细胞评分(somatic cell score,SCS)的关系.结果发现,TLR6基因mRNA序列中存在T853A、G855A、G1793A、C1859A、G1934A和A1980G 6个多态位点,其中T853A突变使编码氨基酸由缬氨酸变为谷氨酸,G855A突变使天冬氨酸变为天冬酰胺,A1980G突变使异亮氨酸变为缬氨酸.T853A和G855A突变位点经SSCP检测发现3种基因型AA、AB和BB,其频率分别为0.308、0.490和0.202;X~2检验表明荷斯坦母牛在该位点处于Hardy-Weinberg平衡;BB基因型SCS最小二乘均值极显著低于AB和AA基因型所对应的值(P<0.01);AB基因型SCS最小二乘均值极显著低于AA基因型所对应的值(P<0.01).G1793A突变位点经BsiY Ⅰ酶切产生2种基因型CC和CD,其频率分别为0.332和0.668;荷斯坦母牛在该位点偏离了Hardy-Weinberg平衡,CC和CD基因型SCS最小二乘均值差异不显著(P>0.05).C1859A突变位点经BstX Ⅰ酶切产生2种基因型EE和EF,其频率分别为0.178和0.822;荷斯坦母牛在该位点偏离了Hardy-Weinberg平衡;EE和EF基因型SCS最小二乘均值差异不显著(P>0.05).G1934A突变位点经BsiY Ⅰ酶切产生2种基因型GG和GH,其频率分别为0.356和0.644;荷斯坦母牛在该位点偏离了Hardy-Weinberg平衡;GG和GH基因型SCS最小二乘均值差异不显著(P>0.05).A1980G突变位点经Bcl Ⅰ酶切产生3种基因型II、IJ和JJ,其频率分别为0.260、0.567和0.173,荷斯坦母牛在该位点处于Hardy-Weinberg平衡;JJ基因型SCS最小二乘均值极显著低于IJ和II基因型所对应的值(P<0.01);IJ基因型SCS最小二乘均值极显著低于II基因型所对应的值(P<0.01).对于奶牛乳房炎抗性,BB和JJ是有利基因型,AA和II是不利基因型.本研究结果初步表明TLR6基因T853A和G855A突变位点的B等位基因以及A1980G突变位点的J等位基因是提高奶牛乳房炎抗性的2个潜在的DNA标记.  相似文献

A reverse line blot hybridization (RLB) one-stage nested PCR (nPCR) for Anaplasma centrale and a nested PCR for Anaplasma marginale were used to detect infected cattle grazing within an endemic region in Israel. A novel set of PCR primers and oligonucleotide probes based on a 16S ribosomal RNA gene was designed for RLB detection of both Anaplasma species, and the performance of the molecular assays compared. The immunofluorescent antibody test (IFA) was used to detect antibodies to both Anaplasma species, whereas, a highly sensitive and specific competitive enzyme-linked immunosorbent assay (cELISA) was used to detect antibodies in A. centrale-vaccinated cattle. The RLB and the nested PCR procedures showed bacteremia with sensitivity of 50 infected erythrocytes per milliliter. Up to 93% of the A. centrale vaccinates carried specific antibodies that were detected by cELISA, and up to 71% of the vaccinated cattle were found to be naturally infected with A. marginale according to the PCR and the RLB assays. Nevertheless, no severe outbreaks of A. marginale infection occurred among vaccinated herds in this endemic region. It appears that both, molecular tools and serology are useful for evaluation of the vaccine efficacy. In the light of wide natural field infection with A. marginale, strong recommendations to continue the A. centrale vaccination program regime will continue until a new generation of non-blood-based vaccine will be developed.  相似文献

OBJECTIVE: To determine whether sequelae of infection differed among single versus double infection with Anaplasma phagocytophilum or Anaplasma marginale, with and without tick salivary extract, in cattle. ANIMALS: Eighteen 13-month old steers. PROCEDURES: Treatment groups of 3 cattle each included A marginale inoculated ID followed on day 35 by A phagocytophilum without tick saliva, A phagocytophilum followed on day 10 by A marginale without tick saliva, A marginale followed on day 35 by A phagocytophilum with tick saliva, A phagocytophilum followed on day 10 by A marginale with tick saliva, tissue culture control injection, and tick saliva control injection. Infection was monitored via clinical observations, CBC, serologic testing, and PCR analysis of blood and tissues. RESULTS: Infected cattle had significantly reduced weight gain. Anemia occurred 25 to 32 days after A marginale infection, which was attenuated by tick saliva. Parasitism was greater if cattle had not previously been inoculated with A phagocytophilum. Nine of the 12 treated cattle had positive results of PCR analysis for A phagocytophilum from at least 1 blood sample. Five tissue samples had positive results of PCR analysis for A phagocytophilum; PCR results for A marginale were positive in spleen, lung, lymph node, heart, and ear skin of infected cattle. CONCLUSIONS AND CLINICAL RELEVANCE: Results indicated an important biological interaction between A marginale and A phagocytophilum infection as well as with tick saliva in disease kinetics and severity in cattle, which may be important for interpretation of diagnostic tests and management of disease in areas where both pathogens occur.  相似文献

Bovine coronavirus (BCoV) and group A bovine rotavirus (BRV) are two of major causes for neonatal calf diarrhea. In the present study, a one-step duplex RT-PCR was established to detect and differentiate BCoV and group A BRV from fecal samples. The sensitivity of this method for BCoV and group A BRV was 10 PFU/100 μl and 1 PFU/100 μl, respectively. Twenty-eight diarrhea fecal samples were detected with this method, the result showed that 2 samples were identified as co-infected with BCoV and group A BRV, 26 samples were group A BRV positive, and 2 samples were negative. It proved that this method is sensitive for clinical fecal samples and is worth applying to laboratory diagnosis for BCoV and group A BRV.  相似文献

Abstract

This study investigated the effects of vitamin E and selenium (Se) injections on concentrations of vitamin E, vitamin A, and Se in blood and milk to predict when injections may improve ewes’ performance. Awassi ewes received 0 (control, 13 ewes) or 15 plus 0.05?mg/kg BW of vitamin E and Se (injected, 15 ewes) at 1 and 4 weeks postpartum. Blood and milk samples were collected weekly for 6 weeks. Injections increased vitamin E, decreased Se, and had no effects on vitamin A levels in blood. Blood vitamin E was correlated positively with vitamin A. Injections increased vitamin E, but had no effects on vitamin A or Se levels in milk. Milk vitamin A was positively correlated with vitamin E and Se levels in the control group. Milk vitamins E and A were positively correlated with their corresponding levels in blood. Milk somatic cell count was negatively correlated with blood vitamin A levels.  相似文献

The aim of this study was to examine interactions between Ascaris suum and Oesophagostomum dentatum infections in pigs with regard to population dynamics of the worms such as recovery, location and length; and host reactions such as weight gain, pathological changes in the liver and immune response. Seventy-two helminth-na?ve pigs were allocated into four groups. Group A was inoculated twice weekly with 10000 O. dentatum larvae for 8 weeks and subsequently challenge-infected with 1000 A. suum eggs, while Group B was infected with only 1000 A. suum eggs; Group C was inoculated twice weekly with 500 A. suum eggs for 8 weeks and subsequently challenge-infected with 5000 O. dentatum larvae, whereas Group D was given only 5000 O. dentatum larvae. All trickle infections continued until slaughter. Twelve pigs from Group A and B were slaughtered 10 days post challenge infection (p.c.i.) and the remaining 12 pigs from the each of the four groups were slaughtered 28 days p.c.i.. No clinical signs of parasitism were observed. The total worm burdens and the distributions of the challenge infection species were not influenced by previous primary trickle-infections with the heterologous species. Until day 10 p.c.i. the ELISA response between A. suum antigen and sera from the O. dentatum trickle infected pigs (Group A) pigs were significantly higher compared to the uninfected Group B. This was correlated with a significantly higher number of white spots on the liver surface both on Day 10 and 28 p.c.i. in Group A compared to Group B. The mean length of the adult O. dentatum worms was significantly reduced in the A. suum trickle infected group compared to the control group. These results indicate low level of interaction between the two parasite species investigated.  相似文献

A feedlot trial was conducted to determine the effect of dietary vitamin A concentration and roasted soybean (SB) inclusion on carcass characteristics, adipose tissue cellularity, and muscle fatty acid composition. Angus-crossbred steers (n = 168; 295 +/- 1.8 kg) were allotted to 24 pens (7 steers each). Four treatments, in a 2 x 2 factorial arrangement, were investigated: no supplemental vitamin A, no roasted soybeans (NANS); no vitamin A, roasted SB (20% of the diet on a DM basis; NASB); with supplemental (2,700 IU/kg) vitamin A, no roasted SB (WANS); and with supplemental vitamin A, roasted SB (WASB). Diets included high moisture corn, 5% corn silage, 10 to 20% supplement, and 20% roasted SB in the SB treatments on a DM basis. The calculated vitamin A concentration in the basal diet was < 1,300 IU/kg of DM. Blood samples (2 steers/pen) were collected for serum vitamin A determination. Steers were slaughtered after 168 d on feed. Carcass characteristics and LM composition were determined. Fatty acid composition of LM was analyzed, and adipose cellularity in the i.m. and s.c. depots was determined. No vitamin A x SB interactions were detected (P > 0.10) for cattle performance, carcass composition, or muscle fatty acid composition. Low vitamin A diets (NA) did not affect (P > 0.05) ADG, DMI, or G:F. Quality grade tended (P = 0.07) to be greater in NA steers. Marbling scores and the percentage of carcasses grading > or = Choice(-) were 10% greater for NA steers, although these trends were not significant (P = 0.11 and 0.13, respectively). Backfat thickness and yield grade were not affected (P > 0.26) by vitamin A supplementation. Composition of the LM was not affected (P > 0.15) by vitamin A or SB supplementation. Serum retinol at slaughter was 44% lower (P < 0.01) for steers fed NA than for steers supplemented with vitamin A (23.0 vs. 41.1 microg/dL). A vitamin A x SB interaction occurred (P < 0.05) for adipose cellularity in the i.m. depot; when no SB was fed, vitamin A supplementation decreased cell density and increased cell size. However, when SB was fed, vitamin A supplementation did not affect adipose cellularity. Adipose cellularity at the s.c. depot was not affected (P > 0.18) by vitamin A or SB treatments. Fatty acid profile of the LM was not affected by vitamin A (P > 0.05), but SB increased (P < 0.05) PUFA (7.88 vs. 4.30 g/100 g). It was concluded that feeding NA tended to increase marbling without affecting back-fat and yield grade. It appeared that NA induced hyperplasia in the i.m. but not in the s.c. fat depot.  相似文献

Anaplasma marginale and Anaplasma centrale are rickettsial pathogens responsible for acute disease and mild infections, respectively, in cattle herds. A duplex real-time polymerase chain reaction (PCR) assay with probes labeled with different fluorophores was developed for simultaneous detection and quantification of A. marginale and A. centrale DNA in bovine blood samples. The assay was able to detect as few as 10(1) and 10(2) DNA copies for A. marginale and A. centrale, respectively, with optimal specificity and reproducibility. Analysis by real-time and nested PCR carried out on 54 samples previously tested by reverse line blot hybridization showed that the established duplex real-time PCR assay can detect and quantify the 2 Anaplasma spp., even if present simultaneously in the same blood samples. Such an assay could be used in pathogenesis studies on bovine acute anaplasmosis.  相似文献

Distribution of Pasteurella haemolytica in the respiratory tracts of calves with no apparent clinical signs of illness and those infected experimentally with Dictyocaulus viviparus was determined so as to define carrier sites for this organism. The calves had been positive by nasopharyngeal swab for either P haemolytica A2 or A1 for at least two months or for over a month, respectively, before slaughter. P haemolytica A1 was acquired following horizontal spread from other infected calves. It was observed post mortem that P haemolytica A1 or A2 resided in the tonsils and retropharyngeal lymph nodes of calves of both groups. In addition to these sites, P haemolytica A1 was also isolated from the right cranial lung lobe of one of the calves from the D viviparus infected group although there was no evidence of pasteurella associated pneumonia. It was concluded that tonsil and retropharyngeal lymph nodes appear to be the most important carrier sites for P haemolytica when compared to other tissues of the bovine respiratory tract.  相似文献

Spontaneous, histamine-induced and Concanavalin A (Con A)-induced suppression of Con A mitogenesis of autologous responder cells was studied in normal dogs and in dogs with atopic dermatitis. Histamine-induced suppression was significantly decreased in the atopic dogs, as was the Con A-induced suppression, at supraoptimal concentration of Con A, to a lesser extent. Total numbers of histamine type 1 or type 2 receptors were not different for cells from atopic or normal dogs. The spontaneous suppression was significantly greater for the atopic dogs and this was not accounted for by the effect of non-specific dermatitis, increased macrophage-induced suppression or increased induction by mitogenic factors in the culture medium. Some possible mechanisms for these results are discussed, and the similarities to suppressor cell function in humans with atopic disease are noted.  相似文献
