口蹄疫是一种严重威胁畜牧业发展的重要传染病 ,目前世界上许多国家和地区都有该病的流行与发生。开展新型口蹄疫疫苗的研究一直是口蹄疫防制技术的一项重要内容。鉴于传统疫苗所存在的诸多缺点 ,人们先后研制开发了口蹄疫的第二代基因工程苗和第三代基因疫苗 ,这些新型疫苗都具有一定的优越性 ,显示出进一步发展的潜力。文章着重讨论了在感染性 c DNA的基础上所构建的一类新型基因疫苗—感染性克隆疫苗及其发展前景。感染性克隆疫苗不仅继承了以往基因疫苗的许多优点 ,而且克服了其表达量低 ,免疫效果不理想的缺点 ,为新型疫苗的研制提供了一种新的思路 相似文献

口蹄疫是由口蹄疫病毒(FMDV)感染所引起的偶蹄动物共患的急性、热性、接触性传染病。其控制措施主要是疫苗免疫,在所有易感动物中使用灭活病毒作为免疫原构成了控制及消除本病的基础。传统疫苗在该病的防控中起了重要的作用。基因工程疫苗以安全性高、生产成本低、可制备同一病毒多个成分或多价病毒疫苗等优点,成为FMD疫苗研究的焦点。自20世纪80年代开始,包括基因工程亚单位疫苗、合成肽疫苗、蛋白质载体疫苗、基因缺失疫苗、活载体疫苗、核酸疫苗等研究日趋活跃。本文将口蹄疫传统疫苗与几种新型疫苗进行比较,从研究概况、生产技术要点、疫苗特点及需要改进的问题几方面阐述了新型口蹄疫疫苗的研究进展。 相似文献

近年来,O型口蹄疫病毒已演化出多种谱系.目前应用的疫苗已不能有效保护多谱系口蹄疫的流行,这给我国猪口蹄疫的防控带来了极大的困难.为了进一步发展免疫原性好、抗原谱广的猪O型口蹄疫疫苗候选株,本研究以O/HN/93现用疫苗毒株的感染性克隆为骨架,构建了含VP3(58位)和VP1(43、48、137、139、140、141和142位)氨基酸改造的全长克隆.线性化的全长cDNA和T7RNA聚合酶质粒共转染BHK细胞后获得拯救病毒.RT-PCR和乳鼠致病性试验表明拯救病毒遗传稳定,具有与亲本病毒相似的致病性.用拯救的基因工程病毒灭活疫苗免疫猪28 d后分别用中国谱系、泛亚谱系和缅甸98谱系猪源毒的流行毒攻击,结果均获得了完全保护(16/16).O/HN/93灭活疫苗免疫猪能完全保护泛亚谱系和缅甸98谱系猪源病毒的攻击(16/16),但不能完全保护(12/16)中国谱系猪源病毒的攻击.结果表明基因工程病毒制备的灭活疫苗提高了对中国型猪源谱系病毒的免疫保护,拓展了抗原谱,是具有良好开发前景的疫苗候选株. 相似文献

Changes to foot-and-mouth disease (FMD) control policies since 2001 mean that emergency vaccination must be considered more readily as a control measure in the future. Since field application of vaccine for emergency use has only rarely been applied, the effectiveness of single dose administration, as a control measure in an outbreak situation, is poorly understood. In this review we consider all the available experimental data from studies utilizing either experimental or readily available, commercially produced vaccines, in order to assess their likely effectiveness as an additional means of controlling FMD transmission and spread in an emergency. Overall it is concluded that such vaccines offer an additional and valuable means of FMD control for both ruminants and pigs. They are able to reduce clinical disease, sub-clinical infection and excretion and onward transmission of virus. However, to be most effective, vaccination should be rapidly applied to give maximum opportunity for immunity to develop. We also identify areas for future research and emphasize the importance of vaccine efficacy studies in providing data for models that can help to predict the efficacy of differing FMD control strategies. 相似文献

Presence of antibodies to non-structural proteins of foot-and-mouth disease virus in repeatedly vaccinated cattle 总被引:1,自引:0,他引:1
For the purpose of removing infected animals by detecting humoral immune responses to non-structural proteins of the foot-and-mouth disease (FMD) virus, antibodies induced by contaminated residual non-structural proteins contained in less pure FMD vaccine can be problematic for serological screening. The aim of the present study was to measure the possible presence of antibodies against these non-structural proteins in repeatedly vaccinated calves and beef cattle. Five imported FMD vaccines were examined using two commercial ELISA kits, UBI FMDV NS EIA and Ceditest FMDV-NS, for serological testing. After five doses of vaccination, the serum of one calf tested positive, and two vaccines induced a significant increase in anti-3ABC antibodies in calves. This finding demonstrated that a positive reaction to non-structural proteins due to impurities in the FMD vaccine was detectable using commercial tests. A low percentage of field sera sampled from beef cattle in Kinmen also tested positive, but the key factor resulting in the positive reactions could not be positively identified based on our data. 相似文献

Young calves were simultaneously vaccinated by subcutaneous route against foot-and-mouth disease (FMD) and against infectious bovine rhinotracheitis/adenovirus/parainfluenza-3 (IBR/Adeno/PI-3) by intranasal route. The serological response against the 3 FMD virus types of the FMD vaccine was clearly positive. There was no significant difference between results of simultaneous FMD and IBR/Adeno/PI-3 vaccination and FMD vaccination only. 相似文献

为探索生猪重大疫病的科学免疫方法,确定最佳免疫程序,笔者设计并开展了口蹄疫(FMD)合成肽苗(或灭活苗)、猪瘟(HC)脾淋苗、高致病性蓝耳病弱毒疫苗(或灭活苗)的不同组合注射、单独注射、不同疫苗类型注射的试验研究.结果表明:3种疫苗不同组合注射或单独注射,以及使用不同类型疫苗注射,其免疫副反应均无明显差异;平均免疫抗体效价及群体合格率各组间存有差异;采用猪口蹄疫、猪瘟、猪蓝耳病3种疫苗同时分点注射,以及仔猪首免口蹄疫、猪蓝耳苗,再免疫口蹄疫、猪瘟苗的免疫效果最佳. 相似文献

In the course of vaccine controls, the potency of 25 foot-and-mouth disease (FMD) vaccines was tested quantitatively in parallel in cattle using the intradermal infection and the determination of the SN titres. More than 95% of the vaccinated cattle with SN titres of greater than 1:20 were protected from generalized FMD, regardless of the virus type tested. 61.5% of the vaccinated cattle with SN titres less than or equal to 1:20 were not protected and developed generalized FMD. Comparison of the PD50 values calculated from the results of the intradermal infection and the corresponding SN titres (minimum protection titre greater than 1:20) showed that the results were in complete agreement in 56% of the tested vaccines. In a further 32% of vaccines, the PD50 calculated from the SN titre was slightly below that for the intradermal infection, in the remaining 12% it was somewhat above. The possibility of using the minimum titre determination for testing a vaccine and the significance of this titre as an expression of protection by vaccination are discussed. 相似文献
