Two experiments were conducted to determine 1) the effect of acute feed deprivation on leptin secretion and 2) if the effect of metabolic fuel restriction on LH and GH secretion is associated with changes in serum leptin concentrations. Experiment (EXP) I, seven crossbred prepuberal gilts, 66 +/- 1 kg body weight (BW) and 130 d of age were used. All pigs were fed ad libitum. On the day of the EXP, feed was removed from four of the pigs at 0800 (time = 0) and pigs remained without feed for 28 hr. Blood samples were collected every 10 min from zero to 4 hr = Period (P) 1, 12 to 16 hr = P 2, and 24 to 28 hr = P 3 after feed removal. At hr 28 fasted animals were presented with feed and blood samples collected for an additional 2 hr = P 4. EXP II, gilts, averaging 140 d of age (n = 15) and which had been ovariectomized, were individually penned in an environmentally controlled building and exposed to a constant ambient temperature of 22 C and 12:12 hr light: dark photoperiod. Pigs were fed daily at 0700 hr. Gilts were randomly assigned to the following treatments: saline (S, n = 7), 100 (n = 4), or 300 (n = 4) mg/kg BW of 2-deoxy-D-glucose (2DG), a competitive inhibitor of glycolysis, in saline iv. Blood samples were collected every 15 min for 2 hr before and 5 hr after treatment. Blood samples from EXP I and II were assayed for LH, GH and leptin by RIA. Selected samples were quantified for glucose, insulin and free fatty acids (FFA). In EXP I, fasting reduced (P < 0.04) leptin pulse frequency by P 3. Plasma glucose concentrations were reduced (P < 0.02) throughout the fast compared to fed animals, where as serum insulin concentrations did not decrease (P < 0.02) until P 3. Serum FFA concentrations increased (P < 0.02) by P 2 and remained elevated. Subcutaneous back fat thickness was similar among pigs. Serum IGF-I concentration decreased (P < 0.01) by P 2 in fasted animals compared to fed animals and remained lower through periods 3 and 4. Serum LH and GH concentrations were not effected by fast. Realimentation resulted in a marked increase in serum glucose (P < 0.02), insulin (P < 0.02), serum GH (P < 0.01) concentrations and leptin pulse frequency (P < 0.01). EXP II treatment did not alter serum insulin levels but increased (P < 0.01) plasma glucose concentrations in the 300 mg 2DG group. Serum leptin concentrations were 4.0 +/- 0.1, 2.8 +/- 0.2, and 4.9 +/- 0.2 ng/ml for S, 100 and 300 mg 2DG pigs respectively, prior to treatment and remained unchanged following treatment. Serum IGF-I concentrations were not effected by treatment. The 300 mg dose of 2DG increased (P < 0.0001) mean GH concentrations (2.0 +/- 0.2 ng/ml) compared to S (0.8 +/- 0.2 ng/ml) and 100 mg 2DG (0.7 +/- 0.2 ng/ml). Frequency and amplitude of GH pulses were unaffected. However, number of LH pulses/5 hr were decreased (P < 0.01) by the 300 mg dose of 2DG (1.8 +/- 0.5) compared to S (4.0 +/- 0.4) and the 100 mg dose of 2DG (4.5 +/- 0.5). Mean serum LH concentrations and amplitude of LH pulses were unaffected. These results suggest that acute effects of energy deprivation on LH and GH secretion are independent of changes in serum leptin concentrations.  相似文献

Crossbred gilts were ovariectomized (OVX) at 120, 150, 180 and 210 d of age to determine whether various characteristics of luteinizing hormone (LH) concentrations are influenced by age and reproductive state (prepuberal vs postpuberal). All 120-d-old gilts were prepuberal and all 210-d-old animals were postpuberal, whereas gilts 150 and 180 d old included both prepuberal and postpuberal animals. Blood was collected at 15-min intervals for 2 h, 2 d before OVX (d -2), and 2 (d +2), 8 (d +8) and 14 (d +14) d after OVX. Mean LH concentrations for prepuberal gilts were similar among age groups (P greater than .05) on d -2 and +2; however, LH increased (P greater than .05) from d -2 to +2. No change in LH secretion was found in postpuberal gilts during these two periods. After OVX, LH increased from d +2 to +14 in both prepuberal and postpuberal gilts in all age groups. In postpuberal gilts, LH increased linearly (P less than .05) between d +2 and +14; rate of increase accelerated with advancing age (P less than .01). In prepuberal gilts, LH increased in a nonlinear manner, but it did not increase between d +2 and +8. The increase observed in prepuberal and postpuberal gilts after OVX resulted primarily from an increase in magnitude of peak concentrations of LH. The purpose of this research was to determine whether serum concentrations of steroids, IGF-I, and relative amounts of serum IGF-binding proteins (IGFBP) differ in growing boars (n = 11), barrows (n = 11), and gilts (n = 12) from 70 to 140 d of age. Pigs of similar age and weight were housed in pens of three or four and given ad libitum access to a 17% CP corn-soy diet and water. Pigs were weighed and blood samples were collected every 14 d beginning at 70 d of age. Serum concentrations of IGF-I and steroids were determined by RIA and relative amounts of IGFBP were determined by ligand blot analysis. By 84 d of age and continuing through 140 d of age, mean serum concentrations of IGF-I were greater (P < .05) in boars than in barrows or gilts. Relative amounts of 46-kDa IGFBP3 and 28-kDa IGFBP-4 were similar (P > .05) among pigs at 70 d of age; however, boars and barrows had greater (P < .05) relative amounts of 24-kDa IGFBP-4 and 41-kDa IGFBP-3 than gilts. Relative amounts of IGFBP-2 were greater (P < .01) in barrows than in gilts or boars at 70 d of age. From 84 d of age through 140 d of age, relative amounts of both forms of IGFBP-3 and the 28-kDa IGFBP-4 were greater (P < .05) in boars than in gilts or barrows. Relative amounts of IGFBP2 were greater (P < .05) in barrows than in gilts or boars at 98 d of age, but by 140 d of age relative amounts were greater (P < .05) in boars and barrows than in gilts. Mean serum concentrations of estradiol-17beta were similar (P > .05) between gilts and boars at 70 d of age, but by 98 d of age, and continuing through 140 d of age, mean serum concentrations of estradiol-17beta were greater (P < .05) in boars than in gilts. Mean serum concentrations of testosterone in boars increased (P < .05) with increasing age and were greatest at 128 and 140 d of age. Serum concentrations of testosterone were negatively correlated (P < .01) with relative amounts of serum IGFBP-2 but positively correlated (P < .01) with serum concentrations of IGF-I and estradiol-17beta. Serum concentrations of estradiol-17beta were positively correlated (P < .01) with serum concentrations of IGF-I in boars. Changes in serum concentrations of IGF-I and relative amounts of IGFBP resulting from changes in serum concentrations of estradiol-17beta and testosterone may contribute to growth differences observed among sexes.  相似文献

Three experiments (EXP) were conducted to test the hypothesis that leptin modulates LH, GnRH, and neuropeptide Y (NPY) secretion. In EXP I, prepuberal gilts received intracerebroventricular (i.c.v.) leptin injections and blood samples were collected. In EXP II, anterior pituitary cells from prepuberal gilts in primary culture were challenged with 10(-14), 10(-13), 10(-12), 10(-11), 10(-10), 10(-9), 10(-8), 10(-7), or 10(-6) M leptin individually or in combinations with 10(-10), 10(-9), and 10(-8) M GnRH. In EXP III, hypothalamic-preoptic area (HYP-POA) explants were placed in perfusion system and exposed to 0 (n=5), 10(-12) M (n=4), 10(-10) M (n=4), 10(-8) M (n=4), or 10(-6) M (n=5) human recombinant leptin (LEP) for 30 min. In EXP I, serum LH concentrations were unaffected by leptin treatment. In EXP II, all doses of leptin increased LH secretion except for 10(-12) and 10(-7) M. Only 10(-7), or 10(-13) M leptin in combination with 10(-8) or 10(-9) M GnRH, respectively, suppressed LH secretion. In EXP III, prior to leptin, media GnRH concentrations were similar across treatments. Media GnRH concentrations increased after 10(-12), 10(-10), and 10(-8) M leptin compared to control. Leptin treatment failed to influence NPY secretion across treatments. These results indicate that components of the neuroendocrine axis that regulate GnRH and LH secretion are functional and leptin sensitive before the onset of puberty. Other neural peptides in addition to NPY may mediate the acute effects of leptin on the GnRH-LH system and lastly, the inability of i.c.v. leptin treatment to increase LH secretion may in part be related to stage of sexual maturation and associated change in negative feedback action of estradiol on LH secretion.  相似文献

Two experiments were conducted to determine whether 1) administration of estradiol-173 (E2) implants to barrows elevates serum concentrations of E2 to levels similar to those of adult boars and subsequently affects the anterior pituitary gland IGF system and 2) administration of E2 to barrows increases serum concentrations of E2, serum and anterior pituitary concentrations of IGF-I, and relative amounts of serum and anterior pituitary IGF-binding proteins (IGFBP), vs boars and unimplanted barrows. In Exp. 1, 20 crossbred barrows (150 +/- 6 d, 103 +/- 8 kg) were administered varying number of E2 implants (0, 2, 3, 4; n = 5/group) on d 1. Blood samples were collected weekly by jugular venipuncture, beginning on d 1. Pigs were killed on d 36 when a blood sample and anterior pituitary were collected. Serum concentrations of E2 were increased (P < 0.05) in pigs with 2,3, and 4 implants vs 0 implants, but no difference (P > 0.05) was detected in serum concentrations of E2 among pigs with 2, 3, and 4 implants. Orthogonal contrasts identified that three or four E2 implants were necessary to increase serum concentrations of E2 to that similar to boars. Serum and anterior pituitary concentrations of IGF-I were increased (P < 0.05) in pigs with 2, 3, and 4 implants vs 0 implants. Relative amounts of anterior pituitary IGFBP-2 and - 5 increased (P < 0.05) in response to administration of E2. In Exp. 2, three treatment groups were randomly allotted by litter; boars (n = 11), E2-implanted barrows (n = 9), and unimplanted barrows (n = 12). A blood sample was taken from all pigs on d 1 and every 14 d thereafter. Implanted pigs received four implants on d 1. Pigs were killed on d 91, when a blood sample and anterior pituitary were collected. Mean serum concentrations of E2 were greater (P < 0.05) in implanted pigs vs boars. Mean serum concentrations of IGF-I (ng/mL) were greater (P < 0.05) in boars (238.7 +/- 6.8) than in implanted barrows (170.2 +/- 8.9) and unimplanted (150.4 +/- 6.7) pigs and tended to be greater (P = 0.08) in implanted vs unimplanted pigs. Mean anterior pituitary concentrations of IGF-I (ng/mg tissue) were greater (P < 0.05) in implanted (773.6 +/- 57.0) pigs than boars (251.9 +/- 51.6) and unimplanted (185.6 +/- 49.4) pigs. Relative amounts of serum IGFBP-2 were greater (P < 0.05) in implanted pigs vs boars. Relative amounts of anterior pituitary IGFBP-2 and -5 were greater (P < 0.05) in boars than in implanted and unimplanted pigs. These data suggest that E2 may influence components of the porcine IGF system in the serum and anterior pituitary. Other gonadal factors present in boars may additionally affect the serum and anterior pituitary IGF system.  相似文献

The objective of this study was to determine the effects of castration on short-term growth performance, hormone profiles, and behavior in pigs at 3, 6, 9, or 12 d of age. Ninety intact male pigs were assigned randomly to a treatment age by litter [3, 6, 9, or 12 d of age; n = 9 to 13 pigs per treatment (age) group]. Pigs within a single litter were then assigned to noncastrated (NC) or castrated (CAS) treatment groups according to BW. Pigs were nonsurgically fitted with jugular catheters, and blood samples were drawn immediately before castration (0 h) and at 0.5, 1, 1.5, 2, 24, and 48 h after castration. Body weights were obtained when pigs were catheterized and again at 24 and 48 h after castration. Serum samples were analyzed for cortisol, porcine corticosteroid-binding globulin, and dehydroepiandrosterone sulfate (DHEA-S). No differences were detected in initial BW of pigs, and there was no overall treatment effect on growth performance of pigs at 24 or 48 h posttreatment. A time x treatment interaction was detected (P < 0.01) for serum cortisol concentrations, such that cortisol was greater in CAS pigs than in NC pigs. No overall effect of age at castration was observed on cortisol concentrations. At 24 h after castration, serum cortisol concentrations returned to baseline in all treatment groups; however, at 48 h after castration, overall cortisol concentrations were elevated (P < 0.01) in the 6-, 9-, and 12-d-old pigs in both the CAS and NC groups compared with baseline concentrations. Total cortisol and porcine corticosteroid-binding globulin were used to calculate the free cortisol index (FCI). A time x treatment interaction was observed (P < 0.01) for FCI, such that FCI was greater in CAS males than in NC males. The FCI was also affected by age (P < 0.01). There was a time x treatment x age interaction (P < 0.01) for serum DHEA-S, such that DHEA-S concentrations decreased in CAS animals but increased in NC animals, and DHEA-S concentrations increased with age. During the first 2 h after castration, there was an overall age effect (P = 0.01) on the time that pigs spent standing, such that 3-d-old pigs stood more than 6-, 9-, or 12-d-old pigs. Treatment did not influence the time that pigs spent nursing, lying, standing, or sitting, although there was a trend (P = 0.08) for CAS pigs to be less active than NC pigs. These data indicate that castration is stressful regardless of age; however, the stress associated with handling seems to increase as pigs age.  相似文献

This study investigated the responsiveness of the pituitary-ovarian axis of prepubertal gilts to hourly injections (i.v.) with GnRH. Six gilts each at 70, 100, 150, and 190 d of age were assigned either to treatment with GnRH or saline. Treatments were given until gilts showed estrus or for 7 d, whichever came first. Hourly pulsing with GnRH resulted in gradually increasing concentrations of estradiol-17 beta (E2), a preovulatory surge of LH, and subsequently increased progesterone (P4) concentrations. The increase in serum P4 was preceded by ovulation and corpora lutea (CL) formation in two gilts 70 d of age and all older gilts. The interval (h) from start of GnRH treatment to peak E2 (88 +/- 3), peak LH (103 +/- 3), and concentrations of P4 greater than or equal to 1 ng/mL (144 +/- 4) did not differ (P greater than .50) for 18 gilts between 100 and 190 d of age. In two ovulating, 70-d-old gilts, the interval from onset of GnRH treatment to peak E2 (171 +/- 6), peak LH (186 +/- 0), and P4 greater than or equal to 1 ng/mL (216 +/- 4) was lengthened (P less than .001). Peak concentrations of E2 (pg/mL) were higher (P less than .01) at 190 d (48 +/- 2) and 150 d (49 +/- 2) than at younger ages and lower (P less than .01) in gilts 70 d of age (31 +/- 1) than in gilts 100 d of age (41 +/- 2). Peak LH (nanograms/milliliter) was higher (P less than .01) in gilts 100 d of age (12.7 +/- 6) than in older gilts. Concentrations of P4 were similar (P greater than .20) for all ovulating gilts. The number of CL (12.7 +/- .7) did not differ (P greater than .20) for 18 gilts 100 d of age or older but was higher (P less than .01) than that (4.5 +/- 1.1) for two gilts 70 d of age. Corresponding endocrine responses or ovulations were not observed in four 70-d-old gilts treated with GnRH or in gilts given saline. These findings indicate that the functional integration of the pituitary-ovarian axis is completed between 70 and 100 d of age. Hourly treatment with GnRH is an adequate stimulus to induce ovulation in prepubertal gilts as early as 70 d of age. Also, the number of follicles reaching ovulatory competency was similar (P greater than .20) in gilts between 100 and 190 d of age, when GnRH was given on a BW basis.  相似文献

The effects of n-methyl-d,l-aspartate (NMA), a neuroexcitatory amino acid agonist, on luteinizing hormone (LH), prolactin (PRL) and growth hormone (GH) secretion in gilts treated with ovarian steroids was studied. Mature gilts which had displayed one or more estrous cycles of 18 to 22 d were ovariectomized and assigned to one of three treatments administered i.m.: corn oil vehicle (V; n = 6); 10 micrograms estradiol-17 b/kg BW given 33 hr before NMA (E; n = 6); .85 mg progesterone/kg BW given twice daily for 6 d prior to NMA (P4; n = 6). Blood was collected via jugular cannulae every 15 min for 6 hr. Pigs received 10 mg NMA/kg BW i.v. 2 hr after blood collection began and a combined synthetic [Ala15]-h GH releasing factor (1-29)-NH2 (GRF; 1 micrograms/kg BW) and gonadotropin releasing hormone (GnRH; .2 micrograms/kg BW) challenge given i.v. 3 hr after NMA. NMA did not alter LH secretion in E gilts. However, NMA decreased (P < .02) serum LH concentrations in V and P4 gilts. Serum LH concentrations increased (P < .01) after GnRH in all gilts. NMA did not alter PRL secretion in P4 pigs, but increased (P < .01) serum PRL concentrations in V and E animals. Treatment with NMA increased (P < .01) GH secretion in all animals while the GRF challenge increased (P < .01) serum GH concentrations in all animals except in V treated pigs. NMA increased (P < .05) cortisol secretion in all treatment groups. These results indicate that NMA inhibits LH secretion and is a secretagogue of PRL, GH and cortisol secretion with ovarian steroids modulating the LH and PRL response to NMA.  相似文献

One-hundred-twenty crossbred gilts from two experiments were assigned randomly to a 2 X 5 factorial experiment. Gilts were reared in two environments (confinement or outside) and assigned to be slaughtered at 4, 5, 6, 7 or 8 mo of age. Beginning at 6 mo of age, blood samples were taken at weekly intervals from each gilt via venipuncture. Serum concentrations of progesterone were analyzed to determine when gilts attained puberty. On the day prior to slaughter, six pigs within a treatment group were cannulated and blood samples were taken at 20-min intervals for 4 h. At slaughter, follicular fluid (FF) was aspirated and the volume determined from those follicles having a diameter of at least 4 mm. No effect of environment was found on the proportion of gilts that attained puberty by 8 mo of age. For the 12 gilts that reached puberty during the study, the age at puberty for gilts reared in outdoor lots (202 +/- 5 d) was less (P less than .05) than those reared in confinement (224 +/- 8 d). Mean concentrations of serum luteinizing hormone (LH; P = 98) and number of secretory spikes of LH (P = .76) were similar between gilts reared in confinement and those reared in outdoor lots. No differences in average serum concentrations of follicle stimulating hormone (FSH) or number of secretory spikes of FSH were found between gilts subjected to these environments (P = .95). The possible roles of dietary protein level and feed restriction in regulating mammary development of prepubertal gilts were investigated. Cross-bred gilts were fed a commercial diet until 90 d of age and then divided into four nutritional regimens based on two pelleted diets (as-fed basis): a high-protein diet (HP = 13.8 MJ of ME, 1.0% total lysine, 18.7% CP) and a low-protein diet (LP = 13.8 MJ of ME, 0.7% total lysine, 14.4% CP). Nutritional regimens were as follows: 1) HP ad libitum until slaughter (n = 22, T1); 2) HP ad libitum until 150 d of age followed by LP until slaughter (n = 20, T2); 3) LP ad libitum until slaughter (n = 21, T3); and 4) HP with a 20% feed restriction until slaughter (n = 19, T4). Gilts were weighed, their backfat thickness was measured, and jugular blood samples were obtained on d 90, 150, and at slaughter to determine concentrations of prolactin, IGF-I, leptin, and glucose. Gilts were slaughtered 8+/-1 d after their first or second estrus (202.7+/-14.5 d of age). Mammary glands were excised, parenchymal and extraparenchymal tissues were dissected, and composition of parenchymal tissue (protein, fat, DM, DNA, protein/DNA) was determined. The T4 gilts weighed less (P < 0.01) and had less backfat (P < 0.01) than did gilts on other treatments on d 150 and at slaughter. Treatments had no significant effects on prolactin, IGF-I, or glucose concentrations, but there was a treatment x day interaction (P < 0.01) for leptin, with concentrations being lower at slaughter in restricted-fed (T4) vs. LP (T3) gilts (P < 0.05). There was less extraparenchymal mammary tissue (P < 0.01) in T4 gilts than in gilts from the other groups and a tendency (P = 0.13) for the amount of parenchymal tissue to be lower in T4 gilts. In conclusion, a lower lysine intake during prepuberty did not hinder mammary development of gilts, but a 20% feed restriction decreased mass of parenchymal and extraparenchymal tissues. The effect of feed restriction on extraparenchymal tissue is most likely associated with the lower fat deposition.  相似文献

To evaluate morphological characteristics and development of the immune system at different ages in neonatal pigs, 4 piglets were euthanized at 7, 14, and 18 d of age for collection of blood, bile, and intestinal tissue for morphological measurements. Blood was collected for differential cell counts, lymphocyte blastogenesis, immunoglobulin (Ig) concentrations, cytokine concentrations, and flow cytometric analysis. Bile was collected for quantification of Ig-A and Ig-M. Villus width and crypt depth from duodenum sections, as well as ileum crypt depth, were reduced (P < or = 0.08) in 18-d-old pigs compared with 7-d-old pigs. No age-related differences (P > or = 0.11) were observed in the number of goblet cells with neutral and acidic mucins, serum or enteric Ig concentrations, IL-2, IL-4, spontaneous lymphocyte proliferation, or leukocyte concentrations. When measured as counts per minute (cpm) and as a stimulation index (SI), lymphocyte proliferation responses to phytohaemagglutinin increased (P = 0.05) between 7 and 14 d of age; no changes (P = 0.10) occurred at 18 d of age. No age-related changes (P = 0.39) were observed in response to pokeweed mitogen (PWM) when measured as cpm; however, the SI from PWM-induced lymphocytes decreased (P = 0.04) 4-fold between 7 and 18 d of age. The CD4+:CD8+ and populations of lymphocytes expressing CD2+CD4+CD8- (T helper cells) and CD25+CD4+CD8- (activated T helper cells) were greater (P > or = 0.04) at 7 d of age than at 14 and 18 d. Populations of T lymphocytes, cytotoxic T cells (CD2+CD4-CD8+), activated lymphocytes (CD25+), and activated cytotoxic T cells (CD25+CD4-CD8+) were greater (P > or = 0.02) in 18-d-old pigs compared with 7-d-old pigs, whereas CD2+CD4-CD8- [double negative cells] were lower (P = 0.08) in 18-d-old pigs compared with 14-d-old pigs. The percentage of CD2+ T cells was 8.4% at 7 d of age, and by the time the pigs reached 18 d of age, the percentage of CD2+ T cells was 33.8%. Moreover, the percentage of gammadelta T cells was greater (P = 0.02) in 18-d-old pigs than in 7-d-old pigs (74.8 vs. 46.1%, respectively). Results indicate that the porcine immune system and gut are continuously changing as the young pig matures. Changes occurred in lymphocyte phenotypic expression and functional capabilities, as well as morphology and mucin production, and their role may be to further protect the neonate from antigenic challenge as protection from passive immunity declines.  相似文献

Serum concentrations of pituitary and adrenal hormones were determined in lactating sows and ovariectomized (OVX) gilts exposed to 8 h (8L:16D) or 16 h of light (16L:8D). In addition serum prolactin (PRL) concentrations were determined after a thyrotropin releasing hormone (TRH) challenge. At 103 +/- 2 d of gestation or 3 wk after ovariectomy of nulliparous gilts on d 7 to 9 of the estrous cycle (d - 10), blood samples were collected from jugular vein cannulae at 30-min intervals for 8 h beginning at 0800 h. Immediately after the last sample, 13 sows and five OVX gilts were assigned to 8L:16D and 14 sows and five OVX gilts were assigned to 16L:8D/d and placed in two identical chambers in the farrowing house. Blood sampling was repeated on d 7, 14 and 21 of lactation in the sows and on d 7, 14, 21 and 28 in the OVX gilts. In Exp. 1, serum cortisol (C) concentrations were similar for sows exposed to 8L:16D (n = 7) and 16L:8D (n = 6) treatments, whereas in Exp. 2, serum C concentrations for sows exposed to 8L:16D (n = 6) were lower than those exposed to 16L:8D (n = 6) on d 7, 14 and 21. The objectives were to determine relative ADG, ADFI, behavior, and endocrine responses in weaned pigs receiving exogenous ghrelin. Twenty-four barrows weaned at 18 d of age (d 0 of the experiment) were catheterized via the jugular vein, weighed, and assigned to either a ghrelin (n = 12) or saline (control; n = 12) infusion group. Initial pig BW did not differ between treatments (7.87+/-0.39 vs. 7.92+/-0.35 kg for ghrelin and control treatments, respectively). Pig BW and feed intakes were measured once daily throughout the experiment. Starting on d 1, the ghrelin pigs were intravenously infused three times daily for 5 d with 2 microg/kg BW of human ghrelin, and the control pigs were similarly infused with saline. Activity observations and blood samples were taken at -15, 0, 15, 30, 60, 90, 120, 240, and 480 min relative to the first infusion and then three times daily (0800, 1600, and 2400) for 8 d. Weight gain during the 5-d infusion period was greater by the ghrelin than by control pigs (0.57+/-0.10 vs. 0.21+/-0.13 kg, respectively; P < 0.04); however, there was no increase in feed intake. During two behavioral observation periods, more pigs in the ghrelin treatment were observed eating compared with control pigs (P < 0.05). The initial infusion of exogenous ghrelin increased serum ghrelin, GH, insulin, and cortisol concentrations (P < 0.05). Endogenous serum ghrelin increased from d 1 to 8 of the experiment in control animals (P < 0.05). Serum IGF-I initially fell in both treatment groups from d 1 to 2 (P < 0.05) but then increased from d 5 to 8 (P < 0.05). Peripheral concentrations of glucose in the ghrelin pigs were greater on d 2, 3, 7, and 8 than on d 1 (P < or = 0.05). In both treatment groups, peripheral concentrations of leptin increased from d 7 to 8, and cortisol decreased from d 1 to 5 of the experiment. These observations provide evidence that ghrelin may positively influence weight gain and concomitantly increase GH, insulin, and cortisol secretion in weaned pigs.  相似文献

The objective of this study was to evaluate the relationship between serum concentrations of the hormone leptin with growth and carcass traits insix distinct breeds of pigs entered into the 2000 National Barrow Show Sire Progeny Test. Breeds evaluated were Berkshire (n = 131), Chester White (n = 33), Duroc (n = 40), Landrace (n = 23), Poland China (n = 26), and Yorkshire (n = 41). Serum samples were collected and assayed for concentrations of leptin at entry into test (On-Test Leptin) at 34 +/- 6.7 kg of live weight and again 24 h prior to harvest (Off-Test Leptin) at 111 +/- 3.1 kg of live weight. Carcass measurements taken included hot carcass weight, carcass length, backfat, longissimus muscle area (LMA), longissimus pH, Hunter L-value, chemically determined intramuscular fat (IMF), and subjective color, marbling, and firmness scores. Average daily gain, IMF percentages, and water-holding capacity (WHC) were also determined. On-Test Leptin concentrations were not different (P > 0.10) between swine breeds; however, Off-Test Leptin concentrations did differ (P < 0.001) across genotype. Berkshire had the greatest Off-Test Leptin concentrations (6.58 +/- 0.43 ng/mL), and Duroc and Yorkshire had the lowest (3.49 and 3.96 +/- 0.68 ng/mL; respectively). In addition, Off-Test Leptin concentrations were correlated with average daily gain (r = 0.29; P < 0.001), last-rib fat thickness (r = 0.48; P < 0.001), 10th rib backfat (r = 0.52; P < 0.001), LMA (r = -0.33; P < 0.001), percent fat-free carcass lean (r = -0.51; P < 0.001), and WHC (r = 0.15; P < 0.05). Off-Test Leptin concentrations also differed by gender, with barrows having greater (P < 0.001) serum concentrations of leptin than gilts (6.55 +/- 0.48 vs 3.35 +/- 0.44). Differences exist between breeds of pigs in a manner consistent with breed-specific traits for growth, leanness, and quality; thus, leptin may serve as a useful marker for selection or identification of specific growth and carcass traits.  相似文献

Two experiments (EXP) were conducted to test the hypothesis that porcine leptin affects GH, insulin-like growth factor-I (IGF-I), insulin, thyroxine (T4) secretion, and feed intake. In EXP I, prepuberal gilts received intracerebroventricular (ICV) leptin injections. Blood was collected every 15 min for 4 hr before and 3 hr after ICV injections of 0.9% saline (S; n = 3), 10 μg (n = 4), 50 μg (n = 4), or 100 μg (n = 4) of leptin in S. Pigs were fed each day at 0800 and 1700 hr over a 2-wk period before the EXP. On the day of the EXP, pigs were fed at 0800 hr and blood sampling started at 0900 h. After the last sample was collected, feeders were placed in all pens. Feed intake was monitored at 4, 20, and 44 hr after feed presentation. In EXP II, pituitary cells from prepuberal gilts were studied in primary culture to determine if leptin affects GH secretion at the level of the pituitary. On Day 4 of culture, 105 cells/well were challenged with 10−12, 10−10, 10−8, or 10−6 M [Ala15]-h growth hormone-releasing factor-(1-29)NH2 (GRF), 10−14, 10−13, 10−12, 10−11, 10−10, 10−9, 10−8, 10−7, or 10−6 M leptin individually or in combinations with 10−8 and 10−6 M GRF. Secreted GH was measured at 4 hr after treatment. In EXP I, before injection, serum GH concentrations were similar. Serum GH concentrations increased (P < 0.01) after injection of 10 μg (21 ± 1 ng/ml), 50 μg (9 ± 1 ng/ml), and 100 μg (13 ± 1 ng/ml) of leptin compared with S (1 ± 2 ng/ml) treated pigs. The GH response to leptin was greater (P < 0.001) in 10 μg than 50 or 100 μg leptin-treated pigs. By 20 hr the 10, 50, and 100 μg doses of leptin reduced feed intake by 53% (P < 0.08), 76%, and 90% (P < 0.05), respectively, compared with S pigs. Serum IGF-I, insulin, T4, glucose, and free fatty acids were unaffected by leptin treatment. In EXP II, relative to control (31 ± 2 ng/well), 10−10, 10−8, and 10−6 M GRF increased (P < 0.01) GH secretion by 131%, 156%, and 170%, respectively. Only 10−6 M and 10−7 M leptin increased (P < 0.01) GH secretion. Addition of 10−11 and 10−9 M leptin in combination with 10−6 M GRF or 10−11 M leptin in combination with 10−8 M GRF-suppressed (P < 0.05) GH secretion. These results indicate that leptin modulates GH secretion and, as shown in other species, leptin suppressed feed intake in the pig.  相似文献

A study was conducted to determine if Se source fed during gestation and lactation affects passive transfer of immunoglobulins. Sixty days prior to breeding (d -60), gilts were randomly assigned to one of three treatments prior to breeding and throughout gestation: control (Control, no supplemental Se; n = 8), inorganic Se (Inorganic Se, 0.3 ppm; n = 4) and organic Se (Organic Se, 0.3 ppm; n = 4). Blood was collected on d -60, 57 and 113 of gestation and on d 21 of lactation and milk was collected at d 0, 1, 7, 14, and 21 of lactation. Blood was collected from piglets at d 0, 1, 7, 14, and 21 of age. Gilts fed organic Se had greater (P 〈 0.05) circulating concentrations of Se than Inorganic and Control gilts. Regardless of treatment, circulating concentrations of Se were greatest (P 〈 0.05) at d -60 compared to all other days. Serum concentrations of IgG were greatest (P 〈 0.05) in gilts at d 57 of gestation compared to d 113. Serum concentrations of IgA were greatest (P 〈 0.05) on d 113 of gestation and d 21 of lactation compared to d -60 and 57. Serum concentrations of IgM were greater (P 〈 0.05) at d 57 compared to d -60. Inorganic gilts had greater (P 〈 0.05) colostral and milk concentrations of IgG and IgM than Organic or Control gilts. Circulating concentrations of Se in piglets were greatest (P 〈 0.05) at d 14 and 21 of age compared to all other days. Piglets from gilts supplemented with organic Se had greater (P 〈 0.05) circulating concentrations of Se on d 1 versus piglets from gilts supplemented with no additional Se. The immunoglobulin concentrations of IgG, IgA, and IgM were lowest (P 〈 0.05) on d 0 and then different sources of Se did not affect the immunoglobulin ncreased when compared to d 1. Secretion into the uterine lumen follows a precise pattern during early pregnancy. Near the end of the second week of pregnancy and coincident with elongation of conceptuses, retinol, retinol binding protein (RBP), estradiol (E2), and prostaglandins E (PGE) and F (PGF) increase in the uterine lumen, and RBP mRNA increases in the endometrium. In the present studies the potential for E2 (0.1 microM) and retinol (10 microM) to regulate RBP and PG production by cultured luminal (LEC) and glandular (GEC) epithelial cells collected from postpubertal females and LEC from prepubertal gilts was examined. Endometrial tissue was collected surgically from cyclic and pregnant females (n = 8) on d 10 and 13 postestrus (first day of estrus = d 0) and from 120- and 150-d-old prepubertal gilts that were treated with progesterone (P4) (2.2 mg x kg(-1) x d(-1), n = 6) or corn oil (n = 6) for 14 d prior to tissue collection. The LEC from postpubertal females responded to retinol with increased (P < 0.05) RBP, PGE, and PGF in culture medium and increased (P < 0.07) RBP mRNA but E2 decreased (P < 0.05) RBP and RBP mRNA and had no effect on prostaglandins. No E2 or retinol effects on secretions of GEC occurred in vitro, but a day x pregnancy status interaction (P < 0.06) affected PGE output by the GEC. Secretion of PGE was greater when GEC were collected on d 10 of pregnancy than from d-10 cyclic or d-13 pregnant or cyclic females. Both E2 and retinol stimulated (P < 0.05) secretion of RBP by LEC isolated from prepubertal gilts, but their effects were not additive. In vivo treatment of prepubertal gilts with P4 increased (P < 0.05) RBP and decreased (P < 0.05) PG production by LEC in vitro. Therefore responses to E2 and retinol differ between pre- and post-pubertal females, and retinol may function in the regulation of endometrial RBP and PG secretion.  相似文献

Full term crossbred sows were selected to study the interaction of the immune system, hypothalamus-pituitary-adrenal axis, and growth in pigs born by Cesarean section (c-section; n=4 sows) or vaginal birth (n=4 sows). Gestation length and birth weight did not differ between vaginal birth and c-section pigs (P=0.34 and 0.62, respectively). Blood and tissue samples were collected from 44 pigs at birth. Forty-five pigs were weaned at 13 d. On d 14, pigs received an i.p. injection of lipopolysaccaride (LPS; 150 microg/kg) or saline at min 0, and blood samples were collected at -20, -10, 0, 5, 10, 20, 40, 60, 90, and 120 min. Vaginal birth pigs had 21% greater average daily gain than c-section pigs on d 14 (P<0.01). Basal serum concentrations of adrenocorticotrophin (ACTH) and cortisol were greater in c-section than vaginal birth pigs at birth (P<0.01) but were not different at 14 d (P=0.99 and 0.80, respectively). LPS increased serum concentrations of ACTH, cortisol, interferon-gamma (IFN-gamma), and tumor necrosis factor-alpha (TNF-alpha; P<0.01) but the response was not different between c-section and vaginal birth (P>0.22). Basal serum concentrations of TNF-alpha tended to be greater in c-section vs vaginal birth pigs at 14 d (P=0.0967); however, basal serum concentrations of IFN-gamma tended to be lower in c-section pigs vs vaginal birth pigs at 14 d (P=0.0787). Expression of interleukin (IL)-6, IL-6 receptor, IL-1beta, and TNF-alpha mRNA did not differ between vaginal birth and c-section pigs but changed in an age and tissue dependent manner. Thus, reduced growth rate of c-section pigs is associated with altered immune system function.  相似文献

研究早熟与晚熟品种母猪下丘脑-垂体-卵巢轴Kiss1和GPR54基因的表达差异。选用8头梅山母猪与12头长大(LY)母猪为研究对象,梅山母猪于70 d和100 d屠宰,LY母猪于70、100 d和199 d屠宰,收集血清及下丘脑、垂体、卵巢组织样品。ELISA检测血清瘦素(Leptin)和雌二醇(E2)水平,PCR克隆梅山与LY母猪Kiss1基因编码序列,实时荧光定量PCR检测母猪下丘脑、垂体、卵巢组织Kiss1和GPR54基因表达水平。结果表明:梅山与LY母猪Kiss1基因编码序列相似性为100%;梅山与LY母猪初情期下丘脑Kiss1基因表达量极显著高于垂体与卵巢(P<0.01),卵巢GPR54基因表达水平显著高于下丘脑与垂体(P<0.05)。梅山母猪下丘脑-垂体-卵巢轴Kiss1基因表达水平都显著高于相同日龄和初情期LY母猪(P<0.05),梅山母猪血清Leptin水平极显著高于相同日龄LY母猪(P<0.01)。而E2水平显著高于100 d与初情期LY母猪(P<0.01)。Leptin与下丘脑Kiss1和GPR54基因表达呈显著正相关(P<0.05),而E2仅与下丘脑Kiss1基因表达有极显著正相关关系(P<0.01);梅山与LY母猪Kiss1基因编码区序列相似性为100%,梅山母猪下丘脑-垂体-卵巢轴上kiss1基因表达量较LY母猪高,主要原因可能是初情日龄早,下丘脑Kiss1基因表达水平的高低与血清Leptin和E2浓度密切相关。  相似文献
