利用H/L值分析鸡抗应激能力差异   总被引:1,自引:0,他引:1  
通过H/L值的比较,对不同品种鸡的抗应激能力进行评价。随机抽取如皋鸡、文昌鸡以及安卡鸡各80只,分别检测了41、56、71和89日龄的异嗜性粒细胞(H)、淋巴细胞(L)及H/L值。结果表明:(1)在41、56日龄时,如皋鸡与文昌鸡、安卡鸡相比H、L及H/L值呈极显著差异(P<0.01);89日龄的H/L值在文昌鸡和安卡鸡之间呈极显著差异(P<0.01)。(2)如皋鸡在41日龄和71日龄间H、L及H/L值呈极显著差异(P<0.01),其余差异均不显著。文昌鸡H、L及H/L值在各日龄间变化较大,41和71日龄间呈显著差异(P<0.05),41和89日龄、56和71日龄以及56和89日龄间呈极显著差异(P<0.01)。安卡鸡各值只在56和89日龄间呈显著差异(P<0.05),其余差异均不显著。研究初步表明家禽的抗应激能力存在品种间差异,如皋鸡的抗应激能力优于安卡鸡,文昌鸡晚期表现出较好的抗应激能力,随着日龄的增加,品种之间的抗应激能力差异会减小。  相似文献

随机抽取如皋、文昌和安卡鸡各80只,经新城疫和禽流感免疫后分别于30、41、56、71和89日龄检测抗体水平变化。结果表明,如皋鸡公母抗体水平无明显差异,而文昌鸡和安卡鸡在性别之间差异显著。文昌鸡新城疫抗体在所有检测日龄均低于如皋鸡和安卡鸡,而安卡鸡禽流感抗体在所有检测日龄均极显著低于如皋鸡和文昌鸡(P<0.01)。本研究初步表明,就新城疫和禽流感而言,不同鸡种以及性别之间的免疫性能存在明显差异。  相似文献

本研究通过检测鸡NLRC5基因编码区域单核苷酸多态位点,以期分析与免疫性状相关的遗传标记。采用PCR-SSCP方法对3个鸡品种NLRC5基因编码区SNPs进行检测,仅在外显子8区域发现多态位点,其中在文昌鸡中检测到AA、BB和AB 3种基因型,如皋鸡中检测到AA和AB 2种基因型,安卡鸡中仅存在AA基因型。序列分析结果显示,NLRC5基因外显子8存在G43A同义突变。测定部分免疫性状,分析不同鸡品种之间以及不同基因型与免疫性状间的关联性,结果表明:安卡鸡H/L值、ND抗体滴度、IL-1浓度均显著高于如皋鸡和文昌鸡(P0.05),AI抗体滴度极显著低于其他2个品种(P0.01),如皋鸡SBRC抗体滴度最高,但3个品种之间差异不显著(P0.05);不同基因型同免疫性状之间,AA型个体各项性状值均优于AB和BB型个体,但是差异不显著(P0.05)。综合分析,中国地方品种文昌鸡、如皋鸡的抗性优于安卡鸡种,AA型可能是NLRC5基因在机体免疫应答能力的有利基因型。  相似文献

随机选取地方鸡种牡丹红鸡和引进蛋鸡品种白来航鸡各30只,在相同饲养管理和免疫程序下饲养,于49日龄腿肌注射25%绵羊红细胞(Sheep blood red cell,SKBC)0.5mL,分别检测了41、56、71、89日龄的异嗜性性粒细胞(Heterophil)量、淋巴细胞(Lymphocyte)量和异嗜性粒细胞/淋巴细胞比率(Heterophil/Lymphocyte,H/L),56、71、89日龄的SRBC抗体滴度和30、41、56、71、89日龄新城疫(New castle disease,ND)的免疫抗体滴度等免疫性状的差异。结果显示,牡丹红鸡在H/L值、SR.BC抗体滴度极显著高于白来航鸡(P〈0.01),ND免疫抗体滴度上显著高于白来航鸡(P〈0.05)。结果表明,家禽的免疫性状在品种间存在遗传差异,中国地方鸡种比外来鸡种在多个免疫性状上存在不同程度的优势。  相似文献

选取同期孵化出雏的中国地方鸡种北京油鸡(Beijng-You Chicken,BYC)和引进蛋鸡品种白来航鸡(White Leghorn,WL)各250只母雏,在相同饲养条件和免疫程序下饲养,测定和对比异嗜性粒细胞(Heterophil)数量、淋巴细胞(Lymphocyte)数量和异嗜性粒细胞/淋巴细胞比率(Heterophil/Lymphocyte,H/L)、白细胞总数(Blood white cell count,BWCC)、绵羊红细胞(Sheep blood red cell,SRBC)免疫抗体滴度和不同日龄的禽流感(Avian Influenze,AI)、新城疫(Newcastle disease,ND)的免疫抗体滴度等免疫性状的差异.试验结果表明①北京油鸡的H/L值为0.36±0.21,白来航蛋鸡为0.52±0.51,二者呈极显著差异(P<0.001);北京油鸡的白细胞总数明显高于白来航蛋鸡(P<0.001);②SRBC抗体滴度在两品种之间差异不显著(P>0.05);③14、28日龄两次免疫禽流感疫苗(H5N1毒株)后,在免疫后21、41、71、105天的4个测定日中,两个品种的禽流感免疫抗体滴度值均呈现先升高后降低的变化趋势,北京油鸡在4个测定日龄的AI免疫滴度值都均高于白来航蛋鸡,差异均达到极显著水平(P<0.001);④同期免疫新城疫疫苗(LaSota毒株),在免疫后21、41、71和105天测定ND抗体滴度,北京油鸡在免疫后21天时达到最高,之后随日龄的增长而呈逐渐降低趋势;白来航蛋鸡在免疫后41天时抗体滴度水平达到最高,以后随日龄增长而下降.4个测定日龄的ND滴度水平北京油鸡均高于白来航蛋鸡.该研究结果初步显示,中国地方鸡种北京油鸡和引进白来航蛋鸡品种的免疫性状具有明显的遗传差异,北京油鸡的综合免疫性能和抗应激能力优于白来航蛋鸡.  相似文献

采用PCR-SSCP对12个不同鸡种glypican-1(GPC1)基因外显子8的单核苷酸多态性进行检测,同时检测如皋鸡、安卡鸡和文昌鸡3个鸡种56日龄的5个免疫性状,进一步分析SNP位点与免疫性状的关系。结果表明:12个鸡种中均出现3种带型,存在g.155 C>A错义突变,引起苏氨酸突变为精氨酸。12个鸡种群中,AA、AB和BB基因型平均频率分别为0.259、0.545和0.211;平均观察杂合度为0.466,平均Shannon信息指数为0.671。除乌骨鸡、大骨鸡偏离了Hardy-Weinberg平衡外(P<0.05),其余群体均处平衡状态。基于该位点构建的UPGMA聚类图将12个鸡种分为3大类,反映了12个鸡种在抗性方面存在的差异。经过关联分析,如皋、安卡鸡中AB型个体的IL-1浓度、H/L值与BB型存在显著差异,与AA型差异不显著;在其他免疫性状中各基因型之间均无显著差异。安卡鸡的一般抗性总体低于如皋鸡和文昌鸡。  相似文献

本研究根据鸡Apo VLDL-Ⅱ基因内含子Ⅰ的基因序列设计3对引物,分别采用PCR-RFLP、PCR-SSCP技术对文昌鸡、如皋黄鸡、丝羽乌骨鸡、安卡肉鸡及红色原鸡5个鸡种进行多态性研究。结果表明,鸡Apo VLDL-Ⅱ基因内含子Ⅰ存在4处突变,分别为:1950处G/A、2522处A/C、2705处A/C及2793处C/T。统计结果显示:安卡肉鸡基因型频率的分布与如皋黄鸡、红色原鸡存在显著差异,文昌鸡基因型频率的分布与乌骨鸡差异不显著,除安卡肉鸡外所有鸡种均处于Hardy-Weinberg平衡状态。  相似文献

按中兽医组方原则配制加工成黄荆复方添加剂A和B,分别按0.5%、1.0%、2.0%三个浓度添加,并设抗生素和空白对照,对15日龄土杂鸡随机分组进行饲养试验,分别于28、42、56、70日龄测定ND和AI抗体水平。结果显示:黄荆复方添加剂A在1.0%添加量时,能显著加快ND和AI抗体上升速度,提高抗体滴度,延长抗体高峰期。  相似文献

将鸡新城疫、禽流感(H9亚型,SY株)二联灭活疫苗与市售同类对照苗分不同剂量皮下注射接种30日龄SPF鸡,免疫后定期采血,分离血清,通过检测血清中ND和AI的HI抗体水平比较各组鸡抗体产生期、抗体高峰及免疫持续期,并进行不同血凝抗原检测HI抗体结果的对比。试验鸡血清样品检测结果显示,免疫鸡抗体产生期为免后2周内,免后8周和5周ND和AI的HI抗体滴度分别达到峰值,抗体至少可持续28周以上,两种灭活苗之间免疫效果基本相当。  相似文献

本试验系统观察了甲硝唑对鸡回忆性体液免疫应答反应的作用,并与咪唑类化合物左旋咪唑进行了比较。28日龄鸡,免疫注射绵羊红细胞 牛血清白蛋白(SRBC BSA)悬液,或稀释的布氏杆菌(BA)抗原。56日龄时,以10.0、100.0mg/kg甲硝唑或10.0mg/kg 左旋咪唑饮水给药,连用3d,给药第二天同时进行第二次免疫注射。结果表明甲硝唑(10.0~100.0mg/kg)可使鸡体内对颗粒性胸腺依赖性抗原SRBC和非胸腺依赖性抗原BA抗体滴度回忆性免疫应答反应明显升高;但使鸡对可溶性胸腺依赖性抗原BSA抗体滴度回忆性免疫应答反应明显降低。与甲硝唑比较,左旋咪唑(10.0mg/kg)也存在着非常类似的作用。  相似文献

A total of 376 chickens from different ecotypes were immunized with the non-pathogenic multi-determinant antigen sheep red blood cells (SRBC). The ecotypes included indigenous chickens from various locations in Tanzania (n=102), India (n=86) and Bolivia (n=89). In addition, eight German Dahlem Red (GDR) chicken lines with different major genes (dwarf, naked neck and frizzled) of tropical interest were also immunized with SRBC. Immune competence of the breeds was assessed by measuring complement haemolytic activity, both from the classical calcium-dependent complement pathway (CPW) and alternative calcium-independent complement pathway (APW), alongside IgTotal, IgG and IgM antibody responses to SRBC at 7 days post immunization. Large variations in complement activity and antibody responses to SRBC were observed within and between the indigenous breeds. Many indigenous chickens, especially from Bolivia, showed decreased complement activity (APW) following immunization with SRBC. Breeds from India showed the highest CPW activity and humoral (especially IgM) responses to SRBC, suggesting high immune competence. In contrast, Bolivian chickens were characterized by low CPW activity, low APW activity and low antibody levels to SRBC suggesting an overall low immune competence. In the GDR chickens, characterized by high CPW activity and high IgG antibody responses to SRBC, the major genes for naked neck, frizzling and dwarfism had no significant effect on the antibody responses and complement activity to SRBC.  相似文献

Our group has established two lines of meat-type chickens divergently selected for early (HC line) and late (LC line) antibody responsiveness at 10 days of age to immunization with inactivated pathogenic Escherichia coli bacteria. The question addressed in the study presented here is whether this selection has changed other immunological responses, increasing the overall 'early' immunocompetence. Broilers of the third and fourth generations (S3 and S4) of the selected lines (HC and LC) and a control, unselected line (CT) were vaccinated at 10 days of age with E. coli vaccine, Newcastle virus vaccine (NDV), sheep erythrocytes (SRBC) or bovine serum albumin (BSA). Line-HC chicks exhibited higher antibody titers to E. coli, NDV and SRBC than CT or LC chicks. At 20 days of age HC chicks demonstrated a higher total protein and a higher beta- and gamma-globulin levels in their serum. At 21 days of age, HC chicks cleared carbon particles faster than LC chicks. Peripheral blood lymphocytes (PBL) from HC chicks vaccinated with E. coli vaccine, proliferated in vitro more actively in the presence of the stimulating antigen than the PBL of LC chicks. Peripheral blood lymphocytes (PBL) obtained from HC-line chicks exhibited a higher proliferative response to concanavalin A (Con A)-, phytohemagglutinin (PHA)- or pokeweed mitogen (PWM)-stimulation than LC PBL. These results demonstrate that the selection for high or low antibody response to E. coli at a young age resulted also in a significant change in the response of other parameters of the immune system. The high response to E. coli was found to be associated with a high antibody response to other antigens (NDV and SRBC), increased phagocytic activity and increased proliferative response to antigen or mitogens. This study was conducted to investigate the effects of probiotic (Primalac), prebiotic (TechnoMos) and synbiotic (Primalac + TechnoMos) supplementation on performance, immune responses, intestinal morphology and bacterial populations of ileum in broilers. A total of 240 one‐day‐old broiler chicks were randomly divided into four treatment groups which included 60 birds. Control group did not receive any treatment. The chicks in the second, third and fourth groups were fed probiotic (0.9 g/kg), prebiotic (0.9 g/kg) and probiotic (0.9 g/kg) plus probiotic (0.9 g/kg; synbiotic), respectively, at entire period. Daily feed intake, daily weight gain and feed conversion ratio were evaluated. The birds were immunized by sheep red blood cell (SRBC) on days 12 and 29 of age and serum antibody titres were measured on days 28, 35 and 42. Newcastle vaccines administered on days 9, 18 and 27 to chicks and blood samples were collected on day 42. Intestinal morphometric assessment and enumeration of intestinal bacterial populations were performed on day 42. The results indicated that consumption of probiotic, prebiotic and synbiotic had no significant effect on daily feed intake, daily body weight gain, feed conversion ratio, carcass traits, intestinal morphology and bacterial populations of ileum (p > 0.05). Consumption of prebiotic increased total and IgM anti‐SRBC titres on days 28 and 42 and antibody titre against Newcastle virus disease on day 42 (p < 0.05). Synbiotic increased only total anti‐SRBC on day 28 (p < 0.05). It is concluded that consumption of prebiotic increased humoral immunity in broilers. Therefore, supplementation of diet with prebiotic for improvement of humoral immune responses is superior to synbiotic supplementation.  相似文献

Antibody responses of hens and their progeny were studied in commercial broiler nuclear lines. Starting at 168 days of age, individually housed pullets from lines A and B were fed a 16% crude protein and 2752 kcal metabolizable energy/kg mash diet supplemented with either 10 or 300 IU/kg of vitamin E fed as dl-alpha-tocopherol acetate. Fifty-eight days later (226 days of age), 12 hens per line-vitamin E subclass were inoculated i.v. with 0.1 ml of a 2.5% suspension of sheep red blood cells (SRBC). Plasma antibody titers were measured 6, 20, 40, 54, 70, and 88 days after inoculation. Hens from both lines were artificially mated to males from line C, and progeny from eggs collected 9-15, 25-30, and 65-70 days after inoculation were tested for antibodies to SRBC. Hens were reinoculated i.v. with 0.1 ml of 0.25% SRBC 88 days after the first inoculation, and their antibody levels were measured 3, 6, and 20 days later. Eggs laid 10-13 days after reinoculation were incubated, and antibody titers of chicks were measured at hatch. Antibody response of hens to an initial inoculation of SRBC was line-diet-time after inoculation specific. In line A, titers were greater for hens fed the lower than the higher vitamin E diet, whereas diet had no effect on the antibody levels in line B. Line effects (A > B) were observed on days 6 and 20 after inoculation but not thereafter. After the second inoculation, dietary vitamin E level had no effect on antibody levels of hens within lines, whereas a between-line difference (A > B) was observed for the lower but not the higher level of dietary vitamin E. Although there was no difference between diets for antibody transferred to progeny by line B, there was a difference (lower > higher) for line A. After reinoculation of their dams, antibody titers of chicks from line A, but not line B, reached levels similar to those after the first inoculation. Antibody levels were higher for chicks at hatch than in 16-day embryos or 10 days posthatch. The results of this research suggest genetic variation in response to immune stimulation by dietary vitamin E.  相似文献

The effects of the lipopolysaccharide-protein complex (LPS) and crude capsular antigen (CCA) prepared from Pasteurella multocida serotype A isolated from a duck in the Philippines, on antibody responses to sheep red blood cells (SRBC) and Brucella abortus (BA) and delayed type hypersensitivity (DTH) responses to bovine serum albumin (BSA) in the chickens were studied. Chickens injected subcutaneously with LPS and CCA at 1 and 2 weeks of age and immunized intravenously with the mixed antigens of SRBC and BA, at 3 and 4 weeks of age showed significantly increased antibody responses against both SRBC and BA, when evaluated at 7 days after each immunization. In addition, these chickens sensitized intramuscularly with the emulsion of BSA in complete Freund's adjuvant at 5 weeks of age, and then injected into the wattle with BSA at 7 weeks of age also showed significantly increased DTH responses against BSA, when evaluated at 24 and 48 hr after challenge. These results indicate that LPS and CCA of P. multocida serotype A have a property enhancing humoral and cell-mediated immune responses.  相似文献

SUMMARY: Two lines of White Leghorn chickens selected divergently for high (HH) or low (LL) antibody response 5 days after an injection with 0.1 ml of 0.25% suspension of sheep red blood cell (SRBC) antigen were used to produce parental, reciprocal F(1) , F(2) and backcross progeny. At 36 days of age males and females of the various progeny types were injected with SRBC suspension and antibody titres measured at 5 and 12 days later. Progeny of the high antibody line had higher titres at both 5 and 12 days after inoculation with SRBC than those of the low line. Reciprocal effects for SRBC titres were important only for female progeny suggesting sex-linked effects of the Z chromosome. Titres for F(1) progeny were intermediate and different from the parental lines at both 5 and 12 days after inoculation. Antibody titres 5 days after inoculation exhibited heterosis which emanated from the homogametic sex. Although maternal effects generally had no influence on antibody titres, maternal heterosis in the selected trait was due to sex-linkage. Recombination effects were negligible for both traits. In Alter von 36 Tagen wurde den Tieren eine SRBC Suspension injiziert und Antik?rpertiter 5 und 12 Tage sp?ter bestimmt. Nachkommen der HH Linie hatten an beiden Tagen h?here Titer als LL Nachkommen. Reziproke Effekte für SRBC Titer waren nur bei weiblichen Nachkommen wichtig, bedingt wohl durch geschlechtsgekoppelte Z-Chromosom Einflüsse. Die Titer der F(1) Nachkommen waren zwischen denen der Elternlinien. Antik?rpertiter 5 Tage nach Injektion zeigten Heterosis, die im homogametischen Geschlecht auftrat. Obwohl im allgemeinen maternale Wirkungen keinen Einflu? zeigten, war maternale Heterosis beim selektierten Merkmal auf Geschlechtskopplung zurückzuführen. 试验旨在研究红景天提取物对高原缺氧条件下肉鸡接种法氏囊疫苗和绵羊红细胞后产生的特异性抗体水平的影响。随机选取100只1日龄健康AA肉仔鸡,随机分为5组。其中,对照组只饲喂基础日粮,其余4组分别在基础日粮中添加0.10%、0.15%、0.20%和0.40%红景天水提物干粉。每个组取半数鸡只于13日龄腹腔注射接种绵羊红细胞,另一半鸡只于20日龄口服接种法氏囊疫苗。研究发现,免疫接种法氏囊疫苗后2 d和19 d,相比于对照组,0.20%和0.40%红景天处理组显著提高了法氏囊疫苗抗体水平(P<0.05)。在腹腔注射绵羊红细胞后5 d、8 d和12 d时,相比对照组,0.20%和0.40%红景天处理组显著提高了绵羊红细胞抗体水平(P<0.05)。该试验表明红景天提取物可以提高高海拔缺氧条件下肉鸡特异性抗体水平,因此其可能作为高海拔缺氧条件下肉鸡日粮的一种免疫调节性添加剂。  相似文献

Chicks from lines selectively bred for either high or low antibody response post-primary immunization (PPI) to sheep erythrocytes (SRBC) and their reciprocal crosses were immunized intravenously with 0.1 ml of 0.25% SRBC antigen in Trials 1 and 3 and with 0.1 ml of 0.025%, 0.25%, or 25% SRBC antigen in Trial 2. All initial immunizations were made at 35 days of age, and 0.1 ml of 0.25% SRBC booster dose was given 24 days later. Results showed that (a) population differences appeared by day 4 PPI and persisted through day 24 PPI, (b) regardless of population, peak titers occurred at about the same time for primary and secondary immunizations, (c) dosage of antigen influenced differences among populations in antibody response to primary immunization, and (d) both selected lines had similar responses in 2-mercaptoethanol-sensitive and 2-mercaptoethanol-resistant antibodies to primary but not secondary immunization.  相似文献
