Changes in the content of biogenic amines (BA; e.g., putrescine, tyramine, and histamine) and the mesophilic and psychrotrophic bacterial counts of tiger-tooth croaker (Otolithes ruber) were monitored during ice storage for 18 days. BA and bacterial loads gradually increased (p < 0.05) with storage time. On the first day of storage, no amines were detected, and no histamine was detected up to 6 days. The best linear regressions between BA and bacterial counts were found between putrescine and psychrotrophic bacteria. Furthermore, it was found that psychrotrophs were the dominant microorganisms during storage and showed significant (p < 0.05) differences with storage time. The initial concentration of putrescine was 1.27 μg g?1 and reached 18.8 μg g?1 when the psychrotrophic bacterial load was over 108 CFU g?1. The results suggest that the level of putrescine is a suitable indicator of freshness for O. ruber, and the shelf life of O. ruber stored in ice as determined by BA content and microbiological data was 10 days.  相似文献

Lytic peptides have been shown in vitro to be toxic to a wide range of fish bacterial pathogens, including Edwardsiella ictaluri, the causative agent for enteric septicemia. Fingerling channel catfish, Ictalurus punctatus, were challenged with an injection of Edwardsiella ictaluri, and lytic peptide was administered with a single intraperitoneal injection 24 hours later. In a second group, lyptic peptide was also administered by osmotic pump 7 days before the bacterial challenge. A range of peptide concentrations (20-400µg/g fish) was used. Mortalities were recorded for 7 days after injection of bacteria, and E. icraluri was isolated and quantified by standard plate counts from livers. Various tissues were examined for abnormalities due to bacteria and peptide treatment. Fish injected with lytic peptide at 400 µg/g had up to a 95% reduction in bacteria and a 65% reduction in mortality compared to complete mortality in bacteria-injected controls. Fish implanted with osmotic pumps had 99.3% reduction in bacterial counts and had 6% mortality. Tissue samples from infected fish showed changes typical of reaction to immunologic stimuli and infection, while control fish injected with saline or peptide had normal gills, spleens, and kidneys. This study shows the potential for enhancing channel catfish disease resistance to E. ictaluri and other diseases by transferring the gene coding for the lytic peptide into channel catfish.  相似文献

Bacteriophage isolated from the semi-intensive culture of Pacific white leg shrimp Litopenaeus vannamei infects the luminous bacteria Vibrio harveyi. Lytic activity and lytic spectrum results revealed that the isolated phage had strong lytic activity in V. harveyi, V. parahaemolyticus, and V. vulnificus. Biofilm inhibition activity was performed against different pathogenic vibrios on high-density polyethylene (HDPE) template and the result revealed that the phage effectively inhibited the biofilm formation in V. harveyi. Spectrophotometric assay performed for lytic activity of the isolated phage in V. harveyi liquid culture showed that the phage significantly decreased the V. harveyi cell densities at different time intervals (P?<?0.05). To study the stability of phage at different temperature and pH revealed that the phage withstands the temperature ranged between 40 and 70 °C and the pH of 4 and 9 at a significant level (P?<?0.001). One-step growth curve depicted that the burst size gradually increased to a significant level and reached the maximum of 90% at 180 min (P?<?0.05). This study concluded that the isolated phage had specific activity against pathogenic V. harveyi infections.

Quantitative and qualitative analyses of bacterial flora associated with pond water, gills, and intestine of polycultured healthy common carp (Cyprinus carpio) and African catfish (Clarias gariepinus) were carried out and identified to species level where possible. Total viable bacterial counts in the pond water ranged from 9.2?±?5.5?×?103 to 6.6?±?5.1?×?104 colony-forming units (cfu)/mL; in the gill filaments of carp and catfish, 3.3?±?3.8?×?106 to 7.9?±?5.6?×?106 and 1.1?±?4.6?×?105 to 2.3?±?5.2?×?106 cfu/g, respectively; and in the intestine of carp and catfish, 1.4?±?2.9?×?1010 to 1.7?±?6.0?×?1011 and 2.7?±?3.4?×?1010 to 1.0?±?4.5?×?1011 cfu/g, respectively. Gram-negative rod-shaped bacteria dominated the populations: 90% in carp, 89% in catfish, 80% in water, and 86% in the total populations. Altogether, 14 bacterial species of 10 genera were identified in total populations. Pond water bacteria had a reflection on the bacterial composition of the gills and intestine of carp and catfish. Aeromonas hydrophila, Shewanella putrefaciens, Vibrio cholerae, Staphylococcus sp., and Vibrio vulnificus appeared as the common bacteria in the populations, where the first three were highly significantly abundant (P?<?0.0001). Moreover, A. hydrophila was the most significantly dominant bacteria (32%; P?<?0.005) among the total populations. Pantoea sp. and Pasteurella pneumotropica were present only in carp and catfish, respectively, but Corynebacterium urealyticum and Micrococcus sp. were present only in pond water.  相似文献

Asian seabass, Lates calcarifer juveniles were fed for 30 days to evaluate the efficacy of cellulolytic (Bacillus sp. DDKRC1.) and amylolytic (Bacillus subtilis DDKRC5.) bacteria supplemented diets. Four experimental groups were maintained in triplicate of which group I was the control group and received no bacterial supplementation. Groups II, III and IV were fed diet supplemented with B. subtilis DDKRC5. (14.25 × 107 CFU/mL), Bacillus sp. DDKRC1. (2.94 × 107 CFU/mL) and mixture of both the microbes (1:1 ratio), respectively, @ 1% (v/w). After 30 days of feeding, fish reared as group IV showed significantly higher (P < 0.01) weight gain (141.42%), survival (91.50%), protein efficiency ratio (1.18) and significantly lower (P < 0.01) FCR (2.15) than that of other groups. Nutrient digestibility parameters were significantly (P < 0.01) higher in group IV followed by groups III, II and I. Digestive cellulase, amylase and protease activities in the gastrointestinal tract were also significantly (< 0.01) higher in group IV. Amylolytic and cellulolytic microbial count in the gastrointestinal (GI) tract were significantly (P < 0.01) higher in groups II, III and IV as compared with that of control. It can be concluded that feed supplemented with mixture of B. subtilis DDKRC5. and Bacillus sp. DDKRC1. showed better performance in Asian seabass juveniles.  相似文献

Quantitative and qualitative estimation of bacterial flora present in pond water, sediments, gills, and intestine of healthy common carp Cyprinus carpio cultured in Saudi Arabia were performed and identified to species level where possible. Mean total viable bacterial counts in pond water ranged from 1.2?±?2.9?×?104 to 2.5?±?3.5?×?105 cfu/mL; in sediments, 9.3?±?2.1?×?107 to 2.7?±?3.5?×?109 cfu/g; in gills filaments, 4.3?±?2.9?×?106 to 1.6?±?3.9?×?107 cfu/g; and in intestine, 8.7?±?4.1?×?109 to 5.4?±?3.2?×?1010 cfu/g. Gram-negative rod-shaped bacteria dominated (76%) the populations. In total, 12 bacterial genera and 15 species were identified. Pond water and sediment bacteria had the reflection on bacterial composition of gills and intestine of carp. Intestinal bacteria showed more diversification in contrast to gill bacteria. Aeromonas hydrophila, Shewanella putrefaciens, Vibrio cholerae, Corynebacterium urealyticum, Vibrio vulnificus, Vibrio sp., and Staphylococcus sp. were the common bacteria in all the populations. In pond water and carp intestine, A. hydrophila, S. putrefaciens, V. cholerae, and C. urealyticum were the most dominant bacteria (prevalence ≥ 10%) where pond sediments and the carp gills experienced with more one dominant bacterium V. vulnificus. Only the A. hydrophila covered one fourth (25%) of the total bacterial populations.  相似文献

The experiment was conducted to investigate the effects of oral administration of probiotic Halomonas sp. B12 (previously isolated from the intestine of Fenneropenaeus chinensis) on the intestinal microflora, immunological parameters, and midgut histological structure of F. chinensis. Shrimp (initial weight: 4.00 ± 0.10 g) were fed diets containing Halomonas sp. B12 at 0 (control), 3.68 × 107 (T1), and 7.18 × 1010 (T2) colony‐forming units per gram for 6 wk, respectively. The results showed that the total bacterial counts significantly increased (P < 0.05) with supplementation of dietary probiotic B12. However, Vibrio spp. counts significantly decreased (P < 0.05) in the intestine of shrimp with increasing dietary probiotic B12. Hemocyte counts in the shrimp fed the diets supplemented with probiotic B12 were significantly higher (P < 0.05) than the control group. Both phenoloxidase (PO) activity in plasma and hemocyte lysate supernatant were higher in the shrimp fed diets supplemented with probiotic B12 compared with the control group. No significant difference was found in PO activity in plasma between the T1 and the control (P > 0.05). PO activity in plasma was higher in T2 than that in T1. Cumulative shrimp mortality after 10‐d white spot syndrome virus (WSSV) challenge test significantly decreased with increasing dietary probiotic B12 (P < 0.05). No significant difference was found between T1 and control in cumulative shrimp mortality after challenge test (P > 0.05). The oral administration of probiotic B12 to F. chinensis was beneficial to protect the integrity of shrimp intestinal mucosal layer. In summary, even though the low dose (T1) had some effects on bacterial counts and immunological parameters, only the high dose (T2) significantly increased the resistance of the shrimp to WSSV.  相似文献

Microbiological conditions in 200 L cultures of microalgae Chlorella minutissima in polyethylene sleeves were examined. The influence of addition of antibiotic (nitrofurantoin) was studied. Samples were taken 2, 5, 9, and 14 d after inoculation and were spread on solid Zobell medium and on thiosulfate citrate bile salts sucrose agar for the enumeration of total numbers of bacteria and numbers of presumptive Vibrio bacteria, respectively. Microalgae grew well in the cultures and high numbers of culturable bacteria (about 107 bacteria/mL) were present in all microalgae cultures. Addition of antibiotic in 200‐L cultures of C. minutissima did not result in a significant decrease of total counts of bacteria per unit volume (P > 0.05), while the numbers of presumptive Vibrio were higher in cultures added antibiotic compared with the control treatment (P < 0.05). The numbers of presumptive Vibrio per unit volume and the percentage of fast‐growing bacteria were higher in microalgae supernatant than in noncentrifuged samples. Representative colonies were taken from all samples and in total 649 bacterial strains were isolated during this study. A range of phylotypes was identified by amplification and sequencing of the 16S rDNA gene and phylogenetic trees were constructed. Several members of the Roseobacter clade were dominant among the culturable isolates.  相似文献

The aim of this study was to evaluate the effects of Bacillus subtilis and Bacillus licheniformis on growth, gut microbiota, and digestive enzyme activities of Artemia urmiana. Three diets containing 102 (T1), 104 (T2), 106 (T3) CFU of probiotics/g feed, and a control diet (C) without probiotic were used through a completely randomized design (treatments with triplicates). Twelve plastic tanks with the capacity of 60- l and density of 20 nauplii/ml were used and the trial continued for 15?days. Results showed that probiotics significantly increased the total length of A. urmiana (P?<?0.05). Although the total aerobic gastrointestinal bacteria count showed no significant differences among the treatments, the total Bacillus count significantly increased in experiments (P?<?0.05). The ratio of TCBS to total aerobic bacteria count was significantly lower in T1 (0.31?±?0.05), T2 (0.27?±?0.15), and T3 (0.25?±?0.05) compared to the control (0.76?±?0.34) (P?<?0.05). The probiotics were able to increase the protease and amylase activities (P?<?0.05). No significant effect on lipase activity. The study determined T2 and T3 as the most effective treatments for improving growth, bacterial flora, and digestive enzyme activities. As less probiotic needed in T2, using 104 bacteria per g diet is recommended for rearing Artemia up to the maturity stage.  相似文献

The effect of Ultraviolet radiation type C (UV-C) radiation (0.100 ± 0.010 J/cm2) on shelf life of Arapaima gigas fillets stored at 4 ± 1°C for 18 days was investigated. The samples were analyzed for total aerobic mesophilic bacteria (TAMB) counts; total aerobic psychrotrophic bacteria (TAPB) counts; Enterobacteriaceae; purge loss; pH; lipid oxidation; total volatile bases (TVB-N); ammonia; biogenic amines (BAs); and L*, a*, and b* values. UV-C radiation increased (< 0.05) the generation time of all bacterial groups evaluated, increasing the shelf life by 2 days. UV-C-treated fillets showed lower (< 0.05) L* values, purge loss, pH, TVB-N, ammonia, and BAs levels than their control counterparts throughout the storage period. Moreover, UV-C treatment did not result (P > 0.05) in detrimental effects on lipid oxidation or a* and b* values. UV-C at 0.100 J/cm2 demonstrated a good potential for use in A. gigas fillets and, therefore, it could be applied at industrial scale.  相似文献

The probiotic activity of 15 bacterial isolates that inhibit Saprolegnia parasitica in vitro was tested for the biocontrol of saprolegniosis in rainbow trout (Oncorhynchus mykiss Walbaum), adding the bacteria to tank water for 14 days at a concentration of 106 bacteria ml?1 water. Pseudomonas fluorescens LE89 and Pseudomonas fluorescens LE141 were effective in controlling experimental infection with S. parasitica since of the fish treated with LE89, 24.5% ± 16.27% (p < 0.05) became infected, as did 42.8% ± 8.41% (p < 0.05) of those treated with LE141. Given their protective effect when administered in water, their effect was also studied when administered in feed before and after experimental infection. Both bacterial isolates survived low pH levels and the action of bile, grew in skin and intestinal mucus, were resistant to several antibiotics and survived in feed; however, neither of the two isolates prevented S. parasitica infection when administered in feed.  相似文献

The antibacterial activities of essential oils and herbal extracts have been demonstrated against a range of bacterial species. In this study, the antibacterial effects of a new combination of essential oils from the herbs Thymus vulgaris, Salvia officinalis, Eucalyptus globulus and Mentha piperita were assayed against common bacterial isolates (Staphylococcus aureus, Escherichia coli and Pseudomonas aeroginosa) and fifteen novel marine bacteria isolates. The sensitivity of different isolates to antibacterial activity of the essential oils was determined using well diffusion assays. Minimum inhibitory concentrations (MIC) and minimum bactericidal concentrations (MBC) were quantified by micro dilution assays. The data demonstrate that the combination of essential oils had potent antibacterial effects and marine bacteria were more sensitive to growth inhibition (P < 0.05). MIC rates were 0.77–6.18 mg/ml, and MBC rates were 1.67–12.30 mg/ml. This indicates that the combined essential oils (CEO) can be a new source of antibacterial agents for use in marine aquaculture.  相似文献

This study investigated postmortem changes of yellowfin tuna (Thunnus albacares) and water migration upon storage at 0°C and 4°C as determined by low-field nuclear magnetic resonance (LF-NMR 1H). It clearly showed that with increasing storage time, sensory scores and physical evaluations (hardness, chewiness), proton density of the water, and moisture content decreased, while total volatile basic nitrogen (TVB-N), K value, aerobic plate counts (APCs), and psychrotrophic bacterial counts increased. The results of LF-NMR showed that T21 (trapped water) had extremely significant correlations with sensory evaluation, texture profile analysis, TVB-N, K value, APC, and psychrotrophic bacteria (< 0.01) and established multiple linear regression. It was concluded that the LF-NMR was an important technique to evaluate storage time and temperature of yellowfin tuna’s quality in terms of water migration during low-temperature storage.  相似文献

This study investigated the effects of manipulating carbon–nitrogen (C/N) ratio and fish stocking density on pond productivity: total heterotrophic bacteria counts, plankton biovolume and benthic macro‐invertebrates. Labeo victorianus juveniles were reared for 72 days in 18 hapas suspended in six ponds measuring 150 m2 at densities of 10, 15 and 25 fish m?2. Fish in hapas received a locally formulated and prepared feed containing 295 g kg?1 crude protein, and ponds were treated with a C/N ratio of either 10 or 20. All treatments were carried out in triplicate. Increasing C/N ratio from 10 to 20 increased phytoplankton by 13% and zooplankton biovolume by 25% in the water column (P < 0.001). Total benthic macro‐invertebrates biovolumes were also 30% higher (P < 0.05) with a C/N ratio of 20 compared to 10. Total heterotrophic bacteria counts increased both in water and sediment by 29% while net yield increased by 15% from 1534 (C/N 10) to 1821 (C/N 20) kg ha?1 72 day?1. C/N ratio of 20 and a stocking density of 25 fish m?2 led to the highest yield, survival, production and net benefits. It is suggested that polyculture may lead to better utilization of pond communities to further improve pond productivity.  相似文献

The bacterial flora of the rearing pond water and sediment as well as the gills and intestine of healthy hybrid tilapia cultured in Saudi Arabia was estimated quantitatively and qualitatively, the isolates being identified at genus or species level. Total viable counts of bacteria (measured as colony‐forming units, cfu) were in the range 5.6 ± 0.8 × 103 to 2.4 ± 1.2 × 104 cfu mL?1 in pond water; 9.3 ± 1.1 × 106 to 1.9 ± 1.5 × 108 cfu g?1 in sediment; 7.1 ± 0.7 × 105 to 8.7 ± 1.1 × 106 cfu g?1 in the gills of tilapia; and 3.4 ± 1.8 × 106 to 5.8 ± 0.4 × 107 cfu g?1 in the intestine of tilapia. In total, 15 bacterial genera and 18 species were identified. Pond water and sediment bacteria reflected the bacterial composition in the gills and intestine of tilapia. In contrast to gill bacteria, more diversification was observed in intestinal bacteria. Corynebacterium urealyticum, Shewanella putrefaciens and Aeromonas hydrophila predominated in all samples. In pond water, C. urealyticurn, S. putrefaciens, A. hydrophila, Flavobacterium sp. and Pseudomonas sp. were the most predominant bacterial species (prevalence > 10%), whereas A. hydrophila, C. urealyticum, S. putrefaciens and Escherichia coli were predominant in pond sediment, and C. urealyticum, S. putrefaciens and A. hydrophila were predominant in both the gills and intestine of tilapia.  相似文献

A 20‐week feeding trial was conducted to measure growth, nutrient utilization and faecal/gut bacterial counts in triplicate groups of red hybrid tilapia, Oreochromis sp., when fed diets supplemented with 0.5% organic acids blend (OAB), 1.0% OAB, 0.5% oxytetracycline (OTC) or a control diet (no additives). At the end of the feeding trial, tilapia were challenged with Streptococcus agalactiae for 22 days. Fish fed the OTC diet had significantly higher (P < 0.05) growth than the control treatment, while growth between fish fed the OTC or OAB diets was not significantly different (P > 0.05). Phosphorus, dry matter and ash digestibility were significantly higher in the 1.0% OAB diet than the control diet. Fish fed the OAB diets had significantly lower colony‐forming units of adherent gut bacteria compared to the control or OTC treatments while those fed the 1.0% OAB diet had the lowest total faecal bacterial counts. Tilapia fed the 0.5% OTC or OAB diet had significantly higher resistance to S. agalactiae than those fed the control diet. This study indicates that dietary organic acids can potentially replace OTC as a growth promoter and antimicrobial in tilapia feeds.  相似文献

An assay for the simultaneous detection of three major bacterial pathogens of Atlantic cod with multiplex polymerase chain reaction (mPCR) was developed. Primers targeting the flanking regions of genes groEL, gyrB and amiB were standardized to diagnose francisellosis, furunculosis and vibriosis respectively. The detection limit of Francisella piscicida, Aeromonas salmonicida and Vibrio anguillarum in the mPCR assay ranged from 10 ng to 100 pg of bacterial DNA mL?1, 10 μg to 50 ng of bacterial DNA mL?1 and 1 μg to 100 ng of bacterial DNA mL?1 respectively. These primers were found to be specific in the determination of the respective genes in the individual pathogen without cross‐reaction, thus making the assay an efficient tool to detect the presence of these pathogens simultaneously or individually.  相似文献

Strategies for sampling sediment bacteria were examined in intensive shrimp. Penaeus monodon (Fabririus), ponds in tropical Australia. Stratified sampling of bacteria at the end of the production season showed that the pond centre, containing flocculated sludge, had significantly higher bacterial counts (15.5 × 109 g-1 dw) than the pond periphery (8.1 × 109g?1 dw), where the action of aerators had swept the pond floor. The variation in bacterial counts between these two zones within a pond was higher than that between sites within each zone or between ponds. Therefore, sampling effort should be focused within these zones; for example, sampling two ponds at six locations within each of the two zones resulted in a coefficient of variation of ± 5%. Bacterial numbers in the sediment were highly correlated with sediment grain size, probably because eroded soil particles and organic waste both accumulated in the centre of the pond. Despite high inputs of organic matter added to the ponds, principally as pelleted feeds, the mean bacterial numbers and nutrient concentrations (i.e. organic carbon, nitrogen and phosphorus) in the sediment were similar to those found in mangrove sediments. This suggests that bacteria are rapidly remineralizing particulates into soluble compounds. Bacterial numbers were highly correlated with organic carbon and total kjeldahl nitrogen in the sediment, suggesting that these were limiting factors to bacterial growth.  相似文献

The objective of these experimental trials was to determine the effect of a mannan oligosaccharide (MOS) derived from the outer cell wall of Saccharomyces cerevisiae strain 1026 on the growth performance and immune status of rainbow trout. Two experiments were conducted, one with eight net cages and the other with eight raceways. The net cage experiment (42 days) involved 14,400 fish with an initial average weight of 30 g. The raceway experiment (90 days) was conduced with 40,000 fish with an initial average weight of 101 g. Both experiments compared a commercial extruded diet with and without 2,000 ppm MOS supplementation. The calculated daily feed was supplied in six equal rations. Body weight, feed intake, and mortality were recorded and samples were taken for analysis of indicators of immune status. All data were subjected to ANOVA, with a net cage or raceway regarded as an experimental unit. Significantly improved performance and immune status were observed in the net cage trial—improved weight gain of 13.7% (P < 0.01), reduced feed conversion ratio (FCR) (P < 0.05), reduced mortality (P < 0.01), and improved indicators of immune status (P < 0.01) for fish fed the MOS supplement compared with controls. Similar significantly improved performance was observed for the MOS-treated groups in the raceway trial—9.97% improved weight gain (P < 0.01), lower FCR (P < 0.01), and reduced mortality compared with the control treatment. In the raceway trials, however, only the indicators of immune status lysozyme concentration, APCA, and CPCA were significantly improved by MOS treatment (P < 0.05). These experimental trials demonstrated the ability of MOS to improve the growth performance, survival, and immune status of rainbow trout produced in net cages or raceways.  相似文献

The everted gut sac technique has been used to investigate the effect of Vibrio vulnificus on water and electrolyte (Na+, K+, Cl, HCO3 ) transport on the intestine of sea bream (Sparus aurata L.). Both the anterior and the posterior intestine were incubated in a medium containing 108 V. vulnificus cells ml−1 at 25°C for 2 h. The presence of V. vulnificus resulted in a significant reduction (P < 0.05) of water absorption in the anterior intestine, while sodium absorption in the anterior (P < 0.01) and posterior (P < 0.05) intestine was elevated. Chloride absorption was increased, but the changed was not significant, while potassium absorption decreased significantly (P < 0.05), but only in the posterior intestine. Incubation the sea bream intestine with V. vulnificus did not affect carbonate secretion in the anterior segment, whereas high secretion was stimulated in the posterior segment (P < 0.01). Histological evaluations demonstrated damage in the anterior intestine of sea bream that was characterized by the detachment of degenerative enterocytes, alterations in the microvilli, and the presence of a heterogenous cell population, indicating inflammation. Based on our results, we conclude that V. vulnificus caused cell damage to the intestine of sea bream and that the anterior intestine is more susceptible than the posterior part of the intestine. Several hypotheses are suggested to explain our observations, such as the presence of higher numbers of villosities in the anterior intestine than in the posterior one and/or the presence of endogenous bacteria in the posterior intestine which may have a protector role.  相似文献
