The Sydney rock oyster breeding programme began in 1990 and initially focused on faster growth and resistance to winter mortality disease (WM, Bonamia roughleyi) before including QX disease (QX, Marteilia sydneyi) resistance in 1997. Four generations of oysters have now been exposed to these diseases at three sites in the Georges River, NSW, Australia (Lime Kiln Bar, LKB; Woolooware Bay, WB; and, Quibray Bay, QB). Non‐selected control oysters and lines developed at each site (Line 1 – LKB, Line 2 – WB, Line 3 – QB) were placed at the three sites alongside a new QX‐resistant line (Line 4) bred from QX survivors in other NSW estuaries. Line 1 oysters grew to a marketable size (>50 g) in two years with minimal losses following QX disease outbreaks: 28% mortality compared with 97% in controls. Losses in Line 3 oysters selected for WM resistance at QB were reduced by more than half (23% versus 52%). Line 2 performed best at WB, but this line showed excellent resistance to QX at LKB and WM at QB, indicating that breeding for resistance to both diseases is effective. However, selection of oysters for QX did not confer resistance to WM and vice versa.  相似文献

QX disease causes mass mortalities among Sydney rock oysters (Saccostrea glomerata). To overcome commercial production losses, Industry & Investment NSW has been developing mass selected QX disease‐resistant breeding lines since 1997. This breeding programme has significantly reduced QX‐associated mortality in the Lime Kiln Bar (LKB) breeding line relative to non‐selected, wild‐type (WT) oysters. The current study assessed mortality in families produced by single‐pair mating between LKB and WT oysters. When these families were grown in a QX disease‐prone area, the progeny of LKB × LKB crosses had significantly lower mortality compared with LKB × WT or WT × WT families. Mortality in the different crosses was associated with infection by sporulating Marteilia sydneyi, the parasite responsible for QX disease. Overall, the study identified a strong association between parentage and mortality resulting from QX disease.  相似文献

In 1990, NSW Fisheries initiated a mass selection programme in Port Stephens, NSW, with the aim of breeding faster growing Sydney rock oysters Saccostrea glomerata (Gould 1850). After two generations of selection, an average weight for age advantage of 18% (range 14–23% per breeding line) was achieved. This equates to a reduction of 3 months in the time taken to reach market size. Experiments are planned to determine how much of this 3 months advantage is additive to the 6 months advantage this laboratory has already obtained using triploid S. glomerata. A parallel set of S. glomerata breeding lines was established on the Georges River, NSW, to include selection for resistance to the protistan parasite Mikrocytos roughleyi, the causal agent of winter mortality. The programme was disrupted by the outbreak of QX disease Marteilia sydneyi, another protistan parasite, in 1994. In 1997, the breeding programme was reorganized and expanded. New lines were bred from oysters that had survived both QX and winter mortality. After one generation, a small improvement in resistance to QX has been recorded; however, the exposure of further generations to M. sydneyi will be required to confirm an increase in resistance.  相似文献

A breeding program for Sydney rock oysters Saccostrea glomerata (Gould, 1850) has been selecting oysters for resistance to QX disease (Marteilia sydneyi) and winter mortality (Bonamia roughleyi) for three generations at three sites in Georges River, New South Wales, Australia. The experimental sites are located at the upper, middle and lower reaches of oyster growing areas in the estuary. QX disease mainly occurs in the middle and upper reaches and is most severe at the latter. Winter mortality on the other hand occurs mainly at the lower and middle reaches and is most severe at the former. Progeny of third‐generation Sydney rock oyster breeding lines were evaluated for resistance to both QX disease and winter mortality against a non‐selected control. Line 1, selected for QX disease resistance at the upper estuary site, had excellent resistance to one season of exposure to disease, but suffered high mortality during the second season of exposure. However, these oysters had already reached market size of 50 g whole weight, with low mortality at 2 years of age, before the second episode of QX disease. Line 2 showed good improvement in resistance to both diseases, whereas Line 3, was the most resistant to winter mortality. Selection for resistance to QX disease did not appear to confer resistance to winter mortality and the converse also applied.  相似文献

The black‐lipped pearl oyster, Pinctada margaritifera, is the most important farmed species in French Polynesia and the basis of the most valuable export industry. Mass production of black pearls relies on a surgical operation requiring tissue from a donor pearl oyster to be grafted, together with a nucleus made of shell, into the gonad of a recipient oyster. Improving pearl size through family selection remains one of the main challenges for future aquaculture development. This study analyses the relative contribution of donor and recipient oysters to pearl size. To this end, hatchery‐produced donor oysters of two batches, large and small (based on shell height), were used to supply grafts for recipients, which were then monitored individually for their growth performance by recording shell height, width, and thickness, and total live weight (flesh + shells) every 6 months (four biometric measurement times) over 20 months of culture. Pearls issued from the two batches of donors showed no significant differences in nacre weight or thickness. In contrast, recipient oyster shell height and total weight were increasingly positively correlated with these pearl size parameters over the culture period, becoming significant at 8 months post‐grafting. Potential therefore exists to use shell height and oyster weight as phenotypic indicators for selective breeding of recipient oysters with high growth performance to increase pearl size in P. margaritifera.  相似文献

The level and extent of linkage disequilibrium (LD) and effective population size (Ne) were studied in three selected lines and a wild population of Pacific oyster, Crassostrea gigas, using 61 single‐nucleotide polymorphisms. Significant differences were detected in the average r2 between the selected lines and wild population for both syntenic and nonsyntenic loci with LD beyond population‐specific critical values (P<0.05). Moreover, the proportions of syntenic and nonsyntenic loci with the expected LD level in the wild population were lower than that in the selected lines. Taken together, the LD level of the selected lines was higher than that of the wild population. The extent of LD analysis showed that a short range of LD (0–0.23 cM) was detected in the four populations, and the decay distance was lower in the wild population than in the selected lines. Ne values ranged from 47.6 to 58.5 in the selected lines and ranged from 527.9 to 709.6 with infinite upper limits in the wild population. Further variance analysis of LD demonstrated that genetic drift and epistatic selection might account for the increased LD levels in selected lines. The LD information will be valuable for further association study and marker‐assisted selection in oysters.  相似文献

The progeny of four breeding lines of Sydney rock oysters, Saccostrea commercialis, selected for faster growth over two generations, were compared with controls in an 18-month farming experiment. Mean whole weights of oysters from all four selection lines did not differ significantly from each other but, were significantly greater (P<0.05) than mean whole weight of controls (spat produced from two groups of non-selected oyster populations). The mean increase in weight for the selection lines over the controls of 18% (range 14–23%) after two generations of selection (third generation) compares extremely favourably with the 4% reported previously after one generation of selection.  相似文献

Growth is an important aquaculture trait, which has been one of the most important traits for genetic improvement in aquatic species. Numerous fast‐growing domesticated aquaculture organisms are being cultured, but the physiological processes underlying growth remain largely unexplored. The Pacific oyster (Crassostrea gigas) is one of the most widely farmed bivalves. In order to improve the productivity traits, a fast‐growing selective strain of C. gigas was established. In this study, we compared physiological energetics of the fast‐growing selective strain and an unselected hatchery population of C. gigas under different conditions of temperature. Components of energy gain (clearance rates, ingestion rates, absorption efficiency and absorption rates), energy loss (metabolic rates) and resulting energy budget (scope for growth) were analysed at four temperatures (15, 20, 25 and 30°C). The results indicated that selective breeding oysters had a higher energy gain compared to unselected oysters, but the selected strain and hatchery population of oysters did not significantly differ in metabolic rates at all tested temperatures (> 0.05). The enhanced energy gain resulted in almost two times higher scope for growth in selective breeding oysters compared with unselected oysters. Physiological characteristics of C. gigas were greatly affected by temperature and showed a fluctuation with the increase in temperatures. The selective breeding oysters outperformed those of hatchery population in growth performance by an increased energy gain along with a similar energy loss. This study provided insight into physiological energetics accounting for growth improvements in fast‐growing selective strain of C. gigas.  相似文献

为比较企鹅珍珠贝[(Röding,1798)]海南金唇选育系(GLHH)、海南白唇选育系(WLHH)、广西金唇选育系(GLGG)和广西白唇选育系(WLGG)的生长存活差异,及了解4个选育系的生长规律,自2018年1月底至2018年12月底连续监测养殖于海南陵水黎族自治县新村港的4个企鹅珍珠贝选育系F3的存活率及其壳长、壳高、壳宽、铰合线长和体重的生长情况。使用单因素方差分析和生长模型拟合等方法对数据进行分析。结果显示,4个企鹅珍珠贝选育系的生长及存活率存在显著差异(<0.05),海南金唇选育系的生长及存活率最优,广西白唇选育系的生长及存活率最差,表明企鹅珍珠贝的生长存活可能与环境、贝龄、壳色和地理种群有关。SGompertz生长模型为企鹅珍珠贝5个生长性状的通用生长模型。研究表明,4个企鹅珍珠贝选育系的生长规律存在差异,且企鹅珍珠贝5个生长性状的生长规律存在差异,其中壳长和铰合线长的生长拐点出现得最早,分别出现在7.9(GLHH)~8.5(WLHH)月龄和7.9(GLHH)~9.1(WLGG)月龄;其次是壳高和壳宽,生长拐点分别出现在11.6(GLHH)~15.9(WLGG)月龄和11.8(GLHH)~14.3(WLGG)月龄时;体重的生长拐点出现得最晚,在15.8(GLHH)~18.7(WLGG)月龄。该研究结果表明海南金唇选育系早期生长阶段具有高存活率和优良的生长性能,广西白唇选育系可能具有最大的生长潜力(生长性状的生长极限最大),本研究可为企鹅珍珠贝的良种选育、健康养殖及优质珍珠的生产提供借鉴。  相似文献

为了评价红、金黄、白和黑壳色系F3育珠性状,设立两个实验,分别利用马氏珠母贝红、金黄、白和黑壳色系F3作为植核(受体)贝和小片(供体)贝进行植核育珠。实验Ⅰ:利用红、金黄、白和黑壳色系F3为插核贝和小片贝建立16个组合,另外利用以普通养殖群体为插核贝、金黄壳色系F3为小片贝建立了1个组合,共建立17个组合,育珠期为24个月;实验Ⅱ:分别利用红、金黄、白和黑壳色系F3为植核贝和小片贝建立16个组合,育珠期为18个月。结果表明,育珠期结束后实验Ⅰ和Ⅱ的4个壳色系F3平均壳高和成活率均存在显著性差异(P<0.05),实验Ⅰ和Ⅱ的黑壳色系F3具有最大的平均壳高,实验Ⅰ金黄壳色系F3具有最高的成活率,实验Ⅱ黑壳色系F3具有最高的成活率。实验Ⅰ:各组合间的留核率、商品珠率、优良珠率和育珠绩效均存在显著性差异(P<0.05),珍珠层厚度差异不显著(P>0.05);BW组具有最大的留核率、商品珠率、珠层厚度和育珠绩效值,RB组具有最大的优良珠率;实验Ⅱ:各组合的留核率、商品珠率、优良珠率、珍珠层厚度和育珠绩效均存在显著性差异(P<0.05)。黑壳色系F3作为插核贝具有较好的育珠效果,经进一步测试其育珠性能,可在珍珠生产中推广应用。  相似文献

This paper provides a review and summary of strain comparison and selective breeding work carried out on redclaw crayfish Cherax quadricarinatus (von Martens) at the Queensland Department of Primary Industries, Freshwater Fisheries and Aquaculture Centre, Walkamin. Redclaw crayfish are a highly marketable, environmentally tolerant, freshwater crayfish with a moderately fast, but variable, growth rate. Five strains of the species were assessed morphologically and in terms of age at maturity. A non‐replicated production trial was also carried out before two strains, from the Gilbert and Flinders rivers in North Queensland, were chosen to be part of an experimental selective breeding programme for improved growth. Initially, 14 families of each strain were randomly mated and grown out. At this point, a size‐related selection took place, with the largest animals from each family forming a selected line, and individuals from around the mean forming a control line. Within‐family selection and reciprocal mating between families was performed. Data were collected after each of two generations of growout and analysed by analysis of variance. Significant differences (P < 0.05) were measured between strains, sexes and between selected and control lines. Selected individuals grew 9.5% faster than the controls. Recommendations for selective breeding research involving freshwater crayfish are presented.  相似文献

The eastern oyster Crassostrea virginica can change sex which makes self‐fertilization possible if sperm can be cryopreserved. In this study, small (~1 year old) and large (~2–3 years old) oysters were biopsied for sperm collection. Survival of the biopsied oysters after 1 year was 50% for small oysters and 17% for large oysters. Oocytes were collected from sex‐reversed females, and self‐fertilized with cryopreserved sperm. Of the 24 cryopreserved samples, 14 individuals had ≤1% fertility when crossed with oocytes from unrelated females, indicating that the cryopreserved sperm had reduced fertility. The other 10 individuals had a fertility of 39 ± 25% when crossed with oocytes from unrelated females (non‐selfing), but showed a significantly lower success of self‐fertilization (12 ± 16%) (P = 0.008), while aliquots of the same oocytes had a fertilization of 83 ± 11% when crossing with fresh sperm. Larvae were produced at day 3 in the self‐fertilized families (12–94% of the fertilized oocytes), and survived to eyed‐larvae stage at days 11–14. Genotyping with 9 microsatellite markers confirmed that the larvae resulted from self‐fertilization in four families. This study demonstrated the feasibility of creating self‐fertilized inbred lines of oysters by use of non‐lethal sperm collection and cryopreservation.  相似文献

Selective breeding efforts have yielded oyster strains, Crassostrea virginica, with improved survival and resistance against Haplosporidium nelsoni (MSX); however, because of susceptibility to the oyster pathogen Perkinsus marinus (Dermo), their utility has been limited in areas where the two parasites co-occur. Dual resistance to H. nelsoni and P. marinus was achieved through four generations of artificial selection of wild Delaware Bay oyster progeny at a site in the lower York River, Virginia, USA where both diseases are enzootic. During 1993–1995, survival, growth, and disease susceptibility of third generation Delaware Bay (F3-DEBY) oysters were evaluated at the York River site in comparison to that of similarly selected third generation James River, Virginia oysters (F3-JR), and first generation Louisiana oysters (F1-LA), whose parents were naturally selected in the wild for resistance to P. marinus. During 1997–1999, the performance of F4-DEBY was evaluated at three sites in Virginia in comparison to two groups of first generation oysters whose parents originated from Mobjack Bay and Tangier Sound, Virginia where both P. marinus and H. nelsoni are enzootic. In the presence of high infection pressure from both H. nelsoni and P. marinus, the F3-DEBY stock showed significantly higher survival and growth than either the F3-JR or F1-LA strain. After 15 months of deployment, 79% of F3-DEBY, 11% of F3-JR and 17% of F1-LA oysters were market size (≥76.2 mm) and cumulative mortality was only 16% in F3-DEBY as compared to 42% in F3-JR and F1-LA. At the termination of the study, F3-DEBY oysters exhibited 22% lower mortality than the F1-LA stock, which outperformed the F3-JR stock. Relative performance in respect to disease varied considerably with sample date; however, average H. nelsoni weighted prevalence varied such that F3-DEBY1-LA=F3-JR and average P. marinus weighted prevalence varied such that F1-LA3-DEBY3-JR. In the 1997–1999 trial, F4-DEBY oysters experienced 34–61% lower mortality, greater growth rate, and consistently lower prevalence and intensity of P. marinus than either Mobjack Bay or Tangier Sound oysters. H. nelsoni prevalences were very low (<12%) in all three stocks. This is the first study to demonstrate that reduced susceptibility to both P. marinus and H. nelsoni can be achieved through selective breeding.  相似文献

The Australian edible oyster industry has been severely impacted by disease and declining yields since the 1970s. Selective breeding of Saccostrea glomerata is one measure addressing these problems by producing fast‐growing, disease‐resistant oysters. Farmers report that selected oysters have different growth characteristics than their wild counterparts using conventional grow‐out methods. This study investigated how different grow‐out methods influence commercially valuable oyster characteristics including shell length, shape, surface growth deformities and meat condition. In June 2015, selectively bred S. glomerata spat were deployed in two estuaries (Hawkesbury River and Georges River) in NSW, Australia, using three grow‐out methods (fixed trays, Stanway cylinders and floating baskets). In November 2015, oysters were transferred among grow‐out methods to test for the effects of changing grow‐out methods on oyster growth patterns. Oysters transferred from baskets to cylinders and from trays to cylinders had, on average, deeper and wider shells, a higher meat condition and fewer shell surface deformities than oysters in other grow‐out method combinations. However, these oysters were smaller than oysters not grown in cylinders. While there were some differences in growth patterns between the estuaries, overall it was the grow‐out methods that most influenced oyster characteristics. This was attributed to differences in the amount and magnitude of movement oysters experienced in the grow‐out methods, as recorded by motion sensors. This study demonstrates how grow‐out methods can be managed to achieve desired growth trajectories and therefore improve marketability among selective bred S. glomerata.  相似文献

In order to study individual growth variability and its relationship with survival in juvenile Crassostrea gigas, parental oysters were sampled at four sites located along the French Atlantic coast and bred under controlled hatchery conditions. Four groups of larvae were obtained by crossing five males and five females from each of the four sites, and a fifth group by crossing these 20 males and 20 females together in a pool. Larvae were reared under conditions allowing the maintenance of a maximum variability of size and gave five experimental groups. Oysters were individually monitored for growth and survival from 3 to 10 months after fertilization. The individual growth performances were relatively stable over time and no noticeable compensation for growth occurred. Early growth rate was a very good predictor of size later in life: 66% of variation in the live weight at 10 months could be explained by variation in the initial growth rate calculated between 3 and 4 months. A significant group effect was observed on survival and on growth rate. Mortality mostly occurred between 3 and 5 months and appeared to affect the slow-growing animals more. However, two groups can be distinguished among those which died during the experimental period, one which showed a decrease in weight and the other whose growth was similar to surviving oysters. These results are discussed in the light of usual oyster farming practices and selective breeding. This work aimed to evaluate the whole weight (g), shell height (mm), mortality (%) and reproductive aspects (condition index and histology) of triploid (3N) Pacific oysters (Crassostrea gigas) cultivated in southern Brazil. To accomplish this, a comparative experiment was performed with a diploid (2N) control group in two cultivation sites on Santa Catarina Island: North Bay (NB) and South Bay (SB). Whole weight, height and cumulative mortality were evaluated monthly for eight months and condition index (CI) for six months. Histological analysis was performed every two weeks for four months to investigate reproductive aspects. At the end of the experiment, whole weight and height were similar for 3N and 2N oysters. However, while more growth in whole weight and height was observed in the initial phase of cultivation in NB, such growth was not detected in SB until intermediate and final grow‐out phase. At harvest, cumulative mortality in 3N oysters was similar that in 2N oysters. The results suggest that diploid and triploid oysters grown in southern Brazil have similar developmental patterns based on whole weight and height, as well as reproductive cycle. In addition, cultivation sites have more influence on growth and mortality than ploidy levels.  相似文献

We studied the effect of strain and selection lines on the fecundity of the cyclopoid copepod Paracyclopina nana. We used six strains (HL, YL, BS, SL, TH, TS) and six isofemale lines (including three higher and three lower fecundity lines). As for the selection of strain, total nauplii production per female of SL strain was the highest in the individual culture (231 nauplii per female) and population growth rate in HL and SL strains showed higher values than the others in combined culture. For the selection of line, the total nauplii production per female of HF2‐2 (high fecundity and second generation) line was the highest (302 nauplii per female) but those of LF2‐1 (low fecundity) (80 nauplii per female), and LF2‐3 (46 nauplii per female) lines were lower than the others. In F4 generation, three selected HF2‐2 line showed that the total nauplii production were higher than those of three selected LF2‐3 lines. In the combined culture on the different selection lines (F8), population growth rate of two selected high fecundity lines showed higher values than that of the non‐selection group and selected low fecundity line. Our results demonstrate that the production of P. nana can be improved by selection of the line with high fecundity.  相似文献

The common pearl oyster, Pinctada fucata (Gould), is the most important species that is cultured for production of marine pearls in China. Heavy mortality and the decline of pearl quality have resulted in a breeding programme being established in recent years. In this study, we conducted selective breeding for the second generation of pearl oyster P. fucata (JCS-2) by mass selection for shell height (SH) with a selection intensity of 1.614, and analysed the growth of the selected line (JCS-2) and the non-selected control line (JCC) during a 1-year grow-out period. The results show that the selected line grew faster than the control one in the SH and total weight (TW) ( P <0.05), and there were higher proportion of larger sized oysters. Coefficient of variation for SH of JCS-2 was smaller than that of JCC. The current genetic gains and realized heritability for JCS-2 averaged 16.03 ± 4.79% and 0.713 ± 0.208 at 3–15 months of age respectively. The findings indicated the selection response to faster growth for SH is markedly effective in the second generation, and there was a high correlated response of TW when selecting for SH.  相似文献

Black‐lip pearl oyster, Pinctada margaritifera broodstock was collected from the wild. Egg production, hatching rate and larval development were compared between oysters induced to spawn within 2 days after collection in the wild (T1), oysters fed a pure microalgae diet during 24 days before spawning (T2) and oysters fed the same microalgal diet in which 10% of the algae were replaced with 2 μm polyunsaturated fatty acid (PUFA)‐rich microspheres (T3). Administration of lipid microspheres resulted in larger sized eggs, a higher percentage of D‐larvae and larger sized 48‐h‐old larvae (P<0.05). The total and neutral lipid contents of the gonad increased after oysters were fed with microalgae only or with supplementary diet. The major neutral and polar fractions of saturated fatty acid (SFA) were 16C and 18C fatty acids, and not influenced by the diet (P>0.05). The gonads of oysters fed supplementary PUFA contained more docosahexaenoic acid (DHA) and less monounsaturated fatty acids. Higher level of DHA in gonads of T3 was associated with oogenesis and embryogenesis success. The n‐3/n‐6 ratio in the neutral lipid fraction provides a good indication of the spawning condition and predicting egg size and hatching rate.  相似文献

Two control (C1 or first control generation, and C4 or fourth control generation) and three selected (S1 or first selected generation, S2 or second selected generation, S4 or fourth selected generation) stocks of Chitralada Nile tilapia were analysed for microsatellite variation to determine the effect of size‐specific mass selection on genetic variability. Genetic variation based on five microsatellite loci (UNH123, UNH147, UNH172, UNH222 and UNH216) showed a slightly higher allelic diversity in the selected stocks (7.4–10 alleles) than in the control stocks (6.8–8.8 alleles). Apparent reductions in the mean number of alleles and He values were noted in successive generations of both control and selected lines. Significant deviations from Hardy–Weinberg equilibrium because of an excess of homozygotes indicated inbreeding in all control and selected stocks. Although estimated inbreeding levels were not significantly different among selected and control lines based on Welch's t‐tests, the increase in the degree of inbreeding within the selected line was higher (107.9%) than the control line (64.2%) after four generations. The implications of these results on the management and conservation of genetic diversity in improved breeds are discussed, while the importance of monitoring and minimizing inbreeding are likewise emphasized.  相似文献
