These studies were conducted to evaluate the efficacy of a live attenuated Edwardsiella ictaluri vaccine against enteric septicemia of catfish. In one study channel catfish fingerlings (72 d of age post hatch) were immersed for 30 min in water containing E. ictaluri RE-33 at dosages of 1 × 106, 1 × 107 and 2 × 107 CFU/ML of water. No mortalities were observed following vaccination. Following exposure to virulent Edwardsiella ictaluri the cumulative mortality of fish vaccinated with dosages of at least 1 × 107 CFU/mL were significantly lower than that of non-vacccinated fish in both laboratory and field challenges. Vaccination with 1 × 106 CFU RE-33mL provided some protection during the laboratory challenge but failed to protect fish under field conditions. In a second study, vaccination of 6 full-sib families of channel catfish at a vaccine dosage of 1 × 107 CFU/mL resulted in a relative percent survival among families ranging from 67.1 to 100%. Significant differences in mortality were found among families and between vaccinated and unvaccinated groups, but there was no family by vaccine interaction. Families with the highest mortality after vaccination were also shown to have the highest mortality without vaccination (r = 0.82; P = 0.04).  相似文献

Enteric septicemia of cattish (ESC), caused by the bacterium Edwardsiella ictaluri, has become the most significant disease problem affecting the commercial channel catfish, Ictalurus punctatus, industry in the United States. Although antibiotics are used extensively for the control of ESC, there are inherent problems associated with their use. Consequently, experiments were initiated to evaluate the effectiveness of vaccination program that used immersion and oral delivery methods to administer a killed E. ictaluri vaccine to fry and fingerling channel cafish. In a preliminary pond study with laboratory challenge, mortality in a group vaccinated with a combination of immersion and oral procedures was only 5.0% in both high- and low-dose challenges. This was significantly different (P c 0.01) from non-vaccinated controls, which had 46.7%mortality in the lowdose challenge and the 6 1.7% mortality in the highdose challenge. This corresponds to relative percent survival (RPS) values of 89.3 and 91.9 respectively. Subsequent field trials further indicated the efficacy of a vaccination program for the prevention of ESC in channel catfish. In 1987-1988, a field study was conducted using 12 commercial ponds, with three replicates of four treatments. The four treatments included vaccination by immersion only, oral only, a combination of both immersion and oral procedures, and non-vaccinated conwols. Relative percent survival was 57.4 for the immersion only treatment, 50.3 for the oral only treatment, and 53.5 for the combination immersion and oral treatment. In 1989-1990, no significant difference was found between vaccinated and non-vaccinated fish. However, in 1989-1990, a vaccine-oil emulsion was topcoated on a floating feed, rather than incorporating vaccine in a sinking pellet. In 1990-1991, overall mortality in vaccinated fish was significantly less (P < 0.05) than non-vaccinated fish, with 41.2% mortality in vaccinates compared to 63.5% in non-vaccinated fish, for an RPS of 35.1. In examining RPS values for individual farms, two farms had excellent results, with RPS values of 81.3 and 76.9; two farms had only moderate success, with RPS values of 26.6 and 15.4; and one location had greater mortality in the vaccinated fish than in the non-vaccinated fish. However, that farm had only two ponds in the study and experienced significant losses to proliferative gill disease in the pond with vaccinated fish.  相似文献

Abstract.— Four different vaccination regimes, including non-vaccinated controls (C), immersion/oral combination (IO). late immersion (LI), and injection (IJ), were evaluated for antibody production and protection from challenge in channel catfish Ictalurus punctatus that were hatched and raised in recirculating culture systems over a 28-wk period. For IO, fry were vaccinated by immersion on day 12 post-hatch and given an oral booster during week 10 post-hatch. The LI and IJ delivered vaccine was also administered at week 10 post-hatch. No titers were detected in the IO group prior to the oral booster at week 10. Mean titers rose sharply after the week 10 vaccinations in all treatment groups, maintained a high level for about 8 wk, and slowly declined over the next 4 wk. Injected fish had significantly higher titers during the primary response period than either LI or IO fish. There was no statistical difference in titers between LI and IO fish during the primary response. After an oral booster delivered during week 22, titers in LI and IO fish increased significantly at week 25, while titers in IJ fish declined despite the booster delivery. An immersion booster delivered during week 25 resulted in significant increases in titers in all vaccinated groups. Of multiple challenges, the only one not marred by concurrent columnaris infections revealed a low, but significant level of protection in IJ fish at 12 wk post-vaccination.  相似文献

Abstract. The pathogenicity of Renibacterium salmoninarum to carp, Cyprinus carpio L., and rainbow trout, Salmo gairdneri Richardson, was investigated. All carp injected with 4·8 × 108 cells/fish, or 4·8 × 107 cells/fish survived for 38 days. R. salmoninarum was isolated from all moribund fish, but not from the kidney of surviving fish, although R. salmoninarum antigen was detected in several of these fish by the dot blot assay. On the other hand, mortality in rainbow trout was 95% in the fish injected with 4·8 × 108 cells/fish, and 15% in those which received 4·8 × 107 cells/fish. R. salmoninarum antigen was detected by the dot blot assay in all surviving rainbow trout. The number of R. salmoninarum cells was immediately decreased by carp or rainbow trout serum, and the serum bactericidal activity of carp was higher than that of rainbow trout. Carp blood leucocytes had higher phagocytic activity than those of rainbow trout.  相似文献

Abstract. One thousand each of pond and hatchery reared 0+ brown trout, Salmo trutta L., were fin-clipped and released in a 1300-m2 large earthen pond. The pond was drawn down 5 days after the introduction, and descending individuals were caught in a trap at the outlet. A total of 904 pond-reared and 890 hatchery-reared fish were recaptured, i.e. a loss of 9·6 and 110% respectively, A total of 25 pond-reared and 16 hatchery reared fish were recorded stranded in the pond during the draw-down, accounting for 2·7 and 1·8% of total recoveries.
Pond-reared fish descended significantly earlier than did hatchery fish. Most individuals descended during the first 4·5h (75-83%). However, the final recoveries were made 10 days (233h) after the first descent.  相似文献

Abstract. During three 5-month experiments in Thailand, earthen ponds of approximately 370m2 surface area were stocked with male Nile tilapia, Oreochromis niloticus (L.), fingerlings of 4–12g weight at densities of 0·5 to l·6fish/m2. Stocking and fertilization (with chicken manure, urea and TSP) in triplicated depth treatments of 0·6,1·0 and 1·5m were proportional to pond volume in two experiments (wet and dry seasons) and to pond area in the other (dry season).
Depth showed no direct effect on fish yields of 0·9–6·3t/ha/year, on survival rates of 66 to 98%, nor on final individual weights of 96–313 g/fish. Greater yields were obtained from deeper ponds when they received proportionally greater stocking and fertilizer inputs. Inputs per unit area were the most important factor accounting for yield variation.
Temperature, dissolved N and P, and suspended solids showed little or no relation to depth treatments. Time-averaged chlorophyll concentrations and photosynthetic production of dissolved oxygen were greater in treatments receiving greater inputs of nitrogen per unit pond volume.
Deeper ponds produced the greatest areal yields of fish, when fertilized in proportion to their volumes. Shallow ponds produced fish and dissolved oxygen at least as efficiently per unit input as did deep ponds, which is consistent with models of photosynthesis-depth relations.  相似文献

Abstract. Cyprinid herpesvirus 1 (CHV) or Herpesvirus cyprini was virulent for carp, Cyprinus carpio L., fry following 1 h immersion in water at 20 °C. Cumulative mortality for carp fry was 86–97% in 2-week-old common carp, 20% in 4-week-old fancy carp, and 0% in both 8-week-old common and fancy carp. The virus did not produce mortality in fry of crucian carp, grass carp or other cyprinids. It was also oncogenic in carp, inducing papillomas to the extent of 55% among both common and fancy carp fry. The neoplasms appeared 5–6 months after carp had been exposed to the virus by immersion and recurred at an incidence of 83% in carp 7·5 months post-desquamation of the tumour. The CHV was reisolated from all moribund fish and from all survivors. It also induced papillomas at an incidence of 13% in adult mirror carp and at 10% in adult fancy carp 5 months after intraperitoneal inoculation of 105 TCID50 ml-1 fish. The virus was rcisolated only from the ncoplastic tissue and not from internal organs. The neoplasms were normally located on fin, skin or mandible, at the intraperitoneal inoculation site. Specific fluorescence for CHV antigen was frequently detected in the gills, liver, kidneys and intestine of 2-week-old fry from 3 to 21 days following challenge with CHV. It was found in greater concentrations in experimentally induced papillomata on 2-week-old carp fry survivors examined 24 weeks after challenge than in naturally occurring neoplasms.  相似文献

This study evaluated the efficiency of differently prepared vaccines against Aeromonas hydrophila in the hybrid surubim (Pseudoplatystoma corruscans × P. reticulatum). Survival and haemato‐immunological parameters were compared between the treatments: non‐vaccinated fish (C); bacterin‐vaccinated fish (B); bacterin plus oral booster vaccinated fish (B+O); bacterin and toxoid‐vaccinated fish (B+T) and bacterin, toxoid and oral booster‐vaccinated fish (B+T+O). Fourteen‐days vaccinated fish from B+O and B+T+O were fed with an oral booster for 4 days. After 1 week, the fish were intraperitoneally challenged with 2 × 108 CFU mL?1 of A. hydrophila. Fish from the treatment B+T+O showed the lowest cumulative mortality (11.36%) 96 h after challenge, compared with other treatments (22.72–44.04%), and a relative survival of 74%. Serum immunoglobulin in B+T+O fish was higher than in other treatments. All vaccinated fish showed an increased agglutination titre when compared with non‐vaccinated fish, both before and after challenge. Fish fed with oral booster showed an increase in phagocytic percentage before and after challenge. It can be inferred that the oral booster vaccination was efficient in reducing mortality in hybrid surubim by enhancing the response against haemorrhagic septicaemia due to A. hydrophila infection.  相似文献

Abstract. Viral haemorrhagic septicaemia (VHS) is reported for the first time in sea-water cultured rainbow trout. Heavy mortalities with typical signs and lesions A VHS virus (serotype 1) was isolated from the diseased fish. The mortalities were caused only by the VHS virus and 80 days post transfer of trout to sea-water the mortalities reached 85%, of the initial population.
The disease was experimentally transmitted to rainbow trout, both in sea-water 3·104 pfu/ml of virus or by intramuscular injection of various doses of VHS1 (7·101 7·104 or 7·104 pfu per fish). Death occurred in all infected groups and started earlier in sea-water. Typical signs of VHS were observed in moribund fish. Viral multiplication was demonstrated to have occurred in fish organs.  相似文献

Uptake and clearance of Edwardsiella ictaluri in the peripheral blood of channel catfish Ictalurus punctatus fingerlings were monitored for 216 h after exposure to E. ictaluri for 4 h and 8 h under static conditions. Most fish exposed to E. ictaluri developed bacteriemia 24 h post-exposure, and by 72 h post-exposure E. ictaluri was recovered from all the blood of all exposed fish. The number of E. ictaluri colony forming units (CFU) in the blood of moribund fish ranged between 1.7 × 103 to 1.6 × 105 CFU/50 μL whole blood. Clearance of bacteria from the blood was observed by 216 h post-exposure and all fish surviving bacterial exposure developed agglutinating antibody against E. ictaluri . The pathogenesis of the infection was accompanied by the shedding of viable E. ictaluri into the water which may serve as a mechanism by which fish to fish transmission occurs.  相似文献

Abstract. The effect on the growth of freshwater angelfish, Pterophyllum scalare (Lichtenstein) (Pisces; Cichlidae), of different densities of population and diet compositions was studied. The results show that increasing the population density from 50 fish/m2 to 200/m2 significantly decreased the rate of growth of fingerlings, and that this effect is correlated to the size of fish. No significant differences were observed in growth between diets containing 37%, 41% or 47% protein. However, the addition of live food (artemia) to the diet significantly raised the growth rate, especially in the higher protein diets.
Body composition shows no changes during the growth periods, with a mean of 64·68% body protein, 35·47% dry weight fat and no significant differences in the percentage of residual ash through the growth period.
The food conversion rate was very low, ranging from 1·15 to 1·31. Total production increased with density, from 4·0g/m2/day (50 fish) to 13·7g/m2/day (200 fish) over a period of 60 days.  相似文献

Bacterial cells of the marine fish pathogen Photobacterium damsela subsp. piscicida were grown in novel culture media. A mixture of whole cells and extracellular components was inactivated and used in bath, intraperitoneal (i.p.) and oral vaccination of sea bass, Dicentrarchus labrax, employing two sizes of fish. A commercial vaccine was used for comparative purposes. Control and immunized fish were either bath or intraperitoneally challenged 6 and 12 weeks post-vaccination. Small fish had significantly higher relative percentage survival with the novel vaccine mixture both at 6 and 12 weeks post-vaccination by bath, in comparison with the commercial vaccine. No protection was afforded at 6 or 12 weeks post-immunization by either vaccine after challenge via i.p. injection. Sea bass (1.5-2 g) intraperitoneally vaccinated with various adjuvanted vaccine mixtures were not protected against pasteurellosis. In contrast, larger sea bass (20 g) benefited from vaccination with the novel vaccine mixtures. Intraperitoneal challenge with the pathogen resulted in protection in both fish groups vaccinated with novel vaccine mixtures, whereas control fish suffered high mortalities (> 80%). Orally vaccinated fish were immersion challenged with the pathogen. At 6 and 12 weeks post-vaccination the control fish had a high mortality and the fish vaccinated with the novel vaccine mixture achieved good protection.  相似文献

Abstract. Growth and survival response of Penaeus stylirostris (Stimpson) to fertilization, pelleted feeding and stocking density were analysed for the conditions prevailing on the Baja California peninsula (Mexico). Three 0·25-ha earthen ponds were longitudinally divided in half with mesh structures. One half received inorganic fertilization (trial F) and the other supplementary feeding using a pelletized ration (trial PF). The initial stocking rates were approximately 5, 10 and 15 PL5/m2 in both trials. After 210 days, final mean weights ranged from 15·0 to 5·3g and survival from 48·6 to 17·8%. Growth and survival were analysed by means of a reparameterization of the von Bertalanffy's growth model and the exponential mortality equation. Results indicated that: (1) final mean weight was dependent on density in both trials ( P < 0·01) although growth rate was only affected in trial PF ( P < 0·01); (2) pelleted feeding produced both greater final mean weight and increased growth rate at densities higher than 5 PL/m2 ( P < 0·005); (3) the ratio tail weight/total weight was influenced by density in trial F ( P < 0·015); (4) the mortality equation served to explain differences in survival to a limited extent. An increase in growth rate after 170 days occurring at the intermediate density in trial PF was attributed to an increase in mortality, which allowed the remaining individuals to attain larger sizes. Further research to increase biomass yields should focus on the optimization of pond depth, the supply of vigorous post-larvae and on the possible influences of seasonal variations of planktonic communities of the lagoons on the productivity of ponds.  相似文献

Abstract. Mutants of Aeromonas salmonicida strains lacking either the A-protein, O-antigen or both of these major surface antigens were tested in rainbow trout, Oncorhynchus mykiss (Walbaum), for their suitability as live vaccines (LV). All of these mutants were shown to be attenuated, as fish receiving ∼5 × 107 of the respective strains showed no clinical signs of furunculosis. Immersion vaccination of fish in 5 × 107 cfu ml-1 of these strains with an identical immersion dose 14 days later resulted in significant protection by all strains from challenge with a heterologous virulent strain of A. salmonicida 5 weeks later. The levels of protection conferred were all greater than or equal to that provided by an injected bacterin using the same vaccination schedule. With one exception, all LV strains that still possessed a functional O-antigen provided protective indices (PI) four- to seven-fold greater than the PI for the fish injected with bacterin. When antibody responses of vaccinated fish were compared, it was found that only vaccination by bacterin gave rise to a measurable agglutinating litre. Western immunoblots using the immune fish sera failed to reveal any major differences in antigen recognition in fish that received any of the vaccines tested. These data suggest that the immune response generated by the use of live vaccine strains is different from that generated by a bacterin, and that these useful mutations may be incorporated into existing furunculosis LVs for further attenuation.  相似文献

Abstract. A 91-day cage trial was conducted with juvenile seabass, Lates calcarifer (Bloch), and grouper, Epinephelus tauvina (Forskal), to ascertain the capacity of kerosene pressure lamps and fluorescent electric lamps as night-lights above the cages to attract pelagic food organisms into the cage and consequently sustain the survival and growth of the cultured fish. The experiment was conducted using 1×1×1·5m floating cages with three different net mesh sizes (1, 13, and 19mm) and four fish stocking densities (seabass—10, 20,30 and 40/m3; grouper —10/m3). A positive growth response and survival was observed with seabass and to a lesser extent with grouper with increasing net mesh size and decreasing fish stocking density. At the lowest tested density of 10 fish/m3 seabass survival increased from 5·0 to 95·0% and total cage fish biomass increased from −95·1% to +56·9% with an increase in net mesh size from 1 to 19mm over the 91-day culture trial, respectively. The results are discussed in relation to the current commercial marine finfish cage farming practices employed in Indonesia.  相似文献

Abstract. Two different commercial vaccines against furunculosis, caused by Aeromonas salmonicida subsp. salmonicida , were tested in Atlantic salmon on seven fish farms. Both vaccines were based on formalin-inaclivated bacterins containing aluminium salts as adjuvants. The fish were vaccinated by intraperitoneal injection in the spring approximately one month prior to transfer to sea water, and they were challenged by natural outbreaks of furunculosis. During the first year, six of the farms experienced disease outbreaks. The overall mortality was 7·14% in vaccinated fish and 21·7% in unvaccinated controls, giving a relative percentage survival (RPS) of 67%. In the seventh farm, outbreaks of furunculosis more than one year after vaccination revealed that there was still a trend towards lower mortality in vaccinated fish, though the mean RPS fell to 22%. The use of adjuvants in the vaccines resulted in local lesions in the abdominal cavity of vaccinated fish. However, the severity of the lesions declined gradually, and they did not influence fish quality at the time of slaughtering. Vaccination also had a moderately adverse impact on fish weight gain in most cases.  相似文献

Abstract. Fifty each of F2-generation gold ( gg ), bronze ( Gg ), and black ( GG ) Oreochromis mossambicus (Peters) and either zero, four or eight largemouth bass, Micropterus salmoides , (Lacepède) were stocked in 20-m2 concrete tanks. After 7 days, water in all tanks was drained, and fish in each tank were censused. Largemouth bass ate 18% of the tilapia in the four-bass treatment (1 tilapia/bass/day) and 60% of the tilapia in the eight-bass treatment (1·6 tilapia/bass/day); the difference was significant (P=0–05). In the four-bass treatment there was a greater observed mean predation rate on gold than that on black or on bronze tilapia, but the difference was not significant: largemouth bass ate 25% of the gold tilapia, 16% of the bronze tilapia, and 13% of the black tilapia. In the eight-bass treatment, predation on the gold tilapia was significantly greater than that on both bronze (P=0·05) and on black (P=0·06) tilapia; predation on bronze and black tilapia was similar: largemouth bass ate 80% of the gold tilapia, 48% of the bronze tilapia, and 51% of the black tilapia. Overall average total predation (both treatments combined) on gold tilapia was significantly (P=0·06) greater than that on both bronze and on black tilapia, which did not differ: largemouth bass ate 52% of the gold tilapia, 32% of the bronze tilapia, and 32% of the black tilapia. The increased vulnerability of gold tilapia to predation was a negative pleiotropic effect of the gg genotype.  相似文献

Hatchery-reared Atlantic salmon, Salmo salar L., were vaccinated intraperitoneally (i.p.) with a live attenuated Cryptobia salmositica vaccine (either 100 000 or 5000 parasites fish−1) and 4 weeks later were challenged with the parasite (either 100 000 or 5000 parasites fish−1). Unvaccinated, infected salmon had high parasitaemias and were anaemic. Fish given a high dose (100 000 parasites fish−1) had higher parasitaemias than fish given the lower dose. Vaccinated fish had low parasitaemias and a mild anaemia, but recovered quickly after challenge. Complement-fixing antibody increased in vaccinated fish after challenge and was highest at 2 weeks post-challenge. The cell-mediated response (both T cells and B cells) was depressed in infected fish until 4 weeks after infection. In vaccinated fish, the humoral response (i.e. B-lymphocytes) was greater than the cell-mediated response (i.e. T-lymphocytes). In contrast, infected fish had a greater cell-mediated than humoral immune response.  相似文献

Abstract – Age and growth of juvenile tilapia, Oreochromis niloticus , from Lakes Zwai, Langeno and Chamo (Ethiopia) were studied from microincrements in otoliths. Growth in length was best described by the Gompertz model. Average growth rate of the fish was most rapid in Lake Chamo (0.39 mm  ·  day−1, 1.14%  ·  day−1), intermediate in Lake Zwai (0.20 mm  ·  day−1, 0.72%  ·  day−1) and slowest in Lake Langeno (0.16 mm  ·  day−1, 0.62%  ·  day−1). Similarly, back-calculation from otolith increment widths gave growth rates of 0.28 to 0.43 mm  ·  day−1, 0.15 to 0.32 mm  ·  day−1 and 0.11 to 0.28 mm  ·  day−1 for Chamo, Zwai and Langeno fish, respectively. In addition, Fulton's condition factor was largest for Chamo tilapia and smallest for Langeno tilapia; the difference between fish from Langeno and Zwai was small. Abstract. Six groups of tropical freshwater catfish, Mystus nemurus (Cuvier & Valenciennes)(mean weight. 20·45 ± l·5g), were reared in 0·34m3 fibreglass tanks at different stocking densities (105, 195, 285, 375, 465 and 555 specimens/m3 water) for 84 days. The objective of the study was to determine the effect of various stocking densities on the growth, nutrition, biochemical composition and survival of M. nemurus. The lowest growth rate appeared in fish at the highest density and the highest was observed in fish stocked in moderate density of 285 and 375 fish/m3 water. Fish production was also lower at relatively low stocking densities of 105 and 195 fish/m3. Food conversion ratio (FCR), protein efficiency ratio (PER) and biochemical composition of M. nemurus indicate that there exists an optimum stocking density which lies between 285 and 375 fish/m3.  相似文献
