为优化H3N2亚型犬流感病毒(CIV)的血凝抑制(HI)试验方法,本研究应用不同种类红细胞进行CIV的血凝(HA)试验,应用不同种类红细胞和不同血清处理方法进行CIV的HI试验,评价其对HA和HI试验的影响。结果表明,H3N2亚型CIV对鸡和犬红细胞的凝集性最好,对小鼠、猪和牛红细胞的凝集性较差。HI试验应用鸡红细胞悬液效果最好,受体破坏酶(RDE)和高碘酸钾处理可以有效去除犬血清中非特异血凝抑制素,但高碘酸钾对血清特异性抗体有损耗。本研究筛选出H3N2亚型CIV HI试验的最佳方法,为犬流感的血清学诊断提供技术支持。  相似文献

用哈尔滨维科生物技术开发公司生产的批号为200901的鸡减蛋综合征京911株油乳剂灭活疫苗,以0.5mL/只剂量接种120日龄SPF鸡10只,7d后采血分离血清,以后每隔一定时间进行采血、分离血清。同时又采集了发病鸡场的15份发病鸡血清。分别用血凝抑制试验和琼脂扩散(AGP)试验对自制的阳性血清和发病鸡血清样品进行血清学检查。结果表明:当血清HI效价等于或大于28时,两种方法的符合率可达100%;当血清的HI效价为26~27时,AGP与HI的符合率为73.33%~86.67%;当血清的HI效价为24~25时AGP的检出率很低或完全呈阴性。从两种方法的符合率比较还可得出HI试验检测的灵敏度要高于AGP,而且HI法简单、特异、快速、实用、方便,不但能定性还可以定量。  相似文献

为观察猪流行性腹泻病毒(PDEV)的血凝特性,进而为建立PEDVHA和HI试验,评估疫苗免疫抗体水平奠定基础,将CV777、DR13、YN13和YN144共4株PEDV毒株,在37℃下,分别用0、5、10、50、100μg/mL胰蛋白酶处理30min,然后用鸡、兔和猪红细胞进行血凝(HA)试验;用已经鉴定为PEDV抗体阳性的血清和乳清分别进行血凝抑制(HI)试验。结果显示:4株PEDV毒株只与兔红细胞发生凝集反应;CV777、DR13毒株的HA现象需要用5μg/mL的胰蛋白酶进行预处理,但是YN13和YN144毒株的HA现象不需要胰蛋白酶作用;该凝集现象能够被PEDV抗体特异性抑制。结果表明,PEDV能够凝集兔的红细胞,不能凝集鸡和猪的红细胞,因而可以用兔红细胞建立PEDV的HA和HI试验方法。  相似文献

新城疫病毒能够与鸡红细胞发生凝集现象,这种红细胞凝集现象可被特异性免疫血清所抑制,即红细胞凝集抑制试验(HI)。由于血凝反应可被特异性抗体所抑制,抗体与病毒结合后,血凝素不能吸附于红细胞表面的受体上,出现凝集抑制现象。该方法具有以下优点:①敏感性强,可以检测到微量的抗体,结果也较为准确,是较敏感的血清学反应之一;②特异性强,病毒凝集红细胞只能被特异的抗体所抑制;③检测速度快,1次HI试验只需2 h左右,即可判定结果;④HI试验对环境要求不高,操作简单,1次还可检测大量的样品。因此血凝抑制试验用于水禽血清,可以检测血清中的抗…  相似文献

A simple adhesion-hemadsorption inhibition (AHAI) test was developed for the detection of antibodies to Mycoplasma gallisepticum in the chicken sera. The AHAI antibody was detected simultaneously with HI antibody from sera of chickens intratracheally inoculated with viable cells of M. gallisepticum. A good correlation between HI and AHAI antibody titers was obtained with 382 (84.7%) of 451 sera from chickens reared on farms spontaneously contaminated with M. gallisepticum, whereas the remainder, 69 sera, was positive for HI but negative for AHAI test. It was not apparent whether the latters exhibited a non-specific reaction or the discrepancy was due to the lower sensitivity of AHAI reaction. The AHAI test does not require a great amount of antigen, special reagents or instruments, or pre-absorption treatment of test sera, and, therefore, it may serve as a simple serological test for detecting antibodies to M. gallisepticum.  相似文献

Both Mycoplasma gallisepticum (MG) and M. synoviae (MS) antigens prepared for the routine haemagglutination inhibition (HI) test were diluted and absorbed to the separate pieces of durapore membrane for the measurement of dot-immunobinding (DIB) titers of test sera. Besides, durapore strips bearing both antigens were employed for a DIB test with chicken sera definitely diluted 100-fold. Shortening of reaction time of chicken sera with antigens as well as with the secondary serum markedly eliminated non-specific DIB reactions exhibited at low dilutions although the same condition was not so effective on the elimination of non-specific reactions among rabbit hyperimmune sera. Rapid and specific development of DIB antibody which continued at high titer up to 1:640 for 10 weeks postinoculation was proved in the sera of SPF chickens inoculated with MG or MS, while DIB titers of sera from uninoculated chickens remained 1:20 or lower. Non-specific reactions, which occurred in the routine serum plate agglutination test with a part of sera from the inoculated chickens, were not exhibited in the DIB as well as in the HI test with the same sera. Results of the DIB test with serum samples from 287 conventionally reared chickens definitely diluted 100-fold coincided with the results of HI test at a level of 90% with MG and 89% with MS antigen. This technique seems to be useful for a rapid, simple and specific diagnosis of avian mycoplasmosis.  相似文献

A recombinant hemagglutinin-neuraminidase (rHN) protein from Newcastle disease virus (NDV) with hemagglutination (HA) activity was expressed in Spodoptera frugiperda cells using a baculovirus expression system. The rHN protein extracted from infected cells was used as an antigen in a hemagglutination inhibition (HI) test for the detection and titration of NDV-specific antibodies present in chicken sera. The rHN antigen produced high HA titers of 213 per 25 µL, which were similar to those of the NDV antigen produced using chicken eggs, and it remained stable without significant loss of the HA activity for at least 12 weeks at 4℃. The rHN-based HI assay specifically detected NDV antibodies, but not the sera of other avian pathogens, with a specificity and sensitivity of 100% and 98.0%, respectively, in known positive and negative chicken sera (n = 430). Compared with an NDV-based HI assay, the rHN-based HI assay had a relative sensitivity and specificity of 96.1% and 95.5%, respectively, when applied to field chicken sera. The HI titers of the rHN-based HI assay were highly correlated with those in an NDV-based HI assay (r = 0.927). Overall, these results indicate that rHN protein provides a useful alternative to NDV antigen in HI assays.  相似文献

Addition of anti-immunoglobulin M (anti-IgM), G (anti-IgG) and A (anti-IgA) sera to the haemagglutination-inhibition (HI) test (anti-Ig HI test) forMycoplasma gallisepticum resulted in 2- to 8-fold increases in the HI titres. On investigating the anti-Ig HI reaction using IgM and IgG antibodies separated by affinity chromatography, it was confirmed that, in the enhanced HI titres, specificity existed between the chicken Ig classes having antibody activity and the antisera used in the test. Four days after inoculation ofM. gallisepticum, the anti-Ig HI reaction was markedly enhanced by anti-IgM antiserum in the intravenously inoculated chickens and by anti-IgA serum in the nasally inoculated chickens. Ten days after inoculation ofM. gallisepticum marked enhancement of the reaction was produced by anti-IgG serum in both intravenously and nasally inoculated chickens, but the enhancement of the anti-Ig HI reaction diminished from the second week after inoculation.  相似文献

An immunoperoxidase monolayer assay (IPMA) has been developed to detect antibodies against swine influenza A virus (SIV) in pig sera. The test was evaluated by using sequential sera from pigs experimentally infected with H1N1 subtype of SIV. Two hundred field serum samples that had been examined by the hemagglutination inhibition (HI) test were also tested. Antibodies specific to SIV were detected as early as 3 days postinoculation (dpi) in the IPMA test as compared with 7 dpi by the HI test. Unlike HI, no serum treatment was required in the IPMA test. Regardless of the virus used in the test, IPMA detected antibodies to both H1N1 and H3N2 subtypes of SIV whereas HI detects antibodies against either H1N1 or H3N2, depending upon the virus used in the test. Results of this study indicate that IPMA is a useful test for screening of pig sera for SIV antibodies.  相似文献

为进一步完善中国大肠杆菌菌体微量凝集试验定型血清库,改良微量凝集试验用大肠杆菌O抗原定型血清的制备工艺,本试验选取大肠杆菌O50、O60、O117、O131血清型进行定型血清制备方法的探究。首先用大肠杆菌不同血清型的参考菌株制备灭活抗原,而后多次免疫家兔以采血获得粗血清,用微量凝集试验的方法确定不同粗血清的交叉凝集素及凝集效价,再通过交叉凝集素吸收法结合血清稀释法消除非特异性凝集,最终研制出特异性良好的微量凝集试验用大肠杆菌O抗原定型血清。本研究确定了大肠杆菌O50、O60、O117、O131定型粗血清的主要交叉凝集素,最终成功研制出特异性良好的微量凝集试验用大肠杆菌O50、O60、O117、O131定型血清共4种,凝集效价在1:256至1:2 048之间,其中微量凝集试验用大肠杆菌O50、O60定型血清无交叉凝集素,为单因子定型血清,微量凝集试验用大肠杆菌O117、O131定型血清存在1~2种非特异性的交叉凝集素O14和O107。本研究作为大肠杆菌O抗原定型血清制备工艺摸索过程中的重要部分,为完善中国大肠杆菌菌体微量凝集试验定型血清库,进一步改良微量凝集试验用大肠杆菌O抗原定型血清的制备方法提供了重要理论依据。  相似文献

Serum samples from 446 randomly selected persons belonging to different age groups and locations in Nigeria were tested for the presence of WSLV IgM using the flavivirus haemagglutination-inhibition (HI) test adopted to the solid-phase immunosorbent technique (SPIT). 61 (14%) persons had IgM to WSLV only, while 9 (2%) persons had heterologous IgM to WSLV and two other flaviviruses, namely yellow fever and Uganda S viruses. There was a high prevalence of IgM in people of younger age groups than those in older groups. The majority of the IgM positive sera (67 (96%) of the 70 positive sera reacted to high titres (>21:80). With the conventional HI tests, 314 (70%) of the total sera tested had HI antibodies to one or more flaviviruses (yellow fever, West Nile, Potiskum, Zika and Uganda S) out of which 305/314 (97%) had antibodies to 3 or more flaviviruses used in the tests. Although SPIT may not be as sensitive as the conventional HI test, it was found to be more specific and could be adopted for the detection of early WSLV infections in flavivirus hyperendemic environments.  相似文献

实验将 60只 1日龄健康雏鸡随机分为A、B、C 3组。A组为CALT缺失组 ,B组为免疫组 ,C组为非免疫对照组。A组于 1日龄手术切除下眼睑结膜 ,A、B两组于 1 2日龄结扎鼻泪管后进行点眼免疫接种新城疫克隆 30疫苗。2 3日龄时 ,收集A、B、C各实验组鸡的血清和泪液 ,做红细胞凝集抑制试验 (HI试验 )、酶联免疫吸附试验(ELISA)。比较观察各实验组雏鸡血清和泪液抗体HI效价与IgA、IgG、IgM含量的变化特点。结果 ,1日龄手术切除下眼睑结膜后造成鸡CALT缺失 ,CALT缺失鸡点眼免疫后 ,泪液抗体HI效价与IgA、IgG、IgM含量比免疫组雏鸡低。但对雏鸡的抗体生成的规律并不产影响。故实验证实了CALT对眼区局部抗体的产生及其种类有重要的调控作用 ,协同哈氏腺完成局部免疫的功能  相似文献

Serum samples from 191 ostriches (Struthio camelus) in Japan were tested for antibodies to Newcastle disease virus (NDV) and avian influenza virus (AIV). Twenty-two (12%) contained NDV-specific neutralizing antibodies by a virus-neutralization (VN) test without vaccination. Antibodies to AIV were not detected in the any sera by an agar gel precipitation test. Seven serum samples that had vaccinated with live NDV by eye drop were all positive by the VN test at 1 month post vaccination. A haemagglutination inhibition (HI) test for NDV seemed not to be suitable for ostriches because of non-specific agglutination of chicken red blood cells. No haemagglutinating viruses were isolated. This is the first report on detection of antibodies against NDV in ostriches in Japan.  相似文献
