Rhamnose-binding lectin (RBL) is one of the animal lectin categories which take part in the innate immune responses of fish. Osmerus lanceolatus lectin (OLL) from shishamo smelt eggs is an RBL composed of two tandem-repeated domains, both of which are considered to be a carbohydrate-recognition domain. SAL, catfish (Silurus asotus) egg RBL composed of three domains, binds to Burkitt’s lymphoma Raji cells through globotriaosylceramide (Gb3) carbohydrate chain and to reduce cell size and growth by altering membrane composition without causing cell death. In this experiment, we tried to compare the binding effects of these two RBLs on Raji cells. Flow cytometric and fluorescence microscopic analyses revealed that OLL also directly bound to and shrunk Raji cells with ten times less reactivity than SAL but reduced cell growth with decreasing cell viability. Anti-Gb3 antibody completely blocked the binding of SAL to Raji cells but not that of OLL. In addition, the direct bindings of OLL and SAL to Raji cells were comparably inhibited by melibiose, but lactose was more effective inhibitor for the binding of OLL than that of SAL. These results suggest that OLL has slightly different cell-binding property compared with SAL and binds not only to Gb3 but also to the other carbohydrate receptor-bearing β-galactoside chains. The quantitative RT-PCR analysis revealed that SAL induced the expression of TNF-α but not of IFN-γ, IL-1β, and IL-10. Thus, SAL-induced cytostatic effect on Raji cells might be partially caused by TNF-α-mediated signaling pathway.  相似文献

In this study, we investigated the gross structure, secondary structure, and antitumor and mitogenic activity of GANL, a lectin from the gill of bighead carp (Aristichthys nobilis). We used infrared spectroscopy, β-elimination, and circular dichroism spectroscopy to determine the structure of GANL. We measured antiproliferation activity against six human tumor cell lines and mitogenic activity against murine splenocytes using the MTT assay. Based on infrared spectroscopy and β-elimination, we conclude that GANL is a glycoprotein. The protein and carbohydrate moieties are joined by O-glycosidic linkage. A circular dichroism spectroscopic analysis revealed that the secondary structure of GANL consists of α-helices (34.8 %), β-sheets (12.1 %), β-turns (24.5 %), and unordered structures (33.0 %). GANL exerted potent antitumor activity against the HeLa cell line (IC50 = 11.86 μg/mL) and a mitogenic effect on murine splenocytes in the MTT assay. GANL, a lectin that is isolated from the gills of bighead carp, is a glycoprotein with potent antitumor and mitogenic activity.  相似文献

This study was conducted to evaluate dietary protein and carbohydrate requirement of juvenile Hawaiian limpets Cellana sandwincensis. A total of 64 juvenile limpets (3.12 ± 0.86 g) were fed five different dietary protein levels ranging from 270 to 470 g kg?1 for 90 days. Carbohydrate and lipid levels were held constant at 180 and 49.7 g kg?1, respectively. Weight gain and growth rates of the animals did not differ significantly (P > 0.05) among the protein levels ranging from 270 g kg?1 (0.30 % day?1) to 470 g kg?1 (0.23 % day?1). Next, opihi were fed four diets with protein levels from 210 to 500 g kg?1 with a constant carbohydrate level at 120 g kg?1. Weight gain and specific growth rates of opihi increased with increasing dietary protein from 210 to 350 g kg?1, and significantly (P < 0.05) decreased at the 500 g kg?1 diet. Highest weight gain, growth rates, and protein efficiency ratio were achieved at 350 g kg?1. Elevated carbohydrate levels (180–370 g kg?1) produced a significant difference (P < 0.05) in growth. The fastest growth rates of animals were obtained with 270 g kg?1 (0.27 % day?1) and 320 g kg?1 (0.26 % day?1). The weight gain of animals fed 180 and 370 g kg?1 carbohydrate diets were significantly (P < 0.05) lower than those of animals fed 270 and 320 g kg?1. We conclude that about 350 g kg?1 protein and 320 g kg?1 carbohydrate levels could be used for opihi.  相似文献

The effect of irradiance and temperature on the photosynthesis of two Japanese agarophytes, Gelidium elegans and Pterocladiella tenuis (Gelidiales), was determined using dissolved oxygen sensors and pulse amplitude modulated (PAM) fluorometry. Gross photosynthesis and dark respiration rates were determined over a range of temperatures (8–36 °C). The highest gross photosynthetic rates were 40.3 and 37.0 mg O2 g ww ?1  min?1 and occurred at 24.3 and 25.5 °C [95 % Bayesian credible interval (BCI) 20.7–28.0 and 23.4–28.3 °C], respectively. The dark respiration rate in G. elegans and P. tenuis increased with increasing temperature at a rate of 0.10 and 0.31 mg O2 g ww ?1  min?1 °C?1 , respectively. Modeling the net photosynthesis–irradiance (PE) responses of G. elegans and P. tenuis at 20 °C revealed that the net photosynthetic rates quickly increased at irradiance levels below the estimated saturation irradiance of 88 and 83 µmol photons m?2 s?1, with a compensation irradiance of 14 and 19 µmol photons m?2 s?1, respectively. The highest value of the maximum effective quantum yield (Φ PSII) occurred at 20.1 °C (BCI 18.9–21.5 °C) and 21.3 °C (BCI 20.2–22.5 °C) for G. elegans and P. tenuis and was 0.49 and 0.45, respectively. These optimal temperatures of Φ PSII were relatively lower than those determined by the photosynthesis–temperature model of oxygen evolution. The temperature response of these species indicates that they are probably well adapted to the current range of seawater temperatures in the study site but that they are near the boundary of their tolerable limits.  相似文献

Biofloc technology based 45-day indoor growth trial was conducted to evaluate the effect of carbohydrate (CHO), molasses supplementation on two developmental stages viz, juvenile (J) and sub-adult (SA) of Penaeus monodon in zero-water exchange system. P. monodon juveniles (1.56 ± 0.04 g) and sub-adults (14.32 ± 0.22 g) were stocked in fibre-reinforced plastic tanks (1,000 L) with soil base. Carbohydrate supplementation significantly reduced (p < 0.05) the total ammonia nitrogen (TAN), nitrite-N (NO2-N) and nitrate-N (NO3-N) over the time periods in sub-adult group. However, no significant difference in TAN and NO2-N was observed in juvenile-based treatments. Supplementation of carbohydrate significantly increased (p < 0.05) the water and soil total heterotrophic bacterial (THB) count in both juvenile and sub-adults. Despite increase in total Vibrio count (TVC), lower TVC/THB % was noticed in the water column of J + CHO (0.82) and SA + CHO (0.73) compared to control groups, J ? CHO (1.48) and SA ? CHO (1.21). Supplementation of carbohydrate increased the final body weight in J + CHO (6.51 ± 0.44 g) and SA + CHO (22.52 ± 0.98 g) compared with respective controls, J ? CHO (5.05 ± 0.45 g) and SA ? CHO (20.00 ± 0.33 g). Similarly, significantly lower (p < 0.05) feed conversion ratio and better protein efficiency ratio were recorded in carbohydrate-supplemented juvenile and sub-adults treatment groups. The present study demonstrates that supplementation of carbohydrate reduced the nitrogenous metabolites and significantly enhanced the growth performance of juvenile and sub-adult of P. monodon.  相似文献

To assess the growth-promoting potential of three medicinal herbs, Alteranthera sessilis, Eclipta alba and Cissus quadrangularis, a 90-day feeding trial was conducted on Macrobrachium rosenbergii post-larvae (PL) fed with these herb-incorporated feeds. The growth parameters (survival rate, weight gain, specific growth rate and protein efficiency ratio) were significantly (P < 0.05) higher in experimental-diets-fed PL when compared with control. These herbs have acted as appetizer and significantly enhanced the activities of digestive enzymes (protease, amylase and lipase). This in turn significantly (P < 0.05) influenced the parameters of energy utilization (feeding rate, absorption rate, conversion rate, metabolic rate and NH3 excretion rate), which ultimately reflected on significant (P < 0.05) increases in concentrations of biochemical constituents (total protein, amino acid, carbohydrate and lipid, and essential amino acids and unsaturated fatty acids). Therefore, it was believed that synthesis of protein was activated due to the active principles of these herbs and resulted in growth promotion. In this study, the higher (P < 0.05) minerals (Na+ and K+) recorded indicate the fact that their balance was maintained. Similarly, the non-enzymatic antioxidants (vitamin-C and -E) status were also improved significantly (P < 0.05) due to herbal incorporation, which provide additional protection. Further, the haemocytes count and population was also found to be higher (P < 0.05), which ensures prevalence of good host-defence mechanisms in herb-incorporated-feeds-fed PL. Among three herbs tested, C. quadrangularis-incorporated-feed-fed PL showed the best survival, growth and protection as well as production followed by E. alba and A. sessilis. Therefore, these herbs can be incorporated in artificial feed formulations for sustainable development of Macrobrachium culture.  相似文献

Fairy shrimp is known as a nutritional food for fish and crustaceans in aquaculture. In most hatcheries, the microalga Chlorella sp. appears to be the most common, suitable, and nutritious food to feed fairy shrimp. In this study, we attempted to determine other alternative algal diets for cultivation of fairy shrimp Branchinella thailandensis. Seven experimental diets including three treatments of dried Spirulina sp. at 0.75 (S1), 1.5 (S2), and 3.0 mg dry weight individual?1 (S3); three treatments of Chlorococcum humicola at 5 × 105 (Ch1), 1 × 106 (Ch2), and 2 × 106 cells mL?1 (Ch3); and a control diet (Chlorella vulgaris at 1 × 106 cells mL?1) were fed to 5-day-old shrimp for 15 days. Evaluation of growth performance, egg production, survival percentage, and nutritional and carotenoid content of the experimental fairy shrimp revealed that Ch3 is the most suitable algal diet. Our results suggest that C. humicola is the best alternative food source for the cultivation of B. thailandensis. In addition, dried Spirulina powder is also a good choice when live algae are not available and can be used as an alternative feed in fairy shrimp cultures.  相似文献

The present study was conducted to evaluate the effect of dietary supplementation of probiotic bacterium Bacillus licheniformis on the gut microbiota, growth and immune response in Macrobrachium rosenbergii juveniles (4.0 ± 0.02 g). Five isocaloric experimental diets supplemented with different concentrations of B. licheniformis, viz. Control (without probiotic supplementation), T1 (1.0 × 106 cfu g?1feed), T2 (1.0 × 107 cfu g?1feed), T3 (1.0 × 108 cfu g?1 feed) and T4 (1.0 × 109 cfu g?1 feed) were used for the experiment. After 60 days of feeding trial, the growth performance increases significantly in all the experimental groups compared with control, and highest weight gain, specific growth rate and lowest feed conversion ratio were recorded from T4 (P > 0.05). However, juvenile survival was not found to be significantly affected by probiotic supplementation (P > 0.05). The total bacterial count and Bacillus spp. count in the gut were significantly increased with concurrent decrease in pathogenic Aeromonas spp. and Pseudomonas spp. in all the experimental groups compared with control (P < 0.05). Compared with control, immune parameters were increased significantly in all the experimental groups (P < 0.05). Highest haemocyte count, phenol oxidase, superoxide dismutase and antibacterial activity in haemolymph were recorded from T4 (P < 0.05). When prawns were challenged with Vibrio alginolyticus, the cumulative mortality was significantly lower in T4 (P < 0.05). These results indicate that dietary supplementation of B. licheniformis can manipulate the gut microbiota and increase the growth and immune response of M. rosenbergii. In the present study, concentration response on the probiotic effect of B. licheniformis was observed which showed higher effects at a concentration of 1.0 × 109 cfu g?1 feed in the laboratory condition.  相似文献

A 60-day feeding trial was conducted to study the effect of dietary microbial levan on growth performance and metabolic responses of Cyprinus carpio fry exposed to sublethal dose (1/10th LC50) of fipronil [(±)-5-amino-1-(2,6-dichloro-α,α,α-trifluoro-p-tolyl)-4-trifluoromethylsulfinylpyrazole-3-carbonitrile]. Two hundred and twenty five fry were randomly distributed in five treatments in triplicates. Four purified diets were prepared with graded levels of microbial levan. Five different treatment groups were levan control L0P0 (basal feed + 0 % levan without exposure to pesticide); pesticide control L0P1 (basal feed + 0 % levan with exposure to pesticide); L0.25P1 (basal feed + 0.25 % levan with exposure to pesticide); L0.50P1 (basal feed + 0.50 % levan with exposure to pesticide); and L0.75P1 (basal feed + 0.75 % levan with exposure to pesticide). Weight gain% and specific growth rate were significantly higher (p < 0.05) in levan fed groups compared to their non-levan fed counterpart. Highest (p < 0.05) content of ascorbic acid in muscle, liver and brain tissues was observed with higher level of dietary levan. Glucose-6-phosphate dehydrogenase activity decreased with the increasing level of dietary levan in the liver and muscle. Aspartate aminotransferase activity exhibited a second order polynomial relationship with the dietary levan, both in liver (Y = ?1.001x 2 + 5.366x + 5.812, r 2 = 0.887) and muscle (Y = ?0.566x 2 + 2.833x + 6.506, r 2 = 0.858) while alanine aminotransferase activity showed third order polynomial relationship both in liver (Y = 1.195x 3 ? 12.30x 2 + 35.23x + 9.874, r 2 = 0.879) and muscle (Y = 0.527x 3 ? 8.429x 2 + 31.80x + 8.718, r 2 = 0.990). Highest (p < 0.05) superoxide dismutase activity in gill was observed in the group fed with 0.75 % levan supplemented diet. Overall results indicated that dietary microbial levan at 0.75 % in C. carpio fry ameliorated the negative effects of fipronil and augmented the growth.  相似文献

In this study, we describe a novel lectin (designated AMUL) from the Northern Pacific starfish Asterias amurensis. AMUL was purified from the coelomic fluid by affinity chromatography followed by size-exclusion chromatography. The native molecular mass of the lectin was found to be approximately 270 kDa by size-exclusion chromatography using a Superdex 200 column. AMUL showed distinct bands at approximately 14 or 18 kDa on SDS-PAGE under nonreducing or reducing conditions, respectively. The hemagglutination activity of AMUL against rabbit erythrocytes was strongly inhibited by the two sialic acids N-acetylneuraminic acid and N-glycolylneuraminic acid. The glycoproteins tested showed no inhibition of hemagglutination. The first 28 amino acid residues of AMUL were determined by automated Edman degradation, showing that AMUL has considerable sequence homology with C-type lectins from other echinoderms. These results show that AMUL is a novel echinoderm-derived sialic-acid-specific lectin that belongs to the C-type lectin superfamily.  相似文献

Lipoic acid (LA) is an antioxidant that also favors glucose uptake in mammals. Until now, there are no studies evaluating the potential effect of this molecule on glycemic control in fish. It was evaluated LA effects on glucose uptake in common carp Cyprinus carpio fed with carbohydrate diets from two carbohydrate sources: glucose (GLU) and starch (STA), and supplemented or not with LA, being the diets: +GLU/?LA (GLU); +GLU/+LA (GLU + LA); +STA/?LA (STA); and +STA/+LA (STA + LA). Carp juveniles (6.5 ± 0.1 g) were fed with each diet ad libitum 4 times a day, during 68 days. Muscle glycogen concentration was higher (p < 0.05) in GLU and GLU + LA than in STA and STA + LA groups. On fish fed with starch, muscle cholesterol and triglyceride concentrations were higher (p < 0.05) in fish fed diets supplemented with LA. Muscle protein levels were higher in fish fed with LA, independent of the diet carbohydrate source. Lipid peroxidation was significantly reduced (p < 0.05) in fish muscle on fish fed the STA + LA diets when compared with the STA diet. Our findings indicate that LA modulates lipid, proteins and carbohydrate metabolism together with the well-known antioxidant effect. Also, LA showed to enhance starch utilization taking into account muscle cholesterol and triglyceride levels.  相似文献

A 45-day feeding trial was conducted to evaluate the effects of dietary citric acid on growth, digestive enzyme and disease resistance of white shrimp, Litopenaeus vannamei. Shrimp with initial body weight of 5.57 ± 0.21 g were fed with basal diet supplemented with 0.0 g kg?1 (control), 1.0, 2.0, 3.0, 4.0 and 5.0 g kg?1 citric acid. Results showed that weight gain was increased by 15.9 % and feed conversion ratio was decreased by 0.17 by 2.0 g kg?1 dietary citric acid compared with control group (P < 0.05). Intestinal protease activity of shrimp fed 2.0 g kg?1 citric acid was significantly higher (P < 0.05) than that of control group. No significant difference was found in intestinal amylase activity among treatments (P > 0.05). The activities of serum phenoloxidase, superoxide dismutase and lysozyme in 2.0 and 3.0 g kg?1 citric acid group were significantly higher, and accumulative mortalities of the two groups on the fourth day after injection of Vibrio alginolyticus were significantly lower than those of control group (P < 0.05). Results above demonstrated that dietary citric acid could improve growth performance, immunity and resistance against V. alginolyticus. The supplementation level of citric acid in diet was suggested to be 2.0–3.0 g kg?1 for white shrimp.  相似文献

In aquatic ecosystems, the stable nitrogen isotope ratio (δ15N) of wakame is affected by the δ15N of its nitrogen source. Thus, the δ15N of wakame is thought to differ between those grown in different aquatic ecosystems. To confirm the authenticity of the geographic origin of Naruto dried wakame (DW), we conducted δ15N analysis. The δ15N (mean ± σ) of Naruto DW model samples (n = 72) was 11.1 ± 1.9 ‰. The δ15N confidence range of Naruto DW from the model was 5.3–16.9 ‰ (mean ± 3σ). The δ15N values of the DW model of Chinese (n = 23) and South Korean origin (n = 22) were 4.2 ± 1.4 and 1.6 ± 2.3 ‰, respectively. Within the δ15N confidence range of Naruto DW, two model samples of Chinese DW and one model sample of South Korean DW were found. From those results, DW of Naruto origin was able to be distinguished from DWs of the Chinese and the South Korean.  相似文献

Abalone culture systems are characterised by high water exchange rates as abalone are sensitive to changes in water quality. To reduce the costs of pumping, serial reuse systems have been tested. As water pH can decline in these systems, this study determined the effect of raising pH through the addition of sodium hydroxide (NaOH?) on weight gain, length gain and feed conversion ratio of H. midae in a serial-use raceway. Dosing with NaOH? increased pH by 0.3, 0.27 and 0.21 units at positions 1, 2 and 3 of a serial-pass system, respectively. Weight gain (Wg) and length gain (Lg) were correlated with pH (Wg: F 1, 16 = 16.35, P = 0.0009, r 2 = 0.51; Lg: F 1, 16 = 16.16, P = 0.0009, r 2 = 0.50), and using pH, dissolved oxygen and free ammonia nitrogen as independent variables suggested that water pH was the best predictor for abalone growth.  相似文献

In the present study, konjac mannanoligosaccharide (KMOS) was evaluated as a prebiotic in yellow catfish. The fish were fed with diets containing KMOS in four concentrations: 0 g kg?1 (C), 1.0 g kg?1 (KM1), 2.0 g kg?1 (KM2), and 3.0 g kg?1 (KM3) for 49 days, respectively. Another group fed with diets containing 3.0 g kg?1 yeast cell wall mannanoligosaccharide (MOS) (M3) was set as positive control. The results indicated that fish receiving the diets supplemented with KMOS or MOS showed higher relative gain rate (RGR), specific growth rate (SGR), and lower feed conversion ratio (FCR) with significantly differences (P < 0.05) than those fed with the basal diets. Moreover, fish receiving the diets with 2.0 g kg?1 KMOS inclusion showed higher RGR, SGR, and lower FCR (P < 0.05) than that feeding the diets supplemented with 3.0 g kg?1 MOS. The quantities of Bifidobacterium spp. were significantly increased (P < 0.05). Meanwhile, Escherichia coli and Aeromonas spp. were significantly reduced (P < 0.05) in the fish-feeding diets with 2.0 g kg?1 KMOS supplement. Compared with the control group, the significantly enhancement of protease and amylase activity (P < 0.05) in intestine and pancreas was observed in fish fed with diets containing KMOS or MOS. Collectively, an optimum level of KMOS inclusion in diets could modulate intestinal microflora, induce digestive enzyme activity, and improve the growth performance of yellow catfish significantly.  相似文献

An indoor trial was conducted for 28 days to evaluate the effects and interactions of biofloc and seaweed Ulva lactuca in water quality and growth of Pacific white shrimp Litopenaeus vannamei in intensive system. L. vannamei (4.54 ± 0.09 g) were stocked in experimental tanks at a density 132 shrimp m?2 (566 shrimp m?3) and the U. lactuca was stocked at a density 0.46 kg m?2 (2.0 kg m?3). Biofloc with seaweed (BF-S) significantly reduced (P < 0.05) total ammonia nitrogen (TAN) by 25.9 %, nitrite–nitrogen (NO2–N) by 72.8 %, phosphate (PO 4 3 -P) by 24.6 %, and total suspended solids by 12.9 % in the water and significantly increased (P < 0.05) settleable solids by 34.2 % and final weight of shrimp by 6.9 % as compared to biofloc without seaweed. The BF-S can contribute by reducing nitrogen compounds (TAN and NO2–N), phosphate (PO 4 3 -P), and total suspended solids in water and increased final weight of shrimp.  相似文献

We determined through morphological and rbcL phylogenetic analyses that a previously unidentified, but introduced species of macroalga, which has been easily cultivated in indoor tanks in Japan, is Agardhiella subulata (Solieriaceae). Additionally, the photosynthetic biology of this alga was examined by inducing photosynthetic activity under a variety of water temperatures and photosynthetic active radiation (PAR) to clarify the optimal conditions needed for its efficient cultivation. Photosynthetic activity was evaluated by using both dissolved oxygen (DO) and pulse amplitude modulated-chlorophyll fluorometric (PAM) techniques, and focused on elucidating temperature and PAR levels that would potentially maximize productivity. The DO method revealed that the net photosynthetic rates at 24 °C quickly increased as PAR increased, and approached a P max of 27.8 mg O2 g ww ?1  min?1 (95 % Bayesian credible interval, BCI 23.8–32.1). The maximum gross photosynthetic rate occurred at 26.7 °C (BCI 24.4–28.3 °C). However, PAM experiments indicated that for the maximum quantum yield, the optimal temperature was 23.7 °C (BCI 22.7–24.6) and the maximum relative electron rates occurred when the water temperature was 31.0 °C (BCI 30.6–31.5). This study suggests that the broad tolerance of maximal photosynthetic activity to temperature (22.7–31.5 °C) is one of the main reasons why this alga can be successfully cultivated year-round.  相似文献
