ABSTRACT:   The distribution of zinc (Zn) concentration in the blood and erythrocytes of common carp, grass carp, silver carp and tilapia was studied. It was found that in whole blood, the average Zn concentrations in these four species of fish (approx. 6–14 μg/[ml whole blood]) were relatively similar to those in other species of fish and mammals. However, the mean Zn concentration in the erythrocytes of common carp is approximately two times higher than the other three species of fish (approx. 5 vs approx. 2 μg/[ml whole blood]). It was found that approximately 70% of the Zn in the common carp whole blood came from its erythrocytes. In addition, approximately 43% of the Zn in the erythrocytes of common carp was found to be located on its outer plasma membranes. When an antibody against a 43 kDa Zn-binding protein, isolated from the digestive tract tissue of common carp, was used, significant quantities of the protein were shown to be present on the erythrocyte plasma membranes of common carp by an indirect immunofluorescent staining. High Zn on the outer plasma membrane of the common carp erythrocyte most probably comes from the 43 kDa Zn-binding protein.  相似文献

ABSTRACT: The concentrations of Zn and sulfhydryl (SH) groups in the digestive tract tissue of common carp and some aquatic animals were studied. It was found that Zn and bound SH groups could be used as indicators for detecting the Zn-binding protein in the digestive tract tissue of common carp. The digestive tract tissue of the fish underwent subcellular fractionation, and it was found that the nuclei/cell debris fraction contained most of the DNA (85%), Na+/K+-ATPase (82%), organic phosphate (90%) and the Zn-binding protein (79%), but only part of the 5'-nucletidase and alkaline phosphatase (<23%). The nuclei/cell debris fraction of the digestive tract tissue of common carp was treated with either collagenase type I or type IV, and subfractionated by sucrose density centrifugation. It was found that treatment with collagenase type IV could release more than 50% of the Zn-binding protein, Na+/K+-ATPase and organic phosphate from collagen. Sections of digestive tract tissue of common carp were stained for Zn. It was observed that Zn can be found mainly on the edge of the epithelial layer, and everywhere in the 'membrane-like' portion of the submucosal and muscular layers. It is proposed that most of the Zn-binding protein in the digestive tract tissue of common carp is located on the basolateral plasma membranes of the epithelial cells and on the surrounding muscle cells that are attached to the collagen type IV of basal laminae.  相似文献

High zinc diet of 2000 mg kg–1 was fed to common carp (Cyprinus carpio), grass carp (Ctenopharyngodon idellus) and tilapia (Oreochromis aureus) for 8 weeks to compare the accumulation of Zn in fish. It was found that accumulation of zinc from diet by grass carp and tilapia was low when compared to common carp. The largest difference occurred in digestive tract tissue. The digestive tract tissue of common carp had a large pool of stored zinc, the mean storable capacity is about 1500 g Zn g–1 fresh tissue, and the time needed to saturate the tissue when feeding 2000 mg Zn kg–1 diet is about 8 weeks. The accumulated Zn in the digestive tract tissue of common carp was released when the dietary Zn was reduced to normal level (50 mg kg–1) or deficient level (4 mg kg–1) for 4 weeks.Subcellular fractionation results indicate that Zn accumulated in digestive tract tissue of common carp was accumulated mainly in the nuclei/cell debris fraction. Exposure to a high Zn diet induced some metallothionein-like substance in the digestive tract tissue and the hepatopancreas of common carp, but the amount was very low when compared with the amount of Zn accumulated in the nuclei/cell debris fraction.  相似文献

To separate the connective tissue cells expressing the 43-kDa zinc-binding protein from the common carp, mucosa of the digestive tract of the fish was removed by scraping with a glass slide, and the de-mucosa tissue was digested with a collagenase type IV solution. The cells collected from the collagenase-treated suspension were rather homogeneous, and more than 90% of the cells were round with a diameter of approximately 6 μm. Significant quantities of the 43kDa zinc-binding protein were shown to be present on the cell surface of the approximately 6-μm cells. The mean zinc concentration in the cells was found to be 2.21 μg zinc/106 cells, which is approximately 20—30 times higher than that found in the other three fish species (grass carp, silver carp, tilapia), studied. The present work may provide a basis for the culture of these cells.  相似文献

ABSTRACT:    This study incorporated the 43 kDa Zn-binding membrane protein isolated from common carp into liposome. The specificity and strength of the binding of 65Zn to the 43 kDa protein-liposomes, and the binding of the 65Zn-labeled 43 kDa protein-liposomes to laminin were studied. It was found that 65Zn was bound to the external side of the 43 kDa protein-liposomes. Specific binding of 65Zn to the protein-liposomes was detected. The binding parameter of Zn to the protein was found to be: maximum binding site (Nmax), 76.7 pmole/µg protein (approx. 3 mole of Zn2+/mole); and equilibrium dissociation constant (Kd), 0.19 µM. Of the cations introduced (Ca2+, Cd2+,Co2+, Cr2+, Cu2+, Fe2+, Hg2+, Mg2+, Mn2+, Ni2+, Pb2+), only Co2+ competed significantly with Zn. The protein-liposomes were also found to bind specifically to laminin with a Nmax of 1.1 pmole/µg laminin, and Kd of 4.79 µM. No significant protein-liposome binding occurred to other extracellular matrix proteins (fibronectin, fibrinogen or vitronectin). Furthermore, the binding was specifically inhibited by the Arg-Gly-Asp (RGD) peptide or GRGDSPG, while two other analogs (GRGESPG and GRADSPG) were without effect.  相似文献

ABSTRACT:   Ten lots of common carp and six lots of crucian carp (each lot of 100 fish) were treated under anoxia for five days at water temperatures of 25–31°C. The average per group mortality of common carp was 17%, but none of the 600 crucian carp died. The digestive tract tissues of the common carp that died had significantly lower zinc concentrations than those that survived (59 ± 41 vs 142 ± 60 μg/[g fresh tissue], P  < 0.001). The digestive tract tissues of the crucian carp had mean zinc concentrations of 652 ± 458 μg/(g fresh tissue). One lot of common carp that had low tolerance for anoxia was fed a high zinc diet (2000 mg zinc/kg diet) for 1, 2 or 6 months and then subjected to 5 days anoxia. The survival rates of those fed the high zinc diet 1 and 2 months increased from 0 to 50%, respectively; all of fish that had fed a high zinc diet for 6 months survived. Thus, anoxia survival in common carp and crucian carp is closely related to the high concentrations of zinc in their tissues.  相似文献

We studied the link between high zinc levels and the extreme stress tolerance of common carp. Fish under stress showed much higher plasma cortisol levels than controls. Stress or cortisol injection induced large changes in zinc levels in the common carp but not in grass carp, silver carp or tilapia. The effect of 5 days of anoxia and 4 subsequent days of recovery on cortisol and zinc contents in the common carp was investigated. Elevated plasma cortisol resulting from anoxia was correlated with decreased zinc in digestive tract tissue and increased zinc in the head kidney. Zinc was mobilized in the common carp while under stress. Changes in cortisol and zinc contents were reversible during the subsequent recovery from anoxia. Under stress, protein-bound zinc levels increased in the head kidney cell nuclei of common carp as cortisol increased. Zinc and cortisol were bound to the same protein, which was bound to DNA. The protein is likely a glucocorticoid receptor. An increase in immature red blood cells in stressed common carp was observed. Zinc was involved in the stress erythropoiesis response. Zinc may play an important role in stress defense in the common carp via the glucocorticoid receptor.  相似文献

本文通过组织解剖与切片法测量了鲤、草、鲢、鳙的相关生物学参数,系统观察了四种鱼的消化道肌肉层、肠绒毛以及粘液细胞的显微结构及分布特点,探究其与食性的相互关系。结果显示:鲤比肠长1.0,环行肌较纵行肌发达,肠绒毛丰富,粘液细胞均匀分布在整个消化道中;草鱼比肠长2.13,消化道组织结构均一,肌肉壁中纵行肌所占比例高,粘液细胞体积小,分散于整个消化道中,肠绒毛极发达;鲢比肠长8.49,环行肌极发达,粘液细胞集中于消化道前段,肠绒毛由前至后逐渐呈短粗状;鳙比肠长4.58,环行肌极为发达,粘液细胞数量多且均匀地分布于消化道前、中段,肠绒毛较为稀疏短粗,粘液细胞较少。这四种鱼消化道的组织结构特征与各自食性密切相关。  相似文献

采用聚丙烯酰胺凝胶电泳方法研究了鲤、鲢、鳙、草鱼亲鱼消化道消化酶的种类;用比色法及滴定法分别测其消化酶相对活性。结果表明:鲤、鲢、鳙、草鱼4种鱼消化道的蛋白酶、淀粉酶活性与食性有明显的相关性。其中,蛋白酶活性依次为鲤〉鲢〉鳙〉草鱼;淀粉酶活性依次为草鱼〉鲤〉鳙〉鲢。脂肪酶活性与食性的关系不明显。四种鱼消化酶的表达与消化道的组织结构特征相一致,消化道前2/5部位(G1、G2)的消化酶活性最高,是无胃鱼消化道的主要消化场所。消化道中央部位(G3)的消化酶活性次之,能将未消化完全的食糜进一步消化,而消化道后段(G4、G5)消化酶活性最低。可见,消化酶活性及消化功能呈递减形式,食物在消化道中随着消化、吸收,逐渐排出体外。本研究为鲤、鲢、草鱼、鳙4种鱼的营养生理学、高效饲料配比的研制及种质资源的科学管理提供理论依据。  相似文献

选择草鱼(C tenopharyngodon idellus)和青鱼(Mylopharyngodon piceus)作试验鱼,对它们消化道的酸性蛋白酶、碱性蛋白酶、淀粉酶以及三酰基甘油脂酶沿消化道及肝脏和胰腺中的活性分布进行了比较研究。研究表明:这些酶在消化道中呈现不同的分布规律,在肝脏和胰腺中酶的活性高于在消化道中相应酶的活性;同种鱼不同消化酶的分布有所差别;两种鱼消化组织中的各种酶活性分布规律也有差别;青鱼和草鱼的主要消化酶在不同消化器官中的分布有较大差别。  相似文献

This work provides a comparative study of the proteolytic and amylase activities in six species of fish with different nutritional habits: rainbow trout (Oncorhynchus mykiss), gilthead seabream (Sparus aurata), European eel (Anguilla anguilla), common carp (Cyprinus carpio), goldfish (Carassius auratus), and tench (Tinca tinca). Trout and carp showed the highest digestive proteolytic activity. When proteolytic activity was determined in a wide range of pHs, the highest values in the digestive tract of all species were found at alkaline pHs, except in eel where activity could be detected only at acid pHs. Eel showed the lowest digestive proteolytic potential among all the species studied. With respect to amylase activity, the omnivorous species presented higher activity than did the carnivores. Among the carnivorous species, the lowest activity was found in trout. The ratio of total amylase:total proteolytic activity was higher in omnivorous fish species, the carp having the greatest value, whereas in trout this ratio was lower than one. Digestive enzyme activity declined as the incubation temperature decreased, but this trend varied depending on the fish species and the tissue analyzed.  相似文献

A growth trial was conducted to estimate the optimum requirement of dietary zinc (Zn) for grass carp (Ctenopharyngodon idella). Triplicate groups of grass carp (3.97 ± 0.05 g) were fed diets containing graded levels (13, 25, 34, 53, 89 and 135 mg kg?1) of Zn for 8 weeks. Grass carp fed with dietary Zn levels higher than 34 mg kg?1 significantly increased final body weight, weight gain and specific growth rate (P < 0.05). For body composition, fish fed with dietary Zn levels higher than 53 mg kg?1 significantly decreased the moisture contents but increased the lipid contents of whole body and liver. Whole body, scales, vertebrae and liver mineralization were all affected significantly (P < 0.05) by dietary Zn levels. Zn contents in whole body, scales, vertebrae and plasma were linearly increased up to the 53 mg kg?1 dietary Zn and then remained stable beyond this level. Grass carp fed with dietary Zn levels higher than 53 mg kg?1 significantly increased triacyglyceride and total cholesterol contents and plasma alkaline phosphatase activity in plasma (P < 0.05). Broken‐line analysis indicated that 55.1 mg kg?1 dietary Zn was required for maximal tissue storage and mineralization as well as optimal growth of grass carp.  相似文献

Characteristics and functional efficacy of digestive proteases of Catla catla, catla, Labeo rohita, rohu and Hypophthalmichthys molitrix, silver carp were studied. Total protease activity was significantly (P < 0.05) higher in rohu (1.219 ± 0.059 U mg protein−1 min−1) followed by silver carp (1.084 ± 0.061 U mg  protein−1 min−1), and catla (0.193 ± 0.006 U mg  protein−1 min−1). Trypsin activity of silver carp and rohu was 89–91% higher than catla. Chymotrypsin activity was significantly (P < 0.05) higher in silver carp compared with rohu and catla. The protease activity of rohu and silver carp displayed bell‐shaped curves with maximum activity at pH 9; whereas in catla, maximum activity was found between pH 8 and 11. Inhibition of protease activity with soybean trypsin inhibitor (SBTI), phenylmethylsulfonyl fluoride (PMSF) revealed the presence of serine proteases and inhibition of activity with N‐α‐p‐tosyl‐L‐lysine‐chloromethyl ketone (TLCK) and N‐tosyl‐L‐phenylalanychloromethane (TPCK) indicated the presence of trypsin‐like and chymotrypsin‐like enzymes in all these three carps. SDS‐PAGE showed the presence of several protein bands ranging from 15.3 to 121.9 kDa in enzyme extracts of catla, rohu and silver carp. The substrate SDS‐PAGE evidenced the presence of various protease activity bands ranging from 21.6–93.7, 21.6–63.8 and 26.7–98.5 kDa for catla, rohu and silver carp respectively. In pH‐stat hydrolysis of Chilean fishmeal showed significantly (P < 0.05) higher degree of hydrolysis compared with soybean meal, silver cup (a commercial fish feed of Mexico) and wheat flour, with enzyme preparations of three fishes. The rate of hydrolysis was significantly (P < 0.05) higher in silver carp compared with others.  相似文献

A feeding trial was conducted for 8 weeks to evaluate the effects of supplemental phytic acid (PA) on the apparent digestibility and utilization of dietary amino acids (AAs) and minerals in juvenile grass carp. Five experimental diets consisted of graded levels of PA (0.2, 4.7, 9.5, 19.1 and 38.3 g kg?1, named as P0, P5, P10, P20 and P40). Triplicate groups of fish (initial weight, 22.37 ± 0.16 g) were fed twice daily (08:00 and 16:00 h). The crude protein content in whole body significantly (< .05) decreased in fish fed with 19.1 and 38.3 g PA kg?1 diet. Supplemental PA (>4.7 g kg?1) significantly reduced the apparent digestibility coefficient (ADC) of AAs (Asp, Thr, Ser, Glu, Gly, Ala, Cys, Val, Met, Ile, Leu, Phe, Lys, Pro, His and Arg) and the ADC of minerals (P, Ca, Mg, Zn, Cu, Fe and Mn) in grass carp. The contents of minerals (P, Ca, Mg and Zn) in whole body and bone were also found to be significantly reduced in dietary PA > 4.7 g kg?1, while the bone ash, serum Alkp and Zn contents were found to be significantly decreased when the PA supplementation level was above 9.5 g kg?1, and the contents of serum Ca and Mg were found to be markedly altered in higher PA‐supplemented groups. The results indicated that supplemental PA decreased the apparent digestibility and utilization of AAs and minerals, and thus reduced the feed utilization of grass carp, suggesting that the level of total PA should be below 4.7 g kg?1 in grass carp diet.  相似文献

The present investigation was carried out to study gut content and accompanying digestive enzyme profiles of the three carp species, golden mahseer Tor putitora, snow trout Schizothorax richardsonii and hill trout Barilius barila. The fishes were captured from the River Ladhiya, Champawat district, Uttarakhand, northern India. The digestive tract length was on average 1.8 and 2.3 times longer than the body, while members of Bacillariophyceae, Chlorophyceae, Cyanophyceae and Zygnematophyceae made up 95.15% and 99.63% of gut content in the golden mahseer and snow trout, respectively. In hill trout, the digestive tract being only half the body length, gut content was insects (72.73%), eggs of aquatic organisms (22.73%) and nematodes (4.55%). Estimated gut enzyme activities demonstrated that amylase activity was significantly (P < 0.05) higher in golden mahseer and snow trout compared to hill trout, while total protease, trypsin and chymotrypsin activities were all significantly (P < 0.05) higher in the hill trout compared to other species. A strong co‐relation was found among relative gut length, food composition and digestive enzyme activities of each species. This may guide the future formulation of feed to these important species of carps kept in farm.  相似文献

Biochemical assays and substrate SDS-PAGE were conducted to partially characterize and identify various types of proteases present in the digestive tract of PL15 giant freshwater prawn ( Macrobrachium rosenbergii ). Casein hydrolytic assay of the enzyme extracts showed major proteolytic activities at pH 3.0, 6.0 and 9.0, while assay of preincubated enzyme extracts with phenylmethylsulphonyl fluoride (PMSF), a serine protease inhibitor produced a 33.17% reduction in alkaline protease activity. When specific inhibitors tosyl-lysine chloromethyl ketone and tosyl-phenylalanine chloromethyl ketone were used, they resulted in a reduction in activity of proteases in the enzyme extracts by 82.41% and 55.03%, respectively, confirming the presence of trypsin and chymotrypsin, while ethylenediamine tetraacetic acid produced protease activity reduction in 33.92% showing the presence of metalloproteases in the digestive tract of the prawn. Further characterization of the alkaline proteases using SDS-PAGE technique, after incubating the extract in the presence or absence of specific inhibitors, produced six bands corresponding to molecular masses of between 13.48 and 136.1 kDa; two trypsin bands of 13.48 and 36.4 kDa, three chymotrypsin bands in the range of 23.0–73.4 kDa and one for metalloprotease of 136.1 kDa, all of which were identified from a zymogram. This study suggests that protein digestion in M. rosenbergii is initiated by an acid protease followed by a combination of action of alkaline proteases: trypsin, chymotrypsin and metalloproteases.  相似文献

ABSTRACT:   Bioaccumulation and distribution of Mn and Zn in the total softtissues, digestive glands, residuals and adductor muscles of thehorse mussel Modiolus modiolus from three sites, includingindustrialized and non-industrialized locations in Eastern Canada,were investigated. Extremely high digestive gland metal concentrationswere found in individual mussels, as high as 1819 µg/gMn wet weight and 1964 µg/g Zn wet weight,with mean values from 358 to 404 µg/gMn and from 399 to 614 µg/g Zn for thecollection sites. High Mn to Zn interrelationships were observed inall types of tissues and at all sites. Between different tissues,Zn was interrelated by linear relationships, and Mn was best describedby power curve relationships for all tissue types. In the totalsoft tissue, Mn and Zn interrelations were fitted to power regressioncurves with different slopes between the three study sites. Thisindicated that horse mussel was exposed to different metal levels inthe environments and could be useful for monitoring these metals.The uptake of both metals at extremely high concentrations, thelack of regulation and the occurrence of interactions all suggest thatMn and Zn may play a biological role in horse mussels. Zn and Mninteractions, surprisingly, were not disrupted at the very highconcentrations of either metal, which proves that the mechanism ofmetal interactions does not involve a detoxification role.  相似文献

This study was conducted to investigate the effects of dietary inulin on growth performance, diet utilization, survival rate, carcass composition and digestive enzymes activities (amylase, lipase and protease) of carp (Cyprinus carpio) fry (0.55 ± 0.02 g). After acclimation, fish were allocated into 9 tanks (40 fish per tank) and triplicate fish groups were fed, control diet (0 g) or diets containing 5 g and 10 g inulin kg?1 for 7 weeks. No significant effect on growth performance and diet utilization of fish fed inulin compared with the control group was observed. However, supplementation of inulin significantly increased survival rate and carcass lipid content, while carcass protein content significantly decreased. Dietary inulin had no significant effects on digestive lipase, protease and amylase activities.  相似文献
