AIM: To identify and quantify concentrations of the isoflavones genistein, daidzein, biochanin A and formononetin in commercially- prepared feline diets sold in New Zealand. METHODS: Feline diets (n=138) were collected from supermarkets, pet stores and veterinary clinics in New Zealand. Diets were classified into five categories based on the following criteria: the presence/absence of soy, the presence/absence of non-soy plant material, and dry matter (DM) content. A high performance liquid chromatography (HPLC)-based assay was developed and validated to identify and quantify concentrations of the isoflavones genistein, daidzein, biochanin A and formononetin. RESULTS: Isoflavones were detected in all categories of diet, and at quantifiable concentrations in 104/138 (75%) of the diets tested. More dry diets (127/138; 92%) contained isoflavones at quantifiable concentrations than moist diets (83/138; 60%, p<0.001). Of the isoflavone-positive diets, moist diets that contained soy had the highest median isoflavone content (71.1 mg/kg DM and 0.018 mg/kcal metabolisable energy; ME) whilst moist meat-only diets had the lowest (3.24 mg/kg DM and 0.0004 mg/kcal ME). Isoflavone contents varied similarly between categories of diet whether evaluated on a DM or ME basis. High isoflavone contents were associated with diets containing soy and those of low cost. Biochanin A and formononetin were found at concentrations above the detection limit of the assay in 25/138 (18%) and 7/138 (5%) of the diets analysed, respectively. The estimated median daily isoflavone exposure for cats consuming feline diets sold in New Zealand ranged from 0.03 mg/kg bodyweight (BW) for moist meat-only diets to 1.47 mg/kg BW for moist diets containing soy, whilst the diet with the highest isoflavone content would provide daily exposure of 8.13 mg/kg BW. CONCLUSIONS: Isoflavones are present in commercially-prepared feline diets sold in New Zealand at concentrations that have elicited physiological responses in the reproductive, endocrine, and immune systems of other mammalian species. Since isoflavones appear to be a common constituent of feline diets, further investigation of the biological activities of these dietary compounds in cats is warranted.  相似文献

Dietary isoflavones, capable of influencing reproductive parameters in domestic cats (Felis catus), have been detected in commercial diets fed to captive cheetahs (Acinonyx jubatus). However, the absorptive and metabolic capacity of cheetahs towards isoflavones has not yet been studied. Experiments were designed to describe the plasma concentration-time curve, metabolite profile, and urinary and fecal excretion of genistein and daidzein in cheetahs following consumption of isoflavones. Four adult cheetahs were administered a single oral bolus of genistein and daidzein, and five juvenile cheetahs consuming a milk replacer formula found to contain isoflavones were also included. Urine was collected from all animals, and blood and feces were also collected from adult cheetahs following isoflavone exposure. Samples were analyzed for isoflavone metabolite concentration by liquid chromatography-electrospray ionization-multiple reaction ion monitoring mass spectrometry and high-performance liquid chromatography. Sulfate conjugates were the primary metabolites detected of both genistein and daidzein (60-80% of total isoflavones present) in the plasma and urine of cheetahs. A smaller proportion of daidzein was detected as conjugates in the urine of juvenile cheetahs, compared to adult cheetahs. Other metabolites included unconjugated genistein and daidzein, O-desmethylangolensin, and dihydrodaidzein, but not equol. Only 33% of the ingested genistein dose, and 9% of daidzein, was detected in plasma from adult cheetahs. However, of the ingested dose, 67% of genistein and 45% of daidzein were detected in the feces of adults. This study revealed that cheetahs appear efficient in their conjugation of absorbed dietary isoflavones and only a small fraction of ingested dose is absorbed. However, the capacity of the cheetah to conjugate genistein and daidzein with sulfate moieties appears lower than reported in the domestic cat. This may confer greater opportunity for biologic activity of isoflavones in the cheetah than would be predicted from findings in the domestic cat. Sloths are renowned for their low metabolic rate, low food intake and low defecation frequency. We investigated factors of digestive physiology and energy metabolism in four captive individuals (mean body mass 10.0 ± SD 3.7 kg) of a hitherto mostly unstudied sloth species, Linné's two‐toed sloth (Choloepus didactylus), in a 2‐week digestion recording and 23‐h respiration experiment on animals fed a standard zoo diet of vegetables and starchy components. Dry matter intake, defecation frequency and particle mean retention time (MRT) in the gastrointestinal tract (GIT) were 12 ± 3 g/(kg0.75 day), once every 5 days and >140 h in three individuals, but 53 g/(kg0.75 day), daily and 82 h in one individual that was apparently compensating for a period of weight loss prior to the experiment. In all animals, solute marker was eliminated at a faster rate than the particle marker, indicating ‘digesta washing’ in the sloths’ GIT. The overall metabolic rate calculated from oxygen consumption matched the metabolisable energy intake in three individuals [173 ± 22 vs. 168 ± 44 kJ/(kg0.75 day)] but not in the fourth one [225 vs. 698 kJ/(kg0.75 day)], supporting the interpretation that this animal was replenishing body stores. In spite of the low food intake and the low‐fibre diet (209 ± 26 g neutral detergent fibre/kg dry matter), methane production was rather high accounting for 9.4 ± 0.8% of gross energy intake (2.7% in the fourth individual), which exceeded literature data for ruminants on forage‐only diets. These results corroborate literature reports on low intake, low defecation frequency, low metabolic rate and long MRT in other sloth species. The long MRT is probably responsible for the comparatively high methane production, providing more opportunity for methanogenic archaea than in other non‐ruminant mammals to produce significant amounts of methane.  相似文献

We investigated the effect of diet on intake of energy and fibre in a group of three captive adult giraffe by weighing offered diet items and leftovers for 7 days after an adaptation period of 7 days. Digestion coefficients were calculated using, as internal marker, the acid detergent lignin content of a faecal sample pooled from subsamples taken during the last 5 days of intake measurement. Two lucerne hay-only diets of differing quality (L1, L2) were fed, as well as the regular diet of lucerne hay and concentrates (L2C), and the regular diet supplemented with 3 or 6 kg of edible, fresh browse material (L2CB3, L2CB6). The proportion of roughage in the ingested diets L2CB6 (45 +/- 5% dry matter), L2CB3 (35 +/- 3%) and L2C (37 +/- 10%) did not differ significantly. Digestible energy intake was low on the hay-only diets [L1: 0.28 +/- 0.06 MJ/kg body weight (BW)0.75; L2: 0.33 +/- 0.10 MJ/kg BW0.75] and increased from L2C (0.60 +/- 0.13 MJ/kg BW0.75) to a significant increase with L2CB3 (0.72 +/- 0.17 MJ/kg BW0.75); no further increase was obvious for L2CB6 (0.63 +/- 0.15MJ/kg BW0.75). The results confirm that giraffes are unlikely to meet energy requirements on lucerne hay-only diets. In a feeding scenario where both lucerne hay and the concentrate component of the diet are fed ad libitum, the animals tended to exchange hay for browse when browse was added. Only the higher level of browse supplementation led to a potentially beneficial increase in fibre intake. Whether additional browse supplementation will lead to increased intakes in a feeding scenario with restricted concentrate provision can be suspected but remains to be demonstrated.  相似文献

The aim of this study was to evaluate the effect of the level of alfalfa in the diet on feed intake and digestibility of two types of rabbits, wild (Oryctolagus cuniculus algirus) vs. domestic (O. cuniculus cuniculus). Ten wild (W; mean LW = 927 g) and 10 domestic (D; mean LW = 4,645 g) adult rabbit does were fed ad libitum two pelleted diets: a control diet (C) with 15% of dehydrated alfalfa hay (as feed basis) and a test diet (A) with 36% of dehydrated alfalfa hay (as feed basis), according to a change‐over design. Wild does dry matter (DM) intake per kg live weight (BW) was 55% higher (p < .001) than the intake of the D ones (58 g vs. 37 g DM per kg BW respectively). However, no difference (p > .05) was found when intake was expressed per kg0.75 BW (ca. 56 g DM) and tended to be higher (p = .07) in D does when expressed per kg0.67 BW (62 g vs. 55 g DM). Domestic does showed a higher (p < .05) DM, organic matter, crude energy and neutral detergent fibre digestibility (3; 2; 3; 3 percentage points respectively) than W does. The amount of nutrients and energy digested by D does was lower per kg BW (p < .001), similar per kg0.75 BW (p > .05) and tended to be higher per kg0.67 BW (p < .1) than in W does. The diet content of alfalfa did not affect (p > .05) the feed intake nor the diet digestibility. This study suggests that W rabbits exhibit a higher intake per kg BW and a lower digestibility than their D counterparts, which results in similar digestible nutrient and energy intake per kg BW powered to 0.75. The nutritive value of dehydrated alfalfa for rabbits, evaluated through intake and digestibility, seems to be equivalent to their base diets (forage plus concentrate).  相似文献

To evaluate the effects of dietary supplementation with a marine algae product (MA) containing a high level of docosahexaenoic acid (DHA) on heavy pig production performances (including meat and subcutaneous fat quality), a trial was conducted on sixty Landrace × Large White barrows with a starting average body weight (BW) of 118 kg. Pigs were allocated to four experimental groups: a control group (A) fed a maize/soybean diet and three treatment groups in which MA was added at 2.5 g kg− 1 over the last 8 weeks prior to slaughtering (group B) or at 5 and 2.5 g kg− 1 over the last 4 weeks prior to slaughtering (groups C and D, respectively). Vitamin E was added at 12.5 mg kg− 1 in the diets for groups B and D and at 25 mg kg− 1 in the diet of group C. The four diets were formulated so as to supply the same amount of nutrients, with exception of vitamin E. Pigs were fed at a rate of 9% of their metabolic BW (BW0.75) up to a maximum of 2.9 kg of dry matter per pig per day. Pigs were slaughtered at about 160 kg BW. The dietary treatment did not affect either the growth or slaughtering parameters of pigs. No significant differences were observed with respect to pH values, meat colour, loin composition and the iodine number of subcutaneous fat. Pigs on MA diets showed a significant (P < 0.01) improvement in DHA levels both in loin and in subcutaneous fat. No significant differences in relative fatty acid composition were observed between pigs receiving the MA supplement at the lower level (2.5 g kg− 1) over an 8-week period (group B) and pigs receiving the higher MA supplement (5.0 g kg− 1) over a 4-week period prior to slaughter (group C).  相似文献

We conducted two experiments with heavy Iberian pigs to determine the ileal digestibility of amino acids (AA) in acorns and freshly cut herbage, and the effects of adding fresh herbage upon the supply of ileal digestible AA when pigs were fed on holm‐oak acorns. In Experiment 1, carried out in cannulated pigs of 107 kg bodyweight (BW), daily intake of acorns reached 44.9 g DM/kg0.75 BW. Arg, His and Thr showed the lowest apparent ileal digestibility (AID) values, whereas Met, the branched‐chain AA and Phe had the highest coefficients. The AID of total EAA was 0.716 but only 0.222 for NEAA. Most of the digestive and absorptive processes of acorn protein occurred before the hindgut. Acorn provides (per kg DM) 2.27 g apparent ileal digestible Lys and 22.7 g apparent total digestible AA. Standardized ileal digestibility (SID) values for EAA, NEAA and total AA were 0.924 ± 0.020, 0.784 ± 0.041 and 0.860 ± 0.029. In Experiment 2 fresh herbage was given to six cannulated Iberian pigs of 140 kg either as a single feed (13.7 g DM/kg0.75 BW) or as a supplement to acorns (28.4 g DM/kg0.75 BW). When only freshly cut forage was offered the AID of the EAA, NEAA and total AA was close to 0.65 and supplied (per kg DM ingested) 5.61 g AID Lys and 91.7 g digestible AA. Standardized ileal values were 0.744 ± 0.023, 0.912 ± 0.038 and 0.831 ± 0.030 respectively. The addition of fresh forage to the acorns led to a significant decrease in AID of AA in acorn due to digesta transfer to the hindgut: His (p < 0.01), Met (p < 0.001), Phe (p = 0.092), Thr (p < 0.05) and Val (p < 0.05), but Arg, Lys and the branched‐chain AA remained unaffected. The main contribution of herbage to AA nutrition of the grazing Iberian pig relies mainly on increasing the supply of digestible AA for pig tissues.  相似文献

Two experiments were conducted to determine the effect of soy isoflavones on growth, meat quality, and carcass traits of growing-finishing pigs. In Exp. 1, 36 barrows (initial and final BW, 26 and 113 kg, respectively) were used and each treatment was replicated four times with three pigs each. The dietary treatments were 1) corn-soybean meal (C-SBM), 2) corn-soy protein concentrate (low isoflavones, C-SPC), or 3) C-SPC + isoflavones (isoflavone levels equal to those in C-SBM). Daily gain and ADFI were increased (P < 0.10) in pigs fed the C-SPC relative to pigs fed the C-SPC + isoflavone diet in the late finishing period; otherwise, growth performance was not affected (P > 0.10) by diet. Longissimus muscle area, 10th-rib fat depth, percentage muscling (National Pork Producers Council), 24-h pH and temperature, color, firmness-wetness, marbling, drip loss, and CIE L*, a*, and b* color values were not affected (P > 0.10) by diet. Dressing percentage, carcass length, weight and percentage of fat-free lean in ham and carcass, lean gain per day, lean:fat, and ham weight were increased (P < 0.10), and ham fat and percentage fat in ham and carcass were decreased (P < 0.10) in pigs fed the C-SPC + isoflavone diet compared with pigs fed the C-SPC diet. Pigs fed the C-SPC + isoflavone diet had similar (P > 0.10) carcass traits as pigs fed the C-SBM diet, except carcass length, percentage ham lean and thaw loss were greater (P < 0.10), and total ham fat was less (P < 0.10) in pigs fed the C-SPC + isoflavone diet. In Exp. 2, 60 gilts (initial and final BW, 31 and 116 kg, respectively) were used, and each treatment was replicated five times with four pigs per replicate. The treatments were 1) C-SBM, 2) C-SBM + isoflavone levels two times those in C-SBM, and 3) C-SBM + isoflavone levels five times those in C-SBM. Daily feed intake was linearly decreased (P < 0.10) in the growing phase and increased (P < 0.10) in the late finishing phases as isoflavone levels increased; otherwise, growth performance was not affected (P > 0.10) by diet. Diet did not affect (P > 0.10) carcass traits; however, CIE a* and b* color scores and drip loss were decreased (P < 0.06) as isoflavone levels increased. Soy isoflavones decreased fat and increased lean in barrows when fed within the dietary concentrations found in typical C-SBM diets but not when fed to gilts at concentrations above those present in C-SBM diets.  相似文献

A nitrogen (N) balance digestion trial was conducted to determine the protein requirement of collared peccaries (Pecari tajacu). In a 4 × 4 Latin square design, four captive adult male peccaries were fed four isoenergy diets containing four different levels of N (11.7, 16.3, 22.8, and 26.7 g N/kg of dry matter—DM). After 15 days of adaptation, a total collection of feces and urine was carried out for five consecutive days. Regression analyses between N intake and N in feces and urine allowed to calculate the metabolic fecal nitrogen (MFN = 2.3 g N/kg of dry matter intake—DMI) and daily endogenous urinary N (EUN = 185 mg N/kg0.75). Likewise, by regression analyses between consumption of nitrogen and the nitrogen balance (NB = N ingested ? N excreted, mg N/kg0.75), a daily requirement of 514 mg N/kg0.75 was calculated. Therefore, if food intake is unrestricted, collared peccaries require a minimum in their diet of about 5.4% crude protein on DM basis. These values are almost as low as those found for browsing and frugivorous wild ruminants, which reinforce the proposition that peccaries’ digestive physiology is nearer to that of domestic and wild ruminants than domestic pigs. This relatively low protein requirement of collared peccary and its great ability to digest protein reveal the relevance of the forestomach for the species on nitrogen/protein metabolism and allow the use of diets with lower crude protein levels than the commercial ones used for the domestic pig, which reduces feed costs.  相似文献

试验选用30周龄罗曼公、母鸡72只,分为12个处理,每个处理6个重复,每个重复1只试鸡,单笼饲养,分别饲喂蛋氨酸水平为理想需要量的36%、50%、65%、75%、85%和100%的半纯合梯度日粮,建立蛋白质和蛋氨酸沉积量对蛋氨酸食入量间的回归方程,来确定30周龄罗曼公母鸡的可利用蛋氨酸维持需要量。结果表明:公鸡蛋白沉积量显著高于母鸡(P<0.05)。分别以蛋白质和蛋氨酸沉积量为因变量,所得公鸡可利用蛋氨酸维持需要分别为每天39.34mg/kgBW0.7(5或表示为266.81mg/kgBPM)和1.93mg/kgBW0.7(5或表示为13.11mg/kgBPM)。母鸡可利用蛋氨酸维持需要分别为每天52.32mg/kgBW0.7(5或表示为282.14mg/kgBPM)和7.76mg/kgBW0.7(5或表示为41.87mg/kgBPM);由蛋白质沉积量为因变量所得的回归方程外推零蛋白质沉积时的蛋氨酸维持需要量要显著高于以零蛋氨酸沉积所得的维持需要量(P>0.05),母鸡可利用蛋氨酸维持需要高于公鸡的测定结果。  相似文献

An isotope dilution method using [1‐13C]sodium acetate was applied to determine the effect of feeding ensiled hop (Humulus lupulus L.) residues on plasma acetate turnover rate in six adult crossbred sheep. The sheep were fed 63 g/kg body weight (BW)0.75/day of either mixed hay of orchardgrass (Dactylis glomerata L.) and reed canarygrass (Phalaris arundinacea L.) and round bale silage at 3:1 ratio (Hay‐diet), or another where round bale silage was replaced by ensiled hop residues (Hop‐diet) with a crossover design each of a 3‐week period. The isotope dilution method was performed on day 21 of each dietary treatment. Dry matter digestibility was similar between diets, and nitrogen (N) digestibility was lower (P = 0.001) for Hop‐diet than Hay‐diet. However, N retention did not differ between diets. Plasma acetate concentration was lower (P = 0.04) for Hop‐diet than Hay‐diet, and the turnover rate of plasma acetate did not differ between diets. Plasma concentration of lactate and non‐esterified fatty acids were similar between diets. Hop‐diet was found almost comparable to Hay‐diet on plasma acetate turnover rate in the present experimental conditions. Therefore, it could be concluded that hop residues partially could be used as an alternative to traditionally used round bale silage for rearing sheep.  相似文献

The effect of dietary energy levels on the urinary excretion of purine derivatives (PD) was investigated in sheep fed hay and concentrate diets. In addition, the possibility of using PD concentrations in spot samples of urine as a useful tool for determining daily PD excretion was also examined. Three female lambs (mean bodyweight (BW) ± standard error, 31.4 ± 1.0 kg) were fed three diets consisting of timothy hay (20 g dry matter/kg BW per day) and rolled barley at three different energy levels (low‐energy (LE), medium‐energy (ME) and high‐energy (HE) at 1.0‐, 1.2‐ and 1.5‐fold greater than the maintenance level of energy (450 KJ/BW0.75/day), respectively) in a 3 × 3 Latin square design. The dietary protein level was almost constant (380 mg nitrogen/BW0.75/day as digestible crude protein base) and was adjusted using soybean protein. Digestible organic matter intake (DOMI) increased (P < 0.05) with an increase in the dietary energy level and there was a difference (P < 0.05) in the organic matter digestibility between the LE and HE diets. For urinary PD excretion, the differences between the LE and ME diets and the HE diet were significant (P < 0.05), although there was no clear difference between the LE and ME diets. There was a positive correlation (r = 0.47, P < 0.05) between urinary PD excretion and DOMI. There was also a correlation (r = 0.59, P < 0.05) between the urinary PD level at 7–8 h after the morning feed and the daily total PD excretion, suggesting that the PD level in spot urine samples could be useful for estimating microbial protein synthesized in the rumen and/or digested in the lower gut.  相似文献

OBJECTIVE: To determine the absolute and relative soy isoflavone content in commercial cat foods. SAMPLE POPULATION: 14 dry, 6 semimoist, and 22 moist commercial cat foods. PROCEDURE: Soy isoflavone content of each food was determined by use of acid-methanol hydrolysis and high-pressure liquid chromatography with ultraviolet absorbance detection. Isoflavones were identified and quantified by reference to authentic standards. RESULTS: Genistein and daidzein were the major soy isoflavones identified in 24 of 42 foods, with concentrations ranging from 1 to 163 microg/g of food. Foods labeled as containing soybean solids (16/42) had isoflavone concentrations > 11 microg/g. More dry (13/14) and semimoist (6/6) foods contained isoflavones than moist foods (5/22). Isoflavone content and food cost were negatively correlated for dry and semimoist foods but not for moist foods. Total amount of isoflavone consumed by cats fed these soy-containing foods as a sole maintenance diet was estimated to be between 0.6 and 4.5 mg/kg of body weight/d, which is comparable to concentrations in humans that result in a measurable although modest effect on serum concentrations of steroid and thyroid hormones. CONCLUSIONS AND CLINICAL RELEVANCE: Genistein and daidzein are common constituents of commercial cat foods. Predictors of isoflavone content included ingredient labeling, food type, and food cost. Soy isoflavones in some commercial cat foods were detected in amounts predicted to have a biological effect.  相似文献

选用1日龄健康樱桃谷鸭128只,随机分为对照组、试验Ⅰ组、试验Ⅱ组、试验Ⅲ组,大豆异黄酮的添加剂量分别为0mg/kg、5mg/kg、10mg/kg和15mg/kg,每组4个重复,每个重复8只,公母各半,分别于21和42日龄屠宰,测定屠宰指标和脏器系数,同时对试鸭的日增重进行统计分析.结果10 mg/kg剂量添加组显著增加了21日龄樱桃谷鸭胃重(P<0.1),同时显著提高了生长速度(P<0.1);15 mg/kg添加组显著增加了42日龄樱桃谷鸭的腿肌率、瘦肉率(P<0.1),其他指标和对照组相比,差异均不显著(P>0.1).提示高剂量的大豆异黄酮对樱桃谷鸭的生长更有利,大豆异黄酮对樱桃谷鸭生长和生产性能的影响主要是通过提高早期生长速度,缩短生产周期,从而提高饲料转化率来完成的.  相似文献

Objective

The butorphanol-azaperone-medetomidine fixed-dose combination (BAM, respectively, 30-12-12 mg mL?1) with subsequent antagonism by naltrexone-atipamezole was evaluated for reversible immobilization of captive cheetahs (Acinonyx jubatus).

Study design

Prospective, clinical trial.

Animals

Twelve cheetahs (six males and six females, weighing 37–57 kg) housed in enclosures, were immobilized at Hoedspruit Endangered Species Centre in the Republic of South Africa.

Methods

BAM volume dose rate was 0.009–0.014 mL kg?1 (mean ± standard deviation 0.010 ± 0.001 mL kg?1). Total dose in all animals was 0.5 mL. The actual doses were as follows: butorphanol (0.29 ± 0.04 mg kg?1), azaperone (0.12 ± 0.01 mg kg?1) and medetomidine (0.12 ± 0.01 mg kg?1). Physiologic variables and quality of immobilization were recorded every 5 minutes beginning at 15–20 minutes after darting. Arterial blood samples were collected three times at 20, 30 and 40 minutes after darting from all animals for analysis of blood oxygenation and acid-base status.

Results

The inductions were calm and smooth and mean induction time was 4.0 ± 1.1 minutes. Heart rate (50 ± 9 beats minute?1) and respiratory frequency (20 ± 3 breaths minute?1) were stable throughout immobilization. The recovery time after reversing with naltrexone and atipamezole was 9.1 ± 3.6 minutes.

Conclusions

and clinical relevance BAM proved to be a reliable and cardiovascular stable drug combination for immobilization of cheetahs.  相似文献

Two long-acting neuroleptics were used to tranquilize nine captive cheetahs (Acinonyx jubatus). Perphenazine enanthate (3.0 mg/kg) and zuclopenthixol acetate (0.6 mg/kg) were each administered to separate groups of three cheetahs in a double blind trial. Both products were administered together to a third group of three animals at the same dosages. Behavioral effect, duration of effect, and possible side effects were observed by a predefined protocol. Under standardized holding conditions, the cheetahs were observed 5 days before drug administration and 14 days after administration. Daily activity was defined and statistically evaluated by a U-test. A significant reduction of activity was observed after administration in all three trials. Zuclopenthixol acetate at 0.6 mg/kg alone and in combination with perphenazine enanthate caused inappetence, ataxia, extra pyramidal reactions, akathisia, and prolapse of the third eyelid. Zuclopenthixol acetate should not be used in cheetahs. Perphenazine enanthate did not cause inappetence, reduced appetite, or any of the previously mentioned side effects when used alone. It produced satisfactory tranquilization and is suitable and safe for cheetahs at 3.0 mg/kg. This dosage should be varied depending on health, age, and temperament of the individual cheetah.  相似文献

A 4 × 6 Youden square design was used to study manipulating effect of Yucca schidigera with or without nisin on rumen fermentation characteristics and microbial protein synthesis in sheep fed silage‐ and hay‐based diets. Four ruminally canulated wethers were given isonitrogenous diets of timothy silage and concentrate (85:15) (S), and timothy hay and concentrate (60:40) (H), on DM basis. The six experimental diets were: (i) S; (ii) S + 240 ppm of Yucca schidigera (SY); (iii) S + 240 ppm of Yucca schidigera + 6 mg/kg bodyweight (BW)0.75 of nisin (SYN); (iv) H; (v) H + 240 ppm of Yucca schidigera (HY); and (vi) H + 240 ppm of Yucca schidigera + 6 mg/kg BW0.75 of nisin (HYN). Rumen pH was higher (P < 0.01) in sheep fed SYN diet than those fed S diet. Concentration of ammonia‐N (NH3‐N) was significantly lower (P < 0.05) in sheep fed SY, SYN, HY, HYN than those of S and H. Total volatile fatty acids concentration was significantly lower (P < 0.05) in HYN than in H diet and tended to be higher in sheep fed H diets than those fed S diets. There was a significantly higher (P < 0.01) efficiency of microbial nitrogen synthesis in the rumen of sheep fed on HY diet than those fed on H diet.  相似文献

ObjectiveTo establish a safe, reliable and reversible immobilization protocol for captive juvenile crocodiles.Study designProspective, randomized, clinical study.AnimalsThirty male estuarine crocodiles (body mass 1–12.1 kg) and 10 male Australian freshwater crocodiles (body mass 4.1–12.8 kg).MethodsAn optimized dose of medetomidine (0.5 mg kg?1) was administered intramuscularly (IM) into the tail (Group 1; n = 5), pelvic limb (Group 2; n = 5) and thoracic limb (Groups 3 and 4; n = 5 in each group) of estuarine crocodiles weighing 3–12.1 kg. Their heart and respiratory rates and degree of immobilization were monitored every 15 minutes until recovery and daily thereafter for 3 subsequent days. In Group 4 (n = 5), medetomidine was antagonized with an optimized dose of atipamezole (2.5 mg kg?1) given IM into the thoracic limb and time to recovery recorded. The effects of increasing doses of medetomidine given IM in the thoracic limb (n = 4) and intravenously (n = 6) were determined in 1–2 kg estuarine crocodiles. Australian freshwater crocodiles (4.1–12.8 kg) were administered medetomidine IM into the thoracic limb in divided doses at 0.5 mg kg?1 (n = 5) and 0.75 mg kg?1 (n = 5) and similarly monitored.ResultsImmobilization was achieved only in the estuarine crocodiles >3 kg and when medetomidine was administered into the thoracic limb. Immobilization was achieved within 30 minutes and the duration of immobilization lasted approximately 90 minutes. Immobilization in estuarine crocodiles was readily reversed with atipamezole. A dose of 0.75 mg kg?1 was required to immobilize Australian freshwater crocodiles and the onset of immobilization was longer and the duration shorter than seen in the estuarine crocodiles. The heart and respiratory rates of all immobilized animals decreased significantly and arterial blood pressure became undetectable in the animals in which it was measured.Conclusions and clinical relevanceMedetomidine administered in the thoracic limb of captive estuarine and Australian freshwater crocodiles, ranging from 3 to 12.8 kg, provides a predictable onset and duration of immobilization sufficient for physical examination, sample collection, short minor procedures and translocation of the animals. Atipamezole administered in the thoracic limb results in complete reversal of the effects of medetomidine in the estuarine crocodile and a rapid return to normal behaviour.  相似文献

This study aimed to estimate the essential amino acid profile and the ideal ratio for the maintenance of poultry by deletion method. A nitrogen balance (NB) trial was conducted using 198 adult roosters, housed individually in metabolic cages. The treatments were 33 purified diets being 11 diets with an amino acid mixture providing high protein intake of 500 mg N/BWkg0.75 per day, 11 diets providing medium protein intake of 250 mg N/BWkg0.75 per day (in each diet, one amino acid tested was diluted 50%) and 11 diets providing low protein intake of 125 mg N/BWkg0.75 per day (made by omitting the amino acid tested). Each treatment had six replicates. After 48 h of fasting receiving water plus sucrose, the roosters were fed 40 g of the diets by tube once a day for 3 days. The excreta were collected within 72 h after the first feeding. The diets and excreta were analysed for nitrogen content. For each amino acid studied, a linear regression was fitted by NB and amino acid intake (AAI). The maintenance requirements were estimated as the AAI to maintain the NB equal to zero. The daily amino acid requirements for maintenance were estimated to be Lys 11, Met 29, Thr 23, Trp 5, Arg 50, Val 29, His 6, Gly 54, Phe 49, Leu 78 and Ile 21 mg/BWkg0.75 per day. Therefore, the amino acid ratio for maintenance was concluded to be Lys 100, Met 276, Thr 220, Trp 48, Arg 467, Val 275, His 60, Gly 511, Phe 467, Leu 735 and Ile 198% independent of the scale. The essential amino acid profile and the ideal ratio for the maintenance of poultry estimated in this study contributed to improve the factorial model for estimating essential amino acid requirements for poultry.  相似文献

The relationships among BW, backfat depth, and body physical and chemical composition were evaluated in response to dietary protein and DE balance in breeding gilts from 30 kg of BW to weaning of the first litter. Large White (sire) x Landrace (dam) F1 hybrid (White; n = 75) and Landrace (sire) x (Meishan x Large White; dam) (Meishan; n = 19) hybrid gilts were received at 30 kg of BW. Five gilts were taken as the initial slaughter group at 30 kg of BW, and the remaining gilts were fed diets differing in total lysine to DE ratio, high (H) vs. low (L), from 30 kg of BW to mating (rearing), and during gestation and lactation, allowing factorial investigation of dietary treatment effects and interactions during rearing, gestation, and lactation. Gilts were slaughtered at approximately 50 and 90 kg of BW, and at mating, farrowing, and weaning. Gilts fed L diets during rearing were lighter at mating (117.9 vs. 133.6 kg of BW, P = 0.035) due to a reduction in gain (592 vs. 720 g/d, P = 0.002) and a restriction in protein accretion (83 vs. 117 g/d, P = 0.001). During rearing, lipid accretion did not differ between L- and H-fed gilts (208 vs. 198 g/d, P = 0.60), but the ratio of lipid to protein accretion was about 1.5-fold greater in L-fed gilts, where lipid mass expressed as a percentage of BW was increased at mating (26.0 vs. 21.9%, P = 0.005). Effects of L diets on lipid accretion during rearing were transient; no residual effects on body lipid mass (P > 0.17) were found at farrowing or weaning. Overall, Meishan hybrids carried greater lipid mass (P < 0.001) than White hybrid gilts. Whereas the rate of body lipid and protein accretion and body lipid and protein mass can be nutritionally influenced and can vary according to growth stage, reproductive status, and genotype, this study established that body protein mass expressed as a proportion of the lipid free empty BW remains inflexible. A value for this measure of 0.188 +/- 0.0052 was found in White and Meishan hybrid gilts ranging from 28 to 203 kg of BW and 3 to 36 mm backfat depth, covering growth, pregnancy, and lactation, and offered diets differing in protein and energy balance. Body protein mass can be predicted as approximately 0.2 of the lipid free empty BW once body lipid mass is estimated accurately from physical measurements, such as backfat depth (P2, mm) and BW (kg), by regression using lipid (kg) = - 8.14 (SE, 1.302) + 0.167 (SE, 0.010) BW + 0.883 (SE, 0.065) P2 (residual SD = 3.51; R2 = 0.912).  相似文献
