通过对热带小奥德蘑深层发酵培养基碳源、氮源和无机盐的筛选 ,利用正交试验得到了优化发酵培养基配方 ,在此基础上对发酵条件进行了探索。试验结果表明 ,热带小奥德蘑的适宜发酵培养基为 :葡萄糖 3%,蛋白胨 0 .4 %,KH2 PO40 .1%,MgSO4·7H2 O 0 .2 %,VB10 .0 1%。其适宜发酵温度为 2 8℃ ,起始pH5 .0~ 6.0 ,摇瓶装量 60mL 2 5 0mL ,接种量 10 %。 相似文献

榆黄蘑(Pleurotus citrinopleatus Si-ng.)是北方著名食菌,已人工栽培。为不断提高栽培榆黄蘑的产量和品质,对其子实体的形成与孢子散布进行初步研究。材料与方法供试菌株系本所自长白山林区分离的菌株。采用瓶栽法,培养料取阔叶树木屑4份,麦麸1份(按重量比)充分混匀,加水适量(加干料重的100-160%的清水),装罐头瓶,每瓶约装湿料6两,加压灭菌(15磅,2小时)接种后置于24±2℃条件下培养,约经3周左右,观察原基形成与子实体形态变化。 相似文献

对采自广东省鼎湖山的一株野生小奥德蘑(Oudemansiella sp.)进行分类鉴定与栽培,研究其液体培养菌丝体的水提液、醇提液和发酵液的抗氧化活性。采用形态学特征与rDNA-ITS序列分析相结合的方法,将其鉴定为厚褶小奥德蘑(O.crassifolia)。利用栽培料(58%杂木屑、30%棉籽壳、10%麸皮、2%碳酸钙,含水量65%,pH自然),在(25±1)℃条件下遮光培养20 d,菌丝满袋后再培养15~16 d,转入出菇房,在25~28℃、相对湿度85%~90%条件下,每日300~500 lx光照10 h,菇蕾产生后,每日通风3~4次,出菇3~4潮,生物学转化率达到79.15%。本研究中,厚褶小奥德蘑菌丝体水提液的还原力和对羟自由基的清除能力明显高于菌丝体醇提液和发酵液,与对照维生素C相比,厚褶小奥德蘑菌丝体水提液的还原力低于维生素C,而对羟自由基的清除能力是维生素C的1.81倍。 相似文献

本文对木屑栽培香菇优质、高产、稳产技术进行初步研究,得出:(1)Cr20、Cr04、L26、0.87、L7等五个菌株是木屑栽培香菇高产优质的菌株;(2)以阔叶树木屑76%、麦皮20%、蔗糖1.5%、碳酸钙2%、过磷酸钙0.2%、尿素0.3%为较佳配方,生物效率高达150%;(3)装袋前培养料先堆制发酵处理是控制污染的最有效方法;(4)菌袋喷施浓度25ppm的柠檬酸、10ppm的三十烷醇、20ppm赤霉素溶液,能促进香菇子实体原基分化,增产效果显著。 相似文献

以北京市怀柔区宝山镇野生墨汁鬼伞(Coprinus atramentarius)为试验材料,对其母种、原种培养基配方进行优化筛选,并采用不同栽培方式研究了野生墨汁鬼伞人工驯化的培养条件。结果表明:实验范围内供试菌株的菌丝体生长最适碳源为葡萄糖+玉米淀粉,最适氮源是蛋白胨,最适pH值为9。培养料配方以棉籽壳、麸皮、玉米粉、石灰、锯末、石膏的配方最好,出菇以菌棒立栽覆土效果最好,覆土后55d现菇蕾,3~5d可采收。第一潮出菇生物学效率可达40%左右,条件适宜可出3~4潮菇。 相似文献

AIM:To study the inhibitory effect of IGF binding protein-4 (IGFBP-4), especially in corporation with heparin on the pathopoiesis of insulin-like growth factor-1 (IGF-1) in alveolitis and fibrosis. METHODS:The diploid human embryonic lung (HEL) fibroblasts were incubated respectively with control, 100 μg/L IGF-1, 100 μg/L IGF-1+100 μg/L IGFBP-4, 100 μg/L IGF-1+200 μg/L IGFBP-4, 100 μg/L IGF-1+100 μg/L IGFBP-4+100 μg/L heparin, 100 μg/L IGF-1 + 100 μg/L IGFBP-4+200 μg/L heparin, 100 μg/L IGF-1+100 μg/L heparin, 100 μg/L IGF-1+200 μg/L heparin for 24 h. Then the content of glucose transporter-4 (GLUT-4), hexokinase-2 (HK-2), collagen-4 and elastin were detected, respectively. RESULTS:Compared with control group, HK-2, GLUT-4, elastin and collagen-4 expressed in IGF-1 group were increased obviously. The expression in the group of IGFBP-4 plus IGF-1 was more than that in IGF-1 group. However, all expression was depressed strikingly when heparin was added. CONCLUSION:(1) IGF-1 apparently stimulates HK-2, GLUT-4, elastin and collagen-IV secretions from lung fibroblasts. (2) The intact IGFBP-4 associated with heparin can inhibit the pathopoiesis of IGF-1. 相似文献

在碳素含量为2%、氮素含量为0.06%的条件下,采用液体静止培养方法,研究了4个猴头菌菌株对碳、氮营养的利用。结果表明,菌丝营养生长对有机的利用比对无机氮强,有机氮中,其对复合氮素加蛋白胨、干酪素、酵母膏的利用比对单一氮如尿素、谷氨酰胺强。无机氮中,其对铵态氮利用较强,硝态氮基本上不能被利用。猴头菌对碳素营养的利用广泛,在多糖中对糊精的利用最好,其次是可溶性淀粉,对纤维素的利用较差,单糖中的甘露糖 相似文献

AIM: To explore the histochemical changes of diabetic skin and the pathogenesis of impaired wound healing in diabetes. METHODS: 54 male Sprague-Dawley (SD) rats weighing 200-220 g were randomized into control and STZ-induced diabetic groups. The shaved skin specimens from the back of rats were collected in 4, 8 and 12 weeks post STZ-induction, respectively. Hematoxylin-eosin dye was used for histological examination. Meanwhile, the skin glucose contents were measured by Beckman’s autoanalyzer. Skin AGEs concentrations were assessed by detecting total fluorescence in tissue collagen and immunohistochemistry assay. RESULTS: The skin thickness in diabetic animals was decreased, with the features of multilayer epithelium structure disappeared in epidermis and collagen fibers atrophied, swollen and degenerated in dermis; The inflammatory responses in the dermis of diabetic animals were increased obviously. The results also revealed that skin glucose contents in diabetic rats [(2.64±1.03)mg/g skin] were 2-3 times higher than those in the controls [(0.74±0.33)mg/g skin] (P<0.01). The collagen fluorescence and AGEs positive expressions in diabetic skin enhanced significantly when compared with age-matched controls over the whole experimental course (P<0.05). CONCLUSIONS: A histochemical changes had already been occurred in diabetic skin before injury, which may be result from the local biochemical factors such as high concentrations of glucose and AGEs. These might be an important mechanism in the pathogenesis of impaired wound healing in diabetes. 相似文献

Chlorosis and necrotic spotting develop on the foliage of particular cultivars of potato (Solanum tuberosum L.) when grown under constant light. 'Kennebec', a cultivar severely injured by constant light when propagated from tissue-cultured plantlets, also was injured when plants were propagated from small tuber pieces (approximately 1 g). However, plants did not develop injury when propagated from large tuber pieces (approximately 100 g). Plants from large tuber pieces grew more rapidly than plants from small tuber pieces. The role of plant vigor and carbohydrate translocation in controlling injury development is discussed. 相似文献

以悬浮培养细胞及叶片作为分离原生质体的起始材料 ,对影响苹果原生质体分离和培养的因素进行了系统研究。优化的原生质体培养基为 :改良MT 维生素C 5mg/L BA0 .5~ 1mg/L 2 ,4 D 0 .2mg/L 蔗糖 0 .6 5mol/L 谷氨酰胺 50 0mg/L CH 10 0mg/L ME50 0mg/L ;以葡萄糖代替蔗糖作渗透压调节物质效果好 ,且在培养过程中不需降压 ;适宜的低密度培养 (0 .5× 10 5/mL)可减少褐变。悬浮系原生质体培养 5~ 6d出现第 1次细胞分裂 ,15~ 2 0d形成多细胞团 ,4 0~ 50d形成肉眼可见的小愈伤组织。经培养诱导出不定芽 ,并进一步诱导生根长成完整植株。获得了平邑甜茶、M2 6 、嘎啦 3种基因型的原生质体再生植株。 相似文献

对采自西藏自治区日喀则市高海拔地区的野生双孢蘑菇(Agaricus bisporus)进行驯化。结果表明:试验范围内,西藏野生双孢蘑菇的最适母种固体培养基配方为200.0g去皮马铃薯、20.0g葡萄糖、3.0g蛋白胨、1.0g KH_2PO_4、0.5g MgSO4、20.0g琼脂、1.0L水、pH自然,菌丝生长速度最快为0.21cm/d。最适原种培养基配方为93%麦粒、5%牛粪粉、1%石膏、1%石灰,原种的生长周期是28d。在栽培试验中,子实体的单位面积产量是8.71kg/m~2及平均单菇重47.56g,子实体颜色以浅褐色为主。 相似文献

以云南省临沧市蚁巢伞中分离获得的菌株0820-003为供试材料,根据形态学特征和ITS序列分子鉴定分析,确定其分类学地位,并设计3种不同液体培养基,筛选其最佳液体培养条件。结果表明,该菌株0820-003为小果蚁巢伞Termitomyces microcarpus (Berk.&Broome) R. Heim,在3种配方中,适合菌株0820-003生长的液体条件为配方C,即土豆200 g、葡萄糖20 g、酵母菌2 g、蛋白胨1 g、KH2PO41.0 g、MgSO41.0 g、维生素B110 mg,加水至1 000 mL,pH自然。 相似文献

以8个不同采集地来源的暗褐网柄牛肝菌(Phlebopus portentosus)菌株为试验材料,分别进行了菌丝固体培养、液体培养初筛和出菇复筛试验。结果表明:8个供试菌株在M1琼脂培养基(200g马铃薯、20g葡萄糖、2g酵母膏、1g MgSO_4、1g KH_2PO_4、20g琼脂、1000mL水)上菌丝长势均粗壮浓密,在M1液体培养基(M1琼脂培养基不加琼脂)中,除菌株Cy3和Cy6(因菌丝生长缓慢或几乎不继续生长而被淘汰)外其它6个菌株均生长良好;出菇试验中,菌株Cy4表现优良栽培性状:菌丝在培养料中长势强,子实体产量高[单菇重(101.0±5.2)g,成熟率95%],商品性状好(菌盖黑褐色、菌柄黄色粗壮和基部稍膨大)。 相似文献
