Oral administration of 17β-oestradiol at 20 mg/kg diet for a period from 0 to 60 days following “swim-up” resulted in the sex reversal of male brook trout and the production of essentially all-female populations. The gonads of these animals were indistinguishable from the ovaries of untreated fish.Oral administration of this hormone at the same concentration but for the period from 0 to 40 days resulted in a population which comprised 67% females, 21% males and 12% bisexual animals.Within the control group, the growth rate of males was significantly greater than that of females. Growth in the all-female stocks produced by dietary oestradiol administration (0–60 days) was significantly lower than that of the untreated population taken as a whole but was identical to that of the control females.The growth of brook trout during the first year was closely comparable to that of rainbow trout reared under identical conditions. During the second year, the growth of the rainbow trout exceeded that of the brook trout. At 19 months of age, all the animals in the control and oestradiol treated brook trout populations were fully mature; maturity in the rainbow trout was substantially confined to males.  相似文献

Plasma cortisol levels and the number (Nmax) and affinity (Kd) of specific hepatic cortisol-binding sites were determined in rainbow trout subjected to chronic confinement stress for 14 days. Confinement significantly elevated plasma cortisol levels to 47.3 ± 13.5 ng ml–1 within 24h and although levels declined to 8.0 ± 3.0 ng ml–1 after 14 days, they were significantly higher throughout than levels in unstressed control fish (< 2.0 ng ml–1). There was a 60% reduction in cytosolic Nmax in stressed fish during the first 24h of confinement (35.8 ± 7.9 cf. 129.0 ± 15.2 fmol mg–1 protein), a decline which was sustained at 7 days after the onset of stress but, although numbers of binding sites in the liver of stressed fish were still lower than in unstressed fish, the difference was no longer significant after 14 days of confinement. There was an accompanying significant rise in the Kd of cortisol binding in stressed fish during confinement, from 4.0 ± 0.6 nM at time 0 to 8.4 ± 0.8 nM after 24 h confinement. This increment in Kd was sustained at a level significantly higher than in control fish throughout the 14 day confinement period, despite marked reductions in cortisol levels and increases in Nmax in stressed fish. Throughout the study, specific binding of cortisol could not be consistently detected in high-salt nuclear extracts from stressed or unstressed fish, suggesting either that high-affinity binding sites for cortisol were absent from these preparations, that receptors were present but unable to interact with ligand because they were occupied, or that receptors were present but not being extracted. These possibilities were investigated using a range of extraction procedures, by varying the temperature of incubation, by employing dexamethasone as ligand and by examining binding in purified, intact, nuclei. Estradiol was employed as a methodological control throughout and substantial amounts of specific estradiol binding were detected in all compartments and preparations. Specific cortisol-binding sites were detected in intact nuclei of both stressed and unstressed fish, at levels an order of magnitude lower than estradiol binding in the same preparations. These data demonstrate that activation of the pituitary-interrenal axis leads to significant changes in the nature of target-tissue binding of cortisol in rainbow trout, and reveal a clear difference in the subcellular distribution of binding-sites for estradiol and cortisol, which reflects the situation in mammalian tissues.  相似文献

The magnitude of the cortisol response to a standardised stressor is a heritable trait in salmonid fishes, and selection for stress responsiveness induces differences in both behaviour and neuroendocrine function. For instance, in laboratory studies, fish with a high cortisol response show a greater propensity for stress induced anorexia. Some authors have, however, commented that controlled studies encompassing relatively small groups of fish hold little or no relevance to practical aquaculture. This notion may be flawed, since understanding the mechanisms behind the behaviour of individuals is a proviso to predict behaviour in groups even with the caveat that some behaviors may be modified by group size. As an example, optimal feeding regimes should be easier to predict in a population consisting of individuals whose appetite is relatively less affected by external stressors. In a fluctuating and potentially stressful environment, such a population should also generate less feed waste, if kept on fixed rations. In the present experiment, we tested this hypothesis by monitoring feed waste and feed conversion efficiency in lines of rainbow trout selected for a low (LR) or high (HR) cortisol response to stress. The study was carried out after fish had been transported between rearing sites in the United Kingdom and Norway. There was significantly more feed waste from rearing units containing HR fish, and these fish also showed lower feed efficiency (growth per unit feed consumed). The difference in feed waste became more apparent with increasing time after transport, as rations increased. Simultaneously, size was more variable and growth was slower in HR rearing units. These results suggest that there are several potential benefits of selection for low stress responsiveness in aquaculture.  相似文献

Hybridization experiments were carried out by crossing females of common rainbow trout with the males of albino rainbow trout. Survival rates in egg, hatchling, fry and yearling stages, and the annual growth rate were higher than those of either parent.Reciprocal crosses with females of albino rainbow trout and males of common rainbow trout showed poor survival rate. The effects of a 21 day infestation with the ectoparasite Argulus foliaceus on the response of rainbow trout Oncorhynchus mykiss to an acute stressor (confinement) were investigated. Fish were divided into a control and two parasitized groups, with one parasitized group having received cortisol feeding 72 and 24 h before infection. Prior to confinement only control fish showed a significant increase in body mass and condition factor. Following 4 h confinement, plasma glucose levels in the normal- fed/infested group were significantly higher than in the controls, while the cortisol-fed/infested group had significantly lower plasma levels of adrenocorticotropic hormone (ACTH) and glucose. Both infested groups, but not controls, had higher plasma ACTH and cortisol levels 48 h after confinement when compared with their pre-confinement levels. Stressor-induced immunomodulation was more evident in the parasitized fish after confinement, as was concluded from decreased lysozyme activity, alternative complement activity and oxygen radical production either immediately after, or 48 h post 4 h confinement. Circulating lymphocyte numbers were significantly reduced in the control and normal-fed/infested fish. Results indicate that low ectoparasitic infestations can, for a prolonged period, lead to a stress experience with effects that impair the response to subsequent stressors. Such effects may be further confounded by a previous elevation of plasma cortisol levels.  相似文献

Triploid rainbow trout exhibit improved survival and extended growth during sexual maturation, compared to their diploid counterparts. However, there have been few benefits demonstrated prior to sexual maturation. This study was undertaken to investigate the possibility of improving growth and survival parameters in triploids through interstrain crosses. Triploids were induced by heat shocking fertilized eggs from intra- and interstrain crosses of two rainbow trout strains. The four triploid groups and their diploid counterpart groups were reared to 233 days post-hatching and analyzed for growth and survival characteristics. Compared with diploids, triploids had significantly (P < 0.05) higher mortalities during the first 100 days post-fertilization, primarily just prior to and after hatching. However, during the remainder of the study triploids exhibited significantly (P < 0.05) lower mortalities than diploids. During the first 50 days of rearing all four triploid groups were significantly shorter and lighter than their diploid counterparts. The growth of the triploid groups later in the study varied considerably. At the conclusion of the rearing phase, one interstrain triploid group was significantly (P < 0.05) longer than its diploid counterpart, although not significantly heavier. The other triploid groups were either significantly smaller than, or equal to the diploids. Analysis of variance indicated that the growth of triploid rainbow trout was significantly affected by maternal strain effects. These results suggest that the use of specific strains and crosses may improve the growth of triploid rainbow trout.  相似文献

Virgin female rainbow trout, Oncorhynchus mykiss, were unilaterally ovariectomised at various stages of ovarian development to investigate the effect of the removal of one ovary on subsequent recruitment and growth of follicles in the remaining ovary. The right ovary was removed from groups of 12–15 fish, 12, 7 and 4 months before they were due to ovulate, and the gonadosomatic index and follicle number and size determined just prior to ovulation. There were no differences in fecundity or follicle size in fish unilaterally ovariectomised at 12 and 7 months prior to ovulation compared to the controls. However, in the females unilaterally ovariectomised 4 months prior to ovulation, the remaining ovary either had the normal number of follicles for a single ovary but of a significantly larger size than follicles in the controls, or alternatively had almost 70% more than the normal number of vitellogenic follicles but comprising two distinctly different size populations. Differences in plasma oestradiol-17β concentrations at the final sample were seen only in females unilaterally ovariectomised 4 months prior to ovulation, where the levels were significantly lower than both the sham operated and control fish (p < 0.05). These data show that in the rainbow trout, complete compensatory ovarian hypertrophy following unilateral ovariectomy can occur throughout a major part of ovarian development, but that follicle recruitment is limited to stages up to (and therefore fecundity is determined by) mid-vitellogenesis.  相似文献

Northern Lake Huron (Ontario, Canada) supports the largest concentration of freshwater salmonid cage culture farms in Canada and receives inputs of domestic‐strain rainbow trout (Oncorhynchus mykiss) through escapement. We assessed the potential effects of these domestic fish on the naturalised rainbow trout of this ecosystem by comparing their respective ecologies. Mature adults were sampled from spawning tributaries, primarily in spring, and to a lesser extent in fall. Fish of domestic origin comprised ~80% of rainbow trout sampled from sites near cage culture facilities but <20% of rainbow trout sampled from more distant sites. Domestic adults in spawning condition (gametes free‐flowing) were present in the spawning tributaries in both spring and fall sampling seasons, whereas naturalised fish in spawning condition were only observed in the spring. Domestic adults were younger and smaller (in length), appeared to have shorter lifespan and had a higher male to female ratio compared with their naturalised counterparts. Growth rates (change in length with age) of naturalised and domestic females in the wild were similar, but domestic males grew more slowly than naturalised males. Domestic females also produced smaller eggs than naturalised females. Food web positions (inferred from δ13C and δ15N) of domestic and naturalised fish were very similar but varied more strongly with body size in the former. Domestic‐strain rainbow trout of cage culture origin can survive, grow and attempt to spawn in northern Lake Huron and have the potential to compete for food, mates and spawning habitat with naturalised rainbow trout.  相似文献

The present study investigated the effects of propolis and vitamin E supplementation in diets of juvenile rainbow trout subjected to two different flow rates (0.9 and 2.1 L min?1) on growth performance, and vitamin A, C and E concentrations in tissues as well as malondialdehyde (MDA) levels. Juvenile rainbow trout were fed with diets containing 10 and 30 g propolis kg?1, 60 mg kg?1 vitamin E (Rovimix E‐50 adsorbate; min.%50 dl‐α‐tokopherly acetate) and without supplemented basal diet for 12 weeks. Weight gain (WG) in the C group was significantly lower (P < 0.05) than P10, P30 and E60 groups at both flow rate treatments. At 2.1 L min?1, specific growth rate (SGR) in the C group was significantly lower (P < 0.05) than other groups, but at 0.9 L min?1, SGR of fish did not differ among the diets groups (P > 0.05). Survival rate (SUR) was higher in propolis and vitamin E supplemented diet groups (P < 0.05) than control diet group at 0.9 L min?1. Fish fed on diet E60 had higher (P < 0.05) tissue vitamin E concentration than fishes fed on other diets groups. Vitamin C concentration in rainbow trout tissues was significantly affected by the 30 g propolis supplemented diet group (P < 0.05), followed by the 10 g propolis supplemented diet group (P < 0.05). MDA level of E60 group was found significantly decreased instead of different than other groups (P < 0.05). The results of Student's t‐test revealed that WG, SGR, SUR values, vitamin (A, C, E) concentrations and MDA levels of tissues were negatively affected by 0.9 L min?1flow rate treatment in juvenile rainbow trout.  相似文献

This study was conducted to evaluate the effect of dietary eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) levels on the fatty acid composition, salinity tolerance and antioxidant status of rainbow trout (Oncorhynchus mykiss). Four diets were formulated with total EPA and DHA contents of 5.41, 9.55, 13.97 and 17.88 g/kg (abbreviated as ED‐5.41, ED‐9.55, ED‐13.97 and ED‐17.88 respectively). Rainbow trout (initial weight of 90.61 ± 9.25 g) were fed the experimental diets for 8 weeks to accumulate significant differences in fatty acid composition and subsequently underwent salinity acclimation. Our results indicated that high dietary EPA and DHA significantly improved the EPA and DHA content in fish tissues. The serum osmolality of fish returned to their freshwater values in the ED‐9.55, ED‐13.97 and ED‐17.88 groups. The Na+, K+‐ATPase (NKA) activity of fish in the ED‐13.97 group changed dramatically to adapt the fish to the hypertonic environment. Moreover, there was no significant difference in the serum cortisol concentration and liver catalase (CAT) activity of fish in the ED‐13.97 group during salinity acclimation. The liver superoxide dismutase (SOD) activity in the ED‐13.97 group was significantly higher than that in ED‐5.41 and ED‐9.55 groups at the end of salinity acclimation. The muscle malondialdehyde (MDA) content in the ED‐13.97 group was significantly lower than that in the ED‐17.88 group before salinity acclimation and significantly lower than the ED‐5.41 and ED‐17.88 groups on day 7 of acclimation. The results of this study indicate that the rainbow trout in the ED‐13.97 group exhibited optimal salinity acclimation performance.  相似文献

Measuring baseline cortisol in wild salmonids is problematic because of the stress induced by most capture methods. The cortisol response to different periods of confinement stress was modelled in a laboratory population of rainbow trout [Oncorhynchus mykiss (Walbaum)]. A quadratic model was used to predict baseline (precapture) cortisol levels in these fish. A similar quadratic response pattern of cortisol was observed in wild sea trout (Salmo trutta L.). These were captured in timed sequences on four dates by seine netting in Clew Bay, west coast of Ireland. The estimated precapture cortisol for these fish indicated higher levels soon after migration to sea and a positive correlation with the level of infestation by Lepeophtheirus salmonis (Krøyer). Curvilinearity of the quadratic model was correlated with levels of lice infestation, indicating an enhanced cortisol response to confinement stress during capture when parasitic abundance was high. Estimated precapture cortisol for individual sea trout was positively correlated with numbers of lice although the linear regression had low predictive power. The method provides, for the first time, a means to estimate baseline cortisol levels in wild salmonids, and addresses one of the most difficult problems in working with wild fish. Flavobacterium psychrophilum is one of the most important pathogens affecting cultured rainbow trout (Oncorhynchus mykiss). Recent information from UK salmonid farms showed country‐wide distribution of genetically and serologically divergent clones, which has hampered the development of a vaccine for rainbow trout fry syndrome. The current study assessed the efficacy of an injectable polyvalent vaccine containing formalin‐inactivated F. psychrophilum in rainbow trout. The vaccine was formulated with an oil adjuvant (Montanide ISA 760VG) or formalin‐killed cells alone. Duplicate groups of trout (60 ± 13 g) were given phosphate‐buffered saline or vaccine formulated with Montanide by intra‐peritoneal (i.p.) injection and challenged by intra‐muscular (i.m.) injection with a homologous and a heterologous isolate of F. psychrophilum at 525 degree days post‐vaccination (dd pv). Significant protection was achieved in vaccinated fish (p = 0.0001, RPS 76% homologous, 88% heterologous). Efficacy of the adjuvanted vaccine was also demonstrated by heterologous challenge at 1155 dd pv resulting in 100% protection, whereas survival in the un‐adjuvanted group was not significantly different from control fish. Levels of specific antibody at 1155 dd pv, as measured by ELISA, were significantly higher in the fish vaccinated with adjuvant when compared with unvaccinated fish.  相似文献

The winter is often considered as a survival bottleneck for stream‐living fish. Juvenile salmonids generally become less active during this period, and while food intake continues to some extent, growth rates are typically low. Here we present the results of an over‐winter field experiment where energy levels were manipulated in late autumn. Three groups of juvenile (age 1+) brown trout, from an anadromous population, were monitored with respect to over‐winter growth rate and survival (as indicated by recapture rates). Two groups were fed either high (HR), or low (LR) food rations in the laboratory for a month (October); the third group remained in the stream (STR). Over‐winter growth rates were relatively low in all groups, and no growth compensation could be detected. Compared to HR and LR, STR fish had higher recapture rates after winter, indicating that laboratory housing may have affected the subsequent stream survival negatively. Comparing the two laboratory‐housed groups, the LR group reached similar condition as the HR group in early spring, without indications of differences in survival. However, the initiation rate of body silvering (indicating initiation of smoltification) was lower in the LR group. Thus, it appears that food restriction during late autumn affect the onset of smoltification in juvenile brown trout. The results support previous laboratory studies indicating that salmonids modify their over‐winter foraging behaviour to avoid too low energy levels at the end of winter. This modification appears to delay smoltification, but may not necessarily be costly in terms of over‐winter mortality.  相似文献

We studied partial replacement of fish meal (FM) by krill meal (KM) and how fluoride from KM enriched-muscles and bones of rainbow trout reared in freshwater affected these fish. Diets that replaced FM with KM at proportions of 0, 7, 15 and 30% (control, KM7, KM15, and KM30) were fed to groups of rainbow trout for 92 days and growth was observed. In fish fed KM7 and KM15, weight gain (WG), feed intake (FI) and specific growth rate (SGR) were unchanged compared with fish fed the control diet, but in fish fed KM30, WG, FI and SGR significantly decreased. After the experiment, fluoride concentration in dorsal muscles of each experimental group was below the detected limit (1 mg/kg), but in vertebral bones, the fluoride concentration increased with each increase in KM inclusion in the diets: 490 mg/kg (control), 755 mg/kg (KM7), 1100 mg/kg (KM15), and 2400 mg/kg (KM30). Tissue specimens of liver of each experimental group had no histopathological changes. Therefore, accumulation of fluoride in vertebral bones apparently adversely affected growth of the vertebral bones thus affecting the growth performance as shown by the decrease in WG, FI and SGR.  相似文献

Thermal shock‐induced triploid and unshocked control hybrids between rainbow trout (Oncorhynchus mykiss) and Black Sea trout (Salmo labrax) and their parental species were produced under hatchery condition by using heat shocks. Triploidization reduced eyed egg rate and alevin yield in all groups. Low survival rate was observed in both shock‐induced triploid hybrid and non‐shock‐induced control hybrids. Although hybrids demonstrated low body weight during the first feeding stage, they reached higher body weight by day 200 when compared with Black Sea trout and rainbow trout. A higher specific growth rate was calculated as 3.60 in the triploid hybrid groups, 1.41 in the triploid Black Sea trout groups and 2.27 in the triploid rainbow trout groups between days 110 and 200. A lower condition factor was determined in the hybrid than in the diploid parental species. A negative value of mid‐parent heterosis (MPH) performance was deected for condition factor, and a favourable MPH was detected for specific growth rate and weight in hybrids.  相似文献

Rainbow trout were fed a diet containing phytase-sprayed and phytase-pretreated soybean meal with different phytase levels. The single factor random block design was used to analyze the effects on rainbow trout of dietary phytase supplementation on growth performance, nutritional ingredient digestibility and nutrient excretion. After 90 days, the results showed that feed conversion ratio (FCR) and protein efficiency ratio (PER) were significantly improved and specific growth rate (SGR) was not affected by spraying phytase, but SGR, FCR and PER were not significantly improved by phytase pretreatment. A digestibility trial conducted after the feeding trial showed that apparent digestibility coefficient (ADC) of diet protein and minerals was increased with phytase supplementation. However, there was a negative effect of phytase on the ADC of lipid. The excretion experiment showed that the supplementation of phytase resulted in decreased nutrient excretion in feces, but lipid excretion was slightly increased with phytase supplementation. In addition, the results of P excretion and ADC of P analyzed by t-test showed that phytase pre-treatment method should be a more rational method than the spraying method. The results of SGR, ADC of P and P excretion analyzed by quadratic regression indicated that 2,000–3,000 U/kg levels by the spraying method could be a rational range of phytase supplementation, and about 1,000 U/kg should be an optimal level by the pretreatment method. Thus, use of phytase in rainbow trout feeds can have economic and environmental benefits.  相似文献

This study was conducted to evaluate the effects of Echinacea purpurea (EP) on growth and some hematological and blood biochemical indices of rainbow trout fingerlings. A basal diet was supplemented with 0 (control), 0.25, 0.5, 1, and 2 g EP kg?1 to formulate five experimental diets. Each diet was randomly allocated to triplicate groups of fish with an initial average weight of approximately 8 g. After 8 weeks of feeding trial, fish fed diets with 0.25 and 0.5 g EP kg?1 showed highest final weight and SGR and fish fed with the control diet indicated the lowest final weight and SGR. There was a significant (P < 0.05) decrease in FCR between the control group and the groups fed with diets of 0.25 and 0.5 g EP kg?1. Biochemical parameters such as serum total protein content, albumin content, globulin content, and albumin/globulin ratio in the fish were evaluated. There were significant differences between hematological parameters including RBC, WBC, HB, lymphocyte, and neutrophil percentage in fish fed with dietary nucleotide compared with control treatment (P < 0.05). The results suggest that EP administration at 0.25 and 0.5 g EP kg?1 exerted positive effects on growth and biochemical and hematological indices in rainbow trout.  相似文献

Long‐day photoperiods are considered as an effective managerial tool in manipulating of reproduction and somatic growth in a number of fish species. In this study, the effects of three different artificial photoperiods on the gonadal development and somatic growth of rainbow trout (Oncorhynchus mykiss L.) were investigated. Two years‐old immature female rainbow trout (279.94 ±2.25 g) were exposed to three artificial photoperiod regimes of 24L:0D, 18L:6D and 6L:18D and natural light (NL) regime for 5 months. The highest gonadosomatic indices were recorded in NL and 6L:18D groups while the rates were significantly lower in fish maintained under 18L:6D and 24L:0D photoperiods (< 0.05). Mean oocyte diameters in fish exposed to 24L:0D and also to 18L:6D were significantly lower than the 6L:18D and NL groups. Photoperiods with 24L:0D and 18L:6D regimes resulted in significantly higher mean final weights and specific growth rates (SGR) than NL regime. The highest mean final weight (635.45 ± 16.19 g) and SGR (1.03 ± 0.04% day?1) were obtained under 24L:0D photoperiod. Fish exposed to 24L:0D and 18L:6D showed the highest condition factor as 1.44 ± 0.01 and 1.44 ± 0.02 respectively, when compared with the NL (1.27 ± 0.01) and 6L:18D (1.34 ± 0.02) groups. Basically, the results suggested that continuous artificial lightning can be used as an influential factor in delaying gonadal development and enhancing somatic growth in rainbow trout during gonadal growth phase.  相似文献

Basal levels of plasma cortisol in unstressed salmonid fish are normally in the range 0–5 ng ml−1. An acute stress such as handling or 1 h confinement caused a temporary elevation of the plasma cortisol levels of both brown trout,Salmo trutta L., and rainbow trout,Salmo gairdneri Richardson, in the range 40–200 ng ml−1 with a return to basal levels within 24–48 h. The extent of the cortisol elevation in response to an acute stress was dependent upon both the species and strain of trout. Chronic stresses, such as prolonged confinement or crowding, resulted in an elevation of plasma cortisol levels to approximately 10 ng ml−1. Under these circumstances, blood cortisol levels remained elevated for periods of up to 4 weeks before acclimation finally occurred. It is shown, by means of intraperitoneal implantation of cortisol, that chronic elevation of plasma cortisol levels in the brown trout results in a dose-dependent increase in mortality due to common bacterial and fungal diseases. This effect is apparent at plasma cortisol levels as low as 10 ng ml−1, levels below those often reported as being representative of ‘unstressed’ fish. These findings are discussed in relation to the known immunosuppressive effects of corticosteroids in teleost fish.  相似文献
