To further improve the technology used in Pacific oyster farming, information is required on the response of different sized and aged oysters to various environmental changes. In this study a neutral red retention (NRR) assay was used to investigate the effects of size and age on the response of Pacific oysters to changes in water temperature and their recovery after exposure to different air temperatures. Results from moving oysters directly between water temperatures of 5°C and 15°C, 10°C and 20°C and 15°C and 25°C demonstrated that different water temperature change affect the lysosomal membrane integrity differently. The NRR times of large and small oysters transferred directly between 10°C and 20°C initially decreased significantly, and then increased to levels corresponding to the new temperature. In addition, NRR times in large oysters responded at a significantly slower rate than small oysters when they were transferred from 5°C and 25°C to 15°C water and between 10°C and 20°C water. Results from the air exposure experiments showed that, after exposure to air temperatures of 5°C, 15°C or 25°C, the lysosomal membrane integrity of large oysters recovered at a slower rate in 15°C water compared to small oysters. It therefore appears necessary to develop different management strategies for large (old) and small (young) oysters. Results from this and previous research also indicate that the NRR assay could potentially be used to develop a model to monitor and predict the performance of oysters on farms.  相似文献

This study compares the effect of two anti‐fouling treatments, hot water immersion (15 s at 60°C) and air drying (72 h) on the physiological status of the Eastern oyster Crassostrea virginica. The negative impact of hot water immersion was greater than that of air drying, but varied depending on the initial size of the oysters (40 vs. 60 mm shell height) and the time of the year (June vs. August). Groups treated with hot water exhibited a higher proportion of haemocytes with destabilized lysosomal membranes (HDLM; 47.5 ± 3.1%) than those exposed to air drying (37.5 ± 2.9%). This suggests that the oyster immunocompetency may be lowered by hot water immersion. Overall, the large oysters had lower HDLM values (32.9 ± 3.5%) than the small individuals in June (45.7 ± 2.8%) but similar values in August (46.6 ± 3.5%). Small oysters subjected to hot water immersion in June exhibited a 50% reduction in shell growth and a 50% mortality rate after one month. Our results indicate that air drying is more suitable than hot water immersion as an anti‐fouling treatment for <45 mm oysters.  相似文献

In this study the effects of exposure to two air temperatures (15 and 27.5 °C) and three graders commonly used in South Australia (Rotary, Flat Bed and Inside/Out) on Pacific oysters were investigated using post-exposure or post-grading catecholamine levels as a stress indicator. During the 48 h experimental period the circulating dopamine and noradrenaline levels of oysters exposed to 15 °C air temperature did not differ significantly from controls sampled directly from the 15 °C water tank. Conversely, in oysters exposed to 27.5 °C air temperature, dopamine and noradrenaline became significantly different from controls after 8 and 12 h air exposure, respectively, and then returned to the control levels at 24 h.Significant increases (P < 0.05) in circulating noradrenaline and dopamine levels were observed in oysters graded by the Flat Bed and the Inside/Out graders. The circulating noradrenaline concentrations in oysters subjected to the Flat Bed grader were significantly higher (P < 0.05) than those observed in the Rotary graded oysters and close to being significantly higher (P = 0.052) than those levels measured in the Inside/Out graded oysters. The circulating noradrenaline levels in oysters subjected to the Inside/Out grader were, in turn, significantly higher than those recorded in the Rotary graded oysters. These results suggest that in relation to the three graders used in South Australia, the Flat Bed induces the highest stress levels in oysters and the Rotary grader the lowest.  相似文献

The trade of live Cancer pagurus in Portugal is totally dependent on imports, mainly from UK. Animals are transported for 48–58 h in conditions that promote high mortalities (40–60%) due to cumulative stressors. The present work aimed to optimize transport, by comparing the physiological responses of edible crab during different simulated live shipment systems for 48 h and recovery conditions: (a) immersed crabs at 12°C; and (b) air‐exposed crabs at 4°C and subsequent recovery at 16°C with and without pre‐acclimation. Both simulated transport systems induced physiological stress, culminating in 25% mortality in immersed conditions and only 4% in air‐exposed conditions. Both systems elicited anaerobic responses marked by hyperglycaemia and increased l ‐lactate concentration in haemolymph. At the end of the recovery period only air‐exposed animals resumed to l ‐lactate and pH control values, the lowest mortality was achieved with crabs acclimated at the lowest temperature (from 6 to 16°C at 1.5°C/h). Overall, results showed that crabs can be transported in air‐exposed conditions at 4°C and acclimated from 6 to 16°C and reach similar physiological conditions than the traditional transport in immersed conditions at 12°C.  相似文献

Episodes of summer mortality of the Kumamoto oyster Crassostrea sikamea are a major problem for its cultivation. Expression of the heat shock protein 70 (HSP70) is induced by various environmental stresses, including heat. We cloned and sequenced hsp70 complementary DNA from C. sikamea to investigate the relationship between hsp70 expression and heat tolerance in this oyster. Quantitative real-time polymerase chain reaction was performed using gill tissue dissected from oysters before and after heat shock for 1 h. The results showed hsp70 expression was faster and greater in oysters cultured at 20–22 °C than at 10–12 °C, and survival was lower among oysters cultured at 20–22 °C than at 10–12 °C. Moreover, heat tolerance was investigated by a 1-h pre-heat treatment, followed by exposure to heat shock conditions 5 days later. Survival was higher and hsp70 expression was notably lower in oysters that received the pre-heat treatment compared with those that did not. We conclude that a pre-heat treatment of only 1 h may be useful for inducing heat tolerance in C. sikamea, and that a low level of hsp70 expression after heat shock is an important index in selecting for high heat tolerance in these oysters.  相似文献

Light spatfalls of the Pacific oyster, Crassostrea gigas, have occurred in the estuaries of the Rivers Exe, Teign and Dart (Devon) and in the Menai Strait (Gwynedd), near to sites of commercial cultivation of this species. Comparisons of the sizes of the naturally settled oysters with hatchery oysters suggests that the spat from the River Teign and Menai Strait settled in 1989 and 1990. The springs and summers of these years were exceptionally warm with mean monthly air temperatures 0.8°-1.3°C above the 1951–1980 average for the period April to August. Although sitings of Pacific oyster spat were made in Loch Sween and Emsworth Harbour over 20 years ago, there is no evidence that self-sustaining populations have developed. The spatfalls which occurred in some coastal waters of Devon and Gwynedd in 1989 and 1990 are unlikely to sustain natural fisheries and are likely to die out as did the Portuguese oysters which occasionally settled in the River Blackwater, Essex.  相似文献

This study investigated potential application of lactic acid bacteria (LAB) in depuration for reducing Vibrio parahaemolyticus in oysters. Lactobacillus plantarum ATCC 8014, which exhibited strong bactericidal effects against V. parahaemolyticus in vitro, was added to artificial seawater for depuration of Pacific oysters (Crassostrea gigas) inoculated with V. parahaemolyticus BE 98-2029 (O3:K6) to levels of about 104 MPN/g at 15 ± 1 and 10 ± 1°C. Application of L. plantarum ATCC 8014 treatment (107 CFU/mL) in oyster depuration did not enhance reductions of V. parahaemolyticus in oysters depurated at 15 ± 1°C but significantly decreased (p < 0.05) levels of V. parahaemolyticus in oysters depurated at 10 ± 1°C after 5 days (3.40 log reductions) when compared with controls (2.75 log reductions). It is not clear if a competitive exclusion by LABs to compete with V. parahaemolyticus binding sites in oyster tissues plays a role in the reduction of V. parahaemolyticus in the oysters. Further studies utilizing different types of LABs in oyster depuration might provide additional knowledge for application of LAB in depuration for decontaminating V. parahaemolyticus in oysters.  相似文献

Tetraploid induction has been conducted on temperate oysters but not on tropical oysters. In this study, different heat shocks (32, 35 and 38°C) and cold shocks (1, 4 and 7°C) were used to induce tetraploidy in two tropical oyster species, Crassostrea belcheri and Crassostrea iredalei, through meiosis I inhibition. Temperature shocks were applied on the newly fertilized eggs at 8–10 min post fertilization and terminated when second polar bodies began to form in the control eggs. The ploidy of the larvae and spat was determined via direct chromosome count. The percentage of larval survival until Day 20 was low (between 0.4% and 42.9%) for both temperature shocks and oyster species. No surviving larva was recorded for induction at 1, 4 and 38°C. Tetraploid spat was only recorded in C. iredalei but the percentage is low through heat shock induction of 32 and 35°C. This study shows that the tetraploid induction success rate was slightly higher in C. iredalei compared to C. belcheri. No surviving tetraploid spat were recorded for both oyster species through the cold shock method. This study shows that heat shock can be used to inhibit meiosis for the production of tetraploids but more experiments need to be conducted to determine the optimum temperature when dealing with tropical oysters.  相似文献

Atlantic sturgeon (Acipenser oxyrhynchus), which are bottom dwelling and migratory fish, experience environmental hypoxia in their natural environment. Atlantic sturgeon, acclimated to either 5 or 15°C, were subjected to a 1 h severe (<10 mm Hg) hypoxia challenge in order to document their physiological responses. We measured hematological parameters, including O2 transport (hemoglobin, hematocrit), ionic (chloride, osmolality), and metabolic (glucose, lactate) variables under normoxic conditions (~160 mm Hg), immediately following a 1 h exposure to hypoxic water, and following a further 2 h of recovery from this challenge in normoxic water. In a second experiment, we assessed the opercular beat frequency before, during, and after hypoxic exposure. Hemoglobin concentrations and hematocrit were significantly different between fish held at 5°C vs. 15°C and also significantly different between normoxia prior to hypoxia and following recovery. Plasma lactate concentrations increased following hypoxia at both temperatures, indicative of an increase in anaerobic metabolism. In contrast, a significant increase in plasma glucose concentrations in response to hypoxia only occurred at 5°C, suggesting different fuel demands under different temperatures. Changes in opercular beat frequency (OBF) were dependent on temperature. At 5°C, OBF increased upon exposure to hypoxia, but returned to pre-exposure levels within 35 min for the remainder of the experiment. During hypoxia at 15°C, OBF increased very briefly, but then rapidly (within 20 min) decreased to levels below control values. Following a return to normoxia, OBF quickly increased to control levels. Overall, these findings suggest that Atlantic sturgeons are relatively tolerant to short-term and severe hypoxic stress, and the strategies for hypoxia tolerance may be temperature dependent.  相似文献

The production of the flat oyster Ostrea edulis (L.) natural spat in Europe has decreased almost by 60% in the past ten years. Thus, the importance of the production of oyster spat in hatcheries is evident. One of the critical steps in hatchery production is broodstock conditioning, especially difficult in autumn, when gonadal development is in resting period. Conditioning is influence by temperature, photoperiod and nutrition. In this work, the effects of two temperature and three photoperiod regimes on the conditioning of O. edulis were studied for three years by stereological analyses and registering number and dates of spawning and larval yield. Temperature had a positive effect on the gonadal development of O. edulis during conditioning. The percentages of germinal cells in oysters conditioned with a gradient of temperature (14–18°C) were double compared to oysters conditioned at 15°C. Oysters conditioned with longer photoperiods showed higher percentages of germinal cells. There was no interaction between temperature and photoperiod. Spawning was observed in the oysters treated with daylight (8–16 h) ten weeks from the beginning of conditioning. Flat oysters conditioned with 8 h and 8–12 h of daylight delayed the first spawning for a month. Total larval production was higher in the oysters treated with the longest daylight gradient. Gonadal and gametogenic development was a non‐synchronic process and the spawning extended for around two months. A protocol for flat oyster broodstock conditioning in autumn by using both a gradient of temperature (14–18°C) and daylight (8–16 h) is proposed.  相似文献

Oysters, Ostrea chilensis, infected with Bonamia exitiosus were held under stressful conditions for 14 days, and then under normal conditions for 3.5 months, to determine the affects of stress on B.exitiosus infection levels. The stressors used were (1) air exposure for 8 h daily; (2) hot water (25–26 °C), or (3) cold water (7 °C) for 1 h daily; (4) hyposaline (15‰), or (5) hypersaline (39–40‰) water; (6) starvation in filtered sea water; and (7) vigorous stirring four times a day. A control tank held oysters in static sea water, which was changed daily. Oysters were also (8) kept in a trough among heavily infected oysters (trough exposure), or (9) in a trough without other oysters (control), for 4 months. Oysters in hyposaline conditions all died within 3 weeks, but this was apparently unrelated to Bonamia infection. Otherwise, cumulative mortalities were highest in trough exposure, cold, hypersaline and hot treatments. There was a significant difference in prevalence between treatments, and the stir, hot, cold, hypersalinity and trough exposure treatments had significantly higher intensities of infection than controls. The mean intensity of Bonamia infection was significantly higher among female and spent oysters than in male and hermaphrodite oysters.  相似文献

In the present study, we investigate the short‐term adaptive physiological strategies to polycyclic aromatic hydrocarbon (PAH) of Pacific oysters, Crassostrea gigas, by exposing the oysters to varying levels of PAH (0, 50, 100 and 200 μg L−1) for 7 days with a 3‐day acclimation period under laboratory conditions. The filtration rate (FR) and respiration rate (R) increased significantly at 50 μg L−1 PAH and decreased at 100 and 200 μg L−1 compared with the control. The absorption efficiency (Abs. eff.) was significantly impaired at 200 μg L−1 PAH. Ammonia excretion (E) increased with increasing PAH levels, with a significant elevation at 200 μg L−1. Although a significantly elevated FR was observed, oysters exposed to 50 μg L−1 PAH showed scope for growth (SFG) similar to the control. This indicates that even at nominal levels, PAH contamination is a possible cause of reduced oyster production because of increased food demand. The oysters exposed to 200 μg L−1 PAH showed negative SFG values, which could be a possible cause of growth stagnation or even mortality when the exposure is chronic.  相似文献

1. Foveaux Strait, a narrow seaway that is exposed to heavy wave action and strong tidal currents, has been the subject of an oyster fishery for over 130 years. Before the oyster fishery commenced the seafloor was extensively covered by epifaunal reefs that were tidally‐oriented, linear aggregations of patch reefs. 2. Patch reefs are formed by the bryozoan Cinctipora elegans cemented by encrusting bryozoa, ascidians, sponges, and polychaetes. The molluscan epifauna is dominated by the oyster, Tiostrea chilensis and bysally attached bivalves. Mortality of oysters is probably lower and recruitment and growth may be higher within the reef habitat. 3. Fishers found commercial densities of oysters occurred only on epifaunal reefs. Fishers exploited local groups of reefs. These groups form the patchily distributed oyster beds characteristic of this fishery. 4. Dredging for oysters progressively modified reefs until oysters were the only epifauna remaining. Dredges caught oysters more efficiently after the catch bag no longer became saturated with other epifauna. This heightened efficiency allowed fishers to rapidly reduce oyster density to commercial extinction. Oyster density has not rebuilt on oyster beds abandoned by fishers. 5. The rate of modification of epifaunal reefs was slower during the early years of the fishery but has accelerated, especially over the last 37 years. Frequency of disturbance increased as the numbers of vessels fishing grew and fishers developed speedier dredging methods. Intensity of disturbance also increased as heavier dredges were introduced and allowed focused fishing of reefs. 6. Oysters became reduced to low densities in the eastern and central areas that fishers then abandoned. The commercially exploited area subsequently expanded to the limits of Foveaux Strait. 7. With accelerated modification of oyster habitat, disease mortality has become more important. 8. This is the first evaluation of growth and survival of spat of the Cortez oyster Crassostrea corteziensis (Hertlein) produced under controlled conditions in a coastal area in the state of Sonora, Mexico for aquaculture purposes. A suspended culture technique, used for the Pacific oyster C. gigas, was used. The Cortez oyster has an isometric shell growth during the first 13 months, reaching 71.3±1.9 mm length, 52.6±1.3 mm thickness and 25.1±0.8 mm width. Allometric growth was found between total weight and length, thickness and width (survival was 70%). The relationships between particulate organic, inorganic material, chlorophyll a and environmental parameters with growth are described. Growth rates of C. corteziensis were affected by temperature with retardation at less than 18°C. For aquaculture purposes, it is recommended that spat be sowed after winter, and oyster harvest occur at the end of autumn. According to the von Bertalanffy equation, Cortez oysters would reach the traditional exploitation size of 65 mm (mean length) at harvest. Finally, the results of this study have shown that C. corteziensis is a good candidate for aquaculture projects in this region.  相似文献

An ozonated water treatment system was designed for the sanitation of shucked oysters (Crassostrea plicatula). The effects of ozone (O3) on shelf life were examined under refrigerated conditions (4 ± 1°C). Microbiological, chemical, and color quality were also evaluated by determining total aerobic plate counts (APC), total volatile basic nitrogen (TVB-N), thiobarbituric acid (TBA), and L, a, and b values. Single and orthogonal experiment results showed that oysters treated with 9 mg/L of O3 in aqueous solution (oyster: ozonated water = 1:6, w/v) for 10 min at 5°C significantly reduced the total APC by about 2-log—from 4.97 ± 0.11 to 2.93 ± 0.17 log CFU/g (p < 0.05). O3 treatment did not cause significant changes in TVB-N, TBA, a, and b values, although L values were significantly affected. During refrigerated storage, O3-treated samples expressed lower TVB-N, TBA, a, and b values and higher L values than control samples. The shelf life of O3-treated samples was 20 to 25 days, while that for the controls was 5 to 10 days. The current study confirms that treatment with ozonated water can effectively prolong the shelf life and maintain the quality of oysters.  相似文献

The benefits of exposing eastern oysters to air during commercial culture have not been well‐characterized. An adjustable longline system (ALS) designed in Australia and recently adopted by the nascent aquaculture industry in the northern Gulf of Mexico, allows growing oysters at any position in the water column and is perfectly suited to study the benefits of air exposure. Four‐month old diploid oysters were deployed in an ALS and divided into three groups: 1) oysters exposed to air daily for 8–12 hr during low tide, 2) oysters exposed to air weekly (~24 hr once a week), and 3) oysters kept subtidally. Oyster mortality and growth rates, Perkinsus marinus load and condition index were then determined every 3 months over 2 years, while refrigerated shelf life and Vibrio vulnificus load were determined in summer and early fall of the second year. Summer mortalities were delayed, P. marinus infection intensities tended to be lower and condition index was significantly higher in oysters exposed to air daily compared with oysters exposed to air weekly or held subtidally. Shell heights of oysters exposed to air daily were lower for most of the study due to a lower growth rate during the initial sampling interval following deployment. No consistent differences were found in V. vulnificus loads or refrigerated shelf lives between the groups. It is recommended that ALS be set so that oysters are kept subtidally fall through spring to promote growth, and exposed to air daily during summer to delay P. marinus proliferation.  相似文献

为探讨干露胁迫对海产贝类基因组DNA甲基化的影响,应用荧光标记甲基化敏感扩增多态性(fluorescencelabeled methylation sensitive amplified polymorphism,F-MSAP)技术,比较了不同干露条件下(0 d、0.5 d、1 d、3 d、5 d、7 d、9 d和11 d)长牡蛎(Crassostrea gigas)基因组DNA甲基化的变化。结果表明,对照组(干露处理0 d)闭壳肌与鳃组织的总体甲基化水平分别为29.76%和29.82%;干露处理0.5 d、1 d、3 d、5 d、7 d、9 d和11 d的长牡蛎全基因组甲基化水平呈现先增高后降低的趋势,其中,闭壳肌组织的总体甲基化水平分别为36.59%、38.86%、43.02%、39.30%、51.13%、46.79%和35.06%,鳃组织总体甲基化水平分别为39.39%、42.13%、39.36%、43.54%、56.19%、38.57%和28.99%;干露处理7 d的长牡蛎甲基化水平明显高于其他时期(P0.05),11 d时甲基化水平基本恢复至初始状态。甲基化变异模式分析发现,闭壳肌与鳃组织DNA甲基化变异位点存在差异,甲基化升高位点变化程度较大(P0.05)。以上结果表明,长牡蛎通过改变DNA甲基化模式来应答干露胁迫,发生了不同程度的甲基化与去甲基化反应,DNA甲基化与长牡蛎的抗逆性状密切相关。  相似文献

The neutral red retention (NRR) assay was used to evaluate the effects of air exposure on lysosomal membrane integrity in the haemolymph of blacklip abalone, Haliotis rubra, and its subsequent recovery in water. After acclimation in 16°C water for 7 days, abalone were exposed to an air temperature of 7, 16 or 23°C for 12 h in the air exposure experiment or to these three air temperatures, e.g., for 12, 24 or 36 h, followed by re‐immersion in 16°C water in the lysosomal membrane stability recovery experiment. Statistical analyses of the air exposure experiment showed that when abalone were exposed to different air temperatures (7, 16 or 23°C), the lysosomal membrane stability was significantly affected by the air temperature, the exposure duration and their interaction. Air temperature similar to the acclimation temperature had a significantly lower impact on the lysosomal membrane stability within the initial 4.5 h in comparison with the other two temperatures in the same period. The lysosomal membrane stability recovery experiment showed that after air exposure durations of 12, 24 or 36 h, the re‐stabilization of the lysosomal membrane was faster in the animals exposed to lower temperatures than those exposed to higher temperatures. The recovery of the lysosomal membrane stability in abalone exposed to lower 7°C air temperature was not significantly affected (F2, 66=0.251, P=0.779) by the exposure durations (12, 24 and 36 h) used in this study. Alternatively, the lysosomal membrane stability in abalone exposed to higher air temperatures of 16 or 23°C recovered at a faster rate when subjected to shorter durations of air exposure (F2, 66=3.663, P=0.031 and F1, 44=17.057, P<0.001 for 16 and 23°C respectively).  相似文献

Abstract. A quantitative gonadal index was developed for oysters, Crassostrea virginica (Gmelin. 1791), using polyclonal antibodies from eggs and sperm. Percoll used in the purification of oyster eggs and sperm greatly improved the purity of antigens compared to filtering the egg or sperm through a fine mesh only. The antigen-antibody reaction was tested with indirect sandwich ELISA using alkaline phosphatase-conjugated goat anti-rabbit igG as a secondary antibody. Rabbit anti-oyster egg IgG and anti-oyster sperm IgG initially exhibited a weak cross-reactivity over somatic tissue. Absoring with acetone-dried oyster tissue powder removed this cross-reactivity. Both antisera exhibited strong specific immunological reactions to oyster eggs or sperm respectively. The quantity of eggs or sperm was measured using ELISA and a quantitative gonadosomatic index (dry wt of egg or sperm/dry wt oyster) (GSI) calculated. GSI from ELISA correlated with gonadal stage measured histologically. Monthly mean GSI of female oysters was highest during late spring to early summer (0·157–0·201) and lowest during early winter to early spring (0·002-0·000). Maximum GSI observed during the study was 0·422 for female oysters and 0·446 for male oysters. Female oysters produce 3·7–65·4 million eggs, with an average of 21·1 million during each spawning. A positive correlation was observed between the number of eggs produced and oyster size; the number of eggs in the gonad increased as oyster size (i.e. total dry wt) increased (r= 0·67); however, the relationship was non-linear. Large oysters contained proportionally fewer eggs. Prevalence of Perkinsus marinus parasitism was high, 90–100%, during the study, as was weighted incidence, 1·33 to 2·67, No statistically significant correlation was observed between infection intensity and the per cent weight of oyster eggs or egg number.  相似文献
