Exposure to heat‐shock protein (Hsp) stimulating factors induces Hsp accumulation and confers tolerance to lethal ammonia stress on the common carp Cyprinus carpio. This study investigated whether a non‐lethal heat shock bestowed similar protective effects against ammonia and induced thermotolerance, both thought to be rendered by increased amounts of Hsps. The 30‐min lethal temperature (30 min LHT) and 1‐h lethal ammonia concentration (1 h LCT) for this species occurred at 41°C and 14.2 mg/L NH3 respectively. Heating juvenile carp (5 cm) from 28°C to 32, 34 and 38°C, with a subsequent 8‐h recovery period augmented tolerance to lethal heat and ammonia perturbation by two to threefold as compared with animals held at 28°C. Protection occurred in conjunction with Hsp70 accumulation in gills, substantiating the role of this Hsp in enhancing the stress tolerance of common carp.  相似文献

The effects of heat-shock during pelagic stages on growth and survival of juvenile sea cucumber Apostichopus japonicus were investigated to test the possibility to acquire high thermal tolerance individuals after the heat-shock selection. Larvae at the stages of blastula, gastrula, and auricularia were heat-shocked at selected temperatures (21.5, control; 26, 28, and 30°C) for 45 min and returned to 21.5°C for continuous rearing. There was a significant difference in thermal sensitivity among different developmental stages, and thermal tolerance of the larvae was correlated with the expression of heat shock protein 70 (Hsp70). Juveniles after the heat-shock selection at pelagic stages showed higher induced thermotolerance than those without heat-shock. Therefore, heat-shock application at early pelagic development stages is potentially a useful way to select high-thermotolerance variety of the sea cucumbers.  相似文献

Using Hsp70 as a biomarker, thermal stress impinges on reproductive organs, ovary and hepatopancreas were being analyzed by determining the expression of Hsp70 mRNA inside the organs after the adult inter‐molt females were subjected to thermal treatment at 35, 30 and 28°C (Control). Results showed the expression of Hsp70 mRNA under thermal treatment of 35°C after 2 hr recovery in ovary were upregulated at 2, 4, 6, 12, 24 hr and 30 days compared to control whereas in hepatopancreas under similar treatment, the expression of Hsp70 mRNA were significantly higher than control at 6, 24 hr and 30 days. Frequency of reproductive molt at 35°C showed the ovary of females were failed to develop and only entered common molt along three consecutive molt cycles. For 30°C thermal treatment, the expression of Hsp70 mRNA was significantly higher than control after 2 hr recovery but returned to normal afterwards until 30 days’ thermal treatment. Maternal heat shock for 2 hr at 35°C were found to give significantly lower frequency of reproductive molt and longer duration of ovarian development and incubation period whereas maternal heat shock for 2 hr at 30°C gave lower frequency of reproductive molt, slower development of embryo and lower hatching success compared to untreated control. This study suggests that short and long‐term thermal stress at 30 and 35°C were found to affect the induction of Hsp70 mRNA in reproductive organs of Macrobrachium rosenbergii and also influence their reproductive performance.  相似文献

Rainbow trout Oncorhynchus mykiss is a cold-water aquaculture species, and a thermally selected strain has been developed by multigenerational high-temperature breeding in Japan. We examined the expression of heat-shock proteins as candidates responsible for thermotolerance in rainbow trout using F2 offspring from F1 hybrids produced between thermally selected and normal strains. From F2 offspring, two groups were selected for western blot analysis, namely, low- and high-thermotolerance groups (times to loss of equilibrium were <30 and ??60?min, respectively). We demonstrated that the expression levels of Hsp70, Hsp60, and Hsp40 in tail fin tissues were significantly higher in the individuals with high thermotolerance than in those with low thermotolerance under non-heat-shock conditions. In particular, Hsp70 was expressed only in the individuals with high thermotolerance. These results suggest that Hsp70 is a major protein responsible for conferring thermotolerance in rainbow trout.  相似文献

Interspecific allo-tetraploidy was induced by a combination of hybridization of Megalobrama amblycephala ♀× Megalobrama terminalis ♂ and thermal shock. A thermal shock at 40 °C for 2 min applied to eggs 33 or 26 min (τ0: 1.40) after fertilization resulted in a significantly ( P <0.01) higher yield of allo-tetraploids than hybrid crosses without heat shock. The yield of allo-tetraploids in embryos from mid-gastrula to hatching stages was 15.7–24.4% in different thermal shock treatment groups. The allo-tetraploidy rate in 330-day-old juveniles was 9.3% and 6.7% in different year treatment groups. After 2 years of rearing, allo-tetraploidy males attained sexual maturity with milk-like milt at the age of 2+, while allo-tetraploidy females at the age of 2+, and even at 3+, did not show any signs of sexual maturation exhibiting obviously delayed maturity. Only a few allo-tetraploidy females (three individuals) showed maturation characters at the age of 4+ and these females were induced to spawn.  相似文献

ABSTRACT:   In order to understand the characteristics of burnt meat in cultured yellowtail Seriola quinqueradiata , fish were kept at two different temperatures (13 and 30°C) and slaughtered by either spinal cord destruction (SCD) or suffocation in air (SA). Early postmortem changes during storage at 32°C were analyzed by rheological, biochemical, and histological methods. The burnt meat (with lightness parameter, L* ≥ 55) was observed at 1-h storage in the SA 30°C group, at 2 h in SCD 30°C, and at 4 h in SA 13°C; meat was normal for the SCD 13°C group until 6 h of storage. Breaking strength scores were higher for the normal meat (200 g/cm2) than burnt meat (70 g/cm2) at 4 h of storage. Expressible water content was higher for the burnt meat than for the normal meat. Adenosine triphosphate concentrations for the SCD groups were higher than for the SA counterparts. Moreover, pH decrease was much faster in the 30°C groups, showing pH 5.6 at 2 h of storage. A negative correlation between the pH and lactic acid contents in muscle ( P  < 0.001) was found. Histological analysis evidenced a larger pericellular area (40%) in the burnt samples than in the normal samples (16%). It was confirmed that a higher fish-keeping water temperature and a stressful slaughter method (faster glycolytic process) were determinative factors that influence the occurrence of burnt muscle in yellowtail, and that the effect of the former is larger than the latter.  相似文献

ABSTRACT: Lethal limits of high temperature were studied to clarify the effects on the survival of the endangered Japanese crayfish species Cambaroides japonicus and the alien species Pacifastacus leniusculus . After the acclimation period for 2 weeks at 16°C, the temperature was raised at a rate of 1°C per week. As a result, the ultimate upper lethal temperatures of C.   japonicus and P.   leniusculus were 27.0 and 31.1°C, respectively, and the lethal temperature for P.   leniusculus was significantly higher than for C.   japonicus . The natural distributions of these two species are discussed in terms of the temperature tolerance.  相似文献

Abstract –  The effects of constant (12, 18, and 24 °C) and cyclical (daily variation of 15–21 and 12–24 °C) thermal regimes on the growth and feeding of Lahontan cutthroat trout ( Oncorhynchus clarki henshawi ) of variable sizes were examined. Higher constant temperatures (i.e., 24 °C) and more variable daily temperatures (i.e., 12–24 °C daily cycle) negatively affected growth rates. As fish mass increased (from 0.24 to 15.52 g) the effects of different thermal regimes on mass growth became more pronounced. Following 14 days exposure to the thermal regimes, feeding rates of individual fish were assessed during acute exposure (40 min) to test temperatures of 12, 18, and 24 °C. Feeding rate was depressed during acute exposure to 24 °C, but was not significantly affected by the preceding thermal regime. Our results indicate that even brief daily exposure to higher temperatures (e.g., 24 °C) can have considerable sublethal effects on cutthroat trout, and that fish size should be considered when examining the effects of temperature.  相似文献

The growth and mortality of Neptunea arthritica juveniles hatched from different egg masses (controlled conditions and field) reared at 10 and 15 °C, and provided with sardine (D1) and polychaete (D2) as foods were evaluated. Results showed that diet type and water temperature significantly affect the growth and mortality. Interaction between these factors reflected robust relationship among them, but restricted exclusively to growth. Through the study period, juveniles treated at 10 °C showed maximum increments of 2.5 mm (shell length), 2.3 mm (shell width) and 0.25 g (body weight) from their initial size. While maximum values of those treated at 15 °C for the above-mentioned parameters were 7.6 mm, 5.5 mm and 1 g respectively. In both cases, these increments were recorded for individuals fed with sardine. Mortality was significantly affected by diets and water temperature, which significantly increased under D2 [10 °C (20–67%), 15 °C (70–87%)] or 15 °C [D1 (30–83%), D2 (70–87%)]. According to the results, juveniles of N. arthritica could be maintained under controlled conditions, which allows growth improvement. Mortality appears to be the limiting factor, but this could be improved by the implementation of efficient culture method, basically oriented to increase the water quality.  相似文献

ABSTRACT:   The effects of temperature and salinity on growth of green algae Chlorella ellipsoidea and Nannochloris oculata were determined to compare the optimum culture conditions. A four-temperature (15, 20, 25, and 30°C) × three-salinity (10, 20, and 30) factorial design with triplicates was applied. Specific growth rate (SGR), maximum density, and duration to reach maximum density of C. ellipsoidea were significantly affected by both temperature and salinity. The highest SGR was observed in C. ellipsoidea at 25°C and salinity 10, but the maximum density was very low. The highest maximum density was achieved in C. ellipsoidea at 15°C and 10. The slope constant of the linear relationship between semilogarithmic growth of C. ellipsoidea and day of culture was highest at 15°C and 10. The SGR and duration to reach maximum density of N. oculata were significantly affected by both temperature and salinity. However, maximum density of N. oculata was significantly affected by temperature, but not salinity. The highest maximum density was achieved in N. oculata at 25°C and 30, but SGR was significantly lower than that of N. oculata at 25°C and 10. The slope constant of the linear relationship between semilogarithmic growth of N. oculata and day of culture was highest at 25°C and 30. Based on these results, the condition of 15°C and salinity 10 seemed to be optimal for maximum density of C. ellipsoidea , and the condition of 25°C and 10 and 30 for SGR and maximum density for N. oculata , respectively.  相似文献

Abstract. Infectivity experiments were undertaken by water-borne inoculation of kuruma shrimp, Penaeus japonicus Bate, larvae with BMN virus. Mysis stage larvae were inoculated with the virus by exposure for 2h in sea water containing a homogenized and filtered preparation (450nm) of naturally BMN virus-infected shrimp stored for about 7 weeks at −80°C. Inoculated shrimp stocked in rearing jars were examined to determine whether nuclear hypertrophy of the mid-gut gland epithelial cells characteristic for BMN virus infection could be observed in fresh squash preparations under dark field illumination equipped with a wet-type condenser. Four days post-inoculation at 25–30°C incubation temperature were considered to be satisfactory for the experimental trial.  相似文献

从刺参(Apostichopus japonicus)低盐转录组数据库中选取与应激(热休克蛋白70基因)和离子传递(甘氨酸转运蛋白基因、锌转运蛋白基因、神经乙酰胆碱受体基因)相关的4个差异表达基因,利用qRT-PCR技术分析这4个基因在不同组织中的表达水平及低盐对其表达丰度的影响.结果表明甘氨酸转运蛋白基因在刺参呼吸树中表达水平最高,肠次之,体腔液表达较低;锌指蛋白基因和神经乙酰胆碱受体基因均在体腔液中表达最高,肠次之,在呼吸树中不表达;热休克蛋白70基因在体腔液中表达量最高,其次是呼吸树和肠组织.低盐胁迫下这4种基因的表达量均随着胁迫时间的延长呈波动性增减,其中甘氨酸转运蛋白在体腔液中的表达低于正常表达水平,在胁迫后3h时达到最低表达量.神经乙酰胆碱受体基因除在体腔液中48 h出现明显的上调外,在体腔液其他时间点和肠组织中处于下调表达状态.低盐胁迫下这4个基因表达丰度的变化,说明这些基因或作为功能蛋白直接参与机体的代谢调节,或作为调控蛋白调节胁迫功能蛋白的表达和活性来提高刺参对低盐胁迫的耐受能力.研究结果可为刺参盐度调节适应机制的研究奠定基础.  相似文献

ABSTRACT:   The modori phenomenon is defined as heat-induced myofibrillar degradation caused by endogenous serine protease(s) of fish muscle during Kamaboko fish meat gel production. This study was undertaken to analyze myofibrillar proteolysis of white croaker Argyrosomus argentatus muscle, which is an ingredient of high quality Kamaboko, by myofibril-bound serine protease (MBSP) under conditions corresponding to the modori phenomenon. White croaker MBSP was stable between pH 2–11 and below 65°C, and about 60% of its initial activity remained after incubation for 2 h under the conditions at 65°C and pH 7.5. About 60% of the enzyme activity was suppressed by 0.5 M NaCl. White croaker MBSP degraded various myofibrillar proteins between 40 and 70°C and pH 6.0–9.0, and preferentially degraded myosin heavy chain rather than other myofibrillar proteins. The enzyme degraded the myosin heavy chain most strongly at 55°C and pH 7.0, and a major part of the bands of myosin heavy chain and its degradation products disappeared for a period of 2 h. These degradation characteristics are very similar to those observed during the modori phenomenon, indicating that MBSP could be a modori-inducing protease involved in the modori phenomenon of white croaker Kamaboko production.  相似文献

Water temperature is a significant environmental stressor that affects physiology and biochemical activities of bivalves. Here, temporal variations in physiological and biochemical parameters of Zhikong scallop, Chlamys farreri, under three water temperature treatments were investigated. For the first treatment, scallops were transferred from rearing temperature (15°C, control temperature) to 5, 10, 20 and 25°C, respectively. The second group of scallops was gradually acclimated to above experimental temperatures at a rate of 1–2°C day?1. The third group was transferred directly between 15 and 7°C every 12 h and for six times. Results showed that significantly higher oxygen consumption and ammonia‐N excretion together with a significant lower ingestion rate was observed for the acute temperature change treatment compared to those in the equivalent gradual temperature variation treatment (< 0.05). In acute temperature change treatment, hepatopancreas antioxidant enzyme activities, that is, superoxide dismutase (SOD) and catalase (CAT), immune enzyme activities, that is, acid phosphatase (ACP) and lysozyme (LSZ), and heat‐shock protein 70 gene (Hsp70) expression levels of scallops increased substantially within 48 h. Significant increases in SOD, CAT, ACP and LSZ activities, and malondialdehyde content occurred under exposure to fluctuating temperatures (< 0.01). Gill and hepatopancreatic Hsp70 expression levels also increased significantly in response to water temperature fluctuations (< 0.05). The study provides basic knowledge about thermal stress in Zhikong scallop and may contribute to the management of scallop mortalities.  相似文献

Flavobacterium psychrophilum heat shock proteins (Hsp) 60 and 70 are highly immunogenic and were therefore investigated as potential vaccine candidates. Recombinant Hsps were purified from Escherichia coli and rainbow trout ( Oncorhynchus mykiss ) were intraperitoneally injected with phosphate buffered saline/Freunds complete adjuvant (FCA), 8 μg of rHsp60/FCA, rHsp70/FCA or a combination of 4 μg each of rHsp60 and rHsp70/FCA. Antibody responses against recombinant Hsp60 and Hsp70 8 weeks post-immunization were observed, but only fish immunized with rHsp70 exhibited highly elevated antibody levels against F. psychrophilum whole cell lysate. Some cross reactivity occurred, which may have been due to the V5 tag common to both proteins. Protection against F. psychrophilum challenge was not observed in any treatments at 8 weeks post-immunization. To further investigate any protective effect of these proteins, hsps were polymerase chain reaction amplified and cloned into pVAX1. Rainbow trout were intramuscularly injected with 8 μg of pVAX1hsp60, pVAX1hsp70 or a combination of 4 μg each of pVAX1hsp60 and pVAX1hsp70. Antibody responses at 4 weeks post-immunization were low and protection was not observed following challenge at 6 or 10 weeks post-immunization. Although Hsps of F. psychrophilum have been shown to be immunodominant, these antigens do not appear to be good vaccine candidates when delivered alone or in combination.  相似文献

Juvenile mirror carp were fed diets containing 303.4, 321.7, 341.2, 361.0 and 379.1 g kg?1 proteins, respectively, and reared at different water temperatures (18, 23 and 28°C) for 60 days. Gene expression of heat shock protein gene (Hsp70) and the warm temperature acclimation‐related 65 kDa protein gene (Wap65), immunity and antioxidant status in the carp were investigated. Results indicated that the contents of serum complement 3 (C3), complement 4 (C4) and immunoglobulin M (IgM), as well as activities of liver superoxide dismutase (SOD) and lysozyme (LSZ) were significantly enhanced with increasing dietary protein (< 0.05), while content of malondiadehyde (MDA) decreased. Gene expression level of Wap65 in the liver significantly increased with dietary protein, while gene expression of Hsp70 decreased. The contents of C3, C4 and IgM, the activities of SOD and LSZ and gene expression level of Wap65 in the liver significantly increased with temperature. These results suggest that: Serum immune parameter, antioxidant enzymes and Hsp70 and Wap65 expression interact in fish to improve ability to adapt to the environment; and the optimal conditions for the immunity of carp are 348.1?354.5 g kg?1 protein at 18°C, 352.3?364.9 g kg?1 at 23°C and 360.2?364.3 g kg?1 at 28°C, and the optimum temperature for carp is 23°C.  相似文献

The effects of temperature and salinity on the settlement, subsequent survival and development of the copepodids of Lepeophtheirus salmonis on Atlantic salmon were investigated experimentally. There was a significantly greater settlement and survival of copepodids at 10 days post-infection (dpi) at 12 °C compared with at 7 °C at a constant salinity of 34‰. Development of L. salmonis was also more rapid at 12 °C. Settlement was significantly greater at a salinity of 34‰ than at 24‰. In one experiment, survival at 10 dpi was significantly greater at 34‰; however, a second experiment found that there was no significant difference between the two saline levels. This may have been because of a rise in water temperature for 2 dpi, which appears to have overridden the effect of low salinity. Development of L. salmonis was more rapid at 34‰. Copepodids settled on all of the external surfaces of the salmon, although the proportion on different surfaces varied between experiments. The gills, particularly at low temperatures, the body surface, and the pectoral and dorsal fins were especially favoured.  相似文献

ABSTRACT:   The effects of temperature changes on the endogenous rhythm of oxygen consumption in the Japanese flounder Paralichthys olivaceus which were acclimated to 3, 14 or 20°C in advance were investigated. The oxygen consumption rate (OCR) of individuals (total length >28 cm) that were acclimated to 3°C, maintained a relatively low value of 6.9 ± 2.3 mL (mean ± SD) O2/kg ww (wet weight)/h till 5.8°C, which is considered a 'minimum' rate. Thereafter, the OCR gradually increased up to 18.7 mL O2/kg ww/h at the temperature of 10.6°C. The OCR of individuals acclimated to 14°C stabilized with 124.2 ± 24.1 mL O2/kg ww/h, when tested at the relatively constant temperature of 14.4 ± 0.17°C. Maximum entropy spectral analysis revealed that the peak OCR occurred at 23.9 h intervals, which could be a circadian rhythm. The mean OCR showed a peak at 06.00–08.00 hours. For the temperature above 25°C in which the fish were acclimated to 20°C, the rhythmic patterns of OCR were observed until the experimental temperature reached 26.4°C. For the temperature above 28.4°C, the metabolic activity rhythm of the OCR was dampened and then the OCR abruptly rose again. It is assumed that the oscillator of the endogenous rhythm lost the governing of normal metabolic activity in Japanese flounders above 28.4°C. These results suggest that the temperatures of around 5.8, 26.4 and 28.4°C are the 'lower limit temperature', 'upper incipient lethal temperature', and the 'critical thermal maximum', respectively, for the endogenous rhythm of oxygen consumption in the Japanese flounder.  相似文献

ABSTRACT:     The effects of parental exposure to 17β-estradiol (E2) and 4-nonylphenol (4-NP) on life history parameters of three successive generations of the euryhaline cladoceran Diaphanosoma celebensis were assessed under laboratory conditions. Less than 24-h-old neonates (P) were exposed to five sublethal concentrations of E2 (0.1, 1, 10, 100 and 1000 μg/L) and 4-NP (0.01, 0.1, 1, 10 and 50 μg/L) at 25°C. Age at first reproduction, reproductive period, fecundity and lifespan were investigated. Successive generations (F1–F3) were monitored further in the absence of toxicants. Results showed that cladocerans exposed to 10–1000 μg/L E2 produced more neonates at a younger age compared to the control. The same effects were also observed in the F1 and F2 but ceased in F3. Cladocerans exposed to 1 μg/L 4-NP also produced significantly more neonates compared to the control, but this effect was not found in successive generations. The results suggest that vertebrate hormone can modify the reproduction of D. celebensis , and the effects are multigenerational.  相似文献
