The objective of this study was to determine the effect of two transport journeys (3 and 16 h) and four lairage times at the abattoir (3, 6, 12 and 24 h) on some stress indicators in steers to validate the current Chilean legislation of a minimum lairage period of 12 h for cattle arriving at abattoirs.The study was carried out twice, 80 steers in the winter and 80 in the summer of 1999. Forty steers were transported for 3 h and 40 for 16 h. At arrival in the slaughterhouse, the 40 transported steers were randomly divided in four lairage groups. Blood samples were collected before loading at arrival at the slaughterhouse and at slaughter during exsanguination.Mixed models were used to determine the associations between the stress indicators and the independent variables while correcting for the repeated observations on animals. Lairage increased plasma cortisol and PCV values independent of the transport duration. Glucose concentrations in steers transported for 16 h further increased after 3 and 6 h of lairage but decreased after 12 and 24 h of lairage. In steers transported for 3 h, glucose concentrations only increased after 24 h of lairage. After 24 h of lairage, the steers transported for 16 h had increased βHB concentrations compared with those transported for 3 h. Plasma CK activity was increased in steers after transport but was not affected by lairage. The study shows that there is no beneficial effect on the welfare of the animals by a long lairage time at the abattoir.  相似文献

Comparisons were made of the behaviour, physiology and meat quality of 24-week-old male veal calves (mean live weight 234 kg) that were slaughtered either at the veal unit (the control group) or following 3 h transportation at space allowances of either 0.7 or 0.95 m2 calf−1 and 1.5 h of lairage. Compared with nontransported calves, transported calves had a greater heart rate during the journey and during lairage; greater plasma cortisol concentration immediately after the journey, but not after 1.5 h of lairage; and greater plasma creatine kinase activity immediately after the journey and after 1.5 h of lairage. The transported calves did not lie down during the journey. There was no effect of space allowance on either the heart rate or frequency of potentially traumatic events during the journey or the plasma cortisol concentration and plasma creatine kinase activity sampled after the journey. There was no effect of transport on either carcase bruising or muscle pH 24 h after slaughter. Although compared with ‘on-farm’ slaughter, the transport and handling associated with the journey to the slaughterhouse were stressful to the calves; there were no adverse effects of transport on either carcase bruising or meat quality.  相似文献

The objective of this study was to determine the effects of lairage time (short vs. long), behavior (aggressive vs. non‐aggressive) and gender (gilts, barrows and boars) on stress and meat quality parameters in pigs (n = 100). In the group with short lairage, aggressive behavior of gilts and boars was observed. At exsanguination, blood samples were collected and levels of blood lactate and plasma cortisol were determined. Post mortem pH value, temperature, drip loss, sensory and instrumental color and meat quality class were determined on the muscle Longissimus dorsi. Higher blood lactate concentration (P < 0.05) and skin blemish score (P <0.001), lower temperature (P < 0.05) and drip loss (P < 0.01), as well as darker, less red and yellow meat color (P < 0.001) were found after long lairage compared to short lairage. Gilts had higher plasma cortisol concentrations than barrows (P < 0.05), while barrows had lower skin blemish scores (P < 0.05) than gilts and boars after long lairage. The highest incidence of PSE (pale, soft and exudative) meat was observed in barrows (100.00%) and boars (91.67%) after short lairage, while the lowest incidence of PSE meat was found in barrows (28.89%) after long lairage. Aggressiveness affected meat temperature, as aggressive pigs had higher meat temperature (P < 0.05) than non‐aggressive pigs.  相似文献

This study examined the effect of blood sampling time (farm, transport, lairage) on welfare physiological indicators in two groups of different age/weight Manchega breed lambs (suckling vs light). Blood sampling time had a different effect on both types of lamb: In light lambs, there was an increase (P < 0.001) in red blood cells (RBC), haemoglobin, hematocrit and urea after lairage; but adrenaline, glucose and creatinine increased after transport. In suckling lambs, there were higher values of cortisol and potassium on the farm, both glucose and sodium increased after transport while adrenaline did after lairage. In general, significant differences (P < 0.001) were found between both types of lamb in the parameters analysed. Light lambs showed higher values of RBC, haemoglobin, hematocrit, adrenaline, noradrenalin, LDH, CK, sodium and lactate than younger animals in all blood sampling times. Only after lairage were urea and leucocytes lower (P < 0.05) in suckling than in light lambs, while cortisol was higher on the farm and lower after lairage in younger animals.  相似文献

Two experiments were carried out in which a total of 602 pigs were slaughtered after being held in lairage for periods ranging from less than one hour to 21 hours. In the first experiment the pigs were handled under ideal conditions and slaughtered at the University of Bristol slaughterhouse; in the second the pigs were killed at a commercial plant. Blood samples collected at exsanguination were analysed for indices of stress. There were no consistent effects of time in lairage on the levels of lactate and creatine phosphokinase. Plasma cortisol and beta-endorphin levels were reduced by lairage for three hours or more in the first experiment and cortisol was reduced by lairage for two hours or more in the second; beta-endorphin was not measured in the second experiment. A period of rest in lairage allowed the pigs to recover from transport and the associated handling and the recovery appeared to be complete within two to three hours.  相似文献

The aim of this study was to assess the effect of the Chilean rodeo exercise on some blood constituents, classically used to assess welfare, during rest; training with and without steer; and before and after official competitions. During the training season, 13 horses were assessed at their farms of origins, and samples were taken at rest and after training with and without a steer; then during the competition season, 16 horses were assessed at four different times, one before and three after the competition. The blood constituents assessed were packed cell volume (PCV), total proteins, fibrinogen, cortisol, neutrophil:lymphocyte ratio, creatine phosphokinase, aspartate transaminase, glutathione peroxidase, and serum amyloid A. Analysis of variance for repeated measures was applied using the Statistix 8.0 software, and a significance level of P < .05 was applied. For horses assessed during exercise with and without a steer, significant differences were observed for PCV, which were significantly lower at rest than after both exercises, and total proteins, which showed a significant increase only after exercise with steer. In the case of official competitions, PCV was significantly higher immediately after rodeo, and the lowest value for cortisol was observed the evening after competition (P < .05); among the enzymes assessed, creatine phosphokinase plasma concentrations immediately after rodeo and the evening after competition were significantly higher than concentrations the next morning. The use of physiological indicators did not seem to be sufficient to assess the overall effect of rodeo on the welfare of these horses, and other types of indicators should be considered in future studies.  相似文献

Two identical experiments, using Standardbred and Thoroughbred horses (experiment A, n = 31; experiment B, n = 17) on a swimming regimen, were performed 1 week apart to evaluate short-term heart rate, hematologic, and endocrinologic changes. Horses were placed in 4 categories based on duration of swimming (1 to 5, greater than 5 to 10, greater than 10 to 15, and greater than 15 minutes). Heart rate, PCV, and plasma concentrations of total protein, cortisol triiodothyronine, thyroxine, insulin, and glucose of each horse were evaluated before, immediately after, and 1 hour after swimming. For experiment A, there was a main effect of time of sampling on PCV, total protein, triiodothyronine, thyroxine, and insulin. There was a main effect of duration of swimming on insulin concentration and an interaction of duration of swimming and time of blood collection on heart rate and cortisol concentration. For experiment B, there was a main effect of time of blood collection on all the variables except glucose.  相似文献

Plasma sodium concentrations, packed cell volume, total plasma protein concentration and dermal skinfold thickness were used to assess the state of hydration of 113 transported calves before and after lairage and in 30 control calves on the farm of origin. Skin thickness increased significantly (P less than 0.05) and total protein increased slightly during transport and decreased during lairage. These changes suggest that transport may cause dehydration and lairage may help in recovery. Plasma potassium concentration decreased during transport, but the effect was inversely related to the distance travelled, and the concentration increased during lairage. These changes are consistent with recovery from initially high cortisol levels at loading. The resting behaviour of 150 transported calves was recorded hourly for six hours. More time was spent resting and sleeping by the transported calves than has been reported for non-transported calves and more still by small transported calves, suggesting that transport is exhausting, that lairage helps recovery and that small calves are more adversely affected. During lairage the numbers of calves asleep decreased to values reported for normal calves, suggesting that 10 hours lairage was adequate. However, small calves did not return to their normal rest patterns within the observation period.  相似文献

OBJECTIVE: To compare physiologic, hematologic, and selected serum and plasma biochemical variables obtained from horses competing in 48-, 83-, or 159-km endurance rides before competition and at the same cumulative distance points. ANIMALS: 83 horses. PROCEDURE: Weight and rectal temperature measurements and blood samples were obtained from horses before, during, and after 1 of 3 rides conducted on the same day. Plasma protein (PP), lactate, WBC, serum electrolyte, and calcium concentrations; PCV; and creatine kinase (CK) activity were determined. Assessments were made to determine whether any differences among groups, with respect to total distance competed, could be explained by differences in lap speed or conditioning and to investigate the effect of time in transit or on-site prior to competition on results of blood analyses or competition outcome. RESULTS: Horses in the 159-km distance group had the lowest preride serum sodium, chloride, bicarbonate, and calcium concentrations. As hours in transit increased, preride PP concentration was significantly greater; serum sodium, chloride, and bicarbonate concentrations were lower; CK activity at 159 km was greater; and horses were more likely to be eliminated. The preride sodium was significantly greater in horses that completed the ride, compared with those eliminated. CONCLUSIONS AND CLINICAL RELEVANCE: Among distance groups, distance ridden, speed, level of fitness, and years of experience of horses had little effect on the variables examined. Electrolyte and water supplementation and earlier arrival at the event may be beneficial for horses that are transported long distances to endurance competition.  相似文献

Leptin is a hormone that coordinates food intake, energy expenditure, and metabolic rate; it is, however, unknown how exercise influences plasma leptin concentration in horses. The aim of the study was to evaluate the exercise-induced changes in plasma leptin concentration in purebred Arabian horses competing in races and endurance rides. A total number of 26 horses (12 purebred Arabian racehorses aged 3–5 years and 14 endurance horses aged 7–14 years) were studied during performing routine physical exercise. From each horse, blood samples were collected at rest and immediately after the exercise session. Concentrations of plasma leptin, cortisol, lactic acid, uric acid, triacylglycerols, glycerol and free fatty acids were determined. Exercise-induced increase in cortisol and free fatty acid values were higher in endurance horses than in racehorses. Neither endurance nor race exercises influenced the plasma leptin concentration. Further research is needed to elucidate the effects of different types of exercise on leptin synthesis and secretion in horses.  相似文献

Physical activity and stress both cause an increase in cortisol release ratio. The aim of this study was to evaluate the use of saliva samples for the determination of cortisol concentrations indicating the work‐load level in horses during race training. Twelve Purebred Arabian horses aged 3–5 years were studied during the routine training session. After the warm‐up, the horses galloped on the 800 m sand track at a speed of 12.8 m/s. Three saliva samples, and three blood samples were collected from each horse. Both types of samples were taken at rest, immediately after return from the track and after 30 min restitution. The concentrations of blood lactic acid (LA), and cortisol in saliva and plasma samples were measured and analyzed. Blood LA, plasma and salivary cortisol levels increased significantly after exercise (P < 0.05). Salivary cortisol concentration determined 30 min after the exercise correlated significantly with plasma cortisol level obtained immediately after exercise (P < 0.05) as well as measured 30 min after the end of exercise (P < 0.05). The determination of cortisol concentration in saliva samples taken from racehorses 30 min after the end of exercise can be recommended to use in field conditions to estimate the work‐load in racehorses.  相似文献

The objectives of the present experiment were 1) to study the effects of transport conditions and lairage duration on stress level, muscle glycolytic potential, and pork quality; and 2) to investigate whether the negative effects of high stress immediately preslaughter are affected by preceding handling factors (transport and lairage). In a 2 x 2 x 2 factorial design, halothane-free pigs (n = 384) were assigned to either short (50 min) and smooth or long (3 h) and rough transport; long (3 h) or short (<45 min) lairage; and minimal or high preslaughter stress. Pigs were processed in eight groups (48 pigs per group) on various days at a commercial plant. Blood samples were taken at exsanguination to measure plasma cortisol and lactate concentrations. Muscle pH and temperature were measured at 30 and 40 min, respectively, and both were measured at 3 h, postmortem. A LM sample was taken 135 min postmortem to estimate glycogen content and rate of glycolysis. Pork quality attributes were assessed 23 h postmortem. Short transport increased cortisol when followed by short lairage (transport x lairage; P < 0.01). Long transport, but not lairage (P > 0.30), tended to increase (P = 0.06) muscle glycolytic potential. Long transport tended to increase (P = 0.08) electrical conductivity, and decreased a* (P < 0.01) and b* (P < 0.02) values. Decreasing lairage from 3 h to <45 min decreased (P < 0.05) the L* value, but it did not (P > 0.10) affect other pork quality traits. High stress decreased (P < 0.001) muscle glycolytic potential, and increased (P < 0.001) plasma lactate, cortisol, muscle temperature, rate of pH decline, and ultimate pH. Except for decreased (P < 0.001) b* values, pork color was not (P > 0.40) affected by high stress, but water-holding properties (measured by electrical conductivity, filter paper moisture, and drip loss) were impaired (P < 0.001) by high stress. Fiber optic-measured light scattering and Warner-Bratzler shear force were not (P > 0.12) affected by any treatment. Comparisons with the "optimal" handling (short transport, long lairage, and minimal stress) revealed that, with regard to water-holding properties, the negative effects of high stress were aggravated by suboptimal transport and lairage conditions. High stress alone increased electrical conductivity by 56%, whereas high stress in combination with short lairage led to an 88% increase. However, high preslaughter stress contributed most and was the major factor responsible for reductions in pork quality.  相似文献

选取体质量约为100 kg、氟烷基因型为NN的长×大二元杂交猪60头,随机分为3组:不休息组,休息3 h组,休息3 h+玩具组,公母各半.根据评分标准分别记录休息过程中猪的精神状况.放血时收集血样,用于分析血浆中皮质醇、促肾上腺皮质激素(ACTH)浓度,乳酸脱氢酶、肌酸激酶活性,乳酸、血糖浓度;添加EDTA-K<,2>抗凝剂的血样用于血细胞分析.同时在放血时,测定血温.屠宰后.测定背最长肌24 h滴水损失;背最长肌(LM)、半膜肌(SM)45 min以及24 h的pH、色值、导电率.结果表明:①休息3 h+玩具组在3个时间段的精神评分要极显著高于休息3 h组,随着时间的延长猪都表现出趋于平静的趋势;②休息3 h+玩具组和休息组3 h组在皮质醇、促肾上腺皮质激素要显著低于不休组(P<0.05),相反乳酸脱氢酶、肌酸激酶要显著高于不休息组(P<0.05),而休息3h+玩具组与休息3 h组差别不显著(P<0.05);③3个休息处理组在肉质指标上没有出现显著差别(P<0.05);④休息3 h+玩具组、休息3 h组在红细胞数(RBC)、血红蛋白含量(HGB)均要显著低于不休息组(P<0.05);休息3h+玩具组在白细胞数(WBC)上显著高于不休组(P<0.05);休息3 h组的淋巴细胞绝对值(W-SCC)显著高于不休息组(P<0.05);而休息3 h+玩具组与休息3 h组在所有的免疫指标上都没有显著差异(P>0.05).根据上述的结果显示,宰前一定要让猪有一个休息的过程,这种措施能有效缓解猪的应激,并能恢复其免疫能力;在休息的过程中,添加部分福利玩具,至少对猪的精神状态有明显的好处.  相似文献

Both physical activity and stress result in an increase in plasma cortisol level. The measurement of cortisol in plasma requires taking blood samples, which is stressful itself. Therefore, the aim of this study was to evaluate the use of saliva sampling for the determination of cortisol concentrations, indicating the intensity of exercise in horses during race training. Twelve Thoroughbred horses aged 2-3 years were examined during their speed training sessions. The horses galloped on the 1,200-m sand track at a speed of 14.4-15.3 m/s. Three saliva samples and three blood samples were collected from each horse. Both types of samples were taken when the horse was at rest, immediately after returning from the track and 30 minutes after the end of exercise. Blood lactic acid (LA) concentration was determined using the enzymatic cuvette test. The concentrations of cortisol in saliva and plasma samples were measured by enzyme immunoassay methods. Statistically significant correlations were found between salivary cortisol level determined 30 minutes after the end of exercise and blood LA concentration obtained immediately after exercise (P = .003) and between salivary and plasma cortisol levels measured 30 minutes after the end of training session (P = .015). The measurement of cortisol concentration in saliva samples taken from race horses 30 minutes after the end of exercise can be recommended for use in practice under field conditions to estimate the level of relative intensity of exercise in race horses.  相似文献

The mean plasma concentration of cortisol at the time of the first treatment for hypocalcaemia at calving was significantly higher in 17 cows which did not recover, than in 53 cows which recovered. Healthy periparturient cows had significantly lower cortisol levels than cows with hypocalcaemia. There was a negative correlation between plasma cortisol and plasma calcium at the time of the first treatment. After adjustment for differences in plasma calcium there was no significant difference between cortisol concentrations in healthy cows and paretic cows which recovered. Plasma cortisol was positively correlated with both packed cell volume (PCV) and serum creatine kinase (CK). At first treatment cows which did not recover had higher levels of PCV and serum CK than cows which recovered, and the difference between the mean plasma cortisol concentrations of these two groups was related to differences in plasma calcium, PCV and serum CK. Plasma cortisol concentrations remained high in cases of protracted paresis.  相似文献

Investigations were carried out on the response of slaughter pigs to CO2 stunning with two different gas concentrations (80 vol%, 90 vol%, 73 s) under practical conditions in a one gondola-dip-lift system. EEG measurements were performed and blood constituents such as catecholamines (adrenaline, nor-adrenaline) and lactate as well as clinical reactions (nasal septum and corneal reflex, heart beats) investigated. Special EEG-electrodes were adapted for the measurement on pigs and a mobile data logger was prepared for the use in the lairage of the slaughter house and in the stunning unit. The CO2 concentrations were measured continuously close to the head of the pigs when transported up and down in the stunning gondola. The results show that the technique is suited to monitor the effects of different CO2 gas concentrations on the EEG of the pigs under practical conditions. There is strong evidence that CO2 concentrations of 80 vol% applied over 70 s as required by law are not sufficient to stun pigs properly. A large part of the animals still showed typical reflexes when leaving the stunning pit. When an atmosphere of 90 vol% CO2 is applied, most animals are already dead before bleeding commences. This may create problems in respect to meat hygiene. The blood analysis revealed very high concentrations of catecholamines after stunning. The values for adrenaline and nor-adrenaline in the sticking blood rose by a factor of about 1000 as compared to the concentrations in blood samples taken in the lairage before stunning. It seems necessary to revise the current legislation on gas stunning and to look in greater detail in the effects of CO2 stunning on the welfare of slaughter pigs.  相似文献

OBJECTIVE: To determine clinical and clinicopathologic abnormalities in horses administered a blood transfusion and evaluate effects of blood transfusion on these variables. DESIGN: Retrospective case series. ANIMALS: 31 adult horses that received > or = 1 blood transfusion. Procedures-Medical records of horses receiving a blood transfusion were reviewed to obtain clinical findings, laboratory test results before and after transfusion, adjunctive treatments, transfusion type and volume, response to transfusion, results of donor-recipient compatibility testing, adverse reactions, and outcome. RESULTS: 31 horses received 44 transfusions for hemorrhagic anemia (HG; n = 18 horses), hemolytic anemia (HL; 8), or anemia attributable to erythropoietic failure (EF; 5). Tachycardia and tachypnea were detected in 31 of 31 (100%) and 22 of 31 (71%) horses, respectively, before transfusion. The PCV and hemoglobin concentration were less than the reference range in 11 of 18 horses with HG, 8 of 8 horses with HL, and 5 of 5 horses with EF. Hyperlactatemia was detected in 16 of 17 recorded values before transfusion. Heart rate, respiratory rate, and PCV improved after transfusion, with differences among the types of anemia. Seventeen (54%) horses were discharged, 9 (29%) were euthanized, and 5 (16%) died of natural causes. Adverse reactions were evident during 7 of 44 (16%) transfusions, varying from urticarial reactions to anaphylactic shock. CONCLUSIONS AND CLINICAL RELEVANCE: Abnormalities in clinical and clinicopathologic variables differed depending on the type of anemia. Colic, cold extremities, signs of depression, lethargy, tachycardia, tachypnea, low PCV, low hemoglobin concentration, and hyperlactatemia were commonly detected before transfusion and resolved after transfusion.  相似文献

OBJECTIVE: To evaluate plasma epinephrine and norepinephrine concentrations and serum cortisol concentration in horses with colic and assess the relationship of these variables with clinical signs, routinely measured clinicopathologic variables, and outcome in affected horses. DESIGN: Prospective observational study. ANIMALS: 35 horses with colic. PROCEDURE: Blood samples were collected within 30 minutes of arrival at the veterinary hospital from horses referred because of colic. Plasma and serum samples were analyzed for cortisol, epinephrine, norepinephrine, lactate, and electrolyte concentrations and acid-base variables. Heart rate at admission and outcome (survival or nonsurvival) were recorded. Univariate logistic regression was used to calculate crude (unadjusted) odds ratios and 95% confidence intervals. RESULTS: Of the 35 horses with colic, 26 survived. Higher plasma epinephrine, plasma lactate, and serum cortisol concentrations were significantly associated with increased risk of nonsurvival, but plasma norepinephrine concentration was not associated with outcome. Plasma epinephrine concentration was significantly correlated with heart rate (r = 0.68), plasma lactate concentration (r = 0.87), blood pH (r = -0.83), anion gap (r = 0.74), and base excess (r = -0.81). CONCLUSIONS AND CLINICAL RELEVANCE: The risk of death appears to be greater in colic-affected horses with high circulating concentrations of epinephrine and cortisol. The correlation of epinephrine with other biochemical markers of illness severity and with heart rate indicates that the degree of sympathetic activation in horses with colic can be inferred from routinely measured variables.  相似文献

Transportation stress has been implicated as a predisposing factor to respiratory disease in horses. Cross-tying horses individually in stalls is common practice for transporting show and racehorses, but horses also travel in small groups or individually without being restricted by tying. The objective of this study was to compare physiological responses of horses travelling cross-tied or loose during 24 h of road transport. Ten horses were used in a cross-over design consisting of two 4 day trials. In the first trial, 6 horses were cross-tied, while 2 pairs of horses were loose in enclosed compartments. Treatments were reversed in the second trial. Baseline samples were collected on Day 1, horses transported on Day 2, and recovery data collected on Days 3 and 4. Blood samples were collected daily at 0800, 1100 and 2000 h. The mean responses in all horses of serum cortisol, lactate, glucose, alpha1-acid glycoprotein, and total protein concentrations, packed cell volume (PCV), white blood cell (WBC) counts and aminotransferase and creatine kinase were was elevated significantly from baseline during the 4 day study. The response of white blood cell counts, neutrophil to lymphocyte ratios and glucose and cortisol concentrations was significantly elevated in the cross-tied compared to the loose group during transport and recovery. This study supports the recommendation of allowing horses during long-term transportation to travel loose in small compartments, without elevating their head by cross-tying.  相似文献

This study compares the effects of 2 journeys of different lengths (50 km and 200 km) on horses. The first journey lasted about 1 hour, the second about 3 hours. Twelve Standardbred horses were used for the test. Blood samples were collected and analyzed (packed cells volume [PCV], cortisol, aspartate transaminase, alanine transaminase, creatine kinase, lactate dehydrogenase, creatinine, glucose, triglycerides, cholesterol, blood urea nitrogen, calcium, phosphorus, chlorine, total proteins, albumin, and alkaline phosphatase), and heart rate (HR), respiratory rate, and temperature were measured at 5 different experimental times: at rest, at loading, at unloading, and at 2 and 4 hours after return from the journey. Behavior was recorded by direct observation of loading and unloading, within the vehicle, and 2 and 4 hours after arrival in a new stall. The same parameters were recorded at the same times (excluding loading and unloading) in a control group of horses that had not travelled. All the data were analyzed using a repeated measures analysis (analysis of variance).Loading always caused an increase of HR and PCV. The different journey lengths produced significant increases in serum glucose, cortisol concentration, PCV, and HR. Serum cortisol concentrations after short transport were considerably higher than values registered at rest and after long transport. The long journey caused an increase of creatine kinase 2 hours after return. On arrival in the new stalls, the horses that had travelled sniffed less and snorted later than the control group. They ate immediately and more quickly, and those that had travelled 200 km also showed more interest in water than those that travelled for 50 km and the control group. The other behaviors, including exploratory, rest, and play activities, were concentrated in the second 2-hour observation period when the horses tended to feed less. The present trial showed that adaptation to a new stall could be facilitated by offering horses food and fresh water when they arrive, and that for horses that must make a 3-hour journey before a race, it would be better to arrive at their destination at least 4 hours before beginning physical activity.  相似文献
