目的 探讨铁调素(Hepcidin)在慢性牙周炎性牙槽骨吸收过程中的作用,阐明铁离子及其相关调节蛋白在牙槽骨吸收中的重要生物学意义,以期为探索出有效地防治牙槽骨吸收的方法提供理论基础。方法 检测慢性牙周炎患者龈沟液(GCF)中铁离子、核因子KB受体活化因子配体(RANKL)、骨保护素(OPG)的表达水平,分析铁离子的含量与RANKL和OPG表达的相关性以及铁离子的含量与牙周炎性牙槽骨吸收的关系。结果 20例慢性牙周炎组受检牙GCF中铁离子、Hepcidin和RANKL的浓度高于健康对照组(P<0.05),OPG的浓度低于健康对照组(P<0.01)。慢性牙周炎组GCF中RANKL和OPG的浓度与该受检牙4项临床指标:牙周探诊深度(PD)、附着丧失(AL)、菌斑指数(PLI)、龈沟出血指数(SBI)无相关(P>0.05),铁离子浓度与Hepcidin浓度呈正相关(P<0.01),与RANKL浓度呈正相关(P<0.05),与OPG浓度无相关(P>0.05)。结论 慢性牙周炎导致的牙槽骨吸收与局部Hepcidin升高和发生铁过载有关。  相似文献

Successful repair after tissue injury and inflammation requires resolution of the inflammatory response and removal of extracellular matrix breakdown products. We have examined whether the cell-surface adhesion molecule and hyaluronan receptor CD44 plays a role in resolving lung inflammation. CD44-deficient mice succumb to unremitting inflammation following noninfectious lung injury, characterized by impaired clearance of apoptotic neutrophils, persistent accumulation of hyaluronan fragments at the site of tissue injury, and impaired activation of transforming growth factor-beta1. This phenotype was partially reversed by reconstitution with CD44+ cells, thus demonstrating a critical role for this receptor in resolving lung inflammation.  相似文献

为构建猪hepcidin毕赤酵母(Pichia pastoris)分泌型表达载体,实现其真核表达,本文采用RT-PCR方法获得猪肝脏中的铁代谢调节基因hepcidin,通过连接酶使其与真核表达载体pGAPZaA相连构建重组表达质粒pGAPZaA-hepcidin。利用高压电击处理,使质粒载体转入到毕赤酵母中,用含Zeocin抗生素的YPD平板筛选阳性转化子,并做PCR鉴定。结果表明:hepcidin基因准确地插入表达载体pGAPZaA,并已整合到酵母基因组中,并成功构建了酵母重组表达质粒。  相似文献

运用RT-PCR和快速扩增cDNA末端(rapid amplication ofcDNA Ends,RACE)技术获得草鱼(Ctenopharyngodon idellus)hepcidin基因全长cDNA,为774 bp,包括ORF 282 bp、5’UTR 117 bp和3’UTR 383 bp,3’UTR存在1个多聚腺苷酸加尾信号(AATAAA)和1个mRNA不稳定基序(ATTTA)。推定编码93个氨基酸,与其它鱼类hepcidin的序列同一性为27.9%~51.6%;SignalP 4.0软件预测信号肽位于1~24位。在邻接(neighbor-joining,NJ)法构建的系统进化树中,草鱼hepcidin前体肽和其他已报导的鱼类hepcidin前体肽聚为一枝。实时定量PCR(quantitative real-time polymerasechain reaction,qPCR)检测结果显示hepcidin基因mRNA主要表达于肝脏、脾脏、头肾和眼等组织;柱状黄杆菌注射后4~48 h,hepcidin基因在肝脏、脾脏和头肾中表达均显著上调。研究亮点:克隆到草鱼抗菌肽hepcidin一个新基因的全长cDNA,阐明了其所编码的hepcidin与其它脊椎动物hepcidin具有类似的结构与功能;证明其广泛分布于各组织中,参与了对细菌的免疫应答,是固有免疫的重要组成部分。  相似文献

A defining feature of inflammation is the accumulation of innate immune cells in the tissue that are thought to be recruited from the blood. We reveal that a distinct process exists in which tissue macrophages undergo rapid in situ proliferation in order to increase population density. This inflammatory mechanism occurred during T helper 2 (T(H)2)-related pathologies under the control of the archetypal T(H)2 cytokine interleukin-4 (IL-4) and was a fundamental component of T(H)2 inflammation because exogenous IL-4 was sufficient to drive accumulation of tissue macrophages through self-renewal. Thus, expansion of innate cells necessary for pathogen control or wound repair can occur without recruitment of potentially tissue-destructive inflammatory cells.  相似文献

Iron oxides occur ubiquitously in environmental, geological, planetary, and technological settings. They exist in a rich variety of structures and hydration states. They are commonly fine-grained (nanophase) and poorly crystalline. This review summarizes recently measured thermodynamic data on their formation and surface energies. These data are essential for calculating the thermodynamic stability fields of the various iron oxide and oxyhydroxide phases and understanding their occurrence in natural and anthropogenic environments. The competition between surface enthalpy and the energetics of phase transformation leads to the general conclusion that polymorphs metastable as micrometer-sized or larger crystals can often be thermodynamically stabilized at the nanoscale. Such size-driven crossovers in stability help to explain patterns of occurrence of different iron oxides in nature.  相似文献

The chemokine-mediated recruitment of effector T cells to sites of inflammation is a central feature of the immune response. The extent to which chemokine expression levels are limited by the intrinsic developmental characteristics of a tissue has remained unexplored. We show in mice that effector T cells cannot accumulate within the decidua, the specialized stromal tissue encapsulating the fetus and placenta. Impaired accumulation was in part attributable to the epigenetic silencing of key T cell-attracting inflammatory chemokine genes in decidual stromal cells, as evidenced by promoter accrual of repressive histone marks. These findings give insight into mechanisms of fetomaternal immune tolerance, as well as reveal the epigenetic modification of tissue stromal cells as a modality for limiting effector T cell trafficking.  相似文献

[目的]构建黄鳝抗菌肽hepcidin基因大肠杆菌表达载体。[方法]根据Genbank中黄鳝hepcidin基因序列设计引物进行PCR扩增,将测序正确的基因片段连接入载体pET-28a中。[结果]以黄鳝cDNA为模板,扩增出hepcidin基因片段,长度为291 bp,连接到pET-28a上。经PCR扩增、双酶切验证,证实成功构建原核表达载体。[结论]成功构建了黄鳝hepcidin基因大肠杆菌表达载体。  相似文献

通过同源克隆结合RACE的方法从大菱鲆(Scophthal mus maximus)肝脏中克隆了大菱鲆hepcidin抗菌肽基因全长cDNA(GenBank accession number AY994074)。大菱鲆hepcidin基因cDNA全长778bp,包含有1个273bp的开放阅读框,编码1个长90氨基酸的前体肽。将大菱鲆hepcidin抗菌肽基因的成熟肽序列重组至原核表达载体pGEX-4T-1中,用IPTG诱导后,SDS-PAGE电泳显示在29ku处有特异性蛋白条带出现,表达的融合蛋白主要以包涵体形式存在,融合蛋白约占总蛋白的26%。Western-blotting分析发现表达的融合蛋白可以特异性地与GST抗体相识别。这些结果表明大菱鲆hepcidin抗菌肽基因成功实现了原核表达。  相似文献

根据已知鱼类抗菌肽hepcidin的同源性设计简并引物,利用同源克隆结合RACE法从黄鳝(Monopterus albus)肝脏中扩增得到了黄鳝hepcidin基因全长cDNA序列(GenBank accession number FJ436808).结果表明,黄鳝hepcidin基因cDNA全长712 bp,5′端非翻译区有133 bp,3′端非翻译区有306 bp,包含1个273 bp的开放阅读框,编码1个长90氨基酸的前体肽.同源性分析表明,黄鳝hepcidin推测的氨基酸序列与其他鱼类报道的hepcidin具有较高的同源性,并具有8个半胱氨酸这一典型特征,是hepcidin家族的1个新成员.采用半定量PCR法对该基因在健康成体不同组织的转录本和脂多糖(LPS)对该基因表达的影响进行了分析.结果发现,该基因在肝脏中表达量最高,其次是肾脏,在心脏、皮肤、脑、血液、小肠、脾脏和胃中的表达量较低,而在肌肉中没有检测到该基因的转录本;LPS注射24 h后,各组织的hepcidin基因表达量都有明显升高.  相似文献

[目的]为动物趾叶炎的预防和治疗提供免疫学基础。[方法]对昆明种小白鼠注射二磷酸组织胺制备趾叶炎小鼠病理模型,以注射生理盐水为对照。分别于给药后2、14、24、48、72 h及15、30 d取小鼠3~5 mm长的小肠组织块,检测肠组织中炎性细胞、杯状细胞、肥大细胞及其脱颗粒数、P物质表达水平的动态变化。[结果]趾叶炎小鼠的肠腺结缔组织内及其绒毛上P物质表达异常丰富,且P物质呈单根或片状走行,并与肥大细胞串联及相邻排列,随给药时间的延长,P物质表达逐渐增强,肥大细胞及其脱颗粒数也急剧增多 炎性细胞、杯状细胞、腺体数量与肥大细胞的变化趋势一致 给药72 h后,血管内壁出现裂隙,肠粘膜出现水肿。[结论]肥大细胞参与小鼠趾叶炎的发病过程,并影响小鼠肠粘膜的通透性。  相似文献

通过RT-PCR从斑点叉尾鮰肝脏中克隆编码hepcidin原前体肽的基因"pIH"。与参考序列相比,显示两个氨基酸残基的变异。通过对编码hepcidin原前体肽的基因添加EcoR I和HindⅢ酶切位点,选择含"trxA"融合头的pET32a(+)作为表达质粒、E.coliBL21(DE3)作为工程菌,成功构建"pET32a-pIH"原核表达系统。阳性克隆经1 mmol.L-1IPTG诱导、在37℃培养12 h后,表达的"trxA-pIH"融合蛋白70%以上可溶。Tricine-SDS-PAGE分析表明,细胞经超声破碎后的上清通过固化金属离子亲和层析(IMAC)纯化后,可获得高纯度的目的蛋白。  相似文献

Raman spectra of hexagonal close-packed iron (varepsilon-Fe) have been measured from 15 to 152 gigapascals by using diamond-anvil cells with ultrapure synthetic diamond anvils. The results give a Gruneisen parameter gamma(0) = 1.68 (+/-0.20) and q = 0.7 (+/-0.5). Phenomenological modeling shows that the Raman-active mode can be approximately correlated with an acoustic phonon and thus provides direct information about the high-pressure elastic properties of iron, which have been controversial. In particular, the C(44) elastic modulus is found to be lower than previous determinations. This leads to changes of about 35% at core pressures for shear wave anisotropies.  相似文献

Experimental elimination of mast cells from the peritoneal tissues of the rat by distilled water treatment inhibited the increase of vascular permeability which normally follows a passively induced antigen-antibody reaction in peritoneal tissue. 为了解生理性缺铁对库尔勒香梨叶片解剖结构的影响,利用透射电镜和石蜡切片法,比较研究了库尔勒香梨在缺铁状态下不同发育期的叶片解剖结构.结果表明,与正常叶相比,缺铁黄化叶片厚度、叶片栅栏组织厚度均极显著降低(P <0.01),叶片上表皮与下表皮厚度均极显著增加(P <0.01).缺铁黄化叶片维管束细胞排列凌乱,导管严重变形,口径变小,生长后期表现更为突出.黄化叶片叶肉栅栏细胞中叶绿体变形且明显变小,片层系统不能堆垛形成基粒而使基粒片层大部分消失;海绵组织细胞排列松散,细胞内结构解体,甚至整个细胞空泡化.研究认为,香梨叶片外部表现黄化症状是由叶片内部及组织结构变化控制的,对比缺铁导致的标志性显微结构异常以及亚显微结构中特定细胞器的损伤,可以初步区分不同缺素或多重缺素对香梨叶片解剖结构造成的影响.  相似文献

将大菱鲆(Scophthalmus maxinus)hepcidin基因成熟肽序列经过部分改造后采用PCR方法克隆到毕赤酵母胞内表达载体pGAPZB中,构建了重组胞内表达载体pGAPZB—tbhep6his。用BlnI线性化重组质粒后,电击转化毕赤酵母SMD1168。经过Zeocin筛选和PCR扩增转化子基因组筛选,得到了数株基因工程酵母菌。通过SDS—PAGE和Western blot鉴定,发现目的蛋白与预期蛋白的分子量相吻合,并能与特异抗体antihis特异性结合,表明大菱鲆抗菌肽hepcidin基因在酵母菌中进行了表达。大菱鲆抗菌肽酵母表达载体的构建和表达为进一步研究该抗菌肽的功能并开发新型鱼类饵料奠定了理论基础。  相似文献

The normal synovium forms a membrane at the edges of joints and provides lubrication and nutrients for the cartilage. In rheumatoid arthritis, the synovium is the site of inflammation, and it participates in an organized tissue response that damages cartilage and bone. We identified cadherin-11 as essential for the development of the synovium. Cadherin-11-deficient mice have a hypoplastic synovial lining, display a disorganized synovial reaction to inflammation, and are resistant to inflammatory arthritis. Cadherin-11 therapeutics prevent and reduce arthritis in mouse models. Thus, synovial cadherin-11 determines the behavior of synovial cells in their proinflammatory and destructive tissue response in inflammatory arthritis.  相似文献
