Specimens of hoof horn from 187 horses were examined for a possible relationship between clinically affected hooves and the occurrence of pathogenic fungi. Specimens were taken from the coronary band and from the stratum externum and medium of the coronary horn and transferred on to Sabouraud dextrose agar, with and without cycloheximide, and incubated at 28 degrees C. Dermatophytes and mould fungi were identified by their macroscopic and microscopic characteristics. The 732 isolates could be assigned to 26 species of moulds, two different species of the dermatophyte Microsporum and three different species of the dermatophyte Trichophyton. Depending on their pathogenic potential they were assigned to three groups: (i) fungi known to be keratinopathogenic (Acremonium blochii, Alternaria alternata, Alternaria chlamydospora, Geotrichum candidum, Microsporum ferrugineum, Microsporum gypseum, Scopulariopsis brevicaulis, Trichophyton species, Trichophyton mentagrophytes, Trichophyton sch?nleinii, 57 isolates), (ii) a group of uncertain pathogenicity (223 isolates), and (iii) a group of non-pathogenic species (452 isolates). Eighty per cent of the samples from horses with hoof horn lesions and 66.7 per cent of the samples from horses with slightly affected hoof horn contained fungi of the keratinopathogenic group, whereas only 8.9 per cent of the samples from horses with healthy hoof horn contained fungi of this group. There were no significant correlations between the clinical data and the age, sex or breed of the horses or their bedding and hygiene. Twelve species of fungi were isolated from the air in the horses' stables, but none of them belonged to the keratinopathogenic group.  相似文献

Horn samples were taken from the hooves of eight horses with clinical signs of equine onychomycosis in at least one hoof capsule. None of the horses had a documented mycological history. The predominant alterations of the horn capsules were sand cracks, white line disease, brittleness (especially around the nail holes), parakeratosis and bruising. The horn samples were stored in sterile tubes for transportation and transferred onto Sabouraud Dextrose Agar and dermatophyte test agar for mycological examination within 6 h. Fungal cultures were incubated for 30 days at room temperature. Fungal identification was based on colonial morphology and microscopic examination of conidia. Horn samples were also stored at ?80°C until used for scanning electron microscopy (SEM). The fungal culture revealed that the hoof horn from all eight horses was infected with keratinophilic fungi. The keratinopathogenic fungi Trichophyton spp and Scopulariopsis brevicaulis were also detected in six horses. SEM revealed severe alterations of the horn structure in horn samples infected with keratinopathogenic fungi compared to horn samples from a sound hoof. The most evident changes were deterioration of the tubular structure of the horn wall, disruption of the horny layers, superficial lysis of cornified cells and the presence of fungal elements. Samples without dermatophyte or Scopulariopsis infection, in contrast, were similar to healthy hoof horn.  相似文献

Carbohydrate moieties of cell membranes in the epidermis of equine skin and hoof were investigated using lectin-histochemistry. The loss of desmosomes during keratinization leads to a reduced lectin affinity. Lectin binding capacity of the intercellular matrix in the stratum corneum of the epidermis is weak compared to that of the intercellular kitt of the hoof. Qualitative differences in lectin binding of the intercellular kitt of different hoof segments correlate with the quality of the horn in the respective areas.  相似文献

Fungi of the genus Geotrichum are commonly found in the environment and, in some circumstances, they may cause diseases in humans and animals. Although these fungi have been isolated from skin lesions of some animal species, their pathogenic role in horses remains uncertain. With the aim to investigate the role of Geotrichum candidum as etiological agent of dermatomycoses, a retrospective study of 64 horses presenting skin lesions and suspected to have fungal infections was carried out. For each animal, anamnestic data were recorded and fungal culture were performed using hair. Out of 64 cases, 18 (28.1%) were positive for G. candidum and only two (3.1%) for dermatophytes (i.e., one for Microsporum equinum and the other for Microsporum canis). Alopecia, desquamation, and pruritus localized mainly on head and neck were frequently observed in G. candidum infected animals. Most of G. candidum infections were recorded during spring (44.4%). Out of the 18 animals presenting G. candidum infections, eight were treated using a disinfectant with antifungal properties. After one month of treatment, the clinical lesions were healed and fungal cultures resulted negative. The high prevalence of G. candidum in skin lesions of horses and the clinical recover following antifungal treatment indicated that these yeast-like fungi might play a role as etiological agents of horse cutaneous mycoses.  相似文献

Locomotion scoring, lying behaviour and lesion recording during hoof trimming are all ways of evaluating hoof health in dairy cows. The objective of this study was to evaluate the relationship between these measures in a random sample of 1340 cows from 42 Danish dairy herds. The hypothesis was that locomotion scoring and/or the monitoring of lying behaviour could be used as tools to identify cows with hoof lesions, either of the horn or of the skin. Cows were locomotion scored, lying behaviour recorded and data on hoof lesions seen during hoof trimming collected. The results were analysed using logistic regression with hoof lesion as the outcome and locomotion score (1-5), mean duration of lying bouts, parity and lactation stage as explanatory variables. This analysis was undertaken for all types of lesions, for hoof horn lesions only and for skin lesions only. Odds of all hoof lesions and of skin lesions increased with increasing locomotion score and increasing mean duration of lying bouts. Odds of horn lesions also increased with increasing locomotion score, but there was no significant association between horn lesions and the mean duration of lying bouts. It was concluded that locomotion scoring and duration of lying bouts may be used as tools in the management of hoof health in dairy herds.  相似文献

The examination of 2066 skin scrapings from domestic animals in the period from July 1981 to June 1984, revealed dermatophytes in 439 samples. Dermatophytes isolated were: M. canis in dog, cat, cattle, horse, swine, goat, rabbit and hamster, M. equinum in dog and horse, M. gypseum in horse, T. equinum in horse and cattle, T. mentagrophytes in dog, cat, cattle, horse, guinea pig and rabbit, T. verrucosum in cattle and E. floccosum in dog.  相似文献

Four surveys were conducted on different animal groups in the metropolitan area of Perth. Microsporum canis was the only fungus isolated from cats and was prevalent in these animals. Dermatophyte infections in dogs were less common and were caused by M. canis, M. gypseum and Trichophyton mentagrophytes. T. mentagrophytes was isolated from laboratory guinea pigs but not from native animals.  相似文献

Three free-ranging Florida panthers (Felis concolor coryi) were diagnosed with clinical dermatophytosis; two were infected with Trichophyton mentagrophytes, and one was infected with Microsporum gypseum. Two of these panthers were juvenile males that were diagnosed with focal to focally coalescing dermatophytosis; one caused by M. gypseum and the other by T. mentagrophytes. These animals were not treated, and clinical signs resolved spontaneously over 6 mo. The third panther, an adult male from southern Florida, presented with a diffuse dermatophytosis due to T. mentagrophytes infection. Initially, the panther had alopecia, excoriations, ulcerations, and multifocal pyoderma of the head, ears, neck, rear limbs, and abdominal region that progressed to lichenification of the skin and loss of nails from two digits. When topical therapy applied in the field at 45-day intervals was ineffective in clearing the infection, the animal was placed in captivity for intensive oral therapy to prevent further development of dermal mycosis, loss of additional nails, and spread of infection to other panthers. The panther was treated orally with itraconazole (9.5 mg/ kg) in the food s.i.d. for 6 wk. After treatment, nail regrowth occurred but the multifocal areas of alopecia remained. The panther was released back into the wild after two skin biopsy cultures were negative for fungal growth. Temporary removal of a free-ranging animal of an endangered species from its habitat for systemic treatment of dermatophytosis requires consideration of factors such as age, reproductive potential, holding facilities, treatment regimen, and the potential for successful reintroduction of the animal.  相似文献

The structure of the hoof epidermis is the link between nutrition and horn quality. The aim of this study was to demonstrate the relationship of single structures in the process of keratinization and cornification of bovine hoof epidermis to certain nutritional factors such as lipids, minerals and vitamins. Furthermore, we wanted to show the structural changes in the dyskeratotic epidermis caused by an insufficient supply of keratinizing epidermal cells. For our study we used samples of hoof epidermis from 25 dual-purpose dairy cattle, with ages ranging between 2.5 and 4 years. We also obtained a complete set of hooves from a biotin-deficient calf. All samples were investigated by light and transmission electron microscopy, using routine methods as well as histochemical and enzyme-histochemical techniques. We focused on epidermal structures that have a major influence on horn quality and are known to be related to single nutritional factors. The strength of the keratin filament bundles is determined by their cross-linking via sulphur-containing amino acids. Essential fatty acids are required for the synthesis of an intercellular cementing substance connecting the horn cells and establishing a permeability barrier in the stratum corneum. Minerals, in particular calcium, are essential for activation of enzymes that are a prerequisite for physiological keratinization and cornification. Furthermore, vitamins such as biotin are essential in the metabolism of the keratinizing epidermal cells.  相似文献

To gain a basis for diagnosing the frequent pathological changes of the porcine hoof, the fine structure of normal hoofs taken from landrace pigs was examined with the light microscope, the polarizing microscope and the electron microscope. The hoofs were taken mostly from 6 to 7 month-old slaughtered pigs, but also from younger animals, beginning at birth. The hoofs destined for light microscopical study were not softened before frozen sections were made; these sections were stained while floating on the stain. The hoof horn of the pig consists of mostly flat cells which are shaped like pancakes. The horn tubules often have oval cross sections and do not present a distinct boundary with the adjacent intertubular horn. The harder horn of the hoof has more horn tubules per mm2 than the softer horn.  相似文献

Shelly hoof in sheep occurs when the hoof wall becomes detached from the laminar corium; it often then becomes impacted with debris leading to infection, pain and lameness. The problem of shelly hoof is under-reported and is often confused with classical footrot. A study was conducted using data on 9,169 Blackface and Texel sheep from 22 farms in the UK. The aim of the study was to estimate the prevalence of shelly hoof in these breeds, to quantify the genetic basis to shelly hoof, and to describe a pilot trail to characterise the physical properties of horn using Transmission Electron Microscopy (TEM). The results showed that shelly hoof has a high prevalence (47% for Blackface and 24% for Texel ewes) and is under moderate genetic control (h2 = 0.3). The TEM images showed fragmented dorsal wall horn, microscopic crevices leading deeper into the hoof accompanied by bacteria and other micro-organisms. The invasion of bacteria was facilitated by separation of the cells in a characteristic ‘un-zipping’ phenomenon, poor attachment of the keratin to the cell membranes, poor cell membrane quality with a ‘lace-chain’ effect and incomplete keratinisation of the cells.  相似文献

Twenty-three horses with persistent hoof horn defects were treated topically with a hoof disinfectant as part of a hoof care programme for a year. The active ingredients of the disinfectant were a poloaximer-iodine complex, ethylenediamine dihydriodide, isopropyl alcohol and propylene glycol. Hoof trimmings were taken at the start of the study and every six weeks, and examined by scanning and transmission electron microscopy. At the beginning of the study all the horn samples contained large numbers of bacteria, and samples from eight of the horses also had fungal hyphae intermingled with the bacteria. After the application of the hoof disinfectant and adjustments to their diet, there were rapid improvements in the gross appearance of the feet of all the horses; some of them improved within two to three weeks and by 12 weeks the horn quality of all the horses had greatly improved.  相似文献

Diseases of the hoof horn are the most common cause of lameness in the UK dairy herd, and diet, particularly starch intake, is commonly implicated in their development. This study assessed whether replacing starch carbohydrate with sugar beet pulp, in a diet containing 50 per cent of the dry matter as forage, would significantly reduce the development of hoof horn haemorrhages in dairy heifers after calving. It also investigated the effect of replacing wood shavings as cubicle bedding with chopped straw. The heifers fed the high-fibre diet had a significantly lower yield of milk protein and a significantly higher yield of milk fat than the heifers fed the high-starch diet, indicating that the change of diet had affected rumen fermentation. However, the change had no significant effect on the development of hoof horn haemorrhages. The change of bedding material also had no significant effect on the development of hoof horn haemorrhages.  相似文献

This study aimed to carry out a comprehensive morphological examination of the structure and ultrastructure of the membrane-coating granules (MCG) and membrane-coating material (MCM) common to all hoof segments as well as segment-specific features in the MCG and MCM regarding their synthesis, structure and biochemical composition. Tissue samples from the hooves of the left front and right hind limb of 20 adult riding horses were used. Samples were preserved and embedded for histological routine methods as well as for histochemical techniques and tracer techniques for light and electron microscopy. The MCM shows a segment-specific structure, biochemical composition and function, which were demonstrated for the different parts of the hoof. The MCM and its properties are related to the segment-specific horn quality. Besides intercellular structural factors the MCM is of major importance for the structural integrity and the biomechanical properties of the hoof horn. The physiological structure and composition of MCM provides explanation for the existence of clinically well-known sites of predilection for hoof disorders such as the White Line Disease. Alterations of the MCM play a role in dysfunction and diseases of the hoof epidermis; they are particularly involved in the pathogenesis of infectious diseases of the hoof.  相似文献

Reasons for performing study: Abnormal epidermal stem cell regulation may contribute to the pathogenesis of equine chronic laminitis. Objective: To analyse the involvement of p63, a regulator of epidermal stem cell proliferative potential, in chronic laminitis. Methods: Epidermal tissues from skin, coronet and lamellae of the dorsal foot were harvested from 5 horses with chronic laminitis and 5 control horses. Tissues were analysed using histopathology, immunofluorescence microscopy and quantitative immunoblotting Results: Hoof lamellae of laminitic horses had a lower frequency of p63 positive cells than control lamellae, particularly in the distal region. Quantitative immunoblotting confirmed reduced p63 expression in the laminitic distal lamellar region. The decreased p63 expression in laminitic epidermal lamellae was most apparent in the abaxial region adjacent to the hoof wall and highly associated with the formation of terminally differentiated, dysplastic and hyperkeratotic epidermis in this region, whereas lamellae from control horses maintained high p63 expression throughout the axial‐abaxial axis. Conclusions: Expression of p63 in equine skin resembles that reported in other species, including man and rodents, suggesting that p63 can serve as a marker for the proliferative potential of equine epidermal stem cells. p63 expression was significantly lower in the chronic laminitic hoof than in that of control horses, suggesting laminitic hoof epithelium has more limited proliferative potential with a shift towards differentiation. This may reflect reduced activity of epidermal stem cells in laminitic hoof. It is proposed that p63 contributes to the maintenance of hoof lamellae and that misregulation of p63 expression may lead to epidermal dysplasia during lamellar wedge formation. Potential relevance: This study suggests that loss of epidermal stem cells contributes to the pathogenesis of equine laminitis. Autologous transplantation of p63‐positive epidermal stem cells from unaffected regions may have regenerative therapeutic potential for laminitic horses.  相似文献

The study was undertaken to determine whether the claw horn from cows suffering from vertical fissures (sandcracks) is weaker with regard to fracture formation than claw horn from cows without vertical fissures. Fracture toughness was determined using a composite testing procedure previously developed for use in equine hoof horn. Twenty cows with vertical fissures of the lateral front claw were compared with 20 case controls. Due to the calibration technique needed as part of the testing method results were obtained for only five animals in each group; cows with and without vertical fissures had median J-integrals of 8182J/m(2) and 8483J/m(2), respectively, but the difference was not statistically significant. Although the numbers are small it is suggested that since no difference existed in the fracture toughness of the claw horn from the two groups of cattle, vertical fissure formation is not directly dependent on the biomechanical property of the horn itself.  相似文献

研究苦参、白鲜皮和土荆皮提取物在石膏样小孢子菌、犬小孢子菌、须癣毛癣菌3株皮肤癣菌体外联合药敏试验中的相互作用,为临床用药提供参考依据。采用棋盘微量稀释法测量3种中药提取物对皮肤癣菌的联合抑菌指数(fractional in-hibitory concentration index,FICI)对其联用效果进行评价。结果发现,苦参醇提物与白鲜皮醇提物两者联合应用,对3株皮肤癣菌FICI值分别为1,0.75,1,FICI值均≤1,对3株真菌的抑制效果呈相加作用;土荆皮醇提物与苦参醇提物、白鲜皮醇提物联合应用,FICI值均≥2,对3株真菌的抑制效果呈无关或拮抗作用。  相似文献

To evaluate the presence of keratinophilic fungi in the environment, 400 samples were collected from the floors of 50 private veterinary clinics using 55-mm-diameter contact plates, containing mycobiotic agar. After incubation for 15 days at 25°C, the following species were isolated: Microsporum canis, Trichophyton terrestre, Chrysosporium keratinophilum, Chrysosporium sp., Microsporum gypseum, Trichophyton ajelloi, Chrysosporium tropicum, Trichophyton mentagrophytes, Chrysosporium state of Arthroderma tuberculatum and Chrysosporium pannorum. It is concluded that the keratinic material shed by infected pets may contribute to the development and propagation of dermatophytes and related fungi in veterinary clinics. Therefore, such veterinary clinics may represent sites where pets and humans are exposed to risk of infection with keratinophilic fungi from the environment.  相似文献

By causing poorer horn quality, subclinical laminitis is considered to be a major predisposing cause of other hoof problems, particularly sole ulcers in newly calved heifers. In this study the hind hooves of 136 female Friesian/Holstein cattle aged between four months and two years were examined to discover at what age the signs of subclinical laminitis appeared. Sole haemorrhages were found in the hoof horn of calves as young as five months. The consistent finding of these lesions in heifers of all ages indicated that subclinical laminitis of varying degree was a common condition during the early growing period of young dairy heifers.  相似文献

Sequential removal of the dorsal hoof wall was examined in-vivo on a laminitic horse at the walk, revealing an increase in solar force under the margin of the distal phalanx as measured through the use of an in-shoe force measuring system.  相似文献
