The Chinese Meishan pig is prolific breed and it is considered that this pig has a capacity with higher rates of embryonic survival. The oviductal secretory cells may affect the embryonic development and survival. The aim of the present study was to investigate the ultrastructural features of secretory cells in the various regions of the Chinese Meishan pig oviduct during the follicular and luteal phases of the oestrous cycle. In the ampullar secretory cells, numerous secretory granules with moderately electron-dense matrices were present in the supranuclear cytoplasm and exocytosis of secretory granules was observed. The number of secretory granules was dramatically reduced in the ampullar secretory cells in the luteal phase. During the follicular phase in the fimbrial epithelium, the secretory cells contained rough endoplasmic reticulum and Golgi apparatus, but most cells had few small granules. In the luteal phase, the secretory cells in the ampullar and fimbrial epithelia extended beyond the luminal border of the ciliated cells. In the isthmus, many granules were present in the cytoplasm of secretory cells throughout the oestrous cycle, but the number of secretory granules was reduced in the luteal phase. The cytomorphometric data revealed that the height of ciliated cells decreased substantially in the fimbriae and ampulla at the luteal phase, while that of non-ciliated cells was less affected. These results suggest that the drastic reduction of cell height of ciliated cells cause the extrusion of most secretory cells beyond the ciliated cells in the fimbriae and ampulla during the luteal phase. In summary, our ultrastructural observations of Chinese Meishan pig oviduct revealed marked cyclic changes in the ultrastructural features of secretory cells. In particular, the ultrastructural features and the numbers of secretory granules were distinctive for each particular segment. These findings should provide insight into the regional and cellular differences in functions of secretory cells of the Chinese Meishan pig oviduct.  相似文献

The purpose of this study was to investigate the morphological changes in the epithelium of Thai swamp buffalo oviducts at the follicular and luteal phases by histological technique and scanning electron microscopy. The samples from the infundibulum, ampulla, isthmus and uterotubal junction (UTJ) of the oviduct were taken immediately after slaughter at the local abattoir. Noticeable cyclic changes were observed on the epithelial surface of the infundibulum and ampulla, but few changes were present in the isthmus and UTJ. At the follicular phase, the epithelium of infundibulum and ampulla were densely covered with ciliated cells whose cilia concealed the apical processes of the secretory cells. In contrast, the secretory cells dominated in the epithelium at the luteal phase and most of the ciliated cells were hidden by the bulbous processes of these cells. In the isthmus and UTJ at the follicular and luteal phases, the secretory cells were almost flat or gently rounded and covered with numerous microvilli at their apical surface. In conclusion, the histological and ultrastructural observation of Thai swamp oviduct epithelium revealed marked cyclic changes in the cellular differences associated with the main functions of segmental variations.  相似文献

The aim of the present study was to investigate morphological changes and proliferative activities in the epithelium of the canine oviduct with regard to the part of the oviduct – possibly indicating the existence of a locally restricted sperm reservoir – and the stage of the oestrous cycle. Nine healthy adult nulliparous bitches were submitted to ovariohysterectomy at three stages of the cycle: anoestrus (n = 3), late follicular phase (n = 3) and mid‐luteal phase (n = 3). The whole oviduct ranging from the utero‐tubal junction (UTJ) to the infundibulum (IN) was collected, divided into UTJ, IN plus six segments of equal length, i.e. eight oviductal specimens per animal were studied by light microscopy. Morphological characteristics of ovaries and endometrium were recorded macroscopically and verified histologically. The height of oviduct epithelial cells and percentage of ciliated cells (CC) were assessed and the respective data analysed statistically. Proliferative activity was immunohistochemically visualized by means of Ki‐67 antigen detection. Blood was collected and concentrations of oestradiol‐17β and progesterone (P4) were measured. Within the IN and five of the six tissue samples collected from the ampulla and isthmus in anoestrous bitches, the oviductal surface epithelium consisted of low cuboidal cells demonstrating a uniform dark staining intensity. Only a very few scattered lighter staining CC could be detected. Under the influence of oestrogens during late follicular phase, the oviductal epithelium was highly differentiated. Lighter stained CC with apically located nuclei were easily distinguishable from basophilic secretory cells with apical cytoplasmic protrusions. Cell height and percentage of CC were significantly higher than in anoestrus (p ≤ 0.05). During mid‐luteal phase, high levels of P4 were associated with differentiated and dedifferentiated cells as well as cells in regression seen in the mucosal folds of all samples. The percentage of CC and cell height were significantly lower than during late follicular phase (p ≤ 0.05). Further signs of dedifferentiation consisted of a loss of cilia, a pinching off of the apical cytoplasm as well as the presence of debris and macrophages within the oviductal lumen. In the oviductal part of UTJ and the caudal isthmus hormone‐dependent variations in cellular morphology were less distinct. Changes in cell height were minimal and did not differ significantly throughout the oestrous cycle. Hypertrophic cells with large nuclei were predominantly present at these sites, but did not consistently demonstrate signs of ciliation or secretion. Sporadic proliferating activity, visualized by means of Ki‐67 antigen, was mainly seen in some cells of the late follicular phase samples. Thus, overall proliferative activity is generally very low or may occur within a relatively short period of time. It therefore cannot be excluded, that periods exhibiting higher mitotic rates are not included in the present study. It should, however, be mentioned that cells demonstrating morphological signs of apoptosis can only be seen very sporadically within a few specimens during mid‐luteal phase, thus, reflecting low proliferative capacities and minimal cellular turnover found during this study. The results of the present study strongly indicate that oestrogens cause hypertrophy and differentiation, whereas P4 induces gradual dedifferentiation or regression of the oviductal epithelium. Furthermore, they reveal clearly visible changes in the morphology of the tubal epithelium during the oestrous cycle. Depending on the tubal segment, these are, however, variably expressed. Whether the low degree of cellular variation of the UTJ and caudal isthmus is caused by specific hormone concentrations at these sites or specific regulatory mechanisms and may be associated with specific functional properties such as the formation of a locally restricted sperm reservoir needs further investigations.  相似文献

A three-dimensional model has not been clearly established for the porcine oviductal mucosa. The oviducts of 12 cyclic sows were examined by Scanning Electron Microscopy to study the structure and nature of the mucosal arrangement of the oviduct. Epithelial cyclic changes were also studied. The oviductal infundibulum is an asymmetric funnel-shaped structure surrounding the ostium, in which a wide and a narrow side can be distinguished. The mucosa is more complex in the narrow side, showing numerous and tortuous longitudinal primary folds, while the mucosa becomes simpler in the wide side. Around the ostium abdominale wide secondary folds form cul-de-sacs, with their opening pointing in ovarian direction. Areas between folds throughout the lumen of the oviduct show a high-degree of complexity. Inter-fold spaces are occupied by a system of irregular grooves and pockets, with the presence of basal crypts in the caudal oviduct. Marked variations were observed in the oviductal epithelium depending on the oviductal segment, basal or apical areas of the folds, and phase of the oestrous cycle. Cyclical changes were observed in the infundibulum and ampulla, so that prominent and numerous ciliated cells lined apical areas of the folds in the follicular phase; whereas secretary cells were predominant throughout all areas of epithelial surface in the luteal phase. The oestrous cycle phase appeared do not affect the epithelial population cells of the caudal segments of the oviduct: ciliated and secretory cells uniformly lined apical and basal areas of the folds. The topography of the oviduct provides a complex system of regulation, which may influence not only the passage of cells, but also movement of fluid within the oviductal canal.  相似文献

The objective of this work was to study cellular changes in the epithelium of the mare’s oviduct. Oviductal samples were taken from mares at different reproductive stages for optical microscopy and Hoechst 33258 staining. Glandular-like structures were observed in 100% of the oviducts. These structures were of the tubular type and were formed by ciliated and nonciliated epithelial cells arranged in a way similar to the epithelial surface. The amount of structures decreased progressively from the ampulla to the isthmus, but did not change through the different reproductive stages. Histological changes in the epithelium of the oviduct were observed associated with the reproductive stages. In the ampulla, the amount of ciliated cells decreased in the anovulatory phase compared with other reproductive stages. Cords of connective tissue lined by epithelium (trabeculae) and dividing completely the lumen of the oviduct were found in 50% of the oviducts. Epithelial cells projected toward the lumen as large vesicles of cytoplasm, sometimes containing a nucleus. The amount of cells presenting nuclear protrusion varied throughout the oviduct, with highest incidence in the ampulla, decreasing progressively toward the isthmus (P < .05). In addition, nuclear protrusions were higher in number during the anovulatory and luteal phases than in the other reproductive stages (P < .05). These nuclear protruding cells appeared to be extruding from the epithelium and showed no signs of apoptosis based on the histological and fluorescent stains used. The existence of these gland-like structures in the oviductal mucosa should be considered when studying the oviductal physiology in mares.  相似文献

Tissue from the infundibulum region of the uterine tube (oviduct) of guinea pigs, cattle, sheep, and swine was examined by electron microscopy. In all specimens, cilia and ciliary rootlets were present in variable numbers in the ciliated cells during both the follicular and the luteal phases of the estrous cycle. True degeneration of cilia was not evident during luteal phase or pregnancy. The ciliary rootlets in ruminant and nonruminant species had structural similarities to those described in people and rhesus monkeys. These organelles measured approximately 1 mum long in most species studied, but in the cow, the rootlets, extending downward into the cytoplasm from the proximal end of the basal body, reached a length of 2 mum. The rootlets had a cross-striation of thick and thin bands, the period measuring 40 to 60 nm. Mitochondria were closely associated with the rootlets. The rootlets usually formed a small angle to the axis of the cilium. These organelles probably function as anchoring or stabilizing structures for the motile cilia. Results indicated that the rootlets are more widely distributed in mammalian uterine tube cilia than previously postulated. Polyribosomes, microfilaments, microtubules, and electron-opaque fibrous granules were frequently seen in the cytoplasm of the ciliated cells. The presence of fibrous granules in close association with the basal bodies indicate that these granules have a role in the development of cilia and rootlets. Cilia and precursor fibrous granules were also seen in porcine fetal uterine tube epithelial cells.  相似文献

Ultrastructural and ultracytochemical features of the uterine tube (oviduct) infundibulum were studied in 8 Hereford cows, which were slaughtered in pairs on days 1 (estrus), 3, 9 or 10, and 18 of the estrous cycle. Fibrous granules (60 to 80 nm), which are supposedly related to basal body replication, were observed in the apical cytoplasm of ciliated cells. Close association between basal bodies and fibrous granules was apparent, especially during the follicular phase. Cilia were observed throughout of estrous cycle, although degeneration of cilia was not observed at any phase of the cycle. Prominent, striated rootlets were observed during both the follicular and luteal phases of the cycle. Maximum secretory cell differentiation was apparent during the follicular phase, at which time these cells were characterized by having a well-developed, rough endoplasmic reticulum with dilated cisternae, numerous ribosomes, and secretory granules of varied size and density. A prominent feature of the secretory granules was their membranous structure, consisting of concentric lamellae of equal dimensions. During the luteal phase, cytoplasmic protrusions were prominent, and extruded nuclei along with other cytoplasmic organelles were present in the tubal lumen. The presence of a well-developed, rough endoplasmic reticulum and numerous secretory granules during the follicular phase indicates that secretory activity of the uterine tube infundibulum may be stimulated by estrogen. During estrus, the cytoplasm of the stromal cells displayed abundant, rough endoplasmic reticulum with dilated cisternae. The increased and extensively dilated rough endoplasmic reticulum at the time of estrus probably indicates increased protein synthesis by the stromal cells. The presence of adenosine triphosphatase activity on the membrane of cilia suggests that this enzyme is involved in energy-forming reactions related to the vigorous action of cilia. The presence of acid phosphatase activity on the cell membrane of the epithelium, microvilli, and secretory granules may indicate involvement in the secretory mechanism of the cell.  相似文献

This study describes ciliated, nonciliated and mitochondrial luminal epithelial cells of the isthmus in laying and moulting domestic fowls using histological and ultrastructural techniques. The ciliated cells were nonsecretory, while numerous electron‐dense secretory granules were present in the nonciliated cells of laying birds. Mitochondrial cells, occurring in two morphologically distinct forms, constituted the third type of epithelial cell present in the isthmus. The SEM study showed that the luminal epithelium was dominated by ciliated cells, the cilia of which partially obscured adjacent nonciliated cells. The involution of the luminal epithelium in moulting birds occurred via autophagy, apoptosis and necrosis. Autophagic inclusions, which included autophagosomes and autolysosomes, were present in the early degenerative phases of ciliated, nonciliated and mitochondrial cells. Nonciliated cells underwent degeneration via apoptosis, which was characterized by nuclear and cytoplasmic condensation. Apoptotic and necrotic ciliated cells were evident during the intermediate and advanced stages of regression. The presence of apoptotic cell death was confirmed using the TUNEL assay. Loss of cilia via the formation of cilia packets was observed using TEM and SEM. Necrotic cell death occurred in mitochondrial cells during the intermediate and late stages of degeneration. In conclusion, the findings of the study on isthmus involution in moulting birds suggest that autophagy is a process confined to the early stages of degeneration, while apoptosis and/or necrosis occur in the terminal stages of regression.  相似文献

This study was undertaken to describe the surface features of the peritoneal mesothelium covering the genital tract and adjacent ligaments of the cow during the oestrous cycle. The relationship between mesothelial surface and spermatozoa was also evaluated after intra-uterine and intraperitoneal insemination. Surface features of mesothelial cells from 25 cyclic cows were examined by scanning electron microscopy and by image analysis. Presence of spermatozoa was evaluated by scanning electron microscopy in seven additional cows. In the external side of the infundibulum, the oviductal mucosa exceeds the free margin, forming a continuous band measuring 2.5-10 mm in width. This oviductal epithelium shows cyclical variations with a predominance of ciliated cells during the follicular phase. In respect of the mesothelium, no clear morphological differences were observed associated with the side of ovarian bursa (internal versus external), or with the phase of the oestrous cycle. Mesothelial cells covering the uterus and mesometrium have a higher microvilli density and length and a smaller cell surface area than in the oviduct and adjacent structures. The presence of solitary cilia in the mesosalpinx and mesotubarium superius (infundibulo-cornual ligament) of some specimens was also observed. When samples were processed without postfixation in osmium tetroxide, a layer of amorphous material covered all surfaces. After intra-uterine insemination of five cows, no spermatozoa were found on their peritoneal mesothelium. Numerous spermatozoa were found after intraperitoneal insemination being attached throughout mesothelial surfaces. These results indicate that there are morphological differences between regions, but no cyclic changes, in the surface features of mesothelial cells covering the genital tract and adjacent ligaments of the cow, and that spermatozoa can bind to mesothelial surfaces after intraperitoneal insemination.  相似文献

This study was conducted to determine the staining properties, the light and electron microscopic appearance and numerical distribution of mast cells from different regions of the bovine oviduct during the estrous phase and luteal phase. Ten oviducts from bovines in the estrous phase, and 10 from bovines in the luteal phase were used as study samples. In three regions of the oviduct (infundibulum, ampulla, isthmus), mast cells that showed metachromasia with toluidin blue and Ab(+), SO(-) with the combined Ab/SO stains were encountered. Electron microscopic studies of the mast cells showed them to contain two types of granules; a dense homogeneous type and a tiny particulate type. In all three regions of the oviduct, the luteal phase was observed to have a higher number of mast cells per mm2 area than the estrual phase with the isthmus having the most dense population.  相似文献

In this study, we investigated the ultrastructural changes during the prenatal differentiation of oviductal epithelium in 16 bovine embryos and fetuses from CRL of 18.0 cm to a CRL of 94.0 cm. Ciliated and secretory cells of bovine uterine tube, a derivative of the Müllerian duct, differentiate to distinct development stages in the prenatal period. The typical cellular pattern, which is generally characteristic for the adult bovine oviduct, is also obtained during fetal life. In the early stages (CRL 18.0/20.4 cm), the bovine oviductal epithelium appears mostly undifferentiated. The epithelial cells show only a few mitochondria, some cisternae of rough endoplasmic reticulum (rER) and a small Golgi-complex. Most of the cytoplasm is filled with a large amount of glycogen, which decreases during later development. Interspersed between the undifferentiated epithelial cells, a few cells undergoing ciliogenesis can be observed. Ciliogenesis increased significantly during the later prenatal developmental stages. At a CRL of 55.0 cm, ciliated cells appear fully differentiated with mature cells covering their luminal surface. Formation of cilia usually use the acentriolar pathway. Fibrous granules occurred initially in association with the Golgi-apparatus and r(ER) in the supranuclear cytoplasm. Fibrous granules later fuse with deuterosomes and give rise to procentrioles, which are translocated to the luminal plasma membrane. There they become arranged in a line just beneath the apical cell membrane and further differentiate to basal bodies from which the formation of cilia and striated rootlets take place. Clear signs of differentiation of secretory cells were first seen in our material in fetuses with a CRL of 51.0 cm and 64.0 cm. These cells contain a well developed rER and Golgi-apparatus with dilated cisterns. In the supranuclear cytoplasm, the number of secretory granules continuously increases during later development and the cells adapt to the morphology of mature secretory cells at the CRL 94.0 cm.  相似文献

The effect upon the reproduction of total resection of the oviductal isthmus, including the uterotubal and ampullary-isthmus junctions followed by end-to-end ampullo-cornual anastomosis was studied in pigs. Normal cycling gifts of proved fertility were submitted to bilateral isthmus deletion (Group I). Other gilts were submitted only to unilateral isthmic resection, while their contralateral medial isthmus was transversally cut and reanastomosed (Group II). A significantly lower nidation index was obtained after bilateral isthmic resection. The gilts in Group I which did not become pregnant and those in Group II were successfully mated during forthcoming standing oestruses, and were in association to that slaughtered on days 3, 4, or 5 of the cycle, and the characteristics and location of ova were determined. Spermatozoa fertilized the ova, and those cleaved normally in the isthmic-resected tube as well as in the sham-operated oviducts. The isthmic-resected oviduct, however, did not transport cleaved ova into the uterus at the time expected as normal. The findings suggest that the fertilized pig ova cannot reach the uterus at the normal expected time in case of total absence of the isthmic part of the oviduct, resulting in impaired fertility.  相似文献

This study was carried out to compare the lectin-binding pattern in the normal and pathological oviduct of sows during the ovarian cycle. Lectin-binding patterns showed differences between segments, phases of ovarian cycle and presence of morphologic lesions. In the infundibulum, it was observed that the cysts, in the follicular phase, reduced Ricinus communis-I (RCA-I) and Ulex europaeus-I (UEA-I) binding. Furthermore, in the pathological oviducts of the luteal-phase group, there is a reduction of Concanavalia ensiformis (Con-A) reactivity in this segment of the tube having wall cysts, adenomyosis and diverticulus. The Arachis hypogaea (PNA) binding in the infundibulum, during the luteal phase, decreased in the tube having adenomyosis. In animals with wall cysts, the Con-A, Triticum vulgaris (WGA) and RCA-I reactivity was minor in the glycocalyx of the isthmus epithelium during follicular phase. Con-A and Dolichos biflorus (DBA) binding pattern was minor in the luteal-phase isthmus of the tube having wall cysts, adenomyosis and diverticulus. In the ampulla, the wall cysts impaired the Con-A reaction only in the basal region of the epithelium, in the follicular phase. Binding with Con-A was decreased in the ampulla of animals in the luteal phase in the tube lesions with cysts and diverticulus. In addition, the diverticulus observed in the ampulla, during the luteal phase, reduced the PNA tubaric binding. The results of this study showed that the morphologic alterations modify the sugar pattern in the oviduct of sows. These modifications in glycoconjugates may be one of the reasons for the failure of fertility in sows.  相似文献

This study was aimed to determine staining intensity, cellular localization and distribution of the nitric oxide synthase (NOS) enzymes during the sexual cycle in the cow oviduct. Oviduct samples belonging to 20 cows, 10 of which were in the estrual phase and 10 in the luteal phase of the sexual cycle, were examined by an immunohistochemical procedure to determine the presence of the NOS enzymes. In the epithelial cells of the isthmus, endothelial NOS (eNOS) expression showed a strong positive reaction during the estrual phase and a weak positive reaction during the luteal phase in the endothelium and smooth muscle of the blood vessels found in the serosa and lamina propria. eNOS expression was not observed in the epithelium of either the ampulla or the fimbria in the two particular phases of the sexual cycle. The eNOS reactions observed in the blood vessel wall in these regions were stronger during the estrual phase. eNOS activity was not observed in the tunica muscularis in any of the regions of the oviduct. During the estrual phase, it was observed that inducible NOS expression showed a stronger positive reaction in the epithelium and muscle layer of the isthmus and ampulla and in the epithelium of the fimbria, compared to the luteal phase. Neuronal NOS immunoreactivity was observed in the epithelial cells of all oviduct regions and in the muscle layer of the isthmus and ampulla and did not display any significant difference between the estrual and luteal phases.  相似文献

本研究旨在探讨神经生长因子(nerve growth factor,NGF)在母牛卵泡期及黄体期子宫、卵巢、输卵管3种组织中的表达情况。采用FQ-PCR方法和Western blotting技术对成年母牛卵泡期及黄体期的子宫、卵巢、输卵管中的NGF mRNA和蛋白质表达进行了检测。结果表明,NGF mRNA和蛋白质在卵泡期和黄体期的子宫、卵巢、输卵管中均有表达,且卵泡期子宫中NGF mRNA表达量显著高于卵泡期卵巢和黄体期输卵管中NGF mRNA表达量(P0.05),其它组织间无显著差异。  相似文献

为研究促性腺激素受体(FSHR、LHR)在广西雌性水牛生殖器官中的分布情况,运用免疫组化SABC法对处于不同发情周期(卵泡期、黄体期)成年水牛的卵巢、子宫、输卵管中FSHR、LHR分别进行染色定位。结果表明,FSHR/LHR阳性细胞在卵巢主要见于卵巢内膜细胞及卵泡颗粒细胞;子宫主要见于子宫内膜上皮细胞和腺体细胞;输卵管主要见于柱状上皮纤毛细胞。其中,随着发情周期不同,FSHR、LHR的表达量也有所差异,卵巢中卵泡期FSHR、LHR的表达量均高于黄体期;子宫中FSHR的表达量卵泡期高于黄体期,LHR的表达量黄体期高于卵泡期;而输卵管并没有显著差异。  相似文献

Lysophosphatidic acid (LPA) is a naturally occurring simple phospholipid which in the bovine reproductive system can be produced in the endometrium, corpus luteum, ovarian follicle and embryo. In this study, we examined the possibility that LPA receptors are expressed, and LPA synthesized, in the bovine oviduct. We found that the concentration of LPA was highest in infundibulum in the follicular phase of the oestrous cycle and was relatively high during the early‐luteal phase in all examined parts of the oviduct. We also documented that LPA synthesis engages both available pathways for LPA production. The autotaxin (ATX) protein expression was significantly higher in the infundibulum compared to the isthmus during the follicular phase of the oestrous cycle. During the early‐luteal phase of the oestrous cycle, ATX and phospholipase A2 (PLA2) protein expression was highest in ampulla, although the expression of LPARs was not as dynamic as LPA concentration in the oviduct tissue, and we presume that in the bovine oviduct, the most abundantly expressed receptor is LPAR2. In conclusion, our results indicate that the bovine oviduct is a site of LPA synthesis and a target for LPA action in the bovine reproductive tract. We documented that LPAR2 is the most abundantly expressed in the bovine oviduct. We hypothesize that in the bovine oviduct, LPA may be involved in the transport of gametes, fertilization and cellular signalling between the oviduct and cumulus–oocyte complex.  相似文献

Sperm reservoirs in South American Camelids would be crucial for successful fertilization. Since ovulation occurs approximately 36 h after mating, the maintenance of the sperm viability in the oviduct waiting for the ovum is a critical reproductive event. Our study aimed at determining whether the isthmus or the utero tubal junction (UTJ) could function as a sperm reservoir in llama by means of in vivo and in vitro experiments. For the in vivo experiments, the oviducts of adult females with a dominant follicle bigger than 7 mm were examined for the presence of sperm at 6, 18, 24, 28 and 35 h after mating. The results using scanning and transmission electron microscopy showed ultrastructural differences between isthmus and UTJ with respect to (1) predominance of secretory cells in the UTJ and ciliated cells in the isthmus epithelium and (2) cytoplasmic bulbous projection of the secretory cells in the UTJ. Sperm adhered by a mucus‐like substance were seen only in the UTJ at 6, 18, 24 and 28 h post‐mating. Lack of sperm adhered to oviductal mucosa was observed around ovulation (35 h). In vitro experiments demonstrated higher ability of UTJ epithelial cell explants with respect to isthmus explants to bind sperm in a co‐cultured system. The anatomical features and the presence of a sperm bonding agent in the UTJ together with the in vitro differential binding of sperm to UTJ explants strongly suggest that both may be feasible mechanisms that facilitate sperm storage in this oviductal region in llama.  相似文献

旨在探究整合素αv在牦牛繁殖周期输卵管不同部位的表达差异,为进一步了解整合素αv在牦牛生殖过程中发挥的作用提供理论依据。本研究采集卵泡期、黄体期和妊娠期健康母牦牛(3~6岁)的输卵管伞部、壶腹部和峡部样品,将卵泡期、黄体期和妊娠期输卵管分别按照伞部、壶腹部和峡部分为9组,采用实时荧光定量PCR(qRT-PCR)和蛋白免疫印迹法(Western-blotting,WB)检测整合素αv基因和蛋白的表达情况,运用免疫组织化学法检测整合素αv的定位。结果表明:1)qRT-PCR结果显示,在卵泡期,整合素αv基因在输卵管伞部的表达量最高,极显著高于壶腹部和峡部(P<0.01);在黄体期,整合素αv基因在输卵管峡部的表达量最高,极显著高于伞部和壶腹部(P<0.01);在妊娠期,整合素αv基因在输卵管伞部的表达量最高,极显著高于壶腹部和峡部(P<0.01)。2)Western-blotting结果显示,在卵泡期,整合素αv蛋白在输卵管伞部的表达量最高,极显著高于壶腹部和峡部(P<0.01);在黄体期,整合素αv蛋白在输卵管峡部的表达量最高,极显著高于伞部和壶腹部(P<0.01);在妊娠期,整合素αv蛋白在输卵管峡部和伞部的表达差异不显著,但二者极显著高于壶腹部(P<0.01)。3)免疫组织化学结果显示,整合素αv主要分布于输卵管伞部、壶腹部和峡部的纤毛细胞、分泌细胞、基细胞、肌层和浆液腺中。结果显示,整合素αv在牦牛繁殖周期输卵管不同部位中的表达存在显著差异,说明其可能参与了牦牛受精及早期胚胎发育等一系列生殖过程,为牦牛的繁殖性能研究提供了基础资料。  相似文献
