The effect of a heat-stable enterotoxin of Escherichia coli was compared with that of theophylline on ion transport in the pig jejunum, using both in vivo and in vitro techniques. The maximal electrical response to heat-stable enterotoxin was only one-half that of theophylline even though the magnitude of the net secretory response was similar. A net, active secretion of HCO3 was partially responsible for the secretory response induced by heat-stable enterotoxin, whereas theophylline induced an active secretion of chlorine which could account for the entire secretory response. Heat-stable enterotoxin elevated tissue cyclic guanosine monophosphate levels, whereas theophylline elevated both cyclic adenosine monophosphate and cyclic guanosine monophosphate. Cyclic guanosine monophosphate levels induced by heat-stable enterotoxin were markedly potentiated by theophylline. Results suggest that HCO3 secretion in the pig jejunum may be controlled by the cyclic guanosine monophosphate system and this system also activates a neutral secretory process which at high heat-stable enterotoxin doses accounts for the bulk of the net secretion observed. Conversely, the chlorine secretion elicited by theophylline is entirely electrogenic and is consistent with results obtained in other species.  相似文献

Fourteen strains of Escherichia coli of serogroups characteristic of porcine class 2 enterotoxigenic E. coli isolated from pigs or calves were selected for genetic studies. The strains were examined for their ability to cotransfer a number of plasmid-mediated properties during conjugation with E. coli K-12. These properties were antibiotic resistance, and the production of heat-stable enterotoxin, the K99 antigen and colicin and the ability to ferment raffinose. Distinction was made between the two types of heat-stable enterotoxin, STa and STb. All 14 strains were antibiotic resistant and 11 of them cotransferred antibiotic resistance and heat-stable enterotoxin. One strain which transferred heat-stable enterotoxin also transferred the raffinose gene. Among six K99-positive strains which transferred heat-stable enterotoxin, five always cotransferred K99. Three strains had 100% cotransfer of colicin as well as heat-stable enterotoxin and K99. Drug resistance determinants were cotransferred at high frequency with heat-stable enterotoxin for six of eight multiple drug resistant enterotoxigenic E. coli. A 100% cotransfer of combinations of heat-stable enterotoxin, K99, colicin and antibiotic resistance was often associated with a single plasmid band on agarose gel electrophoresis. For some strains, the genes for STa and STb were on the same plasmid and for others they were on separate plasmids. The enterotoxin plasmids ranged in size from 5.2 to 85 Mdal. Heterogeneity in molecular size occurred among enterotoxin plasmids in E. coli of the same serogroup and recovered from the same animal host species.  相似文献

Epizootiologic investigations of a diarrheic syndrome in fattening pigs   总被引:2,自引:0,他引:2  
A diarrheic syndrome linked to Clostridium perfringens was observed in fattening pigs. A good correlation was observed between the onset and the severity of diarrhea and the fecal passage of C perfringens enterotoxin. Intestinal fluids from affected pigs had activity comparable with that detected in a purified C perfringens enterotoxin. Seven pigs excreting enterotoxin were shown to develop serum antibodies to C perfringens enterotoxin.  相似文献

Presence of Escherichia coli enterotoxin genes LT (heat-labile enterotoxin), STaP (heat-stable enterotoxin a, porcine genotype), STaH (heat-stable enterotoxin a, human genotype), and STb (heat-stable enterotoxin b) among 874 swine isolates of E coli was determined, using DNA probes and the DNA colony hybridization technique. Of the 874 isolates evaluated, 45% hybridized with at least one of the enterotoxin gene probes and were designated as enterotoxigenic E coli (ETEC). Eighty-five percent of the ETEC were from pigs with enteric colibacillosis. The remaining 15% were from pigs with edema disease or various other diseases, and from healthy swine. Seventy-four percent of the ETEC hybridized with the STb probe, 52% with STaP, and 31% with LT; ETEC did not hybridize with the STaH probe. Most of the ETEC hybridized with more than one enterotoxin gene probe. Isolates that hybridized with the LT probe also hybridized with STb. The most prevalent gene combination was LT-STb. However, 35% of the ETEC from neonatal (less than or equal to 1 week old) swine with enteric colibacillosis were of the STaP-only genotype, and 33% of the ETEC from older swine with enteric colibacillosis were of the STb-only genotype.  相似文献

Prevalence and characterization of Staphylococcus aureus in young goats   总被引:2,自引:0,他引:2  
Thirty-six Staphylococcus aureus isolates recovered from 35 of 204 young goats at slaughter were characterized. All isolates were susceptible to cephalothin, clindamycin, chloramphenicol, gentamicin, kanamycin, and amikacin. All but 2 were susceptible to erythromycin and tetracycline, and 19 and 20 were susceptible to penicillin and ampicillin, respectively. Thirteen isolates were classified as biotype A, 9 isolates were classified as biotype B, 8 isolates were classified as biotype C, and 6 isolates were classified as intermediate between B and C or were not biotypable. Six biotype A isolates were enterotoxigenic; 4 produced enterotoxin B, 1 produced enterotoxin C, and 1 produced enterotoxin D. Two biotype B strains produced enterotoxin B, and all 8 biotype C isolates produced enterotoxin C and the toxic shock syndrome toxin-1.  相似文献

Thirty-two Escherichia coli colonies were taken from the primary step of cultivation of the jejunal contents of each of 10 dead piglets which had suffered from diarrhea. The organisms of each colony were examined for the presence of adhesion fimbria (F4 (K88) and F5 (K99)), production of heat-stable and heat-labile enterotoxin and of colicins.The presence of heat-labile enterotoxin in the intestinal content of the necropsied pigs was also tested, and results correlated with enterotoxin production of the isolated E. coli strains. In all but 3 pigs, 50–80 % of the E. coli strains were found to produce one or both of the enterotoxins and to possess the F4 of the F5 antigen. All bacteria producing both heat-labile and heat-stable enterotoxin proved to belong toi O group 149 and to possess the F4 antigen. Strains from 1 pig belonged to O group 64 and possessed the F5 antigen; these bacteria produced heat-stable enterotoxin only. Most of the enterotoxin-producing E. coli also produced colicins.After each subcultivation, the strains produced less heat-labile enterotoxin, some becoming negative when assayed.  相似文献

The effect of cholera toxin, heat labile and heat stable Escherichia coli enterotoxin on mucosal cyclic AMP concentrations was determined on the proximal jejunum of weanling pigs and young rabbits. Ligated loops were injected with solutions containing no enterotoxin for control and either cholera toxin, heat labile or heat stable E. coli enterotoxin. The loops were drained after either two, four or six hours incubation at which time accumulated fluid was recorded and mucosal samples removed for determination of cyclic AMP concentration. In the rabbit, cholera toxin and heat labile, but not heat stable E. coli enterotoxin stimulated intestinal secretion while in the pig all three enterotoxins induced net fluid accumulation. Cholera toxin and heat labile, but not heat stable E. coli enterotoxin elevated rabbit mucosal cyclic AMP concentrations. In the pig these enterotoxins had no significant effect on mucosal cyclic AMP concentrations. The results are inconsistent with the hypothesis that the adenyl cyclase system is an essential step for enterotoxin induced intestinal secretion. The activation of intestinal adenyl cyclase by bacterial enterotoxins may only be an associated and not a necessary event for the stimulation of intestinal secretion.  相似文献

One hundred and sixty-nine staphylococcal strains of human origin have been tested for production of enterotoxin A, B or Ci, coagulase activity, DNase activity, typical growth on ETGP-agar, hemolysin production and the breakdown of mannitol under aerobic conditions. Very good correlation was observed between enterotoxin production and coagulase activity, in that 82 % of the enterotoxin producing strains also synthesized coagulase. The correlation between DNase activity and positive reaction in mannitol to enterotoxin production was also good (80 % of the enterotoxic strains produced both DNase and aerobic acid from mannitol). Of the enterotoxin producing strains 66 % hemolysed bovine erythrocytes and 61 % were ETGP-positive. However, the frequency of hemolysing respectively ETGP-positive but non-enterotoxin producing strains was very high, viz. 46 % respectively 32 %. It is concluded that enterotoxin production can not to a satisfactory degree of security be predicted by means of the other biochemical characters.  相似文献

作者对自然状态发酵牛肉、接种金黄色葡萄球菌的发酵牛肉和接种发酵剂(植物乳杆菌、戊糖片球菌)与金黄色葡萄球菌的发酵牛肉中金黄色葡萄球菌及其肠毒素等指标在加工过程中的变化规律做了研究。研究结果表明,自然发酵牛肉中没有金黄色葡萄球菌及其肠毒素;接种发酵剂和金黄色葡萄球菌的发酵牛肉中金黄色葡萄球菌先是上升然后快速下降,且金黄色葡萄球菌肠毒素为阴性;接种金黄色葡萄球菌的发酵牛肉中金黄色葡萄球菌及其肠毒素都显著高于接种发酵剂的发酵牛肉(P<0.05)。由试验结果可知,发酵剂植物乳杆菌和戊糖片球菌对金黄色葡萄球菌有很强的抑制作用。  相似文献

OBJECTIVE: To serotype an enterotoxin gene from Escherichia coli isolated from cows, pigs, and chickens in Korea. SAMPLE POPULATION: Isolates from 37 cows with mastitis, 51 diarrheic pigs, and 5 diarrheic chickens. PROCEDURE: Serogroups and serotypes were identified by slide agglutination testing, using pathogenic E coli sera. Detection of E coli enterotoxins by use of reversed passive latex agglutination and ELISA was compared by proving existence of the gene by polymerase chain reaction (PCR) analysis. RESULTS AND CONCLUSIONS: Detection of E. coli enterotoxin by either method was positive for 1 strain (O20:H10; heat-labile enterotoxin [LT+], heat-stable enterotoxin [STa+]; isolation rate, 2%) and 3 other strains (O111:H10, O119:H9, and O125:H6, STa+; isolation rate, 5.9%) isolated from fecal specimens obtained from diarrheic pigs. The E coli enterotoxin genes were identified by use of PCR analysis in 1 strain containing the 417- and 163-base pair (bp) genes (LT+, Sta+; O20:H10) and in 3 strains containing only the 163-bp gene (STa+; O111:H10, O119:H9, and O125:H6). CLINICAL RELEVANCE: Serotyping of E coli enterotoxin may be used to analyze patterns of transmission among species of domestic animals.  相似文献

In an assay procedure, purified Clostridium perfringens enterotoxin induced the accumulation of fluid in the ileal loop of the axenic pig. The smallest amount of enterotoxin causing a positive response was 25 micrograms (1:32 titer by counterimmunoelectrophoresis [CIEP] ), and 100 micrograms (1:128 CIEP titer) caused a marked response. Possibly, diarrhea in pigs with 1:32 CIEP titer or more of enterotoxin in the feces may be associated with enterotoxigenic C perfringens.  相似文献

A toxoid prepared from the toxin of Vibrio cholera was adjuvanted with aluminium hydroxide and used for immunisation of pregnant gilts. Litters of these and of non-vaccinates were experimentally challenged with Escherichia coli producing either heat labile and heat stable (LT and ST) enterotoxins or ST enterotoxin only. Both the challenge strains of E coli produced high rates of mortality (64 and 68 per cent) and morbidity (80 and 100 per cent) in litters of non-vaccinated dams. Statistically highly significant protection against the LT/ST enterotoxin producing strain of E coli was obtained accompanied by the absence of colonisation of the small intestine by the pathogen. No protection against the ST enterotoxin producing strain was found. It is suggested that this vaccine would not confer passive protection to piglets against K99 and 987-positive E coli which usually produce ST enterotoxin only.  相似文献

Heat-stable E. coli and V. cholerae enterotoxins and E. coli endotoxin were tested on the following smooth muscle preparation: vascular; rabbit aorta; rat mesenteric arterioles, and intestinal, rabbit jejunum; pig duodenum; dog jejunal lamina propria smooth muscle. The results indicated that in most preparations used, the prime action of heat-stable enterotoxin from pathogenic strains of E. coli consisted in neutralizing the effects of both alpha or beta adrenergic agonists. In this respect the effect of enterotoxin appeared similar to that of alpha or beta adrenergic blockers. Using the same models, it was found that this enterotoxin did not interfere with the effects of cholinergic agonists or of biogenic amines. Control heat-stable enterotoxin preparations and other tested substances proved inactive, suggesting that different receptor sites might exist for these agents in our models. It appears that smooth muscle preparations are well suited for bioassay of active heat-stable E. coli enterotoxin.  相似文献

Six tests were applied to 39 strains of Yersinia enterocolitica of various serotypes and from several sources in an attempt to relate the test to pathogenicity of the strains. The tests that were used were the pig gut loop test and the infant mouse test for heat stable enterotoxin, the Sereny and HeLa cell tests for invasiveness, inhibition of growth on magnesium oxalate agar, and the ability to cause diarrhea in infant mice. The pig gut loop test was found to be unsuitable for detection of heat stable enterotoxin but 20 strains produced heat stable enterotoxin that was detected in infant mice. None of the strains was positive in the Sereny test but 21 invaded HeLa cells. The growth of 20 strains was inhibited at 37 degrees C on magnesium oxalate agar and, in the orally-infected mice, 23 strains caused diarrhea or death. These findings indicate a discrepancy between the infant mouse test and the ligated intestine test in pigs for heat stable enterotoxin and a significant difference in Y. enterocolitica heat stable enterotoxin compared with Escherichia coli heat stable enterotoxin because the former failed to elicit a significant response in pig intestine.  相似文献

The role of Escherichia coli as a causative agent of diarrhea in cats was investigated. Isolates of E coli from healthy and diarrheic cats were serotyped and investigated for their biochemical characters, production of cytotoxin activity on Vero cells, heat-labile enterotoxin, heat-stable enterotoxin, and hemagglutination of erythrocytes from other animal species. None of 48 investigated strains produced heat-labile enterotoxin or heat-stable enterotoxin, nor did they agglutinate erythrocytes. Most strains were hemolytic and belonged to O-serotypes 2 and 6. Cytotoxin activity on Vero cells was significantly more common and produced in greater amounts among E coli strains isolated from diarrheic cats, and was neutralized by anti-Shiga-like toxin I serum.  相似文献

Eleven Salmonella strains known to produce enterotoxin under aerobic culture conditions in deferrated (DF) medium at 37°C were shown to produce enterotoxin with and without aeration at 22, 28, 37 and 42°C. Heat-labile enterotoxin was generally produced with growth temperatures up to 37°C irrespective of aeration. Heat-stable enterotoxin was produced up to 42°C, mainly aerobically, as indicated by infant mouse assay (IMA), by six of the eleven strains tested. Nine strains produced heat-stable rapid permeability factor (RPF) in rabbit skin.Cholera anti-toxin neutralized reactivities of Salmonella heat-labile enterotoxin in four different biological assays. Mixed gangliosides also neutralized this activity in the cell—test systems.With guinea-pig erythrocytes, all strains underwent mannose-resistant hemagglutination (MRHA) irrespective of growth temperatures, i.e. 22 and 37°C or medium, i.e., DF, tryptose soy broth (TSB) and colonization factor antigen (CFA) agar. At both growth temperatures, CFA agar-grown cells of each strain caused MRHA of bovine erythrocytes. Excepting three Salmonella typhimurium strains, DF broth-grown cells gave MRHA of bovine, chicken and human group A erythrocytes, CFA agar-grown cells caused MRHA of chicken and human blood, whereas TSB-grown cells caused few MRHA reactions.Salmonellae producing both heat-stable, (ST) and heat-labile, (LT) enterotoxins adsorbed to Phenyl Sepharose whereas salmonellae that produced only LT enterotoxin did not.The presence of MRHA adhesions did not correlate with cell-surface hydrophobicity. However, mannose-resistant hemagglutinins may occur more commonly among salmonellae than has been previously recognized.  相似文献

Three hundred and fifteen E. coli strains isolated from piglets dead due to neonatal E. coli diarrhea or traumatic lesions, were examined for presence of enterotoxin production, K-antigens 88 and 99, antibiotic resistance and colicin-production.Almost 100 % of the E. coli strains positive on enterotoxin production produced colicin, while approx. 17 % of the non-enterotoxigenic strains did so.Ninety-nine % of the E. coli strains harbouring K-antigens were found to produce colicin, while approx. 23 % of the strains without detectable K-antigens did so.In strains where neither K-antigens nor enterotoxin production was found, resistance against tetracycline, neomycin and ampicillin, and sensitivity against streptomycin, was more commonly found among colicin-producing strains, than among strains without colicin production.  相似文献
