Highly lung metastasizing model of canine osteosarcoma in nude mice was established from five subcutaneous implantation cycles of lung tumor deposits. The selection of cells with increased metastatic properties from the parent POS canine osteosarcoma cell line recovered medium sized and polygonal Highly Metastasizing POS cells (HMPOS). The doubling time of HMPOS and POS in culture averaged 30 +/- 1.2 hr and 32 +/- 1.3 hr respectively, and their cell growth patterns in vitro were comparable to their in vivo growth patterns. HMPOS cells produced more tumor deposits (> 20 nodules, > 1 -mm in diameter) of various sizes with replacement of lung tissues at 12 weeks after implantation. POS cells produced fewer and smaller lung deposits (< 10 nodules, 1-mm in diameter). Tumor size and number of metastatic tumor deposits showed a regular association. HMPOS cells developed an osteoblastic type of cellular differentiation subcutaneously and in the lungs. HMPOS micrometastasis along the alveolar walls and blood vessels at 4 weeks averaged 6-7 small tumor locus. Each micrometastatic locus contained an average of 5-7 tumor cells, and developed a pleomorphic osteoblastic type of cellular differentiation. An average of 4 macrometastatic nodules could be seen at 6 weeks, composed of an average of 23 tumor cells, 10 nodules at 8 weeks, 12 nodules at 10 weeks and 20 nodules at 12 weeks. These model provides an opportunity for the evaluation of new treatments against canine lung metastatic osteosarcoma in a nude mice model.  相似文献

Articular chondrocytes are phenotypically unique cells that are responsible for the maintenance of articular cartilage. The articular chondrocytic phenotype is influenced by a range of soluble factors. In particular, members of the bone morphogenetic protein (BMP) family support the articular chondrocytic phenotype and stimulate synthesis of cartilaginous matrix. This study was carried out to determine the importance of BMPs in supporting the differentiated phenotype of articular chondrocytes in vitro. Exogenous BMP-2 supported expression of collagen type II and aggrecan in monolayer chondrocyte cultures, slowing the dedifferentiation process that occurs under these conditions. In contrast, BMP-2 had little effect on expression of these genes in three-dimensional aggregate cultures. Endogenous BMP-2 expression was lost in monolayer cultures, coincident with the down-regulation of collagen type II and aggrecan mRNAs, whereas BMP-2 mRNA levels were stable in aggregate cultures. Antagonism of endogenous BMP activity in aggregate cultures by Noggin or a soluble form of the BMP receptor resulted in reduced expression of collagen type II and aggrecan mRNAs, reduced collagen type II protein and sulfated glycosaminoglycan (GAG) deposition into the aggregate matrices and reduced secretion of GAGs into the culture media. These results indicate that endogenous BMPs are required for maintenance of the differentiated articular chondrocytic phenotype in vitro. These findings are of importance to cell-based strategies designed to repair articular cartilage. Articular chondrocytes require conditions that will support endogenous expression of BMPs to maintain the specialized phenotype of these cells.  相似文献

骨形态发生蛋白4 (bone morphogenetic protein-4,BMP-4)属于转化生长因子超家族成员(TGF-β),参与机体的细胞生长、分化、凋亡、迁移及细胞外基质形成等众多病理、生理活动的调控。BMP-4基因与哺乳动物的繁殖性能密切相关,尤其在卵泡的生长分化过程中起着重要的调控作用,是原始卵泡和卵母细胞生存的必需因子。作者就骨形态发生蛋白4的基因结构、受体种类、信号调控机制及该基因与哺乳动物卵泡发育的关系作一综述。  相似文献

Bone morphogenetic proteins (BMPs) are emerging as a family of proteins crucial in the regulation of fertility and ovulation rate. We have shown that porcine theca cells express BMP receptors, however, there is a paucity of information regarding the effect(s) of BMPs on theca cell function. The purpose of this study was to investigate the effects of BMP-2 and -6 on theca cells cultured under serum-free conditions in terms of steroidogenesis, cAMP release and proliferation. The study was further extended to determine whether BMP responses in theca cells are affected by the addition of granulosa cells to the culture system. Both BMPs suppressed progesterone and androstenedione synthesis by theca cells (P < 0.05) after 144 h in culture. Oestradiol synthesis was suppressed (P < 0.05) by BMP-2, but not BMP-6, and theca cell proliferation was stimulated (P < 0.05) by BMP-6, but not BMP-2, after 144 h in culture. Both BMP-2 and -6 inhibited cAMP release (P < 0.05) by theca cells. Furthermore, progesterone and androstenedione synthesis by co-cultured theca and granulosa cells were suppressed (P < 0.05) whereas cell proliferation was stimulated (P < 0.05). These results provide strong evidence for a functional BMP system in the porcine ovary and that theca cells are responsive to BMPs in terms of steroidogenesis and proliferation. BMP-2 and -6 may have a role as luteinisation inhibitors in this polyovular species.  相似文献

Extraskeletal osteosarcoma is extremely rare in mice. This case report demonstrates a spontaneous murine extraskeletal osteosarcoma that exhibited various histological growth patterns in an ICR mouse. At necropsy, the tumor mass was located in the abdominal wall and was 45 × 30 × 25 mm in size. Histopathologically, the tumor showed the following four growth patterns: a solid pattern of polygonal cells embedded in an osteoid eosinophilic matrix with calcification, an irregular sheet pattern of short spindle cells accompanying some eosinophilic multinucleated cells, a fascicular pattern of spindle cells and a cystic pattern lined by short spindle cells. Immunohistochemically, most of the tumor cells were positive for vimentin, proliferating cell nuclear antigen and osterix. The multinucleated cells mentioned above were desmin positive and were regarded as regenerative striated muscles but not tumor cells. Since no clear continuity with normal bone tissues was observed, the tumor was diagnosed as an “extraskeletal osteosarcoma.”  相似文献

Seventy-three mammary tumors and three mammary tissue specimens were examined to elucidate the expression of bone morphogenetic protein (BMP)-6 in the myoepithelial cells of canine mammary gland tumors. Morphologically, the myoepithelial cells were classified into four types: resting and proliferating cells inside the basement membrane, and spindle- and star-shaped cells proliferating in the outer area of the basement membrane. The characteristics of these myoepithelial cells were confirmed by immunohistochemistry using antibodies raised against keratin, cytokeratin 19, alpha-smooth muscle actin, and vimentin. In simple adenoma, a small number of resting myoepithelial cells was immunopositive for BMP-6. In complex adenomas and benign mixed tumors, all types of myoepithelial cells, depending in some cases on their specific location within the tumor, were immunopositive for BMP-6, but almost all of the tubular epithelial cells were immunonegative. Foci consisting of a proliferation of BMP-6-positive star- and spindle-shaped cells had mucinous stroma with marked hyaline and chondroid changes. In contrast, the foci with BMP-6-negative spindle- and star-shaped cells tended to have mucinous stroma without chondroid change. Several types of mesenchymal cells including chondrocytes, osteoblasts, and fibroblastlike cells in the mixed tumors, showed an intense immunopositive reaction for the BMP-6 antibody, and were located close to the ectopic cartilage and bone matrix. No significant immunoreactivity for BMP-6 was observed in most of the malignant mammary tumors; only one malignant mixed tumor was examined. All of these findings indicate that BMP-6 expression in myoepithelial cells may increase in complex adenomas and benign mixed tumors in canine mammary glands, and that BMP-6 expression is most intense in the vicinity of chondroid matrix in these tumors.  相似文献

骨形态发生蛋白-2(BMP-2)能够促进成骨细胞分化和骨组织的再生。将制备的羟基磷灰石/丝素蛋白(HA/SF)复合支架在50μg/mL BMP-2溶液中浸渍4 h后,获得了载入BMP-2的HA/SF复合支架。体外检测在原核表达系统中表达、纯化的BMP-2有促进细胞增殖的生物活性;BMP-2从HA/SF复合支架中的释放可持续9 d,并保持其生物活性,其中BMP-2在第1天的释放率即达到了43%;细胞实验结果显示,与空白HA/SF复合支架相比,载入BMP-2的HA/SF复合支架可以促进人骨肉瘤细胞(MG-63)的增殖与分化;RT-PCR检测BMP-2的加入使细胞内骨钙素基因mRNA转录水平明显提高,即骨钙素基因的表达增强。研究结果提示:HA/SF复合支架可吸附BMP-2并使其缓慢释放,从而促进细胞的增殖与分化,具有良好的细胞相容性,是一种较理想的骨组织缺损修复材料。  相似文献

Bone morphogenetic protein‐4 (BMP‐4) and BMP‐7, theca cell‐derived growth factors, directly affect the granulosa cell function. The aim of this study was to examine the involvement of BMP‐4 or BMP‐7 in vascular endothelial growth factor (VEGF) expression in bovine granulosa cells. Granulosa cells were collected from small follicles (4–6 mm) and seeded at a density of 2–5 × 105 cells per well in Dulbecco's modified Eagle's medium (DMEM)/F12 medium with BMP‐4 or BMP‐7. The expression of VEGF messenger RNA and protein was the maximum when 1.0 ng/mL of BMP‐4 was added to the culture medium. On the other hand, 10 ng/mL of BMP‐7 significantly increased the expression of the VEGF gene and protein. In addition, BMP‐4 stimulated the expression of Smad1 and Smad5 genes in granulosa cells, whereas BMP‐7 stimulated the expression of Smad5 gene. These results suggested that BMP‐4 and BMP‐7 may be associated with VEGF expression via several specific Smads in bovine granulosa cells: BMP‐4 via Smad1/Smad5 and BMP‐7 via Smad5. In conclusion, theca cell‐derived BMP‐4 and BMP‐7 might contribute to follicular vasculature and development by inducing VEGF expression in granulosa cells.  相似文献

Granulosa cells (GCs) play important roles in the regulation of ovarian functions, and in vitro culture is a relevant model for the study of steroidogenesis in ovarian follicles. Thus, growth factors secreted by the oocyte, like Growth and Differentiation Factor 9 (GDF9) and Bone Morphogenetic Protein 15 (BMP15), play an important part in the luteinization of granulosa cells. The aim of this work was to express GDF9 and BMP15 genes in bovine GCs in vitro and evaluate their effects on the luteinization process. Samples of culture medium and GCs transfected with GDF9 and BMP15 were obtained for 21 consecutive days to analyse the steroidogenic hormones' concentration (progesterone (P4) and estradiol (E2)) and the expression of STAR, GDF9 and BMP15 and their respective receptors. The results demonstrated an inhibitory effect of GDF9 and BMPF15 on P4 secretion in bovine GCs cultured in vitro. Moreover, our study demonstrated the entire expression of their respective receptors (TGFBR1, BMPR1B and BMPR2) and the inhibition of the steroidogenic marker, STAR gene. This work sheds light on a novel biological function of BMP15 and GDF9 in bovine GCs physiology, which could elucidate a non-described biological role for GDF9 and BMP15 in bovine granulosa cells' metabolism.  相似文献

为分析绵羊已知多羔主效基因BMPR1B、BMP15和GDF9的多态性与鲁中肉羊产羔数之间的关系,采用Sequenom MassARRAY■SNP技术检测鲁中肉羊BMPR1B、BMP15和GDF9基因中已知主效位点多态性,并与产羔数进行关联分析。结果表明:BMPR1B基因在鲁中肉羊中存在FecB突变,GDF9基因6个分型位点仅4个位点(G1、G3、G4、G5)在鲁中肉羊中存在多态,BMP15基因中5个分型位点在鲁中肉羊中均不存在多态性。FecB基因3种基因型(CC、CT、TT)突变频率分别为0.16、0.58和0.26,此位点多态性较高(0.25相似文献

BMP‐6 has been found to be important to ovarian cells and oocyte, as well as to uterus. Thus, this study investigated the effect of bone morphogenetic protein (BMP‐6) and recombinant follicle‐stimulating hormone (rFSH) alone or in combination on the in vitro culture (IVC) of isolated caprine secondary follicles (Experiment 1) and the mRNA levels for BMP receptors/Smad signalling pathway (BMPR1A, BMPR2, SMAD1, SMAD4, SMAD5, SMAD6, SMAD7 and SMAD8) in vivo and in vitro using BMP‐6 (Experiment 2). Secondary follicles were cultured in αMEM+ alone (control medium) or supplemented with BMP‐6 at 1 or 10 ng/ml and rFSH alone or the combination of both BMP‐6 concentrations and rFSH. The results from Experiment 1 showed that the antrum formation rate was higher in the BMP‐6 at 1 ng/ml (p < 0.05) than in MEM. In Experiment 2, the mRNA expression for BMPR2, SMAD1, SMAD5 and SMAD6 was detected in non‐cultured control and after in vitro culture (MEM and 1 ng/ml BMP‐6); while the expression of SMAD7 and SMAD8 mRNA was only detected after IVC, SMAD4 was only detected in the BMP‐6 at 1 ng/ml treatment. In conclusion, the low BMP‐6 concentration positively influenced antrum formation and ensured normal mRNA expression for BMP receptor and Smads after IVC of caprine secondary follicles.  相似文献

To study the ectopic chondrogenesis in canine mammary mixed tumors, the expression of bone morphogenetic protein-6 (BMP-6) and specific BMP receptors (BMPRs), BMPR-IA, BMPR-IB, and BMPR-II, was examined using immunohistochemical and immunoblot analysis in 39 canine mammary gland tumors. Immunohistochemically, BMP-6 and all three types of BMPRs were coexpressed in the myoepithelial cells and chondrocytes in six of eight benign mixed tumors. In complex adenomas, myoepithelial cells showed an expression pattern of BMP-6, BMPR-IA, and BMPR-II similar to those in benign mixed tumors, whereas immunoreactivity for BMPR-IB was very mild. The myoepithelial cells proliferating within the basement membrane showed more intense immunoreactivity for BMP-6 and all BMPRs as compared with those proliferating in the interstitial areas. Western blotting analysis revealed immunopositive bands at 40-45 kDa for BMP-6 in the samples from simple and complex adenomas and benign mixed tumors. The BMPR-IB-specific bands at 45 kDa were most detected in benign mixed tumors. Because among BMPRs, BMPR-IB is thought to be the major receptor for BMP-6 for primary chondrogenesis, these findings suggest that the expression of BMP and its receptors on the myoepithelial cells might play a role in the ectopic cartilage formation in canine mammary gland tumors, especially in benign mixed tumors.  相似文献

旨在研究BMP4对睾丸支持细胞增殖的调控。本试验选取0、1、2和3月龄组的健康大足黑山羊公羊各3只,采集睾丸支持细胞,每次试验均设3个生物学重复和3次技术重复,通过体外培养、细胞免疫荧光、基因干扰、过表达、qPCR和Western blotting等技术对BMP4是否通过Id2对山羊睾丸支持细胞进行调控以及它们的调控关系进行验证。结果发现,BMP4在2月龄组大足黑山羊睾丸支持细胞中的表达量极显著高于0、1月龄组(P<0.01);在一定范围内,随着BMP4浓度的增加睾丸支持细胞的增殖活性有所增强,BMP4浓度为200 ng·mL-1时其增殖能力最强(P<0.05);干扰BMP4后,细胞增殖活力显著下降(P<0.05),但在72 h后回到正常水平(P>0.05),PCNA的表达极显著降低(P<0.01),表明干扰BMP4后细胞的增殖能力受到限制,细胞增殖指数测定结果进一步说明,干扰BMP4后细胞的增殖能力受到限制;过表达BMP4后,Id2基因表达水平极显著增加(P<0.01),表明BMP4对Id2具有正向调控作用;相对过表达BMP4再干扰Id2试验组,干扰Id2试验组的PCNA基因的表达水平显著降低(P<0.05),这进一步证明BMP4可以调控该基因和蛋白表达。综上所述,山羊睾丸支持细胞的增殖活力在一定范围内与BMP4的浓度呈正相关;且BMP4能够正向调控Id2的表达,并通过促进Id2的表达进而促进支持细胞的增殖。本研究为阐明BMP4调控山羊睾丸支持细胞的分子机制和生理功能提供了基础。  相似文献

Bone morphogenetic proteins (BMPs) play important roles in controlling fertility and ovulation rate. There is however, little information on the BMP system in the ovary of a large polyovular species. The aims of the present study were to investigate BMP-2 and -6 protein expression in the porcine ovary, their effects on granulosa cells in culture and their mechanism of action. Cells and oocytes were recovered from healthy antral follicles 2–6 mm in diameter. When assessed by Western blotting, oocytes and follicular fluid contained BMP-2 and -6. In addition, BMP-2 and -6 were observed in granulosa cells and BMP-2 was also found in theca cells. Granulosa cells were cultured in a serum-free system for 144 h in the presence of increasing doses (0, 3, 30 and 100 ng/ml) of BMP-2 or BMP-6. Both BMPs suppressed progesterone production in a dose-dependent manner after 48 h (P < 0.001) and 144 h (P < 0.05). Only BMP-6 stimulated cell proliferation at 100 ng/ml (P < 0.05). Investigation into the mechanism of action found that BMP-2 and -6 decreased cyclic adenosine monophosphate (cAMP) production (P < 0.01), expression of 3β-hydroxysteroid dehydrogenase (3β-HSD) protein (P < 0.001) and steroidogenic acute regulatory protein (StAR) (BMP-6 only; P < 0.05). This supports the hypothesis that BMP-2 and -6 act as luteinization inhibitors. In conclusion, these findings provide evidence for the presence of a complex signalling mechanism in the porcine ovary and suggest that both BMP-2 and -6 may act in a paracrine manner to control granulosa cell function in this large polyovulatory species.  相似文献

Before dogs with lung tumors were treated by adoptive immunotherapy, the ability of canine blood lymphocytes (PBL) from the peripheral circulation to differentiate in vitro in the presence of human recombinant interleukin-2 (rIL-2) and become tumoricidal was investigated. The PBL from healthy dogs (n = 6) and dogs with lung tumors (n = 5) were grown in culture medium alone, in the presence of rIL-2 to generate lymphokine-activated killer (LAK) cells, or with phytohemagglutinin (PHA) and rIL-2 to generate autologous-stimulated lymphocytes (ASL). After 4 days, cytotoxicity by the ASL, LAK, and PBL was determined in a 4-hour 51chromium-release assay. Target cells in the assay were short-term cultured enzyme digests of autologous (self), allogeneic (genetically different) primary tumors, and Raji, the xenogeneic human lymphoma cell line. The PBL cultured without rIL-2 were not cytotoxic against any tumor. However, when a dog's PBL were activated in vitro, they killed the dog's own tumor, ASL more effectively than LAK cells. Pulmonary adenocarcinomas and an osteosarcoma metastasis to lung were among the autologous tumors assayed. Against an allogeneic canine osteosarcoma, ASL generated from healthy dogs were significantly more cytolytic than LAK from healthy dogs, or than ASL generated from tumor-bearing dogs. Cytotoxicity was greater against allogeneic tumor than against Raji. Lectin-dependent cellular cytotoxicity, tested by including PHA in the assay medium with lymphocytes and Raji cells, by ASL and LAK was greater than cytotoxicity of Raji without PHA. Because ASL were more cytolytic than LAK against all targets in vitro, they may be more beneficial than LAK for immunotherapy of canine tumors.  相似文献

 以控制部分绵、山羊品种高繁殖力的BMPR IB、BMP15和GDF9基因为候选基因,采用PCR RFLP方法分析闽东山羊BMPR IB,BMP15 和GDF9 基因多态性与繁殖性状的关系。研究发现:多胎品种闽东山羊及南江黄羊在BMPR IB 基因的相应位置上并未发生与Booroola Merino 羊相同的突变,同时也未检测到BMP15 的FecXI, FecXH , FecXB基因及GDF9的FecGH 基因,因此排除了这5个突变位点影响闽东山羊高繁殖力性状的可能性。然而,由于闽东山羊的BMPR IB、BMP15、GDF9 的全基因序列信息的缺乏,尚不能完全断定3个基因对闽东山羊高繁殖力性状没有影响。  相似文献
