The effects of Hg, As, Pb, Cu, Cd, and Cr (1, 2, and 5 mg L?1 each) on Azolla pinnata R. Br. were analyzed. The treatments (2 and 5 mg L?1) of the heavy metal pollutants decreased Hill activity, chlorophyll, protein and dry wt, and increased tissue permeability over control values. The effects were most pronounced with the treatment of 5 mg L?1. The harmful effects of the metals were, in general, found by the treatments in the order: Cd 〉 Hg 〉 Cu 〉 As 〉 Pb 〉 Cr. There was no significant change in these parameters at 1 mg L?1 of the metals over control. Thus Azolla pinnata shows tolerance to the heavy metals tested up to 1 mg L?1 each.  相似文献

Pure cultures of E. coli, Klebsiella and Enterobacter, obtained from hospital patients and from natural waters were tested for their growth patterns by spread plate and membrane filtration procedures at the following temperatures; 35°, 41.5°, 43°, 44.5°, and 35°C for 4 h followed by 18 h at 44.5°C. Results indicated that 44.5°C incubation produces the lowest population estimate and that the application of the membrane filtration technique also reduced the potential population. Three water samples collected during June, August and November were tested for fecal coliform and E. coli populations, with 11 different media (broth and agar) and incubation temperatures of 35°, 41.5°, 43°, 44.5°, and 35°C for 4 h followed by 18 h at 44.5°C. During the study, isolates were collected from all positive MPN tubes at each temperature and from each MF medium-temperature regime, 24 to 50 isolates were collected. From the isolate data corrected coliform (oxidase negative), fecal coliform and E. coli population estimates were made. A sample of feces was diluted in lake water and maintained at 20°C for 56 days. Samples were collected at various times and tested for fecal coliform densities using five media and the same temperature regime as for the lake water samples. Data from these studies indicate that, depending on the age of the population being measured, the temperature of the water sample, and the temperature-media-procedure combination used, fecal coliform and E. coli population estimate techniques measure from 5 to 100% of the potential population.  相似文献

Chromium occurs naturally at trace levels in most soils and water, but disposal of industrial waste and sewage sludge containing chromium compounds has created a number of contaminated sites, which could pose a major environmental threat. This study was conducted to enumerate and isolate chromium-resistant microorganisms from sediments of evaporation ponds of a metal processing plant and determine their tolerance to other metals, metalloids and antibiotics. Enumeration of the microbiota of Cr-contaminated sediments and a clean background sample was conducted by means of the dilution-plate count method using media spiked with Cr(VI) at concentrations ranging from 10 to 1000 mg L?1. Twenty Cr(VI) tolerant bacterial isolates were selected and their resistance to other metals and metalloids, and to antibiotics was assessed using a plate diffusion technique. The number of colony-forming units (cfu) of the contaminated sediments declined with increasing concentrations from 10 to 100 mg L?1 Cr(VI), and more severely from 100 to 1000 mg L?1 Cr(VI). The background sample behaved similarly to 100 mg L?1 Cr(VI), but the cfu declined more rapidly thereafter, and no cfu were observed at 1000 mg L?1 Cr(VI). Metals and metalloids that inhibited growth (from the most to least inhibitory) were: Hg > Cd > Ag > Mo = As(III) at 50 μg mL?1. All 20 isolates were resistant to Co, Cu, Fe, Ni, Se(IV), Se(VI), Zn, Sn, As(V), Te and Sb at 50 μg mL?1 and Pb at 100 μg mL?1. Eighty-five percent of the isolates had multiple antibiotic resistance. In general, the more metal-tolerant bacteria were among the more resistant to antibiotics. It appears that the Cr-contaminated sediments may have enriched for bacterial strains with increased Cr(VI) tolerance.  相似文献

The Algal Assay Procedure Bottle Test was used to investigate the effect of Cd, Cu, and Cr (VI) on the growth of Nile water algae. The results of this investigation indicated that Cd has slight inhibitory effects on algal growth at low concentration (0.05 mg L?1), while it was inhibiting algal growth at higher concentration (> 1.0 mg L?1). In contrast, Cu and Cr did not affect algal growth in all investigated concentrations. Combinations of Cd with Cu and Cr seemed to interact synergistically. The synergism between Cd and Cu was more pronounced than between Cd and Cr. Clear changes in the diversity and redundancy of algal structure took place after metal addition. The inhibitory effect of the studied metals was in the following order: Cd-Cu > Cd - Cr > Cd > Cu > Cr.  相似文献

Land applications of manure from confined animal systems and direct deposit by grazing animals are both major sources of bacteria in streams. An understanding of the overland transport mechanisms from land applied waste is needed to improve design of best management practices (BMPs) and modeling of nonpoint source (NPS) pollution. Plots were established on pasturelands receiving phosphorus-based livestock waste applications to measure the concentrations of Escherichia coli (E. coli), fecal coliform (FC), and Enterococcus present in overland flow at the edge of the field. The flow-weighted bacteria concentrations were highest in runoff samples from the plots treated with cowpies (1.37×105 colony forming units (cfu)/100 ml of E. coli) followed by liquid dairy manure (1.84×104 cfu/100 ml of E. coli) and turkey litter (1.29×104 cfu/100 ml of E. coli). The temporal distribution of fecal bacterial concentrations appeared to be dependent upon both the animal waste treatment and the indicator species, with peak concentrations occurring either at the beginning of the runoff event or during peak flow rates. BMPs could be selected to reduce peak flows or first flush effects depending upon the litter or manure applied to the land. The commercial Biolog System was used to identify the dominant species of Enterococcus present in the cowpie source manure (Enterococcus mundtii 55%) and in the runoff collected from the transport plots treated with cowpies (Enterococcus faecalis 37%). The identification of predominant species of Enterococcus that are associated with specific sources of fecal pollution could greatly assist with identifying the origins of NPS pollution.  相似文献

From the results of in vitro studies carried out on wastewater from a stabilization pond it can be concluded that pH values close to 9, as a single factor, do not play any essential role in the die-off of fecal coliform bacteria. The percentage of elmination that occurs in pond water inoculated with 1.1×1010 cells of E. coli mL?1 where the pH is maintained at 8.2 (original pH) is 99.997%, a much greater reduction than that observed (94.1%) in distilled water inoculated with the same concentration of E. coli and where pH is increased up to 9.  相似文献

The effects of cadmium and copper on larval metamorphosis and mortality of cultured Capitella sp Y were investigated. Metatrochophore larvae were exposed to 0.013 (control), 0.030, 0.080, 0.120, and 0.170 mg Cd L?1 and 0.017 (control), 0.025, 0.060, and 0.075 mg Cu L?1 for 34 days. Larval mortality significantly (p < 0.01) increased with increasing Cd and Cu concentrations (Kruskal-Wallis test), with similar effects for both metals for day 10 (LC50 = 0.035 mg Cd L?1 and 0.035 mg Cu L?1). Metamorphosis was inhibited in larvae exposed to the two metals in the absence of food, the effect of Cu being the most severe. This study suggests that Cd and Cu exposure could be harmful to early developmental stages of Capitella sp Y producing a delay in recruitment into natural populations.  相似文献

A cross-disciplinary research project has been implemented because of increased awareness of the potential environmental effects caused by dispersion of metals from external applications into the environment. The work comprises a 4-year (1998–2002) field exposure of grades 304 and 316 stainless steels, and a laboratory percolation study simulating 20–25 years of chromium and nickel containing runoff water interactions with soil. Total metal annual release rates varied between 0.2 and 0.7 mg m?2 yr?1 for Cr, between 0.1 and 0.8 mg m?2 yr?1 for Ni and between 10 and 200 mg m?2 yr?1 for Fe. Most Cr and Ni is present in an ionic form as a result of the limited presence of organic matter at the immediate release situation. Metal ion concentrations in the runoff water are far below reported ecotoxic concentrations. Studies of the environmental interaction between runoff water from stainless steel and soil show the majority of released Cr and Ni to be retained and their concentrations in percolation water to be very low (0.5–1 μg L?1 and 1–5.5 μg L?1 for Cr and Ni, respectively). Speciation calculations showed Cr to be primarily complexed to dissolved organic carbon while Ni also was present in an ionic form in the solution phase. In this work, the acute effects of copper, chromium and mixtures of both metals on females of the marine crab Carcinus maenas were investigated. The parameters indicative of toxicity were mortality and the activity of the enzymes acetylcholinesterase (AChE), lactate dehydrogenase (LDH) and glutathione-S-tranferases (GST). LC50 values determined at 96 hr were 51.8 mg L-1 for copper, 49.8 mg L-1 for chromium and 15 mg L-1 Cu + 43.6 mg L-1 Cr for the mixture of both metals. Copper, chromium and the mixtures of both metals significantly inhibit haemolymph AChE, ovarian LDH and hepatopancreas GST after an in vivo exposure of 96 hr to concentrations lower than NOEC values for mortality. Furthermore, the mixtures tested showed a more pronounced effect than individual agents.  相似文献

This nutrient solution experiment investigated the effects of zinc (Zn) and cadmium (Cd) on winter wheat growth and enzymatic activity. Twelve nutrient solution treatments were prepared of four zinc levels (0, 0.5, 5 and 50 mg L?1) and three cadmium levels (0, 5 and 50 mg L?1). Cadmium concentrations ≥5 mg L?1 decreased plant growth, superoxide dismutase activity, and leaf and stem zinc concentrations, but increased plant cadmium concentrations, proline content, and peroxidase and catalase activities. Root activity and zinc concentration were highest in the 5 mg L?1 treatment and lowest in the 50 mg L?1 treatment. Zinc concentrations ≥5 mg L?1 inhibited plant growth, but increased proline content and cadmium concentration in stems and leaves. Low levels of zinc (0.5 mg L?1) increased cadmium-induced toxicity in wheat plants but high levels of zinc (50 mg L?1) reduced. In conclusion, these results indicated that the addition of zinc alleviated cadmium toxicity if the zinc/cadmium ratio was >10/1. Additional study needs to be done to quantify zinc content before zinc is supplied to alleviate cadmium toxicity.  相似文献

Thlaspi caerulescens J. and C. Presl is a Zn-hyperaccumulatingplant which has aroused considerable interest with respect to its possible use for phytoremediation of Zn-contaminated soils. In this work, a British population of T. caerulescens, from a soil which was found to have relatively high concentrations of water-extractable Cu (0.22 mg L-1), Pb (0.99 mg L-1) and Zn (6.49 mg L-1), was studied. Its ability to grow and accumulate Zn from hydroponic nutrient solution in the presence of elevated concentrations of Cu and/or Pb was investigated. The chosen concentrations of Cu, Pb and Zn were based on reported water-soluble concentrations of these heavy metals in contaminated soils. When supplied with 32.7 mg L-1 Zn, plants accumulated 19 780 mg kg-1 Zn in their shoot dry matter. This concentration declined by 9.3, 87 and 84% respectively when 5.0 mg L-1 Pb, 1.0 mg L-1 Cu or 2.0 mg L-1 Cu were included in the nutrient solution. Despite the apparent adaptation of this population of T. caerulescens to a Zn/Pb/Cu-contaminated soil, these Cu treatments strongly inhibited growth, but the Pb treatment did not affect growth significantly.  相似文献

A Gram-negative bacterium strain LWD09, capable of growing aerobically on 3,4-dichloroaniline (DCA) as the sole carbon and energy source, was isolated from the farm field. This bacterium was identified as Myroides odoratimimus strain by morphological, physiological, and biochemical characteristics as well as 16S rDNA sequence. Analysis of culture pH, temperature, cells growth, and DCA concentration demonstrated that strain LWD09 could effectively degrade DCA without a lag phase. The kinetics of DCA degradation was well described using the Andrews equation, and the kinetic parameters were as follows: q max?=?1.74?h?1, K s?=?43.5?mg?L?1, and K i?=?230.3?mg?L?1. In addition, strain LWD09 was found to be moderately halophilic and showed the highest power of DCA degradation in 5% NaCl (w/w, %). With initial concentrations of 30, 100, and 200?mg?L?1, 100%, 80.4%, and 33.2% of DCA were transformed after 96?h in 5% NaCl, respectively. These results suggest that strain LWD09 has the potential to degrade DCA in saline wastewater. To date, this is the first report on the degradation of DCA by a M. odoratimimus strain with moderate salinity tolerance.  相似文献

The European Union has introduced a new bathing water directive where future classification of recreational waters will be based on the microbial parameters Escherichia coli, and intestinal enterococci. Introduction of enterococci as a new quality parameter may pose a challenge in some areas because relatively less is known about these organisms compared to E. coli. In the present study, the relative abundance of intestinal enterococci, E. coli, and ten fecal sterol and stanol biomarkers were investigated in water and sediment at two estuarine beach sites affected by fecal pollution. In the bathing water, enterococci were relatively more abundant at low E. coli concentrations. In the sediment, enterococci were generally more abundant than E. coli with surface concentrations between 1.0?×?102 and 4.5?×?103 CFU cm?3. Enterococci populations were relatively similar in water and sediment, and were phenotypically different from that of nearby pollution sources. The putative human specific genetic marker esp in Enterococcus faecium was not detected in water or sediment samples despite occasional inputs of human waste from storm water overflows. Sterol and stanol profiles suggested a direct link between water and sediment pollution profiles on days with wind conditions that facilitated resuspension. Sediment resuspension may occur at wind speeds exceeding 6–8 m s?1, and could contribute significantly to enterococci concentrations in the overlying water. The study emphasized that recontamination of the water column due to wind induced resuspension should be considered when evaluating indicator levels and microbial hazards in estuarine recreational waters.  相似文献

Changes in the soil chemical environment can be expected to increase the leaching of trace metals bound in soils. In this study the mobility of trace metals was monitored in a column experiment for two contaminated urban soils. Four different treatments were used (i.e. rain, acid rain, salt and bark). Leachates were analysed for pH, dissolved organic carbon (DOC) and for seven trace metals (cadmium (Cd), chromium (Cr), copper (Cu), mercury (Hg), nickel (Ni), lead (Pb) and zinc (Zn)). The salt treatment produced the lowest pH values (between 5 and 6) in the effluent whereas the DOC concentration was largest in the bark treatment (40–140 mg L?1) and smallest in the salt and acid treatments (7–40 mg L?1). Cadmium, Ni and Zn were mainly mobilised in the salt treatment, whereas the bark treatments produced the highest concentrations of Cu and Pb. The concentrations of Cu, Cr, and Hg were strongly correlated with DOC (r 2?=?0.90, 0.91 and 0.96, respectively). A multi-surface geochemical model (SHM-DLM) produced values for metal dissolution that were usually of the correct magnitude. For Pb, however, the model was not successful indicating that the retention of this metal was stronger than assumed in the model. For all metals, the SHM-DLM model predicted that soil organic matter was the most important sorbent, although for Pb and Cr(III) ferrihydrite was also important and accounted for between 15 and 50% of the binding. The results confirm the central role of DOC for the mobilization of Cu, Cr, Hg and Pb in contaminated soils.  相似文献

The toxic effects and accumulation of the heavy metals, Cd, Cu, and Zn by the sheath forming blue-green alga Chroococcus paris were investigated. All three of the metals were bound rapidly. Approximately 90% of the total amount of the added metal was bound within 1 min. Further significant binding occurred at a slower rate. The maximum metal binding capacity, as determined by filtration studies, was determined to be 53, 120, and 65 mg g?1 dry algal weight for Cd, Cu, and Zn, respectively. Binding curves for the metals followed the Langmuir adsorption isotherm model. The amount of metal bound increased with increasing pH. Metal binding increased significantly when pH was increased from 4 to 7. Nearly all of the metal was found to be rapidly EDTA extractable. Metals were found to be increasingly toxic to growing cultures in the order, Zn, Cd, and Cu. All of the metals studied exhibited toxic effects at concentrations greater than 1.0 mg L?1. The lowest concentrations used which showed detectable toxicity were 0.1 mg L?1 for Cu and >0.4 mg L?1 for Cd and Zn.  相似文献

The effects of four concentrations (0.5, 1, 5 and 10 μg mL?1) of the heavy metals Hg, As, Pb, Cu, Cd, and Cr on some senescence variables of Cuscuta reflexa Roxb. were studied. All of the treatments, except 0.5 μg mL?1, decreased Hill reaction activity, chlorophyll and protein contents and dry matter percentage in biomass and increased tissue permeability over control data. The harmful effects of the metals were best visible at 10 μg mL?1. The general order of sensitivity was As > Cd > Pb > Hg > Cu > Cr (absolute metal concentration). The data suggest that Cuscuta reflexa shows tolerance to the heavy metals tested up to 0.5 μg mL?1.  相似文献

Long-term irrigation with untreated industrial sewage effluents causes accumulation of high concentrations of chromium (Cr) and other heavy metals in soil and subsequently in crop plants (especially leafy vegetables), which can be phytotoxic to plants and/or a health hazard to animals and humans. Greenhouse experiments were conducted to determine the effects of Cr application on the growth of spinach (Spinacia oleracia L.) and to develop critical toxic ranges of Cr in plants and in soil. The study involved growing of spinach variety ‘Punjab Green’ in a greenhouse on silty clay loam and sandy soils equilibrated with different levels of applied Cr (0, 1.25, 2.5, 5, 10, 20, 40, 80, 160, and 320 mg Cr kg? 1 soil). Plants were harvested at: three growth stages 45, 60, and 90 days after sowing (DAS). Critical toxic ranges were estimated by regressing and plotting data on ammoniumbicarbonate-diethylenetriaminepenta-acetic acid (AB-DTPA) extractable Cr in soil or Cr concentration in plants versus dry-matter yield (DMY) of spinach at the three growth stages. Toxic ranges, i.e., slightly toxic (80%–90%), moderately toxic (70%–80%), and extremely toxic (< 70%) in terms of DMY relative to the attainable maximum DMY, were established for both soils and for plants at all three growth stages. There was no germination of spinach with applied Cr at 320 mg Cr kg? 1 rate in silty clay loam soil and at 40 mg Cr kg? 1 rate in sandy soil due to Cr toxicity. Roots accumulated more Cr in comparison with shoots. Chromium concentrations of 0.47–1.93 mg Cr kg? 1 soil in silty clay loam soil, 0.13–0.94 mg Cr kg? 1 soil in sandy soil, 1.08–5.40 mg Cr kg? 1 plant DM in silty clay loam soil and 0.54–11.7 mg Cr kg? 1 plant DM in sandy soil were found to be toxic. The critical toxicity ranges of Cr thus established in this study could help in demarcating Cr toxicity in soils and in plants such as spinach and other leafy vegetables due to irrigation of soils with untreated sewage water contaminated with chromium.  相似文献

Most tidal creeks in North Carolina are closed or partially closed to shellfishing. These creeks often remain closed due to the inability to determine sources of fecal pollution. This study was designed for intensive fecal coliform monitoring of Futch Creek, N.C., to try and determine sources(s) of fecal pollution. Futch Creek is a mildly polluted tidal creek, with marginal levels of fecal coliforms and could potentially be reopened. Problems in interpreting levels of fecal coliforms and pollution risks are two fold and were extremely pronounced in this study. First, several environmental factors have been shown to influence levels of fecal coliforms. Therefore, effects of temperature, salinity, tidal cycles, and rain events on fecal coliform counts were examined. There were higher fecal coliform levels in the warmer temperatures. There was a strong inverse relationship with salinity, with highest fecal coliform counts in the 10–14 g L-1 range for both the mFC and mTEC counts with no apparent source of pollution. This trend was also observed in three other tidal creeks. Tidal cycles did affect fecal coliform counts with substantially higher counts during low tide and appeared to be more important than rain events. It is apparent that when evaluating several stations in a creek, samples must be taken during the same tidal cycle stage in order to have comparative data. Counts obtained using the mTEC method were consistently higher than mFC counts in all salinity ranges. Basic taxonomic tests were performed on fecal coliforms isolated from three salinity regimes: 0 g L-1, 10–14 g L-1, and 23–26 g L-1. The mFC method in the 10–14 g L-1 (45%) and 23–26 g L-1 (70%) salinity range had the highest incidence of false-positive counts (non E. coli). The mTEC method also had the highest incidence of false-positive counts in the 23–26 g L-1 (27%) and 10–14 g L-1 (24%), none as high as the mFC method. Therefore, the mTEC method appears to be the better of the two but is still not an ideal approach.  相似文献

Algal bioassay studies on the effect of F concentrations on diatoms Nitzschia palea (freshwater) and Amphora coffeaeformis (brackishwater) were conducted in the laboratory. Significant enhancement of growth occurred in N. palea at F concentrations between 10 and 110 mg L?1. In A. coffeaeformis statistically significant stimulation occurred at 70 mg L?1 F and above 90 mg L?1 F the growth declined. The results indicate that the diatom N. palea and A. coffeaeformis can tolerate high F concentration.  相似文献

Split root solution culture experiments were conducted to study the effects of the rare earth element lanthanum (La) on rice (Oryza sativa) growth, nutrient uptake and distribution. Results showed that low concentrations of La could promote rice growth including yield (0.05 mg L?1 to 1.5 mg L?1), dry root weight (0.05 mg L?1 to 0.75 mg L?1) and grain numbers (0.05 mg L?1 to 6 mg L?1). High concentrations depressed grain formation (9 mg L?1 to 30 mg L?1) and root elongation (1.5 mg L?1 to 30 mg L?1). No significant influence on straw dry weight was found over the whole concentration range except for the 0.05 mg L?1 treatment. In the pot and field experiments, the addition of La had no significant influence on rice growth.Lanthanum had variable influence on nutrient uptake in different parts of rice. Low concentrations (0.05 mg L?1 to 0.75 mg L?1) increased the root copper (Cu), iron (Fe), and magnesium (Mg), and grain Cu, calcium (Ca), phosphorus (P), manganese (Mn), and Mg uptake. High concentrations (9 to 30 mg L?1) decreased the grain Ca, zinc (Zn), P, Mn, Fe and Mg, and straw Ca, Mn, and Mg uptake. With increasing La concentration, root Zn, P, Mn, Cu, and Ca concentrations increased, and grain Ca and Fe, and straw Mn, Mg, and Ca concentrations decreased. Possible reasons are discussed for the differences between the effects of La in nutrient solutions and in pot and field experiments.  相似文献
