Serum antibody is shown by passive immunization and absorption experiments to be an important component of defense mechanisms against experimentally induced vibriosis in rainbow trout (Salmo gairdneri). Antigen from heat-killed Vibrio anguillarum injected into trout with Freund's complete adjuvant induced much higher agglutinin titers than formalin-killed bacteria with or without adjuvant, or heat-killed bacteria without adjuvant. Passively transferred trout anti-Vibrio anguillarum serum provided protection against experimental vibriosis in trout for more than 2 months. Passively transferred anti-Vibrio anguillarum rabbit serum also protected trout.  相似文献

The variation in semen production among farmed Atlantic salmon (Salmo salar) and rainbow trout (Salmo gairdneri) has been studied. Both species were stripped at weekly intervals, the Atlantic salmon four times and the rainbow trout three times.The individual variation in volume of semen was very high, particularly in rainbow trout. The total volume of semen obtained was 137 ml (20 ml/kg body weight) in Atlantic salmon and 23 ml (5 ml/kg body weight) in rainbow trout. The intraclass correlation for volume of semen was estimated at 0.73 in Atlantic salmon and at 0.59 in rainbow trout. The correlations between volume of semen and body size (weight and length) were all positive. They were all significant and medium in Atlantic salmon whereas in rainbow trout they were all low and significant only for volume of semen at first stripping.The number of males needed to supply the Norwegian fish farming industry with semen is discussed. It is concluded that the possibility of disseminating genetic improvement throughout the whole population of farmed Atlantic salmon and rainbow trout by transport of semen from selected males is considerable.  相似文献

In this study, different traits that have been associated with bacterial virulence were studied in Yersinia ruckeri. Two isolates that had been shown to cause disease and mortality in experimentally infected rainbow trout were compared with five avirulent isolates. Both virulent isolates showed high adhesion to gill and intestinal mucus of rainbow trout, whereas the majority of non‐virulent strains demonstrated significantly lower adhesion. A decrease in adherence capability following bacterial treatment with sodium metaperiodate and proteolytic enzymes suggested the involvement of carbohydrates and proteins. All strains were able to adhere to and invade chinook salmon embryo cell line (CHSE‐214), fathead minnow epithelial cell line (FHM) and rainbow trout liver cell line (R1). One non‐virulent strain was highly adhesive and invasive in the three cell lines, whereas the virulent strains showed moderate adhesive and invasive capacity. The internalization of several isolates was inhibited by colchicine and cytochalasin‐D, suggesting that microtubules and microfilaments play a role. For all strains, intracellular survival assays showed a decrease of viable bacteria in the cells 6 h after inoculation, suggesting that Y. ruckeri is not able to multiply or survive inside cultured cells. Analysis of the susceptibility to the bactericidal effect of rainbow trout serum demonstrated that virulent Y. ruckeri strains were serum resistant, whereas non‐virulent strains were generally serum sensitive.  相似文献

Coldwater Vibrio species isolated from Atlantic salmon, Salmo salar L., during winter ulcer disease outbreaks at saltwater sites in Norway and Iceland were characterized phenotypically, tested for virulence, and used to evaluate the efficacy of multivalent, oil-adjuvanted vaccines. The intraperitoneal (i.p.) injection of rainbow trout, Oncorhynchus mykiss (Walbaum), in fresh water with one bacteria species isolated during winter ulcer outbreaks, V. ‘viscosus’, produced rapid mortality and disease signs which resembled those observed during natural outbreaks [105 colony-forming units (cfu) fish??1]. Another species, V. ‘wodanis’, was not virulent to rainbow trout (103–106 cfu fish??1). Although vaccination of rainbow trout with a mineral-oil-adjuvanted, injectable vaccine containing V. anguillarum (serotypes 01 and 02), V. salmonicida and Aeromonas salmonicida did not provide protection against injection challenge with V. viscosus, vaccines which included V. viscosus produced significant protection in Atlantic salmon and rainbow trout. Atlantic salmon vaccinated with an oil-adjuvanted vaccine containing V. viscosus, V. wodanis and atypical A. salmonicida produced a relative percentage survival (RPS) of 97% when challenged i.p. with V. viscosus, demonstrating cross-protection between strains from Iceland and Norway. Short-term efficacy was demonstrated in rainbow trout by injection challenge at 21 and 43 days post-vaccination with an oil-adjuvanted vaccine containing V. viscosus, V. anguillarum (01/02), V. salmonicida and A. salmonicida, which produced an RPS of 96–99%. Rainbow trout challenged with V. viscosus at 52 and 362 days post-vaccination produced an RPS of 93% and 79%, indicating that vaccination provided long-term protection. In a similar manner, rainbow trout injected i.p. with 0.2 mL of a vaccine containing the five bacteria species and infectious pancreatic necrosis virus produced a 90% RPS when challenged with V. viscosus 66 days later. The high RPS under a severe challenge burden, along with disease signs in experimental freshwater challenges which resembled the saltwater disease condition, indicated that V. viscosus is a contributing factor to winter ulcer and that vaccination will protect against the disease.  相似文献

This study was conducted to examine the effect of ethanolic ginger (Zingiber officinale) extract on growth performance and some skin mucus immune parameters in rainbow trout (Oncorhynchus mykiss). A total of 600 rainbow trout fingerling (10.07 ± 0.21 g) were randomly assigned to four groups in triplicate and fed for 45 days with 1, 2.5 and 5 g kg?1 of ginger extract and with unsupplemented basal diet as the control. The total protein content and lysozyme, alkaline phosphatase and protease activities of skin mucus were investigated following 15, 30 and 45 days of feeding. Results showed that administration of 2.5 and 5 g kg?1 of ethanolic ginger extract for 45 days improved weight gain, feed utilization efficiency and feed conversion ratio of O. mykiss (P < 0.05). Significantly higher (P < 0.05) skin mucus lysozyme, alkaline phosphatase and protease activity and protein level were observed in the 2.5 and 5 g kg?1 ginger extract groups on the 30th and 45th day compared to the controls. The findings suggest that ethanolic extract of ginger enhances growth performance and skin mucus immune parameters of rainbow trout.  相似文献

The non‐native rainbow trout (Oncorhynchus mykiss) has been introduced worldwide for angling purposes and has established self‐reproducing populations in many parts of the world. Introduced rainbow trout often have negative effects on the native salmonid species, ranging from decrease abundance, growth and survival, to their local extinction. Assessing the effects of introduced rainbow trout on the native species is thus crucial to better set up conservation programmes. In this study, we investigated the effects of non‐native rainbow trout on the diet of native marble trout (Salmo marmoratus) living in the Idrijca River (Slovenia). An impassable waterfall separates the stream in two sectors only a few hundred metres apart: a downstream sector (treatment) in which marble trout live in sympatry (MTs) with rainbow trout (RTs) and an upstream sector (control) in which marble trout live in allopatry (MTa). Specifically, we investigated using stable isotopes the effects of rainbow trout on dietary niche, diet composition, body condition, and lipid content of marble trout. We found dietary niche expansion and niche shift in marble trout living in sympatry with rainbow trout. Compared to MTa, MTs had higher piscivory rate and showed higher body condition and prereproduction lipid content. Our results indicate that the presence of rainbow trout did not have negative effects on marble trout diet and condition and that changes in dietary niche of marble trout are likely to be an adaptive response to the presence of rainbow trout, and further research is needed to better understand.  相似文献

Gastric uptake of recombinant growth hormone in rainbow trout   总被引:1,自引:0,他引:1  
The aim of the present study was to investigate gastrointestinal uptake of recombinant growth hormone in rainbow trout. Recombinant human GH was used in this study to avoid cross reactivity with the endogenous trout GH molecule. Using both in vitro and in vivo approach, the results demonstrate that stomach is the main site of uptake for orally administered intact recombinant human growth hormone (hGH) in rainbow trout. The presence of hGH was verified by western blot analysis in the stomach tissue as well as in the serum after 30 minutes of intubation. No traces of the hGH could be detected after 90 minutes of administration, indicating rapid clearance of the hormone in rainbow trout. Furthermore, no immunoreactive hGH was detected in other tissues including mid-gut, muscle and liver at the time points tested. The findings provide novel information on the gastric uptake of hGH in rainbow trout, and would be of relevance to the application of GH in aquaculture.  相似文献

In 1972 a hybridization experiment with salmonids was initiated at the Fish Breeding Experimental Station, Sunndalsøra. The purpose of this experiment was to study the viability, performance traits and the fertility of hybrids. Hybrids were produced from the following members of the salmon family: Salmon (Salmo salar), sea trout (Salmo trutta), brown trout (Salmo trutta), rainbow trout (Salmo gairdneri) and arctic char (Salvelinus alpinus). All crosses attempted, except those involving the rainbow trout, were relatively successful. Considerable differences in the hatching rates of the different hybrids were observed. All crosses with the arctic char were heavier than the better pure bred specimens at 11 months. In all other crosses the hybrids were lighter than the purebred specimens. The experiment is continuing, it is evident that more research is necessary before the potential value of these hybrids for fish farming can be properly assessed.  相似文献

The treatment of amoebic gill disease (AGD) in cultured Atlantic salmon, Salmo salar L., using mucolytic agents has been previously reported. The agent L‐cysteine ethyl ester reduces salmonid mucus viscosity and potentially increases the flushing of the gill. In the present study, the effects of the mucolytic agent N‐acetyl cysteine (NAC) were assessed. Cutaneous mucus from rainbow trout, Oncorhynchus mykiss Walbaum, and Atlantic salmon was shown to have reduced viscosity when mixed in vitro with 100 or 200 μg/mL NAC. Saltwater‐acclimated rainbow trout and Atlantic salmon were fed an oil‐incorporated, NAC‐medicated diet (8 g NAC/kg diet) for up to 24 d and challenged with inoculation of 300 cells/L Neoparamoeba spp., the etiological agent of AGD. Control fish were fed normal oil‐coated pellets and received no NAC. NAC medication failed to reduce the severity of gill lesions associated with AGD even though the mucus viscosity from medicated fish was less than that of controls. Oral NAC medication does not appear to be an effective method for controlling AGD in salmonids despite reducing cutaneous mucus viscosity.  相似文献

Effects of porcine bile extracts added at three different dietary concentrations 0, 10 and 20 g kg?1 were studied on astaxanthin serum concentration in rainbow trout (mean weight 200 ± 7 g). Astaxanthin from micro‐algae Haematococcus pluvialis and synthetic astaxanthin (CAROPHYLL® pink) were incorporated in diets of rainbow trout at a rate of 100 mg astaxanthin kg?1 of feed. Fish were hand fed twice a day. After 5 days of feeding there was a significant effect of the pigment source on the ratio (total blood astaxanthin per unit body weight to cumulative astaxanthin intake per unit body weight). Trout receiving synthetic astaxanthin showed a significantly (P < 0.05) higher ratio than trout fed algal astaxanthin. Increasing dietary bile extract did not lead to produce any effect on this ratio. The power of the statistical analysis is discussed. Therefore, the interaction (pigment source × dietary bile concentration) showed no more effect.  相似文献

Triploid hybrids between female rainbow trout (Oncorhynchus mykiss Walbaum) and male brown trout (Salmo trutta L.) were tested for farming performances, with reference to parental species. The main drawback of hybrids lay in embryonic and larval mortalities, amounting to 60% on average, and displaying a large variability between spawns. Further survival was inferior to that of diploid, but similar to that of triploid rainbow trout. Hybrid body weight was intermediate between weights of rainbow and brown trout of the same age, mainly as a consequence of differences in precocious growth. Analysis of relative growth rates from 6 to 18 months showed that hybrids were surpassed by rainbow controls in common rearing, but not in separate rearing. Hybrid behaviour was similar to that of rainbow trout. These results are discussed in the scope of providing fisheries managers with original and sterile game fishes. The present study was conducted to evaluate the effects of three commercial astaxanthin preparations (100 mg kg?1 diet) with different solubilities in water, from DSM (Dutch State Mines), BASF (Badische Anilin and Soda Fabrik) and Wisdom Company on pigmentation of flesh and antioxidation of flesh, serum and liver in rainbow trout with an initial weight of 52.07 g. After 60 days of feeding, there were no significant differences in growth or flesh proximate composition of rainbow trout among groups (P > 0.05); the Salmo Fan score, redness and astaxanthin content of flesh in rainbow trout fed diets supplemented with astaxanthins were higher than those of the control group (P < 0.05). At 0, 12, 24, 48 and 72 h after thawing, the flesh malondialdehyde (MDA) content of the three astaxanthin groups was lower than that of the control group (P < 0.05). The total antioxidation capacity (T-AOC) of liver in the three astaxanthin groups was significantly higher, but serum catalase (CAT) activities were lower, than that of the control group (P < 0.05). The results indicate that addition of 100 mg kg?1 astaxanthin from DSM, BASF or Wisdom to the diet could improve flesh redness and liver T-AOC, reduce serum CAT, SOD and flesh MDA and extend the shelf life of flesh, in spite of the different solubilities of the three sources in water.  相似文献

Methemoglobin is hemoglobin containing ferric iron. Methemoglobin cannot bind to oxygen and at high concentrations causes tissue hypoxia. Brook trout (Salvelinus fontinalis) develop significantly greater methemoglobinemia than Atlantic salmon (Salmo salar) or rainbow trout (Oncorhynchus mykiss) following general anesthesia with benzocaine or tricaine methanesulfonate. The objective of this study was to compare the activity of the major methemoglobin reducing enzyme, NADH-dependent cytochrome b5 reductase (CB5R), in brook trout erythrocytes to the activity of CB5R in Atlantic salmon and rainbow trout erythrocytes. Methemoglobin levels were compared using co-oximetry following in vitro incubation of erythrocytes with sodium nitrite (NaNO2). The CB5R activity was measured using a ferricyanide assay. There was significantly greater methemoglobin at time 0 in brook trout erythrocytes than in rainbow trout or Atlantic salmon erythrocytes (2.79 ± 0.29 %, 2.19 ± 0.23 %, 2.08 ± 0.14 %), (P < 0.001). There was significantly greater methemoglobin induction by NaNO2 in brook trout erythrocytes (33.14 ± 3.32 %) than in rainbow trout or Atlantic salmon erythrocytes (28.73 ± 2.92 % and 24.85 ± 1.40 %, respectively), (P < 0.001). The CB5R activity was significantly less in brook trout erythrocytes (median of 3.05 μmol/min/μl) than in rainbow trout erythrocytes (median of 6.73 μmol/min/μl). The CB5R activity in Atlantic salmon erythrocytes (median 4.09 μmol/min/μl) was not significantly different than in brook or rainbow trout erythrocytes. Total methemoglobin at any one time is a balance between induction by oxidants and reduction by antioxidants. Lower CB5R activity in brook trout erythrocytes may contribute to a species-specific sensitivity to methemoglobin induction; however, there are likely additional factors.  相似文献

Plasma levels of insulin in rainbow trout,Oncorhynchus mykiss, Atlantic salmon,Salmo salar, and Pacific coho salmon,Oncorhynchus kisutch and plasma circulating levels of glucagon and glucagon-like peptide, in rainbow trout and Atlantic salmon, were measured by homologous radioimmunoassays. Hormonal levels were compared against the average body weight of the same group of fish. Plasma insulin levels were significantly correlated (r=0.56, 0.46 and 0.42 respectively) with body weight in all three salmonid species. Moreover, rainbow trout from fast-growing families had significantly higher (p<0.005) plasma insulin levels than did fish from slow-growing families. The first evidence of proliferative kidney disease (PKD) in an Austrian river (the River Kamp) was documented in 2016, and no information on the PKD infection status of trout in other rivers was available. Since then, brown trout (Salmo trutta fario) and rainbow trout (Oncorhynchus mykiss) have been collected from rivers in Upper and Lower Austria for different diagnostic purposes. In this study, we summarize the recent findings of a first survey concerning the distribution of Tetracapsuloides bryosalmonae, the causative agent of proliferative kidney disease (PKD), from these samples. Between September 2015 and October 2017, a total of 280 brown trout and 39 rainbow trout were collected from 21 rivers in the provinces of Upper and Lower Austria. T. bryosalmonae was detected by PCR of kidney tissue in 17 of 21 sampled rivers and in 138 of 280 brown trout as well as in 11 of 39 rainbow trout. Pathological signs of PKD (e.g., hypertrophy of the kidney) were observed in 33 analysed brown trout and six rainbow trout samples. No correlations between fish infected by Tbryosalmonae and the parameters size and age class, condition factor, geological origin of the streams and distribution within the river course were found, while positively tested fish are significantly increased at sampling sites exceeding water temperatures of 15°C for median periods of 115 days. The prevalence within the affected streams or stream sections is highly variable, and in single rivers, infection rates of up to 90% are confirmed.  相似文献

The main aim of the present study was to assess the effects of chronic high stocking density on liver proteome of rainbow trout. Rainbow trout juveniles (42.6 ± 2.3 g average body weight) were randomly distributed into six tanks at two stocking densities (low stocking density (LD) = 20 kg m?3 and high stocking density (HD) = 80 kg m?3). Both treatments were performed in triplicate tanks for a period of 60 days. High stocking density caused a reduction in the growth performance compared with LD fish. Lysozyme activity increased with stocking density, while serum complement activity presented the opposite pattern. Serum cortisol and total protein levels did not show significant differences (P > 0.05) between experimental groups. The fish reared at high stocking density showed significantly lower osmolality and globulin values but higher albumin level. The HD group had significantly higher activities of catalase, glutathione peroxidase and superoxide dismutase, and malondialdehyde content in the liver when compared to the LD group. Comparative proteomics was used to determine the proteomic responses in livers of rainbow trout reared at high stocking density for 60 days. Out of nine protein spots showing altered abundance (>1.5-folds, P < 0.05), eight spots were successfully identified. Two proteins including apolipoprotein A-I-2 precursor and mitochondrial stress-70 protein were found to increase in HD group. The spots found to decrease in the HD group were identified as follows: 2-peptidylprolyl isomerase A, two isoforms of glyceraldehydes-3-phosphate dehydrogenase, an unnamed protein product similar to fructose-bisphosphate aldolase, 78 kDa glucose-regulated protein, and serum albumin 1 protein.  相似文献

An experiment was conducted to determine the effects of supplemental dietary l-carnitine, chromium picolinate (Cr-Pic) and their combination on growth performance and serum total protein, cholesterol, triglyceride and glucose of rainbow trout (Oncorhynchus mykiss). A total of 276 rainbow trout were randomly allocated to four groups. Fish (average initial body weight = 151 ± 1.69 g) were fed a basal diet without supplemental l-carnitine and chromium picolinate in the control group. The basal diet was supplemented with either 500 mg/kg l-carnitine, 1.6 mg/kg chromium picolinate or 500 mg/kg l-carnitine plus 1.6 mg/kg chromium picolinate for experimental groups 1 (C), 2 (Cr-Pic) and 3 (C + Cr-Pic), respectively. Fish were fed twice a day to apparent satiation for 58 days. Weight gain, growth rate, feed consumption and feed conversion ratio (feed/gain) were calculated for the whole period. At the end of the study, six fish were chosen randomly from each tank to represent the experiment and frozen at −20°C for subsequent dorsal muscle analysis. The results showed no significant differences in final body weight, weight gain or feed conversion ratio among groups. There were also no significant differences in serum total protein, cholesterol, triglyceride or glucose concentrations among groups. Significantly higher lipid concentration of dorsal muscle was observed in experimental groups 1 and 3. In conclusion, supplemental dietary l-carnitine, chromium picolinate and their combination have no beneficial effects on improving growth performance and feed conversion ratio in rainbow trout. However, dietary l-carnitine slightly increased lipid concentration in dorsal muscle of rainbow trout.  相似文献

Attempts were made to complete all possible interspecific hybrid crosses between Atlantic salmon (Salmo salar), rainbow trout (Salmo gairdneri), brook trout (Salvelinus fontinalis), lake trout (Salvelinus namaycush), and Arctic char (Salvelinus alpinus). Survival was appreciable only in (giving female first) lake × brook, salmon × char, brook × char and char × brook hybrids. Growth rates over a weight increment of 3–30 g averaged 2.14% wet weight/day for all species and hybrids (14.5–17.0°C — excess rations). The highest growth rate, 2.74%, was obtained with the brook (female) × char (male) hybrid and the lowest, 1.63%, in lake trout. Salmon (female) × char (male) hybrids grew faster than salmon; brook trout (female) × char (male) hybrids grew faster than brook trout. Salmo species grew no faster than Salvelinus species but became silvery and had greater salinity tolerance at a smaller size and earlier age. Intergeneric hybrids between female Salmo spp. and male Salvelinus spp. more closely resembled the female parent in processes related to smoltification (silvering and salinity tolerance).  相似文献
