The ability of stable flies (Stomoxys calcitrans), horn flies (Haematobia irritans), and tabanids (Diptera: Tabanidae) to transmit bovine leukosis virus (BLV) was investigated. Stable flies and horn flies were fed on blood collected from an infected cow, and the flies' mouthparts were immediately removed, placed in RPMI-1640 medium, ground, and inoculated into sheep and calves. Infection of sheep occurred with mouthparts from as few as 25 stable flies or 25 horn flies. However, sheep were not infected when removal of stable fly mouthparts was delayed greater than or equal to 1 hour after blood feeding. Infection of calves occurred after inoculation of mouthparts removed immediately after feeding from as few as 50 stable flies or 100 horn flies. Infected blood, applied by capillary action to the mouthparts (labella) of 15 deer flies (Chrysops sp) and a single horse fly (Tabanus atratus) caused infection in each of 2 sheep. Infection did not occur in 2 calves inoculated daily for 5 days with mouthparts from 50 horn flies collected after feeding on a BLV-infected steer. Four calves receiving bites from 75 stable flies interrupted from feeding on a BLV-positive cow also were not infected. Seronegative cattle held for 1 to 4 months in a screened enclosure with positive cattle in the presence of biting flies were not infected with BLV. The feeding behavior of each insect is discussed to assess their potential as vectors of BLV.  相似文献

House flies (Musca domestica) and little house flies (Fannia canicularis) were examined for their ability to take up and harbor a velogenic strain of exotic Newcastle disease virus (ENDV) (family Paramyxoviridae, genus Avulavirus). Laboratory-reared flies were allowed to feed on evaporated milk containing ENDV at a virus concentration of 10(8.3) egg infectious dose (EID)50/0.1 ml or on poultry feces containing an ENDV titer of 10(5.8) EID50/0.1 g. Flies exposed to either infectious food source for 24 hr became transiently infected with virus. Virus persisted predominantly in the mid- and hindgut, with relatively little virus isolated from the remainder of the fly body. Virus persisted similarly in both fly species that were fed evaporated milk containing ENDV, with a maximum ENDV titer of 10(5.98) EID50/fly for the house fly and 10(4.78) EID50/fly for the little house fly at 1 day postexposure; titers decreased on subsequent days to 10(2.38) EID50/fly for house fly and > or = 1 EID50/fly for little house fly at 5 days postexposure. Both fly species acquired viral titers greater than the infective dose for a susceptible chicken (10(3.0) EID50-10(4.0) EID50). In addition, flies fed evaporated milk containing a high titer of ENDV maintained viral titers above the infective dose for up to 4 days postexposure to the infectious food source. Flies fed on infective feces retained a chicken infective dose for only one day. The decrease in viral titer over time was significantly explained by logistic regression for both fly species (P < 0.05). The slope of the regression line was not different for the two fly species (P < 0.05), indicating a similar rate of virus loss.  相似文献

Most in vivo studies with equine infectious anemia virus (EIAV) have been performed in horses and ponies (Equus caballus) with little published information available detailing the clinical responses of donkeys (Equus asinus) to infection with this virus. Consequently, donkeys were inoculated with two strains of EIAV (EIAV(PV) and EIAV(WY)) which have been documented to produce disease in E. caballus. Four ponies, 561, 562, 564 and 567 and two donkeys, 3 and 5 were infected with EIAV(PV) and one horse (94-10) and one donkey (4) were infected with EIAV(WY). Although the horse and ponies all experienced clinical signs of disease, which in some cases were severe, the donkeys remained asymptomatic throughout a 365-day observation period, except for mild transient reductions in platelet counts. The results from serological assays, virus isolation from plasma and detection of plasma-associated viral RNA by RT-PCR, indicated that initial replication of EIAV(PV) and EIAV(WY) was lower in donkeys than in horses and ponies. This conclusion was confirmed using competitive RT-PCR, in which viral RNA levels in the plasma of EIAV(PV)-infected ponies was up to 100,000-fold higher than in infected donkeys during the first 20 days post-infection (dpi). Similar results were obtained in the EIAV(WY)-infected animals, in which viral RNA burdens in the donkey at 20 dpi were 1000-fold less than in the horse. However, infection of donkey and horse monocyte-derived macrophage cultures with EIAV(PV) demonstrated that these cells in vitro were equally susceptible to virus-induced cytopathic effects and yielded similar levels of progeny virus. This result suggests that factors other than host cell permissiveness mediate the clinical differences observed between horses and donkeys infected with EIAV(PV) or EIAV(WY).  相似文献

A successful attempt was made to mechanically transmit bovine leukosis virus (BLV) from a BLV-infected cow with a normal lymphocyte count to sheep by inoculation with horse fly (Tabanus abactor) mouthparts. After interrupted natural feeding, horse flies were anesthetized with CO2. Mouthparts were severed and pooled into a tissue grinder containing medium. Five inocula containing the mouthparts of 10 flies each, and 5 inocula containing the mouthparts of 20 flies each, were prepared and inoculated SC in the right axilla of 10 BLV antibody-negative sheep. Five additional sheep served as controls. Serum samples were collected at 2-week intervals and tested by agar gel immunodiffusion for BLV antibodies. One sheep injected with 20 mouthparts developed antibodies to BLV at 10 weeks after inoculation. Six months after inoculation with fly mouthparts, 1 BLV antibody-negative sheep was randomly selected from each treatment group and injected, in the left axilla, with 3 ml of blood from the donor cow to confirm susceptibility of the sheep. All 3 sheep developed antibodies to BLV within 4 weeks.  相似文献

Aujeszky's disease virus was isolated from the brain of a horse which had shown severe neurological signs, including excessive sweating, muscle tremors and periods of mania. Pathological examination revealed a non-suppurative meningoencephalitis. The virus was propagated in cell culture and inoculated into the conjunctiva and nostrils of two ponies. The ponies developed fever seven days after inoculation and subsequently started to behave abnormally, showing severe neurological signs on the ninth day after inoculation. One pony became excited and the other was depressed. One pony died on the ninth day after inoculation and the other was euthanased on the 10th day. Both ponies had a significant increase in serum antibody titre against the virus. The virus was recovered from several parts of the brains and the eyes of the ponies. Aujeszky's disease in horses therefore fulfils Koch's postulates. Although horses do not appear to be very susceptible to the virus, Aujeszky's disease should be included in the differential diagnosis of horses with fatal or transient neurological signs of disease in areas where the virus is endemic.  相似文献

A horse whose serum reacted equivocally in the agar gel immunodiffusion (AGID) test for equine infectious anemia was studied over a 3-year period. The horse remained afebrile and virus was detected in only 1 of 6 horse inoculation tests. The intensity of AGID test reactions increased temporarily following this evidence for virus. Although the AGID test reaction was equivocal and 5 of the 6 transmission attempts failed, the 1 successful transmission proved the horse was infected.  相似文献

The objectives of the study were to determine the duration of porcine reproductive and respiratory syndrome virus (PRRSV) survival in houseflies (Musca domestica Linnaeus) following feeding on an infected pig, and to determine whether the virus was present on the exterior surface or within the internal viscera of the fly. A total of 210 laboratory-colonized houseflies were allowed to feed to repletion on a pig, experimentally infected with PRRSV on day 7 postinoculation, and then maintained alive under laboratory conditions (27°C). Two subsets (A and B) of 30 flies were collected at each of the following sampling points; 0, 6, and 12 hours post feeding (pf). Subset A contained an extra group of 30 flies collected at 24 hours pf due to the availability of extra flies. Flies in subset A were processed as whole fly homogenates, while the exterior surface washes and digestive organs were collected from flies in subset B. Whole fly homogenates, collected at 0, 6, and 12 hours pf, were positive by both polymerase chain reaction (PCR) and swine bioassay. Digestive organs, collected at 0 and 12 hours pf, were positive by PCR and swine bioassay. The PRRSV RNA was detected by PCR from the exterior surface wash of subset B flies collected at 0, 6, and 12 hours pf; however, only the subset collected at 0 hour pf was swine bioassay-positive. This study indicates that infectious PRRSV can survive within the intestinal tract of houseflies for up to 12 hours following feeding on an infected pig, but only for a short period on the exterior surface of the flies.  相似文献

The enzyme-linked immunosorbent assay (ELISA) antigen-positive and agar-gel immunodiffusion test (AGID)-negative horses do not have infective equine infectious anemia (EIA) virus. The ELISA testing of horse leukocyte culture (HLC) supernatants did detect EIA virus in a HLC that was infected with the Wyoming strain of EIA virus and in HLC derived from horses in febrile, acute, or subacute stages of EIA infection. In supernatants of HLC derived from chronic and inapparent carrier horses, EIA virus was not detected with ELISA. Direct fluorescent antibody tests detected EIA virus in HLC infected with 10(6)TCID50 of the Wyoming strain of EIA virus and in 50% of the HLC from febrile acute or subacute horses. The direct fluorescent antibody testing of HLC derived from chronic and inapparent carrier horses did not detect cell-associated EIA virus. The pony inoculation test proved to be the most reliable and accurate method for detecting infective EIA virus in horses in various stages of EIA infection and accurately correlated with the AGID test.  相似文献

The objectives of the study were to determine the site of porcine reproductive and respiratory syndrome virus (PRRSV) in individual houseflies, to assess whether an individual housefly could transmit PRRSV to a susceptible pig, and to compare the ability of PCR, virus isolation and a pig bioassay to detect PRRSV in houseflies. In the first experiment 26 houseflies were fed on a pig infected experimentally with PRRSV; 13 were processed as a whole fly homogenate, while an exterior surface wash and a gut homogenate were collected from the other 13. Infectious PRRSV was recovered from nine of the whole fly homogenates, 12 of the gut homogenates and one of the exterior surface washes. In the second experiment, two of 10 individual houseflies, which had fed on an infected pig, transmitted PRRSV to a susceptible pig in a controlled manual transmission protocol. In the third experiment, single flies or pools of 30 flies were immersed in different concentrations of a PRRSV inoculum, then tested by PCR, virus isolation and bioassay. The virus was detected at a concentration of 10(1) TCID50/ml by PCR, 10(2) TCID50/ml by the bioassay and 10(3) TCID50/ml by virus isolation.  相似文献

Brain stem auditory-evoked response in the nonanesthetized horse and pony   总被引:3,自引:0,他引:3  
The brain stem auditory-evoked response (BAER) was measured in 10 horses and 7 ponies under conditions suitable for clinical diagnostic testing. Latencies of 5 vertex-positive peaks and interpeak latency and amplitude ratio on the 1st and 4th peaks were determined. Data from horses and ponies were analyzed separately and were compared. The stimulus was a click (n = 3,000) ranging from 10- to 90-dB hearing level (HL). Neither horses nor ponies responded with a BAER at 10 dB nor did they give reliable responses at less than 50 dB. The 2nd of the BAER waves appeared in the record at lower stimulus intensities than did the 1st wave for the horse and pony. Horses and ponies had a decreasing latency for all waves, as a result of increasing stimulus intensity. Latencies were shorter for the ponies than for the horses at all stimulus intensities for the 1st, 2nd, 3rd, and 4th waves, but not the 5th wave. At 60-dB HL, the mean latencies for the 1st through 5th wave, respectively, for the horse were 1.73, 3.08, 3.93, 4.98, and 6.00 ms and for the pony 1.48, 2.73, 3.50, 4.56, and 6.58 ms. Interpeak latencies, 1st to 4th wave, averaged 3.22 ms (horse) and 3.11 ms (pony) for all stimulus intensities from 50- to 90-dB HL and had a tendency to decrease slightly as stimulus intensity increased. Amplitude ratios (4th wave/1st wave) were less than 1 for all stimulus intensities in the horse. In the pony, the ratio was less than 1 at greater than or equal to 70-dB HL and greater than 1 at less than or equal to 60-dB HL.  相似文献

Nowadays, ponies are used more and more for competitive equine sports, driving and racing. The locomotor performance of ponies, however, has been evaluated in only a few kinematic and kinetic studies. To compare the longitudinal development of locomotor characteristics in the pony to the same process in the horse, the kinematics of 23 Shetland ponies were recorded at the trot on a treadmill (3 m/s) at ages 4 and 30 months using a modified CODA-3 apparatus. Furthermore, the influence of specific feeding and training regimes on this development was evaluated. It appeared that stride and stance duration increased with age, but the swing duration and pro/retraction range were similar at foal and mature age. Fore- and hindlimb maximal joint flexion at swing was larger in the mature ponies compared to foals. The elbow and shoulder joint were kept in a more extended position and the stifle joint more flexed, while scapula and pelvis were kept in a more vertical position. These effects are similar to those found in horses. The majority of both temporal and angular kinematic variables correlated significantly between foals and mature ponies. Ponies that were restricted in their feeding, resulting in a low body condition score, demonstrated a flatter gait than ponies fed ad lib (4 cm more height, 60 kg more weight), which trotted at the same velocity with significantly more joint flexion. Extra sprint training on top of free paddock exercise did not significantly influence the longitudinal development of kinematics. From these data, it can be concluded that the longitudinal development of locomotor characteristics in ponies is similar to that in horses. Fatter animals show a more animated gait, while extra sprint training added to free paddock exercise has no detectable effect on the development of kinematics. Aside from training or feeding regimes, in the Shetland pony breed, it appears possible to recognise 'good movers' at a young age.  相似文献

To investigate the influence of overweight and dietary fat supplementation on lipid and insulin glucose metabolism of Shetland ponies, eight Shetland pony geldings were fed a hypercaloric (30 MJ DE/150 kg bwt. and day) fat diet (10% fat as soybean oil) or a carbohydrate control diet for nine months until ponies gained an overweight of 15%. Afterwards oral glucose tolerance tests (oGTT; 5, 6 mmol/kg bwt.) were performed after a 12 hour fast and after a fast which led to an increase of plasma triglyceride concentrations to a threshold of 3 mmol/l (36-65 hrs.). Plasma concentrations of glucose, insulin, triglycerides and non esterified fatty acids (NEFA) were determined for 480 minutes after the glucose load. Ponys having had received the control diet tended to a higher insulin secretion in case of both oGTTs, whereas the glucose tolerance was similar in both groups but lower than in ponies of normal weight. During the oGTTs after fasting leading to the plasma triglyceride threshold, triglyceride concentrations decreased significantly (p < 0.05) faster and stronger in fat fed ponies. Additionally, fat fed pony showed significantly (p < 0.05) lower NEFA levels. The results of this study demonstrate a positive effect of fat feeding on the triglyceride clearance of overweight Shetland ponies.  相似文献

Identifying reservoirs and transmission routes for bovine viral diarrhea virus (BVDV) are important in developing biosecurity programs. The aim of this study was to evaluate BVDV transmission by the hematophagous horn fly (Haematobia irritans). Flies collected from four persistently infected cattle were placed in fly cages attached to principal (n?=?4) and control (n?=?4) BVDV-naïve calves housed individually in isolation rooms. Flies were able to feed on principal calves, but a barrier prevented fly feeding from control calves. Flies were tested for BVDV by RT-PCR and virus isolation at time of collection from PI cattle and after 48 h of exposure on BVDV-naïve calves. Blood samples were collected from calves and tested for BVDV infection. Virus was isolated from fly homogenates at collection from PI animals and at removal from control and principal calves. All calves remained negative for BVDV by virus isolation and serology throughout the study. Bovine viral diarrhea virus may be detected in horn flies collected from PI cattle, but horn flies do not appear to be an important vector for BVDV transmission.  相似文献

Eight normal ponies placed in direct contact with ponies experimentally infected with Ehrlichia risticii for 30 to 90 days did not develop signs of Potomac horse fever. They also did not seroconvert, and they remained susceptible to IV infection. One of 8 ponies that were force fed fresh feces from infected ponies while in direct contact with ponies experimentally infected with E. risticii developed Potomac horse fever and seroconverted. The other 7 remained asymptomatic, did not seroconvert, and were susceptible to IV infection. Six of 9 ponies inoculated with E. risticii via nasogastric intubation and oral drench developed Potomac horse fever and serocon verted. The other 3 remained asymptomatic and did not seroconvert. One of these latter ponies and 2 normal ponies that were inoculated via oral drench only developed Potomac horse fever and seroconverted. The high fever, maximum clinical score for decreased feed intake, depressed mental attitude, decreased borborygmal sounds, severity of diarrhea, and the length of illness of the orally infected ponies was not significantly different from those of IV infected control ponies, although the signs occurred significantly later ( P < .05).  相似文献

During 2007, large outbreaks of equine influenza (EI) caused by Florida sublineage Clade 1 viruses affected horse populations in Japan and Australia. The likely protection that would be provided by two modern vaccines commercially available in the European Union (an ISCOM-based and a canarypox-based vaccine) at the time of the outbreaks was determined. Vaccinated ponies were challenged with a representative outbreak isolate (A/eq/Sydney/2888-8/07) and levels of protection were compared. A group of ponies infected 18 months previously with a phylogenetically-related isolate from 2003 (A/eq/South Africa/4/03) was also challenged with the 2007 outbreak virus. After experimental infection with A/eq/Sydney/2888-8/07, unvaccinated control ponies all showed clinical signs of infection together with virus shedding. Protection achieved by both vaccination or long-term immunity induced by previous exposure to equine influenza virus (EIV) was characterised by minor signs of disease and reduced virus shedding when compared with unvaccinated control ponies. The three different methods of virus titration in embryonated hens’ eggs, EIV NP-ELISA and quantitative RT-PCR were used to monitor EIV shedding and results were compared. Though the majority of previously infected ponies had low antibody levels at the time of challenge, they demonstrated good clinical protection and limited virus shedding. In summary, we demonstrate that vaccination with current EIV vaccines would partially protect against infection with A/eq/Sydney/2888-8/07-like strains and would help to limit the spread of disease in our vaccinated horse population.  相似文献

Biting flies were collected from the umbilical areas of Onchocerca lienalis-infected cattle in New York (state) from June through September of 1980. Of the 766 flies collected, 705 were Simulium jenningsi. Microfilariae were detected in the midguts of 37 (50%) of 73 females dissected immediately after the flies had fed. The mean number of larvae per positive fly (fly with microfilariae) was 15.2. Third-stage larvae were recovered from 25 (21.9%) of 114 S jenningsi dissected 8 to 13 days after they had fed on the infected cattle; the mean number of 3rd-stage larvae per positive fly was 3.5. Dissections of flies performed on days 1 through 7 after feeding yielded various numbers of 1st and 2nd-stage larvae from the thoracic muscles. Ovarian dissections performed on 304 S jenningsi attacking cattle indicated an overall parous rate of 58%. Naturally occurring infections with filarial larvae indistinguishable from O lienalis were found in 7.3% of the parous females. Three of these flies, or 1.7% of the parous collection, harbored 3rd-stage larvae. The onset of naturally occurring filarial infections in the population of S jenningsi coincided with a peak in the parous rate in late June. Thereafter, filarial infections were generally detected when the parous rate was above 50%.  相似文献

A tissue culture of Culex pipiens quinquefasciatus Say ovarian cells appeared to support the growth of equine infectious anemia (EIA) virus. Shetland ponies inoculated with 2nd, 7th, 9th, and 11th passages of mediums harvested from infected tissue culture had clinical signs of the disease and became EIA positive on 11, 19, 23, and 43 days after inoculation, respectively.  相似文献

The clinical and immunological response of ponies exposed to a bovine herpesvirus isolate and equine herpesvirus 1 were compared. Each virus was inoculated into two ponies by the intranasal route. One uninoculated pony was used with each group as a contact control. The four inoculated ponies developed a mild rhinitis with an increase in rectal temperature. Virus was recovered from nasal secretions collected from the four inoculated and one contact pony. All ponies developed a serum neutralizing antibody to each virus. The data show that the two viruses are similar.  相似文献

Neurologic disease occurs sporadically in horses infected with the equine infectious anemia virus (EIAV). This report describes a case of clinically severe neurologic disease in a pony experimentally infected with EIAV. This pony did not have fever or anemia, which are the characteristic clinical signs of disease. The histopathologic changes were characterized as lymphohistiocytic periventricular leukoencephalitis. Polymerase chain reaction and in situ hybridization data showed that the brain lesions were directly associated with viral replication and that high-level viral replication occurred selectively within the lesion and not in other tissues. These findings suggest that EIAV-associated neurologic disease is the direct result of viral replication.  相似文献
