为实现亚麻籽饼粕中分离蛋白的综合加工利用,该文以冷榨亚麻籽饼粕为原料,对其进行脱胶脱脂处理,采用超声辅助水提法分别提取亚麻籽分离蛋白和脱胶脱脂亚麻籽分离蛋白,并对理化性质、结构及功能特性进行分析比较。结果表明,冷榨亚麻籽饼粕和脱胶脱脂亚麻籽饼粕中蛋白质质量分数分别为37.52%±0.04%、37.47%±0.02%。采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳测定亚麻籽分离蛋白和脱胶脱脂亚麻籽分离蛋白的分子质量,显示10~55 kDa之间有明显谱带,其中水溶性低分子质量的白蛋白(10、14、15及17 kDa)和盐溶性高分子质量的球蛋白(30、33、35、40、45及55 kDa)谱带最为明显。氨基酸的种类各检测到17种,含有丰富的必需氨基酸和非必需氨基酸。傅里叶变换红外光谱测定的2种蛋白质的二级结构稳定性一般;由扫描电镜和X-射线衍射可知亚麻籽分离蛋白比脱胶脱脂亚麻籽分离蛋白的微观孔隙率低,结构中都较缺乏结晶度或有序排列。2种蛋白的两亲性与大豆分离蛋白相比,亲水/油特性突出,亲油性是大豆分离蛋白的2倍多。通过对不同pH(2~11)和盐离子浓度(0~1.25 mol/L)下溶解度、起泡性、泡沫稳定性、乳化活性及乳化稳定性的测定,显示两者具有良好的碱溶性,脱胶脱脂亚麻籽分离蛋白的起泡性、乳化活性及乳化稳定性均优于亚麻籽分离蛋白,而泡沫稳定性恰好相反,该研究结果有利于拓宽2种分离蛋白在健康食品领域中的应用前景,为食品中的应用提供有益参考和数据支撑。  相似文献

为探明凤丹牡丹花粉、花药壁的营养成分,从而更好地发挥其营养价值,本研究采用凯氏定氮法、电感耦合等离子体发射光谱、高效液相色谱、氨基酸分析仪等对凤丹花粉和花药壁蛋白质、矿物质元素、维生素C、水解氨基酸及游离氨基酸进行了系统地分析和评价。结果表明,凤丹花粉中蛋白质含量高达40.68%(初开期),水解氨基酸总量为355.65 mg·g-1(初开期),花药壁中蛋白质和水解氨基酸的总量虽仅为花粉的一半,但二者必需氨基酸与总氨基酸的比值均为40%左右,属优质蛋白来源。36种游离氨基酸检测结果显示,花药壁中总游离氨基酸含量稍高,但花粉中必需氨基酸、蛋白氨基酸及鲜甜氨基酸含量更高。矿物质元素分析结果表明,凤丹花粉、花药壁含有丰富的常量和微量元素,尤以K、Ca、Mg、Fe、Zn等元素含量较高,是开发高钾低钠和补铁补锌的佳品。Vc测定结果显示,凤丹花粉和花药壁中Vc含量较高,可作为Vc补充剂。同时考察了凤丹花粉、花药壁不同采收时期及破壁对检测成分含量的影响,结果显示二者更适合在初开期采收,且破壁对花粉、花药壁营养成分检测无显著影响。综上,凤丹花粉、花药壁含有丰富的营养成分,本研究将为...  相似文献

Although extracellular proteins may play an important role in the soil environment, these proteins are difficult to isolate because they are immediately degraded by soil microbes, or become associated with clay mineral and humic substances. We developed a method of isolating extracellular proteins from greenhouse soils. Phosphate buffer (pH 6.0) was used to extract protein from soil. A phosphate buffer with higher pH was not recommended because it extracted a large amount of non-proteinaceous organic matter as well as protein and, as a result, the extracted protein was difficult to separate by sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE). After removing cells by filtration, proteins dissolved in the soil extract were recovered by precipitation with 5% trichloroacetic acid (TCA) and isolated by SDS-PAGE. Proteins were detected in 10 of 32 soil samples derived from different greenhouses and the protein bands ranged in apparent molecular mass from 35 to 68 kDa, suggesting that some of soils derived from greenhouse culture contained significant amounts of a specific protein soluble in 67 mM phosphate buffer (pH 6.0). N-terminal amino acid sequence of one of the isolated proteins was found to be a homologue of thermostable cellulase produced by the genus Humicola, a thermophilic fungus.  相似文献

The 85% methanol-soluble proteins are known to specifically contribute to the production of flavor of roasted peanut. To determine the nature of the 85% methanol-soluble proteins, they were isolated from the peanut seed, and the 85% methanol-soluble (MS) and 85% methanol-insoluble (MIS) fractions were characterized using polyacrylamide gel electrophoresis (PAGE) and capillary electrophoresis. The results showed that the 85% MS fraction contained lower amounts (9-10%) of protein than the MIS fraction (15-33%). Protein content of the MIS fraction increased more significantly during seed maturation than it did in the MS fraction. Unlike the protein, free amino acids and soluble sugars levels of the MS fraction decreased significantly during seed maturation. The 85% MS fraction contained predominantly low molecular weight (<20 kDa) proteins/polypeptides, whereas the MIS fraction contained a mixture of polypeptides with molecular weight between 14 kDa and 90 kDa. SDS-PAGE showed no major changes in the polypeptide composition of the MS fraction during seed maturation. Capillary electrophoretic analysis revealed major qualitative and quantitative changes in the protein and polypeptide composition of the MS and MIS fractions during seed maturation. Fatty acid analysis of these fractions indicated that the MS fraction is lipoprotein in nature and rich in oleic and linoleic acids.  相似文献

Comparison of different components released during autolysis with fermented or active dried yeast, which has never been reported previously, is related in this paper. Three autolysates were elaborated with Saccharomyces cerevisiae in a model wine (pH 3.5) at 30 or 18 degrees C. Composition of the autolysate appears to depend on both the growth medium and the physiological state of the yeast. The autolysate obtained from active dried yeast presents a higher total nitrogen concentration (a factor of 1.5-1.9 for the fermented yeast autolysate), a greater proportion of free amino acids (42 vs 16-25%), the lowest proportion of oligopeptides (25 vs 31-37%) and polypeptides (27 vs 45-34%), and an absence of nucleosides. Distribution of amino acids in peptides and proteins is relatively homogeneous in the autolysates but shows significant differences in free amino acids. Parietal macromolecules (mannoproteins and glucans) are present in greater quantities in autolysates elaborated from fermented yeast. The influence of the different fractions of these autolysates (<0.5, 0.5-1, 1-10, and >10 kDa) on bacterial growth has been investigated.  相似文献

The aim of this study was to analyze sulfur content, protein size distribution, and free amino acids in flour mill streams (FMS) and their associations to dough rheology and breadmaking traits. Break FMS had higher nitrogen and sulfur quantities than reduction FMS. The third break FMS had the highest nitrogen and sulfur contents among FMS but low bread loaf volume partly due to high ash content. Sulfur quantity had greater or equivalent correlations with dough rheology and breadmaking properties compared with nitrogen quantity when the effect of percent ash content was removed statistically. FMS also showed significant quantitative variation in HMW polymeric proteins of the SDS‐unextractable fraction that had greater association with sulfur content and dough rheology and breadmaking traits than other protein fractions. Asparagine, which is a major amino acid in flour, was found at higher levels in the third break and third reduction FMS. Ratio of nitrogen to sulfur was significantly correlated with asparagine concentration (r = 0.73, α = 0.01). This study indicates that information on sulfur, protein size distribution, and free amino acid is potentially useful in research for more precise blending of FMS in commercial flour mills to meet customer specifications for high quality flour.  相似文献

Data reported here show that Ganoderma lucidum could biotransform inorganic selenite in the substrate into organic forms by intergrating Se into proteins (56-61%) and polysaccharides (11-18%) and other components. Furthermore, water- and alkaline-soluble protein components were mainly responsible for the storage of organic Se, and Se-Met accounts for only a minor (8.2-18.3%) amount of the selenocompounds present in proteins. The molecular mass of most proteins or protein subunits containing Se was no more than 16 kDa. A low concentration of Se (<100 microg/g) in the substrate facilitated the synthesis of total protein and amino acids in G. lucidum, but a high concentration of Se (>150 microg/g) played a reverse role. Additionally, Se concentration in the culture had no significant effect on the distribution of the amino acids and proteins.  相似文献

Food allergies represent an important health problem in industrialized countries, such that detection and quantitative analysis of the protein considered to be the main allergen is crucial. A dot-blot fluorescent staining method for the detection and quantitative analysis of protein residues in food grade amino acids and nucleic acids is presented. This method combines fluorescence staining with dot-blotting onto PVDF membrane. Several standard proteins, such as bovine serum albumin (66 kDa), lysozyme (14 kDa), ubiquitin (8.6 kDa), bovine insulin (5.7 kDa), and oxidized insulin B chain (3.5 kDa), were detectable at 0.1 ppm using SYPRO Ruby blot stain. Twenty-five different amino acids and two different nucleic acids of food grade were analyzed using this method combined with an internal standard addition method using BSA as an internal standard. All amino acids and nucleic acids were dissolved in 3.6% aqueous HCl and dot-blotted onto PVDF membrane before a large amount of amino acids and nucleic acid were removed. Protein residues and the internal standard protein immobilized on the membrane were stained using SYPRO ruby blot stain. The internal standard in all samples was detectable at 0.1 ppm. Samples were dissolved at 120 or 70 mg/mL, according to their solubility under acidic conditions. The detection limit of protein residues per weight was 0.8-1.4 ppm in amino acids and nucleic acids; residual protein was not detected in any sample.  相似文献

采用国际上通用的非生物学评价方法,研究了不同供锌水平对猪肚菇子实体蛋白质营养价值的影响,以期为猪肚菇栽培技术的改善和完善提供科学依据。结果表明,氨基酸总量以40mg·kg^-1处理的最高,其与对照差异达到显著(P〈0.05),但与其他处理差异不显著(P〉0.05);而Zn含量以10mg·kg^-1处理最高。营养综合评价结果显示,以浓度为30mg·kg^-1Zn处理的培养料栽培的猪肚菇子实体蛋白质营养价值评价的6项指标中氨基酸评分、必需氨基酸指数、氨基酸比值系数分和生物价分别为95.33、91.70、82.52和88.50,均居6种Zn处理之首,化学评分和营养指数分别为73.22和25.49,均居第3位。综合评价的结果显示,以浓度为30mg·kg^-1Zn处理的蛋白质营养价值最高。  相似文献

Total protein content and level of 14 amino acid in mature acorns of 20 Quercus taxa from Turkey were studied. The range of total protein amounts between 2.75 and 8.44% were detected among taxa. Similar values in related species and variety level were observed. The amino acid profiles for each taxon were characteristically different and high variability of individual amino acid concentration was present at variety, subspecies and species levels. Different amino acid concentrations, relative percentages and critical values of some amino acids are thought to be additional considerable parameters for diagnosis of Quercus. Generally higher total quantity of amino acids in section Quercus and lower values in section Ilex were observed. When the essential and non-essential amino acids are expressed as a relative percent of total protein indicating quality of proteins, the ratios varied significantly among taxa (p < 0.05). But, no significant difference at section level were detected. Major amino acids were aspartic acid and glutamic acid showing the largest variations and the lowest levels were detected for methionine. Amino acid concentrations ranged from 1665 for aspartic acid to 13 mg/100 g dry wt. for methionine. All taxa had relatively higher amounts of leucin, lysin and valine than other essential amino acids. The highest quantity of protein and amino acids was recorded for Q. infectoria ssp. boissieri and the lowest for Q. pontica. The level of all essential amino acids in examined taxa was not sufficient compared to FAO scoring pattern for children (1985). But, threonine and valine in Q. infectoria ssp. boissieri and isoleucine in Q. petraea ssp. iberica show remarkable concentrations to the requirements. All examined essential amino acids among taxa generally provide adequate levels for adults according to FAO standard.  相似文献

探讨了锌对大蒜生理生化指标及营养品质的影响。结果表明,当锌处理水平为Zn4(0 3gZnSO4·7H2O/15kg土)时,大蒜叶片硝酸还原酶、过氧化氢酶(CAT)、过氧化物酶(POD)、超氧化物歧化酶(SOD)活性、可溶性蛋白质含量、光合色素含量及光合参数均达最高。Zn3(0 2gZnSO4·7H2O/15kg土)处理水平极有利于大蒜素和蒜薹中Vc的形成。而当处理水平为Zn2(0 1gZnSO4·7H2O/15kg土)时,鳞茎中游离氨基酸含量高出对照11%,蒜薹中其含量却在Zn3处理水平时比对照增加33%;而锌处理水平为Zn4时,有利于提高鳞茎中可溶性糖和可溶性蛋白质含量,分别提高6%和5%;蒜薹中可溶性蛋白质含量要求浓度与游离氨基酸相同。  相似文献

Ara h 1 was purified from raw peanuts (Arachis hypogaea L.) in the presence or absence of protease inhibitors. N-Terminal amino acid sequences were determined after western blotting. Both purification procedures proved to be very consistent and resulted in identical chromatographic and electrophoretic behavior of Ara h 1 and in the isolation of identical proteins of approximately 64 kDa with RS/H_PPGERTRG as the N-terminal amino acid sequence. Consequently, purified Ara h 1 appears to be truncated at the N-terminal side. The observations strongly suggest that Ara h 1 occurs physiologically as a protein of which the first 84 and 78 amino acids, respectively, are cleaved off in planta upon maturation of the protein. On the basis of epitope mapping, the cleaved-off N-terminal peptide contains three allergenic epitopes, of which two are major. These truncated epitopes will go undetected in assays when purified Ara h 1 from peanuts is used as reference material. Patients' sera, however, contain IgE-type antibodies against the epitopes that are contained in the cleaved-off peptide, implying that the peptide, or part of it, is still present in peanuts that are consumed. Possible consequences of this exposure to these three epitopes are discussed. On the basis of literature data the cleaved-off peptide is hypothesized to have antifungal activity.  相似文献

通过溶液培养试验,比较研究了外源有机酸对两种生态型东南景天生长和锌吸收与积累的影响。结果表明,外加柠檬酸或草酸能明显增强非超积累生态型东南景天的耐锌毒能力,叶片、茎和根系的生物量均有增加;而对锌超积累生态型东南景天的生长没有显著性影响。外加柠檬酸、草酸或柠檬酸+草酸,锌超积累生态型根系Zn含量显著增加,但叶片和茎组织中Zn含量无明显变化;而非超积累生态型叶片、茎和根系Zn含量均显著增加,尤其是根系Zn含量,与单独Zn处理相比外源有机酸处理增加4~5倍。同样,外加柠檬酸、草酸或柠檬酸+草酸明显提高Zn在非超积累生态型根系和地上部的积累量,且对地上部Zn积累量的促进程度大于根部;柠檬酸的促进作用又比草酸及2种酸混合处理高1倍。然而,有机酸处理对Zn超积累生态型根系Zn积累量有所增加,但对叶片和茎中Zn积累量无影响。叶片和茎中水溶态Zn含量也受外源有机酸的影响,2种生态型叶片、茎中水溶态Zn含量均受有机酸处理而提高。Zn超积累生态型叶片和茎中水溶态Zn含量占总Zn含量比例也受有机酸处理而增大,但非超积累生态型则显著降低。上述结果表明,柠檬酸和草酸可能作为其Zn吸收的配基,促进根系对Zn吸收与积累,叶片和茎中的Zn可能与可溶性有机化合物或无机盐结合。  相似文献

As a result of methionine deficiency, legume proteins are considered to be incomplete, and therefore there is a need to explore ways to improve legume protein amino acid balance. Using rabbit anti-soybean sulfur-rich protein (SRP) polyclonal antibodies (pAb), sensitive immunoassays (nanogram sensitivity) were developed. The immunoassays detected SRP in all soybean seeds and soybean-based commercial samples examined. In addition, the presence of pAb cross-reactive proteins was detected in certain dry beans and oilseeds. The cross-reactive proteins were isolated using purified IgG-based immunoaffinity column chromatography. Biochemical analyses including N-terminal amino acid sequencing and amino acid composition indicated that the cross-reactive proteins were comparable to soybean SRP. The cross-reactive proteins contained methionine (1.6-2.4 residues/100 residues) and cysteine (2.4-3.6 residues/100 residues), which satisfies the FAO/WHO recommended pattern for sulfur amino acids in both adults and children (2-5 years old). The results suggest the presence of constitutive SRPs in several dry beans and oilseeds.  相似文献

Pecans (cv. Desirable) contained approximately 10% protein on a dry weight basis. The minimum nitrogen solubility (5.9-7.5%) at 0.25-0.75 M trichloroacetic acid represented the nonprotein nitrogen. Among the solvents assessed for protein solubilization, 0.1 M NaOH was the most effective, while borate saline buffer (pH 8.45) was judged to be optimal for protein solubilization. The protein solubility was minimal in the pH range of 3-7 and significantly increased on either side of this pH range. Increasing the NaCl concentration from 0 to 4 M significantly improved ( approximately 8-fold increase) protein solubilization. Following Osborne protein fractionation, the alkali-soluble glutelin fraction (60.1%) accounted for a major portion of pecan proteins followed by globulin (31.5%), prolamin (3.4%), and albumin (1.5%), respectively. The majority of pecan polypeptides were in the molecular mass range of 12-66 kDa and in the pI range of 4.0-8.3. The pecan globulin fraction was characterized by the presence of several glycoprotein polypeptides. Lysine was the first limiting essential amino acid in the defatted flour, globulin, prolamin, and alkaline glutelin fractions. Leucine and tryptophan were the first limiting essential amino acids in albumin and acid glutelin fractions, respectively. Rabbit polyclonal antibodies detected a range of pecan polypeptides in the 12-60 kDa range, of which the globulin fraction contained the most reactive polypeptides.  相似文献

Legumes, including soybeans ( Glycine max ), are deficient in sulfur-containing amino acids, which are required for the optimal growth of monogastric animals. This deficiency can be overcome by expressing heterologous proteins rich in sulfur-containing amino acids in soybean seeds. A maize 27 kDa γ-zein, a cysteine-rich protein, has been successfully expressed in several crops including soybean, barley, and alfalfa with the intent to biofortify these crops for animal feed. Previous work has shown that the maize 27 kDa zein can withstand digestion by pepsin and elicit an immunogenic response in young pigs. By use of sera from patients who tested positive by ImmunoCAP assay for elevated IgE to maize proteins, specific IgE binding to the 27 kDa γ-zein is demonstrated. Bioinformatic analysis using the full-length and 80 amino acid sliding window FASTA searches identified significant sequence homology of the 27 kDa γ-zein with several known allergens. Immunoblot analysis using human serum that cross-reacts with maize seed proteins also revealed specific IgE-binding to the 27 kDa γ-zein in soybean seed protein extracts containing the 27 kDa zein. This study demonstrates for the first time the allergenicity potential of the 27 kDa γ-zein and the potential that this protein has to limit livestock performance when used in soybeans that serve as a biofortified feed supplement.  相似文献

The aim of this work was to study the effect of arbuscular mycorrhizal fungus Glomus mosseae on growth and nitrogen (N) metabolism of durum wheat (Tritcum durum) under various P soil contents. The analyses were extended to macro and micronutrient tissue concentrations, nitrate reductase and glutamine synthetase activities, as well as protein, aminoacids, pyridine dinucleotides and adenine nucleotides. Arbuscular mycorrhiza increased wheat growth in soil in which P availability was low and nitrate was the dominant N form. The root colonization occurred at the highest level in plants grown in limiting soil P and was inversely related to soil P content. The micorrhizal wheat plants contained also the highest concentrations of macro (P, K, Ca, N) and micronutrients (Fe, Zn, Mn) as well as free amino acids, protein, NAD, NADP, AMP, ADP, ATP in roots and leaves. In particular, the micronutrient tissue concentrations (Zn, Mn) supported that mycorrhiza actively modulated their uptake limiting interferences and optimizing growth better than the plant roots, like a very efficient “rootstock”. Control plants grown at the highest soil P did not reach the same concentration as the mycorrhizal plants. Nitrate reductase activities in the roots of mycorrhizal plants were higher than in the control ones, while glutamine synthetase activities were highest in the leaves. Protein and amino acids concentrations, as well as AMP, ADP, ATP, NAD(P), and NAD(P)H were also higher than in the control. Among the free amino acids in the roots, the high levels of glutamine, asparagine, arginine, support the view that ammonium was transferred through the arbuscules to the root cells where it was re‐assimilated in the cortical cells, forming high N : C ratio‐amino acids. They were transferred to the leaves where all the other N compounds could be largely synthesized using the carbon skeletons supplied by photosynthesis.  相似文献

The squid (Loligo pealei) byproduct composed of heads, viscera, skin, fins, and small tubes was subjected to hydrolysis at 55 degrees C and natural pH (6.8) using endogenous proteases. Squid hydrolysate was characterized during the course of hydrolysis for changes in the degree of hydrolysis, viscosity, electrophoretic pattern of proteins and peptides, and amino acid and fatty acid profiles. The change in viscosity can be used to monitor the progress of protein hydrolysis up to the molecular mass of 26.63 kDa. The 2 h hydrolysis resulted in a 2-fold increase in the total free amino acids and yielded hydrolysate with protein molecular mass of < or =45 kDa having feed attractability and good amino acid and fatty acid profiles with high contents of essential amino acids and fatty acids. Such hydrolysis-induced changes can make squid byproduct hydrolysate a good source of aquaculture feed ingredient, especially for a starter diet for larval fish.  相似文献

Characterization of yam bean (Pachyrhizus erosus) proteins   总被引:1,自引:0,他引:1  
Seed proteins from Mexican yam bean seeds (Pachyrhizus erosus L.) were sequentially extracted according to the Osborne classification. Albumins were the major fraction (52.1-31.0%), followed by globulins (30.7-27.5%). The minor protein fraction was prolamins (0.8%). Defatting with chloroform/methanol remarkably affected the distribution of protein solubility classes; albumins were the most affected fraction (4.3-17.5%). Electrophoretic patterns of albumins showed bands at 55, 40, 35, and 31 kDa. After reduction of the globulin fraction exhibited two triplets, one from 35 to 31 kDa and the second from 19 to 21 kDa, these could be compared to the acid and basic polypeptides of 11S-like proteins. Prolamins showed one band at 31 kDa, and glutelins after reduction showed three main bands at 52, 27, and 14 kDa. Trypsin inhibitors were assayed in saline extracts; the values found (1232-2608 IU/g of meal) were lower than those of other legumes. In general, yam bean seed proteins showed an excellent balance of all essential amino acids; albumins contain the highest amount of essential amino acids.  相似文献

采用HEDTA螯合剂缓冲营养液培养法,选用籽粒含锌量有明显差异的2个基因型水稻(BY和Z921),设置4种锌离子活度(pZn2+9.7、10.3、11.0、11.4),研究了锌离子活度对水稻锌积累、分配的影响以及对不同时期水稻叶片中锌的化学形态的影响。结果显示:(1)2个基因型水稻各器官的锌含量都随着锌离子活度的升高而升高,但不同基因型间,同一基因型不同器官间均存在差异,供锌正常的的条件下,锌首先向代谢活性较弱的营养器官分配;缺锌的条件下,锌首先满足籽粒的需要;(2)从籽粒锌分配看,当锌离子活度(pZn2+)小于10.3时,糙米锌含量最高,当pZn2+升高到9.7时,颖壳锌含量则超过糙米,糙米和精米锌含量的比值在0.79~0.90之间,并以pZn2+为9.7时为最小;(3)任一锌离子活度下,BY籽粒锌含量均大于Z921。表明通过筛选籽粒富锌水稻品种来提高稻米锌含量是经济可行的,且通过增加环境锌离子活度来改善水稻的锌营养能显著提高水稻籽粒的锌含量;(4)营养生长前期,水稻叶片中的锌主要以活性较低的醋酸提取态(重金属磷酸盐)存在;营养生长后期,锌主要以乙醇提取态(醇溶性蛋白、氨基酸等)存在。  相似文献
