The aim of this study was to evaluate the efficacy of lyophilised C-strain vaccine in domestic pigs and wild boar after oral application. A new spherical bait form (diameter 3 cm) containing lyophilised vaccine virus and the recent vaccine baits were used for animal experiments. Four vaccination groups were established in experiment 1 (group 1: recent liquid bait vaccine; group 2: spherical baits containing one dose of the lyophilised vaccine; groups 3 (domestic pigs) and 4 (wild boar): spherical baits containing two doses of the lyophilised vaccine) and two groups in experiment 2 (group 1: recent liquid bait vaccine; group 2: spherical baits with two doses of the lyophilised vaccine). Challenge was carried out with the highly virulent virus strain "Alfort 187" (using 100 TCID50 in the first and 1.000 TCID50 in the second experiment). Our results showed that the animals vaccinated with lyophilised C-strain vaccine developed high neutralising antibody titres comparable to those obtained after vaccination with the recent bait vaccine. All pigs which picked up the baits remained healthy after challenge. Neither clinical symptoms nor viremia or virus shedding were observed after infection except in one pig (group 2, experiment 2) which had not consumed the vaccine bait. The surviving domestic pigs and wild boar were tested negative for CSFV and viral RNA at the end of the study. This result demonstrates that lyophilised vaccine may become an effective vaccine formulation for oral immunisation of wild boar against CSF in the near future.  相似文献

The aim of the studies was to fathom the duration and the role of maternal immunity for Aujeszky's disease (AD) and classical swine fever (CSF) in wild boar offspring. In one experiment, two wild boar sows were infected with a low pathogenic pseudorabies virus (PRV) in 1999. A total of 51 offspring was born between 1999 and 2002 and was monitored for PRV maternal antibodies. In a second experiment, the maternal immunity for CSF was analysed. Therefore, a sow was orally vaccinated against CSF using vaccine baits containing the live‐attenuated C‐strain vaccine. The vaccination took place in January 1999. The sow gave birth to four piglets in 2001 and to two piglets in 2002. With respect to maternal immunity for AD, some piglets reacted positive in the ELISA up to 27‐week post‐partum while in the neutralization test antibodies were detected up to 15‐week post‐partum. The calculated half‐life of neutralizing antibodies was 21 days. Regarding CSF, the neutralization titres of maternal antibodies dropped continuously reaching values of ≤10 ND50 20‐week post‐partum. After the 12th week post‐partum, most of the sera reacted negative in the ELISA. However, after the third month, low levels of neutralization titres were still detectable. The results are discussed with respect to the epidemiology and control of both diseases in wild boar populations.  相似文献

Tuberculosis (TB) caused by Mycobacterium bovis, a member of the Mycobacterium tuberculosis complex, is an important health problem worldwide. The control of TB through vaccination of wildlife reservoirs may potentially have advantages over other management strategies. The most practical approach to deliver vaccines to wildlife is using oral baits that are stable under field conditions and effective in reaching the target species. Baits were developed in our laboratory to deliver oral vaccines to wild boar piglets. However, these baits were well accepted by other wild species. Therefore, bait consumption by different M. bovis hosts was evaluated herein. The results showed that the baits were well accepted by cattle, feral pigs, and adult red deer whereas small mammals like badgers and possums showed varying bait acceptance. Bait acceptance by different species has the advantage of targeting more than one wildlife reservoir when they coexist in the same area and need to be vaccinated for TB control. However, bait delivery methods such as the use of selective feeders to target the desired species should be developed to avoid bait consumption by other species.  相似文献

Despite the compulsory test and slaughter campaigns in cattle, bovine tuberculosis (bTB) is still present in Spain, and the role of wildlife reservoirs is increasingly recognized. We provide an update on recent progress made in bTB control in Spanish wildlife, including aspects of epidemiology, surveillance, host-pathogen interaction and wildlife vaccination. At the high densities and in the particular circumstances of Mediterranean environments, wild ungulates, mainly Eurasian wild boar and red deer, are able to maintain Mycobacterium bovis circulation even in absence of domestic livestock. Infection is widespread among wild ungulates in the south of the country, local infection prevalence being as high as 52% in wild boar and 27% in red deer. Risk factors identified include host genetic susceptibility, abundance, spatial aggregation at feeders and waterholes, scavenging, and social behaviour. An increasing trend of bTB compatible lesions was reported among wild boar and red deer inspected between 1992 and 2004 in Southwestern Spain. Sporadic cases of badger TB have been detected, further complicating the picture. Gene expression profiles were characterized in European wild boar and Iberian red deer naturally infected with M. bovis. The comparative analysis of gene expression profiles in wildlife hosts in response to infection advanced our understanding of the molecular mechanisms of infection and pathogenesis, revealed common and distinctive host responses to infection and identified candidate genes associated with resistance to bTB and for the characterization of host response to infection and vaccination. Ongoing research is producing valuable knowledge on vaccine delivery, safety and efficacy issues. Baits for the oral delivery of BCG vaccine preparations to wild boar piglets were developed and evaluated. The use of selective feeders during the summer was found to be a potentially reliable bait-deployment strategy. Safety experiments yielded no isolation of M. bovis BCG from faeces, internal organs at necropsy and the environment, even after oral delivery of very high doses. Finally, preliminary vaccination and challenge experiments suggested that a single oral BCG vaccination may protect wild boar from infection by a virulent M. bovis field strain.  相似文献

To determine the persistence period of C-strain vaccine virus in immunized animals, domestic pigs and wild boars were vaccinated orally and killed on different days post vaccinationem (dpv). Tissue samples were taken at necropsy from both species for detection of C-strain virus. From domestic pigs nasal swabs and faeces were also collected. During the investigation period (2-12 dpv) vaccine virus could never be detected in nasal secretions and in faeces of vaccinated domestic pigs. In contrast, C-strain virus was found in organs until day 8 pv in domestic pigs and until day 9 pv in wild boars. Whereas in domestic pigs virus was detected in tonsils, Ln. mandibularis or in spleen, in wild boar it only was found in tonsils. We conclude that C-strain vaccine virus is not detectable in wild boars longer than 10-12 days after intake of the vaccine baits.  相似文献

Two recombinant Mycobacterium bovis BCG (rBCG) strains carrying the Eimeria tenella rhomboid gene (Rho) delivered by extrachromosomal vector pMV261 and integrative vector pMV361 were evaluated for their ability to protect chickens against E. tenella challenge. The chickens were immunized intranasal with BCG, rBCG pMV261-Rho, or rBCG pMV361-Rho twice at a 2-week interval. All the recombinant BCG immunized chickens developed specific immune responses, and there was a significant increases of the percentages of CD4+ and CD8+ cells compared to the control (P < 0.05). Challenge experiments demonstrated that the two rBCG strains could provide significant protection against E. tenella challenge. But vaccination with rBCG pMV261-Rho induced higher specific antibody titers and produced greater protection rate (56.04%) than rBCG pMV361-Rho group (P < 0.05). These results indicated that M. bovis BCG is a novel vaccine vector to express and present antigens of E. tenella, and rBCG has a potential as vaccine in chickens.  相似文献

The use of transgenic plants as delivery system for antigenic proteins is attractive for its simplicity and increases likelihood for local immune response at sites of infection. The aim of this study was to evaluate the protective effect of oral administration of tobacco seeds, expressing the FedA, the major protein of the F18 adhesive fimbriae, and B subunit of verocytotoxin, against verocytotoxin-producing E. coli (VTEC) strain in piglets. Forty-three early weaned piglets, were randomly divided into 4 experimental groups: 3 test groups and a control. Treatment groups orally received a bolus, with different dose of tobacco seeds on 0, 1, 2, 14 days post primary administration. After challenge, with 1*1010 CFU of O138 Escherichia coli strain, piglets showed clinical scores significantly higher in the control group compared to orally immunized groups (P?<?0.05) and the latter showed a faster recovery than in CG. In conclusion, oral administration of recombinant tobacco seeds expressing antigenic proteins against VTEC strains can induce a protective effect against challenger strain in piglets.  相似文献

Porcine edema disease (ED) is a toxemia that is caused by enteric infection with Shiga toxin 2e (Stx2e)‐producing Escherichia coli (STEC) and is associated with high mortality. Since ED occurs most frequently during the weaning period, preweaning vaccination of newborn piglets is required. We developed stx2eB‐transgenic lettuce as an oral vaccine candidate against ED and examined its protective efficacy using a piglet STEC infection model. Two serially developed Stx2eB‐lettuce strains, 2BN containing ingredient Stx2eB constituting a concentration level of 0.53 mg Stx2eB/g of powdered lettuce dry weight (DW) and 2BH containing ingredient Stx2eB constituting a concentration level of 2.3 mg of Stx2eB/g of powdered lettuce DW, were evaluated in three sequential experiments. Taken the results together, oral administration of Stx2eB‐lettuce vaccine was suggested to relieve the pathogenic symptoms of ED in piglets challenged with virulent STEC strain. Our data suggested that Stx2eB‐lettuce is a promising first oral vaccine candidate against ED.  相似文献

The aim of the study was to determine the apparent consumption of dry matter (DM), gross energy (GE), crude protein (CP) and amino acids (AA) from pasture by European wild boar in a pastoral system. Two pasture-types were used, one consisting predominantly of Lolium perenne L. and the other predominantly of Plantago lanceolata L. The study was conducted in Spring and repeated in Summer. Twelve purebred European Wild Boar of 18.8 ± 0.8 kg (mean ± sem) with nose rings were randomly grouped into six pairs. Each day of the 19-day study, a pair of animals was placed into each of three areas of a pasture strip (1.4 × 6.3 m per area) from each pasture type from 8:30 h until 16:30 h, after which the animals entered a barn and had free access to a commercial diet for 45 min, with each pasture strip being grazed once. Pasture samples were taken on days 4 to 19 from each grazed area pre- and post-grazing and the DM content of these samples was used to calculate DM consumption of the animals. Additional pasture samples were collected and analysed for gross energy, crude protein and amino acids. The wild boar consumed (mean + SEM) 418 ± 72.2 and 210 ± 38.3 g of DM per day in the L. perenne paddock during Spring and Summer, respectively, and 550 ± 85.9 and 226 ± 44.8 g DM per day in the P. lanceolata paddock during Spring and Summer, respectively. The amount of DM, energy, crude protein and amino acids that the animals consumed varied markedly between days, but did not significantly differ in amount between the L. perenne and P. laceolata paddocks. However, the consumptions were significantly lower in Summer than in Spring. It is estimated that the wild boar would have satisfied somewhat less than 90 and 45% of their daily maintenance digestible energy requirements through consumption of pasture when grazing the L. perenne paddock in Spring and Summer, respectively.  相似文献

为研究猪霍乱沙门菌C500(S.C500)运载猪瘟病毒(CSFV)新型基因疫苗口服免疫家兔的体内免疫应答特点,采用电转化法将CSFV新型基因疫苗转化到S.C500,构建重组工程菌,口服免疫接种家兔,检测CSFV、S.C500特异性抗体;并用猪瘟兔化弱毒疫苗及猪伤寒沙门菌野毒株依次进行攻毒试验。结果显示,成功构建CSFV新型基因疫苗重组菌S.C500/pCB-ME2-IL-15,S.C500/pCC-ME2-IL-15。口服免疫家兔可以诱导产生抗CSFV和S.C500的特异性ELISA抗体,且S.C500/pCC-ME2-IL-15略显优势。经三免后免疫家兔能够部分抵抗猪瘟兔化弱毒疫苗与猪伤寒沙门菌野毒株的攻击。试验提示以S.C500为CSFV新型基因疫苗运载体的猪用重组活菌苗具有可行性。  相似文献

Studies were conducted to evaluate the feasibility of using canine herpesvirus (CHV) as a vaccine vector for bait-delivered oral vaccination of wild foxes. To test the viability of CHV in baits, CHV was freeze-dried, incorporated into different baits, stored, and the remaining viral infectivity tested in cell culture after varying periods of time at different storage temperatures. Experimental baits (mouse carcasses) and commercial baits (FOXOFF and PROBAIT) were prepared with either liquid or freeze-dried CHV and tested in two fox trials for their capacity to induce CHV-specific antibodies following oral baiting. Freeze-drying and storage temperatures below 0 degrees C had a stabilizing effect to virus infectivity. When stored at -20 degrees C, freeze-dried CHV retained its full infectivity for up to 3 months in PROBAIT baits, the remaining infectivity in FOXOFF baits was 100-fold less. Oral baiting with CHV induced antiviral serum antibodies in all vaccinated foxes (20/20). None of the vaccinated foxes became ill or shed infectious virus into the environment although viral DNA was detected in body secretions as evaluated by PCR. The results indicate that CHV can be freeze-dried and stored over extended periods of time without loosing much of its infectivity. This is the first report of CHV being used for oral bait vaccination of foxes. It appears that CHV is well suited for use as a recombinant vector for wild canids.  相似文献

The effectiveness of oral immunisation of wild boar against classical swine fever (CSF) was studied in a field trial in Lower Saxony for two years, from 1993 to 1995. This field study was performed in an area of ca. 270 km(2)50% of young boars did not feed on vaccine baits nor become immunised. Therefore, an intensive hunting of this age group is a necessary adjunct to the use of oral vaccination. After the third immunisation period, no virus was detected in the areas where oral immunisation took place.  相似文献

An experimental study was performed to investigate the development of maternal antibodies after oral immunisation of young female wild boar against classical swine fever (CSF) using C-strain vaccine. Our results demonstrated that maternal antibodies do not persist in the offspring for more than 3 months. Based on the neutralising serum antibody titres, we assume that piglets of wild sows vaccinated orally twice or immunised once a long time before conception have protective antibodies for approximately 2 months. Furthermore, it seems that the level and the duration of maternal antibodies in the offspring are depend on the age of the female animals at the moment of vaccination as demonstrated in our experiment. The recent vaccination procedure consists of three double vaccinations in spring, summer and autumn. Especially vaccinations in summer and autumn could be crucial for transfer of high maternal antibody titres to the offspring.  相似文献

The H5 gene of avian influenza virus (AIV) strain A/chicken/Malaysia/5744/2004(H5N1) was cloned into pcDNA3.1 vector, and Esat-6 gene of Mycobacterium tuberculosis was fused into downstream of the H5 gene as a genetic adjuvant for DNA vaccine candidates. The antibody level against AIV was measured using enzyme-linked immunosorbent assay (ELISA) and haemagglutination inhibition (HI) test. Sera obtained from specific-pathogen-free chickens immunized with pcDNA3.1/H5 and pcDNA3.1/H5/Esat-6 demonstrated antibody responses as early as 2 weeks after the first immunization. Furthermore, the overall HI antibody titer in chickens immunized with pcDNA3.1/H5/Esat-6 was higher compared to the chickens immunized with pcDNA3.1/H5 (p < 0.05). The results suggested that Esat-6 gene of M. tuberculosis is a potential genetic adjuvant for the development of effective H5 DNA vaccine in chickens.  相似文献

During two consecutive replicates Iberian sows (n = 20) were subjected either to a conventional lactation (C) or intermittent suckling (IS). Piglets in the C treatment had free access to the dam. Piglets on IS treatment were separated from the sow during 6 h on days 29 and 30, 8 h on days 31 and 32, and 10 h on days 33 and 34. Litters on both treatments were weaned at 35 days of age and offered a starter diet until day 60 of age. The apparent digestibility (ApD) of nutrients and gross energy (GE) of the post-weaning diet was measured using Cr2O3 as external indigestible marker. Litters on IS tended to increase solid feed intake during the milk restriction (125 ± 12 vs. 70 ± 19 g piglet− 1 day− 1; P = 0.08). Feed intake was improved by IS during the second week post-weaning (676 ± 17 vs. 497 ± 12 g piglet− 1 day− 1, P < 0.01), but differences were totally offset after the 7th week of age. Differences in average growth rate of litters on C and IS treatments did not attain statistical significance during the period of restriction and in the first week post-weaning. Litters on IS showed increased growth rate during the second week post-weaning (422 ± 31 vs. 289 ± 33 g piglet− 1 day− 1; P = 0.01), but slower daily gains from the 3rd week post-weaning to 60 day of age than litters on C treatment (371 ± 14 vs. 432 ± 15 g piglet− 1 day− 1; P < 0.05). Nevertheless, neither at weaning nor at 60 days of age average body weight of piglets differed between treatments (7.71 ± 0.31 vs. 7.94 ± 0.30 kg for C and IS, at 35 days of age, 15.62 ± 0.65 vs. 16.20 ± 0.62 kg, at day 60). The apparent digestibility of nutrients of the starter diet offered after weaning remained unaffected by the treatment, except for a trend for higher ApD of GE for the piglets on IS treatment (P = 0.08). ApD for CP and GE was 78.1 ± 0.6 and 78.8 ± 0.3%, respectively. No significant differences in the proportions of total viscera and gastrointestinal tract to empty body weight (EBW) were observed between C and IS piglets at weaning and 60 d of age. In conclusion, the increase in feed intake observed prior and early after weaning as a consequence of intermittent suckling during the last week of 35 d lactation had no effect on nutrient digestibility measured at two weeks after weaning and failed on improving Iberian piglet performance.  相似文献

The present study was conducted to investigate the effects of Acanthopanax senticosus extract (ASE) as a dietary additive on gut health in weanling piglets by examining diarrhea frequency, intestinal microbiota and morphology. A total of 96 Duroc× (Landrace × Yorkshire) piglets weaned at 21 days of age with an average initial body weight (BW) of 5.6 ± 0.4 kg were randomly assigned to 3 treatment groups with 4 duplicates of 8 piglets each. The piglets were fed basal diet to which had been added 0 or 1 g/kg of ASE, or 0.7 g/kg antibiotics, respectively. Fecal consistence was monitored twice daily and the frequency of diarrhea was calculated. On day 21 after the initiation of supplementation, 8 piglets were randomly selected from each treatment group (2 piglets per pen) and slaughtered. The jejunum, ileum, colon and cecum were then excised and fixed in 10% neutral formalin solution to determine villus height and crypt depth, after their contents were collected to determine microbiota. The results showed that dietary supplementation with ASE increased (P < 0.05) the density of bacterial populations that co-migrated with Lactobacillus amylovorus, Lactobacillus salivarius, Bacillus subtilis, and Clostridium lituseburens, but decreased (P < 0.05) those co-migrating with Staphylococcus aureus, Salmonella typhimurium, Ruminococcus forques, and E. coli O157:H7 in the PCR-DGGE profiling analysis when compared with the control group. The villus height of the duodenum, jejunum and ileum increased (P < 0.05) by 14.8, 13.7 and 10.0%, while the crypt depth decreased (P < 0.05) by 17.9, 9.1 and 12.1%, respectively, in response to dietary ASE supplementation. Additionally dietary supplementation with ASE or an antibiotic decreased (P < 0.05) the frequency of diarrhea by 55.6 and 52.2%, respectively, compared with the control group. In conclusion, these findings suggest that dietary supplementation with ASE could regulate the microbiota composition and maintain a normal morphology of gut mucosa in weanling piglets, thereby decreasing diarrhea that resulted from weaning stress.  相似文献

The aim of the study was to evaluate the protection generated in mice against Toxoplasma gondii brain cyst burden by vaccination with T. gondii cytoskeleton proteins using Lactobacillus casei as adjuvant. One hundred and sixty-eight NIH mice were randomly allocated into eight groups of 21 mice each. Animals were immunized as follows: in group 1 with Toxoplasma lysate antigen (TLA) in Freund's modified adjuvant, containing L. casei (FMA), in group 2 with Toxoplasma cytoskeleton proteins (TCPs) in FMA, in group 3 with FMA, in group 4 with phosphate buffered saline (PBS), in group 5 with L. casei dead by heath (Lc), in group 6 with Freund's complete adjuvant (FCA), in group 7 with TLA in FCA, and in group 8 with TCP in FCA. Mean brain cyst burden (±S.E.M.) was assessed in mice 8 weeks after challenge with T. gondii Me49 strain (20 cysts per mouse). The percentages of reduction in cyst burden per brain (P < 0.01) as compared with the group 4 (control: mean 3181 ± 97.5) were 77.25% (724 ± 98) in group 1, 88.02% (381 ± 97.5) in group 2, 38.92% (1943 ± 130.3) in group 3, 44.31% (1771.4 ± 102) in group 5, 59.28% (1295.2 ± 99.1) in group 7 and 55.69% (1409.5 ± 89.9) in group 8. In order of importance, the best protection was obtained in groups 2, 1, 7, 8, 5 and 3. Noticeably the mice inoculated with L. casei alone showed a significant reduction in T. gondii brain cysts (P < 0.01), while those animals treated with FCA alone did not. Additionally, IgM anti-T. gondii antibody levels, as determined by ELISA 2 weeks after challenge, were highest in group 2 (P < 0.01) than in the other seven groups. Results suggest that T. gondii cytoskeleton proteins with L. casei as adjuvant constitute a good anti-toxoplasmosis vaccine candidate.  相似文献

The aim of the present study was to carry out molecular epidemiological investigation on enterotoxigenic Escherichia coli (ETEC) K99 and Salmonella spp. in diarrheic neonatal calves. Fecal samples were obtained from 220 diarrheic calves at 9 farms related to four governorates in central and northern Egypt. E. coli and Salmonella spp. isolates were examined for E. coli K99 and Salmonella spp. using PCR. ETEC K99 was recovered from 20 (10.36 %) out of 193 isolates, whereas Salmonella spp. was recovered from nine calves (4.09%).Multivariable logistic regression was used to evaluate the risk factors associated with both infections. ETEC K99 was significantly affected by age (P < 0.01; OR: 1.812; CI 95%: 0.566–1.769), colostrum feeding practice (P < 0.01; OR: 5.525; CI 95%: 2.025–15.076), rotavirus infection (P < 0.001; OR: 2.220; CI 95%: 0.273–1.251), vaccination of pregnant dams with combined vaccine against rotavirus, coronavirus and E. coli (K99) (P < 0.001; OR: 4.753; CI 95%: 2.124–10.641), and vitamin E and selenium administration to the pregnant dam (P < 0.01; OR: 3.933; CI 95%: 0.703–1.248).Infection with Salmonella spp. was found to be significantly affected by the animal age (P < 0.05; OR: 0.376; CI 95%: 0.511–1.369), Hygiene (P < 0.05; OR: 0.628; CI 95%: 1.729–5.612), and region (P < 0. 01; OR: 0.970; CI 95%: 0.841–1.624).The results of the present study indicate the importance of PCR as rapid, effective and reliable tool for screening of ETEC and Salmonella spp. when confronted with cases of undifferentiated calf diarrhea. Moreover, identification of the risk factors associated with the spreading of bacteria causing diarrhea may be helpful for construction of suitable methods for prevention and control.  相似文献

The efficacy of a new vaccine against neonatal Escherichia coli diarrhoea in piglets containing purified F4ab, F4ac, F5 and F6 fimbriae and detoxified heat‐labile toxin (LT) was tested in challenge experiments by the method described by the European Pharmacopoeia (3rd edn, EDQM, Council of Europe, Strasbourg, France). A group of 11 young sows from a herd without E. coli problems was vaccinated 6–8 and 2–4 weeks prior to expected farrowing and another group of nine young sows were non‐vaccinated controls. Escherichia coli antibody titres were determined in serum samples taken from the sows before first vaccination and before farrowing and in colostrum samples. The newborn piglets were allowed to suckle colostrum from their mother immediately after birth. The piglets were marked with individually numbered ear tags. Approximately 12 h after birth, 118 piglets from vaccinated sows and 79 piglets from non‐vaccinated control sows were challenged by oral instillation of 5 ml of a freshly prepared culture of one of the challenge strains [O8:K87:F4ab (LT+) or O149:K91:F4ac (LT+) or O9:K30:F5 or O9:K103:F6 respectively]. The challenge cultures contained as a mean 6.8 × 109 CFU/ml. After challenge the piglets were observed for 7 days and mortality and morbidity were recorded. Vaccinated sows developed significant levels of antibody titres in colostrum and serum. Control sows stayed at a low/seronegative level. The protective efficacy was excellent because 66.7–87.5% of the piglets from vaccinated sows remained without clinical signs after challenge. Only 0.0–28.0% of the piglets from non‐vaccinated sows remained healthy and more than 47.1% of the piglets in this group died after challenge. It is concluded that the new vaccine is very effective in protection of piglets against neonatal E. coli diarrhoea.  相似文献
