利用来自国际马铃薯中心含有单显性抗性基因的11个鉴别寄主,采用离体叶片测定方法对2007—2008年采自甘肃和内蒙古地区的共91个致病疫霉马铃薯分离物的致病型进行了测定。结果显示:来自甘肃的65株菌中共有28个致病型,有3株可以克服11个抗性基因;总共有超过一半的菌株对抗性基因R7、R9、R11有毒性,而能克服抗性基因R1、R2、R8的菌株数最少。在来自内蒙古的26株菌中,共检测到24个致病型,有2株菌能克服11个抗性基因,共有84.6%的菌株可以对含有抗性基因R7的植株表现出毒性,而R1、R2、R8被克服比例最低。因此,研究结果表明,在这两个地区可优先考虑在菌株采集地种植和选育含有R1,R2和R8这3个抗性基因的马铃薯品种。  相似文献

 由致病疫霉(Phytophthora infestans)引起的马铃薯和番茄的晚疫病是一种世界范围内的重要病害。在致病疫霉与寄主的互作过程中,效应因子对于病原物能否成功入侵与定殖起到了关键作用。本研究检测了来自中国马铃薯主产区的37株致病疫霉菌中编码Avr3a, Avr4和IPI-O三种效应因子的基因组成,研究发现来自国内的所有供试菌株中只有1株分离自马铃薯的和2株分离自番茄的致病疫霉含有无毒基因Avr3a(编码Avr3aKI),其余菌株均含有avr3a(编码Avr3aEM);所有菌株中都仅含有编码截断蛋白的avr4;从37株国内菌株中共检测到了25种ipiO变异型,其中20种为新变异型,聚类分析的结果表明这些变异型仍属于已报道的三类ipiO,而且大部分ipiO变异型为无毒型,可以被抗性基因Rpi-blb1识别。该研究结果表明avr3aavr4普遍存在于中国致病疫霉菌株中,推测大部分菌株可以克服R3aR4抗性基因;而ipiO的无毒变异型仍然普遍存在于国内致病疫霉群体中,可以被马铃薯含有的抗性基因Rpi-blb1识别。  相似文献

由致病疫霉Phytophthora infestans(Mont)de Bary引起的晚疫病是马铃薯生产上可导致绝收的世界性病害,在我国各地均有发生和流行[1].传统的致病型鉴别只局限于表型上的分析,无法对复杂多变的晚疫病菌致病型变化进行快速准确的预测,因此有必要探索新的方法来弥补常规手段的不足,以便更加准确有效地监测其变化趋势.晚疫病菌与寄主马铃薯这一病理体系中的单一抗性基因和相应的无毒基因符合典型"基因对基因"学说,因此基于抗病基因与无毒基因的关系对晚疫病菌分类将更为科学和实用.到目前为止,已知的晚疫病菌无毒基因序列有Avr1、Avr2、Avr3a和Avr4[2,3].本研究试图分析晚疫病菌基于无毒基因的指纹类型与马铃薯鉴别品种划分的致病型之间是否存在对应关系,以期进一步了解病原菌群体结构特征及遗传变异本质,从而更好地指导抗晚疫病育种和品种的合理布局.  相似文献

为探究水杨酸、壳聚糖、麦角甾醇和纤维二糖4种化合物诱导马铃薯抗晚疫病的效果及其作用机理,采用整株喷雾法、挑战接种法、菌丝生长速率法分析4种化合物对马铃薯晚疫病的预防作用及对其病原菌致病疫霉Phytophthora infestans的离体抑制效果,利用实时荧光定量PCR技术测定4种化合物诱导后马铃薯病程相关蛋白和防御酶基因的调控表达情况。结果显示,水杨酸、壳聚糖、纤维二糖和麦角甾醇分别处理马铃薯植株24 h后接种致病疫霉,在多个浓度下对晚疫病均有不同程度的诱导抗性,其中在1 000 μmol/L浓度(壳聚糖浓度为1 000 mg/L)时诱导抗性最好,较对照显著提高,诱抗效果分别为54.73%、61.99%、59.73%和48.59%。壳聚糖对马铃薯晚疫病的诱抗效果高于其它3种化合物,且只有壳聚糖能显著抑制致病疫霉的菌丝生长和孢子囊形成,说明壳聚糖除了能诱导作物产生抗病性外,还具有直接抑菌作用。4种化合物诱导后马铃薯病程相关蛋白基因PR1、过氧化物酶基因POD和多酚氧化酶基因PPO的表达量在施用早期均显著升高。表明这些化合物能诱导马铃薯对晚疫病产生抗性,可能与马铃薯体内过氧化物酶、多酚氧化酶的活性和抗病信号传导途径关键基因PR1的表达有关。  相似文献

 在离体条件下,我国马铃薯和番茄上62株致病疫霉中对甲霜灵和恶霜灵抗性菌株占11.29%,平均抗性水平15022倍和24733倍,未发现抗霜脲氰菌株和抗烯酰吗啉菌株。在活体条件下,比利时马铃薯上66株致病疫霉中对甲霜灵和恶霜灵抗性菌株占27.3%,河北省围场、崇礼马铃薯上217株中对甲霜灵和恶霜灵抗性菌株分别占29.0%和32.7%,河北省徐水番茄上88株中仅发现1株抗甲霜灵和恶霜灵。比利时马铃薯上的66株中4株为A2交配型,占6.1%,1株交配型为A1A2,抗甲霜灵。围场、崇礼马铃薯上的73株中6株为A2交配型,占8.2%,6株A2交配型中3株抗甲霜灵和恶霜灵。徐水番茄上的4 4株中3株A2交配型占6.8%。致病疫霉对甲霜灵和恶霜灵具有交互抗性,而2药与霜脲氰或烯酰吗啉之间无交互抗性关系。由于抗性菌株的产孢能力明显较高,田间抗甲霜灵和恶霜灵菌株明显比中间型菌株和敏感菌株的适合度高。  相似文献

大豆疫霉根腐病是大豆破坏性病害之一.利用抗病品种是防治该病的唯一有效的方法.迄今,已经鉴定了15个抗大豆疫霉根腐病基因(Rps基因),而且大豆部分基因都获得了不同类型的分子标记.本研究根据以前抗病基因推导结果选择26个可能含有Rps1基因座位等位基因或(和)Rps4基因的大豆品种(系),利用分别与Rps1a和Rm4紧密连锁的SSR标记进行抗病基因的分子检测,通过比较含有已知抗病基因对照大豆品种(系)分子标记检测结果并综合以往基因推导结果推断检测品种(系)的Rps基因.在选择的26个品种(系)中,长农14号被证明含有Rpsla,周豆13和铁95068-5含有Rps1α与Rps4基因组合,品系50794、科8924-3和合豆1号含有Rps4;有11个品种(系)存在含有Rps1a的证据、品系50052被推断含有Rps1c与Rps3b基因组合,但不排除这些品种(系)在Rps1座位含有一个新等位基因的可能性.另外.有7个品种(系)的所含抗病基因不能确定,它们可能含有新的Rps基因.  相似文献

利用41个已知抗性基因水稻品种测定2003—2006年从福建省闽东、闽南、闽西、闽北和闽中5个主要稻区采集分离的87个稻瘟病单孢菌株的致病性。结果表明,福建省稻瘟病菌群体含有与所有测试抗病基因相应的无毒基因,其中66.67%的稻瘟病菌株表现较强致病力。病菌群体对水稻抗病基因Pi-d2、Pi-k(1)、Pi-km、Pi-kh、Pi-1(1)、Pi-z5(1)、Pi-z5(2)和Pi-1(2)的毒力频率均低于10%,提示这些抗病基因在福建省可作抗源使用。2003—2006年福建省稻瘟病菌群体中分别出现了40、37、36和38个无毒基因,其中有34个无毒基因在各年份均有分布,有30个无毒基因在5个主要稻区均有分布,Avr-a(2)、Avr-3(2)、Avr-ks、Avr-4b、Avr-b、Avr-kp(C)、Avr-km(C)、Avr-ta(C)、Avr-11(C)、Avr-19(t)、Avr-t和Avr-a(1)无毒基因的出现频率均低于30%,提示与之相对应的抗病基因在福建省水稻品种抗稻瘟病育种中应慎用。含有17、14、23、18和16个无毒基因组合的病菌较多,其组合频率分别为13.79%、10.34%、9.20%、8.05%和8.05%。  相似文献

为明确致病疫霉对甲霜灵的敏感性及其交配型的动态变化,采用菌落生长速率法监测了2013-2014年采自我国北方5省(自治区)的马铃薯致病疫霉对甲霜灵的抗性,并利用标准菌株与待测菌株对峙培养测定了交配型.2013年和2014年检测的致病疫霉均以中抗菌株占主导,分别占当年检测菌株总量的82.7%和75.6%,但高抗菌株从3.3%增加到16.3%.测定甲霜灵对部分敏感、中抗和高抗菌株的EC50,发现该药剂对高抗菌株的EC50分别是中抗菌株和敏感菌株的5倍和400倍.2013年采自内蒙古、黑龙江、辽宁3省(自治区)6个县(市)的92个菌株中,A1交配型占主导地位,还发现了A2和A1A2;2014年采自内蒙古、黑龙江2省(自治区)4个县(市)的52个菌株仍以A1为主,A2所占比例较低,但未检测到A1A2.2013-2014年北方5省(自治区)致病疫霉群体结构呈现复杂化的趋势.  相似文献

 2003~2005年,从云南省德宏州新兴冬作马铃薯主产区采集的马铃薯晚疫病病样中分离纯化到86株致病疫霉菌,对其代表性菌株的表型特征进行了较系统的研究。交配型实验结果表明:所测定的62个菌株都属于A1交配型,未发现A2交配型或自育型菌株。毒性基因鉴定、监测及生理小种组成分析显示:德宏州存在所有已知可鉴定的毒性基因,而且生理小种组成非常复杂;所测定的66个菌株可区分出36个生理小种,其中,出现频率最高的小种为1,2,3,4,5,6,7,8,9,10,11,占被测菌株的18.18%,在主产区潞西县和盈江县均有分布。对60个菌株的甲霜灵抗性测定结果表明:56.67%的菌株表现为敏感,25.00%的菌株中抗,18.33%的菌株对甲霜灵有抗性。本文对此实验研究结果作了初步的分析和讨论。  相似文献

为明确宁夏回族自治区马铃薯主产区晚疫病菌的交配型种类以及生理小种的类型、组成和分布,为宁夏有针对性地选育马铃薯抗晚疫病品种提供科学依据。利用A1、A2交配型标准菌株和含有11个主效抗性基因的鉴别寄主,对2018年-2019年从原州区、泾源县、彭阳县、隆德县、西吉县、盐池县、海原县采集得到的130个马铃薯晚疫病菌菌株进行交配型和生理小种鉴定。结果表明,宁夏马铃薯晚疫病菌群体的交配型和生理小种存在多样性,采集地交配型有A1、A2、SF(自育型)3类,分别占被测菌株的30%、63.8%、6.2%;生理小种有8种类型,其中生理小种1.2.3.4.5.6.7.8.9.10.11发生频率最高,占被测菌株总数的44.62%,是宁夏马铃薯主栽区的晚疫病菌优势小种,在各测试地均有分布;次优势小种为3.4.10和1.2.3.5.6.7.8.11,发生频率分别为12.31%和10.77%。宁夏不同种植区域马铃薯晚疫病菌交配型组成复杂,采集地组成差异较大,但西吉县、原州区交配型年度间的变化基本趋于一致。宁夏马铃薯晚疫病菌群体组成日趋复杂,生产中要合理布局已有抗病品种,挖掘培育抗病水平高的新品种,综合防控马铃薯...  相似文献

以国内外收集的436份豌豆资源为试验材料,在田间自然感病的条件下,每份材料选10株,每株调查上部10片叶的病情级别,计算病情指数,进行白粉病抗性鉴定。同时对鉴定出的抗白粉病豌豆材料进行主要农艺性状的调查记载,旨在筛选出抗白粉病的豌豆优良种质资源,为今后抗性材料的有效利用提供参考,以利于加速豌豆抗性遗传育种进程。鉴定结果显示,436份材料中,抗病资源35份,占8.03%,其中11份高抗(HR),占2.52%;6份抗病(R),占1.38%;18份中抗(MR),占4.13%。综合抗白粉病性与农艺性状优良性,筛选出半无叶型豌豆抗白粉病资源‘RZ-1-1’、‘X9002’、‘1702’、‘MP1811’,普通蔓生型豌豆抗白粉病资源‘DX23’、‘DX25’、‘定褐’、‘黑眉’、‘所427’、‘所430’可进一步加以利用。  相似文献

Differential interactions in tuber blight attack between potato cultivars and Phytophthora infestans isolates were studied using whole tuber and tuber slice assays. Tuber blight incidence and severity were studied in a whole tuber assay, whilst necrosis and mycelium coverage were evaluated in a tuber slice assay. The overall defence reaction of the potato cultivars tested varied considerably. Cultivars like Kartel and Producent showed resistant reactions, whilst Bintje and, to a lesser extent, Astarte reacted more susceptibly after inoculation with aggressive strains of P. infestans . A highly significant cultivar by year interaction was observed when tuber blight incidence was evaluated in two successive years. Differential responses were revealed by changing ranked order of cultivars after exposure to aggressive isolates of P. infestans . The results show that cultivar by isolate interactions existed for all components of tuber blight resistance studied. The quantitative nature of the observed resistance responses suggests the presence of quantitative trait loci governing resistance to tuber blight. The consequences of differential interactions in relation to the stability of tuber resistance are discussed.  相似文献

Competition between genotypes of Phytophthora infestans was studied by inoculating potato cultivars with differing susceptibility to late blight in field experiments over three years in Northern Ireland, UK, and Michigan, USA. Multiple isolates of six genotype groups of P. infestans were chosen from the local populations in both N. Ireland and Michigan for inoculation of separate field trials planted in 2003, 2004 and 2005. Four cultivars were used in each trial; two (susceptible cv. Atlantic and the partially resistant cv. Stirling) were common to both locations, whereas the two additional cultivars (with partial resistance to late blight) were cvs Santé and Milagro in N. Ireland and cvs Pike and Jacqueline Lee in Michigan. Single-lesion isolates of P. infestans were obtained from leaves at 1% level of infection, characterized using pre-assigned markers and re-assigned to their respective genotype groups. Extreme selection occurred within the population of genotypes of P. infestans in N. Ireland in each year, with different genotype groups dominating the infection of different cultivars. Selection was observed on all cultivars tested, but was greatest on the more resistant cultivars. Over the 3 years, all of the 114 isolates obtained from cv. Milagro belonged to a single group, whereas among the 118 isolates from cv. Atlantic all six groups were represented. By contrast, in Michigan, the US-8 genotype dominated infection in all cultivars in each year; only 12 of 374 isolates characterized belonged to other genotypes (11 US-14 and a single US-10 isolate).  相似文献

The aggressiveness of 20 Northern Ireland single-lesion isolates of Phytophthora infestans was compared following their inoculation onto detached leaflets of three potato cultivars chosen on the basis of their differing levels of race-nonspecific resistance to late blight: Bintje (highly susceptible); Cara (moderately resistant); and Stirling (more resistant). Five isolates from outside Northern Ireland were included for comparative purposes: two from the Republic of Ireland; two from the USA (representing the US-1 and US-8 clonal lineages); and one from Mexico. To control the variation between tests, a balanced incomplete block design was used, as opposed to either a complete block design or the method of inclusion of standard isolates, where such variation would have increased the error. Highly significant variation for disease parameters, including latent period, infection frequency, area under the lesion expansion curve (AULEC) and sporulation capacity, was found between isolates. These differences were much more marked on the cultivars exhibiting higher levels of race-nonspecific resistance. There was a significant interaction between isolate and cultivar for all parameters assessed and, overall, no one isolate was the most aggressive across all three potato cultivars. However, a group comprising seven of the 20 Northern Ireland isolates was consistently found to exhibit the highest levels of aggression towards all three cultivars for each of the disease parameters. These results demonstrate that significant variation for foliar aggressiveness exists within the Northern Ireland population of P. infestans , and indicate the importance of selecting appropriately aggressive isolates for evaluation of host resistance to late blight within breeding programmes.  相似文献

 种质资源筛选是抗病育种的前提和基础。以已知抗病水平的5份鉴别品种为对照,对水稻5个类群和1个混合类群的299份品种进行了温室苗期纹枯病抗性鉴定,筛选抗纹枯病水稻新种质。在“雾室/mist-chamber”环境下以改进的带菌木质短棒为接种物、以基于“叶枕高”的“0~9”级病级指标为标准,可有效区分对照品种间的抗病水平。299份品种中未发现免疫和高抗品种,中抗以上品种比例仅为36.5%,多数品种为中感至高感水平。就水稻不同类群而言,AUS类群中中抗以上品种的分布频率最高,超过60%;其次为ARO类群,为54.6%;分布频率最低的为TRJ类群,仅为22.7%。结合各品种苗高及与抗病对照YSBR1间的病级差异,从299份品种中筛选到7份抗病新种质,其中1份的抗性显著高于YSBR1,接近高抗水平。本研究为水稻抗纹枯病遗传育种提供了新的抗源,同时为选择合适的类群间品种杂交以培育抗纹枯病新品种提供了理论依据。  相似文献

葡萄霜霉病是葡萄生产上重要的病害之一,通过对辽宁省不同葡萄品种进行室内离体叶片接种和田间自然发病情况调查,以期为葡萄抗性品种的选育和葡萄霜霉病的防治提供科学依据。结果表明,在供试品种中,没有对霜霉病完全免疫的品种,室内离体叶片接种和田间调查结果基本一致,不同品种间霜霉病的抗性存在差异。供试的65个品种中,室内离体叶片接种评价高抗品种3个,抗病品种23个,感病品种24个,高感品种15个;田间自然发病调查评价高抗品种1个,抗病品种19个,感病品种35个,高感品种10个。欧美杂交品种(系)相对欧亚杂交品种(系)较抗病。  相似文献

1983-2007年,对南方稻区水稻区域试验品种、组合的白叶枯病抗性进行了研究。结果表明,在1 805个参试品种、组合中,78个表现抗白叶枯病,抗病品种频率为4.3%;其中,常规水稻74个,占抗病品种的94.9%。在1 219个籼型杂交水稻中,抗病的组合3个,中抗的76个,抗病和中抗组合频率分别为0.2%和6.2%。不同类型和不同生态型水稻的抗病品种频率差异显著,晚粳>早籼>中籼>晚籼;常规水稻>杂交水稻。籼型杂交水稻中抗病组合频率为:杂交晚稻>杂交早稻>杂交中稻。1990-2004年,17个抗或中抗白叶枯病的优良水稻新品种、组合通过国家审定。  相似文献

The rate of late blight disease was analysed for individuals of a diploid Solanum phureja – Solanum tuberosum dihaploid hybrid population (PD), using three different assessment techniques, in the laboratory, screenhouse, and field. These hybrids expressed low disease rates in the field, comparable to resistance based on intact R genes. However, none of the parents of PD expressed any R genes and the pattern of segregation within the PD population was not indicative of R-gene inheritance. The foliage (or leaflet) area diseased had the largest broad-sense heritability of all criteria analysed, in all tests performed. In the field evaluations, the PD population showed intermediate levels of broad-sense heritability for foliage area diseased, relative to the much larger heritability detected for the group of controls possessing R genes. Resistance in the field of the PD hybrids had very little genotype-environment (G × E) interaction, indicating stability of its expression. All genotypes without R genes exhibited heritable, reduced rates of late blight disease in the field, but they were susceptible with low heritability in screenhouse and laboratory tests. This differential expression of disease indicates that the plants' indirect response to unknown environmental stress in the field may have been measured. The value of the PD hybrids for breeding of late blight resistant potato and the use of the disease rate data for detection of the underlying quantitative trait loci are discussed.  相似文献
