In the arbuscular mycorrhizal (AM) symbiosis, plants take up part of the nitrogen (N) through a mycorrhizal pathway. In this study, we assessed the effect of different N sources on the expression of genes coding for enzymes and transporters of the mycorrhizal N uptake pathway, using Sorghum bicolor and Glomus intraradices as a model. Some of the genes investigated were differentially regulated in the intraradical and in the extraradical mycelium depending on the N source. In AM roots, some fungal and plant genes were co-regulated, suggesting an interdependence of both partners in the mycorrhizal N uptake pathway. Mycorrhizal N transfer may have a preference for glycine (plant growth and N uptake stimulation).

Water scarcity threatens global food security and agricultural systems are challenged to achieve high yields while optimizing water usage. Water deficit can be accentuated by soil physical degradation, which also triggers water losses through runoff and consequently soil erosion. Although soil health in cropping systems within the Brazilian Cerrado biome have been surveyed throughout the years, information about soil erosion impacts and its mitigation are still not well understood; especially concerning the role of cropping system diversification and its effects on crop yield. Thus, the aim of this study was to assess whether ecological intensification of cropping systems –inclusion of a consorted perennial grass and crop rotation– could promote soil coverage and consequently decrease water erosion and soil, water, and nutrient losses. This work studied the effects of crop rotation and consorted Brachiaria, along with different levels of investment in fertilization on soil physical quality and on soil, water, and nutrient losses, and crop yields. Results proved that soybean monoculture (SS) is a system of low sustainability even under no-till in the Brazilian Cerrado conditions. It exhibited high susceptibility to soil, water, and nutrient losses, causing low crop yields. Our results showed that water losses in SS cropping system were approximately 10% of the total annual rainfall, and total K losses would require an additional 35% of K application. Conversely, ecological intensification of cropping systems resulted in enhanced soil environmental and agronomic functions, increased grain yield, and promoted soil and water conservation: high soil cover rate, and low soil, water and nutrient losses. Ecological intensification proved to be an adequate practice to boost crop resilience to water deficit in the Brazilian Cerrado.

Mycorrhizal fungi and earthworms can individually or interactively influence plant growth and heavy metal uptake. The influence of earthworms and arbuscular mycorrhizal (AM) fungi either alone or in combination on maize (Zea mays L.) growth and cadmium (Cd) uptake was investigated in a calcareous soil artificially spiked with Cd. Soils were contaminated with Cd (10 and 20 mg Cd kg−1), inoculated or un-inoculated with the epigeic earthworm Lumbricus rubellus and two AM fungal species (Rhizophagus irregularis and Funneliformis mosseae) for two months of growth under greenhouse conditions. Generally, earthworms alone increased both shoot P uptake and biomass but decreased shoot Cd concentration and root Cd uptake. AM fungi individually often increased total maize P uptake, declined shoot Cd concentration, and consequently produced higher total biomass. However, R. irregularis enhanced shoot Cd uptake at low Cd level and root Cd uptake at high Cd level. In plants inoculated with F. mosseae species, earthworms increased shoot biomass and Cd uptake, decreased root biomass and Cd uptake at all Cd levels, and increased shoot Cd concentration at low Cd level. In plants colonized by R. irregularis species, however, earthworm addition decreased maize biomass only at high Cd level and root Cd concentration and total maize Cd uptake at both Cd levels. Earthworm activity decreased Cd transfer from the soil to maize roots at low Cd level, but this was counterbalanced in the presence of F. mosseae. Mycorrhizal symbiosis significantly reduced the transfer of Cd from roots to shoots, independence of earthworm effect. Overall, it is concluded that L. rubellus and AM fungi, in particular F. mosseae isolate, improved maize tolerance to Cd toxicity both individually and interactively by increasing plant growth and P nutrition, and restricting Cd transfer to the aboveground biomass. Consequently, the single and interactive effects of the two soil organisms might potentially be important not only in protecting maize plants against Cd toxicity, but also in Cd phytostabilization in soils polluted by this highly toxic metal.

Soil aggregate (SA) can be formed and stabilized when soil organic matter (SOM) is decomposed in the soil. However, the relationships between the SA dynamics and SOM with different decomposition rates have not been clarified. Therefore, this study examined the effects of the addition of polysaccharides to soil on SA formation and stability. A Japanese tropical soil was incubated for 99 d at 30 °C in a dark environment following the addition of 0.5% (w/w) starch or cellulose. The decomposition rates of the amendments, and SA formation and stability were evaluated by measuring soil respiration rates, and distribution fractions of soil aggregate sizes and mean weight diameter (MWD) of SA, respectively. The cumulative soil respirations with all treatments rapidly increased until Day 12 of the incubation. The initial slope of the cumulative soil respiration in the soil with starch was significantly higher than that in the soil with cellulose. In either soil with starch or cellulose, the fractions of macro-aggregates (>1000 μm in diameter) significantly increased, respectively, compared with control soil. However, the fractions of meso-aggregates (250–1000 μm) and nano-aggregate (<20 μm) in the soil with starch significantly decreased, while those fractions in the soil with cellulose fluctuated until Day 6. The MWDs reached the maximum on Day 6, indicating the SA formation in the soils with starch or cellulose. The increasing rate of the SA formation in the starch-amended soil was greatly higher than that in the cellulose-amended soil. After Day 6, the MWDs in the soils with either polysaccharide decreased with similar trends with no significant differences between treatments, indicating similar stability of the SA in both treatments. This study showed that the different decomposability of the organic amendments might influence the SA formation differently, but not the SA stability.

Disturbance and change to C inputs can alter microbial community structure and impact ecosystem function. Particularly in temperate regions, seasonal change also has an effect on microbial communities both directly through climate and indirectly through plant function. The temporal change in microbial communities of an undisturbed pasture, disturbed pasture (similar to a single tillage event) and pasture soil amended with two forms of particulate carbon were monitored over eight consecutive seasons after grass was reestablished. The soil microbial community was assessed by a DNA fingerprinting technique (terminal restriction fragment length polymorphism, TRFLP) of bacterial, fungal and archaeal communities, and also from phospholipid fatty acid (PLFA) analysis. The single disturbance had a significant effect on fungal microbial community structure (by TRFLP) and significantly decreased the fungal:bacterial ratio. Though the change was relatively small, it persisted throughout the sampling period. Nitrate was also higher on the disturbed treatment providing evidence for the theory that changes to fungal:bacterial ratios can alter nutrient cycling and retention. Fungal communities were the most altered by the C amendments, and while bacteria were also affected by the C amendments, seasonal change was a greater cause of variation. Correlation to soil and climatic variables explained more of the total variability for PLFA (78% for all treatments) than bacterial (50%), fungal (35%) and archaeal (14%) restriction fragments. Most climate and soil variables explained significant variation for seasonal patterns in the multivariate community structures but measurements of soil moisture were important for all communities while pH was relatively more important for bacteria, temperature for fungi, and soil C:N ratio for archaea. Autumn was particularly distinct from other seasons for bacteria (less so for the fungal community) and although there was seasonal change in pH suggesting pasture management was a factor, the significant correlation of other soil characteristics suggests that plant physiological changes (most probably root exudates) also played a significant role. The large change in the saprotrophic fungal community due to the particulate C addition but minor seasonal change would tend to suggest that the fungal community may be more responsive to changes in litter inputs rather than root exudates while the reverse is true for bacteria.

Earthworms and arbuscular mycorrhizal fungi (AMF) are known to independently affect soil microbial and biochemical properties, in particular soil microbial biomass (SMB) and enzymes. However, less information is available about their interactive effects, particularly in soils contaminated with heavy metals such as cadmium (Cd). The amount of soil microbial biomass C (MBC), the rate of soil respiration (SRR) and the activities of urease and alkaline phosphatase (ALP) were measured in a calcareous soil artificially spiked with Cd (10 and 20 mg Cd kg−1), inoculated with earthworm (Lumbricus rubellus L.), and AMF (Glomus intraradices and Glomus mosseae species) under maize (Zea mays L.) crop for 60 days. Results showed that the quantity of MBC, SRR and enzyme activities decreased with increasing Cd levels as a result of the elevated exchangeable Cd concentration. Earthworm addition increased soil exchangeable Cd levels, while AMF and their interaction with earthworms had no influence on this fraction of Cd. Earthworm activity resulted in no change in soil MBC, while inoculation with both AMF species significantly enhanced soil MBC contents. However, the presence of earthworms lowered soil MBC when inoculated with G. mosseae fungi, showing an interaction between the two organisms. Soil enzyme activities and SRR values tended to increase considerably with the inoculation of both earthworms and AMF. Nevertheless, earthworm activity did not affect ALP activity when inoculated with G. mosseae fungi, while the presence of earthworm enhanced urease activity only with G. intraradices species. The increases in enzyme activities and SRR were better ascribed to changes in soil organic carbon (OC), MBC and dissolved organic carbon (DOC) contents. In summary, results demonstrated that the influence of earthworms alone on Cd availability is more important than that of AMF in Cd-polluted soils; and that the interaction effects between these organisms on soil microorganism are much more important than on Cd availability. Thus, the presence of both earthworms and AMF could alleviate Cd effects on soil microbial life.

To investigate the climate impacts on the different components of ecosystem respiration, we combined soil efflux data from a tree-girdling experiment with eddy covariance CO2 fluxes in a Mediterranean maritime pine (Pinus pinaster) forest in Central Italy. 73 trees were stem girdled to stop the flux of photosynthates from the canopy to the roots, and weekly soil respiration surveys were carried out for one year. Heterotrophic respiration (RH) was estimated from the soil CO2 flux measured in girdled plots, and rhizosphere respiration (RAb) was calculated as the difference between respiration from controls (RS) and girdled plots (RH).Results show that the RS dynamics were clearly driven by RH (average RH/RS ratio 0.74). RH predictably responded to environmental variables, being predominantly controlled by soil water availability during the hot and dry growing season (May–October) and by soil temperature during the wetter and colder months (November–March). High RS and RH peaks were recorded after rain pulses greater than 10 mm on dry soil, indicating that large soil carbon emissions were driven by the rapid microbial oxidation of labile carbon compounds. We also observed a time-lag of one week between water pulses and RAb peaks, which might be due to the delay in the translocation of recently assimilated photosynthates from the canopy to the root system. At the ecosystem scale, total autotrophic respiration (RAt, i.e. the sum of carbon respired by the rhizosphere and aboveground biomass) amounted to 60% of ecosystem respiration. RAt was predominantly controlled by photosynthesis, and showed high temperature sensitivity (Q10) only during the wet periods. Despite the fact that the study coincided with an anomalous dry year and results might therefore not represent a general pattern, these data highlight the complex climatic control of the respiratory processes responsible for ecosystem CO2 emissions.

Successful restoration of an ecosystem following disturbance is typically assessed according to similarity between the restored site and a relatively undisturbed reference area. While most comparisons use the average or mean parameter to represent measured properties, other aspects of the distribution, including the variance of the properties may assist in a more robust assessment of site recovery. Our purpose was to compare soil properties in different ages of reclaimed soils with those in reference areas by incorporating the potentially different distributions according to areas. On two sampling dates, in consecutive years, we examined soil properties on a chronosequence of reclaimed natural gas pipelines spanning recovery ages of <1–54 years, obtaining data on soil moisture, organic carbon, nitrogen, electrical conductivity, pH, and microbial abundance. To make the comparisons, we analyzed our data with a Bayesian hierarchical linear mixed model and obtained posterior predictive distributions for the soil properties. This allowed us to probabilistically quantify the extent to which a soil property from a reclaimed treatment was similar to that from an undisturbed reference. We found that the posterior predictive variance of most soil properties was particularly sensitive to disturbance and reclamation, especially, within the first few years of recovery. Response of this variance to disturbance, reclamation, and recovery was not necessarily accompanied by a shift in the posterior predictive mean value of the property. Patterns for all soil properties changed over time, with posterior predictive distributions of soil properties generally becoming more similar to those of the undisturbed reference sites as recovery time increased. We suspect these trends in altered variability coincide with the degree of spatial heterogeneity in soil properties that results following disturbance and reclamation, which is also coupled to patterns of vegetation recovery.

A pot experiment was conducted to investigate the effect of epigeic earthworm (Eisenia fetida) and arbuscular mycorrhizal (AM) fungi (Glomus intraradices) on soil enzyme activities and nutrient uptake by maize, which was grown on a mixture of sterilized soil and sand. Maize plants were grown in pots inoculated or not inoculated with AMF, treated or not treated with earthworms. Wheat straw was added as a feed source for earthworms. Mycorrhizal colonization of maize was markedly increased in AM fungi inoculated pots and further increased by addition of epigeic earthworms. AM fungi and epigeic earthworms increased maize shoot and root biomass, respectively. Soil acid phosphatase activity was increased by both earthworms and mycorrhiza, while urease and cellulase activities were only affected by earthworms. Inoculation with AM fungi significantly (p?<?0.001) increased the activity of soil acid phosphatase but decreased soil available phosphorus (P) and potassium (K) concentrations at harvest. Addition of earthworms alone significantly (p?<?0.05) increased soil ammonium-N content, but decreased soil available P and K contents. AM fungi increased maize shoot weight and root P content, while earthworms improved N, P, and K contents in shoots. AM fungi and earthworm interactively increased maize shoot and root biomass through their regulation of soil enzyme activities and on the content of available soil N, P, and K.

A pot experiment was carried out to study the effects of three arbuscular mycorrhizal fungi (AMF), including Glomus intraradices, Glomus constrictum and Glomus mosseae, on the growth, root colonization and Cd accumulation of marigold (Tagetes erecta L.) at Cd addition levels of 0, 5 and 50 mg kg-1 in soil. The physiological characteristics, such as chlorophyll content, soluble sugar content, soluble protein content and antioxidant enzyme activity, of Tagetes erecta L. were also investigated. The symbiotic relationship between the marigold plant and arbuscular mycorrhizal fungi was well established under Cd stress. The symbiotic relationship was reffected by the better physiobiochemical parameters of the marigold plants inoculated with the three AMF isolates where the colonization rates in the roots were between 34.3% and 88.8%. Compared with the non-inoculated marigold plants, the shoot and root biomass of the inoculated marigold plants increased by 15.2%- 47.5% and 47.8%-130.1%, respectively, and the Cd concentration and accumulation decreased. The chlorophyll and soluble sugar contents in the mycorrhizal marigold plants increased with Cd addition, indicating that AMF inoculation helped the marigold plants to grow by resisting Cd stress. The antioxidant enzymes reacted differently with the three AMF under Cd stress. For plants inoculated with G. constrictum and G. mosseae, the activities of superoxide dismutase (SOD) and catalase (CAT) increased with increasing Cd addition, but peroxidase (POD) activity decreased with increasing Cd addition. For plants inoculated with G. intraradices, three of the antioxidant enzyme activities were significantly decreased at high levels of Cd addition. Overall, the activities of the three antioxidant enzymes in the plants inoculated with AMF were higher than those of the plants without AMF inoculation under Cd stress. Our results support the view that antioxidant enzymes have a great influence on the biomass of plants, and AMF can improve the capability of reactive oxygen species (ROS) scavenging and reduce Cd concentration in plants to alleviate Tagetes erecta L. from Cd stress.

Earthworms and arbuscular mycorrhizal fungi (AMF) might interactively impact plant productivity; however, previous studies reported inconsistent results. We set up a three-factorial greenhouse experiment to study the effects of earthworms (Aporrectodea caliginosa Savigny and Lumbricus terrestris L.) and AMF (Glomus intraradices N.C. Schenck & G.S. Sm.) on the performance (productivity and shoot nutrient content) of plant species (Lolium perenne L., Trifolium pratense L. and Plantago lanceolata L.) belonging to the three functional groups grasses, legumes and herbs, respectively. Further, we investigated earthworm performance and plant root mycorrhization as affected by the treatments. Our results accentuate the importance of root derived resources for earthworm performance since earthworm weight (A. caliginosa and L. terrestris) and survival (L. terrestris) were significantly lower in microcosms containing P. lanceolata than in those containing T. pratense. However, earthworm performance was not affected by AMF, and plant root mycorrhization was not modified by earthworms. Although AMF effectively competed with T. pratense for soil N (as indicated by δ15N analysis), AMF enhanced the productivity of T. pratense considerably by improving P availability. Remarkably, we found no evidence for interactive effects of earthworms and AMF on the performance of the plant species studied. This suggests that interactions between earthworms and AMF likely are of minor importance.

A greenhouse pot experiment was conducted to study cadmium (Cd) and phosphorous (P) acquisition of upland kangkong (Ipomoea aquatica Forsk.) intercropped with Alfred stonecrop (Sedum alfredii Hance) in a Cd-contaminated soil inoculated with arbuscular mycorrhizal (AM) fungi. There were four treatments, including monoculture of kangkong (control), intercropping with stonecrop (IS), and intercropping with stonecrop plus inoculation with Glomus caledonium (IS + Gc) or Glomus versiforme (IS + Gv). Both kangkong and stonecrop plants were harvested at week 8 after seeding or cutting. Compared with the control, IS tended to decrease Cd and P acquisition by neighboring kangkong via competition for phytoavailable Cd and P. The inoculation of Gc, but not Gv, significantly elevated Cd acquisition by stonecrop, and hence resulted in significantly lower Cd acquisition by kangkong and the subsequent Cd concentrations in both roots and shoots of kangkong. Both Gc and Gv significantly increased mycorrhizal colonization rates in stonecrop roots, as well as acid phosphatase activities and available P concentrations in the soil. However, only Gc significantly elevated P acquisition and shoot biomass of the host plant (stonecrop), while Gv significantly increased P acquisition and shoot biomass of neighboring kangkong rather than of stonecrop, causing a significant dilution effect on kangkong shoot Cd concentration. In addition, both Gc and Gv inoculation significantly decreased soil DTPA-extractable (phytoavailable) Cd concentrations by elevating soil pH. The results showed that Gc and Gv played totally different roles in the intercropping system for vegetable production and phytoremediation of Cd-contaminated soils.

Heavy metal(HM) contamination in soils is an environmental issue worldwide that threatens the quality and safety of crops and human health. A greenhouse experiment was carried out to investigate the growth, mycorrhizal colonization, and Pb and Cd accumulation of pakchoi(Brassica chinensis L. cv. Suzhou) in response to inoculation with three arbuscular mycorrhizal(AM) fungi(AMF), Funneliformis mosseae, Glomus versiforme, and Rhizophagus intraradices, aimed at exploring how AMF inoculation affected safe crop production by altering plant-soil interaction. The symbiotic relationship was well established between pakchoi and three AMF inocula even under Pb or Cd stress, where the colonization rates in the roots ranged from 24.5% to 38.5%. Compared with the non-inoculated plants, the shoot biomass of the inoculated plants increased by 8.7%–22.1% and 9.2%–24.3% in Pb and Cd addition treatments, respectively. Both glomalin-related soil protein(GRSP) and polyphosphate concentrations reduced as Pb or Cd concentration increased. Arbuscular mycorrhizal fungi inoculation significantly enhanced total absorbed Pb and Cd(except for a few samples) and increased the distribution ratio(root/shoot) in pakchoi at each Pb or Cd addition level. However, the three inocula significantly decreased Pb concentration in pakchoi shoots by 20.6%–67.5% in Pb addition treatments, and significantly reduced Cd concentration in the shoots of pakchoi in the Cd addition treatments(14.3%–54.1%), compared to the non-inoculated plants.Concentrations of Pb and Cd in the shoots of inoculated pakchois were all below the allowable limits of Chinese Food Safety Standard. The translocation factor of Pb or Cd increased significantly with increasing Pb or Cd addition levels, while there was no significant difference among the three AMF inocula at each metal addition level. Meanwhile, compared with the non-inoculated plants, AMF inocula significantly increased soil p H, electrical conductivity, and Pb or Cd concentrations in soil organic matter in the soils at the highest Pb or Cd dose after harvest of pakchoi, whereas the proportion of bioavailable Pb or Cd fraction declined in the AMF inoculated soil. Our study provided the first evidence that AM fungi colonized the roots of pakchoi and indicated the potential application of AMF in the safe production of vegetables in Pb or Cd contaminated soils.

Previous greenhouse and field studies have shown arbuscular mycorrhizal (AM) plants usually have greater P uptake and growth when raised in undisturbed soil compared to soil disturbed between plantings, such as by tillage. We report here for the first time that AM fungi able to stimulate shoot P uptake in experimental comparisons to non-mycorrhizal plants differ in their ability to bring about similar responses in undisturbed soil compared to disturbed soil. This outcome indicates a difference in functional character between the two stimulation processes. Three isolates of AM fungi were tested for growth promotion of maize (Zea mays L.) in pots in a soil disturbance experiment that included non-mycorrhizal controls. All three fungi colonized roots well and promoted shoot P uptake compared to non-inoculated controls, but only Glomus mosseae was able to stimulate growth in undisturbed soil compared to disturbed soil. This effect was seen when Glomus mosseae was alone or in combination with Gigaspora margarita. However, the presence of Glomus aggregatum in combination with Glomus mosseae prevented any stimulation, presumably due to domination by Glomus aggregatum. The ability of AM fungi to be beneficial to plants in comparison to non-mycorrhizal situations likely relates to the spread of mycelium in the soil and the capacity for nutrient transfer to the root. The ability of an AM fungus to promote growth in undisturbed soil appears to be related to these features and, in addition, a capacity for persistence and retention of functional capacity of the extraradical mycelium from one plant generation to the next.

Plants inoculated with arbuscular mycorrhizal (AM) fungi utilize more soluble phosphorus from soil mineral phosphate than non-inoculated plants. However, there is no information on the response of soil microflora to mineral phosphate weathering by AM fungi and, in particular, on the catabolic diversity of soil microbial communities. The AM fungus, Glomus intraradices was examined for (i) its effect on the growth of Acacia holosericea, (ii) plant-available phosphate and (iii) soil microbial activity with and without added rock phosphate. After 4-months culture, AM fungal inoculation significantly increased the plant biomasses (by 1.78× and 2.23× for shoot and root biomasses, respectively), while mineral phosphate amendment had no effect in a sterilized soil. After 12-months culture, the biomasses of A. holosericea plants growing in a non-sterilized soil amended with mineral phosphate were significantly higher than those recorded in the control treatment (by 2.5× and 5× for shoot and root biomasses, respectively). The fungal inoculation also significantly stimulated plant growth, which was significantly higher than that measured in the mineral phosphate treatment. When G. intraradices and mineral phosphate were added together to the soil, shoot growth were significantly stimulated over the single treatments (inoculation or amendment) (1.45×). The P leaf mineral content was also higher in the G. intraradices+mineral phosphate treatment than in G. intraradices or rock phosphate amendment. Moreover, the number of fluorescent pseudomonads has been significantly increased when G. intraradices and/or mineral phosphate were added to the soil. By using a specific type of multivariate analysis (co-inertia analysis), it has been shown that plant growth was positively correlated to the metabolization of ketoglutaric acid, and negatively linked to the metabolisation of phenylalanine and other substrates, which shows that microbial activity is also affected.G. intraradices inoculation is highly beneficial to the growth of A. holosericea plants in controlled conditions. This AM symbiosis optimises the P solubilization from the mineral phosphate and affects microbial activity in the hyphosphere of A. holosericea plants.

The effectiveness of reforestation programs on degraded soils in the Mediterranean region is frequently limited by a low soil availability and a poor plant uptake and assimilation of nutrients. While organic amendments can improve the nutrient supply, inoculation with mycorrhizal fungi can enhance plant nutrient uptake. A pot experiment was conducted in 2004 to study the influence of inoculation with an arbuscular mycorrhizal (AM) fungus (Glomus intraradices Schenck & Smith) or with a mixture of three AM fungi (G. intraradices, G. deserticola Trappe, Bloss. & Menge, and G. mosseae (Nicol & Gerd.) Gerd. & Trappe) and of an addition of composted sewage sludge or Aspergillus niger–treated dry‐olive‐cake residue on plant growth, nutrient uptake, mycorrhizal colonization, and nitrate reductase (NR) activity in shoot and roots of Juniperus oxycedrus L. Six months after planting, the inoculation of the seedlings with G. intraradices or a mixture of three AM fungi was the most effective treatment for stimulating growth of J. oxycedrus. There were no differences between the two mycorrhizal treatments. All treatments increased plant growth and foliar N and P contents compared to the control plants. Mycorrhizal inoculation and organic amendments, particularly fermented dry olive cake, increased significantly the NR activity in roots.
