We conducted a population genetic analysis of Penaeus stylirostris to describe the genetic variability in two wild samples (Guaymas and Peñasco) and in two cultivated strains (A and B), and to determine the relationship among samples. Seventeen enzymatic systems and general proteins were used to visualize 31 loci. Of the 31 loci, 11 polymorphic loci (35%) were detected in all the organisms, with an average heterozygosity of 0.10 and an average of 1.8 per locus. There were no significant differences among the four samples of observed heterozygosity or percentage of polymorphic loci. According to Hardy–Weinberg, both cultivated strains showed disequilibrium in more loci than the wild samples. Guaymas showed linkage disequilibrium in five pairs of polymorphic loci, Peñasco samples in two pairs, A in seven pairs, and B in nine pairs. Peñasco and both cultivated lineages showed evidence of recent reductions in their effective populations size. Based on the comparisons of Fst values, and the distribution of the allelic frequencies, the cultivated strains were significantly different from the two wild populations. There is evidence that strain B originated from A and the discrepancies between the strains are probably the result of genetic drift and a founder effect.  相似文献

利用9个微卫星位点对文蛤红壳(RS)、黑斑(BS)、细纹(TC)、暗纹(DF)4个壳色花纹品系共120个个体进行遗传差异分析。结果共检测到105个等位基因,每个位点平均观察等位基因数为11.7个,其中WG07位点等位基因数最多(21个),WG11位点最少(7个)。每个品系均具有特异等位基因类型,不同品系中相同微卫星位点的等位基因分布规律不同。4个文蛤品系的观测杂合度、期望杂合度和多态信息含量变异范围分别为0.110~1.000、0.486~0.867、0.433~0.834,均属高度多态性位点。Hardy-Weinberg平衡的卡方检测发现,大多数位点偏离平衡状态;聚类分析表明,4个品系间存在较明显的遗传差异,TC品系与其它品系的遗传距离最大,DF品系和RS品系的遗传距离相对较小。  相似文献

Genetic variation fuels selective change in natural and captive populations. In establishing a broodstock for selective improvement, the level of genetic diversity is an important consideration because it provides an indication of the scope for selective progress. Three domesticated strains of rainbow trout, Oncorhynchus mykiss (Walbaum), were examined at nine polymorphic microsatellite loci to assess detectable levels of allelic diversity and heterozygosity within and differentiation among the strains. A total of 126 alleles were observed to segregate into unique multilocus genotypes for each of the 152 individuals assayed. There was an average heterozygosity of 71.5% at these nine loci, and an average of 14 alleles at a locus. Each locus was represented by alleles unique to at least two of the three strains. Deviations from Hardy–Weinberg expectations of genotype frequencies were detected in each strain. Subsequent analysis indicated sub‐structuring within strains leading to Wahlund effects that caused these deviations. Significant differences in genotype frequencies and pairwise FST values demonstrated that all strains were unique. The overall FST of 0.089 provides additional evidence of unique genetic diversity present in each strain, and agrees well with the degree of genetic variation found in rainbow trout across broad geographical ranges. The genetic diversity contributed by each population suggests that there is greater scope for selective improvement of numerous traits within a synthetic strain combining these three strains than within any individual strain.  相似文献

Abstract. An examination of biochemical-genetic variation at seven polymorphic loci was carried out among five year classes of wild Atlantic salmon, Salar L., in the R. Bush and in a hatchery strain derived from the wild population. Within some of the year classes, gene frequencies at several loci differed significantly between wild and artificially reared salmon. Highly significant temporal variation in gene frequencies was detected among successive year classes of the hatchery strain, while this was less significant among the wild salmon. Samples of wild salmon taken as 0+ and 1 + parr in the river showed no significant temporal variability in allelic frequencies. Heterozygosity levels among the wild and hatchery-reared salmon were comparable, averaging 0·185 and 0·176 respectively. The genetic variability of the artificially reared salmon is discussed in relation to numbers of broodstock and breeding regime used at the hatchery.  相似文献

采用磁珠富集和PCR筛选相结合的方法,得到番红砗磲的19个多态性微卫星标记。利用新开发微卫星标记对西沙群岛2个野生群体的遗传多样性进行比较分析,七连屿海域野生群体和永兴岛海域野生群体的平均观测等位基因数(Na)分别为11.105、11.895,平均有效等位基因数(Ne)分别为6.274、6.173,平均期望杂合度(He)分别为0.776、0.788,平均多态性信息含量分别为0.730、0.744,发现2个野生群体的遗传多样性都处于高度多态水平,说明其有效群体大小仍然保持在较高水平。Bonferroni校正后,在2个群体中各有4个位点偏离Hardy-Weinberg平衡。另外分析了这些引物在近缘种中的通用性情况,发现鳞砗磲中有7对微卫星引物具有通用性,6对具有多态性;无鳞砗磲中有3对微卫星引物具有通用性,1对具有多态性;诺瓦砗磲中有5对微卫星引物具有通用性,5对具有多态性;长砗磲中有9对微卫星引物具有通用性,8对具有多态性;砗蚝中有2对引物具有通用性,2对具有多态性。  相似文献

Coho salmon, Oncorhynchus kisutch (Walbaum), is intensively cultured in Chile. An increasing proportion of the eggs necessary to sustain the culture are locally produced by some hatcheries. However, there is no information about the origin or the genetic variability in these strains. The present study analysed allozymic variability and its distribution within and between some commercial strains of coho salmon in southern Chile. The genetic variability was estimated using horizontal starch gel electrophoresis. Samples of coho salmon were obtained randomly from four Chilean hatcheries. Twenty‐five enzymatic systems were examined, representing 51 enzymatic loci. Eight loci showed variability in at least one strain, which represented a total polymorphism (P) of 15.7%. Only the PGM‐1* locus was variable in all strains. The remaining loci were fixed in at least one strain. Total heterozygosity (HT) and within population heterozygosity (HS) were 0.35% and 0.36% respectively. The index of genetic diversity (GST) was 1.5%. The results confirm previous reports of low genetic variability in cultured strains of coho salmon in Chile, below that observed in their native range, which suggests loss of the genetic variability caused by genetic drift or other causes in these strains.  相似文献

We examined the amount and distribution of molecular variation at microsatellite loci in 21 broodstocks of Arctic charr Salvelinus alpinus derived from the Fraser River, Labrador and the Nauyuk Lake, Nunavut, Canada. Our goal was to assess the amount of genetic diversity and differentiation as broodstocks are subdivided and propagated in different hatcheries and grow‐out facilities. We observed significant heterogeneity across pairs of ancestral and descendant broodstocks in the mean numbers of alleles at microsatellite loci. We detected a significant decrease in the observed heterozygosity between ancestors and descendants but the amount of decrease did not depend on either the degree of removal from the wild (number of sequential transfers) or the strain (Fraser vs. Nauyuk). Based on allele frequency distributions, there was little genetic evidence of bottlenecks during the creation of subsequent broodstock populations after the initial founding events. All broodstock samples were significantly differentiated from each other but those within the same strain were more similar to each other than to broodstocks from different strains. Broodstocks from the Nauyuk Lake broodstocks showed greater differentiation from each other than did Fraser River broodstocks, which could be attributed to differences in the number of founders.  相似文献

Ten enzymatic systems were analyzed to determine allozyme genetic differentiation among three hatchery strains (A, B and C) of white shrimp, Penaeus (Litopenaeus) vannamei, commonly used in shrimp farming in northwest Mexico. A wild population from northern Sinaloa was used as a reference. Fifteen loci were detected, nine of which were polymorphic (ACP-1*, ACP-2*, AKP-2*, EST-2*, EST-3*, EST-4*, EST-5*, LAP*, and LDH*). Polymorphism of A, B and C were 53, 53, and 40%. The mean observed heterozygosity per locus was 0.071, 0.093, and 0.050 without any significant difference among them or with respect to the wild population (0.056). In all samples observed heteroxygosity was smaller than expected. The mean number of alleles per locus was 1.8 in A and 1.87 in both B and C. Strain A was the only sample without rare alleles. Only EST-3* and LAP* of strain A were in Hardy-Weinberg equilibrium; the other loci were in disequilibrium in all samples. High inbreeding values and heterozygote deficiencies were detected in all samples. Distribution of allelic frequencies was heterogeneous among the samples (G-test), involving all polymorphic loci and suggesting a genetic differentiation. According to Fst, a moderate genetic differentiation (7.4%) was detected among the samples. Greater differences were between strains A and C. Based on genetic distance, the samples were grouped into two pairs, B-C and A-wild. Strain A is a young strain related to the wild sample, whereas strains B and C have a different geographic origin than the wild sample.  相似文献

Molecular markers have significant potential for use in precise breeding programmes in aquaculture. This paper reviews the use of DNA markers to estimate inbreeding depression and heterosis in the guppy Poecilia reticulata. Full‐sib matings revealed that inbreeding causes declines in survival and salinity tolerance, but not in undwarfism, growth and high water temperature tolerance, indicating the effects of inbreeding differ among fitness‐related traits. Salinity tolerance was used to quantify the level of inbreeding depression and heterosis because the trait is strongly sensitive to inbreeding and shows a linear decrease with an increase in inbreeding coefficient. A positive correlation was observed between heterozygosity at microsatellite loci and salinity tolerance among 17 guppy populations. This indicates that heterozygosity estimated from microsatellites is a useful indicator for the estimation of inbreeding depression, suggesting that overall heterozygosity is important for fitness‐related traits that show inbreeding depression. Use of DNA markers to estimate the amount of heterosis in various strain combinations was examined using diallele and reciprocal crosses among four domestic strains. The amount of heterosis differed among the strain combinations and correlated with Nei's genetic distance measured by microsatellites and also by dissimilarity using random amplified polymorphic DNA (RAPD) analysis. This indicates that microsatellite and RAPD markers are useful for estimating the amount of heterosis in various strain combinations, further suggesting that the amount of heterosis depends on the genetic differences between the strains. The present study showed that DNA markers are useful tools for estimating inbreeding depression and heterosis in guppy breeding.  相似文献

Growth and variation in size of turbot and sole larvae originating from different genetic sources were compared at different stocking densities, including genetically based information that might lead to a better knowledge of heterogeneity in cultivated populations. There was no effect of density on growth rate in either species. However, between the two sole batches there was an earlier metamorphosis of faster growing fish from Norway. In sole groups after metamorphosis, variation was stable and therefore no development of size variation was observed. There was no density effect on size variation within turbot batches before, or after metamorphosis. Spreading rate and growth rate were positively correlated in sole larvae indicating that growth of the smaller fish was not equal to the larger ones, but size variation followed a different pattern in turbot. No evidence for either overall significant heterozygosity excess, or deficiency was evidenced in the farmed strains of either species . However, significant differences between farmed populations in terms of genetic variability (θ) were reported, which was rather attributed to within differentiation than between strains. One could safely argue that different strains of Dover sole originated from different genetic sources (factorial analysis). Moreover, when the analysis was extended to individual loci to assess whether specific loci had any consistently larger, or smaller effect on heterozygosity, locus specific significant trends were discerned in turbot. A positive Heterozygosity Fitness (growth rate) Correlation (HFC) suggests that heterozygotes reported enhanced growth rates compared with homozygous ones in flatfish larvae, thus special precautions should be taken into account whenever different genetic structure patterns emerge, because of various flatfish broodstock geographical origin and year class samples.  相似文献

为研究中国沿海地区棘头梅童鱼群体的遗传多样性,利用微卫星标记技术,采用9对微卫星引物对中国连云港(LYG)、大丰(DF)、崇明(CM)、舟山(ZS)、温州(WZ)、宁德(ND)、厦门(XM)棘头梅童鱼7个野生群体的遗传多样性和遗传结构进行了分析。结果显示,实验检测到63个等位基因,各位点等位基因数为3~13个,有效等位基因数为1.7510~8.0317;各位点的观测杂合度(Ho)为0.3596~0.7854,期望杂合度(He)为0.4300~0.8780;多态信息含量(PIC)为0.3604~0.8631,其中有2个位点表现为中度多态(0.25PIC0.5),7个位点表现为高度多态(PIC0.5),具有较丰富的遗传多样性水平。Hardy-Weinber平衡分析显示,7个群体的大部分位点未偏离平衡。基于群体间Nei氏标准遗传距离构建的7个野生群体UPGMA系统进化树结果显示,ND和WZ群体遗传关系最近,ZS和WZ群体遗传关系最远,WZ和ND聚为一支,但总体上没有显著的遗传分化。  相似文献

本文利用15对微卫星分子标记,对德国镜鲤选育系F4天津群体中的高背型和长条型德国镜鲤的遗传结构进行分析,结果表明:两种体型德国镜鲤F4群体平均等位基因数分别为5.0和5.1,平均期望杂合度分别为0.5527和0.5591.平均观测杂合度分别为0.5906和0.5824,平均多态信息含量分别为0.5653和0.5470。以卡方检验估计群体Hardy—Weinberg平衡显示有大部分位点发生了偏离。两体型间遗传相似性为0.8330,相似性较高。群体间遗传分化微弱(Fst=0.0242),群体内变异占总变异的97.58%。说明高背型德国镜鲤和长条型德国镜鲤在遗传结构上差异较小。由天津群体的等位基因数为5.05,期望杂合度为0.5559,观测杂合度为0.5865,多态信息含量为0.5562,可知天津群体属于中度多态,遗传多样性保持较高水平。  相似文献

为了探讨壳白长牡蛎人工选育对群体遗传变异的影响,实验利用4个多重PCR组合共10个微卫星标记分析了连续3代壳白长牡蛎人工选育群体和野生群体及基础群体的遗传多样性。结果发现,6个群体的平均等位基因数量为7.2~12.6,等位基因丰度为6.8~11.0,期望和观测杂合度分别为0.672~0.769和0.486~0.542;与野生群体相比,3代选育群体的平均等位基因数显著降低,但平均期望杂合度并无显著差异。哈迪—温伯格平衡检验结果显示,在60个群体—位点组合中有39个群体—位点组合显著偏离哈迪—温伯格平衡,近交系数F_(is)范围为0.215~0.342。群体间遗传分化指数F_(st)范围为0.005~0.076,处于中—低等的遗传分化水平。研究表明,虽然连续选育对群体的遗传多样性和遗传分化造成了一定程度的影响,但人工选育群体依然表现为较高的遗传多样性,仍可以一定的选择压力对选育群体进行人工选育。  相似文献

采用水平凝胶电泳技术对星突江鲽(Platichthys stellatus)养殖群体进行了同工酶分析。筛选了9种酶,检测到15个位点,在0.99水平上,仅PGM^*位点表现为多态,LDH^*、SDH^*、G3PDH^*、GPI-1^*、GPI-2^*、GPI-3^*、IDHP^*、SOD-1^*、SOD-2^*、SOD-3^*、MDH-1^*、MDH-2^*、MPI-1^*和MPI-2^*均为单态,多态位点比例为6.67%,实际观测杂合度为0.0098,预期杂合度为0.0303,平均有效等位基因数目为1.0557。结果表明,星突江鲽养殖群体的遗传多样性水平较低。  相似文献

An allozyme survey was conducted in three naturalized populations of rainbow trout, Oncorhynchus mykiss (Walbaum), from rivers in two Andean lakes of southern Chile. The analysis was based on 32 presumed loci, 10 of which were variable, and revealed that the mean percentage of polymorphic loci (P= 21.8%; range = 15.6-28.1%), the mean number of alleles per locus (n=1.26; range = 1.2-1.3) and the mean observed heterozygosity (HO=7.0%; range = 6.0-8.2%) are high, although within the range reported for rainbow trout. Although the mean genetic distance between the samples was low (D = 0.003), and only 5.2% of the total observed genetic variation can be accounted by interpopulation differences (FST= 0.052), eight out of 10 polymorphic loci exhibited significant allelic heterogeneity. Some loci, notably EST-1*, IDHP-3,4*, MEP-1* and PGM-1*, showed considerable discriminatory capacity among the samples. Rainbow trout has a longer history in Chilean waters since salmonid introduction started earlier this century. Under current intensive farming practices, the preliminary information provided in the present paper is likely to be helpful in setting up a baseline for further characterization and/or monitoring of naturalized populations.  相似文献

Several tilapia species in Mexican reservoirs have been used as food source for more than 40 years but little effort has been devoted to the conservation and genetic selection of those species. Our objective was to evaluate genetic variability in feral (n = 59) and hatchery-produced (n = 58) Oreochromis niloticus stocked in the Benito Juarez dam. Allozyme analyses for 13 enzymatic systems and general proteins yielded 27 genetic loci. Seventeen loci were polymorphic in at least one sample. The observed heterozygosity was lower than expected and the imbalance in the Hardy–Weinberg equilibrium in almost all loci was caused by heterozygote deficiency. The inbreeding coefficient was 84%. Although the evidence suggests moderate genetic differentiation, it is probable that a great proportion of the source of hatchery organisms is from feral organisms. According to our results, management of the hatchery strain is inadequate, therefore other breeding strategies are needed to increase the genetic variability of O. niloticus from Oaxaca.  相似文献

运用微卫星DNA技术对仿刺参烟台群体、威海群体、大连群体的3个野生群各20个个体进行了遗传分析。对9个基因位点进行了扩增,共获得了59个等位基因。评估了9对微卫星引物的多态信息含量(PIC),范围在0.5129~0.8794之间。结果表明:3个野生群体的平均杂合度观测值分别为0.6416、0.6595、0.5824,平均杂合度期望值分别为0.7641、0.7161、0.7364;在Hardy-Weinberg平衡条件下,进行了P检验,发现3个群体均有位点发生了显著偏离;对3个群体进行了配对Fst值计算,发现3个群体之间遗传分化较弱。从变异贡献率来看,95.68%的变异来自个体之间,4.32%的变异来自群体之间。经过聚类分析,发现威海群体和大连群体之间亲缘关系较近,烟台群体与前两群体亲缘关系较远。  相似文献

采用不连续聚丙烯酰胺凝胶垂直电泳技术对两种壳色虾夷扇贝的闭壳肌进行了同工酶检测分析与比较。8种同工酶(MDH、ADH、ME、SOD、EST、GDH、SDH和α-AMY)共检测出17个位点,其中白色贝有7个多态位点,褐色贝有6个多态位点,多态位点(P0.99)的比例分别为41.18%和35.29%,白色贝和褐色贝平均每个位点的等位基因有效数目(Ac)分别为1.1421和1.1040,预期杂合度分别(Hc)为0.0919和0.0674,实际杂合度(Ho)分别为0.1275和0.0907,多态位点的遗传偏离指数表明,白色贝具有更高的遗传变异水平和多样性水平。住点Est-3可以作为鉴定白色贝和褐色贝的特异性蛋白质分子标记。  相似文献
