Physicochemical changes of Japanese cockle foot parts occurring during cooking and cold storage at 4°C for 10 days were examined. Raw samples decreased in weight by 27% during cooking at 85°C for 15 s. Both raw and cooked samples decreased in weight to the same extent of 22–23% during the 10-day cold storage. The levels of ATP and related compounds, as well as those of free amino acids, were higher in raw than in cooked samples, although the former were found by sensory evaluation to have a stronger fishy odor. Raw samples started to decrease in ATP content after 5 days of cold storage, showing K' values of 18 and 42% after 5 and 7 days, respectively. The L* value in the Hunter scale color evaluation increased during the cold storage of raw samples, revealing a gradual disappearance of its original purple–gray color. Meanwhile, the L* value of cooked samples remained constant during the cold storage. The tensile strength of cooked samples was significantly higher than those of both raw samples before and after the cold storage, indicating increased toughness by cooking.  相似文献

ABSTRACT

To understand biochemical characteristics, storage stability, and freshness indicators of white shrimp (Litopenaeus vannamei), changes in extractable nitrogenous compounds, microbial count, and sensory rating of white shrimp during storage at 25 and 4°C were investigated. Free amino acids showed a slow increase during storage at 25°C, but no obvious change was found at 4°C. Adenosine triphosphate (ATP) and adenosine diphosphate (ADP) were found at initial stage and decreased rapidly after storage. Both inosine 5′-monophosphate (IMP) and adenosine monophosphate (AMP) increased and then decreased during storage. Inosine, hypoxanthine, and the K-value gradually increased with time. The levels of total volatile basic nitrogen (TVB-N), NH3, and trimethylamine (TMA) of white shrimp increased with storage time at 25 and 4°C. The TVB-N, NH3, TMA, inosine, hypoxanthine, and K-value could be considered as freshness indicators of white shrimp during storage. However, the total plate count did not corroborate the acceptability recommended limits for white shrimp during storage. The sensory evaluation, associated with TVB-N, TMA, and K-value, showed the quality was unacceptable after 6 h storage at 25°C and 7 days at 4°C.  相似文献

ABSTRACT:   Small abalone meats were heated at 80°C and 98°C for 0–120 min and the differences in chemical, physical and sensory changes of the cooked meats were investigated. The decrease in moisture and weight and the increase in browning and Hunter's b -value were relatively higher for cooking at 98°C than at 80°C. After cooking for 20–120 min, the total amount of adenosine triphosphate and its related compounds on a dry weight basis decreased by 17–27% at 80°C and by 30–39% at 98°C; the total amount of free amino acids on a dry weight basis changed insignificantly at 80°C but decreased by 22–35% after cooking at 98°C. The meats cooked at 80°C were higher in cutting force whereas the levels in the samples cooked at 98°C did not decrease until samples had been cooked for 60 min. The hydroxyproline content showed little change during cooking except for in samples cooked at 98°C for 120 min, in which the content was found to be low. The extended cooking at 80°C improved the acceptability of small abalone meat, whereas only the acceptability score of aroma increased significantly for cooking at 98°C.  相似文献

ABSTRACT:   The effect of storage temperature (0, 5, 10, and 15°C) and killing procedure (instant, struggled, temperature shocked, and spinal cord destruction killing) on post-mortem changes in the muscle of the horse mackerel caught near Nagasaki, Japan, was investigated. Temporal changes in adenosine triphosphate (ATP), inosine monophosphate (IMP), and lactic acid concentrations were slowest at 10°C storage temperature. The increase in K -values was slower at 10°C and below than at 15°C storage temperature. In addition, 10°C storage temperature was most suitable for maintaining a constant breaking strength value of the muscle. Regarding the effect of killing procedure, temporal changes in ATP, IMP, and lactic acid concentrations were slowest when killed by spinal cord destruction. The increase in K -value and the decrease in breaking strength were also slowest in the spinal cord destruction group. From these results, it was considered that storage at 10°C temperature and spinal cord destruction killing procedure were most effective in delaying post-mortem change in the horse mackerel.  相似文献

将池塘捕获的体重(1.70±0.80)kg的草鱼(Ctenopharyngodon idella)饲养在室内水族箱内,分别测定了空腹(禁食1 d,26样本)、饥饿15 d(15样本)、饥饿30 d(15样本)的草鱼血清游离氨基酸含量。结果显示:草鱼血清游离氨基酸总量基本恒定,空腹、饥饿15 d和饥饿30 d的氨基酸总量无显著差异;处于饥饿状态的草鱼血清游离必需氨基酸总量极显著地高于空腹状态,非必需氨基酸总量极显著地低于空腹状态;血清赖氨酸、缬氨酸、异亮氨酸、亮氨酸和胱氨酸含量因饥饿显著升高,而组氨酸、苯丙氨酸、天冬氨酸、丝氨酸、谷氨酸和酪氨酸因饥饿显著降低;饥饿15 d和饥饿30 d各氨基酸含量均未见差异。草鱼血清游离氨基酸含量在正常摄食情况下主要受饲料蛋白质影响,处于饥饿状态下主要来源于肌体蛋白质的分解,比较正常摄食后和饥饿状态下血清游离氨基酸的组成可评价饲料蛋白质的质量。  相似文献

We report the effect of salinity and temperature on the viability of stored culture-based subitaneous eggs of the calanoid copepod Acartia tonsa for use of copepods in fish larvae culture. Quiescence induction was recorded at 17 and 25 °C, in salinities from 0 to 30. Quiescence was strongly induced at 0 salinity and partially at 5 in both temperatures. Eggs incubated at 0 salinity for up to 12 days at both temperatures showed a decline in the fraction able to be induced into quiescence by abrupt salinity changes. The hatching success of eggs that were able to enter quiescence stabilized after a 1-day incubation and remained ∼25% viable for 12 days in 17 °C. On the contrary, the 25 °C trial showed a gradual decline in viability until stabilizing ∼10% at day 7 and onwards. Longterm 17 °C incubation for 35 days showed that eggs remained quiescent with a viability of ∼14%. Hence, we recommend salinity storage of A. tonsa subitaneous eggs as a relevant shortterm technique, and a suitable alternative to the recently proposed cold storage of eggs when eggs are to be shipped from the copepod producer to a given fish larvae hatchery.  相似文献

ABSTRACT:   Tropical minute rotifer strains (SS-type) induce mixis at 30–35°C but sexual reproduction and resting egg formation do not proceed well due to rapid environmental change. The present study examined the effect of temperature regulation on rotifer Brachionus rotundiformis (Langkawi strain, SS-type) resting egg formation in small (500 mL in culture volume)- and large-scale (500 L in culture volume) experiments. Rotifers were cultured at 30°C in 15–17 p.p.t. seawater with an initial density of 1 individual (ind.)/mL. After 4 days, when cultures were in exponential growth stage with active mixis induction, the culture temperature of the experimental rotifers was changed to 25°C. Control rotifers were cultured at 30°C throughout the experiment. Fresh or frozen Nannochloropsis oculata and condensed freshwater Chlorella vulgaris were used as the rotifer diets in the small- and large-scale experiments, respectively. Significantly higher resting egg production was observed with the experimental rotifers (30→ 25°C) versus the control rotifers. In the large-scale trial, experimental rotifers produced 2.6 × 106 resting eggs during a 9-day experiment, which was 1.6-fold more than the control rotifers. Moreover, the efficiency of resting egg formation was found to increase by a factor of 1.8. The present study indicates that decreasing culture temperature from 30 to 25°C after active mixis increased resting egg formation in B. rotundiformis (SS-type).  相似文献

Nuclear magnetic resonance ‐based metabolomics was applied to study effects of egg aging on ovarian fluid metabolites in rainbow trout, Oncorhynchus mykiss. The eggs of three females were pooled and then assigned to three plastic vials for 18 days in vitro storage at 4°C. Ovarian fluid samples were taken 0, 6, 12 and 18 days after storage. Three groups of metabolites including amino acids, osmolytes and energy metabolites were found to change during storage period. The glucose levels of ovarian fluid showed significant decreases on days 12 and 18 after storage (< .05). For acetoacetate and acetate, significant increases were observed, respectively, on days 12 and 18 after storage (< .05). The creatine levels of ovarian fluid increased significantly on days 12 and 18 after storage (< .05). Lactate levels in ovarian fluid elevated during storage period (< .05). Glycerol levels showed a significant increase on days 12 and 18 after storage (< .05). The values of osmolytes in ovarian fluid (e.g., betaine, taurine, trimethylamine‐N‐oxide, N,N‐dimethylglycine) showed a decreasing trend on day 12 which continued until the end of storage on day 18 (< .05). Almost all amino acids elevated on days 12 and 18 after storage (< .05). After an apparent elevation in isoleucine levels on days 6 and 12, this amino acid decreased on day 18 after storage (< .05). The osmolytes might act as antioxidant against free‐radicals produced as a result of over‐ripening. Glucose can be used as energy resource for eggs and bacteria during ova storage. Also, change pattern of amino acids indicate hydrolysis of proteins as the time of storage increases.  相似文献

Atlantic salmon (Salmo salar L.) with an average weight of 411 ± 16 g were fed after a period of 7 days without food and the free amino acid concentrations in the pylorus, liver and white muscle measured before and at 3, 6, 9, 15 and 24 h after feeding. There were few significant postprandial changes in tissue free amino acid concentrations. In the white muscle, concentrations of six indispensable amino acids were significantly (p < 0.05) higher after 9 (Ile, Leu, Phe, Thr) or 15 h (Val, Met). Individual feed intake was measured and there were significant positive correlations between amino acid intake and amino acid concentrations in white muscle free pools for total amino acids (p < 0.001), total indispensable amino acids (p < 0.001) and individual indispensable amino acids (Ile, Leu, Lys, Met, Phe, Val). These relationships were due to relatively low feed intake (0.28% body weight) that followed 7 days without feeding. The indispensable amino acid profile of the white muscle free pool was compared with that of standard proteins (the feed, whole body and white muscle), as well as with indispensable amino acids requirements. At different times one of two indispensable amino acids, Phe (at 0, 3, 6, 9, 15 h after feeding) or Trp (at 24 h after feeding), was present at the lowest relative concentrations compared to the other indispensable amino acids. This showed that although changes in tissue free amino acid concentrations following feeding were small the amino acid profile (relative concentrations) in the white muscle free pool changed. It is proposed that the lowest relative concentration of an indispensable amino acid in the white muscle free pool should be considered in relation to its potential to limit the efficiency of protein synthesis and retention.  相似文献

ABSTRACT: An ice-nucleating bacterium, designated MACK-4, was isolated from ice-stored mackerel ( Scomber australasicus ) and identified as a Pseudomonas fluorescens . The optimal temperature and pH for its growth in nutrient broth with 2.5% glycerol (NB-G) were 15°C and 6.5, respectively. The maximal ice-nucleating activity (INA) was obtained after 54 h incubation at 15°C. However, the INA was almost completely lost after 48 h incubation at 25°C or higher. The growth and INA decreased with increase of NaCl added in NB-G within 0.0–4.0%. The INA of MACK-4 was very stable at 5–25°C, pH 4.0–9.5, while that of isolated ice-nucleating matter from MACK-4 was stable at 5–25°C, pH 5.5–9.0.  相似文献

ABSTRACT: For the development of a stepwise cryopreservation technique for larvae of the Pacific oyster Crassostrea gigas , various conditions were examined. Larvae at 9, 12, 15, 18 and 21 h after insemination were cooled at a rate of −1°C/min (seeding at −8°C for 15 min) and then plunged into liquid nitrogen at −35 or −40°C using 1.5 M dimethyl sulfoxide (DMSO) and 250 mM trehalose as cryoprotectants. Among these larvae, 15 h after insemination (the trochophore stage before formation of the shell gland) showed the highest motility and the best external appearance after thawing. Trochophore larvae were cryopreserved in preservation media containing different dilutions (1/4, 1/6, 1/8, 1/10 and 1/30) of seawater. Larvae preserved in the 1/4 seawater medium showed the highest appearance of shelled larvae 4 days after thawing. Trochophore larvae reared in seawater at 21, 25 or 29°C were cryopreserved for 8 months and then reared at 26°C after thawing. Larvae reared at 25°C showed the highest survival rate and normal larval ratio at day 6 after thawing, although larvae reared at 21°C showed the highest rates until day 4. Abstract. At 9°C juvenile sockeye salmon, Oncorhynchus nerka , inoculated intraperitoneally with 105 Cryptobia (= Trypanoplasma ) salmositica began to succumb about day 15, with mortalities exceeding 90% by day 28. Fish inoculated at 8°C and acclimatized to 13°C began to die at about the same time but total mortalities were only about 75%. At 5°C the infection progressed more slowly, with the first mortalities occurring about day 25, but the disease seemed equally as lethal as at 9°C. All fish inoculated at 8°C and acclimatized to 20°C within 11 days survived the infection. Changes in salinity from fresh to sea water (30%o salinity) at 9°C or 13°C had almost no effect on the course of infection either when presmolts were slowly acclimatized to sea water over a period of 6-8 days or when smolts were transferred to sea water quickly (within 1-2 days). However, there were differences in susceptibility to disease among three species of salmon of smoking age. The order of decreasing sensitivity was chinook salmon, Oncorhynchus tshawytscha , with 100% mortality, sockeye salmon with 56 to 74% mortality and coho salmon, Oncorhynchus kisutch , with no mortality.  相似文献

Thirty percent flour was substituted with dolphinfish (Coryphaena hippurus) and unicorn leatherjacket (Aluterus monoceros) paste in a cake recipe to evaluate the quality and shelf life of fish chiffon cakes stored at 4℃ and 25℃. Proximate composition, free amino acid (FAA), aerobic plate count (APC), total volatile basic nitrogen (T-VBN), and texture profile and sensory evaluation of fish chiffon cakes were conducted. Fish paste addition increases proteins. Cakes made with dolphinfish paste had the highest FAA content, followed by the cake made with unicorn leatherjacket. Histidine is the predominant FAA in dolphinfish chiffon cake, and it is significantly higher than the other two cakes. T-VBN values and APC increased with the addition of fish paste and was lower than the upper limit of acceptable fish products during storage period. The hardness of the two fish cakes was slightly lower than the control after 1 and 2 days storage at 25℃ or 3 and 5 days storage at 4℃ and increased during storage. Sensory evaluation showed no significant differences among the ordinary cake and two fish chiffon cakes for odor, texture, flavor, and overall acceptability. Therefore, incorporation of white fish meat is feasible for chiffon cake.  相似文献

The use of live prey is still necessary for a large number of raised fish species. Small sized rotifers are usually used as live preys during the first days of feeding in small mouth fish. An alternative to this is the use of copepods as prey for first feeding. In this study, the sizes, weight and biochemical contents of two copepods and one rotifer species raised on similar algal food conditions were compared. Rotifers contained a higher proportion of essential amino acids in the free amino acid (FAA) fraction (43%) than copepods (30–32%). However, rotifers had lower levels of important fatty acids like DHA (7% compared with 23–32% in copepods) and their DHA/EPA ratio was lower than that in copepods (0.54 compared with 1.35–1.63 in copepods). The FAA pattern of the preys tended to be species‐specific and its implications from an aquaculture point of view is discussed. In contrast, the‐protein bound amino acids tended to be very conservative among the studied organisms. The second part of the work is focused on ‘the price’ of hatching in Acartia tonsa eggs before or after cold storage at 3°C. The fatty acid contents in A. tonsa tended to decrease with the storage time. It also decreased with hatching of the nauplii, but its proportion compared with the dry weight remained constant.  相似文献

Thermal stress responses of a sterile mutant of Ulva pertusa (Chlorophyta)   总被引:1,自引:0,他引:1  
SUMMARY: The thermal stress responses of a sterile mutant of the marine alga Ulva pertusa were investigated at 20°C and 30°C. The amounts of the photosynthetic pigments, β-carotene, chlorophylls a and b , lutein, neoxanthin, and violaxanthin, were 1.4–2.4 times higher in the 30°C-cultivated alga than in the 20°C-cultivated alga. The free amino acids, asparagine, aspartic acid, glutamine, glutamic acid, glycine, and serine, were abundant in the 20°C-cultivated alga, and increased 1.9–10.5-fold in response to thermal stress (30°C). Total carbon and nitrogen contents also increased in the 30°C-cultivated alga. Sodium dodecylsulfate-polyacrylamide gel electrophoretic patterns of total proteins extracted from both temperature-treated algae showed the increases of 20, 25, and 90 kDa proteins in the 30°C-cultivated alga. Isozyme assays for 20 enzymes showed a different banding pattern only in the case of glutamate dehydrogenase (GDH). Although it was observed that both temperature-treated algae possessed NAD+- and NADP+-specific GDH, the 30°C-cultivated alga had an additional NADP+-specific GDH (NADP-GDH). These results suggest that NADP-GDH compensates for the thermally induced decreases in nitrogen assimilation efficiency and thereby regulates nitrogen metabolism under conditions of temperature stress.  相似文献

The effects of size of fish on composition and of time in storage at -18°C on lipid and sensory changes for channel catfish, Ictalurus punctatus, were investigated. Dressed large (1.0 kg) fish (LF) averaged 13.2% fat, while dressed small (0.3 kg) fish (SF) averaged 10.8% fat (P < 0.05). Moisture was lower (P < 0.05) for LF (68.1 vs. 70.8%). Thiobarbituric acid (TBA) values for LF increased during the first ten months and dropped during the 12th month, while TBA values reached their highest level after the 8th month for SF. Free fatty acids did not change for LF but increased (P < 0.05) throughout storage for SF. Lipids for both sizes of fish were 25% saturated, 58% monounsaturated, and 17% polyunsaturated fatty acids. C 14:0, C 18:0, and C 18:3 acids decreased after 12 months of frozen storage. LF were rated higher (P < 0.05) in flavor for the first four months but were not rated significantly different from SF thereafter. These results indicate that although LF contained more fat, they did not oxidize any more than did SF, and both sizes were acceptable to panelists throughout the study. Thus, if channel catfish is handled and stored properly regardless of size, it should be acceptable even after 12 months frozen storage.  相似文献

以南极磷虾虾(Euphausia superba)肉糜为原料,通过测定南极磷虾虾肉糜在反复冷冻和解冻过程中ATP及其关联产物、游离氨基酸等的变化,分析冻融循环对虾肉糜滋味成分的影响。结果显示,随着冻融循环次数的增加,南极磷虾虾肉糜的ATP及其关联产物降解加剧,呈鲜味的核苷酸含量逐渐减少,呈苦味的核苷酸含量逐渐增加。1次冻融对应的鲜味苦味比为3.32;2次及以上的冻融循环会造成鲜苦比的显著降低(P<0.05),同时K值迅速增大,表明鲜度发生大幅下降;3次冻融循环后,K值显著增大(P<0.05);4次冻融对应的K值为(86.88±3.58)%,南极磷虾虾肉糜已不新鲜。冻融循环次数的增加导致游离氨基酸总含量增加,鲜味氨基酸和甜味氨基酸比例逐渐减小,苦味氨基酸的比例逐渐增大,导致虾肉糜的滋味进一步劣化。研究表明,冻融循环次数控制在2次以内,可较好地保持南极磷虾虾肉糜的滋味。  相似文献

ABSTRACT:   The optimal water temperature in seed germination and the upper critical water temperature in seedling growth were determined for Zostera marina collected from Ise Bay, Japan. The relationship between the seed germination rates and seed storage period (0, 30 and 60 days at 0°C) was also examined. The optimal water temperature for seed germination was in the range from 10 to 15°C regardless of the storage periods, in which germination rates ranged from 35 to 57%. Seedlings grown from seed up to 10 cm in total length were cultured for 1 week under various water temperatures to measure their relative growth rates. The optimal water temperature in growth was in the range from 20 to 25°C; relative growth rates ranged from 2.0 to 2.6%. Seedlings could survive up to a water temperature of 28°C, but most seedlings withered at 29 or 30°C. The optimal water temperatures for seed germination and seedling growth were related to the seasonal changes of water temperature at the sampling site. Although seedlings were rarely observed in the field in summer, they can grow at temperatures as high as 28°C. Therefore, Z. marina may extend its distribution as far as where the summer water temperature is lower than 28°C.  相似文献

During refrigeration, lipid oxidation is a major factor contributing to post-mortem deterioration of flesh quality. Polyunsaturated fatty acids (PUFA), especially n -3 PUFA, are present in high concentration in fish tissues, and in oils used in diets, and are readily susceptible to peroxidation. α-Tocopherol (AT) can reduce tissue lipid peroxidation in vivo and post mortem. The effect of increasing the tissue level of AT by dietary supplementation of α-tocopherol acetate (ATA) was therefore investigated. Commercial salmon diets C, M and H, high in lipids, containing 184, 573 and 865 mg ATA kg−1 diet DM (dry matter) were fed to 18 fish per treatment. Dietary AT: PUFA ratios were 2.0, 6.3, and 9.5 mg g−1 for diets C, M and H, respectively. Fish (mean initial live weight 630 g) were slaughtered after 50 and 78 days of feeding. Fillet samples were analysed fresh or after storage at 4 °C for 12 days and –20 °C for 12 months. Lipid oxidation was measured using the thiobarbituric acid test. Colour score, but not carotenoid content, of fillets was significantly higher between 6 and 12 days of fresh storage in fish fed diets M and H compared with those fed diet C. Colour score, carotenoid content and ΑΤ content decreased and the content of lipid oxidation products increased following storage of fillets at –20 °C for up to 12 months, although lipid oxidation was always significantly lower in fish fed diets M and H.  相似文献

ABSTRACT:   To elucidate the utilization of the major yolk nutrient stocks in eggs and larvae of walleye pollock Theragra chalcogramma , the contents of free amino acids (FAA), the major yolk protein (180 kDa lipovitellin originated from vitellogenin B in ovulated eggs: oLv B), and lipids were measured. Most eggs hatched 18 days after fertilization at 5°C, and all larvae absorbed almost all their yolk mass by 28 days. The total FAA content showed no change during the first 6 days, and then decreased to 28% of the initial level by 18 days. The oLv B contents, measured by an enzyme-linked immunosorbent assay using a specific antiserum against oLv B, gradually decreased from 6 to 18 days, followed by a rapid decline. The content of phospholipids (PL) and triacylglycerols (TG) showed no marked change until hatching, and then decreased until disappearance of yolk sac. From these results, it is proposed that there are two main periods for nutrient utilization in embryos and larvae of walleye pollock. In the first period, FAA was mainly utilized until 18 days after fertilization. Active utilization of oLv B and lipids (PL and TG) instead of FAA occurred during the second period from 18 to 28 days.  相似文献
