本研究以鸡传染性支气管炎病毒(infectious bronchitis virus,IBV)S1基因为研究对象,设计特异性引物扩增S1抗原集中区目的基因,长度为198 bp,亚克隆至原核表达载体pGEX-6P-1,构建重组原核表达质粒pGEX-6P-1-S1,转化至宿主菌Rosetta(DE3)后进行诱导表达。SDS-PAGE分析结果显示,表达获得1条特异性蛋白条带,其分子质量大小为33.0 ku,与预期蛋白大小一致。可溶性分析结果表明目的蛋白以包涵体形式存在。Western blotting结果显示,重组蛋白能与IBV鸡阳性高免血清反应,被GST标签单抗识别,结果表明该融合蛋白正确表达并具有良好的抗原性。本研究为制备IBV单克隆抗体奠定了抗原基础。  相似文献

为获得鸡传染性支气管炎病毒(Avian infectious bronchitis virus, IBV)N蛋白的单克隆抗体,通过原核表达IBV N蛋白,纯化后作为免疫原免疫BALB/c小鼠,并按常规方法制备杂交瘤细胞。经ELISA方法筛选阳性杂交瘤细胞,经过3次亚克隆获得3株杂交瘤细胞株,命名为1#、18#、19#,并进行了抗体亚类的鉴定、Western-blot和IFA检测。结果显示:制备的3株单克隆抗体亚型均为IgG1,Western-blot和IFA试验结果表明单克隆抗体均能与IBV发生特异性反应而与其他禽病常见病毒均无交叉反应。本研究成功制备了IBV单克隆抗体,为进一步建立IBV检测方法和深入研究IBV的生物学特性奠定了基础。  相似文献

为筛选日本乙型脑炎病毒(JEV)的E抗原表位,本实验以抗JEV E蛋白的单克隆抗体(MAb)作为固相筛选分子,应用噬菌体表面展示技术,按消减、结合、洗脱、扩增的顺序筛选噬菌体七肽库,挑取噬菌体单克隆培养并采用MAb包被的ELISA鉴定,对阳性克隆测序分析,确定JEV E抗原模拟表位的氨基酸序列.设计合成包含该表位的E抗原15肽(E-365GGADSMSMAGMAVSYE-379)cDNA序列,与pGEX-KG构建重组表达质粒,诱导表达重组多肽并进行western blot验证.经过4轮筛选后,噬菌体得到高度富集,挑取单克隆采用MAb包被进行ELISA鉴定,有22个克隆呈阳性.对重组多肽进行western blot验证,结果表明该重组多肽能够特异结合兔抗JEV多克隆抗体.本实验成功筛选出JEV结构蛋白E的特异性噬菌体模拟表位,为开展用JEV抗原表位探索JEV的防制研究创造了条件,为多肽疫苗研制、药物筛选以及特异血清学诊断方法的建立提供重要依据.  相似文献

Using the whole infectious bronchitis virus (IBV) for detecting the antibody against IBV by enzyme-linked immunosorbent assay (ELISA) is a routine work in poultry industry. To prepare virus is time consuming and tedious. Furthermore, the whole viral antigen detects all antibodies against the viral structural proteins, including spike (S), nucleocapsid, matrix, and other proteins. Among those, S protein is related to neutralization. Thus, to develop and express protein fragment from S gene and to use the protein as a coating antigen for antibody detection against IBV are the purposes of this experiment. A partial S gene fragment (n.t. 1143-1665) was cloned into pRSET vectors and transformed into competent Escherichia coli (E. coli) BL21 (DE3). A 27.5 kDa fusion protein (S-fg, containing S1-F and partial S2-G antigenic sites) was successfully expressed, affinity-purified and detected specifically with chicken anti-IBV serum by Western blot. The expressed S-fg protein was used as a coating antigen for developing an ELISA (S-fg ELISA) for serum antibody detection in anti-IBV antisera from different IBV serotypes and in field sera. The results show that the S-fg fusion protein is highly cross-reactive among different IBV serotypes, and the S-fg ELISA is found to be a convenient, economical, and efficient method for antibody detection against IBV.  相似文献

将传染性支气管炎病毒(IBV)ZJ971 S1基因亚克隆到绿色荧光蛋白(GFP)表达载体pEGFP—C2中,成功构建重组表达质粒pEGFP—ZJ971-S1。重组质粒在脂质体的介导下转染Vero细胞,借助荧光显微镜在转染后4h观察到S1—GFP融合蛋白的瞬时表达。免疫细胞化学染色(ICC)结果显示,抗ZJ971 S1D蛋白单克隆抗体和鸡抗IBVZJ971全病毒血清特异性识别了S1基因转染细胞,表明S1蛋白在Vero细胞中得到有效表达。荧光显微镜观察和ICC均表明,S1表达蛋白主要分布在转染细胞的胞浆内,而胞核中未见分布,提示IBVS1蛋白内可能存在与病毒装配相关的细胞定位信号。  相似文献

Infectious bronchitis virus (IBV) was detected by indirect immunofluorescent assay with a monoclonal antibody (MAb-IFA). The monoclonal antibody was specific for the nucleocapsid protein of IBV strain M41. The MAb-IFA clearly detected IBV with high specificity in infected chicken kidney cells. The assay furthermore detected IBV in tracheal smears and sliced tracheas from experimentally infected chickens. The positive reaction was found to be longer than that in the virus recovery test. These results indicate that MAb-IFA is a useful method for the detection of IBV from chickens suspected to have infectious bronchitis.  相似文献

Infectious bronchitis virus (IBV) poses a severe threat to the poultry industry and causes heavy economic losses worldwide. Vaccination is the most effective method of preventing infection and controlling the spread of IBV, but currently available inactivated and attenuated virus vaccines have some disadvantages. We developed a chimeric virus-like particle (VLP)-based candidate vaccine for IBV protection. The chimeric VLP was composed of matrix 1 protein from avian influenza H5N1 virus and a fusion protein neuraminidase (NA)/spike 1 (S1) that was generated by fusing IBV S1 protein to the cytoplasmic and transmembrane domains of NA protein of avian influenza H5N1 virus. The chimeric VLPs elicited significantly higher S1-specific antibody responses in intramuscularly immunized mice and chickens than inactivated IBV viruses. Furthermore, the chimeric VLPs induced significantly higher neutralization antibody levels than inactivated H120 virus in SPF chickens. Finally, the chimeric VLPs induced significantly higher IL-4 production in mice. These results demonstrate that chimeric VLPs have the potential for use in vaccines against IBV infection.  相似文献

In this study,IBV HH06 complete E gene was firstly cloned and sequenced.According to the hydrophilicity and antigenic index analysis,its partial gene (193 to 327 bp) was subcloned into prokaryotic expression vector pET-32a(+) and eukaryotic expression vector PVAX1.The recombinant plasmid pET-32a-E1 was transformed into E.coli Rosetta (DE3) and induced with IPTG.The recombinant IBV truncated E1 protein with molecular weight of 23 ku was observed as expected.It could be recognized by positive IBV antisera in Western blotting with high reactivity.Then the purified recombinant protein was used as antigen for immunization of rabbit to prepare polyclonal antibody.Indirect ELISA showed that the titer of polyclonal antibody was 220,and it had high reactivity and specialty with recombinant protein.Furthermore,IFA test demonstrated that this polyclonal antibody could react with Hela cells transfected with PVAX-E1 plasmid and IBV-infected CEK cells.The IBV E polyclonal antibody obtained in this study laid a foundation for further functional research of E protein in IBV pathogenesis.  相似文献

Chicks hatched with high levels of maternal antibody had excellent protection (>95%) against infectious bronchitis virus (IBV) challenge at 1 day of age, but not at 7 days (<30%). This protection significantly (P<0.05) correlated with levels of local respiratory antibody and not with serum antibody.A high percentage of both maternal antibody-positive (Mab+) and maternal antibody-negative (Mab-) chicks failed to produce IBV antibody when vaccinated at 1 day of age by the intraocular route. In addition, Mab+ chickens had a weaker virus-neutralizing antibody response to a second IBV vaccination compared to Mab- birds (P<0.05). Mab+ chicks experienced a more rapid decline (P<0.01) in maternal antibody after 1-day-of-age vaccination compared to their unvaccinated counterparts.A monoclonal antibody-based blocking ELISA that measured antibody levels specific to S1 glycoprotein of IBV correlated well with virus-neutralizing antibody titers.  相似文献

采用RT-PCR方法扩增鸡传染性支气管炎病毒(infectious bronchitis virus, IBV)HH06株E基因全长,经抗原性和亲水性分析,将E基因部分序列(193~327 bp)亚克隆于pET-32a(+)和PVAX1载体中。将阳性重组质粒pET-32a-E1转化E.coli Rosetta(DE3)感受态细胞,诱导表达获得大小约为23 ku的重组截短E1蛋白,Western blotting检测重组蛋白与IBV全病毒多克隆抗体能特异性反应。以纯化后的E1蛋白免疫新西兰白兔制备多克隆抗体,ELISA检测抗体效价达到220;Western blotting分析表明,E1多克隆抗体与重组表达蛋白具有良好的反应性;间接免疫荧光试验显示,多克隆抗体可检测到PVAX-E1转染Hela细胞表达的E1蛋白和感染鸡胚肾细胞(CEK)中的HH06株IBV,抗E蛋白多克隆抗体的制备为E蛋白功能的深入研究奠定了基础。  相似文献

以抗鸡传染性支气管病毒(IBV)核衣壳蛋白(N)的单抗株6DH8作为一抗,以辣根过氧化物酶标记的羊抗鼠IgG作为二抗,建立了检测石蜡切片中IBV抗原的单抗免疫过氧化物酶技术(Mc-IP),并对人工攻毒鸡及临床IBV感染疑似鸡进行了检测。在IBVM41株人工攻毒鸡,用该技术于1~12d从气管、2~7d从肾脏可以检测到IBV抗原,阳性染色集中于气管粘膜上皮细胞及肾小管上皮细胞胞浆;临床疑为IBV感染的病鸡,以Mc-IP技术和单抗免疫荧光试验(Mc-IFA)同时进行检测,结果阳性率分别为90.3%及83.9%。  相似文献

本试验将商品化疫苗株H120鸡传染性支气管炎病毒(IBV)直接免疫大白兔进行抗IBV全病毒粒子多克隆抗体的制备,通过琼扩试验及酶联免疫吸附试验检测其抗体效价,同时利用免疫组织化学、免疫荧光及Western blotting技术检测了所制备多抗血清的生物活性。试验结果表明,制备得到的多抗血清具有很高的抗体效价,同时能够与不同形式的IBV相关抗原发生特异性结合。  相似文献

为鉴定传染性支气管炎病毒(IBV)的抗原表位,本研究将IBV ck/CH/LDL/091022株免疫6周龄~8周龄BALB/c小鼠,将小鼠脾脏细胞与骨髓瘤细胞SP2/0融合,经过有限稀释法克隆筛选,得到一株针对IBV核衣壳(N)蛋白的单克隆抗体(MAb)2F2,经鉴定其染色体数目为101条,MAb亚类鉴定其重链属于IgG1,轻链属于K链.用肽扫描方法将IBV ck/CH/LDL/091022株N基因截短为具有相互部分重叠的23段,表达带GST标签的重组蛋白,与MAb 2F2进行western blot和ELISA反应后,鉴定其表位为397INWGDSAL404.将表位序列进行Blast结果表明,本研究鉴定的表位在大部分IBV株中均为保守抗原表位,为进一步建立IBV的检测方法奠定了基础.  相似文献

腺胃病变型鸡传染性支气管炎病毒(IBV)变异株H95提纯样品经SDS-PAGE和Westernbloting,证明H95株单抗DE7和M41株单抗6DH8均能识别H95株54000蛋白多肽。采用单抗、多抗介导间接ELISA试验表明,H95毒株能与抗IBVM41N蛋白单抗6DH8和IBVM蛋白单抗MC发生反应,也能与抗M41、Gray、Holte、T、H52、N115和分离株C9001、DLZ9111的多抗血清反应,同时抗H95株的4株单抗、多抗血清也能与上述毒株反应。卵磷脂酶C处理的H95株的血凝活性能被相应的H95株的单克隆抗体和高免血清所抑制,也能被M41单抗和高免血清所抑制。通过RT-PCR获得了H95毒株的免疫原基因S1,经Southernbloting和IBV的S探针检测呈阳性。  相似文献

为对禽流感病毒(AIV)M1蛋白表(拟)位进行分析,本研究采用针对AIV M1蛋白的型特异性单克隆抗体(MAb),淘选M13噬菌体展示的7肽随机肽库,进行M1蛋白表(拟)位分析。筛选获得共有序列MDRxL或HPR,定位于M1蛋白93~99位(93MDRAVKL99)和222~230位(222HPNSSAGLR230)氨基酸区域。采用ELISA、竞争性ELISA分析不同噬菌体拟位与抗M1的MAb免疫反应性,表明含有MDRxL或HPR基序的噬菌体拟位能够与MAb发生特异性结合,并且其结合能够被天然病毒抗原抑制或阻断,表明拟位多肽真实模拟病毒蛋白上与MAb结合的抗原决定簇或表位,提示M1蛋白93~99位(93MDRAVKL99)和222~230位(222HPNSSAGLR230)氨基酸区域构成AIV型特异性表位。  相似文献

Three panels of monoclonal antibodies (MAbs) were prepared against the spike (S) proteins of infectious bronchitis virus (IBV) strains Arkansas 99, Connecticut 46, and Massachusetts 41. Based on enzyme-linked immunosorbent assay (ELISA), the MAbs were grouped into three categories: 1) group-specific, which reacted with a broad spectrum of homologous and heterologous IBV serotypes; 2) serotype-specific, which reacted only with strains of the homologous serotype; and 3) strain-specific, which reacted "selectively" with only certain strains of homologous and heterologous serotypes. MAbs that displayed serotype specificity were all specific to S1 fractions of the homologous serotype, confirming that epitopes that determine virus serotype are associated with the S1 protein. An excellent correlation was found when the results of IBV serotyping by MAb-based indirect ELISA were compared with those from the conventional virus-neutralization test. This confirms that the MAbs described here will serve as valuable tools in epizootiological studies and serotype-specific diagnosis of IBV infection.  相似文献

The purpose of the present study was to examine the antigenicity of turkey coronavirus (TCV) isolates from various geographic areas with antibodies to different viruses. Seventeen isolates of TCV were recovered from intestinal samples submitted to Animal Disease Diagnostic Laboratory, Purdue University, from turkey farms located in different geographic areas. The prototype TCV Minnesota isolate (TCV-ATCC) was obtained from the American Type Culture Collection. Intestinal sections were prepared from turkey embryos infected with different TCV isolates and reacted with polyclonal or monoclonal antibodies to TCV, infectious bronchitis virus (IBV), bovine coronavirus (BCV), transmissible gastroenteritis virus (TGEV), reovirus, rotavirus, adenovirus, or enterovirus in immunofluorescent antibody staining. All 18 TCV isolates have the same antigenic reactivity pattern with the same panel of antibodies. Positive reactivity was seen with polyclonal antibodies to the TCV Indiana isolate, the TCV Virginia isolate, TCV-ATCC, and the IBV Massachusetts strain as well as monoclonal antibodies to the TCV North Carolina isolate or the membrane protein of IBV. Antibodies to BCV or TGEV were not reactive with any of the TCV isolates. Reactivity of antibodies to unrelated virus, rotavirus, reovirus, adenovirus, or enterovirus with different TCV isolates was all negative, except positive response was seen between enterovirus antibody and a TCV western North Carolina isolate, suggesting coinfection of turkeys with TCV and enterovirus in that particular case. The results indicated that the TCV isolates from these geographic locations in the U.S. shared close antigenicity and were antigenically related to IBV.  相似文献
