An avian adenovirus (AAV) was isolated from liver samples of two 2-wk-old broiler-breeder flocks obtained from grandparents vaccinated at 10 and 17 wks of age with an autogenous inactivated vaccine containing the European AAV 8 (8565 strain) and 11 (1047 strain) serotypes (AAV8/11 vaccine). Affected broiler-breeders exhibited clinical signs and macroscopic and microscopic lesions associated with inclusion body hepatitis (IBH). The isolated adenovirus, identified as Stanford, was molecularly characterized as European serotype 9. The pathogenicity of the Stanford strain was confirmed after inoculation of specific-pathogen-free (SPF) chickens at 1-7 days of age, causing 100% and 20% mortality, respectively. The level of protection against IBH was evaluated in two broiler-breeder progenies from AAV 8/11-vaccinated grandparent flocks and a commercial broiler flock by challenge at 1 or 7 days of age with the AAV 8 and 11 serotypes and/or the Stanford strain. The broiler-breeder progenies and the commercial broiler flock exhibited protection against IBH after challenge. No significant differences in mean body weights were observed at 3 wk of age in any of the evaluated groups. We conclude that broiler-breeder progenies from 30- to 50-wk-old grandparents vaccinated with the AAV 8/11 vaccine were adequately protected against challenge with the AAV 8 and 11 serotypes and the Stanford strain.  相似文献

An adenovirus was isolated from a foal with respiratory tract disease. The virus produced cytopathic effects (CPE) in equine embryo kidney (EEK) cell culture, contained deoxyribonucleic acid (DNA), was resistant to chloroform and pH 3, and was moderately resistant to heat. The virus caused hemagglutination of human (type O) erythrocytes. Viral density was 1.34 g/cm,3 and diameter was 75 nm. An adenovirus-associated virus (AAV) isolated from the infected cell culture was 22 nm in diameter. These viruses are classified as equine adenovirus and equine AAV.  相似文献

Chicken kidney cells, derived from the eggs of white leghorn chickens that had serological evidence of prior exposure to both adenovirus and the adeno-associated virus (AAV), produced AAV antigenic proteins upon challenge with purified adenovirus. Antigen was detected by indirect immunofluorescence using monoclonal antibody to AAV. The number of fluorescent cells were few and did not increase during the course of adenovirus infection. Similar results were obtained using cells prepared from specific-pathogen-free chicks with no previous exposure to adenovirus or AAV. It is postulated that the avian AAV can exist as a latent infection in the germ line of chickens.  相似文献

Coronavirus was isolated in HRT-18 tissue culture from nasal swabs of 4 from 15 calves showing respiratory symptoms. The strains showed cytopathic effects and could be identified as coronaviruses of bovine origin by their mouse erythrocyte agglutinating activity as well as serological specific hemagglutination inhibition and electron microscopical methods. This is the first report on the isolation of coronaviruses in tissue culture from the respiratory tract of calves in the FRG.  相似文献

A bovine adenovirus was isolated from a yearling heifer with systemic adenovirus infection. The virus was characterized as a subgroup 2 bovine adenovirus but could not be typed with reference antisera to the 10 known bovine adenovirus serotypes. A serological survey of cattle in the northern half of the North Island showed the prevalence of precipitating antibodies to bovine adenoviruses to be 43.3%.  相似文献

Two serotypes of adenovirus were isolated from lambs assembled at a ram testing station. The isolates were identified as adenoviruses on the basis of their morphologic and physicochemical characteristics. One isolate (RTS-42) was neutralized by antiserum to ovine adenovirus serotype 5, whereas the other strain (RTS-151) was not neutralized by the antisera to ovine adenovirus serotypes 1 through 5 or bovine adenovirus 1 through 8.  相似文献

An immunocytochemical technique utilizing an avidin-biotin peroxidase complex was developed to detect viral antigens in various tissues following oral administration of a locally isolated serotype 8 avian adenovirus (AAV) in specific pathogen-free (SPF) chickens. A strong color reaction was obtained with tissues from infected birds that contained a minimal amount of AAV antigens as determined by an indirect enzyme-linked immunosorbent assay. No reaction was detected in sections of tissues obtained from SPF chickens, and the reactivity with infected tissues could be removed by prior absorption of the primary antibody with purified AAV. A group-specific antigen common to the 12 serotypes of AAV was demonstrated by this technique. Because of the high sensitivity and broad-spectrum reactivity, this technique could be useful for studying the pathogenesis and laboratory diagnosis of inclusion body hepatitis caused by several serotypes of AAV.  相似文献

Type 1 bovine adenovirus has been isolated from cell cultures prepared from the kidneys of a 7-month-old bovine fetus, taken from a slaughterhouse, and types 4, 6 and 8 from cell cultures prepared from the testicles of four neonatal, precolostral calves. Sera of these pre-colostral calves possessed virus-neutralizing antibodies in titers of 1:4–1:16. Bovine adenoviruses of types 4 and 8 have also been isolated from two bovine fetuses aborted from an unknown cause. Relying upon these findings the possible vertical (transplacental) transmission of bovine adenoviruses is presumed. Though a cause-and-effect relationship between abortion and adenovirus infection could not be detected, it is probable that infection during fetal development cannot be indifferent to the fetus or the neonatal calf.  相似文献

A virus (T94-0353) isolated from the small intestine of a 3-week-old kid with diarrhea and serous ocular and nasal discharge was identified as an adenovirus based on morphologic and physicochemical characteristics. Neutralization tests and restriction endonuclease analysis comparing the caprine adenovirus with the prototype bovine and ovine adenovirus serotypes and a previously isolated caprine adenovirus showed that the caprine isolate was antigenically distinct, produced a unique restriction pattern compared with currently recognized bovine, caprine, and ovine adenoviruses, and represents a new adenovirus type. The role and significance of naturally acquired adenovirus infection in respiratory and enteric disease in goats has not been established. Isolation of adenovirus from goats with disease coupled with seroepidemiologic and pathogenicity studies will help define the role of the adenoviruses in disease production.  相似文献

Twenty-four dairy calves ranging in age from six to 15 weeks were divided so that 10 were inoculated with bovine adenovirus serotype 1 (BA-1), eight were inoculated with bovine adenovirus serotype 3 (BA-3), and six were inoculated with control tissue culture and fluids.

Post-inoculation serum samples from most of the calves presented evidence of adenovirus infection, but none of the animals exhibited signs of respiratory or enteric disease. Histopathological examination of lung tissue was not considered to be specific for adenovirus pneumonia, but was characterized by peribronchial cuffing and slight aspiration pneumonia. BA-1 was not isolated from any of the inoculated calves, and BA-3 was only recovered from the feces of two animals.

Reasons for the discrepancy between these results and those of other workers are considered, and the etiology of peribronchial cuffing is briefly discussed.

Four 3-month-old Jersey calves and three 3-month-old Holstein calves were inoculated with cervid adenovirus and monitored for clinical signs until necropsied between 10 and 42 days postinoculation. The neonatal Jersey calves had received colostrum, and the Holstein calves were colostrum deprived. Preinoculation and postinoculation serum samples were tested for antibodies to the cervid adenovirus, bovine adenovirus type 6, bovine adenovirus type 7, and goat adenovirus type 1. Virus isolation was performed on kidney, nasal secretion, and/or lung homogenates in fetal white-tailed deer lung cells. Negatively stained preparations of feces from Jersey calves were examined weekly using an electron microscope, and weekly blood samples were collected for complete blood counts. Full necropsies were performed on all calves. A complete selection of tissues was evaluated for microscopic changes, and immunohistochemistry was performed on all tissues using a polyclonal antibody to deer adenovirus. No clinical signs were observed in the calves during the study period. Following inoculation, colostrum-deprived calves developed low antibody titers to deer adenovirus, while the Jersey calves that received colostrum did not. Calves that received colostrum had high antibody titers to bovine adenovirus type 7 and goat adenovirus type 1. No consistent gross or microscopic lesions were seen. Adenovirus was not observed in negatively stained preparations of feces. Immunohistochemistry results did not demonstrate virus in all tissues examined microscopically, and virus was not isolated from lungs, nasal secretions, and kidneys.  相似文献

Two calves and a feedlot steer with systemic bovine adenovirus infection had hemorrhagic and necrotizing lesions confined to the digestive tract. In one calf there was severe, multifocal, necrotizing, hemorrhagic rumenitis, omasitis, abomasitis, enteritis and colitis. Small intestinal lesions were predominantly in Peyer's patches. Multifocal necrotizing abomasitis was the principal change in the other calf. Large amphophilic intranuclear inclusions typical of bovine adenovirus were common in swollen vascular endothelial cells. Bovine adenovirus was isolated from the former calf.The steer had diffuse hemorrhagic enterocolitis and large numbers of BAV inclusions in vascular endothelial cells of the intestinal lamina propria. Serum-neutralization tests to bovine adenovirus type 3 on acute and convalescent samples from six clinically affected in-contact animals revealed fourfold elevations in two.  相似文献

In a 10-year survey started in 1980, specimens from 8,995 bovine abortions and stillbirths were submitted to the South Dakota Animal Disease Research and Diagnostic Laboratory. Of these, 8,962 were suitable for some type of examination. Viruses were associated with 948 (10.58%). Bovine herpesvirus-1 (IBR) was detected in 485 (5.41%), and bovine viral diarrhea virus (BVDV) was detected in 407 (4.54%). In 1 year of the survey, BVDV was detected in 8/32 fetuses that had lesions of passive congestion. Bovine herpesvirus-4 was isolated from 47 specimens (0.52%), parvovirus and enterovirus were each isolated from 2, and adenovirus, parainfluenza virus, and pseudorabies virus were each isolated from 1. Malignant lymphoid neoplasia was present in 2 fetuses, and their abortion was assumed to have been caused by the bovine leukosis virus.  相似文献

Sporadic sudden deaths in adult white-tailed deer occurred from November 1997 through August 1998 on an Iowa game farm. Three of the 4 deer necropsied had severe pulmonary edema, widespread mild lymphocytic vasculitis, and amphophilic intranuclear inclusion bodies in scattered endothelial cells in blood vessels in the lung and abdominal viscera. Immunohistochemistry with bovine adenovirus 5 antisera and transmission electron microscopy demonstrated adenoviral antigen and nucleocapsids, respectively, within endothelial cells. Adenovirus was isolated in cell culture from 1 of the affected deer. The isolate was neutralized by California black-tailed deer adenovirus antiserum. These findings indicate that adenovirus should be considered in the differential diagnosis of both black-tailed and white-tailed deer with pulmonary edema and/or hemorrhagic enteropathy.  相似文献

为了研究禽腺病毒1型和4型(AAV-1和AAV-4)六邻体抗原表位和同义密码子的使用情况,运用生物学软件Antheprot 5.0和EMBOSS的CHIPS、CUSP程序分别对AAV-1和AAV-4六邻体进行了分析,并将分析结果与大肠埃希菌、酵母及人的密码子偏爱性进行比较.结果显示,AAV-1、AAV-4六邻体的抗原表位区都主要集中在前段、中段,推测具有型和群特异性表位,可作为诊断型和群的首选蛋白.AAV-1和AAV-4六邻体的Enc值分别为43.553和38.475,在编码密码子使用频率上都存在明显差异,与大肠埃希菌、酵母及人的密码子偏爱性也不同,其表达系统选择大肠埃希菌较为合适.  相似文献

Colostrum deprived calves were experimentally infected with an adenovirus isolated from sheep and related to bovine adenovirus type 2. The calves showed respiratory symptoms and mild diarrhoea from the third day after infection. Laboratory tests revealed the development of leucopenia, lymphopenia, a drop of the pH of the urine and the appearance of pathological changes in the urine. The animals shed the virus in their nasal discharge, faeces and urine. Comparing the clinical and virological findings with the previous experimental infection of lambs it is concluded, that this type of adenovirus is similarly pathogenic for the two ruminant species.  相似文献

A modified version of the test method of the Comité Européen de Normalisation (CEN) was developed using formic acid and three commercial disinfectants to evaluate virucidal activity against three non-enveloped viruses, bovine enterovirus type 1 (ECBO virus), mammalian orthoreovirus type 1 and bovine adenovirus type 1 (BAV 1). Determination of the effects of temperature was carried out at 20 and 10 degrees C. All tests with protein load used bovine serum albumin (BSA) and yeast extract. The investigations were performed in suspension tests and in carrier tests using poplar wood virus carriers. The carrier tests showed that ECBO virus could be inactivated at 20 degrees C with 1% formic acid within a 60 min reaction time. For disinfection of ECBO virus at 10 degrees C within 60 min, a 2% concentration of formic acid was necessary. Formic acid was ineffective against reovirus and bovine adenovirus and cannot be recommended as a reference disinfectant. Inactivation of ECBO virus and adenovirus type 1 using a disinfectant containing aldehydes and alcohols could be achieved, but only at room temperature. The disinfection of reovirus type 1 at room temperature with this product was possible without a protein load. This disinfectant exhibited disinfection ability at 10 degrees C at a concentration of more than 2% or with a longer exposure time. A disinfectant containing aldehydes was effective at room temperature but its effect was reduced in the presence of organic matter. Inactivation at 10 degrees C was found only against adenovirus. The fourth disinfectant, which contained peroxiacetic acid, inactivated all test viruses at a concentration of 0.5% within 15 min independent of temperature and protein load.  相似文献

In double immunodiffusion tests between a bovine subgroup 2 adenovirus (serotype 8) and other mammalian adenoviruses, no group-specific crossing was demonstrated.However, in cross-fluorescent antibody tests (FAT) between bovine subgroup 2 viruses (serotypes 5, 7 and 8) and conventional (non-subgroup 2) adenoviruses from several species (bovine adenovirus serotypes 1 and 2, ovine serotypes 1, 4 and 5, porcine serotype 1, and human serotypes 2 and 5) sharing of antigens was demonstrated. The FAT titres observed when rabbit antisera to conventional adenoviruses were used to stain bovine subgroup 2 viruses were, however, much lower than titres with other non-subgroup 2 viruses. The converse was also true. The crossing was also predominantly one-sided.The low level cross was confirmed using antisera to selected viruses prepared in chickens to exclude interference by possible natural adenovirus infections in the rabbits used to prepare the antisera in the initial experiments.  相似文献

The occurrence of natural in utero adenovirus infection in sheep was examined. Isolation of three adenovirus strains from the kidneys of 174 sheep foetuses is reported; all three isolates belonged to Type 2 bovine adenovirus (BAV 2) Subtype B. Five of 25 blood samples from sheep foetuses contained virus-neutralizing antibodies (1:4-1:24) against BAV 2. These data prove that transplacental transfer of adenovirus infection can occur naturally in sheep.  相似文献

A viral agent, designated Id-1, was isolated from the buffy coat of a calf suffering from weak calf syndrome. The virus replicated on bovine salivary gland cells and caused cytopathic effect within four days after infection-Cytopathic effect was characterized by rounding and clumping of cells. Stained preparations of infected monolayers revealed multiple intranuclear inclusions. The agent was found to be resistant to chloroform, ether, trypsin, sodium desoxycholate, oxytetracycline and a pH range of three to nine. The virus was sensitive to 5-iodo-2'-deoxyuridine and to a temperature of 70 degrees C. Cross neutralization tests with Id-1 antiserum and bovine adenovirus type 7 (strain Fujuroi) antiserum resulted in complete neutralilation of both viruses with four or less antibody units of homologous or heterologous antiserum.  相似文献
