Soybeans in general contain 35-40% protein. Efforts are underway to increase further this protein content, thus enhancing their nutritive value. Even though higher protein is a desirable characteristic, whether such an increase will be accompanied by enhanced protein quality is not known. Soybean protein quality could be significantly improved by increasing the concentration of the sulfur-containing amino acids, cysteine and methionine. To ascertain if a correlation existed between protein quantity and quality, a comparison of the amino acids of soybeans differing in protein content was made. Soybeans with higher protein content had a significantly lower percentage of sulfur amino acids, while those with lower protein exhibited a higher content of cysteine and methionine. Nitrogen application elevated the protein content but lowered that of the sulfur amino acids. Transmission electron microscopy examination of thin sections of low protein soybean seeds revealed several protein storage vacuoles that were partially filled with storage proteins. Fluorescence two-dimensional difference gel electrophoresis of soybean seed proteins revealed that nitrogen application favored the accumulation of the beta-subunit of beta-conglycinin while decreasing the accumulation of Bowman-Birk protease inhibitor (BBI), a protein rich in cysteine. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis of 60% 2-propanol-extracted proteins showed a drastic reduction in the accumulation of BBI with increasing protein content. Northern blot analysis indicated that nitrogen had a negative influence on the expression of the BBI gene. Our results indicate that the negative correlation between total protein and sulfur amino acid content is mostly mediated by the differential accumulation of BBI.  相似文献

The agronomic performance and storage proteins of high seed protein lines of soybeans [Glycine max L. (Merr.)] were investigated to determine if the two major storage proteins, beta-conglycinin and glycinin, contribute to the increased protein content of high seed protein lines. Subunits of these two major storage proteins were estimated by scanning SDS-PAGE gels by scanning densitometry. The relative rankings of the lines with respect to seed size and protein content were not different between years in one environment over 5 years, but oil and total protein and oil contents and the ratio of protein to oil differed. The alpha', alpha, and beta subunits of beta-conglycinin were significantly higher in the high-protein lines except CX797-115, CX804-108, CX804-3, D81-8498, and NC-2-62. The acidic A(3) polypeptide of glycinin was significantly higher in high-protein lines except 76-48773, CX804-108, CX804-3, D81-8498, and NC-2-62, whereas the acidic polypeptides A(1,2,4) of glycinin were significantly higher in all of the high-protein lines. The basic polypeptides of glycinin were significantly higher in all high seed protein lines except D81-8259. In conclusion, high-protein lines appear to contain more beta-conglycinin and glycinin than normal-protein soybean lines, and the amounts of subunits and polypeptides differ among lines.  相似文献

To circumvent drought conditions persisting during seed fill in the mid-south U.S. soybean production region, researchers have developed the early soybean (Glycine max [L.] Merr.) production system (ESPS), which entails early planting of short-season varieties. Because soybean supplies a preponderance of the world's protein and oil and consumption of soy-based foods has been associated with multiple health benefits, the effects of this agronomic practice on seed quality traits such as protein, oil, and isoflavones should be investigated. Four cultivars of soybean, two from maturity group IV and two from maturity group V, were planted in April (ESPS) and May (traditional) in a two-year study at Stoneville, MS. Near-infrared analysis of soybean seed was utilized to determine the percentages of protein and oil. Dependent upon variety, the oil content of the early-planted crop was increased by 3-8%, whereas protein was not significantly changed. Visualization of protein extracts fractionated by sodium dodecyl sulfate-polyacrylamide electrophoresis and fluorescence two-dimensional difference gel electrophoresis revealed that early planting did not affect the relative accumulation of the major seed-storage proteins; thus, protein composition was equal to that of traditionally cultivated soybeans. Maturity group IV cultivars contained a higher percentage of oil and a lower percentage of protein than did the maturity group V cultivars, regardless of planting date. Gas chromatographic separation of fatty acids revealed that the percentages of saturated and unsaturated fatty acids were not significantly altered by planting date. Methanol extracts of seed harvested from different planting dates when analyzed by high-performance liquid chromatography showed striking differences in isoflavone content. Dependent upon the variety, total isoflavone content was increased as much as 1.3-fold in early-planted soybeans. Irrigation enhanced the isoflavone content of both early- and late-planted soybeans as much as 2.5-fold. Accumulation of individual isoflavones, daidzein and genistein, was also elevated by irrigation. Because this cultural practice improves the quality traits of seeds, ESPS provides an opportunity for enhancing the quality of soybean.  相似文献

Application of nitrogen (N) fertilizer at the flowering stage changed the contents of storage compounds in seeds of Soybean (Glycine max L. cvs Enrei and Tamahomare). The effects of the N application on the maturation of soybean seeds were examined by comparing changes in the contents of amino acids, sugars, water, protein, and oil in seeds from N‐dressed plants (NDS) with those from undressed plants (UDS) during maturation. The application resulted in a decrease of contents of total and some amino acids (glutamine and asparagine) in developing seeds except for at the early maturation stage and in a decreased protein content of mature seeds. On the other hand, the N application led to faster accumulation of oil in developing seeds and to an increased oil content of mature seeds. Based on these results, it was concluded that the N application at the flowering stage changed the composition of solutes imported by developing seeds and resulted in variations in the contents of storage compounds. The results indicate that it is possible to improve seed quality by fine control of N application.  相似文献

Seed protein concentration of commercial soybean cultivars calculated on a dry weight basis ranges from approximately 37 to 42% depending on genotype and location. A concerted research effort is ongoing to further increase protein concentration. Several soybean plant introductions (PI) are known to contain greater than 50% protein. These PIs are exploited by breeders to incorporate the high-protein trait into commercial North American cultivars. Currently, limited information is available on the biochemical and genetic mechanisms that regulate high-proteins. In this study, we have carried out proteomic and molecular analysis of seed proteins of LG00-13260 and its parental high-protein lines PI 427138 and BARC-6. Sodium dodecyl sulfate polyacrylamide gel electrophoresis analysis revealed that the high-protein lines accumulated increased amounts of beta-conglycinin and glycinins, when compared with Williams 82. High-resolution two-dimensional electrophoresis utilizing pH 4-7 and pH 6-11 ampholytes enabled improved resolution of soybean seed proteins. A total of 38 protein spots, representing the different subunits of beta-conglycinin and glycinin, were identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry. High-protein was correlated with an increase in the accumulation of most of the subunits representing beta-conglycinin and glycinin. Comparisons of the amino acid profiles of high-protein soybean lines revealed that the concentration of sulfur amino acids, a reflection of protein quality, was not influenced by the protein concentration. Southern blot analysis showed the presence of genotypic variation at the DNA level between PI 427138 and BARC-6 for the genes encoding group1 glycinin, beta-conglycinin, Bowman-Birk inhibitor (BBI), and the Kunitz trypsin inhibitor (KTI). LG00-13260 inherited the allelic variants of the parental line PI 427138 for glycinin, beta-conglycinin, and KTI, while BBI was inherited from the parental line BARC-6. The results of our study indicate that high-seed protein concentration is attributed to greater accumulation of specific components of beta-conglycinin and glycinin subunits presumably mediated by preferential expression of these genes during seed development.  相似文献

The Hibiscus genus encompasses more than 300 species, but kenaf (Hibiscus cannabinus L.) and roselle (Hibiscus sabdariffa L.) are the two most economically important species within the genus. Seeds from these two Hibiscus species contain a relatively high amount of oil with two unusual fatty acids: dihydrosterculic and vernolic acids. The fatty acid composition in the oil can directly affect oil quality and its utilization. However, the variability in oil content and fatty acid composition for these two species is unclear. For these two species, 329 available accessions were acquired from the USDA germplasm collection. Their oil content and fatty acid composition were determined by nuclear magnetic resonance (NMR) and gas chromatography (GC), respectively. Using NMR and GC analyses, we found that Hibiscus seeds on average contained 18% oil and seed oil was composed of six major fatty acids (each >1%) and seven minor fatty acids (each <1%). Hibiscus cannabinus seeds contained significantly higher amounts of oil (18.14%), palmitic (20.75%), oleic (28.91%), vernolic acids (VA, 4.16%), and significantly lower amounts of stearic (3.96%), linoleic (39.49%), and dihydrosterculic acids (DHSA, 1.08%) than H. sabdariffa seeds (17.35%, 18.52%, 25.16%, 3.52%, 4.31%, 44.72%, and 1.57%, respectively). For edible oils, a higher oleic/linoleic (O/L) ratio and lower level of DHSA are preferred, and for industrial oils a high level of VA is preferred. Our results indicate that seeds from H. cannabinus may be of higher quality than H. sabdariffa seeds for these reasons. Significant variability in oil content and major fatty acids was also detected within both species. The variability in oil content and fatty acid composition revealed from this study will be useful for exploring seed utilization and developing new cultivars in these Hibiscus species.  相似文献

To evaluate the extent of the genetic change and its effects on the seed protein composition of soybean cultivars released during the past 60 years, representative ancestral cultivars and those derived from selective breeding were grown in a side-by-side comparison. Total seed protein content, determined by combustion analysis of nitrogen, revealed a decline in the protein content after decades of selection and breeding. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis comparison of protein profiles of the soybean cultivars indicated that relative expression of most of the seed storage proteins had not varied substantially from the ancestral lines to the present commercial cultivars. There was noticeably less beta-subunit of beta-conglycinin, a protein devoid of sulfur amino acids, in the modern cultivars represented by Mustang, Pioneer 93B09, and Asgrow 3602. Comparison of the amino acid profiles of soybean seed, a benchmark of the protein's nutritional quality, revealed that the ancestral progenitor, G. soja, was significantly higher in cysteine, glutamic acid, histidine, and arginine than either the ancestral or the modern cultivars. Selective breeding over the past 60 years minimally affected the overall amino acid composition. The degree of divergence in the DNA sequence of the genes encoding glycinin and beta-conglycinin in the ancestral and modern cultivars was investigated using Southern hybridization and the polymerase chain reaction. Even though some restriction fragment polymorphisms could be detected, overall, the banding patterns were remarkably similar among the ancestral cultivars and those derived from them, suggesting a high degree of conservation of seed-storage protein genes. The results of our study suggest that selection and breeding for yield during the past 60 years had no major influence on the protein composition, ostensibly because of limited genetic diversity among the parental lines.  相似文献

ABSTRACT

We compared the protein and oil contents, seed yields, and natural abundance of 15N (σ15N) of seeds from the plants of the cultivar Enrei, which has moderate nodulating ability (Enrei) with those of its two near-isogenic lines (NILs), a super-nodulating genotype of Enrei, i.e., En-b0-1, and a non-nodulating genotype of Enrei, i.e., En1282. Plants of these three genotypes were grown on four types of field plots with different types of urea coated slow-release nitrogen (N) fertilizers (CUSLNFs) which delivered N in different manners during plant growth . The seed yield of the En1282 plants was similar to that of the Enrei plants but much greater than that of the En-b0-1 plants when the plants were grown on the field to which a CUSLNF with a long lifespan was applied. The amounts of protein per seed were positively correlated with those of oil per seed in the case of En1282 plants irrespective of the field N conditions. The seed protein contents were proportional to the seed weight in both En1282 and Enrei plants. Such a relationship was not observed in the En-b0-1 plants, and the protein and oil contents in the seeds of En-b0-1 plants were negatively correlated with each other. These observations suggested that the N supply to maturing seeds was a key to the protein contents in the En1282 and Enrei plants and that the C supply to maturing seeds was a key to the protein contents in the En-b0-1 plants. The comparison of the σ15N values, protein contents, and seed yields of seeds from En1282 plants suggested that the Enrei plants assimilated considerable amounts of N from the soil during the late plant growth. We concluded that high N2 fixation activity depressed the amounts of oil per seed and changed the protein and oil contents in soybean seeds.  相似文献

Sanchi (Panax notoginseng) root has been widely used as traditional herb to stanch blood, disperse gore and reduce pain in China for centuries. However, the nutritional information about its seed was unknown. Sanchi seeds obtained from Yunnan Province were analyzed for their nutritional composition, fatty acid, amino acid profile and sugar contents. Sanchi seeds were abundant in fats (46.35 %) and proteins (23.90 %). Notably, seven fatty acid compositions were determined and abundant in unsaturated fatty acid (99.56 %), containing 95.71 % oleic acid. Sanchi seed proteins were rich in glutenin, globulin and albumin (28.63, 27.83 and 26.81 %, respectively). Sanchi seed contain 17 kinds of amino acids, of which nine were essential amino acids, accounting for 41.30 %. These nutritional compositions indicate that Sanchi seed has the potentiality to be exploited as human edible oil, industrial use, new medicine or healthcare products for diabetes patients due to low sugar content.  相似文献

Influence of the nitrogen-nutrition on qualitative and quantitative characteristics of seeds of rape (Brassica napus L.) An experiment was carried out under greenhouse conditions to analyse the influence of nitrogen nutrition on qualitative and quantitative characteristics of seeds of rape, variety “Erglu”. Increasing the level of nitrogen fertilization raised the crude protein content during the whole period of seed growth but decreased the lipid content during the main seed growth stages. Nitrogen nutrition only changed the fatty acid composition significantly during the early stages of seed growth. Increasing nitrogen fertilization resulted at maturity in a trend towards higher contents of palmitic and linoleic acids but in a lower content of oleic acid. At maturity the lipid content was not influenced by the level of nitrogen nutrition. The differences in seed-yield per pod between the main branch and axillary branch 5 became smaller when nitrogen nutrition was increased.  相似文献

The variation in the seed shape, colour and yield, and content, yield and fatty acid composition of seed oil of 109 accessions of opium poppy Papaver somniferum, (majority of them Indian land races), was investigated. The seeds were white, pale yellow or light brown in colour, reniform or round in shape and varied in size up to three fold. The oil content, seed and the oil yield varied between 26 to 52%, 1.0 to 7.4 g/plant and 0.4 to 2.7 g/plant, respectively. The % content of palmitic, oleic and linoleic acid in the seed oil ranged between 9.3 to 40.0%, 7.5 to 58.4% and 0.7 to 72.7%, respectively. On average basis, the levels of major fatty acids in the seed oil were: oleic (37.1%) > palmitic (27.3%) > linoleic acid (17.2%). The palmitoleic, stearic and linolenic acids were present in the oils of only some of the accessions. Two of the accessions yielded linoleic acid rich seed oil of about the same quality as soybean and maize oils, and in four accessions, the proportion of palmitic, oleic and linoleic acids was roughly equal. The palmitic acid was relatively less and linoleic acid more in the seed oil from accessions rich in oil content. The oil that contained higher amount of oleic acid also contained higher amount of palmitic acid and relatively lower amount of linoleic acid. The correlation analyses revealed a strong positive relationship between seed yield and oil yield (r = +0.81), oil yield and oil content (r = +0.54) and oleic acid and palmitic acid content in the seed oil (r = +0.49), and a weak positive relationship between oil content and linoleic acid content of oil (r = +0.24), and a negative correlation was observed between oil content and palmitic acid content (r = –0.32), palmitic acid and linoleic acid (r = –0.55) and oleic acid and linoleic acid contents of oil (r = –0.68). The observations have permitted selection of accessions that are high seed and oil yielding and/or rich in linoleic, palmitic and oleic acids or containing palmitic, oleic and linoleic acids in about equal amounts.

Rapeseeds contain cruciferin (11S globulin), napin (2S albumin), and oleosin (oil body protein) as major seed proteins. The effects of oil expression and drying conditions on the extraction of these proteins from rapeseed meal were examined. The conditions strongly affected the extraction of oleosin and only weakly affected the extraction of cruciferin and napin. The protein chemical and physicochemical properties of cruciferin, the major protein present, were compared with those of glycinin (soybean 11S globulin) under various conditions. In general, cruciferin exhibited higher surface hydrophobicity, lower thermal stability, and lower and higher solubility at mu= 0.5 and mu = 0.08, respectively, than did glycinin. At the pHs (6.0, 7.6, and 9.0) and ionic strengths (mu= 0.08 and 0.5) examined, the emulsifying ability of cruciferin was worse than that of glycinin, except at mu= 0.08 and pH 7.6. The emulsifying abilities of cruciferin and glycinin did not correlate with thermal stability and surface hydrophobicity. Higher protein concentration, higher heating temperature, higher pH, and lower ionic strength were observed to produce harder gels from cruciferin. Gel hardness partly correlated with the structural stability of cruciferin.  相似文献

Experiments were conducted in 2009–2010 at the Agricultural and Natural Resources Research Center of Yazd, Iran, to study the effect of nitrogen levels and plant growth-promoting rhizobacteria (PGPR) containing Azotobacter sp. and Azospirillum sp. on seed yield, its components and quality traits of sesame cultivars. Treatments were arranged in a factorial experiment based on randomized complete block design with nitrogen rates (0 (control), 25 and 50 kg N ha?1), cultivars (Darab-14, GL-13 and local) and N-fixing bacteria levels (non-inoculation and inoculation) were applied with three replications. Nitrogen fertilizer significantly increased yield and yield components, but reduced oil content in 2010. Inoculating seeds with PGPR increased yield and yield components of sesame cultivars compared to the control treatment. Seed yield in PGPR inoculation with half a rate of N fertilizer treatment was more than seed yield in the full rate of N fertilizer without a PGPR inoculation treatment. N fertilizer and PGPR application significantly decreased saturated fatty acids (palmitic and stearic acid) while it significantly increased unsaturated fatty acids (oleic and linoleic acid). Oleic acid had a significant negative correlation with linoleic acid (r = ?0.79). The result showed that an application of PGPR could be usefully applied to reduce use of chemical fertilizer.  相似文献

The effect of germination and growth under illuminated and dark environments on canola seed reserves was investigated. Depletion of proteins and lipids in whole seedlings and their top (leaf/cotyledons) and bottom parts (stem/roots/seed coat) was independent of light, whereas the protein solubility increased at a faster rate under an illuminated environment than in the dark. A rapid increase in free fatty acids but a net decrease of dry matter content in seedlings grown in the dark environment was observed. The dry matter content of seedlings grown in the illuminated environment increased due to photosynthetic biomass accumulation.  相似文献

Castor has tremendous potential as a feedstock for biodiesel production. The oil content and fatty acid composition in castor seed are important factors determining the price for production and affecting the key fuel properties of biodiesel. There are 1033 available castor accessions collected or donated from 48 countries worldwide in the USDA germplasm collection. The entire castor collection was screened for oil content and fatty acid composition by nuclear magnetic resonance (NMR) and gas chromatography (GC), respectively. Castor seeds on the average contain 48.2% oil with significant variability ranging from 37.2 to 60.6%. Methyl esters were prepared from castor seed by alkaline transmethylation. GC analysis of methyl esters confirmed that castor oil was composed primarily of eight fatty acids: 1.48% palmitic (C16:0), 1.58% stearic (C18:0), 4.41% oleic (C18:1), 6.42% linoleic (C18:2), 0.68% linolenic (C18:3), 0.45% gadoleic (C20:1), 84.51% ricinoleic (C18:1-1OH), and 0.47% dihydroxystearic (C18:0-2OH) acids. Significant variability in fatty acid composition was detected among castor accessions. Ricinoleic acid (RA) was positively correlated with dihydroxystearic acid (DHSA) but highly negatively correlated with the five other fatty acids except linolenic acid. The results for oil content and fatty acid composition obtained from this study will be useful for end-users to explore castor germplasm for biodiesel production.  相似文献

Abstract

Camelina (Camelina sativa (L.) Crtz., false flax) is described as a species requiring fewer inputs than other oilseed crops thus making it an interesting alternative in sustainable cropping systems. As information on the combined effects of nitrogen and sulphur on camelina yield and quality parameters is meagre, a pot fertilization experiment was carried out with nitrogen applied as NH4NO3 at three increasing rates equivalent to a range from 63 up to 127 kg ha?1. These treatments were combined with sulphur additions applied as (NH4)2SO4 to achieve a soil sulphate content of 25 or 45 mg kg?1, respectively (equivalent to 75 and 135 kg S ha?1). The medium and high nitrogen rates combined with the low sulphur fertilization level increased the number of branches per plant compared with the lowest nitrogen fertilization at the same sulphur rate. Camelina seed yield increased with increasing nitrogen doses at the same low sulphur level, whereas straw yield increased only at the highest rate of nitrogen. Thousand-seed mass increased at the highest nitrogen dose and with the low sulphur application rate. In addition, the increase in nitrogen fertilization reduced seed oil content from 39.8% to 37.1%. A highly negative correlation was observed between oil and protein content of seeds. Nitrogen fertilization increased total oil yield and total protein yield. Differences in crop parameters between the sulphur treatments were not significant statistically, although the higher sulphur treatment tended to increase seed yield as well as oil and protein content compared with the low sulphur treatment. Thus, the combined application of N and S as mineral fertilizers is only recommended when growing camelina on sulphur-deficient soils if the aim is to achieve both high oil and protein production.  相似文献

This paper describes the composition of sesame seed oils obtained from seeds collected from five countries that are major suppliers of traded sesame seed oil. Oils were extracted from the seeds using small-scale industry pressing equipment and analyzed using standard methods for fatty acids, fatty acids in the triglyceride 2-position, tocopherols and tocotrienols, triglycerides, sterols, steradienes, and iodine value. Values for the composition of the sterols, triglycerides, fatty acids, iodine value, and tocopherol composition were generally in good agreement with the results published elsewhere. All of the oils from roasted seeds contained low levels of the sterol degradation products stigmasta-3,5-diene and campesta-3,5-diene, which were probably formed by dehydration of the parent sterols during roasting.  相似文献
