1. Female chicks of a White Leghorn strain were fed three different diets from one day old: control, additional vitamin K3 (10 mg/kg), and a diet containing a combination of additional vitamin K3, sodium fluoride (10 mg/kg) and limestone in particulate rather than powdered form. At 16 weeks photoperiod was increased for half the birds from 8:16 L:D to 16:8 L:D immediately or by one hour per week to the same ultimate photoperiod for the other half. 2. Age at first egg was lower by 4.0 d for birds on the fast lighting regime but there were no overall effects of lighting on bone quality at either 25 or 70 weeks. 3. Additional vitamin K3 resulted in higher proximal tarsometatarsus cancellous bone volumes at 15 weeks and throughout the laying period compared with controls. Plasma osteocalcin concentrations were unaffected by vitamin K3 supplementation during growth. 4. The combination diet resulted in beneficial responses of 12 to 20% in most bone characteristics in hens at 70 weeks. The magnitude of these effects was similar to a previous study involving a particulate calcium source alone (Fleming et al., Poultry Science, 39: 434-440, 1998b). We conclude that the beneficial effects of the combined treatment over the lifetime of the hens were attributable mainly to the presence in the diet of a calcium source in particulate form.  相似文献

1. The effects upon bone quality of feeding limestone in flour or particulate form and housing type (cage or aviary) in lines of hens divergently selected for high (H) or low (L) bone strength over 7 generations were investigated. 2. As in previous generations, highly significant phenotypic differences between lines were observed in all measured bone traits at peak egg production (25 weeks) and towards the end of production (56 weeks) in both cage and aviary systems. 3. At 25 weeks there were no significant effects on bone variables of feeding particulate limestone although a significant reduction in osteoclast number was observed at this age. By 56 weeks osteoclast numbers were further reduced in hens fed particulate limestone and beneficial effects on some bone variables were observed in this treatment group. 4. The genotypic and dietary improvements upon bone quality were independent and additive at both ages. There were very few interactive effects. 5. Hens with the freedom to move in an aviary environment during the laying period had improved bone status compared to caged siblings. Environmental and genotypic effects were additive. 6. There were no effects of line on egg production although H line hens had slightly higher egg production by 56 weeks. Egg numbers were unaffected by diet. Eggshell thickness and strength were unaffected by line but hens fed particulate limestone had thicker- and stronger-shelled eggs over the production period as a whole. 7. We conclude that; (a) genetic selection is extremely effective in improving bone strength and resistance to osteoporosis; (b) allowing hens freedom to exercise can also improve bone strength but may increase the risk of keel damage if they do not have genetically-improved bone status; (c) feeding hens a particulate form of limestone from 15 weeks onwards can also increase bone strength and eggshell quality; (d) genetics, environment and nutrition all have independent and additive effects on bone status in laying hens but the relative effectiveness of these factors is genetics > environment > nutrition.  相似文献

The aim of this study was to evaluate the effect of dietary supplementation of vitamin K (vit K) and Ca in the diets of laying hens on bone and blood parameters and performance during the growing phase and the effects on the laying phase up to 32 weeks. The study utilized 120 Hy‐Line W‐36 pullets in the growing phase (13–18 weeks), 80 laying hens in the production phase (20–32 weeks), distributed in a completely randomized design in a 2 × 5 factorial arrangement, two levels of Ca (0.8 and 1.4%) and five levels of vit K supplementation (0, 2, 8, 16 and 32 mg/kg) with four experimental units. An experimental diet was fed during the growing phase, and the commercial diet was fed during the production phase and were analysed for biochemistry and bone variables at 18 and 32 weeks. In pullets, a significant interaction of levels of vit K and Ca was observed for total serum calcium (mg/dl), Seedor index, Ca in the bones (%) and medullary bone (%). Pullets at 18 weeks of age, receiving diets with 1.4% Ca, displayed a quadratic effect of vit K with better results for these variables with 17.86, 14.59, 14.48 and 16.80 mg/kg of vit K, respectively. For level 0.8% Ca no effect of vit K was observed. The treatments during the growing phase had no effect on egg production, performance and biochemistry serum until 32 weeks. Medullary bone (%), there was a significant interaction for 1.4% Ca and a quadratic effect for vit K, with greater medullary bone areas observed with 6.09 mg/kg vit K in the diet. Supplementation with vit K in a diet with 1.4% Ca during the growing phase allowed for an increase in the area of medullary bone at 18 weeks, which was maintained until 32 weeks.  相似文献

Many studies have described the effects of exercise restriction on the mammalian skeleton. In particular, human and animal models have shown that reduction in weight bearing leads to generalised bone loss and deterioration of its mechanical properties. The aim of this study was to assess the effect of prolonged exercise restriction coupled with heavy calcium demands on the micro-structural, compositional and mechanical properties of the avian skeleton. The tibiae and humeri of 2-year-old laying hens housed in conventional caging (CC) and free-range (FR) housing systems were compared by mechanical testing and micro-computed tomography (μCT) scanning. Analyses of cortical, cancellous and medullary bone were performed.Mechanical testing revealed that the tibiae and humeri of birds from the FR group had superior mechanical properties relative to those of the CC group, and μCT scanning indicated larger cortical and lower medullary regions in FR group bones. Cancellous bone analysis revealed higher trabecular thickness and a higher bone volume fraction in the FR group, but no difference in mineral density. The biomechanical superiority of bones from the FR group was primarily due to structural rather than compositional differences, and this was reflected in both the cortical and cancellous components of the bones. The study demonstrated that prolonged exercise restriction in laying hens resulted in major structural and mechanical effects on the bird skeleton.  相似文献

The effects of different dietary levels of zinc and vitamin A on tissue mineral accumulation and histopathological alterations were examined in laying hens. For this purpose, 130 Hisex brown laying hens aged 56 weeks were divided into two main groups. Each group of hens were fed on diets supplemented with two different levels of vitamin A (0-10,000 IU kg(-1)). Then, laying hens in both of the main groups were subdivided into five treatment groups given different levels of zinc (0, 25, 50, 100, and 200 mg kg(-1)) for 12 weeks. Dietary additions of zinc and/or vitamin A did not significantly affect the zinc concentrations of the pancreas, liver, gizzard, thyroid, and copper concentrations of the pancreas, gizzard, and thyroid. The group fed the highest zinc (200 mg kg(-1)) in the diet had twofold iron concentration in their thyroid tissue compared to the concentration of the control group receiving no extra zinc. Histopathologically, there was slight oedema, degranulation, and vacuolation in exocrine cells and some fibrotic areas in the pancreas and reduced follicular size. Follicular cell hypertrophy and hyperplasia occurred in the thyroids, and gizzard erosions were seen in some cases.  相似文献

The effects of zinc and vitamin A supplementation to the diet on some blood metabolites were evaluated in Hisex brown laying hens from 56 weeks to 68 weeks of age. A total of 130 birds were divided into two main groups according to vitamin A treatment (0 and 3.44 mg retinyl acetate kg(-1) feed, respectively), each consisting of 65 hens. Hens in both of the main groups were then divided into five zinc treatment groups (0, 25, 50, 100 and 200 mg zinc kg diet(-1) respectively) of 13 hens each. It was observed that plasma T4, T3 and total cholesterol levels were affected by only zinc supplementation. While 100 and 200 mg Zn kg(-1) decreased plasma T4 level compared to control value, plasma T3 level was reduced by 100 mg Zn kg(-1) compared to groups fed less Zn. Adding 50 and 200 mg Zn kg(-1) to the diet increased plasma total cholesterol level in the birds compared to other groups. Vitamin A, zinc, and their interaction did not influence the concentration of plasma high-density lipoprotein (HDL)-cholesterol, glucose and egg yolk cholesterol in laying hens.  相似文献

1. The aim of this study was to assess bone characteristics in restricted ovulator (RO) hens. These hens generally are unable to ovulate due to a point mutation in the oocyte VLDL receptor gene whose protein product mediates the uptake of yolk precursors. Because these hens do not have the cyclic calcium (Ca) metabolism associated with egg formation, they could be a useful model for studying bone metabolism. 2. RO hens had greater humerus, femur and tibia ash concentrations than wild-type (WT) and out-of-production (OP) hens. Bone mineral content and density obtained with dual-energy X-ray absorptiometry (DXA) were highly correlated with the results of conventional bone assays. 3. Gross and histological examination of the femurs confirmed the presence of extremely dense medullary bone deposition in the RO hens. However, the composition of non-collagenous protein extracts of medullary bone was similar for the two genotypes. 4. Analysis of medullary bone extracts for glycosaminoglycans (GAG) confirmed the presence of large amounts of keratan sulphate (KS) in the matrix of medullary bone. 5. Plasma Ca, total GAG and KS concentrations of RO hens were markedly higher than WT and OP hens. The changes in plasma calcium and keratan sulphate are probably a reflection of elevated Ca-binding yolk precursor molecules and intensive medullary bone formation in response to increased plasma oestrogen observed by others in RO hens.  相似文献

Aged laying hens at oviposition were given four levels of dietary vitamin D3 and three levels of 1,25-dihydroxyvitamin D3 (1,25[OH]2D3) to determine the effects of the additives on the medullary and endosteal bone. Oxytetracycline hydrochloride labeling and histomorphometry of undecalcified sections were used in the determination of changes in medullary bone and endosteal surface of mid-diaphyseal tibiotarsal bones. Treatment with 1,25(OH)2D3 increased mineralized medullary bone area, endosteal surface length, amount of tetracycline labeling in medullary and endosteal bone, total number of osteoclasts on endosteal and medullary bone, and appositional rate of mineralizing bone. Vitamin D3 increased mineralized medullary bone area and total tetracycline labeling in endosteal bone but decreased endosteal mineral apposition rate. Medullary bone matrix with no mineral was observed only in the group receiving neither supplement. These areas were much larger than typical osteoid seams usually seen in trabecular bone. Both 1,25(OH)2D3 and vitamin D3 were effective in stimulating bone formation and retention of mineral within medullary bone matrix at oviposition.  相似文献

Adult laying hens were fed diets deficient in phosphorus, calcium, calcium and phosphorus, and vitamin D3 to determine their effects on bone histology and parathyroid gland size. The phosphorus deficient diet caused an insignificant decrease in parathyroid size while the other diets caused significant increases. A considerable amount of individual variation in medullary bone volume and osteoid seam width was observed in all groups but, despite this, the calcium, calcium and phosphorus and vitamin D3 deficient diets clearly resulted in increased osteoid. Birds receiving diets deficient in calcium and phosphorus, and in vitamin D3 for longer periods were observed to have partially or completely resorbed medullary bone. Osteodystrophia fibrosa was noted in vitamin D3 deficient birds which had no follicular activity.  相似文献

This study was conducted to evaluate the effects of dietary vitamin K (menadione) on bone quality in cage-raised broilers. Three hundred and sixty male broilers were randomly allotted to one of six treatments, with six replicate pens per treatment and 10 chicks per pen. Broilers were fed one of six diets including a control diet or the control diet plus graded levels of vitamin K (0.5 mg/kg, 2 mg/kg, 8 mg/kg, 32 mg/kg and 128 mg/kg). Water and feed were provided ad libitum during the 7-week experimental period. Results indicated that vitamin K supplementation of broilers diets significantly effected bone quality and feed efficiency. The treatment containing vitamin K at 8 mg/kg improved growth performance (during weeks 6-7) and bone quality (during weeks 0-3). In our study, hydroxyapatite binding capacity of serum osteocalcin (during weeks 0-3), bone breaking strength, bone flexibility, bone ash weight increased linearly (P < 0.05) and bone mineral density, bone mineral content increased quadratically (P < 0.05) with increasing supplementation of vitamin K. In conclusion, to gain optimum bone quality and broiler performance, our studies suggest that the concentration of vitamin K in broilers diets should be 8 mg/kg, 2 mg/kg, and 2 mg/kg, for the starter, grower and finisher phases, respectively. Furthermore, it was shown that the starter period is an important phase for improving bone quality. In addition, this study validated the mechanism of vitamin K effects on bone quality. Vitamin K boosts the carboxylation of osteocalcin and decreases the concentration of serum under-carboxylated osteocalcin enhancing hydroxyapatite binding capacity of serum osteocalcin and improving bone quality.  相似文献

试验旨在探究虾青素和维生素B2在日粮中的添加量对鸡蛋品质的影响。选择1200只体重相近、健康的45周龄海兰褐蛋鸡,随机分成5组,每组4个重复,每个重复60只鸡。对照组蛋鸡饲喂基础日粮,试验Ⅰ、Ⅱ、Ⅲ、Ⅳ组蛋鸡饲喂固定比例的基础日粮和0.25%虾青素,混合不同含量的维生素B2(5.0、8.0、11.0、14.0 mg/kg)。预试期1周和正式试验期4周,预试期各组蛋鸡均饲喂基础日粮,正式试验期分别投放上面试验Ⅰ、Ⅱ、Ⅲ、Ⅳ组的配比的饲料。结果表明,试验组蛋黄颜色、哈夫单位均显著高于对照组(P<0.05)。试验Ⅱ、Ⅲ、Ⅳ组鸡蛋粗脂肪含量显著低于对照组(P<0.05)。试验组鸡蛋平均蛋重、蛋壳厚度、蛋壳强度、蛋比重与对照组相比有增大的趋势。试验表明,日粮添加虾青素和维生素B2能提高蛋品质,添加0.25%虾青素和14.0 mg/kg维生素B2效果最好。  相似文献

The aim of this study was to observe the expression and localization of estrogen receptor (ER) α and ER β mRNA in the medullary bone of laying hens. First, medullary bone, liver, kidney, and shell gland of the oviduct tissues were dissected from laying hens. Then, the total cellular RNA was isolated from each tissue specimen, and the ER α and ER β mRNA expression was observed using semiquantitative RT‐PCR. Second, the localization of ER α mRNA in the medullary bone was detected with in situ hybridization using digoxigenin‐11‐UTP‐labeled cRNA probes. As a result, the expression of ER α mRNA was higher than that of ER β mRNA in the medullary bone, liver, and shell gland of the oviduct from laying hens. In the kidney, ER α mRNA expression was lower than that of ER β mRNA. The expression pattern of ER α and ER β mRNA of the medullary bone was similar to that of the shell gland of the oviduct. Moreover, ER α mRNA was intensively expressed in osteoblasts on the medullary bone surface and bone marrow stromal cells but was not expressed in osteoclasts. These results suggest that in medullary bone, estrogen action may be regulated not by ER β but by ER α.  相似文献

This study was undertaken to determine what difference, if any, occurred when autogenous cancellous bone was placed in the medullary canal of a fresh, 4-cm cortical allograft compared to a fresh cortical allograft alone. Twelve mature dogs were used. A tubular cortical graft was placed in the midshaft of both tibias. The right tibia received the combination graft while the left tibia received a fresh cortical allograft only. Study periods ranged from one week to 17 weeks. The progress of the grafts was assessed by technetium-99m radionuclide bone scans, radiographs, gross and histological examination, and tetracycline labeling. Results indicate that autogenous cancellous bone placed in the medullary canal of a fresh cortical allograft was beneficial to the ingrowth of new blood vessels and incorporation of the cortical graft.  相似文献

试验探讨维生素E对免疫抑制蛋鸡生产性能和蛋品质的调控效应,选取健康的尼克褐产蛋鸡270只,随机等分为5组,每组3个重复,每重复18只。第Ⅰ组为对照组,第Ⅱ~Ⅴ组为免疫抑制组,对第Ⅱ~Ⅴ组鸡在试验第5、6、7 d进行腿部肌肉注射80 mg/kgBW环磷酰胺(CTX),第Ⅰ组注射等量生理盐水。Ⅰ、Ⅱ组饲喂基础日粮(维生素E含量44.59 mg/kg),Ⅲ、Ⅳ、Ⅴ组在基础日粮中分别添加50、100、200 mg/kg维生素E。结果显示:①免疫抑制产蛋鸡的产蛋率、平均蛋重和平均日采食量显著降低(P<0.05),破蛋率显著升高(P<0.05)。基础日粮中添加50 mg/kg维生素E显著提高免疫抑制产蛋鸡的产蛋率、平均蛋重和平均日采食量(P<0.05),但仍低于对照组Ⅰ(P<0.05);显著降低料蛋比和破蛋率(P<0.05)。②免疫抑制产蛋鸡的蛋壳厚度、蛋壳强度和哈氏单位显著降低(P<0.05),蛋形指数和蛋黄颜色无显著改变(P>0.05);50 mg/kg维生素E添加量显著提高免疫抑制产蛋鸡的蛋壳厚度、蛋壳强度和哈氏单位(P<0.05),与对照组Ⅰ无显著差异(P>0.05)。因此,免疫抑制显著降低产蛋鸡的生产性能和蛋品质;基础日粮中添加维生素E对免疫抑制蛋鸡的生产性能和蛋品质具有明显的调控效应,且以50 mg/kg添加量效果较好。  相似文献

1. A 6-week study was conducted to investigate the effects of phytase and hydroalcoholic extract of Withania somnifera root (WS) on productive performance and bone mineralisation of laying hens in the late phase of production.

2. Diets were arranged factorially (3?×?2?×?2) and consisted of a positive control with adequate Ca (4·37%) and nonphytate P (NPP; 0·39%) and a negative control diet with Ca (4·06%) and NPP (0·36 %); three concentrations of Withania somnifera (0, 65 and 130?mg/kg diet); and two concentrations of microbial phytase (0 and 300 U/kg diet).

3. A total of 144 72-week-old Hy-Line W36 laying hens were randomly assigned to the 12 treatment groups. Each treatment was replicated 4 times (4 x 3 hens). Egg production and egg weight were recorded daily, while feed intake and egg quality traits were recorded every two weeks. Bone quality traits were evaluated at the end of experiment.

4. Withania somnifera supplementation increased egg production and lowered egg weight only in the second two weeks of the experiment. Addition of phytase significantly depressed specific gravity of the eggs for the entire experiment period. No dietary treatment effects were observed on egg shell thickness and yolk weight.

5. Withania somnifera at 130?mg/kg did not affect feed intake. The hens fed on the positive control diet had higher albumen weight than the negative control diet in the second two-week period. Supplementation of the positive control diet with 65?mg/kg Withania somnifera in the absence of phytase significantly improved shell weight compared with the negative control (5·779 vs. 5·273?g respectively).

6. Supplementing Withania somnifera significantly improved Ca and P retention in tibia bone. In addition, an increase in tibia bone P was observed with phytase supplementation. There were significant interactions between Withania somnifera content and phytase for tibia bone Ca and P.

7. The results of this experiment indicated that dietary Withania somnifera has beneficial effects on tibia bone Ca and P content, and phytase improved tibia bone P retention without adverse effects on productive performance.  相似文献

Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two trials were conducted to investigate the synthesis and metabolism of AA in layers of different ages and the effects of dietary supplemental AA on the performance and the immune and antioxidant statuses of 78 weeks old hens.Methods: In Exp.1,equal numbers(24 hens) of 35 weeks old(Young) and 75 weeks old(Old) layers were fed the same diet without AA supplementation for 4 weeks.In Exp.2,360 healthy 78 weeks old laying hens were randomly assigned to 4 treatments(basal diet supplemented with 0,0.25,0.5,or 1 g AA/kg diet) in an 8-week feeding trial.Results: The old hens tended to have decreased L-gulonolactone oxidase(GLO) synthase activity in the kidney and liver than that of the young hens(P = 0.07 and P = 0.05,respectively).Compared with the young hens,the old hens had lower hepatic antioxidant capacity allowing for the lower thioredoxin(TXN),thioredoxin reductase(TXNR) and cytochrome b5 reductase(CYB5 R) gene expression(P 0.05),whereas increased sodium-dependent vitamin C transporter(SVCT) 1 expression levels in the ileum and kidney and enhanced splenic and hepatic AA concentrations(P 0.05).Dietary supplementation with AA significantly decreased GLO enzyme activity but increased splenic AA concentration and anti-bovine serum albumin IgG levels(P 0.05) and tended to increase CD4+T lymphocyte numbers(P = 0.06) in serum.Supplementation of 0.25 g AA/kg diet significantly increased hepatic total antioxidant capacity(T-AOC,P 0.05) relative to the control group.Conclusions: Laying hens could synthesize AA in both the kidney and the liver,though the GLO enzyme activities were 100 times greater in kidneys than in livers.The old laying hens had greater absorption and reabsorption capacity and higher AA retention in some tissues that did the young hens.Dietary supplementation of AA can improve the health of old layers by enhancing immunity and antioxidant capacity.  相似文献

试验研究了不同的维生素A(VA)添加水平对肉鸡和蛋鸡的生长性能及机体免疫功能的影响.试验选用1日龄的雌性蛋鸡和雌性肉鸡各270只,分别随机分成3个处理,每个处理6个重复,每个重复15只,分别饲喂含维生素A 0(对照)、4 000和20 000 IU/kg的日粮,饲养试验时间为84 d.试验结果表明,日粮添加维生素A为0 IU/kg处理的肉鸡和蛋鸡的采食最显著高于添加维生素A的各处理(P<0.05);日粮维生素A添加水平对肉鸡和蛋鸡的免疫器官指数(胸腺指数、脾脏指数和法氏囊指数)无显著影响(P>0.05);对照的肉鸡和蛋鸡异嗜粒细胞/淋巴细胞(H/L)高于其他处理(P>0.05);肉鸡和蛋鸡绵羊红细胞(SRBC)抗体滴度和新城疫(ND)抗体滴度随着日粮维生素A添加水平的升高而升高;日粮添加4 000 IU/kg维生素A处理的肉鸡和蛋鸡钥孔戚血蓝蛋白(KLH)抗体滴度、白蛋白浓度和白介素-2(IL-2)的浓度最高.蛋鸡的免疫器官指数和血清白蛋白浓度显著高于肉鸡(P<0.05);肉鸡和蛋鸡的其他免疫指标差异不显著(P>0.05).综上所述,日粮添加4 000 IU/kg维生素A能提高肉鸡和蛋鸡的生长性能和免疫功能,当添加水平达到20 000 IU/kg时,可对生长性能和免疫机能产生不利影响.肉鸡和蛋鸡的主要免疫指标没有最著差异.  相似文献

1. Ipriflavone (IP) has been widely studied in humans and is effective for inhibiting osteoclastic bone resorption and enhancing osteoblastic bone formation. The aim of this study was to investigate the effects of IP on the structural histology and histomorphometric parameters of tibia in laying hens fed on a low-calcium diet.

2. A total of 200 Hy-Line Brown laying hens, 24 weeks of age, were divided randomly into 4 groups which were fed on one of 4 diets (CaN, standard diet; CaL, low-calcium diet; IP1, low-calcium diet + 8 mg/kg of IP; IP2, low-calcium diet + 20 mg/kg of IP) for 60 d after which 10 hens of each group, chosen at random, were killed and the tibiae were processed and assessed under a fluorescent microscope

3. The cortical bone showing resorption cavities had become porous, and medullary bone did not completely fill the marrow cavity in the CaL group compared with the CaN group. However, in the IP2 group, the cortical bone showed a structure comparable to the CaN group with an absence of resorption cavities, and more medullary bone filled the marrow cavity compared with the CaL group.

4. The CaL hens had significantly lower trabecular bone area, trabecular perimeter, per cent trabecular area, trabecular number and trabecular thickness and significantly wider trabecular separation compared with CaN. These results indicated that the low-calcium diet led to increased loss of tibial bone, eventually causing osteoporosis. However, compared with the CaL group, the IP2 group caused a significant increase in trabecular bone area and bone quality.

5. It is concluded that even in hens fed on a low-calcium diet, the addition of 20 mg/kg IP exerted beneficial effects on histological structure and structural histomorphometric parameters of tibia, indicating an alleviation of caged layer osteoporosis (CLO).  相似文献

1. Line differences in the incidence of the presence of radiodense material in the central cavities of humeri of 6 different lines of laying hens were observed using radiographs. 2. Internal humeral radiodense material was observed in over 60% of hens in two lines and 18% of hens in two other lines measured at 65 weeks of age. There was no difference in the incidence (15, 16%) between the last two lines observed at 47 weeks of age. 3. In the 4 lines studied at 65 weeks of age the presence of radiodense material was associated with increased humerus dry weight and ash weight in two lines, while percentage-ash was reduced in all 4 lines. 4. Results from this and previous studies suggest that humeral medullary bone could be an appreciable source of variability in experimental data collected to assess skeletal health.  相似文献
